Abstract:
Ce memoire est consacre a l'obtention d'un materiel vegetal susceptible de fournir des protoplastes vegetaux, dans des conditions parfaites. Des feuilles, cotyledons, hypocotyles, racines et cals sont obtenus a la suite de microbouturages ou de germination de pepins in vitro sur 5 cultivars de la vigne: grenache, cabernet-sauvignon, clairette, carignan, rupestris du lot et un hybride de plantet (5455). L'isolement des protoplastes a ete realise a partir de ces explants eleves dans des conditions d'axenie rigoureuse et rendus plus reactifs que les memes organes preleves en vignoble sur les plantes-meres par cette acclimatation prealable a la culture in vitro. Pour l'isolement des protoplastes foliaires, nous avons utilise un melange enzymatique de maceration associant 1% de cellulase et 1,0% de driselase. Pour les autres organes 2% de cellulase et 0,5-1,0% de macerozyme. Plusieurs facteurs pouvant influencer l'isolement des protoplastes ont ete etudies en particulier pour les protoplastes issus de cals cotyledonnaires. Les protoplastes isoles sont purifies par la technique de flottaison sur une solution de 10% de ficoll centrifugee a 250 g pendant 20 min. Au cours de la culture des protoplastes: la regeneration d'une nouvelle paroi cellulaire, des phenomenes de "bourgeonnement" et de declenchement de trois divisions cellulaires successives, ont ete observes chez des protoplastes isoles a partir d'hypocotyles de grenache cultives dans le milieu liquide de kao & michayluk (8p) modifie par l'addition de 2,0 mg/l 2,4-d, 1,0 mg/l bap et 0,6 mannitol

Abstract:
This creative project and essay for a Masters degree in Media Theory and Practice is focused around the design of a South African TV series called 'Schedule 7' which is set in Cape Town's south peninsula in a (fictional) non-governmental, psychiatric clinic. The premise is that a troubled handful of patients' perspectives on life change after they encounter what they believe to be God's salvation when they accidentally almost burn themselves to death. The format follows their endeavours to follow and serve God, who they term 'GOGO' (God of Goodness Only), with mixed results. The format is an hour slot comedy-drama series designed for television broadcast (in that it is structured around commercial breaks). A comprehensive series bible is included, as well as two full sample episodes: the pilot and episode 3.Accompanying the creative components, the academic research essay firstly looks at the significance of mental illness: particularly in terms of stigma, South African context and places of treatment. It argues that an NGO psychiatric clinic makes for an interesting setting for a TV series as it allows characters from a range of socio-economic backgrounds to be rendered equal in power and united under a 'patient' status. This section also looks at mental illness intersecting with religion or faith. Secondly, the essay gives an overview of trends in and significant instances of on-screen representations of mental illness for which it offers a critique. Thirdly, in light of the issues raised surrounding mental illness, religion and the strengths and weaknesses of on-screen fiction around the subject so far, it offers an explication of the creative choices made in the design of the TV series, which were made with the aim of a resulting TV series that is enjoyable, relevant, diverse, and open-ended in its exploration of both experiences of mental illness and religious perception and practice.

Abstract:
Current planning and scheduling techniques are carried out in an unstructured form with considerable reliance on planners judgement, imagination and intuition. The final product of such techniques is typically a lengthy textural and tabular report and/or symbolic network. This serves as an abstract rather than visual modeling of the real construction process. The availability of advanced computer hardware and software allows us, today, to develop new planning and scheduling techniques to overcome the current limitations. Computer-Aided Design (CAD) is a computing system which makes extensive use of computer graphics. The use of CAD systems in construction presents a great opportunity for integrating engineering and construction processes in a more cost effective way. The combination of computer graphics, animation, and 3D computer modeling can be extremely effective for real-time simulation and visualization to support engineering and construction from the conceptual design to the construction process. This thesis presents a new planning and scheduling system. The system combines a construction scheduling network with 3D computer models to form a Visual Scheduling Simulation (VSS) of the construction process. The VSS system simulates, or put into motion, construction activities so they can be viewed at a graphics display. The VSS system allows the user to view the actual and planned construction sequence. The user has the option to view either: planned schedule; actual schedule; or both schedules shown side by side for quick visual comparison. The user has the option to visually simulate the entire construction project, or any specified time period. This simulation can be viewed for a partial segment or the entire configuration of the project.

We consider the problem of integrating flight schedule design and fleet assignment decisions at airlines. The flight schedule design problem involves selecting and scheduling the set of flight legs that an airline will include in its service network. Fleet assignment involves assigning a particular aircraft type to each flight leg in the schedule. Due to the particularly challenging nature of schedule design problems, we limit our focus to that of incremental schedule design. Incremental schedule design involves the modification of a given flight schedule to produce an improved schedule by adding, deleting, and rescheduling flight legs. We present models and algorithms to achieve incremental schedule design and unlike previous schedule design efforts, we explicitly model flight demand and supply interactions. We present two case studies, using our models and algorithms. The first case study allows flight additions and deletions, while the second allows flights to be rescheduled. Future case studies well integrate these flight modification options. In our first case study, high-yield flights are maintained in the schedule and low-yield flights are dropped. Although the resulting schedule incurs higher spill costs, the savings from flight operating costs are sufficiently large to offset these higher spill costs, resulting in a more profitable schedule. The second case study, allowing flights to be rescheduled, considers several network sizes including the domestic network of a large U.S. airline. We consider Free Flight, a system allowing reduced flying times due to improved utilization of the national airspace. We find that reductions in flying times of about 10% can lead to dramatic cost savings for the airline, including reductions in the number of aircraft needed to fly the flight schedule.

Abstract:
Airline schedule punctuality is a complex problem and one of the major concerns of the airline top management. Flight schedule disturbances may occur as delays and/or cancellations. There are many internal and external reasons for delays. These delays may propagate in the aircraft cycles and cause a large schedule disturbance. This may influences passenger satisfaction and airline resources. The objective of this research is to formulate a systematic approach for schedule punctuality which supports management decision making. The punctuality management system is structured to combine all schedule punctuality components, input and output variables. Five models are incorporated in this system. The first model is the disturbance model which generates random delays based on an estimated Lognormal delay distribution function. The delay analysis is carried out from a one year sample of delay statistics in which general, original , reactionary and other delay types are classified. The second model is the recovery model which incorporates the disturbance model with management strategies to determine delay propagation. A PC based simulation model (SKDMOD) is developed as a prototype which integrates disturbance and recovery models using SIMSCRIPT 11.5. 18 management strategies are simulated covering ground times (30, 40 and 50 minutes), maximum delay times to assign spare aircraft (1, 2, 3, 4, 5, and 6 hours) and spare aircraft using part of the domestic network of Saudi Arabia. The third model is the passengers' attitude model which determines the delay impact functions and the maximum passenger revenue loss based on 262 responses from a passenger interview survey. The fourth model is the revenue model which estimates the passengers' revenue loss. The fifth model is the cost model which estimates the extra cost resulting from implementation of the management strategies. All strategies are evaluated to determine the optimum based on profit and profit margin. OPTIM is the optimization program developed to find the optimum strategy(ies). This approach provides a guidelines for the management of punctuality. It integrates all the tools developed in a decision support system framework.

Abstract:
The education of to-be physicians at Akademiska sjukhuset, Uppsala, includes practical services. The students are divided into groups that each has its own goals. The goals specify (1) what services that group's students should perform, and (2) for each service, a minimum number of times each student should attend that service. It is only possible to perform any service at certain occasions: each occasion offers slots, to be filled by students. The occasions make up a calendar. The challenge is to distribute the students over the calendar, so that the goal isa chieved for each student and service. No occasion is overpopulated, and no student is due to attend two (or more) occasions that collide in time. The algorithm to solve this sets up a table with occasions (expanded horizontally by their number of slots) as the x-axis, and dates (expanded vertically by two: the day parts) as the y-axis. Then, distribution of students is done top-down, left-right. Collision is avoided by having students only appear once per row. Overpopulation won't happen as the allocation of students is done explicitly to slots, not to occasionsin general. MS Access forms make up the UI. My thoughts when I set them up was that each form should boil down to a single purpose, but include everything to fulfill that purpose (and nothing else). Also, I setup an intuitive flow of movements between forms, and I made an effort to setup mnemonic shortcuts (and tab chains) as to minimize mouse use

Scheelea phalerata Mart. ex Spreng (Arecaceae), popularly known as uricuri, is a native amazonian palm tree, which has seeds with significant amounts of oil that can be used in the production of biodiesel. This study aimed to chemically characterize the oilseed of uricuri in order to add value to this important non-timber forest product. Two lots of oilseeds were obtained from Comunidade do Roque, located in the Extractive Reserve of Juruá. The two lots of oilseed were submitted to standard physico-chemical methods used to analyze vegetable oil described by AOAC and Moretto and Fett: acid value, iodine value, peroxide value, saponification number, unsaponifiable matter and specific gravity. Lot 1 presented low acid value, whereas lot 2 a higher value. For both lots the unsaponified value was considered low. The fatty acid composition was obtained by GC-FID by two different methods and it could be observed that most of them were saturated with short carbon chain. In all experiments, biodiesel analysis was performed by HPLC with a UV detector (215 nm). In normal phase, it was not possible to separate all components of biodiesel mixtures. Moreover some of them have very different response to the detector; therefore it was not possible to analyze biodiesel by normal phase HPLC. In the reversed phase, the separation of all components could be accomplished with good separation and time analysis. The ester value was determined by the quantification of FAME by RP-HPLC using the external standard method. Lot 1, with low acid value, was converted into biodiesel by transesterification with methanol (molar proportion of 1:6) employing three different basic catalysts. The best result, considering the yield, the quality determined by the specific gravity and the conversion determined by ester value, was obtained using 1 % NaOH at 50 ºC for 2 h. The acid catalysis in the transesterification process was studied with lot 2 due to its higher acid value. Three variables were evaluated: type of alcohol (methanol vs. ethanol), temperature and time. The best condition was achieved with ethanol at 75 ºC, using as catalyst 1M of HCl. Some of the components of unsaponifiable matter were isolated by chromatography and their characterization was done by MS and NMR experiments (1H and 13C, COSY, HSQC, HMBC). In fraction Fr4 a mixture of hydrocarbons was identified by GC-MS. In fraction Fr7, a mixture of methyl esters was identified by GC-FID. In fraction Fr14, a cis/trans mixture of 9-10-epoxy-stearic acid was identified.
A Scheelea phalerata Mart. Ex Spreng é conhecida popularmente como uricuri, pertence à família Arecaceae, uma palmeira, cujas amêndoas de seus frutos apresentam grande potencial de óleo, podendo ser utilizado como matéria-prima na produção de biodiesel. Este trabalho teve como objetivo caracterizar o óleo extraído da sua amêndoa, a fim de agregar valor a esse produto. Dois lotes de óleo foram obtidos da Comunidade do Roque localizada na Reserva Extrativista do Juruá (AM). A caracterização físico-química do óleo foi realizada utilizando métodos oficiais, índices de acidez, índice de iodo, índice de peróxido, índice de saponificação, determinação da massa específica e do material insaponificável. O lote 1 apresentou baixo índice de acidez enquanto o lote 2 apresentou índice de acidez elevado. O teor de material insaponificável foi baixo para os dois lotes. A composição da cadeia graxa obtida por CG-FID por dois métodos, apresentou a predominância de ésteres metílicos de ácidos graxos saturados e de cadeias curtas. A análise da qualidade do biodiesel foi realizada por CLAE em modo normal e reverso com um detector de UV (215 nm). O método em modo normal desenvolvido não poderá ser empregado na análise de conversão de biodiesel porque os diferentes componentes de uma mistura de biodiesel apresentam resposta diferente ao detector e vários dos FAMEs coeluem. Na análise em modo reverso foi possível identificar os FAMEs, com boa resolução e rapidez de análise. A quantificação dos ésteres de ácidos graxos foi realizada por padronização externa e através do teor de éster foi possível obter a pureza do biodiesel. O lote 1, com acidez baixa foi submetido a reação de transesterificação por catálise básica avaliando o emprego de três diferentes catalisadores pela via metílica na proporção molar de 1:6 (óleo:álcool). O melhor resultado observado em termos de rendimento mássico, densidade e conversão em ésteres foi utilizando 1% de NaOH como catalisador a 50 ºC por 2 horas. A transesterificação por catálise ácida foi realizada para o lote 2, devido seu elevado índice acidez. Três variáveis foram avaliadas: o tipo de álcool, que foi usado, metanol e etanol, na proporção molar de 1:9 (óleo:álcool), a temperatura e o tempo. A melhor condição em termos de rendimento mássico, densidade e conversão em ésteres foi pela via etílica a 75 ºC, utilizando 1 M de HCl como catalisador. A extração do material insaponificável realizada em maior escala gerou um extrato hexânico onde este foi submetido a um fracionamento em coluna de sílica flash. As três frações, Fr4, Fr7 e Fr14 foram caracterizadas por CG-EM e RMN (1H e 13C, COSY, HSQC, HMBC). A fração Fr4 foi identificada como uma mistura de hidrocarbonetos, a Fr7 como uma mistura de ésteres de ácidos graxos e a Fr14 como uma mistura de isômeros metil-cis-9,10-epoxiesteárico e metil-trans-9,10-epoxiesteárico.

Abstract:

Dynamic model driven architecture (DMDA) is a architecture made to aid in the

development of parallel computing code. This thesis is applied to an implementation of

DMDA known as DMDA3 that should convert graphs of computations into efficient

computation code, and it deals with the translation of Platform Specific Models (PSM)

into running systems. Currently DMDA3 can generate schedules of operations but not

finished code.

This thesis describes a DMDA3 module that turns a schedule of operations into a

runable program. Code was obtained from the DMDA3 schedules by reflection and a

framework was build that allowed generation of low level language code from

schedules. The module is written in Java and can currently generate C and Fortran code

for computational tasks. Based on runtime tests for matrix multiplication algorithms the

generated code is almost as fast as handwritten code.

Abstract:
This thesis extends the body of research into the application of Genetic Algorithms to the Project Scheduling Problem (PSP). A thorough literature review is conducted in this area as well as in the application of other similar meta-heuristics. The review extends previous similar reviews to include PSP utilizing the Design Structure Matrix (DSM), as well as incorporating recent developments. There is a need within industry for optimisation algorithms that can assist in the identification of optimal schedules when presented with a network that can present a number of possible alternatives. The optimisation requirement may be subtle only performing slight resource levelling or more profound by selecting an optimal mode of execution for a number of activities or evaluating a number of alternative strategies. This research proposes a unique, efficient algorithm using adaptation based on the fitness improvement over successive generations. The algorithm is tested initially using a MATLAB based implementation to solve instances of the travelling salesman problem (TSP). The algorithm is then further developed both within MATLAB and Microsoft Project Visual Basic to optimise both known versions of the Resource Constrained Project Scheduling Problems as well as investigating newly defined variants of the problem class.

Os parques eólicos devem periodicamente desligar suas turbinas para realizar as manutenções agendadas. Uma vez que esta interrupção afeta a geração de energia e qualquer déficit na produção deve ser coberto por compras de energia no mercado spot, determinar o tempo ótimo para iniciar o trabalho de manutenção em um parque eólico é fundamental para maximizar sua receita, considerando que é função tanto da velocidade do vento esperada como dos preços spot da eletricidade. Neste trabalho, desenvolvemos um modelo para determinar o momento ideal para manutenção em um parque eólico. Analisamos uma janela de oportunidade no período mais provável do ano e realizamos atualizações semanais das velocidades esperadas do vento e previsões de preços de energia. As velocidades do vento são previstas com um modelo ARIMA enquanto os preços spot são simulados sob o modelo de programação estocástica dupla Newave. A decisão de adiar a manutenção para uma data futura é modelada como uma opção real americana. Testamos dois modelos com dados reais de um parque eólico no Nordeste brasileiro e comparamos nossos resultados com a prática atual e com o agendamento de manutenção considerando informações perfeitas para determinar os benefícios do modelo. Os resultados sugerem que esses modelos podem oferecer vantagens significativas em relação a uma decisão de parada que escolhe aleatoriamente uma semana para começar a manutenção dentro da janela de oportunidade e está perto da data de parada ideal, considerando o modelo de informação perfeita.
Wind farms must periodically take their turbines offline in order to perform scheduled maintenance repairs. Since this interruption impacts the generation of energy and any shortfall in production must be covered by energy purchases in the spot market, determining the optimal time to start maintenance work at a wind farm is key to maximizing your revenue, which is a function of both the expected wind speeds and electricity spot prices. In this study we develop a model to determine the optimal maintenance schedule in a wind farm. We analyze a window of opportunity in the most likely period of the year and perform weekly updates of expected wind speeds and energy price forecasts. Wind speeds are forecasted with an ARIMA model, while spot prices are simulated under the Newave dual stochastic programing model. The decision to defer maintenance to a future date is modeled as an American real option. We test two models with actual data from a wind farm in the Brazilian Northeast, and compare our results with current practice and with maintenance scheduling considering perfect information in order to determine the benefits of the model. The results suggest that the models may provide significant advantages over a stopping decision that randomly chooses a week to begin maintenance within the opportunity window and is close to the ideal optimal stopping date considering perfect model.
