Abstract:
Background:in SSc, PAH has a high morbidity and mortality burden. Therefore, screening and early detection are pivotal to achieve an early diagnosis of PAH.Objectives:to search the literature for all screening modalities for SSc-PAH in reference to right heart catheterization as diagnostic gold standard. Methods:papers from 2 previously published SLRs [22 fromGlaude et al(1) – from inception to 19/06/2012 - and 22 fromYoung et al 2018– from 20/06/2012 to 02/10/2017] were included. The articles’ database was integrated with a systematic search on Pubmed, EMBASE, Web of Science for papers published from 03/10/2017 to 31/12/2018. A total of 199 papers were reviewed and 32 were finally extracted. Bias risk was assessed through QUADAS2 tool.Results:167 papers were excluded from data extraction mainly for PAH screening non as main focus or for non-including SSc patients. The 32 papers extracted presented a low bias risk according to QUADAS2. Screening methods reported were:Echocardiographic parameters in 31/32 studies, in particular systolic pulmonary arterial pressure (sPAP) in 22 papers; 40 mmHg was the most frequently used cut-off (in 12/22 papers); sPAP was part of a composite algorithm in 9/22 papers. Among others, tricuspid regurgitation velocity (TRV) was used in 6/31 (as part of composite 5/6) and right atrial pressure (RAP) in 3/31 papers.Pulmonary function tests parameters in 22/32 papers, with % predicted Lung diffusion for carbon oxyde (DLco) in 21 papers, with a 50% cut-off in 11/21 and as part of composite algorithm in 13/21 studies. Moreover, walked distance at six minutes walking test was a screening parameter in 3/32 papers.Serum biomarkers in 12/32 papers, with anti-centromere antibodies (6/12), NT-proBNP (6/12) and uric acid (5/12) being the most frequently reported.Clinical parameters in 15/32 papers, with unexplained dyspnoea in 9/15 and telangiectasias in 5/15 papers.Composite algorithms were used in 18/32 manuscripts: among them, DETECT (5/18), ESC/ERS 2009 (4/18) or 2015 (3/18) guidelines, ASIG (2/18) e ITINER-air (1/18). In different cohorts, DETECT and ASIG showed higher sensitivity and negative predictive value than ESC/ERS 2009.Conclusion:in the literature, the screening of SSc-PAH is largely investigated by echocardiographic parameters. In particular, sPAP and TRV, both as single items or part of a composite algorithm, including also serum biomarkers, clinical and functional parameters, are the most frequent parameters evaluated.(supported by an Actelion Pharmaceuticals unrestricted research grant)References:[1]Gladue H, et al. Abstract:
Background and Objectives: Coal mine injuries commonly occur, affecting both the safety and health of miners, and the normal operation of the coal mine. Accordingly, this study aimed to explore the regularity of injury and injury-related risk factors in coal mines in China so as to establish a scientific basis for reducing the incidence and promoting the prevention and control of injuries. Methods: A meta-analysis of casualty cases and injury-related risk factors from 1956 to 2017 in China was conducted utilizing data from six databases, including CNKI, Web of Science, PubMed, Medline, Embase, and Wanfang data. Summary estimates were obtained using random effects models. Results: There were statistically significant variations in coal mine accident types, types of work, injury sites, age, experience, months, and shifts (p < 0.001). Eight types of accidents were susceptible to the risk of injury, and the greatest risk was presented by roof-related accidents (odds ratio (OR) = 0.46, 95% confidence interval (CI) = 0.32–0.6). Coal miners and drillers were at a greater risk of injury (OR = 0.39, 95% CI = 0.35–0.44; OR = 0.22, 95% CI = 0.17–0.26, respectively). The extremities and the soft tissues of the skin were at the greatest risk of injury (OR = 0.44, 95% CI = 0.3–0.58; OR = 0.23, 95% CI = 0.1–0.48, respectively). Compared with other ages, miners aged 21–30 were at a greater risk of injury (21–30 years, OR = 0.45, 95% CI = 0.42–0.47; 31–40 years, OR = 0.29, 95% CI = 0.25–0.32; <20 years, OR = 0.13, 95% CI = 0.03–0.23; >40 years, OR = 0.17, 95% CI = 0.09–0.25). Compared with other miners, those with 6–10 years of experience were at a greater risk of injury (6–10 years, OR = 0.29, 95% CI = 0.25–0.32; 2–5 years, OR = 0.33, 95% CI = 0.25–0.41; <1 year, OR = 0.22, 95% CI = 0.08–0.33; >11 years, OR = 0.22, 95% CI = 0.17–0.27). During the months of July to September, the risk of injury was elevated (7–9th months, OR = 0.32, 95% CI = 0.25–0.39; 10–12th months, OR = 0.24, 95% CI = 0.16–0.31; 1st–3rd months, OR = 0.22, 95% CI = 0.16–0.28; 4–6th months, OR = 0.21, 95% CI = 0.16–0.27). In the three-shift work system, the risk of injury was higher during night shifts (22:00–06:00, OR = 0.43, 95% CI = 0.3–0.56; 14:00–22:00, OR = 0.3, 95% CI = 0.23–0.27; 06:00–14:00, OR = 0.27, 95% CI = 0.18–0.35). Conclusions: The results of this research study reveal that coal mine injuries are prevalent among coal miners. These injuries are often related to the age, experience, months of work, and the three-shift work system of miners.

Abstract:
The article deals with the notion of manipulation in advertising slogans of fast food restaurants. It focuses on techniques and means of language influence on the recipient. The article shows results of the study of 239 advertising slogans of fast food restaurants in English-speaking countries (a total number of 104 companies). Different language patterns which are used in advertising slogans and aimed at persuasion of customers on the subconscious level, are defined and analyzed; among them the most numerous are language patterns which create statements with distortions, deletions, and generalizations manifesting themselves in a variety of forms. The research also describes manipulative language patterns specifically related to advertising, such as unique selling proposition or life values. The article also shows quantitative proportions of the use of each pattern, as well as patterns used by the most successful and less successful restaurants. The most frequent of the manipulative language patterns are lost performatives, mind reading, and comparisons. The techniques of life values, mind reading, and antithesis are more common in advertising slogans of top restaurants compared to less popular ones. Individual slogans frequently manifest the use of more than one linguistic pattern of manipulation. References Alder, H. (2002). Handbook of NLP: A Manual for Professional Communicators. Abstract:
Computational Thinking (CT) is a set of logical-operational cognitive skills or processes of reasoning, based on Computer Science. Abstraction, pattern recognition, algorithmic reasoning, and decomposition are examples of some of these skills that form the four pillar of CT. Some researchers have considered these skills as useful, and even mandatory to to cognitive development of the schoolchildren. In this paper, we present practical aspects and the possible contributions of CT in the development of competence of reading and interpreting English texts. Didactic interventions were carried out in high school classes of a public school, supported by the Bring Your Own Device (BYOD) approach, in which the students used their own smartphones. During these interventions, the students developed concept maps and podcasts, performed online exercises and the traditional exam, all of that composed the set of evaluation instruments. It was possible to understand that the CT skills are intrinsically present and contributed to the development of the reading and writing skills in English. According to testimonials, we highlight that the BYOD approach provided new conceptions and perspectives on the use of electronic equipment in function of the students’ learning. Keywords: Computational thinking, English teaching, Mobile learning, Computer science education.ReferencesALBERTA Education. Abstract:
Children's stunting is a problem because it is associated with an increased risk of pain and death, suboptimal brain development resulting in late motor development, and inhibition of mental growth. This study aims to examine the factors that influence the occurrence of stunting and risk in children in Indonesia. The research method used is a traditional review. The results of the review found that many factors cause the high incidence of stunting in toddlers, including energy intake, birth weight, maternal knowledge level, family economic status, parental parenting, and food security. WHO estimates the prevalence of stunting worldwide at 22 percent or as many as 149.2 million in 2020. According to the Indonesian Nutritional Status Study (SSGI) results of the Ministry of Health, the prevalence of stunting in children under five in Indonesia reached 24.4% in 2021. The prevalence of stunting of toddlers in Indonesia continues to show a decline. In 2018 the prevalence of stunting toddlers was still 30.8%. Then, it dropped to 27.7 in 2019 and continued to fall to 24.4% in SSGI 2024. The government even targets to drop to 14% by the end of 2024. Furthermore, WHO also recommends reducing stunting by 3.9% per year to meet the target of 40% stunting reduction by 2025. Abstract:
Introduction: Acute myeloid leukemia (AML) is characterized by both aberrant proliferation and differentiation arrest at hematopoietic progenitor stages 1,2. AML relies upon de novo nucleotide synthesis to meet a dynamic metabolic landscape and to provide a sufficient supply of nucleotides and other macromolecules 3,4. Hence, we hypothesized that inhibition of de novo nucleotide synthesis would lead to depletion of the nucleotide pool and pyrimidine starvation in leukemic cells compared to their non-malignant counterparts and impact proliferative and differentiation inhibition pathways. PTC299 is an inhibitor of dihydroorotate dehydrogenase (DHODH), a rate limiting enzyme for de novo pyrimidine nucleotide synthesis that is currently in a clinical trial for the treatment of AML. Aim: We investigated the pre-clinical activity of PTC299 against AML in primary AML blasts and cytarabine-resistant cell lines. To confirm that PTC299 effects are due to inhibition of de novo pyrimidine nucleotide synthesis for leukemic growth, we specifically tested the impact of uridine and orotate rescue. In addition, a comprehensive analysis of alteration of metabolic signaling in PI3K/AKT pathways, apoptotic signatures and DNA damage responses were analyzed by Mass cytometry based proteomic analysis (CyTOF) and immunoblotting. The potential clinical relevance of DHODH inhibition was confirmed in an AML-PDX model. Results: The IC50s for all tested cell lines (at 3 day) and primary blasts (at 5-7 day) were in a very low nanomolar range: OCI-AML3 -4.43 nM, HL60 -59.7 nM and primary samples -18-90 nM. Treatment of AML in cytarabine-resistant cells demonstrated that PTC299 induced apoptosis, differentiation, and reduced proliferation with corresponding increase in Annexin V and CD14 positive cells (Fig.1). PTC299-induced apoptosis and inhibition of proliferation was rescued by uridine and orotate. To gain more mechanistic insights, we used an immunoblotting and mass cytometry (CyTOF) based approach to analyze changes in apoptotic and cell signaling proteins in OCI-AML3 cells. Apoptotic pathways were induced (cleaved PARP, cleaved Caspase-3) and DNA damage responses (TP53, γH2AX) and the PI3/AKT pathway were downregulated in response to PTC299. In isogenic cell lines, p53-wildtype cells were sustained and an increased DNA damage response with corresponding increase in apoptosis in comparison to p53-deficient cells was shown. (Fig.2) In a PDX mouse model of human AML, PTC299 treatment improved survival compared to mice treated with vehicle (median survival 40 days vs. 30 days, P=0.0002) (Fig.3). This corresponded with a reduction in the bone marrow burden of leukemia with increased expression of differentiation markers in mice treated with PTC299 (Fig.3). Conclusion: PTC299 is a novel dihydroorotate dehydrogenase (DHODH) inhibitor that triggers differentiation, apoptosis and/or inhibition of proliferation in AML and is being tested in a clinical trials for the treatment of acute myeloid malignancies. Abstract:
Empirical studies proved that good corporate governance had a positive link to the success of an organization. Good in directing and managing organizational processes, structures, and information mean satisfaction guaranteed was given to all stakeholders. Essential factor like working atmosphere may be considered when looking into corporate governance as well as organizational success. However, limited literature focused on the importance of communication climate and its connection to corporate governance and organizational success. Hence, this study looked into the predicting effect of communication climate to perceived corporate governance and organizational success. A total of ninety-eight rank and file employees voluntarily participated in accomplishing three questionnaires such as the Communication Climate Inventory by Costigen & Schiedler; Corporate Governance Survey adapted from Wickramanayake’s Seven Characteristics of Corporate Governance; and Organizational Success Survey adapted from Ilyas & Rafig used to measure communication climate, corporate governance, and organizational success respectively. Results revealed that communication climate was a predictor of perceived corporate governance and organizational success. This implied that the role of supervisors in setting work conditions was essential on how rank and file employees perceived corporate governance and organizational success.ReferencesAbdu Rashid, M., Azman bin Othman, M., Zainudin bin Othman, M., Ain bt Arshad, F. Abstract:
ABSTRACT The objective of this research was to evaluate advanced potato clones for tuber yield, frying quality, plant vigor, and maturity, grown from minituber seeds, and to assess correlations between these traits. Eight advanced potato clones of Embrapa Potato Breeding Program (F11-09-03, F05-11-03, F54-11-06, F141-11-01, C2718-12-09, C2718-24-09, C2743-09-09, and CL308) and three commercial cultivars (Agata, Asterix and BRSIPR Bel) were evaluated. Experiments were carried out in autumn 2017, in Canoinhas-SC and Pelotas-RS. A randomized complete block design with four replicates was used. We used tuber seeds type IV (minibubers). We evaluated plant vigor, maturity, yield and number of marketable tubers, yield and number of total tubers, average tuber weight, specific gravity, and frying color. Individual and joint variance analyses revealed significant differences (p <0.05) among genotypes for all traits at both locations. The GxE (genotype x environment) interaction was significant for all traits. For the two locations together, clones F05-11-03, C2718-12-09, C2718-24-09 and the control cultivar BRSIPR Bel showed higher marketable tuber yields. The first two clones also showed tubers with high average tuber weight. Clone F54-11-06 showed the later maturity at both locations. Regarding the frying quality characteristics, only clone F141-11-01 showed a pattern comparable to that of the control cultivar Asterix. Also, clones with higher plant vigor had higher tuber yield, but late maturity. Frying quality traits were not correlated between them and among other traits.

Abstract:
Introduction: Cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy (CADASIL), is a non-atherosclerotic, nonamyloid, hereditary cerebral disease of small vessels and capillaries caused by mutations in the NOTCH-3 gene located on chromosome 19. The presence of granular osmophilic material (GOM) deposition in the smooth muscle cells of vessel walls is the pathological hallmark of arteriopathy in CADASIL. GOM deposits in the basal lamina of smooth muscle of small vessels are pathognomonic for CADASIL. The presence of GOM in capillary blood vessels of the skin and muscle in biopsy and genetic studies (NOTCH-3 analysis) plays a key diagnostic role. Biopsy tests have high specificity (up to 100%) and low sensitivity (less than 50%). The NOTCH-3 test has been proposed as the primary diagnostic approach, allowing detection of 90% of affected individuals. CADASIL has an estimated prevalence between 2 and 5 in 100,000, and the phenotypic study demonstrates different clinical symptoms in the course of the disease within the same family. The average age of onset of clinical symptoms varies between 48.3 years in men and 52.2 years in women. Characteristic symptoms of migraine, stroke or TIA (transient ischemic attack), behavioral changes, early and progressive cognitive changes associated with leukoencephalopathy on imaging studies. The large association of symptoms often causes the diagnosis of CADASIL to be delayed. In this case, we had an association of gait ataxia within the framework of motor alterations, demonstrating the wide range of symptomatology of the pathology. This case report presents a familial course that started outside the most prevalent age group in the studies described and with an atypical presentation in an affected generation. Case reports: Case 01: woman, 68 years old, started progressively forgetting to perform household activities after the age of fifty, associated with primarily generalized myoclonic epileptic seizures, evolving rapidly within five years to walking apraxia with the use of a wheelchair and tonic-tonic epileptic seizures. bilateral clonic disorders, comprehension aphasia and bradypsychism. Relatives report previous migraine without chronic aura and REM (rapid eye movement sleep) sleep behavior disorder (RMSD). On neurological examination, severe ataxia with bilateral dysdiadochokinesia associated with bilateral hypometric index-index. Bilateral ROT 4+/4+ with bilateral Hoffman and Babinski signs. MMSS and MMII with FGM 4/5 proximal and distal. Case 02: woman, 36 years old, pastry chef, had episodes of forgetting about everyday activities of her work, progressive in the last three years (cake recipes, budget accounts, orders placed) associated with confusion for spatial location on the way home/ work, evolving to apraxia in writing letters and words and difficulty with calculations associated with monoparesis of the right lower limb for twelve months with progression to paresis of the lower limbs after six months and evolution to paresthesia of the upper limbs for three months. Associated with the condition, he has migraine without chronic aura and RMSD. The neurological examination showed Mini-Mental State Examination 22/30 (expected score of 29), list of animals in one minute: 09 animals; list of words starting with “F”: 03 words; clock test: 2/4; difficulty with calculations and digital agnosia with right/left apraxia; Upper limbs: eutrophic, FMG 4/5, bilateral distal; FMG 5/5 bilateral proximal; Lower limbs: eutrophic, bilateral FMG 4/5 distal and proximal with positive Mingazini; atypical gait with evidenced weakness in heel, toe and tandem gait; Bilateral dysdiadochokinesia with eumetric, slowed indexindex; ROT 4+/4+ in the right side with positive Hoffman and Babinski signs. MRI Brain (17/09/2020): extensive area of hypersignal on T2 and FLAIR (T2- weighted-Fluid-Attenuated Inversion Recovery) involving the periventricular white matter in all lobes without atrophic or expansive effect; Case 03: woman, 45 years old, started behavioral arrest epileptic seizures at the age of thirty-two, progressing to focal dysperceptive seizures with progression to bilateral tonic clonic seizures after eight months and multiple episodes of anterograde amnesia, presenting forgetfulness related to everyday work activities (exchanged worksheets , payments, calculation errors and budgets); associated with the condition presented migraine without chronic aura and RMSD. Genetic Test (04/06/2017): Heterozygous alteration in exon 8 of the NOTHC – 3 gene. Discussion: CADASIL presents a rare cause of cognitive decline and is often overlooked in diagnosis, except in cases of high clinical suspicion in a familial course. Access to imaging tests becomes fundamental for the diagnostic segment and the primordial genetic test for etiological elucidation and family planning, in the report described the family presentation with the same course of satellite symptoms (migraine without aura, and RMSD) associated with cognitive alteration with anticipation of age of onset are hallmarks of clinical thinking. Cases described in the literature show that the clinical symptomatology is not necessarily related to the level of brain injury observed in the imaging exam, which could be explained by personal factors and which exon is affected. The NOTCH-3 gene has 24 exons, in which the literature reports exon 4 as the most common mutation, followed by 3, 5 and 6, mainly in the Caucasian population. In an Asian population, the most affected exon is 4 and 11, which is also found in Italian descendants. The mutation in exon 8, described in the clinical case, is found in a population of Portuguese origin, being the second most common mutation in this nationality, behind the mutation in exon 4. Brain MRI studies have tried to elucidate the most affected brain regions, aiming to trace a line of evolution. Involvement of the temporal lobe, external capsule and corpus callosum are described as probable markers of CADASIL, and can be used as an aid in the diagnosis due to its specificity of 86% and sensitivity of 89%. Studies also show that the frontoparietal area has frequent findings of hyperintensity (100%), followed by the temporal lobe (83%), less frequently affecting the brainstem, occipital lobe and cerebellum. Despite the great advances in the specialized literature, the causes of the important cognitive dysfunction presented in the course of the evolution of CADASIL remain unclear. However, studies have suggested that the process of cognitive decline is more related to the loss of cortico-subcortical connections than to brain atrophy itself, with these disconnections resulting from repeated transient ischemic accidents. Science has been looking for ways to change the prognosis of CADASIL, recent studies in gene therapy and neurogenetics show the importance of thinking about this pathology as a genetic disease of great importance to change the prognosis of this pathology.
