Abstract:
The research for exact solutions of mixed integer problems is an active topic in the scientific community. State-of-the-art MIP solvers exploit a floating- point numerical representation, therefore introducing small approximations. Although such MIP solvers yield reliable results for the majority of problems, there are cases in which a higher accuracy is required. Indeed, it is known that for some applications floating-point solvers provide falsely feasible solutions, i.e. solutions marked as feasible because of approximations that would not pass a check with exact arithmetic and cannot be practically implemented. The framework of the current dissertation is SCIP, a mixed integer programs solver mainly developed at Zuse Institute Berlin. In the same site we considered a new approach for exactly solving MIPs. Specifically, we developed a constraint handler to plug into SCIP, with the aim to analyze the accuracy of provided floating-point solutions and compute exact primal solutions starting from floating-point ones. We conducted a few computational experiments to test the exact primal constraint handler through the adoption of two main settings. Analysis mode allowed to collect statistics about current SCIP solutions' reliability. Our results confirm that floating-point solutions are accurate enough with respect to many instances. However, our analysis highlighted the presence of numerical errors of variable entity. By using the enforce mode, our constraint handler is able to suggest exact solutions starting from the integer part of a floating-point solution. With the latter setting, results show a general improvement of the quality of provided final solutions, without a significant loss of performances.

Cells must sense changes in their environment and respond appropriately in order to survive. A common survival strategy is for cells to translate an environmental signal into the activity of a transcription factor they effects a change in gene expression. In this way, cells can express a gene just-in-time for its biological function. In addition, cells can coordinate the activity of many transcription factors to construct genetic regulatory networks that integrate many inputs to control complex cellular behaviors. In this thesis, I use the model system Caulobacter crescentus to examine the regulation of CtrA, the essential transcription factor at the core of the Caulobacter cell cycle regulatory network. CtrA regulates the activity of approximately 100 cell cycle genes many of which are critical for cell cycle progression. CtrA activity is regulated at the level of abundance, post-transcriptional modification, and here I show that CtrA is also regulated by a novel protein-protein interaction. I identify SciP, a GI specific inhibitor of CtrA transcriptional activity and show that SciP forms a complex with CtrA at CtrA dependent promoters. The SciP/CtrA interaction likely prevents CtrA from recruiting RNA polymerase thereby blocking the activation of transcription. In addition, I show that SciP is restricted to G1 by regulated proteolysis. I identify the Lon as the SciP protease and show that Lon is required for SciP proteolysis in vivo and that E. coli Lon degrades SciP in vitro. Finally I engineer a stable allele of sciP and show SciP proteolysis is critical for proper cell cycle progression.

Abstract:
Tactical radio communications used in military applications hasstrict requirements regarding security and has to be operable inrough environments in which there may be disturbances and disruptionson a radio link. The performance of the Secure CommunicationInteroperability Protocol (SCIP) operating in an asynchronouscommunication network with various levels of packet loss isinvestigated and found inadequate mainly due to problems withcryptographic synchronization between the transmitting and receivingunits. The introduction of additional counter data to each datapacket remedies this problem and allows the receiving units to fillthe holes left by packet losses with filler packets, maintainingsynchronization. The audio quality can then be measured using thePerceptual Evaluation of Speech Quality (PESQ) algorithm.Measurements are performed in an emulated radio link with aconfigurable packet loss ratio developed by Saab. The results showthat parts of SCIP can be used alongside the counter solution withoutimpacting the audio quality. The insertion of filler packets is shownto have a positive effect on the audio quality, while aggregation ofpackets to conserve transmission data rate is shown to have anegative effect.

Stoodley, Lynda. "Profiling Factors and Outcomes of Hyperglycemia After Cardiac Surgeries: An Important Step to Improve a Quality Measure." Diss., The University of Arizona, 2013. http://hdl.handle.net/10150/297030.

Abstract:
Background: Elevated blood glucose in patients undergoing cardiac surgery pose increased risks of sternal incision infections, kidney disease, irregular heartbeats, extended hospital stays, and death. Elevated blood glucose also increases the costs of healthcare from prolonged lengths of stay and increased complications. The Surgical Care Improvement Project (SCIP) #4 performance measure is used to evaluate achievement of a target blood glucose control < 200 milligram/deciliter (mg/dl) post cardiac surgery on postoperative days 1 and 2. In the institution where this study was carried out, blood sugar control in the cardiac surgery patient has presented a challenge. Purpose: The purpose of this practice inquiry was to identify patient characteristics and outcomes in cardiac surgery patients who met the SCIP #4 performance measure versus those patients who did not. Methods: A retrospective nested case-control design was used. Risk factors for postoperative hyperglycemia and in-hospital outcomes were compared between cardiac surgery patients who were SCIP #4 met defined as 6AM BG ≤ 200 mg/dl on postoperative days 1 and 2 and those that were SCIP #4 not met, defined as 6AM BG > 200 mg/dl on postoperative days 1 or 2. Results: Results from this study showed that preoperative hemoglobin AIC and history of diabetes were two major contributors for SCIP #4 not-met status. There was a trend towards a longer length of stay in the SCIP #4 not met group as compared to the met group (9.01 ± 7.33 versus 7.30 ± 4.93 days, respectively; p = . 096). Mortality was 3 times more prevalent in the SCIP #4 not met than the met group (6.2% versus 2.1%, respectively); however, this different did not reach statistical significance (p = .129). Renal failure was four-fold more frequent in patients who were SCIP #4 not-met than who were SCIP #4 met (13.6% vs. 4.1%, respectively; p = 0.003). Conclusions: Results from this study showed that SCIP #4 not met is associated with development of postoperative renal failure in the hospital and a trend towards longer length of stay. History of diabetes and preoperative hemoglobin AIC level should be taken into consideration when evaluating strategies for managing hyperglycemia. Future research is needed to study the relationship between SCIP #4 met status and long-term outcomes. The use of preoperative hemoglobin A1C to identify patients at high risk for uncontrolled postoperative glucose and plan effective glucose control should be studied. Such study may include implementing intravenous insulin on all patients with diabetes and elevated hemoglobin A1C levels and comparing the short and long term outcomes.

Abstract:
Reconstructive Surgery techniques seek to achieve two main objectives, repair body defects in the most appropriate way and minimize the donor area lesion. The microsurgical techniques allowed the development and popularization of the free flaps, enabling reconstructions in areas where previously was not possible. The imaging techniques evolution allowed greater sensitivity and specificity in the identification and selection of the perforating vessels that feed the flaps. The most useful imaging test in this identification is multidetector computed tomography (MDCT). Two of the most complex body areas for reconstruction are the distal portion of the limbs and head and neck. They have a thin skin, with thin adipose paniculus and the lesions in those places require very complex three-dimensional reconstructions. The search for a thin, broad, malleable flap with a constant vascular anatomy and minimal donor site lesion led to the development of the SCIP perforator flap. The inguinal area is a ideal flap donor site that provides the above characteristics. The free classic inguinal flap was described in 1973 (Mc Gregor) with vascular base in the deep branch of the superficial iliac circumflex artery (SCIA). This flap did not undergo any significant changes since its description in 1973 to 2004 (Koshima). This year was designed to be a perforator flap, with a vascular base in the perforators of this deep branch (indirect perforator type D or F of Nakajima). This indirect SCIP perforator flap presents drawbacks that have prevented its popularization such as: short pedicle, medium size and the need for supramicrosurgery techniques. This doctoral thesis aims to clarify the vascular base of the SCIP flap, establish a pre-surgical planning using non-invasive imaging techniques (MDCT) that allow a safe elevation of the flap and a design that provides the maximum benefits minimizing its drawbacks. The anatomical constancy of the superficial branch of the SCIA (direct cutaneous perforator C type of Nakajima) is considered. The use of this branch as a vascular base, of this variant of the SCIP flap, defining it as a direct flap. The location of this superficial branch is proximal and medial to the anterosuperior iliac spine (EIAS), thus providing advantages with respect to the indirect SCIP flap: avoid the use of supramicrosurgery techniques, larger cutaneous island size could be raise, longer vascular pedicle length and direct closure of donor site. All the patients in the study had a positive identification in the MDCT of the superficial branch of the SCIA. A probability of 90% of exit point of this branch, to suprafascial plane, in an area determined by values of X of +-125mm to +-77mm and of values of Y of -130mm to -63mm with respect to the umbilicus is establishes. Converting these data to reference points located on both EIAS facilitate clinical application, we obtain a 90% probability of finding the exit point of this branch in a circumference whose center is medial and distal to both EIAS in values for X of 17mm and for Y of 18mm, and whose radius is 21mm. The clinical application of the direct SCIP flap is detailed in five cases of reconstruction on the distal extremity and head and neck. Therefore, the direct SCIP flap has a constant vascular axis constituted by the superficial branch of the SCIA, giving a medium or large flap with fine and moldable skin and direct donor site closure possibility. This flap being ideal in the reconstruction of tissue defects in areas such as leg, foot, forearm, hand, head and neck.

Abstract:
There are many real cases where a company needs to determine the price of its products so as to maximise its revenue or profit.

To do so, the company must consider customers' reactions to these prices, as they may refuse to buy a given product or service if its price is too high. This is commonly known in literature as a pricing problem.

This class of problems, which is typically bilevel, was first studied in the 1990s and is NP-hard, although polynomial algorithms do exist for some particular cases. Many questions are still open on this subject.

The aim of this thesis is to investigate mathematical properties of pricing problems, in order to find structural properties, formulations and solution methods that are as efficient as possible. In particular, we focus our attention on pricing problems over a network. In this framework, an authority owns a subset of arcs and imposes tolls on them, in an attempt to maximise his/her revenue, while users travel on the network, seeking for their minimum cost path.

First, we provide a detailed review of the state of the art on bilevel pricing problems.

Then, we consider a particular case where the authority is using an unit toll scheme on his/her subset of arcs, imposing either the same toll on all of them, or a toll proportional to a given parameter particular to each arc (for instance a per kilometre toll). We show that if tolls are all equal then the complexity of the problem is polynomial, whereas in case of proportional tolls it is pseudo-polynomial.

We then address a robust approach taking into account uncertainty on parameters. We solve some polynomial cases of the pricing problem where uncertainty is considered using an interval representation.

Finally, we focus on another particular case where toll arcs are connected such that they constitute a path, as occurs on highways. We develop a Dantzig-Wolfe reformulation and present a Branch-and-Cut-and-Price algorithm to solve it. Several improvements are proposed, both for the column generation algorithm used to solve the linear relaxation and for the branching part used to find integer solutions. Numerical results are also presented to highlight the efficiency of the proposed strategies. This problem is proved to be APX-hard and a theoretical comparison between our model and another one from the literature is carried out.

Abstract:

The emergence of real-time applications, like Voice over IP and video conferencing, in IP networks implies a challenge to the underlying infrastructure. Several real-time applications have requirements on timeliness as well as on reliability and are accompanied by signaling applications to set up, tear down and control the media sessions. Since neither of the traditional transport protocols responsible for end-to-end transfer of messages was found suitable for signaling traffic, the Stream Control Transmission Protocol (SCTP) was standardized. The focus for the protocol was initially on telephony signaling applications, but it was later widened to serve as a general purpose transport protocol. One major new feature to enhance robustness in SCTP is multihoming, which enables for more than one path within the same association.

In this thesis we evaluate some of the mechanisms affecting transmission performance in case of a switchover between paths in a multihomed SCTP session. The major part of the evaluation concerns a failure situation, where the current path is broken. In case of failure, the endpoint does not get an explicit notification, but has to react upon missing acknowledgements. The challenge is to distinguish path failure from temporary congestion to decide  when to switch to an alternate path. A too fast switchover may be spurious, which could reduce transmission performance, while a too late switchover also results in reduced transmission performance. This implies a tradeoff which involves several protocol as well as network parameters and we elaborate among these to give a coherent view of the parameters and their interaction. Further, we present a recommendation on how to tune the parameters to meet  telephony signaling requirements, still without violating fairness to other traffic.

We also consider another angle of switchover performance, the startup on the alternate path. Since the available capacity is usually unknown to the sender, the transmission on a new path is started at a low rate and then increased as acknowledgements of successful transmissions return. In case of switchover in the middle of a media session the startup phase after a switchover could cause problems to the application. In multihomed SCTP the availability of the alternate path makes it feasible for the end-host to estimate the available capacity on the alternate path prior to the switchover. Thus, it would be possible to implement a more efficient startup scheme. In this thesis we combine different switchover scenarios with relevant traffic. For these combinations, we analytically evaluate and quantify the potential performance gain from utilizing an ideal startup mechanism as compared to the traditional startup procedure.

Abstract:
Message Passing Interface(MPI)is a popular message passing interface for writing parallel applications. It has been designed to run over many different types of network interconnects ranging from commodity Ethernet to more specialized hardwares including: shared memory, and Remote Direct Memory Access (RDMA) devices such as InfiniBand and the recently standardized Internet Wide Area RDMA Protocol (iWARP). The API itself provides both the point-to-point and remote memory access (RMA) operations to the application. However, it is often implemented based on one kind of underlying network device, namely entirely RDMA or point-to-point. As a result, it is often not possible to provide a direct mapping from the software semantics to the underlying hardware. In this work, we propose a hybrid approach in designing MPI in which network device to use can depend on its functional requirement. This allows the MPI API to exploit the potential performance benefits of the underlying hardware more directly. Another highlight of this work is the design of the MPI middleware to be IP based in order to provide support for both cluster and wide area network environment; this can be achieved via the use of a commodity transport layer protocol, namely Stream Control Transmission Protocol (SCTP). We will demonstrate how SCTP can be used to support MPI with different kinds of network devices and to provide multirailing support from the transport layer.

Abstract:
There are several weaknesses in the popular transport protocol TCP (Transmission Control Protocol). A possible replacement to TCP would be the newer protocol SCTP (Steam Control Transmission Protocol). This thesis presents three different proposed implementations of SCTP: HTTP over SCTP, online games over SCTP and IP mobility over SCTP. The proposed implementations are analyzed, based on relevant literature, and recommendations are issued on the importance of moving forward with them. The result of the thesis is that HTTP over SCTP is recommended. SCTP features such as multi-streaming, multi-homing and the four-way handshake addresses the inherent weaknesses with using TCP for HTTP traffic. IP mobility over SCTP is also recommended since it results in lower delay in the handover process compared to MIPv6 (Mobile IPv6). Online games over SCTP, however, is not recommended since the existing implementations of SCTP results in poor latency for the kind of traffic online games produce.

Abstract:

Abstract:
The research reported in this thesis investigates the performance of the transport layer Stream Control Transfer Protocol (SCTP) for streaming video over CDMA2000 cellphone and data wireless networks. The main measure of performance was quality of the received video at a given buffer size, as cellphones have memory of limited capacity. The hypothesis was that SCTP would be able to improve the quality of streamed video over UDP under the same memory requirements. Our study involved two series of simulation experiments and measurements in a testbed on the Telecom NZ CDMA2000 network, to test the performance of video streaming under SCTP and under UDP. It was found that SCTP did not improve the quality in streamed video with up to 5 second buffers. While other scenarios that have been tested by other people with high packet loss or congestion have shown that SCTP can improve the quality, the CDMA2000 network does not suffer from the impairments that SCTP could neutralise, and because of that, in this scenario, the quality of video streaming under SCTP and UDP are similar. The complexity that SCTP adds does not correspond to an increase in quality.

Abstract:
In current higher education paradigms, art and science are often siloed fields rather than subjects that inform each other. This dichotomy or hyper-specialization of art and science in higher education is a result of the industrial revolution with production as the telos. The product of this educational paradigm divorces art and science from each other. But how should we educate students for jobs that don't exist today? What learning environments are most conducive to creativity and innovation? What are the potential benefits of teaching art and science as one? What disciplines would work best together? Are their patterns in ones perception of the relationship between art and science? Are trans-disciplinary learning environments a possibility or an ideal? This thesis investigates the hypothesis that the walls between art and science exist only in our minds. This research consists of 27 one-on-one interviews conducted with students, professors and other higher education affiliates, who visualize the relationship between art and science.) The interviews use everyday objects as prompts to build a baseline to the investigation. The instrument consisted of seven questions that investigated if the use of quotidian, everyday, objects as prompts expose the false dichotomy between art and science. Additionally, the research tries to uncover the possible patterns that exist in how disciplines visualize/diagram the relationship between art and science. Each participant was asked to draw how they view the relationship between art and science. The researcher used these drawings as data points to lead the analysis. The researcher developed a series of field notes (thinking sketches) as interpretations of the themes of the participant's drawings. These thinking sketches were then translated into four thinking prototypes (three-dimensional models) which later inform the development of four simple yet profound findings called quotidian proverbs.

Abstract:
TCP and UDP are the most popular transport protocols used for end-end data transmission. The rapid growth of internet leads to development of many innovative applications in the current environment. Depending on the functionality of these applications, requirements of transport protocols are changing. TCP is known for its problems with Head Of Line blocking (HOL) and SYN attacks which gives reduced performance, and also doesn’t support Multi-Homing. SCTP is another transport layer protocol similar to TCP which provides end-end communication. It has some unique features like support for Multi-homing and multi-streaming. It also protects better from SYN attacks by using four-way hand shake mechanism during association establishment. As an extension to SCTP, CMT-SCTP was proposed to take full advantage of a multi-homed host by doing load sharing over multiple paths. SCTP is believed to be a next generation transport protocol. This thesis gives an overview of the SCTP protocol and its features focusing on analysing and testing of failover mechanisms provided by SCTP in multi-homed host, evaluating the transmission performance of SCTP vs TCP in a real network environment. This report also gives a theoretical analysis on how SCTP can mitigate SYN attacks by using four-way handshake mechanism and the state of art of CMT-SCTP.

Abstract:
Cirsium arvense (L.) Scop. (Canada thistle) is a persistent perennial pasture weed in Eastern Canada. Single control measures such as cultural, chemical, and biological methods are not effective in long term reduction of the weed population. The objectives of this research project were to evaluate the efficacy of various traditional control methods and to attempt to integrate them.
The effectiveness of Urophora cardui L. as a biological control agent is limited due to a lack of synchrony between the agent and the weed. This can be improved through integration with herbicides or mowing in time or space. Other biological control agents such as Orellia ruficauda Fab., Cassida rubiginosa Muell., and Puccinia punctiformis (Str.) Rhol. resulted in limited suppression of the weed population. Mowing was not effective in sustaining weed reductions for longer than one year. Application of clopyralid or 2,4-D/dicamba effectively reduced the weed population. The application of 3.37 kg a.i. ha$ sp{-1}$ 2,4-D/dicamba or 0.3 kg a.i. ha$ sp{-1}$ clopyralid with the spring and summer mowings were the most effective mowing/herbicide combinations over the two-year period.

Abstract:
This work deals with the comparison between the OFDM (orthogonal frequency division multiplexing) and SCCP (single carrier with cyclic prefix). These systems are analyzed as a function of the modulation order, the sensitiveness to the channel coding rate and to the interleaver configuration. The comparison is accomplished by means of a system model that leads to an universal framework to both analyzed systems. Theoretical results are obtained exploring convexity properties of functions. In addition, some results provided by Monte Carlo simulation are also presented to complement the analysis. The systems are compared mainly in terms of the BER (bit-error rate). Moreover, comparisons in terms of outage probability obtained by cutoff rate and in terms of the effective SNR (signal to noise ratio) at the equalizer output are also provided. Finally, the systems will be compared when channel estimation error are presented at the receiver.

Abstract:
As transistor feature sizes continue to shrink, it will become feasible, and for a number of reasons more efficient, to include multiple processors on a single chip. The SCMP system being developed at Virginia Tech includes up to 64 processors on a chip, connected in a 2-D mesh. On-chip memory is included with each processor, and the architecture includes support for communication and the execution of parallel threads. As with any new computer architecture, benchmark kernels and applications are needed to guide the design and development, as well as to quantify the system performance. This thesis presents several benchmarks that have been developed for or ported to SCMP. Discussion of the benchmark algorithms and their implementations is included, as well as an analysis of the system performance. The thesis also includes discussion of the programming environment available for developing parallel applications for SCMP.

Abstract:

Performance measurements have become very important tools for companies because of ever increasing global competition in today’s market. Delivery dependability is one of the three most important performance measurements for companies to focus on and that makes this analysis, of the dependability, both very interesting and up to contemporary. In this rapport different aspects of delivery dependability will be dealt with, and a weight will be on what to measure and how to measure it together with how to present the outcome of the measurements. A discussion has also been held regarding what goal the company should have on its dependability. The aspects that have been handled in this paper are based on the fact that Sanmina-SCI experiences its dependability measurement as a time costing task and at the same time it is important to understand the meaning of the result. To get the answer to our questions a theory study was made to get an understanding on needs to be measured and how to measure it. A thorough examination on the present situation at Sanmina-SCI was necessary to get the information on what problems the company is faced with and with these facts a result was made. The examination of the company was made with interviews together with our own observations and assessments of today’s measures and reports.

The result from the examinations shows that the measurements that are made manually at Sanmina-SCI can be made automatically instead. The preparation will then be at a minimum. At the same time as common report for the presentation of the delivery dependability has been created to make the reporting to the headquarter easier for all departments. The layout on the common report, that will be created with a macro, is well thought through and some parts of the report are totally new for Sanmina-SCI for example delivery dependability by product line. It has come to light that it’s very important that all employees that are in contact with the dependability result understand what it shows. Based on that fact the common report was made with an explanation added. A significant conclusion that has been drawn is that the result for the deliveries, that is measured against, what has been promised and what has been requested from the customer, not can be compared because of the differences in input. From this conclusions a proposal on a new way of measure has been created. The result of the analysis of Sanmina-SCIs goals of dependability shows that they are on a very high level but the company has not fully analysed the meaning of high goals compared to costs.

An analysis of measurements is something that is valuable for all companies and organisations, which makes this report relevant to read independent in which industries the company or organisation are in. A performance measurement are technical very easy to measure but in practice it’s much harder. Problem with performance, definition of data, analysis and reporting is common.

Abstract:
This doctoral research aims to study the relationship between Governance, Human Resource Management and Strategy of SCOP. These co-operative enterprises have an original and innovative political project and are beneficiaries of a triple linage of economic, political & social nature. Their values which derived from the nineteenth century are of solidarity, collective inter-generational ownership and equity. The identification of risks of tension and disintegration inherent to such an hybrid form of governance calls for a adequate management to run a co-operative organisation where worker members have a majority vote in co-responsibility.This issue emerging from both the field and the literature is studied in an innovative triptych approach based on a qualitative methodology. The analysis of practices and speeches - gathered from managers of 16 micro co-operatives and small & medium co-operatives of Rhône Alpes and from leaders of the co-operative sector – has revealed, on one hand, brakes and levers and on the other hand, contingency elements to a sustainable co-operative development of SCOP.These results have allowed the development of an ideal type of co-operative management and sustainable co-operative development based on co-operative management tools designed to prevent the risk of individual and collective tensions and to strenghthen the link between co-operative governance, co-operative management and co-operative development

Abstract:

Abstract:
Gene addition strategies to correct inherited diseases are showing promise for treatment in the clinic, but an improved approach to treat both dominant and recessive genetic disorders would be to repair the mutant gene by homologous recombination-mediated gene targeting. Recently, gene targeting frequencies have dramatically increased through the development of (i) improved designer nuc1eases, including zinc finger nucleases (ZFN), able to introduce specific double-strand breaks at their target locus; and (ii) efficient DNA delivery tools, including integration-deficient lentiviral vectors (IDL Vs). Even when using these state-of-the-art systems, gene repair frequencies in stem cells are moderate. To obtain proof-of-principle of phenotypic rescue in the haematopoietic system, a model in which corrected cells had selective advantage would be optimum. An ex vivo strategy to correct the classical scid mouse, a model of human DNA-dependent protein kinase catalytic subunit (DNA-PKcs, PRKDC) deficiency, is hereby presented. Donor templates to correct the scid point mutation and a ZFN were produced, optimised and incorporated into IDLVs. Specific ZFN activity in mouse scid fibroblasts and haematopoietic progenitors was demonstrated by Cel-I assay (which detects modifications introduced at the target site upon repair by non-homologous end- joining) and by deep sequencing. ZFN- and template-mediated gene repair of the scid mutation was demonstrated via the incorporation of a selection cassette and/or a diagnostic restriction site from the donor template into the targeted locus. In scid fibroblasts, gene repair led to DNA-PKcs activity rescue and increased resistance to DNA damage. In scid haematopoietic progenitors, gene correction was demonstrated only when the ZFN genes were delivered by integrating lentiviral vectors. Following an optimised ex vivo protocol, transplantation of potentially corrected scid haematopoietic progenitors into irradiated scid recipients has been carried out. Preliminary results of the ex vivo gene repair and transplantation experiment have indicated potential rescue of the T -cell compartment in a fraction of the scid transplant recipients. The results presented in this work highlight the potential of gene repair for future therapy in the haematopoietic system.

Abstract:
Today mobile network operators utilize IP Packet exchange (IPX) carriers to interconnect their networks with other operators. Mobile network operators are free to choose one IPX carrier for their data traffic and another for their control traffic. This thesis examines the case of control traffic, specifically Stream Control Transmission Protocol (SCTP) carrying Diameter protocol traffic arising from users roaming from their home Long Term Evolution (LTE) network to another operator’s LTE network. The thesis project aims to identify a set of SCTP parameter configurations that can provide improved application/service level availability between two Diameter nodes in different network connectivity environments, specifically for IPX carriers who are Diameter service providers. These service providers provide Diameter connectivity for their customers who are mobile network operators. These mobile network operators in turn provide LTE roaming services to their customers. Unfortunately, applying the ‘One size fits all’ configuration recommendations given in the SCTP documentation is unsuitable for different network environments. In addition, the amount of Diameter signaling traffic is growing at a very rapid rate. Therefore, it is valuable to identify suitable parameter selection criteria for Diameter service providers to ensure 100% Diameter connectivity reliability for their customers. In this thesis project, author investigated how tuning SCTP parameter values affect Diameter message transmission in terms of Round Trip Delay and identified its determining parameters for packet loss recovery performance. Both IPX carriers and mobile network operators may use these values as reference when attempting to ensure high availability of Diameter transmissions under reliable, semi reliable, and unreliable network transport conditions.
Abstract: The NK cells secrete the citoquinas INF-gamma and TNF-alpha - molecules associated with the immune protective mediated by cells of the type Th1 - in response to infection for Plasmodium. Instead the induction of these cells with IL- 12 helps to reduce the mortality by P. chabaudi. Although studies was related the NK cells and its citoquinas play a important role in the control of the malaria, it is known that the genetic factors represent one of the largest determinant of resistance by host infected by Plasmodium sp. For testing the malaria immunity by NK cells, we have used two animal models: the C.B-17 SCID/SCID because their absence of T and B cells and the C57BL/6 animals which are deprived for interferon-gamma gene. In this research, the capacity of both to control the infection was evaluated comparing the parasitaemia among them, using the immunocompetent BALB/c/Uni like a control animal. They were infected intraperitoneally with a dose of 106 parasited red blood cells and the parasitaemia was determined in established intervals for several weeks. So the animals SCID as the animals KO have maintained high parasitemia during chronic phase of the infection. The immunocompetents mice have decreased the parasitemia level , controlling the infection. It could be concluded that the NK cells collaborate to the malaria immunity reducing the parasitaemia; however the data indicating they don't act alone and the resistance against the P. chabaudi could be also a genetic factor. The malaria affects the kidney, not only for the immunocomplex deposition, because the mice C.B17 SCID/SCID/Uni unprovided immunoglobulins also developed renal pathologies: tubular cells exfoliation, thrombosis and necrosis in the glomerulus, also observed in C57BL/6 KO Interferon-gamma animals. It was not observed kidney alterations in C57BL/6 probably because resistant genotype

Abstract:

Lowrey, Jacqueline Anne. "Transplantation of Hodgkin's and non-Hodgkin's lymphomas into SCID mice." Thesis, University of Edinburgh, 1996. http://hdl.handle.net/1842/20637.

Abstract:
In an attempt to establish an in vivo animal model to study Hodgkin's disease (HD) and the histogenesis of the neoplastic cell of the disease - the Reed-Sternberg (RS) cell, and to study non-Hodgkin's lymphomas (NHL), severe combined immunodeficient (SCID) mice were transplanted with fresh biopsy material from 17 cases of HD and 25 cases of NHL. Five of the Hodgkin's (3 lymphocyte predominant, 1 nodular sclerosing and 1 mixed cellularity) and 7 of the non-Hodgkin's lymphomas (3 centroblastic, 1 immunoblastic, 1 follicular lymphoma and 2 biopsies from a case of B cell large cell anaplastic lymphoma) produced human high grade B cell lymphomas in the mice. All of the Hodgkin's disease derived SCID tumours expressed the Epstein-Barr virus (EBV) gene products EBER, EBNA-2 and LMP-1, even though only 1 of the original biopsies showed EBV positive RS cells. However, EBV clonality analysis showed that the EBV positive clone present in the original biopsy was not present in the SCID tumour. Morphologically, these tumours resembled polymorphous immunoblastic lymphomas, although in each case there were a few RS-like cells. The HD-derived SCID tumours did not retain the phenotype of the original biopsies, instead exhibiting an activated B cell phenotype with high expression of the CD23 and CD43 antigens. All HD-derived SCID tumours showed clonal Ig gene rearrangements. Further characterisation demonstrated that all of the HD-derived SCID tumours showed Ig secretion and contained the normal diploid DNA content.

Cantin, Amber M. "Proteolytic Regulation of CtrA, the Master Regulator of Cell Cycle in Caulobacter crescentus." 2012. https://scholarworks.umass.edu/theses/896.

Full text
Abstract:
Cell cycle progression in Caulobacter crescentus depends on the master regulator, CtrA. During the transition from swarmer to stalk cell (G1 to S phase), CtrA is degraded by the AAA+ protease ClpXP and levels rise again in the predivisional stage. The focus of this work is to explore how cyclic, regulated degradation is controlled. CtrA is known to bind to the origin of replication, thereby suppressing replication, so we first asked if DNA binding had an effect on CtrA stability. CtrA is readily degraded by ClpXP on its own, but when bound to DNA containing the proper binding sites, degradation is inhibited. Stabilization is dependent on DNA binding, as CtrA mutants deficient in DNA binding show the same degradation regardless of addition of DNA, as does CtrA in the presence of a mutant origin sequence lacking CtrA binding sites. Looking closely at CtrA degradation in the presence of auxiliary factors suggests that higher order complex formation may be a mechanism of protecting critical cell cycle regulators from premature proteolysis. In vivo study of over-expression of CtrA mutants revealed that accumulation of non-degradable CtrA, CtrA-DD, perturbs the cell cycle, leading to filamentation and a G2 arrest. Over-expression of the DNA-binding domain alone showed filamentation but no G2 arrest, suggesting that CtrA-DD is detrimental for reasons including, but likely not limited to, its ability to bind DNA. Exogenously expressing other domains of CtrA may further elucidate the mechanism of its regulated degradation in vivo.

Steyn, Yolinda. "A group resilience-promoting programme for individuals whose partners have acquired a spinal cord injury." Thesis, 2015. http://hdl.handle.net/10394/17034.

Full text
Abstract:
Spinal cord injury (SCI) is an acquired physical disability through traumatic injuries such as car accidents and shooting incidents, and non-traumatic injury such as a tumour on the spinal cord, amongst others. Unlike other parts of the body, the spinal cord does not have the ability to repair itself if it is damaged. Consequently, a person who has acquired an SCI will have a physical disability and will be either a paraplegic – paralysis of the lower part of the body, including the legs, or a quadriplegic, which is paralysis of all four limbs. Acquiring a spinal cord injury (SCI) has devastating long-term negative outcomes for the injured person as well as his/her cohabiting partner on a physical, psychological, psychosocial and socio-economical level. Exposure to such prolonged adversity and resulting negative outcomes calls for resilience, namely the ability to positively adapt despite the adversity being exposed to. Not all individuals have the natural ability to “bounce back”, and consequently resilience promotion is imperative. In South Africa, the focus of service delivery in rehabilitation centres is mainly centred on the injured person and consequently the well-being of the spinal cord injured person’s partner (SCIPP) is neglected. Little information is available on resilience-promoting programmes for SCIPPs; thus the main aim of this study was to design and develop a group resilience-promoting programme (GRPP) for SCIPPs. The researcher mainly followed a qualitative research approach and included a small quantitative component. In the context of applied research, an intervention research model comprising six phases was employed. Phase 1, Problem analysis and project planning, was reported on in section A. Manuscript 1 reports on phase 2 (information gathering and synthesis), and consists of a qualitative research synthesis, and mainly aimed at organizing and synthesizing previous research on resilience-promoting processes in order to inform the design and development of a group resilience-promoting programme (GRPP) for SCIPPs. After a systematic review and quality appraisal a total of 74 papers were selected to be quality appraised after abstracts and titles were assessed for relevance. Twenty-one studies were included and synthesized where after an outline for the content of a GRPP for SCIPPs was formulated. Conclusions and recommendations highlight that the formulated GRPP for SCIPPs needs to be further developed into an intervention that could be implemented with SCIPPs. As such, the researcher therefore proceeded with the study (see manuscript 2), aiming in developing small-group programme content and activities (using knowledge gathered from pre-existing interventions; resilience literature; consultations with experts; people living with spinal cord injury (SCI) and personal experience) to promote resilience in SCIPPs. By means of purposive sampling six advisory panel members from a diverse background were interviewed before and after the pilot study with two SCIPPs and one observer to contribute towards the further development of the GRPP for SCIPPs. A six-session GRPP for SCIPPs was formulated, including the following: (1) Information on SCI and resilience; (2) Help SCIPPs understand that their reactions to/emotions regarding these huge changes are normal; (3) Caretaking and support; (4) My dual role; (5) Own caretaking by SCIPPs; and (6) Termination and way forward. The newly developed GRPP for SCIPPs however had to be formally evaluated. Recommendations were made by professionals in the field that the GRPP for SCIPPs should first be subjected to peer review prior to implementing it with the target population. Therefore in manuscript 3 (reporting on phase 5 – evaluation ) the evaluation purpose was to subject the GRPP for SCIPPs to peer review by means of an empirical study with professional role-players (social workers and psychologists) in the field of spinal cord injury, prior to exhibiting it to the target population. The six group sessions were presented to professional role-players (n=12) working within the field of SCI during two 2-day workshops, whereby they were requested to evaluate the content and procedural elements of the GRPP for SCIPPs mainly by means of qualitative research, with a small numerical (quantitative) component. Thematic content analysis and basic descriptive statistics were employed. Overall positive feedback regarding the newly developed intervention was received, with suggested adjustments that needed to be made to the GRPP for SCIPPs prior to formal evaluation with the target group. The GRPP for SCIPPs will further be subjected to expert review in other provinces in South Africa, as the current participants were all from Gauteng. Furthermore, postgraduate students will be recruited to test the programme with the target-population in South Africa for possible further improvement and suggestions, as well as possible expansion to adjust this intervention to meet the needs of male SCIPPs; SCIPs themselves; children of a parent/s living with SCI; and also for post-injury cohabiting relationships, as this intervention might be a starting point for above-mentioned research-opportunities.
