Abstract:
FUT2 is a gene for a fucosyltransferase that encodes expression of ABO blood group antigens found on gastrointestinal mucosa and secretions. We hypothesized that the fecal microbiomes of healthy subjects, with blood group antigens A, B, and O, have differing compositions. We analyzed 33 fecal and blood specimens from healthy subjects for FUT2 genotype, and the fecal microbiome was determined by 454 pyrosequencing. Our data show that being a blood group secretor is associated with less diversity at higher orders of taxonomy; and the presence of blood group A antigens in the secretor subjects are associated with an expansion families of bacteria within the gut. Furthermore, our study confirms the previous findings that secretors and nonsecretors have differing bacterial taxa. This extends the previous findings by demonstrating that the impact of being a nonsecretor is higher than that of individual blood group antigens. Additionally, we demonstrate that both secretor status and blood group antigen expression especially affect the Lachnospiraceae family of bacteria within the gut microbiome, with lower abundances noted in nonsecretors and higher abundances in secretors of various blood groups. We further note specific differences in blood group A-secretors demonstrating that the genus Blautia is lower in the group A-secretors compared with the non-A-secretors and that this reduction is accompanied by higher abundances of members of the Rikenellaceae, Peptostreptococcaceae, Clostridiales, and Turicibacter. This study offers a first insight into the relationship between the fecal microbiome and blood group antigens in secretors.

Abstract:
The present study was designed to find out the status of the secretor and non-secretor in Rawalakot city of Azad Jammu and Kashmir (AJ&K). Blood and saliva were collected from 200 individuals of the study area during 2021-22. Secretor and non-secretor status were determined by Hemagglutination inhibition method. During this study status of secretor and non-secretor has been observed for gender, caste, and ABO blood group. Out of a total of 200 sampled individuals 142 (71%) individuals were secretors, while the remaining 58(27%) were non-secretors. Females (63.5%) were more secretors as compared to males (36.5%). The highest percentages of secretors were observed in Mughals (79.2%) and the lowest in Hashmi (65.0%). The highest numbers of secretors were observed in blood group B (37.0%), while the lowest in Blood group A (20.5%). The study concluded that secretors were more common in Rawalakot city as compared to non-secretors. This information would be helpful in healthcare management in the area.

Abstract:
Abstract Objectives Background: The FUT2 gene is responsible for the secretion of ABO blood type antigens into the body fluids (saliva, mucous, urine, tears, breast milk, sweat, and semen). Those who secrete the antigens into body fluids are call secretors, those who do not are called non-secretors. Hypothesis: GWAS studies have reported FUT gene variants to be associated with circulating vitamin-B12 (Vit-B12) concentrations. Missense mutations in the FUT2 gene result in a non-secretor phenotype. Thus, the secretory status of an individual may affect circulating vitamin-B12 concentrations over and above the genotype. Methods Materials and Methods: We included 780 participants (271 children, 282 mothers, and 227 fathers) from Pune Maternal Nutrition Study (PMNS). We measured the secretor status of individuals in saliva by hemagglutination test. A total of eight genetic variants including six SNPs from the FUT2 gene (rs492602, rs681343, rs281377, rs601338, rs1800027, and rs602662) and two SNPs from the FUT6 gene (rs3760776 and rs3760775) from our previous GWAS study were correlated with circulating vitamin-B12 levels. We tested the associations of FUT gene variants with secretor status phenotype and of the secretor phenotype with circulating vit-B12, folate, and ferritin concentrations in addition to H.pylori antibody levels. Results Results and Discussion: We found 33% of participants were non-secretors compared to 20% reported in Western Caucasian populations. Non-secretors had higher vitamin-B12 concentrations but not of folate and ferritin, vitamin-B12 associations were over and above FUT genotypes. Non-secretors showed a higher response to Vit-B12 supplementation. We found a FUT2 haplotype () to be strongly associated with Vit-B12 concentrations and non-secretor status. Non-secretors had lower H.pylori antibody concentrations. FUT6 genotype and haplotype were associated with Vit-B12 concentrations but not with secretor status and H.pylori antibody levels. Conclusions Our data suggest that secretor status may influence Vit-B12 concentrations through susceptibility to H.pylori infection and possibly other gut microbiota. Chronic periodontitis is the most prevalent oral inflammatory disease that affect teeth supporting tissues in response to microbial infection. A secretor is aterm applied to anyone who secret his/her blood type antigen into their body fluids as saliva in the oral cavity. Forty eight males were participated in the present research. Their age ranging from 35 to 55 years. The clinical periodontal parameters included Plaque index (PLI), Gingival index (GI), bleeding on probing (BOP) and Probing pocket depth (PPD) were examined also. .The secretory ABO blood groups from saliva by using Blood Typing Kit # 11 were assessed. After that the patients were divided into se cretors and non-secretors and the comparisons were done between t hem. There was highly significance difference between secretory and non-secretory chronic periodontitis patients in all periodontal pa rameters (PI, GI and PPD), from the total sample the Proportional ratio for secretory chronic periodontitis group was 58.33% while the proportional ratio for the non-secretary chronic periodontitis was 41.66%. Group O is the predominant type of ABO for both groups. Chronic periodontitis is more likely to occur in secretor ABO more than non-secretor ABO.

Abstract:
Abstract Anti-class II ag mAb (DR and DQ) inhibited, in a dose-dependent manner, LPS-induced IL-1 and TNF secretions from human monocytes (34 to 95% inhibition). The potentiating effect of IFN-gamma on LPS-induced TNF secretion (15.3 +/- 0.7 to 44 +/- 0.6 ng/ml) was also blocked by anti-class II ag mAb (44 +/- 0.6 to 0.3 +/- 0.03 ng/ml). We also report a relationship between interindividual differences in monocyte IL-1 and TNF secretions and the HLA-D-encoded genetic polymorphism. Heterozygotes were, in general, higher secretors of those cytokines than homozygotes. Analysis of these secretions in heterozygotes demonstrated a differential effect of certain haplotype combinations (i.e., DR2-DR4 vs DR2-DR3) that could be arbitrarily characterized as being "low" or "high" secretors (6,230 +/- 2,950 vs 13,029 +/- 6,541 cpm for IL-1, and 12 +/- 10 vs 25 +/- 15 ng/ml for TNF, p = 0.006 and 0.048). DR-associated Dw subtypes appeared to account for differences within certain haplotype combinations (Dw18 vs Dw19 in DRw13/DR4) (11,227 +/- 3,648 vs 17,166 +/- 3,176 cpm for IL-1, and 13 +/- 9 vs 25 +/- 10 ng/ml for TNF, p = 0.02 and 0.047). Interindividual differences were better explained by differences in LPS sensitivity than by differences in the kinetics of secretion and related not to the secretory process itself but to the rate of cytokine synthesis. Finally, there were no relationships between high secretor genotypes and IDD high risk genotypes. Thus, we conclude that, a) LPS-induced IL-1 and TNF secretions are, at least in part, regulated by class II MHC molecules, b) that HLA-D region-encoded genetic polymorphism accounts for interindividual differences in these secretions, and c) that the HLA-associated risk to develop IDD is not explained by these cytokine secretory differences as previously proposed.

Abstract:
Gastroenteritis is a preventable cause of morbidity and mortality worldwide. Rotavirus vaccination has significantly reduced the disease burden, but the sub-optimal vaccine efficacy observed in low-income regions needs improvement. Rotavirus VP4 ‘spike’ proteins interact with FUT2-defined, human histo-blood group antigens on mucosal surfaces, potentially influencing strain circulation and the efficacy of P[8]-based rotavirus vaccines. Secretor status was investigated in 500 children <5 years-old hospitalised with diarrhoea, including 250 previously genotyped rotavirus-positive cases (P[8] = 124, P[4] = 86, and P[6] = 40), and 250 rotavirus-negative controls. Secretor status genotyping detected the globally prevalent G428A single nucleotide polymorphism (SNP) and was confirmed by Sanger sequencing in 10% of participants. The proportions of secretors in rotavirus-positive cases (74%) were significantly higher than in the rotavirus-negative controls (58%; p < 0.001). The rotavirus genotypes P[8] and P[4] were observed at significantly higher proportions in secretors (78%) than in non-secretors (22%), contrasting with P[6] genotypes with similar proportions amongst secretors (53%) and non-secretors (47%; p = 0.001). This suggests that rotavirus interacts with secretors and non-secretors in a VP4 strain-specific manner; thus, secretor status may partially influence rotavirus VP4 wild-type circulation and P[8] rotavirus vaccine efficacy. The study detected a mutation (rs1800025) ~50 bp downstream of the G428A SNP that would overestimate non-secretors in African populations when using the TaqMan® SNP Genotyping Assay.

BACKGROUND: Head and neck cancers include malignancies of the scalp and neck skin, nasal cavity, paranasal sinuses, oral cavity, salivary glands, pharynx and larynx. The term ABO secretor refers to people who secrete blood group antigens in their body fluids such as saliva, sweat, tears, semen, and serum. Non-secretors refer to those who do not secrete their blood group antigens in their body fluids. The lack of blood type antigens in body discharge increases the susceptibility to certain types of diseases and infection.
 AIM: Our study aimed to investigate the relationship between the secretion of blood groups in the saliva of patients with head and neck cancers.
 MATERIAL AND METHODS: This case-control study was performed on 110 people (57 patients with head and neck cancer who were referred to Imam Khomeini Hospital, Tehran and 53 cancer-free controls). Five ml of non-stimulated saliva were collected by the spitting method. By agglutination or lack of agglutination in the test tubes, we determined the patient’s secretor or non-secretor condition.
 RESULTS: In terms of secretor status, 52.7% of all samples were secretors. In the case group, 19 out of 57 cases (33.3%) were secretors, and 38 were non-secretors (66.7%). In the control group, 39 out of 53 cases (73.6%) were secretors, and 14 cases were non-secretors (26.4%). There was a significant difference in the percentage of non-secretors between the two groups (p = 0.00).
 CONCLUSION: People with non-secretor status may be more prone to develop head and neck cancer. The presence of these antigens in saliva may have a protective effect.

We have recruited 188 participants of age ranging from 18- 40 years, out of which 155 were males and 33 were females. Tube method was used for ABO blood typing and Absorption inhibition method was used to determine the secretor status of ABH antigen using saliva samples.&nbsp; </em> <strong><em>Results: </em></strong><em>Our study showed that </em><em>68.62% of the population was secretors and 31.38% were non-secretors. The frequencies of secretor status in different ABO blood groups were 70.37% in group A, 64.15% in group B, 80.0% in group AB, and 67.21% in group O.</em> <strong><em>Conclusion: </em></strong><em>Our study shows that the secretor status in Peshawar is high than non-secretor. SUMMARYNon-secretors of ABO blood group antigens are over-represented among patients with meningococcal diseases. Lower levels of secretory IgA reported for non-secretors have been suggested to compromise mucosal defences. Total serum and salivary IgG, IgA and IgM and levels of these isotypes specific for Neisseria lactamica and five isolates of meningococci were determined by ELISA for 357 pupils and staff of a secondary school in which an outbreak of meningitis occurred. There were no differences in total or specific levels of serum IgG, IgA or IgM or salivary IgG or IgA of secretors compared with non-secretors. Non-secretors had significantly lower levels of salivary IgM (P=0·022) A similar pattern was observed for levels of IgM specific for N. lactamica and five meningococcal isolates. The significance of these results is discussed with reference to the role of secretory IgM in protection of mucosal surfaces in infants.

Abstract:
Introduction: Secretor status is a critical component of human biology that depends on specific glycoproteins in body fluids and secretions. Its importance lies in its significant impact on health and disease, making it a compelling subject for medical research. This study aimed to determine the prevalence and understanding of secretor status among undergraduates at the University of Calabar, Nigeria. The findings could revolutionize our understanding of secretor status and open new research opportunities. Materials and Methods: The study used across-sectional approach, analyzing blood samples from 100 undergraduate students using the adsorption-inhibition method. Most participants were single (94.0%), and the majority were 100-level students (51.0%). 48 students were in the 21 to 28-year range, while 6.0% were 30 or older. Results: The findings of this study are significant, revealing that a substantial proportion of the participants were secretors, 82 (82.0%), while 18 (18.0%) were non-secretors. Interestingly, most participants (83.0%) were unaware of their secretor status, indicating a potential knowledge gap. Blood group O had the highest number of secretors, 58 (96.7%), followed by blood group A 11 (55.0%), blood group B 7 (63.6%), and the minor blood group AB 6 (66.7%). The most prevalent ethnic group was found among the Efiks (18.1%) followed by Yakurr (16.6%) and the least the Ijaws (3.8%). Conclusion: This study underscores the importance of public education and awareness regarding secretor status and its impact on health and disease.

Brazil is the largest producer of essential oil, however, is what earns less in money value, because, its production is mainly of orange essential oil, with low added value. The essential oil of M. arvensis is rich in menthol (70-80%), a natural products with higher demand in daily life of modern man. Currently, Brazil has become a major importer of menthol which has contributed negatively to the brazilian trade balance. Thus, for the development of this work was chosen to be a specimens of economic interest and social demand, the Mentha arvensis. The purpose of study was to evaluate whether changes in light intensities and different levels of phosphorus promote changes in biomass production and nutrition, with consequences for the biosynthesis of essential oils. We also analyzed the influence of leaf age on the quality of essential oil. To this end, the plant of M. arvensis was grown under the effect of different conditions, which allowed the analysis of the levels of soluble fractions, of photosynthetic pigments, the activities of enzymes related to nitrogen assimilation and the anatomical and histochemical study of trichomes, morphological structure where it is produce the essential oil. The study also included analysis of the expression of genes related to the route of menthol synthesis. The essential oil was obtained by extraction with hydrodestillation in modified Clevenger and the chemical analysis of substances obtained by GC-MS. The Kovats index was used to collaborate in identification of the main substances in essential oil. The results showed that the essential oil quality is determined by physiological conditions and leaf age and that the production is dependent on the synthesis efficiency and density of trichomes on the leaves. In addition, it was found that different concentrations of phosphorus affected some aspects of plant metabolism, which reflected the production of biomass in the soluble fraction, with consequences on production and quality of essential oils. Also showed that variations in light levels affected the expression of six of the eight genes involved in the route of synthesis of menthol, in plants of M. arvensis and that changes in gene expression are probably associated with a transcription factor, which could be acting on the whole pathway. All of this information showed that environmental conditions and / or nutritional affected some aspects of plant metabolism, which reflected the production of biomass, of trichomes and on the expression of genes related to the route of menthol, which reflect on the quality and the production of essential oils<br>O cultivo de esp?cies arom?ticas para a obten??o de ?leos essenciais tem grande import?ncia econ?mica, principalmente, pela crescente demanda nas ind?strias de alimentos, de cosm?ticos e a farmac?utica. Relatively little is known about the molecular mechanisms displayed by parasitic nematodes to infect a host; however, it has been generally recognized that successful parasitic nematode infections rely on their ability to release a variety of products commonly named Excretory-Secretory Products (ESP). To gain a deeper understanding of the mechanisms that lead to the establishment of filarial nematode infections, we collected and analyzed through 1D-SDS PAGE and LC-MS/MS the ESP of Brugia malayi adult females, adult males and microfilariae, one of the etiological agents of human lymphatic filariasis. 228 proteins were identified through this approach, including several proteins with potential immunoregulatory properties. Subsequent work using an immunohistochemical approach allowed for the definition of 3 anatomical expression patterns in B. malayi microfilariae for a representative group of 5 ESP. All of these patterns involved localization in the microfilarial Excretory-Secretory apparatus, a specialized anatomical feature involved in protein release that in this life stage was found to be associated to a muscle structure. Glutamate-gated chloride channels (GluCls), the main target of the antiparasitic drug ivermectin (IVM), were also located at this structure, suggesting that protein release from the Excretory-Secretory apparatus is enhanced by neuromuscular activity regulated by GluCls. Consistent with these observations, a marked reduction in protein release by microfilariae upon in vitro IVM exposure was shown. It is proposed that under in vivo conditions, the rapid microfilarial clearance induced by IVM treatment is the result of the suppression of the parasite's ability to secrete proteins that enable evasion of the host immune system. Finally, it is anticipated that elucidation of specific features associated with each of the different parasitic nematode lifestyles would require the comparison of ESP composition from several nematode species. To overcome the limitations associated with the lack of sequence information in most nematode species required for the database searching strategy in MS-based proteomic analysis, the use of transcriptomic next-generation sequencing (RNA-seq) de novo assemblies is explored to identify the ESP composition of the mouse gastrointestinal (GI) parasitic nematode Heligmosomoides polygyrus. 209 proteins were identified using this strategy. The list also includes proteins with potential involvement in immunoregulation, modulation of signalling pathways and nutrient transport and/or uptake. The results presented here are useful to understand the roles of proteins released by filarial and GI nematodes in immune evasion events and other aspects of the host-parasite relationship.<br>Les mécanismes moléculaires déployés par les parasites pour s'établir dans leur hôtes sont relativement méconnus; néanmoins, il est généralement accepté que le succès des infections par les nématodes parasitaires dépend de leur capacité à libérer une variété de produits dénommés produits d'excrétion-sécrétion (PES). Afin d'acquérir une meilleure connaissance des mécanismes qui conduisent à l'établissement d'infections de nématodes filaires, nous avons recueilli et analysé par électrophorèse sur gel de polyacrylamide (1D-SDS-PAGE) et par chromatographie liquide couplée à la spectrométrie de mases ( LC-MS/MS) les PES des adultes mâles et femelles et des microfilaires de Brugia malayi, l'un des agents étiologique de la filariose lymphatique humaine. Grâce à cette approche, 228 protéines ont été identifiées, incluant plusieurs protéines possédant potentiellement des propriétés immunorégulatrices. Des travaux ultérieurs utilisant une approche immunohistochimique ont permis de définir trois profils d'expression anatomique pour un groupe représentatif de 5 PES chez les microfilaires de B. malayi. Pour tous ces profils, les protéines ont été localisées au niveau de l'appareil d'excrétion-sécrétion des microfilaires. Cette structure anatomique spécialisée est impliquée dans la libération de protéines et, chez ce stade de développement du parasite, elle est associée à une structure musculaire. Nous avons aussi montré que les canaux chloriques glutamate-dépendants (GluCls), la cible principale du médicament antiparasitaire ivermectine (IVM), étaient également localisés dans cette structure, ce qui suggère que la libération de protéines par l'appareil d'excrétion-sécrétion est renforcée par l'activité neuromusculaire régulée par des GluCls. Ces résultats concordent avec l'observation in vitro d'une réduction marquée de la libération de protéines par les microfilaires exposés à l'IVM. Il est proposé que, dans des conditions in vivo, l'élimination rapide des microfilaires induite par traitement avec l'IVM est causée par la suppression de la capacité du parasite à sécréter des protéines lui permettant d'évader le système immunitaire de l'hôte. Finalement, il est prévu que l'élucidation des caractéristiques spécifiques associées à chacun des différents modes de vie des nématodes parasitaires requerrait la comparaison de la composition des PES de plusieurs espèces de nématodes. Ces comparaisons, qui requièrent l'emploi d'une stratégie de recherche des résultats des MS générés lors des analyses protéomiques sur des banques de données des protéines, sont fortement limitées par l'absence d'information sur les séquences pour la plupart des espèces de nématodes. Pour contourner ces limitations, nous avons essayé l'utilisation des assemblages de novo du séquençage transcriptomique de nouvelle génération (RNA-seq) afin d'identifier la composition des PES du nématode gastrointestinal (GI) chez la souris Heligmosomoides polygyrus. 209 protéines ont été identifiées en utilisant cette stratégie. La liste comprend aussi des protéines ayant une implication potentielle dans l'immunorégulation, la modulation des voies de signalisation et le transport et/ou l'absorption de nutriments. Les résultats présentés dans cette thèse permettent de mieux comprendre le rôle des protéines libérées par des nématodes filaires et GI dans les événements d'évasion immunitaire et dans d'autres aspects de la relation hôte-parasite.

As estruturas secretoras florais também foram anatomicamente caracterizadas, com ênfase na cabeça dos estiletes. Uma grande quantidade de secreção esbranquiçada de aspecto gomoso é observada envolvendo ambas as faces dos primórdios foliares e a região adaxial das folhas jovens. Esta secreção é constituída principalmente por mucilagem (detectada pela reação PAS e pelo Vermelho de rutênio) e produzida por um número elevado de coléteres cônicos, os quais apresentam uma variação de comprimento e estão dispostos em duas ou três fileiras paralelas a superfície da base dos primórdios. Estas estruturas apresentam tanto origem axilar quanto marginal, são íntegras e persistem nas folhas adultas ocupando posição intrapeciolar. Coléteres responsáveis pela liberação de uma secreção incolor, constituída principalmente por mucilagem, são observadas nas brácteas folhosas e nas brácteas diminutas das inflorescências. Cada bráctea folhosa possui 9-13 coléteres polimorfos e destes, 2-4 são marginais do tipo standard e os demais são axilares cônicos semelhantes aos foliares. Nas brácteas diminutas é possível notar 2-4 coléteres marginais cônicos. A distensão da cutícula é observada nos coléteres foliares e das brácteas folhosas, e nos senescentes das brácteas diminutas. As estruturas secretoras encontradas nas flores são: epiderme da cabeça dos estiletes, nectários e laticíferos. Coléteres calicinais não foram observados. A epiderme da cabeça dos estiletes produz polissacarídeos e a presença de tricomas longos abaixo dos apêndices apicais diferencia a cabeça da flor funcionalmente masculina da feminina. A cabeça dos estiletes destas flores não apresenta adnação a qualquer dos verticilos florais e corresponde ao ¿tipo 2¿ descrito para Apocynaceae. O nectário é anelar na base e levemente bilobado no ápice; a vascularização é constituída por feixes concêntricos na região basal e por cordões floemáticos na região apical. O parênquima nectarífero é revestido por epiderme unisseriada, com estômatos ocupando preferencialmente a região apical. Os laticíferos são ramificados e estão presentes em todas as peças florais. A presença de um anel nectarífero e ausência de coléteres calicinais são caracteres relevantes em nível de tribo: considerados conjuntamente, eles corroboram a posição de R. sellowii em Vinceae<br>Abstract: The work here presented a morphological and anatomical study of the secretory structures of Rauvolfia sellowii Müll.Arg. The main goal of this study is to confirm the presence of colleters in the vegetative and reproductive organs of this species by analysing the chemical composition of the exsudate they produce and by doing a comparative study of their structural type, number and organization. An anatomical characterization of flower secretory strutures was also conducted, with emphasis on the style head. A substancial amount of a whitish, gluelike secretion was observed surrounding both faces of the young leaf primordia. This secretion is mainly composed by a mucilage (detected by the PAS and Ruthenium red reactions) produced by several, conical colleters organized in two to three rows parallel to the leaf primordia base. The observed colleters can be either axillary and marginal in origin, form an entire and undivided body, and are persistent even in mature leaves, where they have an intrapetiolar position. Other colleters secreting a colorless fluid, mainly composed by mucilage, were also observed on the leafy and minute bracts of the inflorescences. Each leafy bract bears 9-13 colleters, from which 2 to 4 are marginal and standard-type, and the remaining are axillary and resemble morphologically the conical colleters from the leaf primordia. The minute bracts, on the contrary, bear 2 to 4 marginal, conical colleters at the base. A distention of the cuticle was observed in the epidermal cells of the colleters at the leaf primordia and leafy bracts, and also on the senescent colleters at the minute bracts. Three types of secretory structures were found in the flowers of R. sellowii: epidermal cells of the style-head, nectaries and laticifers. No colleters were found at the base of the calyx lobes. The epidermal cells of the style-head produce mucilage, and the presence of long trichomes below the apical appendages of the style-head is a major distinctive characteristic of masculine functionally flowers. Regardless the flower type, the style-head is never fused to the androecium and thus correspond to the ¿type 2¿ organizational level described in Apocynaceae. The nectaries form an annular ring surrounding the ovary base and slightly bilobed at the apex. Vascularization of the organ is constituted by concentric bundles at the basal portion and phloematic bundles at the apical portion. The nectariferous parenchima is covered by a single epidermal layer, with stomata distributed preferentially at the apex. The observed laticifers are branched and are widely distributed in all floral organs. The presence of a conspicuous nectary ring surrounding the ovary and the absence of colleters at the base of the calyx lobes are diagnostic characteristics traditionally used in the tribal classification of Apocynaceae, and their co-ocurrence in R. sellowii corroborates its placement in the tribe Vinceae<br>Mestrado<br>Biologia Vegetal<br>Mestre em Biologia Vegetal

Tais estruturas podem estar localizadas em diferentes partes da flor e estão associadas à atração de polinizadores e/ ou defesa. As flores polinizadas por animais exalam odores que são produzidos e liberados por meio de glândulas de odor ou osmóforos, estruturas que apresentam grande variedade morfológica. Estudos anatômicos de tais glândulas são importantes, pois além de fornecerem informações para o entendimento das interações ecológicas de plantas e seus polinizadores, podem fornecer dados que auxiliem na determinação de relações filogenéticas entre as espécies. Assim, este trabalho objetivou (1) identificar as estruturas secretoras presentes nas flores de espécies de Leguminosae com polinização noturna, (2) levantar caracteres anatômicos florais compartilhados por essas espécies, (3) determinar se existem relações entre a polinização diurna e noturna e a morfologia dos osmóforos em Leguminosae (4) e investigar se os tricomas secretores podem atuar como osmóforos. Botões florais e flores de 14 espécies zoófilas de Leguminosae foram fixados e processados para análises em microscopias de luz, eletrônica de varredura e eletrônica de transmissão. Análises em cromatografia gasosa e cromatografia líquida de alta eficiência foram feitas nos tricomas secretores foliares e florais de Bauhinia. As flores polinizadas à noite apresentam várias características condizentes a este tipo de polinização, como antese com duração de uma noite, coloração inconspícua e forte odor. A prefloração ou fusão do perianto e a presença de hipanto em forma de copo permite o acúmulo de néctar nestas flores. A grande quantidade de néctar produzido nas espécies quiropterófilas tem relação com a estrutura robusta de seus nectários. No perianto, especialmente nas pétalas, encontram-se estruturas responsáveis pela produção do odor nas espécies de Leguminosae com polinização zoófila. A presença de regiões secretoras não especializadas na epiderme e no mesofilo das sépalas e/ou pétalas corrobora a liberação de odor difusa na maioria das espécies. Osmóforos típicos, ou seja, glândulas de perfume com estrutura anatômica especializada na emissão de fragrâncias, restritas a certas regiões da flor, foram observados em quatro das 12 espécies estudadas. Os osmóforos estudados sempre exibiram algum tipo de célula ou tecido secretor de terpeno; porém, compostos fenólicos e proteínas também foram detectados. Outros tipos de estruturas secretoras, frequentemente associadas a mecanismos de defesa, como tricomas, cavidades e idioblastos secretores foram encontrados. Dentre elas, destacam-se os tricomas secretores cavitados em Bauhinia, os quais secretam as mesmas classes de compostos nas folhas e flores e, assim, apresentam função de defesa e não na atração dos polinizadores. Conclui-se que não existe relação entre o tipo de osmóforo (típico ou difuso) e o hábito do polinizador (diurno ou noturno). As estruturas secretoras florais encontradas (exceto os nectários) apresentam significado biológico e/ou taxonômico, mas não podem ser associadas à determinada síndrome. Já a anatomia dos nectários revelou correlações entre seu tamanho, a quantidade de néctar produzido e o tipo de polinizador demonstrando sua grande importância na relação flor-polinizador<br>Abstract: Although the Leguminosae family is rich in species showing wide geographic distribution and a large morphological variation, their representatives have been poorly studied in terms of floral secretory structures. Such structures can be located in different parts of the flower and are associated with attraction of pollinators and/ or defense. Flowers pollinated by animals emit scents that are produced and released by means of scent glands or osmophores, structures that show a great morphological variety. Anatomical studies of these glands are important because besides providing information for the understanding of ecological interactions of plants and their pollinators, they can provide data to assist in the determination of phylogenetic relationships among species. This study aimed (1) to identify the secretory structures present in the flowers of Leguminosae species with nocturnal pollination, (2) to reveal floral anatomical features shared by these species, (3) to determine whether relationships exist between the diurnal and nocturnal pollination and the morphology of osmophores in Leguminosae (4) and to investigate if trichomes can function as osmophores. Flower buds and flowers of 14 zoophilic species of Leguminosae were fixed and processed for analysis in light, scanning electron and transmission electron microscopy. Analysis in CG and HPLC were performed in leaf and floral trichomes in Bauhinia. Flowers pollinated at night have several characteristics consistent with this type of pollination, as anthesis with duration of one night, inconspicuous coloration and strong odor. The aestivation or fusion of the perianth and the presence of cup-shaped hypanthium allows the nectar accumulation in these flowers. The large amount of nectar produced in the chiropterophilous species is related to the robust structure of their nectaries. In the perianth, especially in petals, occur structures responsible for producing the scent in the Leguminosae species with zoophilic pollination. The presence of non-specialized secretory regions in the epidermis and mesophyll of sepals and/ or petals confirms the diffuse release of scent in most species. Typical osmophores, i.e., scent glands with anatomical structure specialized in the emission of fragrances, restricted to certain regions of the flower, were observed in four of the 12 species studied. The studied osmophores always exhibited some type of cell or tissue secreting terpene; but phenolic compounds and proteins were also detected. Other types of secretory structures often associated with defense mechanisms, such as secretory trichomes, idioblasts and cavities were found. Among these are the cavitated trichomes in Bauhinia, which secrete the same classes of compounds in the leaves and flowers and thus have no role in the pollinator attraction but in the plant defense. It is concluded that there is no relationship between the type of osmophore (typical or diffuse) and the habit of pollinator (diurnal or nocturnal). The floral secretory structures found (except nectaries), have biological and/ or taxonomic significance, but they cannot be associated with a particular syndrome. Emend Marques) no estado de São Paulo. Os principais pontos abordados na tese são a caracterização de estruturas secretoras e a elaboração de um inventário de caracteres morfológicos do eixo vegetativo. Destaca-se entre os resultados obtidos: a verificação de que as glândulas presentes no caule de P. laureola tratam-se de nectários extranupciais de origem estipular ocupando posição nodal. Além disso, foi possível constatar que as glândulas da inflorescência de P. laureola também são nectários extranupciais. Já em P. angulata, ductos secretores são observados no caule . Tais estruturas estão sendo identificadas como sítios de síntese de compostos hidrossolúveis - mucilagem. No inventário morfológico realizado nas cinco espécies de Polygala, foi evidenciado que a estrutura primária do caule se apresenta distintiva, enquanto que a estrutura secundária do caule e da raiz se apresenta conservativa. No estudo anatômico foliar, dois padrões estruturais são observados nas espécies estudadas de restinga e de cerrado; P. laureola, P. paniculata e P. violacea possuem características mesomórficas, P. cyparissias e P. angulata apresentam características xeromórficas.<br>Abstract: The purpose of this study is to contribute to the morphological knowledge of five species of Polygala L., three of which occur in the restinga ( P. cyparissias A.St.-Hil. & Moq., P. laureola A.St.-Hil. & Moq. and P. Paniculata L.) and the other two occur in the cerrado vegetation ( P. angulata L. and P. violacea Aubl. emend Marques) of São Paulo state, Brazil. The main points of this research are the characterization of the secretory structures and the survey of morphological features of the vegetative axis of these five species. Among the data obtained, it should be highlighted that the vegetative glands of P. laureola are stipular, extranuptial nectaries occupying a nodal position and that their inflorescence glands are also extranuptial nectaries. Secretory ducts observed in the stem of P. angulata are identified as sites of biosynthesis and secretion of mucilage. The morphological survey of the five Polygala species reveal that the primary stem structure is distinctive, whereas the secondary structure of the stem and the root is conservative. The leaf anatomy of these species shows two structural patterns occurring in the restinga and the cerrado vegetation: P. laureola, P paniculata and P. (Anacardiaceae) conhecida popularmente como pau-pombo é uma espécie importante para uso madeireiro, medicinal e pode ser empregada na recuperação de áreas degradadas e de matas ciliares. Diversas substâncias são produzidas em estruturas secretoras específicas, sendo a presença de canais e cavidades secretoras característica universal da família. Na literatura é relatada a presença de disco nectarífero intraestaminal nas flores de muitas Anacardiaceae. No entanto, estudos anatômicos e histoquímicos recentes não comprovaram a secreção de néctar por esta estrutura. Além disso, nestes trabalhos foi verificada a persistência do disco intraestaminal no fruto maduro de T. guianensis, bem como a presença de um endocarpo secretor em frutos não maduros. Aliado a estes fatores, a ontogenia de frutos e sementes também é um tema pouco explorado frente ao grande número de espécies da família. A anatomia dos frutos de Anacardiaceae tem-se mostrado de grande valor sistemático. Assim, o presente trabalho consistiu em verificar a estrutura e ultraestrutura do disco glandular de flores e frutos de T. guianensis, bem como examinar a natureza da secreção produzida e estudar a ontogenia do fruto e da semente. Com os resultados alcançados foi possível determinar que a glândula floral das flores hermafroditas de T. guianensis é uma glândula de secreção mista, produzindo néctar com predominância de sacarose, lipídios e substâncias fenólicas. Ultraestruturalmente esta glândula passa por mudanças profundas ao longo do desenvolvimento da flor, inicialmente rica em amiloplastos e posteriormente plastídios desprovidos de amido e mitocôndrias repletas de gotículas de óleo, indicando a mudança da secreção antes predominantemente hidrofílica para lipofílica. No que diz respeito à anatomia do fruto, observou-se que o mesmo possui características típicas da tribo Spondieae, porém algumas delas indicam uma condição menos derivada da espécie, como ausência de opérculo e endocarpo relativamente fino quando comparado com outras espécies da mesma tribo. O pericarpo possui grande quantidade de canais resiníferos na região do mesocarpo, epicarpo produzindo substâncias fenólicas e lipídios, e por meio de microscopia eletrônica de transmissão pôde-se observar o modo de secreção, as organelas envolvidas do processo, bem como a estrutura celular. A semente provém de um óvulo único, anátropo, bitegumentado, crassinucelado de funículo longo. A semente não possui camada mecânica, tem a exotesta e endotégmen ricos em substâncias fenólicas, cristais no mesotégmen, desenvolvimento de paquicalaza parcial, hipóstase fenólica e lipídica, permanência de funículo aderido ao envoltório da semente, formando um arilo vestigial. O embrião armazena grande quantidade de proteínas e amido, indicando uma condição de transição da espécie, pois em Anacardiaceae as reservas são constituídas principalmente por proteínas e lipídios. O fruto e semente apresentaram algumas características anatômicas possivelmente relacionadas à dispersão como, formação de aerênquima no mesocarpo interno e na mesotesta, acúmulo de amido no mesocarpo. O trabalho traz contribuições no estudo de glândulas florais em Anacardiaceae e na área de ontogenia de frutos e sementes<br>Abstract: Tapirira guianensis Aubl. (Anacardiaceae) well-known as "pau-pombo" is an important species with uses as timber, in medicine and can be employed recovering degraded areas and riparian forests. Several substances are produced in specific secretory structures and the presence of cavities and ducts are universal features in the family. In literature is reported the presence of an intrastaminal receptacular nectary in the flowers of many Anacardiaceae, however anatomical and histochemical recent studies did not support the nectar secretion by this structure. Furthermore, in other studies were found the persistence of intrastaminal receptacular gland in T. guianensis ripe fruits and the presence of a secretory endocarp in unripe fruits. Allied these factors, the ontogeny of fruits and seeds is also a relatively unexplored subject, in contrast with the large number of species from this family. The fruit anatomy of Anacardiaceae has proved great systematic value. Thus, the present study verified T. guianensis structure and ultrastructure of flowers and fruits intrastaminal gland and examined the nature of substances produced as well the fruit and seed development. With the results obtained it was possible provide that the T. guianensis floral gland of hermaphrodite flowers is a mixed secretion gland, producing nectar with sucrose predominance, lipids and fenolic substances. Ultrastructurally, this gland undergoes deep changes during the development of the flower, originally rich in amyloplasts and subsequently chloroplasts devoids of starch and mitochondria replete with oil droplets, indicating the change in the secretion predominantly hydrophilic to lipophilic. With respect to the fruit anatomy, it was observed characteristics typical of the tribe Spodieae, but some of them indicate a condition less derived of this species, such as lack of operculum and endocarp relatively thin compared to other species of the same tribe. Pericarp has a large number of resiniferous ducts in mesocarp region, and epicarp produces phenolic substances and lipids. By transmission electron microscopy it was possible to observe the way of secretion, the organelles involved in process and the cell structure. Seed comes from a single ovule, anatropous, bitegmic, crassinucelate with longe funicle. Seed does not have mechanical layer, has exotest and endotegmen rich in phenolic substances, crystals in mesotegmen, development of partial pachycalaza, phenolic and lipid hypostasis, permanence of the funicle attached to the seed-coat, forming a vestigial aril. The embryo stores large amounts of protein and starch, indicating a transitional condition in the species, because Anacardiaceae reserves comprehend mainly protein and lipids. Fruit and seed showed anatomical characteristics possibly related to dispersion, like formation of aerenchyma in the inner mesocarp and mesotest, starch accumulations in mesocarp. O uso de metodologia de imunoquimiluminescência para realizar tal diagnóstico permite uma maior sensibilidade que os métodos espectrofotométricos tradicionalmente utilizados. Neste trabalho, um ensaio Dotimunoquimiluminescente (Dot-CLIA) foi proposto para a determinação de IgA Secretora. Os anticorpos anti-IgAS e anti-IgG-peroxidase foram previamente conjugados com éster de acridina (AE). Dot-ELISA e Dot-CLIA foram então realizadas aplicando amostras de IgAS em discos de membranas de nitrocelulose (0,45 μm de diâmetro de poro) e foi medida a atividade da peroxidase e a quimiluminescência (expressa em unidade relativa de luz; URL), respectivamente. O complexo ternário formado por IgAS/anti- IgAS-AE/anti-IgG-peroxidase- AE e o controle (PBS em substituição a IgAS) forneceram valores de 302.255 ± 28.736 RLU e 8.247 ± 3.479 RLU, respectivamente. Dot-ELISA simultaneamente realizada forneceu cor marrom pelo complexo ternário e ausência de cor foi observada para o controle. A relação entre RLU versus quantidade de IgAS utilizando o método proposto mostrou uma curva hiperbólica. O leite materno sem lipídios e caseína purificado por HPLC apresentou três picos de proteína e os que correspondiam a IgAS e ao componente secretor livre apresentaram valores de cerca de 50.000 RLU e 30.000 RLU, respectivamente. Portanto, pode-se concluir que o ensaio Dot-CLIA é capaz de avaliar quantitativamente e especificamente IgAS em fluidos biológicos.

Tricomas secretores são apêndices epidérmicos presentes nas maiorias das plantas desempenhando funções diversas, dentre elas a defesa contra herbivoria. Dentre as famílias que apresentam tricomas secretores destaca-se Cannabaceae, que é amplamente estudada por produzir compostos psicoativos e outros utilizados como aromatizantes de cerveja. Recentemente alguns gêneros pertencentes à Ulmaceae foram inseridos em Cannabaceae, como Celtis e Trema. Tais gêneros são os mais numerosos em espécies da família. Considerando que tricomas secretores são os principais sítios de produção de substâncias de interesse econômico, e que são amplamente utilizados como caráter de valor taxonômico, o presente trabalho comparou a distribuição e morfologia de tricomas secretores de duas espécies de Cannabaceae (Celtis pubescens e Trema micrantha) e três espécies afins de Ulmaceae (Ampelocera glabra, Phyllostylon rhamnoides e Ulmus parvifolia). Os dados foram comparados aos existentes na literatura e discutidos em um contexto sistemático. Os tricomas secretores de Trema micranta foram, ainda, analisados em detalhe quanto à ontogenia, análise química do exsudato (in situ e direto) e ultraestrutura. Para tal, amostras de folhas e flores (pistiladas e estaminadas) foram coletadas, fixadas e processadas para análise de microscopia de luz, eletrônica de varredura e de transmissão. Foram encontrados cinco morfotipos diferentes nas espécies estudadas. C. pubescens e T. micrantha apresentaram tricomas secretores capitados e filiformes, que diferiram quanto ao pedúnculo, unisseriado em C. pubescens e bisseriado em T. micrantha. As três espécies de Ulmaceae apresentaram um único morfotipo (capitado com pedúnculo unicelular). Os tricomas de T. micranta iniciam o desenvolvimento com uma divisão anticlinal, semelhante ao que ocorre em Cannabis sativa. Seus tricomas secretam terpenos, alcaloides e compostos fenólicos. Testes químicos comprovaram a presença de alcaloides e os indicadores de canabinoides sugerem que T. micrantha apresenta potencial para produção desses compostos. A morfologia dos tricomas encontrados nas espécies de Cannabaceae é diferente da observada nas espécies de Ulmaceae, o que apoia a atual circunscrição destas famílias. A diversidade morfológica e de termos utilizados torna difíceis a classificação e comparação entre os tipos de tricomas secretores, sendo necessários esforços no sentido de padronizar a caracterização destas estruturas secretoras, em especial nas famílias que compõem o clado urticoide.<br>Secretory trichomes are epidermal appendages that are present in most plants performing various functions, among them the defense against herbivory. Cannabaceae comprises plants covered with secretory trichomes, and is a well-known family for producing psychoactive compounds and others used as beer flavorings. Recently some genera belonging to Ulmaceae were inserted into Cannabaceae, such as Celtis and Trema, the species-richest genera of the family. As the secretory trichomes are the main plant source of economically important substances and are widely used as taxonomic markers, the present study compared the distribution and morphology of secretory trichomes of two species of Cannabaceae (Celtis pubescens and Trema micrantha) and three related species of Ulmaceae (Ampelocera glabra, Phyllostylon rhamnoides and Ulmus parvifolia). The data were compared to those in the literature and discussed in a systematic context. The secretory trichomes of Trema micranta were also analyzed in detail regarding ontogeny, histolocalization of substances and ultrastructure. For this, samples of leaf and flower (pistillate and staminate) were collected, fixed and processed for analyses of light microscopy, scanning electron microscopy and transmission electron microscopy. For T. micranta chemical tests in situ and direct were carried out; in addition, samples were prepared for ultrastructural study in transmission electron microscopy. Five different morphotypes of secretory trichomes were found in the species studied. C. pubescens and T. micrantha exhibited capitate and filiform secretory trichomes that differed in the stalk, uniseriate in C. pubescens and biseriate in T. micrantha. The three species of Ulmaceae had a single morphotype (capitate with a unicellular stalk). The trichomes of T. micranta originate of a first anticlinal cell division, similar to what occurs in Cannabis sativa. They secrete terpenes, alkaloids and phenolic compounds. Chemical tests have confirmed the presence of alkaloids and cannabinoid indicators suggest that T. micrantha is a putative species for the production of these compounds. The morphology of the trichomes found in the species of Cannabaceae is different from that observed in the species of Ulmaceae, which supports the current circumscription of these families. The diversity of morphology and terms employed makes it difficult to classify and compare the types of secretory trichomes. In this sense, efforts are needed to standardize the characterization of these secretory structures, especially in the families that comprise the urticalean rosids.

A circunscrição de Apocynaceae gerou diversas discussões ao longo da história devido às semelhanças morfológicas de suas flores com as dos representantes de Asclepiadaceae e, atualmente, essas duas famílias foram unidas, sendo subdivididas em cinco subfamílias. Nesse novo conceito, Apocynaceae apresenta uma sinorganização não usual de estruturas florais e somente através de uma análise comparativa das diferentes subfamílias é possível compreender como esse grupo atingiu esse alto grau de elaboração floral. Neste contexto, o presente estudo teve o propósito de caracterizar anatomicamente as estruturas secretoras florais de três representantes de diferentes subfamílias: Tabernaemontana, Mandevilla e Ditassa. Essa análise comparativa demonstrou que os gêneros analisados apresentam diferentes estratégias para atração do polinizador e que houve uma alteração de estrutura e posição do nectário em Apocynoideae em relação à Asclepiadoideae. Enquanto o nectário de Mandevilla é uma projeção glandular ao redor do ovário, em Ditassa apenas a epiderme do tubo dos filetes é nectarífera. Essa alteração de posição está relacionada a diferentes estratégias de dispersão do pólen: mônades em Mandevilla e polínias em Ditassa. A dispersão do pólen em mônades ou em polínias depende diretamente da secreção produzida pela cabeça do estilete que apresenta forma e função distintas nas diferentes subfamílias. O estudo de Tabernaemontana e Mandevilla evidenciou que a cabeça do estilete produz uma secreção viscosa e fluida que auxilia na adesão do pólen ao polinizador durante a coleta do néctar no fundo do tubo floral, além de reter os grãos no nível do estigma, após a visitação de uma outra flor. Por outro lado em Ditassa, a cabeça do estilete produz uma secreção rígida que se adere às polínias e ao polinizador sendo transferida em conjunto durante a coleta do néctar que se encontra no androceu. Além dessas glândulas, coléteres e osmóforos foram encontrados em Ditassa, demonstrando uma maior diversidade de estruturas secretoras associada a uma maior elaboração da morfologia floral dentro do grupo.<br>Apocynaceae circumscription led to several discussions throughout its history because of the morphological similarities of their flowers with those of Asclepiadaceae and currently these two families are united, being subdivided into five subfamilies. In this new concept, Apocynaceae presents an unusual synorganization of floral structures and, only through a comparative analysis of different subfamilies, is possible to understand how this group reached this high degree of floral complexity. In this context, this study aimed to characterize anatomically the floral secretory structures of three representatives of different subfamilies: Tabernaemontana, Mandevilla and Ditassa. This comparative analysis showed that the analyzed genera have different strategies to attract the pollinator and that there was structural and position shifts in Apocynoideae in relation to Asclepiadoideae. While the Mandevilla nectary is a glandular projection around the ovary, in Ditassa only the epidermis on the filaments tube is nectariferous. This change of position is related to different pollen dispersal strategies: monads in Mandevilla and pollinia in Ditassa. The pollen dispersal in monads or pollinia depends directly on the secretion produced by the style head which has distinct shape and function in the different subfamilies. The study of Tabernaemontana and Mandevilla showed that the style head produces a viscous and fluid secretion that aids in the adhesion of pollen to the pollinator, while collecting nectar at the bottom of the floral tube, and retaining the grains in the stigma level after the visitation of another flower. On the other hand, the style head of Ditassa produces a rigid secretion that adheres to pollinia and to pollinator, being moved together while collecting nectar which is in androecium. In addition to these glands, colleters osmophores were found in Ditassa, demonstrating a greater diversity of secretory structures associated with a further elaboration of floral morphology within the group.

As espécies sul-americanas do gênero Viguiera Kunth foram transferidas para o gênero Aldama La Llave com base em estudos moleculares. Porém, a circunscrição de Aldama tenuifolia (Gardner) E.E.Schill. & Panero e A. kunthiana (Gardner) E.E.Schill. & Panero, pertencentes à Seção Paradosa, série Tenuifolieae ainda não está bem estabelecida, pois as espécies são morfologicamente muito próximas. O objetivo deste trabalho foi analisar a anatomia e o rendimento e a composição química do óleo essencial dos órgãos vegetativos das referidas espécies, visando fornecer dados que auxiliem na delimitação das mesmas. Para as análises anatômicas, as amostras de folhas, caules aéreos, xilopódios e raízes de três indivíduos de cada espécie foram fixadas em FAA 50 ou em solução de Karnovsky, infiltradas em hidroxi-etil-metacrilato, seccionadas em micrótomo rotativo, coradas e montadas em resina sintética. Também foram realizados testes histoquímicos em materiais frescos ou fixados incluídos ou não em hidroxi-etil-metacrilato Para a análise da superfície foliar utilizou-se a técnica de dissociação de epiderme e microscopia de luz e microscopia eletrônica de varredura, utilizando os procedimentos usuais de preparação. A extração do óleo essencial (OE) foi realizada por hidrodestilação em sistema do tipo Clevenger, separadamente para cada órgão e espécie. Os óleos essenciais (OE\'s) foram analisados em Cromatografia a Gás acoplada a Detector de Massas com amostras na concentração de 20mg/mL em acetato de etila. No estudo anatômico foi possível verificar que as folhas das duas espécies diferem quanto ao contorno da parede das células epidérmicas em secção paradérmica, ocorrência de canal secretor no floema primário e no parênquima fundamental da nervura central. Em relação ao caule, a diferença mais marcante foi a ocorrência de canais secretores no floema primário e secundário e, em relação à raiz, o grau de participação da atividade cambial no processo de tuberização. O xilopódio não apresentou diferenças estruturais entre as espécies. As duas espécies apresentaram em comum a ocorrência de estruturas secretoras, tais como tricomas glandulares, espaços secretores internos e hidatódios. Os maiores valores de rendimento dos OE\'s foram verificados para os órgãos subterrâneos. Nas folhas, xilopódios e raízes das duas espécies foram encontrados os mesmos compostos majoritários, apenas com variação de porcentagens relativas. Também foram identificados compostos exclusivos às espécies (?-felandreno, ?-patchouleno, ?-isocomeno e longifoleno-(V4) para A. tenuifolia; cipereno para A. kunthiana) que podem, em conjunto com os compostos comuns às duas espécies, fornecer um perfil químico característico para cada uma delas. Portanto, o estudo anatômico e da composição química dos OE´s dos órgãos vegetativos de A. tenuifolia e A. kunthiana, que são consideradas morfologicamente muito semelhantes, podem auxiliar na diferenciação dessas espécies.<br>South American species of Viguiera were transferred to the genus Aldama based on molecular studies. However, the circunscription of Aldama tenuifolia (Gardner) E.E.Schill. & Panero and A. kunthiana (Gardner) E.E.Schill. & Panero, belonging to Section Paradosa, series Tenuifolieae, is not yet well established because these species are morphologically very similar. The aim of the present study was to analyze the anatomy of the vegetative organs and the yield and chemical composition of essential oil of these species in order to provide data to help in their circumscription. For anatomical analyzes, leaves, aerial stems, xylopodia and roots from three individuals of each species were fixed in FAA 50 or in Karnovsky solution, infiltrated in hydroxyethyl methacrylate, sectioned with a rotary microtome, stained and mounted in synthetic resin. Histochemical tests were also performed on fresh or fixed material embedded or not in hydroxyethyl methacrylate. The analysis of the leaf surface was performed by light microscopy using the technique of dissociation of the epidermis and by scanning electron microscopy using the usual procedures of preparation. The extraction of essential oil (EO) was performed by hydrodistillation in a Clevenger-type system, separately for each organ of each species. Essential oils (EO\'s) were analyzed by Gas Chromatography coupled to Mass Detector with samples in 20mg/mL concentration in ethyl acetate. The anatomical study showed that the leaves of the two species differ in the outline of the epidermal cell wall in front view side and in the occurrence of secretory canals in primary phloem and ground parenchyma of the midrib. In relation to the stem, the most remarkable difference was the occurrence of secretory canals in the primary and secondary phloem and in relation to the root was the degree of cambial activity participation in tuberisation process. The highest OE\'s yields were verified to the underground organs. For the leaves, xylopodia and roots of both species, the same major compounds were found, varying only the relative percentages. In addition, unique compounds (?-phelandrene, ?-patchoulene, ?-isocomene and longifolene-(V4) for A. tenuifolia and cyperene for A. kunthiana) were also identified that can, along with the compounds common to all two species, provide a characteristic chemical profile for each of these species. Therefore, the studies of the anatomy and of the chemical composition of OE\'s of the vegetative organs of A. tenuifolia and A. kunthiana, which are considered morphologically very similar, may help differentiate these species.

Abstract:
The plasma concentration of factor VIII is influenced by the ABO blood group. Individuals with blood group 0 have a lower concentration of both factor VIII coagulant activity, factor VIII coagulant antigen (VIIICAg) and factor VIII related antigen (VIHRAg) than individuals with group A, B and AB. Thirty percent of the genetic variance of VIIIRAg concentration is due to the ABO blood group. The Lewis substances Lea and Leb are closely related to the A, B and H substances. We therefore examined the effect of the presence of these antigens on the plasma concentration of VIIICAg and VIIIRAg. The material was 74 monozygotic and 84 dizygotic twin pairs and 58 blood donors with ABO blood group 0. VIIICAg was determined by a radioimmunoassay and VIIIRAg was determined by an electroimmunoassay. A higher mean concentration °f VIIICAg (147%) and VIIIRAg (81%) was found in individuals with the Le antigen on their red cell surface compared to the mean concentration of VIIICAg (101%) and VIIIRAg (66%) in individuals who lacked this antigen. The difference was found in individuals with ABO blood group 0 only. Individuals with red cell Lea antigen are non-secretors and individuals who lack this antigen but have the Leb antigen are secretors of the A, B and H substances. The lowest factor VIII concentration was found in group 0 secretors. The effect of the Lewis phenotype on factor VIII concentration is therefore most probably due to an effect of the secretor locus. This finding may have practical implications for the diagnosis of carriers of hemophilia A. it has been shown that information on the ABO blood group improves the discrimination between carriers and normals. We found that the effect of ABO blood group on the total variance of VIIIRAg was higher in secretors (21%) than in non-secretors (8%). Since the frequency of secretors varies widely, it is possible that the importance of the ABO locus in carrier detection is different in different populations. Lewis blood typing in materials of carriers and normals are necessary to determine the effect of the Lewis phenotype in carrier detection.

This article studied excretory-secretory products of parasitic flatworms aimed at searching for potential immunoregulatory proteins. Immunoregulatory proteins are poorly studied at the moment. Recent years have showed increased interest in identifying immunoregulatory molecules produced by parasitic worms. Potential immunoregulatory proteins will make a significant contribution to the development of medicine and biotechnology and will make it possible to effectively treat allergic and other autoimmune diseases. The study used methods of bioinformatics, proteomics, and transcriptomics. Potential immunoregulatory proteins were identified in the Ligula interrupta secretome proteins and listed. Each protein was analyzed for possible immunoregulatory functions in the parasite-host system. Based on identification data of SEP proteins using parasite transcriptomes, annotation of secretome proteins (SEP) was made in the NCBI and Swissprot international databases. A table was compiled with identified potential immunoregulatory proteins. A functional analysis of each protein was performed. Protein functions were determined based on analysis of scientific articles, patents and publications. By comparing different proteins, it is possible to identify those that are similar in domain structure, phylogeny, and description. The discovered potential immunoregulatory proteins will make a significant contribution to the development of medicine and biotechnology and will make it possible to effectively treat allergic and other autoimmune diseases.

Abstract:
We have previously showed that hirudin abruptly arrests thrombin-induced secretion of acid hydrolase at any stage of its progress, whereas it only affects dense granule secretion only at its initial stages; these results have been interpreted to show that acid hydrolase secretion requires sustained while dense granule secreion ony requires a brief receptor occupancy (Holmsen et al. JBC 256(1981)9393). The requirement for receptor occupancy in thrombin-induced α-granule secretion and secretion during aggregation have been studied. Increasing concentrations of thrombin were added to gel-fitered platelets containing a constant, high concentration of hirudin. Dense granule secretion was initiated at lower thrombin concentration than those required for α-granule secretion and aggregation; acid hydrolase secretion required higher concentrations. A 14-fold exess of hirudin produced abrupt stop of dense granule secretion and α-granule secretion when added to platelets shortly after thrombin; it had no affect after these secretory process had reached 30% of their maximal values. Acid hydrolase secretion was, however, abruptly stopped by hirudin at any stage. When the platelets were allowed to aggregate, all three secretory processes increased their rates and could now be abruptly stopped by hirudin at any stage. Aggregation (optical) occurred slower than dense granule andoαgranule secretion, and was reversed by hirudin when added before it had reached 30% of its maximum. It is concluded thatαgranule secretion, like dense granule secretion, only requires a short receptor occupancy to be completed, in contrast to the requirement for sustained occupancy for hydrolase secretion.α-granule secretion might, however, require longer occupancy than dense granule secretion. It is possible that aggregation potentiates all secretory responses through close cell contact and that the abrupt inhibition by hirudin of all secretions may have been caused by its effect on the slower aggregation.

A partir del incremento de uso de metanfetamina (denominada localmente como “cristal”) por vía fumada, inyectada, esnifada y/o rectal en la Ciudad de México, y cuyos usuarios se autodenominan coloquialmente como “dulceros”, se presenta el reporte de resultados del programa de Reducción de Riesgos y Daños en Clínica Especializada Condesa. En un primer momento se describe el proceso de elaboración de la guía “Secretos de oficio del dulcerx” que tiene por objetivo captar población de hombres que tienen sexo con hombres que mantienen relaciones sexuales bajo el efecto de metanfetamina, ácido gamma-hidroxibutírico (GHB), ketamina, poppers, estimulantes para la erección, entre otras, la cual está sustentada en una postura enfocada en la reducción de riesgos y daños y ha sido distribuída tanto de manera digital como en sitios de encuentro. En un segundo momento se describe el trabajo colaborativo de pares y organizaciones de la sociedad civil en la difusión e implementación de estrategias descritas en la guía adaptadas para la población mexicana encaminadas a la reducción de riesgos y daños ante el uso de sustancias, así como la reducción de la transmisión de Virus de Inmunodeficiencia Humana (VIH), Virus de Hepatitis B (VHB), Virus de Hepatitis C (VHC), Sífilis y otras Infecciones de Transmisión Sexual (ITS). Por último, se describe el funcionamiento del programa de Reducción de Riesgos y Daños en Clínica Especializada Condesa de la Ciudad de México, en el que se contemplan nueve intervenciones diferentes: a) capacitación para el consumo, b) psicoeducación, c) vacunación, d) pruebas de detección y tratamiento para VIH, VHB, VHC, ITS, e) profilaxis pre y post-exposición, f) deshabituación y sustitución de sustancias, g) recolección segura de parafernalia y, h) entrega de kits de reducción de riesgos y daños, i) intervención psicológica.

Abstract:
Mortality in a broiler flock will typically range from 4-5% of the population over the course of 6- 7 weeks in the U.S. and 7-8% of the population in Israel. Suboptimal intestinal maturation and functionality are one of the major factors that contribute to early age mortality and hinder flock body weight uniformity. The development of absorptive and secretory functions is orchestrated by differentiation of cells that arise from stem cells. Supplying compounds by in ovo feeding (IOF) during late embryogenesis provides nutrients that may change the dynamics of stem cell differentiation. We hypothesize that the introduction of specific nutrients or probiotics to the late embryonic chick via IOF will result in an acceleration of the maturation of the small intestine as measured by villus/crypt morphology and the number and distribution of absorptive and secretory cells. A chick that can absorb nutrients more efficiently by increasing the number of cells expressing nutrient transporters and resist enteric pathogens by increasing the number of cells expressing mucin and host defense peptides will be healthier at hatch. This chick may have less need for antibiotics and may show reduced early mortality. The objectives of this proposal are to: 1) develop a model for the development of putative stem cells and absorptive/secretory cells in the small intestine of the late embryonic and early post hatch broiler. 2) determine the ability of IOF of nutrients to modulate the population of differentiated cells in the intestine. 3) determine the ability of IOF of probiotics to modulate the population of differentiated cells in the intestine. 4) reduce early mortality and increase body weight uniformity by IOF of selected nutrients or probiotics. This proposal combines the IOF expertise of Zehava Uni (Hebrew University) with the RNAscope in situ hybridization technique of Eric Wong (Virginia Tech). Previous studies using quantitative PCR to examine expression of genes in the intestine were unable to identify specific cells expressing these genes. RNAscope allows the ability to identify putative stem, absorptive and secretory cells in the small intestine. Thus, we will be able to investigate the effect of IOF on the presence of intestinal absorptive and secretory cells at the cellular level. Understanding the mechanisms for intestinal development and function are key to maintaining peak growth and health of chickens and thus would be of great economic benefit to the poultry industry.

Abstract:
The extracellular matrix of the mineralizing eggshell contains molecules hypothesized to be regulators biomineralization. To study eggshell matrix molecules, a bank of monoclonal antibodies was generated that bound demineralized eggshell matrix or localized to oviduct epithelium. Immunofluorescence staining revealed several staining patterns for antibodies that recognized secretory cells: staining for a majority of columnar lining cells, staining for a minor sub-set of columnar lining cells, intensified staining within epithelial crypts, and staining of the entire tubular gland. Western blotting with the antibody Epi2 on eggshell matrix showed binding to molecules with the apparent molecular weight of eggshell matrix dermatan sulfate proteoglycan (eggshell DSPG) (Carrino, et al., 1997). Immunoblots of cyanogen bromide-cleaved eggshell DSPG revealed broad band of reactivity that shifted to 25 kDa after chondroitinase digestion; indicating that the Epi2 binding site is located on a fragment which contains dermatan sulfate side chains. Immunogold labeling showed that Epi2 binds to secretory vesicles within the non-ciliated cells of the columnar epithelium, while the antibodies Tg1 and Tg2 bind to secretory vesicles of tubular gland cells. Immunogold labeling of demineralized shell matrix showed binding of Epi2, Tg1, and Tg2 to the matrix of the palisades layer, and showed little reactivity to other regions of the shell matrix. Quantification of the immunogold particles within the eggshell matrix revealed that antibodies Epi2 and Tg1 bind all calcified regions equally while antibody Tg2 has a greater affinity for the baseplate region of the calcium reserve assembly.
