Abstract:
The cellular machine Cdc48 functions in multiple biological pathways by segregating its protein substrates from a variety of stable environments such as organelles or multi-subunit complexes. Despite extensive studies, the mechanism of Cdc48 has remained obscure, and its reported structures are inconsistent with models of substrate translocation proposed for other AAA+ ATPases (adenosine triphosphatases). Here, we report a 3.7-angstrom–resolution structure of Cdc48 in complex with an adaptor protein and a native substrate. Cdc48 engages substrate by adopting a helical configuration of substrat

Abstract Background The p97/valosin-containing protein (VCP) complex is a crucial factor for the segregation of ubiquitinated proteins in the DNA damage response and repair pathway. Objective We investigated whether blocking the p97/VCP function can inhibit the proliferation of RepID-deficient cancer cells using immunofluorescence, clonogenic survival assay, fluorescence-activated cell sorting, and immunoblotting. Result p97/VCP was recruited to chromatin and colocalized with DNA double-strand breaks in RepID-deficient cancer cells that undergo spontaneous DNA damage. Inhibition of p97/VCP ind

Maintenance of genomic integrity is dependent on the duplication of chromosomes, only once per cell cycle. Highly conserved mechanisms for the regulation of chromosome replication exists to ensure that the genome is copied only once. The Cdc45-MCM-GINS (CMG) DNA helicase which is the core of the eukaryotic replication complex, has been shown to be extensively regulated by post translational modifications, during its assembly. Therefore, it is not inconceivable that the process to unload the replication complex would also be a conserved and regulated process. In 2014, our lab discovered that th

The practical parts of the thesis are summarised measurements on six samples of the most commonly used asphalt mixtures to determine the segregation depending on the selected parameters. The main new results contained in this thesis are as follows. The Segregated Λ-coalescent has a non-trivial construction which we present here in terms of stochastic flows. Moving beds are devices where a slow flow of particulate solids moving under the influence of gravity is contacted with a flow of gas. Heat transfer rates, mass transfer rates, and consequently reaction rates, for these devices are often modelled using information from well-packed fixed beds. It has been found, however, that moving beds behave differently from well-packed fixed beds (Paterson et al. 1991). The cause of this different behaviour is a bed structure, which is a consequence of the movement of the particles, that yields gas flow maldistribution (Crawshaw et al., 1993).

Education in post-Dayton Bosnia and Herzegovina suffers from the serious consequences of ongoing ethno-national conflict. My focus is segregated education in the Federation of Bosnia and Herzegovina generally, particularly the "two schools under one roof" phenomenon. Segregated education, by being an object of political struggle, seriously blocks the society from the necessary path of ethnic reconciliation.
