Obat Semax® memiliki izin khusus dari Badan Pengawas Obat dan Makanan (BPOM) yang merupakan neuropeptide yang berasal dari fragmen pendek ACTH (Adrenocorticotropic Hormone) dengan rumus kimia Pro8-Gly9-Pro10ACTH(4-10) dan merupakan peptida neuroaktif dengan sifat neuroregulator yang unik. Obat ini telah banyak diresepkan di Rusia dan digunakan untuk mengobati penderita stroke. Perkembangan penggunaan Semax® yaitu pada kasus iskemik di otak, dimana Semax® secara signifikan meningkatkan fungsi saraf, mengurangi ukuran infark dan kerusakan otak permanen. Abstract:
Purpose. To study by Heidelberg diagnostics the efficacy and safety of Semax 0.1 % used as a neuroprotective agent in POAG treatment. Materials and methods. The study involved 36 patients (72 eyes) with primary glaucoma. The first group was represented by patients with POAG receiving Semax in addition to the basic IOP-lowering therapy (24 patients - 48 eyes). The second (control) group consisted of patients with POAG, receiving only the basic IOP-lowering therapy (12 patients - 24 eyes). All patients underwent ophthalmologic examination with Heidelberg diagnostics before the treatment initiation, and 1 month after Semax was prescribed. Results and discussion. The paper presents a study of the efficacy and safety of Semax treatment in primary glaucoma with the Heidelberg diagnostics - scanning laser tomography, HRT-perimetry in the SAP III mode, in-vivo confocal corneal microscopy. During the observation period, no negative changes in the structure of the optic nerve head and in the corneal subbasal nerve plexus were revealed. The functional state of the visual analyzer also did not change, according to perimetry data. No adverse reactions to Semax 0.1 % were observed.

Abstract:
The Semax (Met-Glu-His-Phe-Pro-Gly-Pro) peptide is a synthetic melanocortin derivative that is used in the treatment of ischemic stroke. Previously, studies of the molecular mechanisms underlying the actions of Semax using models of cerebral ischemia in rats showed that the peptide enhanced the transcription of neurotrophins and their receptors and modulated the expression of genes involved in the immune response. A genome-wide RNA-Seq analysis revealed that, in the rat transient middle cerebral artery occlusion (tMCAO) model, Semax suppressed the expression of inflammatory genes and activated the expression of neurotransmitter genes. Here, we aimed to evaluate the effect of Semax in this model via the brain expression profiling of key proteins involved in inflammation and cell death processes (MMP-9, c-Fos, and JNK), as well as neuroprotection and recovery (CREB) in stroke. At 24 h after tMCAO, we observed the upregulation of active CREB in subcortical structures, including the focus of the ischemic damage; downregulation of MMP-9 and c-Fos in the adjacent frontoparietal cortex; and downregulation of active JNK in both tissues under the action of Semax. Moreover, a regulatory network was constructed. In conclusion, the suppression of inflammatory and cell death processes and the activation of recovery may contribute to the neuroprotective action of Semax at both the transcriptome and protein levels.

Abstract:
PURPOSE. It is well established that cholinergic neurons of the basal forebrain degenerate in Alzheimer's dementia. Although recent studies were concentrated on screening molecules that might reduce the concomitant cell loss, little is known about therapeutically promising molecules. We studied the effect of Semax (Met-Glu-His-Phe-Pro-Gly-Pro), a behaviorally active adrenocorticotropic hormone (4–10) analogue, on survival of cholinergic basal forebrain neurons in vitro. Semax is known to stimulate learning and memory and can be successfully used for treatment of ischemic stroke. METHODS. Primary cultures of neuronal and glial cells from basal forebrain of rats were used in all experiments. The stability of Semax in cell cultures was tested by HPLC analysis. Cell survival in neuronal cultures was quantitated using immocytochemical and cytochemical analyses as well as detection of choline acetyltransferase activity. RESULTS. We have shown that Semax may approximately 1.5–1.7 fold increase survival of cholinergic basal forebrain neurons in vitro. Moreover, Semax (100 nM) stimulated activity of choline acetyltransferase in dissociated basal forebrain tissue cultures. However, the numbers of GABA-ergic neurons, total neuron specific enolase neurons were not affected. In concentration from 1 nM to 10 μM, Semax did not affect proliferation of glial cells in primary cultures. CONCLUSION. Implications of these findings with respect to Alzheimer's disease remain to be clarified.

Abstract:
Currently, there are no drugs for the specific treatment of hypertensive retinal changes. The main therapy is for the treatment of a systemic disease – hypertensive disease. Therefore, the search for ways of specific pharmacological correction of hypertensive retinal changes is of great interest. The aim of the study: To evaluate the correction possibility of retinal injuries with Semax in a rat model of hypertensive neuroretinopathy. Materials and methods: The model was performed by injection of N-nitro-L-arginine methyl ester (L-NAME) at a dose of 1.25 mg/100 g of rat mass within 28 days and a single increase in intraocular pressure (IOP) to 110 mmHg for 5 min. The retinoprotective effect of Semax at a dose of 7.2 μg/100 g of rat mass, in comparison with Picamilon at a dose of 3 mg/100 g of rat mass, was estimated by laser Doppler flowmetry (LDF) and electroretinography (ERG). Results: The use of Semax led to an increase in retinal perfusion by 62.7%, p < 0.05, in comparison with the group with the model, and by 9.9%, p < 0.05, in comparison with Picamilon; an increase in the b/a coefficient by 31.4% in comparison with the group with the model, p < 0.05, and by 14.6%, p < 0.05 in comparison with Picamilon. Conclusion: The neuroretinoprotective effect of Semax in correction of hypertensive retinal changes in rats may be due to the presence of neuroprotective, neurometabolic, antioxidant and endothelioprotective effects in Semax. Thus, Semax can be a promising agent in hypertensive neuroretinopathy treatment.

Cerebral ischaemia is the most common cause of impaired brain function. Biologically active peptides represent potential drugs for reducing the damage that occurs after ischaemia. The synthetic melanocortin derivative, ACTH(4-7)PGP (Semax), has been used successfully in the treatment of patients with severe impairment of cerebral blood circulation. However, its molecular mechanisms of action within the brain are not yet fully understood. Previously, we used the transient middle cerebral artery occlusion (tMCAO) model to study the damaging effects of ischaemia–reperfusion on the brain transcriptome in rats. Here, using RNA-Seq analysis, we investigated the protective properties of the Semax peptide at the transcriptome level under tMCAO conditions. We have identified 394 differentially expressed genes (DEGs) (>1.5-fold change) in the brains of rats at 24 h after tMCAO treated with Semax relative to saline. Following tMCAO, we found that Semax suppressed the expression of genes related to inflammatory processes and activated the expression of genes related to neurotransmission. In contrast, ischaemia–reperfusion alone activated the expression of inflammation-related genes and suppressed the expression of neurotransmission-related genes. Therefore, the neuroprotective action of Semax may be associated with a compensation of mRNA expression patterns that are disrupted during ischaemia–reperfusion conditions.

The present work is devoted to the study of the peptides bioregulators of new generation (Cortexin, Semax, Selank) in the compensation of the disturbed Higher Nervous Functions ih the ascending row of mammls (insectivores, rodents, primates). The new data have been established that at the early stage of mammals (in insectivores) Cortexin, Semax and Selank induced the more significant and long lasted effect on inherent forms of behaviour. Their effects on the compensation of the disturbed Higher Nervous Functions have nonspecific, facilatory short time lasted character. Contrary to insectivores, rodents have a clear tendency to compesation for Higher Nervous Function disturbances. At the Semax and Selank background, the delayed conditional reflexes were restored. It has been established that the cerebroprotective effects of Selank exerted more significant influences upon the brain function disturbances in neurotic rats. Contrary to the results found in lower mammals, the application of Cortexin, Semax and Selank to neurotic monkeys exerted differnt effects upon the Higher Nervous Functions disturbances. The compensatory effects of drugs are dose dependant in nature being more effective with intranasal administration and having different effects on the various types of neurosis. The long duration compensation of the mental disturbances (the EEG homeostatic parameters of memory processes) took place during the Selank (30 mkg/ kg) administration. The cerebroprotective effects of Semax are especially significant on the operative memory. Cortexin antiamnestic and cerebroprotective effects especially significant at the inhibitory type of neurosis. The data collected on the compensatory Cortexin, Semax and Selank influeces on the disturbed memory processes and cognitive deficit of monkey’s may serve as neurophysiological basis for the more intensive and specific role of these drugs in the neurological clibic.

Este trabalho analisa os principais métodos ágeis utilizados em empresas startup, como scrum, extreme programming, kanban e lean, isolando suas práticas e mapeando-as no Kernel do SEMAT para escolher os elementos essenciais da engenharia de software que estão relacionados a cada prática de forma independente. Foram identificadas 34 práticas que foram reduzidas a um conjunto de 26 pelas similaridades. Um questionário foi desenvolvido e aplicado no ambiente de startups de software para a avaliação do grau de utilização de cada determinada prática. Através das respostas obtidas foi possível a identificação de um subconjunto de práticas com utilização acima de 60% onde todos os elementos essenciais da engenharia de software são atendidos, formando um conjunto mínimo de práticas capazes de sustentar este tipo específico de ambiente.<br>This work analyses the main agile methods used in startup companies such as scrum, extreme programming, kanban and lean, isolating their practices and mapping them on SEMAT Kernel to choose the essential elements of software engineering that are related to each practice independently. 34 practices were identified and reduced to a set of 26 by similarities. A questionnaire was developed and implemented in software startups environment to evaluate the usage degree of each particular practce. Through the responses obtained it was possible to identify a subset adopted by more than 60% of respondents where all the essential elements of software engineering are met, forming a minimum set of practices capable of sustaining this specific type of environment.

Abstract:
This study investigates the reported language use of one bilingual Semai community, a linguistic minority in Peninsular Malaysia. The Semai is the largest indigenous group in the peninsular and the language, Semai, is generally an oral language. Semai bilingual speakers are faced with choices in language behaviour that will determine whether or not the Semai language will be maintained. Given the fact that the future of the Semai language depends on its actual use, this study was undertaken to examine the reported language use patterns and the factors that contribute to maintenance or shift in one Semai community. Data for this study were collected primarily through self-administered questionnaire as well as semi-structured interviews and participant observation.;Quantitative and qualitative analyses suggest that there are no gender and age effects in the reported pattern of language use and that the community is experiencing stable diglossia. While Semai functions as the in-group language, Malay is reserved for out-group communication and used in formal domains. Additionally, results show that there is intergenerational transmission of the language to the younger generation. The reported language use patterns in the sample population appear to suggest a trend towards maintenance. Findings in the study also reveal that respondents in the sample population have a high perception of Semai vitality and a positive attitude toward their language, which they perceive as being a substantial part of their cultural identity. These findings suggest that socio-psychological factors are important and contribute to maintenance efforts in the community. This study concludes by positing that demographic factors, the values and attitudes of the people and religious homogeneity found in the community play an important role in the maintenance of the Semai language.

A leishmaniose é causada por tripanossomatídeos do gênero Leishmania que infectam preferencialmente macrófagos. Vários factores influenciam a forma e a severidade da doença: a espécies de Leishmania e a resposta imune do hospedeiro. Considerando a importância da ativação dos macrófagos na infecção, o potencial papel de CD100 na modulação da ativação dos macrófagos e os nossos dados anteriores de que CD100 solúvel (sCD100) aumenta a infectividade pelo parasita, pretendemos caracterizar os efeitos do CD100 na infecção por Leishmania (L.) amazonensis. Descobrimos que ambos, promastigotas e amastigotas, são mais infecciosos na presença de sCD100 e que o receptor CD72 é o responsável pelo aumento da infecção. Experimentos in vitro indicaram índice de infecção similares entre macrófagos nocautes para CD100 e selvagens, mas curiosamente, os animais nocautes infectados desenvolveram lesões significativamente menores do que os selvagens, sugerindo que sCD100 presente em outras células pode influenciar a formação da lesão.<br>Leishmaniasis is caused by trypanosomes of the genus Leishmania that preferentially infect macrophages. Several factors influence the form and severity of the disease: the species of Leishmania and the host immune response. Considering the importance of the activation of macrophages in infection, potential role of CD100 in the modulation of macrophage activation and our previous data that CD100 soluble (sCD100) increase the infectivity of the parasite, we intend to characterize the effect of CD100 in infection with Leishmania (L.) amazonensis. We found that both promastigotes and amastigotes, are most infectious in the presence of sCD100 and the CD72 receptor is responsible for the increased infection. In vitro experiments indicated similar infection rate of macrophages to CD100 knockouts and wild type, but interestingly, the infected knockout animals developed significantly smaller lesions than wild type suggesting that sCD100 present in other cells may influence lesion formation.
