The yeast Sen1 protein was discovered by virtue of its role in tRNA splicing in vitro. To help determine the role of Sen1 in vivo, we attempted to overexpress the protein in yeast cells. However, cells with a high-copy SEN1-bearing plasmid, although expressing elevated amounts of SEN1 mRNA, show little increase in the level of the encoded protein, indicating that a posttranscriptional mechanism limits SEN1 expression. This control depends on an amino-terminal element of Sen1. Using a genetic selection for mutants with increased expression of Sen1-derived fusion proteins, we identified mutation

Abstract Two unlinked mutations that alter the enzyme activity of tRNA-splicing endonuclease have been identified in yeast. The sen2-1 mutation does not confer a detectable growth defect, but causes a temperature-dependent reduction of in vitro endonuclease activity. Pre-tRNAs do not accumulate in sen2-1 strains. ABSTRACT The Saccharomyces cerevisiae SEN1 gene codes for a nuclear, ATP-dependent helicase which is embedded in a complex network of protein-protein interactions. Pleiotropic phenotypes of mutations in SEN1 suggest that Sen1 functions in many nuclear processes, including transcription termination, DNA repair, and RNA processing. Sen1, along with termination factors Nrd1 and Nab3, is required for the termination of noncoding RNA transcripts, but Sen1 is associated during transcription with coding and noncoding genes. Abstract:
The relationship between fracture toughness estimated using standard single edge notch bend (SENB), single edge notch tension (SENT) test specimens and fracture toughness associated with a circumferential flaw in a pipe girth weld is explored in terms of constraint using the Q parameter. It is shown that in the elastic-plastic regime, use of standard deeply notched SENB specimens provides a conservative assessment of fracture toughness, for both weld metal and HAZ, because of the high constraint associated with this specimen geometry. Use of specimen geometries and loading modes associated wit

Abstract:
Most materials for offshore applications are tested for brittle fracture resistance at a single temperature related to the minimum design temperature and by a single fracture test method. It is much rarer to perform tests at multiple temperatures to compare the fracture performance across a range of temperatures and testing methods. EWI recently compared the fracture toughness transition behaviors for an X70 steel pipe across Charpy V-notch (CVN), single-edge notched bending (SENB) crack-tip open displacement (CTOD), and single-edge notched tension (SENT) CTOD test geometries. This showed vari

Abstract:
We measure the extent of discrimination against same-sex couples by schools in Colombia using a matched-pair correspondence study. We send requests to visit private schools from several couples of different sexual orientation as conveyed by the names of the parents. We track the response rate from schools, the time to reply and the quality of the reply. We find that schools are 12 percentage points (22.3%) less likely to respond to a request sent by a homosexual couple with respect to one sent by a heterosexual one. When no information about sexual orientation is provided, the response rate de
