Abstract:
A metasomatic zone formed between the contact of a 2940 ± 5 Ma intrusive trondhjemite sheet in the Archean dunite of the Seqi Ultramafic Complex, SW Greenland, consists of three distinct mineral zones dominated by (1) talc, (2) anthophyllite, and (3) phlogopite. These zones supposedly resulted from a process of dissolution of olivine by silica rich fluid residual from the trondhjemite magma, with crystallization of secondary minerals along a compositional gradient in the fluid phase. A zircon crystal inclusion in a large (4 cm) olivine porphyroblast was dated in situ via LA-ICP-MS U–Pb isotope analysis, yielding a weighted mean 207Pb/206Pb age of 2963 ± 1 Ma, which coincides with granulite facies metamorphism and potential dehydration. Considering phase relations appropriate for the dunite composition, we deduced the talc forming conditions to be at temperatures of 600–650 °C and at a pressure below 1 GPa. This is supported by oxygen isotope data for talc, anthophyllite and phlogopite in the metasomatic zone, which suggests formation in the temperature range of 600–700 °C from fluids that had a δ18O of ~8‰ and a Δ’17O0.528 of about −40 ppm, i.e., from fluids that could have been derived from the late stage trondhjemite sheet.

Abstract:
Soil pollution has become one of the most important environmental issues in China. It is very important to evaluate soil environmental quality comprehensively and objectively. This paper proposes a soil environment quality assessment model based on the Driving Force-Pressure-State-Impact-Response (DPSIR) model and data fusion. At first, 18 evaluation indicators are selected, including complex indexes, such as the industrialization index, heavy metal pollution index, organic pollution index, potential ecological risk index, and human health risk index, and single indexes such as population density, fertilizer/pesticide application intensity, annual average air quality index, etc. Then, hierarchical analysis model is constructed, and the weight of each indicator is calculated based on Analytic Hierarchy Process (AHP) method. According to the quartile of indicator values of 32 provincial administrative divisions on the Chinese mainland, the values of each indicator are standardized and graded. Finally, the soil environmental quality index (SEQI) is calculated by the weighted average of the standard values of the 18 indicators. The assessment model is then applied in evaluating soil quality of Hebei Province, China. The results show that the soil environmental quality of Hebei’s agricultural land is in a medium state, and the industrial land is approaching the alert state. The pressure of soil pollution mainly comes from the discharge of industrial pollutants and the application of pesticides and fertilizers. Soil pollutants, such as lead, copper, zinc, benzo[a]pyrene, and benzo[a] should be especially controlled.

Introdução: O intervalo apresenta relação direta com o desempenho das repetições; contudo, suas relações com diferentes métodos de treinamento permanecem desconhecidas. Objetivo: Verificar a influência de diferentes intervalos e métodos de treinamento no desempenho das repetições. Métodos: Dezessete homens (23,50 ± 3,54 anos; 77,44 ± 7,26 kg; 176,35 ± 1,41 cm; 24,92 ± 2,49 kg/m2) realizaram quatro sessões: SEQ1 consistiu em uma série do Tríceps Puxador (TP) e Rosca Bíceps (RB), para três minutos de intervalo; SEQ2: uma série de TP e RB, com 20 segundos entre exercícios; SEQ3: uma série de TP e Abdução de Ombros (AO), com três minutos e SEQ4: uma série de TP e AO, com 20 segundos. Resultados: SEQ2 apresentou um menor número de repetições comparado às demais situações de intervalo e método (p < 0,001). Conclusão: A execução do método agonista/antagonista com intervalos curtos acarretou reduções no desempenho de repetições.

Abstract:
This study focuses on the sequence stratigraphy and the dominated seismic facies in the Central Taranaki basin. Four regional seismic sequences namely SEQ4 to SEQ1 from bottom to top and four boundaries representing unconformities namely H4 to H1 from bottom to top have been traced based on the reflection terminations. This was validated using well logs information. An onlapping feature on the seismic section indicates a new perspective surface separated between the upper and lower Giant formation, which indicates a period of seawater encroachment. This study focused extensively on deposition units from SEQ4 to SEQ1. The seismic facies, isochron map, and depositional environment were determined, and the system tract was established. This study was also able to propose a new perspective sequence stratigraphy framework of the basin and probable hydrocarbon accumulations and from the general geological aspect, SA-Middle Giant Formation (SEQ3) could act as potential traps.

Kekerasan dalam rumah tangga adalah bentuk kekerasan yang paling banyak dialami oleh perempuan berusia 25 – 40 tahun. Kecemasan merupakan salah satu bentuk reaksi emosional yang menyertai perempuan ketika mengalami kekerasan dalam rumah tangga. Kecemasan pada perempuan yang menjadi korban kekerasan dalam rumah tangga diukur dengan Hamilton Anxiety Rating Scale (HARS) dan melihat gejala kecemasan dari segi kognitif, somatis, motorik dan afektif. Penelitian ini menggunakan metode kualitatif untuk menggambarkan kecemasan dan metode kuantitatif untuk melihat penurunan tingkat kecemasan dengan terapi seni pada perempuan korban kekerasan dalam rumah tangga. Subyek penelitian ini adalah dua perempuan yang telah bercerai dan mengalami kekerasan dalam rumah tangga dalam bentuk fisik, seksual, psikis atau verbal dan penelantaran rumah tangga. Teknik pengambilan sampel dengan menggunakan metode purposive sampling. Pemberian terapi seni pada masing-masing subyek dilakukan sebanyak tujuh sesi. Dalam penelitian ini, terapi seni telah terbukti dapat mengurangi kecemasan pada perempuan korban kekerasan dalam rumah tangga dengan menunjukan perubahan tingkat kecemasan sebelum dan sesudah intervensi. Keberhasilan terapi seni ini juga dipengaruhi oleh adanya kesadaran pada masing-masing subyek untuk konsisten menjalani terapi.

Gedung seni pertunjukan, merupakan wadah bagi para seniman untuk berkarya, baik seni musik, seni peran, maupun seni rupa. Pada umumnya, gedung seni pertunjukan bisa berupa kawasan yang merupakan pusat seni pertunjukan. Semakin banyaknya musisi/seniman yang lahir di Indonesia maupun di dunia, mendorong banyaknya kebutuhan akan fasilitas pertunjukan para musisi tersebut. Di abad ke-21, sudah tercatat hampir 50 pusat seni pertunjuan iconic di dunia yang diakui tidak diragukan lagi kualitasnya. Dari ke-limapuluh gedung tersebut, hanya 5 dari Asia, itu pun di China dan Singpura. Sedangkan di Indonesia, baru beberapa pusat seni pertunjukan. Namun, belum ada yang iconic dan memiliki kualitas akustik yang memadai. Dan dari segi jumlah wisatawan seni pertunjukan Jakarta barat menempati jumlah paling sedikit di antara kawasan lainnnya, ironisnya di Jakarta barat terdapat pusat seni pertunjukan yaitu Kawasan Kota Tua, namun di lihat dari berbagai faktor banyak kekurangan yang kurang mendukung untuk berkembangnya seni di kawasan itu. Atas permasalahan tersebut, maka diperlukan perancangan pusat seni pertunjukan dengan program ruang yang lengkap dari segi tata ruang, aksesibel dari segi pencapaiannya, kualitas akustik yang baik dari segi interior dan dari segi eksterior. Perancangan GEDUNG SENI PERTUNJUKAN JAKARTA ini akan di integrasikan denagn ruang public sekitar, serta bagian eksterior mangambil tema kawasan dan konsep kawasan agar dapat selaras dengan lingkugan sekitar.

Abstract The only known elements encoding enterotoxins in coagulase-negative staphylococci are composite Staphylococcus epidermidis pathogenicity islands (SePIs), including SePI and S. epidermidis composite insertion (SeCI) regions. We investigated 1545 Staphylococcus spp. genomes using whole-genome MLST, and queried them for genes of staphylococcal enterotoxin family and for 29 ORFs identified in prototype SePI from S. epidermidis FRI909. Enterotoxin-encoding genes were identified in 97% of Staphylococcus aureus genomes, in one Staphylococcus argenteus genome and in nine S. epidermidis genomes. All enterotoxigenic S. epidermidis strains carried composite SePI, encoding sec and sel enterotoxin genes, and were assigned to a discrete wgMLST cluster also containing genomes with incomplete islands located in the same region as complete SePI in enterotoxigenic strains. Staphylococcus epidermidis strains without SeCI and SePI genes, and strains with complete SeCI and no SePI genes were identified but no strains were found to carry only SePI and not SeCI genes. The systematic differences between SePI and SeCI regions imply a lineage-specific pattern of inheritance and support independent acquisition of the two elements in S. epidermidis. We provided evidence of reticulate evolution of mobile elements that contain elements with different putative ancestry, including composite SePI that contains genes found in other coagulase-negative staphylococci (SeCI), as well as in S. aureus (SePI-like elements). We conclude that SePI-associated elements present in nonenterotoxigenic S. epidermidis represent a scaffold associated with acquisition of virulence-associated genes. Gene exchange between S. aureus and S. epidermidis may promote emergence of new pathogenic S. epidermidis clones.

Abstract Rearrangement of the MLL gene at chromosome 11q23, which occurs in 80% of infants and 5% of older children with acute lymphoblastic leukemia (ALL), confers a dismal prognosis despite maximally intensified chemotherapy and allogeneic transplantation. Novel therapies are clearly needed for these patients. The FLT3 tyrosine kinase represents an attractive potential target in these leukemias, due to the marked overexpression of FLT3 in cases of MLL-rearranged ALL. We have previously shown that MLL-rearranged infant and childhood ALL leukemic cells express high levels of constitutively activated FLT3 receptor protein, and that these cells are selectively killed by exposure to the FLT3 inhibitor CEP-701. However, FLT3 inhibition is unlikely to represent curative therapy for these patients if used as monotherapy. Thus, we examined various combinations of CEP-701 with chemotherapy to look for potentially synergistic therapeutic strategies. We performed MTT cytotoxicity and annexin V binding (AVB) apoptosis assays on MLL-rearranged ALL cell lines (SEM-K2 and HB-1119) and three primary patient samples (all of which we show to express high levels of constitutively activated FLT3) after exposure to multiple dose combinations of CEP-701 and six chemotherapy agents (vincristine, l-asparaginase, dexamethasone, daunorubicin, etoposide and cytarabine). Since we have previously demonstrated that the nature of the interactions between CEP-701 and chemotherapy agents (i.e., synergistic, additive or antagonistic) are sequence-dependent in AML cells, we performed the MTT and AVB assays in three sequences designed to mimic the potential clinical uses of the combinations (Seq1: chemotherapy followed by CEP-701; Seq2: simultaneous exposure to both; and Seq3: CEP-701 followed by chemotherapy). The nature of the interaction between CEP-701 and each chemotherapy agent was determined using the median effect method of Chou and Talalay, which calculates a combination index (CI) for each combination (CI &lt; 0.9 - synergistic; CI 0.9-1.1 - additive; CI &gt; 1.1 antagonistic). A striking pattern of sequence-dependent synergy was observed: Seq1 was markedly synergistic (mean CI +/− SD = 0.59 +/− 0.18), Seq2 was additive (CI = 0.99 +/− 0.29) and Seq3 was antagonistic (CI = 1.23 +/− 0.40). The sequence-dependence is attributable to the G1 cell cycle arresting effects of CEP-701 in these cells. An MLL-rearranged cell line that expresses low levels of minimally activated FLT3 (RS4-11) demonstrates minimal cell cycle arrest when treated with CEP-701, and mere additivity in all 3 CEP-701/chemotherapy combination sequences (Seq1: CI = 0.95 +/− 0.10; Seq2: CI = 1.01 +/− 0.09; Seq3: CI = 1.06 +/− 0.10), confirming that the observed pattern of synergy is mediated through the effects of CEP-701 on FLT3 signaling. These results can help guide the combination of CEP-701 with existing chemotherapy regimens for infants and children with MLL-rearranged ALL, with the goal of improving the dismal outcome for these patients.

AbstractThe purpose of this study was to examine the acute effects of multi- to single-joint or the reverse exercise order on repetition performance and perceived exertion for the pectoralis major. Fourteen trained men (24.05 ± 4.17 yrs, 78.85 ± 3.51 kg, 175.42 ± 4.01 cm) underwent two different training sequences (SEQ1 and SEQ2). In SEQ1, all subjects performed 5 sets for maximal repetitions, with a 2-min rest interval, of the bench press followed by the machine chest fly with 10 repetitions maximum load. In SEQ2, the same procedures were repeated, but with the reverse order. The t-test did not show any differences (p = 0.140) in total workout repetitions between SEQ1 (62.22 ± 11.00 repetitions) and SEQ2 (55.40 ± 8.51 repetitions). Conversely, the total repetition number for the bench press exercise was significantly greater (p = 0.001) following SEQ1 (34.36 ± 4.68 repetitions) compared to SEQ2 (25.85 ± 6.73 repetitions). In contrast, the total repetition number for the machine chest fly exercise following SEQ2 was significantly greater (p = 0.001) (33.50 + 4.11 repetitions) compared to SEQ1 (27.85 ± 6.52 repetitions). Despite no significant differences found for the rating of perceived exertion (RPE) values between SEQ1 and SEQ2 for the barbell bench press in all sets (p ≥ 0.083), significantly higher RPE values for the machine chest fly were observed over the first three sets following SEQ1 compared to SEQ2 (p < 0.01). In conclusion, the total workout repetitions were not significantly different when performing the traditional multi- to single-joint or the reverse exercise order when training the pectoralis major muscle.

Introdução: entre as diversas variáveis do treinamento de força (TF), a ordem dos exercícios provavelmente é uma das mais ignoradas pelos praticantes e treinadores na sua prescrição. Objetivo: investigar o efeito de diferentes ordens de exercícios nas respostas da pressão arterial (PA) após uma sessão de TF. Métodos: oito mulheres normotensas com experiência prévia em TF realizaram três protocolos distintos: SEQA foi adotado uma ordem de alternância dos segmentos; SEQB os exercícios foram realizados dos membros inferiores para os membros superiores; e SEQC a ordem foi oposta ao SEQB. Após o treinamento, os valores pressóricos (sistólico, diastólico e médio) foram analisados ao longo de 60 minutos a partir de um aparelho digital. Resultado: a sessão de TF não promoveu efeito hipotensivo pós esforço redução em nenhuma das diferentes ordens e não houve diferença significativa entre os protocolos. Conclusão: a ordem dos exercícios no TF não é uma variável que influencie na PA pós esforço.

Learning new information is crucial in daily activities and occurs continuously during a subject’s lifetime. Retention of learned material is required for later recall and reuse, although learning capacity is limited and interference between consecutively learned information may occur. Learning processes are impaired in Parkinson’s disease (PD); however, little is known about the processes related to retention and interference. The aim of this study is to investigate the retention and anterograde interference using a declarative sequence learning task in drug-naive patients in the disease’s early stages. Eleven patients with PD and eleven age-matched controls learned a visuomotor sequence, SEQ1, during Day1; the following day, retention of SEQ1 was assessed and, immediately after, a new sequence of comparable complexity, SEQ2, was learned. The comparison of the learning rates of SEQ1 on Day1 and SEQ2 on Day2 assessed the anterograde interference of SEQ1 on SEQ2. We found that SEQ1 performance improved in both patients and controls on Day2. Surprisingly, controls learned SEQ2 better than SEQ1, suggesting the absence of anterograde interference and the occurrence of learning optimization, a process that we defined as “learning how to learn.” Patients with PD lacked such improvement, suggesting defective performance optimization processes.

The ability to perform accurate sequential movements is essential to normal motor function. Learning a sequential motor behavior is comprised of two basic components: explicit identification of the order in which the sequence elements should be performed and implicit acquisition of spatial accuracy for each element. Here we investigated the time course of learning of these components for a first sequence (SEQA) and their susceptibility to interference from learning a second sequence (SEQB). We assessed explicit learning with a discrete index, the number of correct anticipatory movements, and implicit learning with a continuous variable, spatial error, which decreased during learning without subject awareness. Spatial accuracy to individual sequence elements reached asymptotic levels only when the whole sequence order was known. Interference with recall of the order of SEQA persisted even when SEQB was learned 24 h after SEQA. However, there was resistance to interference by SEQB with increased initial training with SEQA. For implicit learning of spatial accuracy, SEQB interfered at 5 min but not 24 h after SEQA. As in the case of sequence order, prolonged initial training with SEQA induced resistance to interference by SEQB. We conclude that explicit sequence learning is more susceptible to anterograde interference and implicit sequence learning is more susceptible to retrograde interference. However, both become resistant to interference with saturation training. We propose that an essential feature of motor skill learning is the process by which discrete explicit task elements are combined with continuous implicit features of movement to form flawless sequential actions.

Merujuk dari latar belakang masalah tersebut maka penulis tertarik untuk mengkaji lebih dalam mengenai pelaksanaan kegiatan ekstrakurikuler seni baca Al-Qur’an di SMAN 1 Tanjung Mutiara. Penelitian ini bertujuan untuk mengetahui pelaksanaan kegiatan, faktor pendukung dan faktor penghambat dalam pelaksanaan kegiatan ekstrakurikuler seni baca Al-Qur’an. Jenis penelitian yang penulis gunakan dalam penelitian ini adalah Field Research (penelitian lapangan). Sedangkan metode penelitian yang penulis gunakan adalah metode deskriptif dengan pendekatan kualitatif. Hasil penelitian ini menyatakan bahwa proses pelaksanaan dari kegiatan seni baca Al-Qur’an, dari segi pelaksanaan terbagi atas tiga yaitu persiapan, pelaksanaan dan evaluasi, dari segi pelaksanaan dan evaluasi guru sudah melaksanakan dengan baik, namun dari segi persiapan masih kurang karena guru tidak menggunakan perencanaan dalam proses kegiatan tersebut sehingga pembelajaran tidak terarah dari segi waktu. Adapun faktor pendukung dari pelaksanaan kegiatan seni baca Al-Qur’an adalah dari dukungan kepala sekolah yang memberikan izin pelaksanaan kegiatan ekstrakurikuler seni baca Al-Qur’an, lingkungan belajar, bakat dan minat peserta didik. Sedangkan faktor penghambat dalam kegiatan ini kurangnya fasilitas belajar dari pihak sekolah seperti tidak adanya Al-Qur’an dan pengeras suara, dari peserta didik faktor penghambat yaitu dari beberapa orang peserta didik yang tidak memiliki bakat seperti suara yang tidak bagus dan suara pendek. Ketebatasan waktu, dari guru kurangnya persiapan dan metode yang kurang bervariasi. Terakhir kurangnya dukungan dari orang tua. Hal ini yang menjadi penyebab timbulnya masalah-masalah diatas. Penelitian ini menggunakan pendekatan kuantitatif, jenis penelitian deskriptif, teknik pengumpulan data observasi nonpartisipan, sumber data sekunder diperoleh melalui video dari channel youtube PBVSI_OFFICIAL sejumlah 6 pertandingan Proliga Final Four Seri Kediri tahun 2019. Intrumen penelitian adalah rekaman pertandingan final four Seri Kediri tahun 2019 dan form statistik serangan open spike, quick, semi, back attack, serve. Teknik analisa data menggunakan rumus efektivitas. Hasil penelitian menunjukkan bahwa serangan yang paling efektif sebagai berikut: open spike memiliki tingkat efektivitas serangan sebesar 15,9%, semi memiliki tingkat efektivitas serangan sebesar 0,5%, quick memiliki tingkat efektivitas serangan sebesar 4,3%, back attack memiliki efektivitas serangan sebesar 6,1% dan serve memiliki efektivitas serangan sebesar 2,8%. Kesimpulan studi ini menyatakan bahwa serangan yang paling efektif digunakan pada pemain bolavoli level profesional yang berlaga di proliga 2019 final four seri Kediri adalah open spike berada diurutan paling pertama, back attack di urutan kedua, quick di urutan ketiga, serve diurutan keempat, dan teknik semi di urutan kelima. Keterbatasan penelitian ini terletak di pengambilan data yakni hanya dilakukan pada satu seri final four proliga bolavoli seri Kediri.

Introdução: A ordem dos exercícios interfere diretamente no desempenho de força. Objetivo: Investigar a influência de diferentes ordens de execução de exercícios em uma sessão de treinamento de força sobre o desempenho de repetições. Métodos: Dezoito homens treinados realizaram dois protocolos experimentais randomizados: SEQA – supino reto (SR), supino inclinado (SI), desenvolvimento na máquina (DM), abdução de ombros (AO), tríceps pulley (TP) e tríceps francês (TF); SEQB – ordem inversa. Três séries com cargas de dez repetições máximas foram realizadas para todos os exercícios. Resultados: Observaram-se diferenças significativas no trabalho total (repetições x séries) para os exercícios SR, SI, DM, AO, e TF entre SEQA e SEQB, exceto no TP. Não houve diferenças significativas entre as sequências (p = 0,07), considerando o somatório de repetições para SEQA (2225 ± 2,44) e SEQB (2337 ± 3,02). Conclusão: Sugere-se que os exercícios prioritários sejam realizados no início da sessão de treinamento, independente do tamanho do grupo muscular.

Abstract:
Background Single-cell RNA-sequencing (scRNA-seq) technology is a powerful tool to study organism from a single cell perspective and explore the heterogeneity between cells. Clustering is a fundamental step in scRNA-seq data analysis and it is the key to understand cell function and constitutes the basis of other advanced analysis. Nonnegative Matrix Factorization (NMF) has been widely used in clustering analysis of transcriptome data and achieved good performance. However, the existing NMF model is unsupervised and ignores known gene functions in the process of clustering. Knowledges of cell markers genes (genes that only express in specific cells) in human and model organisms have been accumulated a lot, such as the Molecular Signatures Database (MSigDB), which can be used as prior information in the clustering analysis of scRNA-seq data. Because the same kind of cells is likely to have similar biological functions and specific gene expression patterns, the marker genes of cells can be utilized as prior knowledge in the clustering analysis. Methods We propose a robust and semi-supervised NMF (rssNMF) model, which introduces a new variable to absorb noises of data and incorporates marker genes as prior information into a graph regularization term. We use rssNMF to solve the clustering problem of scRNA-seq data. Results Twelve scRNA-seq datasets with true labels are used to test the model performance and the results illustrate that our model outperforms original NMF and other common methods such as KMeans and Hierarchical Clustering. Biological significance analysis shows that rssNMF can identify key subclasses and latent biological processes. To our knowledge, this study is the first method that incorporates prior knowledge into the clustering analysis of scRNA-seq data.

Wheat (Triticum aestivum L.) lines containing the 1BL/1RS chromosome translocation yield up to 20% more than established wheat cultivars in some Queensland environments. However, 1BL/1RS germplasm possesses a quality defect known as ‘sticky dough’, which is incompatible with the high-speed dough-mixing processes used for bread production in Australia. Therefore, we investigated whether the 1BL/1RS translocation conveyed a yield advantage to locally adapted germplasm across a wide range of environments that was sufficient to justify attempting to overcome the ‘sticky dough’ defect either through plant breeding or by altering the mixing processes. Three sets of recombinant inbred lines (RILs) that segregated for the presence or absence of the 1BL/1RS translocation were developed from crosses between 1BL/1RS germplasm (Seri and Genaro) and established local cultivars (Hartog and Banks), and grown in 11 environments representing six sites across southern Queensland and northern New South Wales and two years. The effect of the 1BL/1RS translocation on grain yield depended on environment and genetic background. In semi-dwarf genotypes of the Hartog/Seri and Hartog/Genaro crosses, the 1BL/1RS RILs had lower grain yield than the 1B RILs in the three lowest yielding environments. This effect was associated with changes in grain number per unit area, suggesting that the negative yield effect of the translocation is expressed before, or at, anthesis. In the higher yielding environments, the 1BL/1RS translocation conveyed a yield advantage in semi-dwarf genotypes of the Banks/Seri cross, but had no consistent effect on yield in semi-dwarf genotypes of the Hartog/Seri and Hartog/Genaro crosses. The 1BL/1RS translocation was also associated with decreased yield in the double-dwarf genotypes of the Hartog/Seri cross across all environments. We conclude that the 1BL/1RS translocation is not useful for local breeding programs, as it decreased yield among the more advanced, semi-dwarf germplasm in low-yielding environments that potentially represent up to 85% of the target population of environments, and had no consistent positive effect on yield in this germplasm in higher yielding environments.

386 Background: Current standard combination first line palliative chemotherapy regimens in advanced pancreatic ductal adenocarcinoma (PDAC) have not been compared in head-to-head trials. Data on optimum treatment sequencing is also lacking. Methods: To assess whether first line (1L) treatment with gemcitabine/nab-paclitaxel (Gem/Nab-P)(SEQ1) or FOLFIRINOX (SEQ2) in the palliative treatment setting impacts survival outcomes, data for patients receiving palliative intent combination chemotherapy between 2016 and May 2020 was extracted from PURPLE, a prospective pancreatic cancer registry enrolling consecutive patients across multiple institutions. Results: Of 637 patients, 180 (28%) who received 1L single agent therapy and/or palliative radiotherapy were excluded. Of the remaining 449 patients, 132 (29%) had locally advanced disease (LA PDAC), 67 had local recurrence (15%), and 250 (56%) had de novo metastatic disease (mPDAC). Patients receiving 1L Gem/Nab-P (n=376, 84%) were older (median 67 vs 59 years, P<0.001), had a higher Charlson Comorbidity Index (CCI) (CCI ≥2: 18% vs 3%, Odds Ratio [OR] 8.0, P=0.002), and poorer performance status (ECOG≥2: 10% vs 1%, OR 8.4, P=0.01) compared to the 1L FOLFIRINOX group (n=73, 16%). 140 (37%) patients receiving 1L Gem/Nab-P (SEQ1) and 32 (44%) patients receiving 1L FOLFIRINOX (SEQ2) received second line (2L) chemotherapy. SEQ1 2L regimens included FOLFIRINOX (n=14), FOLFIRI (n=49), FOLFOX (n=35) and 5FU alone (n=3). SEQ2 2L regimens included Gem/Nab-P (n=19), Gem/5FU (n=4), Gem/Cisplatin (n=1) and gemcitabine alone (n=5). Median progression free survival (mPFS) did not differ between patients receiving 1L Gem/Nab-P vs 1L FOLFIRINOX (5.7 vs 5.1 months, P=0.54); nor did median overall survival (mOS; 11.3 vs 12.3 months, P=0.37). In the subset of patients who went on to receive 2L chemotherapy, mPFS in 2L was shorter for SEQ1 compared to SEQ2 (2.9 vs 5.2 months, Hazard Ratio [HR] 1.3, P=0.03) but mOS did not differ (15.9 vs 17.3 months P=0.91). In the subset with LA PDAC, mPFS in 2L was comparable (2.9 vs 3.3 months, P=0.55) but mOS was significantly longer with SEQ1 (22.5 vs 13.8 months, HR 0.50, P=0.01). In mPDAC, mPFS in 2L was shorter with SEQ1 (2.3 vs 5.6 months, HR 1.64, P=0.03), but mOS did not differ by sequence (13 vs 17 months, P=0.23). Conclusions: There were no significant differences in survival outcomes between 1L choices of chemotherapy, despite patients offered 1L FOLFIRINOX (SEQ2) being younger and fitter. Survival differences observed for LA PDAC versus mPDAC will be further explored. Given the multiple potential confounders, randomised clinical trials are needed to make firmer conclusions regarding the optimal initial treatment for each patient subset.
