Abstract:
<p>Sherlock BBC (2010) is one of the latest adaptations of Sherlock Holmes. By using the modern era as its sett ing, Sherlock off ers something different compared to previous Sherlock Holmes adaptations. This research focuses on the representation of female characters on Sherlock according to an issue that already chosen which is women and crime. The theories and concepts which are used on this research are gender construction in Victorian Era, the concept of femme-fatale and crime fi ction characteristics. I argue that there are some substantial diff erences of female characters’ representation in Sherlock compared to the original story. Despite those diff erences, female characters in Sherlock are still represented in a similar manner as the crime fictions characteristics in which women are always put in an inferior position and<br />the one who need men’s help.</p>

Abstract:
The aim of this article is to develop the concepts of masks and masking to interrogate the role of institutions in the co-production of ‘untouchable’ celebrity icon status. The empirical focus is the multi-institutional masking of Sir Jimmy Savile OBE KCSG. For decades, Savile was celebrated as one of the UK’s best-loved celebrity icons. One year after his death, he was exposed as a serial sexual predator. We argue that the largely compartmentalised official reports on Savile have presented a partial analysis. They have emphasised the importance of Savile’s celebrity status while taking it for granted, downplayed the significance of his moral standing in British society, and marginalised the proactive, enabling role of the BBC, the NHS and the British establishment. However manipulative the individual, we propose that it was Savile’s cumulative multi-institutional masking as celebrity personality (the BBC), celanthropist (the NHS) and, ultimately, celebrity icon (the British establishment) that co-produced his ‘untouchable’ status and enabled him for decades to deflect and discredit rumour, gossip and allegations about his sexually predatory behaviour. We conclude by reflecting on the ‘researchability’ of powerful elites, and by suggesting how our analysis might inform further research into the power dynamics that have co-produced the ‘untouchability’ of other celebrities subsequently exposed as serial sexual predators.

Abstract:
In 1974 the BBC screened the twelve-part documentary serial The Family. Yet despite the title of the programme, and its promise to open up the gendered terrain of the domestic sphere, The Family has largely been conceptualised with regard to discourses of class rather than gender. Given the famous slogan of second-wave feminism that the ‘personal is political’, The Family provides a fertile terrain upon which to consider how discourses relating to the women's movement at the time were negotiated within a particularly (tele)visible domestic sphere. It was often at the level of the micro-political – the everyday oppressions in women's daily lives – that the second wave often sought to politicise the nature of female subjectivity (Tyler 1997). In 1974, many critics and viewers lamented the fact that the wider social insight promised by The Family's publicity failed to transpire, suggesting that it was ultimately about ‘nothing much’. In challenging this view, this article seeks to contribute to the project of writing women, at the level of representation and critical reception, back into the history of canonical documentary texts, a process which can involve revisiting documentaries that have been untouched by feminist scholarship ( Waldman and Walker 1999 ). In doing so, it draws upon archival research undertaken at the BBC Written Archive Centre as based upon press cuttings, internal production memos and BBC Audience Research reports.

Abstract:
This article explores how nostalgia for both the Cold War and the 1970s in general became a key feature of the BBC drama The Game (2014). It argues that the serial situated the Cold War as a more stable era in international relations in which the enemy played by a specific set of rules, thus leading to a danger that was manageable and more predictable than the terror threat of the twenty-first century. Furthermore, the article argues that the serial presents the 1970s as a golden age which was defined by the continuity of consensus politics and communities of class and family. Finally, the article examines how this nostalgia is reinforced by narrative devices which engage with generic features such as the storyline playing out like a game. However, in the re-imagined Cold War of the twenty-first century, the traditional chess trope has been replaced by the more complex game of Alice Chess.

This article examines two adaptations of John le Carré’s 1974 novel Tinker, Tailor, Soldier, Spy: the 1979 BBC television serial and the 2011 cinema film, in order to investigate critical orthodoxies around the aesthetics of television and cinema. It examines the dialectical relationship between space and place, concluding that filming location acts as a ‘framing place’, shaping production practices and effecting a dialogic interchange between production space and narrative place. Drawing on original research interviews with production team members, it illuminates the process of production and demonstrates the interaction between material space and social space in the interaction of location and practitioners. The article concludes that the television serial's mimetic fidelity to its source novel results in a text which is at times more ‘cinematic’ than the cinema version. The cinema film features a more fully developed visual concept throughout its aesthetic, in the motifs of cages, grids and boxes. However, in its narrative compression and unsubtle use of signifiers of place, the cinema adaptation is at times less ‘cinematic’ than the television serial.

Abstract:
This article analyses three serialised adaptations of John le Carré novels produced by the BBC: Tinker Tailor Soldier Spy (1979), Smiley's People (1982) and A Perfect Spy (1987). It aims firstly to position them in the context of developments and trends during the period of the serials' production. It explores how, on the one hand, they were produced as variants on the classic serial model which aimed for a more contemporary focus and aesthetic in response to concurrent developments in British television drama, and on the other, how they have a complex and ambivalent relationship with the genre of television spy fiction. Secondly, this article builds upon this positioning of the serials to explore how the themes of le Carré’s novels are interpreted specifically for the television medium. Central to this is the issue of temporal displacement, as television's process of ‘working through’, often considered as characteristic of the medium's immediacy and ‘liveness’, is in this case delayed over many years by a cycle of continual adaptation. Here a particular narrative – the defection of Kim Philby in 1963 – resonates across three decades and is worked through in a variety of approaches, initially in the novels and subsequently reworked on television. It then examines how this manifests in the television adaptations in a contemporary heritage aesthetic which is complex and highly troubled.

Screen performance is characterised by the interaction of performance, place and screen, but has suffered from critical neglect. Most accounts of this collaborative process focus on the performer in front of the camera rather than on the activities going on behind it. This article examines how performance is shaped within the BBC’s Outside Broadcast (OB) drama production paradigm of the 1970s and 1980s. The interactions of performers and camera operators combine to construct on-screen performance, since the generation of the screen frame necessarily ‘frames’ performance both literally and figuratively. Camera operators therefore offer ‘invisible’ performance through their manipulation of the frame and the choreography of their interactions with actors. The article’s key case study is The Mayor of Casterbridge (1978) , the BBC’s first drama serial to be made entirely on location on OB videotape, and also considers OB production on Boys from the Blackstuff (BBC 1982) .

Abstract:
This article analyzes the Belfast-set BBC series The Fall (2013–) as an illustrative example of television noir. It aims to use noir scholarship to investigate The Fall’s complex gender politics and genre position, and, more significantly, to use The Fall to illuminate the complex ways in which noir currently operates across Anglo-American television and culture. The Fall is self-conscious, if not self-reflexive, in its mobilization of noir to aspire to the cinematic. This article argues that the series, and others like it, use noir as a legitimation strategy, often to excuse prurient stories of sexualized violence. The act of labeling something noir, particularly a visual fiction, is a way of insisting on its status as art. I conclude that the system of noir (and its associations with art and authenticity) is unable to contain the excesses of the serial killer mythology and Gothic inflection of its postfeminist investigator.

This thesis focuses on the various adaptations of Arthur Conan Doyles novel The Hound of the Baskerville and accentuate the episode The Hounds of Baskervilles from the BBC TV series Sherlock. Thus, this thesis is decribing the theme, which was not fully covered in Czech literature. The author used different sources, books, internet pages and cinematographic pieces. The thesis consists of two parts. In the first part the author is focusing on the theory of adaptation, Doyle's life and art, The Hound of the Baskervilles book and the TV series Sherlock. In the practical part the author narrates about the episode The Hounds of the Baskervilles and searching for definition of the adaptation process, which the makers of this episode used. This part also answers the question, why is this episode so popular and key for the whole series. The crucial final part of the thesis summarizes the main principles, which authors and film makers can use during the process of the adaptation. The thesis deals with contemporary Czech serial production in terms of script development. It focuses primarily on the development of screenplays for the series on Česká televize and on drama series with one-hour episodes. It compares the development in the Czech environment with British and American practice. It maps the current Czech serial environment, creates an overview of contemporary Czech serials in broadcasting and clarifies the basics of narrative serial structure. It contains a comparison of selected pilot parts in terms of their narrative structure. It focuses on both screenwriting practice and development practices from the point of view of the television of public service, in the case of the American production it focuses on the Hollywood system. It uses methods of qualitative analysis of scripts, in-depth semi-structured interviews with creative producers and comparison of script development in individual productions.

Abstract:
Main focus of my Master's Thesis is to conduct a research of mythological elements and themes that appear within the narrative structure of selected episodes of BBC television series Doctor Who, deriving from the initial hypothesis that such mythological patterns are still recurring and repeating even in the structure of stories produced by modern show-business industries, which renders their content intrinsically more attractive for any audience. We analyse selected episodes from both arks of the Doctor Who's story individually - the old one, which began in the sixties, and the new one, which is considered to be a reboot of the original series and aired in 2005. The originally intended educative element of the series and its sudden disappearance is also a part of our research. Main analysis of our paper consists of a semiotic analysis of the text of the television series utilizing the point of view of the critical reader that was introduced by Umberto Eco, and deriving from the definition of modern myth that appears in works of Roland Barthes, archaic myth, studied for example by Carl Lévi- Strauss, and Jungian archetypes.

This study makes a unique contribution to applied game theory and to the studies of Shiji (Records of Grand Historian) and the Warring States Period of ancient China (453-221B.C.) by constructing and analyzing the annual series of alliance structures or partitions of the seven states during the period of two hundred thirty three years. It shows that twenty six of the eight hundred seventy seven possible partitions were observed, and that the three most frequent partitions were the finest partition (146 years), partitions with four singletons and one three-member coalition (63 years), and partitions with five singletons and one two-member coalition (33 years). Such quantitative results have future applications in alliance studies, game theory, and international economics. They also provide a list of future research topics such as the unknown statistical properties of a series of partition of seven elements with 233 observations.

Abstract:
In this study we develop a new method for testing the null hypothesis that the predictor function in a canonical link regression model has a prescribed linear form. The class of models, which we will refer to as canonical link regression models, constitutes arguably the most important subclass of generalized linear models and includes several of the most popular generalized linear models. In addition to the primary contribution of this study, we will revisit several other tests in the existing literature. The common feature among the proposed test, as well as the existing tests, is that they are all based on orthogonal series estimators and used to detect departures from a null model. Our proposal for a new lack-of-fit test is inspired by the recent contribution of Hart and is based on a Laplace approximation to the posterior probability of the null hypothesis. Despite having a Bayesian construction, the resulting statistic is implemented in a frequentist fashion. The formulation of the statistic is based on characterizing departures from the predictor function in terms of Fourier coefficients, and subsequent testing that all of these coefficients are 0. The resulting test statistic can be characterized as a weighted sum of exponentiated squared Fourier coefficient estimators, whereas the weights depend on user-specified prior probabilities. The prior probabilities provide the investigator the flexibility to examine specific departures from the prescribed model. Alternatively, the use of noninformative priors produces a new omnibus lack-of-fit statistic. We present a thorough numerical study of the proposed test and the various existing orthogonal series-based tests in the context of the logistic regression model. Simulation studies demonstrate that the test statistics under consideration possess desirable power properties against alternatives that have been identified in the existing literature as being important.

Thrombin (Th) binds specifically to confluent cultures of bovine aortic (ABAE) and brain capillary (BBC) endothelial cells. Saturation of 125I-Th binding is observed after 1 h exposure to the ligand and at an extracellular concentration of 0.5 and 1.0 µg/ml for ABAE and BBC cells, respectively. Under optimal conditions both ABAE and BBC cultures bind about 2 to 5 ng/106 cells, which represents about 20% of Th binding.to bovine corneal endothelial (BCE) cells. The cell associated 125I-Th in ABAE and BBC cells is internalized and degraded as described in BCE cells. The nature of the cell associated radioactivity is analyzed on SDS-polyacrylamide -gel electrophoresis and in ABAE and BBC cells about 30% of the I-Th appears in a complex with protease nexin I (PN I) while in BCE cells about 70% of the binding is mediated by PN I. ABAE cells possess 3 types of complexes, one which appears only on the cell surface with a molecular weight of 78 kDa, and two other complexes which appear only in the conditioned medium (CM) with molecular weights of 84 and 85 kDa. BBC and BCE cells demonstrate only one type of complex with a molecular weight of 77 kDa which appears both on the cell surface and in the CM. Preincubation of BCE cultures in the presence of Th is known to up-regulate the amount of PN I on the cell surface and in the CM, but this Th induced up-regulation effect is not observed in ABAE or BBC cells.The results described above indicate a difference between ABAE and BBC cells although both cell types growunder similar conditins and demonstrate similar morphological appearance. However, in both vascular endothelial cell types the total amount of PN I and its metabolism is relatively small compared to corneal endothelial cells. It, therefore, may indicate the lower capacity of vascular endothelial cells to control serine proteases activity at or near their cell surfaces as compared to corneal endothelial cells. The study sample consisted of the mango, avocado and lemon production indices 2000-2020. As a result, the models were obtained arima mango (1,0,0) (2,1,2) (AIC=5448.99, BIC=5473.35 and RMSE=19067.93), arima avocado (0,1,3) (2,1,0) (AIC=4687.05, BIC=4707.91 and RMSE=4114.35) and arima lemon (1,0,1) (0,1,1) (AIC=4484.36, BIC=4501.76 and RMSE=2551.96) with a 12 months period, the diagram of boxes and whiskers was also made with which it was identified that atypical data (Outliers) abound in the periods of greatest production.

Introduction: Metaplastic breast carcinoma (MBC) is understood as a heterogeneous group of malignant tumors, which exhibits the transformation of part or its entire glandular carcinomatous component into a nonglandular component by means of epithelial and mixed mesenchymal differentiation. The MBC is a rare and aggressive breast cancer, accounting for approximately 1% of all breast tumors that have a worse prognosis. The aim of this study was to describe four cases of MBC, analyzing the progression of patients in a mean follow-up of 25 months (11–43 months). Case series: All patients were female. The mean age of the four cases of the series was 50 years (40–61 years). All patients had tumors that were histologically classified as metaplastic carcinoma. The mean tumor size of the series was 8.3 cm (2.2–15.5 cm). Two patients had the angiolymphatic invasion, and none had the perineural invasion. In three cases, there was an axillary node involvement. None of the cases had metastasis at the time of diagnosis. All patients underwent surgical treatment. Concerning complementary treatment, all patients underwent chemotherapy, three underwent radiation therapy, and two received hormone therapy. Three patients had systemic recurrence with metastases (i.e., all progressed to death). The study was approved by a Research Ethics Committee, under CAAE Nº 30154720.0.0000.5209. Conclusion: With a mean follow-up of 25 months (11–43 months), three out of four cases (75%) presented systemic recurrence with metastases (i.e., all culminating in death), and one patient is alive without evidence of malignancy.

Abstract In this paper we present a solution to the inverse kinematics problem for serial manipulators of general geometry. The method is presented in detail as it applies to a 6R manipulator of general geometry. The equations used are derived using a linearization method and dialytic elimination. In doing this, all variables except one, a tangent half angle of a joint variable, can be eliminated. The result is a 16 by 16 matrix in which all terms are linear in the suppressed variable. The unique design of this matrix allows the suppressed variable to be solved as an eigenvalue problem. Substituting these values of the suppressed variable back into the equations, all other joint variables can be found using linear equations. The result is the 16 solutions expected for the 6R case. The same technique is also applicable to manipulators with prismatic joints. We present the solution technique for all six possible 5R,P manipulators through numerical examples. The primary distinction between the technique presented in this paper and recently published Raghavan and Roth (90a,b.c) solution is that they removed two known spurious imaginary roots of multiplicity four to obtain a 16th order polynomial for 6R and 5R,P cases. In our formulation, the 16th degree polynomial can be derived directly without having to remove any spurious imaginary roots. Another distinction is that the solution procedure presented in this paper can be reduced to an eigenvalue problem. This results in significant gains in computation time.

Abstract:
The Montney Formation is a highly productive hydrocarbon reservoir with significant reserves of hydrocarbon gases and liquids making it of great economic importance to Canada. However, high concentrations of hydrogen sulfide (H2S) have been encountered during exploration and development that have detrimental effects on environmental, health, and economics of production. H2S is a highly toxic and corrosive gas and therefore it is essential to understand the distribution of H2S within the basin in order to enhance identification of areas with a high risk of encountering elevated H2S concentrations in order to mitigate against potential negative impacts. Gas composition data from Montney wells is routinely collected by operators for submission to provincial regulators and is publicly available. We have combined data from Alberta (AB) and British Columbia (BC) to create a basin-wide database of Montney H2S concentrations. We then used an iterative quality control and quality assurance process to produce a dataset that best represents gas composition in reservoir fluids. This included: 1) designating gas source formation based on directional surveys using a newly developed basin-wide 3D model incorporating AGS's Montney model of Alberta with a model in BC, which removes errors associated with reported formations; 2) removed injection and disposal wells; 3) assessed wells with the 50 highest H2S concentrations to determine if gas composition data is accurate and reflective of reservoir fluid chemistry; and 4) evaluated spatially isolated extreme values to ensure data accuracy and prevent isolated highs from negatively impacting data interpolation. The resulting dataset was then used to calculate statistics for each x, y location to input into the interpolation process. Three interpolations were constructed based on the associated phase classification: H2S in gas, H2S in liquid (C7+), and aqueous H2S. We used Empirical Bayesian Kriging interpolation to generate H2S distribution maps along with a series of model uncertainty maps. These interpolations illustrate that H2S is heterogeneously distributed across the Montney basin. In general, higher concentrations are found in AB compared with BC with the highest concentrations in the Grande Prairie region along with several other isolated region in the southeastern portion of the basin. The interpolations of H2S associated with different phases show broad similarities. Future mapping research will focus on subdividing intra-Montney sub-members plus under- and overlying strata to further our understanding of the role migration plays in H2S distribution within the Montney basin.

The objectives of this project were to: 1) Identify genetic markers linked to sex-determining genes in various experimental and commercial stocks of O. niloticusand O. aureus, as well as red tilapias; 2) Develop additional markers tightly linked to these sex determiners, and develop practical, non-destructive genetic tests for identifying genotypic sex in young tilapia; A third aim, to map sex modifier loci, was removed during budget negotiations at the start of the project. Background to the topic. A major obstacle to profitable farming of tilapia is the tendency of females to reproduce at a small size during the production cycle, diverting feed and other resources to a large population of small, unmarketable fish. Several approaches for producing all-male fingerlings have been tried, including interspecific hybridization, hormonal masculinization, and the use of YY-supermale broodstock. Each method has disadvantages that could be overcome with a better understanding of the genetic basis of sex determination in tilapia. The lack of sex-linked markers has been a major impediment in research and development of efficient monosex populations for tilapia culture. Major conclusions, solutions, achievements. We identified DNA markers linked to sex determining genes in six closely related species of tilapiine fishes. The mode of sex determination differed among species. In Oreochromis karongaeand Tilapia mariaethe sex-determining locus is on linkage group (LG) 3 and the female is heterogametic (WZ-ZZ system). In O. niloticusand T. zilliithe sex-determining locus is on LG1 and the male is heterogametic (XX-XY system). We have nearly identified the series of BAC clones that completely span the region. A more complex pattern was observed in O. aureus and O. mossambicus, in which markers on both LG1 and LG3 were associated with sex. We found evidence for sex-linked lethal effects on LG1, as well as interactions between loci in the two linkage groups. Comparison of genetic and physical maps demonstrated a broad region of recombination suppression harboring the sex-determining locus on LG3. We also mapped 29 genes that are considered putative regulators of sex determination. Amhand Dmrta2 mapped to separate QTL for sex determination on LG23. The other 27 genes mapped to various linkage groups, but none of them mapped to QTL for sex determination, so they were excluded as candidates for sex determination in these tilapia species. Implications, both scientific and agricultural. Phylogenetic analysis suggests that at least two transitions in the mode of sex determination have occurred in the evolution of tilapia species. This variation makes tilapias an excellent model system for studying the evolution of sex chromosomes in vertebrates. The genetic markers we have identified on LG1 in O. niloticusaccurately diagnose the phenotypic sex and are being used to develop monosex populations of tilapia, and eliminate the tedious steps of progeny testing to verify the genetic sex of broodstock animals.
