Abstract Background: Lipophilic estradiol derivatives carried by lipoprotein particles in blood may mediate antioxidant or endocrine effects. We developed a new quantitative method to determine the concentration of circulating lipophilic estradiol fatty acid esters in human early- and late-pregnancy serum and in ovarian follicular fluid. Methods: After extraction from serum or follicular fluid, estradiol fatty acid esters were separated from nonesterified estradiol by Sephadex LH-20 column chromatography. Abstract Context: The 17β-estradiol fatty acid esters are hormone derivatives with long-lasting estrogenic effect. They are transported in serum lipoproteins and thought to be sequestered in adipose tissue. Objective: Our objective was to determine the 17β-estradiol fatty acid ester concentrations in serum and adipose tissue in women of various hormonal states. Design: After several chromatographic steps separating esterified from free estradiol, time-resolved fluoroimmunoassay was used as a quantifying tool. Chronic exposure to cadmium caused significant changes both at the testosterone and estradiol levels. Exposure to cadmium resulted in lesser serum testosterone concentrations and more estradiol in 0.05 and 0.1 ppm. Although not statistically significant, frogs exposed to 0.2 ppm had smaller drop in testosterone and significant more estradiol. The results suggest that cadmium exhibited the non-monotonic dose-response relation

This effect is not explained by current knowledge of ketoconazole's mechanism of action. Ketoconazole's effect on estradiol's binding to either receptors in target tissues (target-cell receptors) or serum-estradiol-carrying proteins in the aforementioned rats was investigated. Male rat mating is a suite of individual behaviors mediated by the actions of two metabolites of testosterone (T), dihydrotestosterone (DHT) and estradiol (E2), on the brain. Individually, neither metabolite fully maintains or restores mating in castrated males, but both combined are as effective as T. Two hormone-responsive areas of the brain, the medial preoptic area (MPO) and the medial amygdala (MEA), are crucial for mating. These studies ask: by what mechanism(s) does E2 act in the MPO and MEA? We blocked the conversion of T to E2 in the MEA of intact male rats and sexual behavior was not
