Observations from recent earthquakes have emphasised the need for a better understanding of the effects of structure-footing-soil interaction on the response of structures. In order to incorporate the influences of soil, a laminar box can be used to contain the soil during experiments. The laminar box simulates field boundary conditions by allowing the soil to shear during shake table tests. An empirical model was developed through response surface methodology to describe the relationship between tested variable (peptone, glucose, soyabeanmeal and pH). Maximum enzyme activity was attained with Peptone at 4 g∕L; temperature at 30 °C glucose at 6 g∕L; 30 °C and pH at 10. We investigate experimentally the turbulent flow through a two-dimensional contraction. Using a water tunnel with an active grid we generate turbulence at Taylor microscale Reynolds number Reλ ~ 250 which is advected through a 2.5:1 contraction. Volumetric and time-resolved Tomo-PIV and Shake-The-Box velocity measurements are used to characterize the evolution of coherent vortical structures at three streamwise locations upstream of, and within the contraction.
