Abstract:
Abstract Cold is a very common physical risk factor in workplaces in circumpolar regions. Cold has many detrimental effects on human health and performance, and on the safety, quality and productivity of work. In this study a systematic general Cold Risk Management Model was developed, applied and evaluated. The model can be integrated in a company’s or an organization’s occupational safety, health, environment and quality (SHEQ) management systems and practices in workplaces. The Cold Risk Management Model and methods were later included in ISO 15743 Ergonomics of the thermal environment – Cold workplaces – Risk assessment and management. The Cold Risk Management Model and methods were applied in two case company’s SHEQ systems and practices in the fields of construction and maritime administration and services. Based on the case studies, the concrete cold risk management activities and the personnel training campaign resulted in immediate positive results and improved attitudes towards further development. At the national level, working in the cold was estimated to increase personnel costs in the construction industry annually by €50M, which is 3% of the industry’s annual personnel costs. This study also showed that the Cold Risk Management Model and methods are profitable. In the case construction company, the savings achieved by cold risk management activities at a construction site were 2.5 time the costs of those activities. A follow-up study in the case company in the field of maritime administration and services showed that implementation and dissemination of the Cold Risk Management Model and methods require systematic work also after the initial development process. The implementation process could and should be enhanced by early establishment of organization-wide guidelines, visible concrete actions, a training campaign and use of necessary external experts. This study also presents a Safety Management Matrix Model for analyzing development and implementation activities during the process time span
Tiivistelmä Kylmä on yksi yleisimmistä työympäristön riskitekijöistä pohjoisissa oloissamme.

Abstract:
Transaction costs, information costs and defaults costs are suspected to partially explain differences in returns which were previously attributed to risk premiums in the financial markets. Two portfolios with identical costs are constructed from a Put, a Call, and underlying S&P 500 stocks, with the first Portfolio being hedged (Put-Call Parity) and the second Portfolio being unhedged (with systematic beta close to 2). The expected stock prices of S&P 500 were calculated based on 52-year historical data using several methods, and returns of the two portfolios were obtained and compared, using 935 observations of S&P 500 from January 2, 1992 through June 30, 1992. A linear regression model adjusted for cross-sectional heteroskedasticity and auto-correlation was used to estimate the expected risk premium rate. Transaction, information and default costs were statistically significant and estimated at 0.6% of value annually. These costs reduce the risk premium estimated by the Capital Asset Pricing Model.

Abstract:
A b s t r a c t. Shea nut butter was extracted from shea nut by cold press method and was investigated as feedstock for the production of biodiesel. Biodiesel yield was used to verify the optimization, while density and viscosity were chosen to serve as an indicator for the effectiveness and completeness of the ester conversion process. Based on the amount of shea butter used, the final product yield obtained was 94.55% mass and the percentage conversion of FFA in shea butter to biodiesel was 92.3% using a methanol/oil ratio of 6:1 and 1.0% mass KOH at 60 min and 55°C, respectively. The important properties of the biodiesel (density, kinematic viscosity, cloud point, pour point, cetane number, neutralization number, iodine value, methyl ester content and high heating value) were compared to those of ASTM and EN standards for biodiesel. The comparison shows that the shea butter methyl ester could be used as an alternative to diesel.

Abstract:
Traditional media has a long history of marginalising women and ethnic minorities by trivialising their contributions and obscuring their involvement. Ethnic minorities have been kept out of top roles in films – as evidenced in 2016, with the #Oscarssowhite controversy, when not a single nomination was extended to a person of colour. Women, too, have been objectified and typecast in supporting and “love interest” roles. This marginalisation, whereby male and white are the norm and everyone else is “the other”, extends far beyond movies. YouTube has transformed from its humble beginnings of a video sharing site into one of the main video-based services, capable of extending producers’ voices across national boundaries. With this change, amateur contributors have professionalised their productions. Analysing four top-rated guitar review YouTube videos using critical discourse analysis, this thesis explores the overrepresentation of white men in online spaces for the reviewing of music instruments and looks specifically at subtle ways in which discourse is used to reinforce social exclusions online. Abstract:
The images included in my thesis work reflect my experience growing up with military propaganda—pictures of cheerful white women in pearls as part of my rural middle American landscape. I do not name the oppressor because I am not here to pick at the thorns, but to get to the root of the oppression. These are some of the servicewomen I’ve met. Their stories parallel but cannot encompass the private experiences of all service women. I am grateful for their generosity; without them there would be no pictures. The battle for equality is much older than Rosie the Riveter but we still ask the same questions we asked Joan of Arc in the 15th century: if she isn’t working miracles, what is she doing on the battlefield?

Abstract:
Articular cartilage injuries on the knee are frequent pathologies and usually caused by trauma. Although some are asymptomatic, these changes may evolve into osteochondral articular cartilage degeneration and osteoarthritis. A progressive degeneration of articular cartilage leads to osteoarthritis (OA), being femorotibiopatelar joint the most affected one. As conventional treatments for OA are not curative, but palliative, cell therapy with stem cells (SC) stands out as an alternative. In this work we used stem cells from human exfoliated deciduous teeth (SHED), grown in association with a biomaterial, to treat osteochondral lesions on sheep\'s knees. Four osteochondral defects were surgically produced to test four conditions: 1) SHED associated with the biomaterial, 2) only the biomaterial, 3) only SHED and 4) no treatment (control). The sheep were followed clinically and through radiographic exams, US and arthroscopy, for 120, 190, 365 and 730 days until euthanasia. The results showed that treatment with biomaterial in combination with SHED produced repair similar to the cartilage structure from 190 days, showing cartilage adjacent features. However, despite the positive results in relation to the repair of osteochondral lesion it was not possible to say whether the improvement was due to direct differentiation of SHED for cells present in cartilage or was a paracrine effect. In time, there was no rejection and no adverse reaction to the xenogenic cell implant during the trial period.

Abstract:
Stem cells (SC) are cells that present self-renewal properties, capable of generating cells types and reconstitute several tissue types. For this reason, they have been seen as a therapeutic alternative to the treatment of many diseases which conventional treatment is not efficient. Besides, because of its plasticity, SC may be used to produce in vitro models in order to study diseases, what is interesting to diseases that affect the nervous system. Human SC from SHED (Stem cell from Human Exfoliated Deciduous teeth, SHED) are described in some papers as being capable of self-differentiation in neuronal cells through the direct transdifferentiation, a simple process that makes possible the differentiation of a cell type in another through the use of chemical agents. Nevertheless, SHED differentiation is refuted as being a artifact of culture due to cytotoxic action of these agents. Based on this premise, the aim of this study was to carry out protocols in order to produce neurons from SHED, using modifications and methods of neuronal differentiation described in the literature and used in the neuronal differentiation. During one of the protocols used, it was possible to verify the morphology of the cells as neuron-like, however, after some days of maintenance, they turn back to their original fusiform format. In this study, we can state, according to the obtained results, that SHED do not generate functional neurons

Abstract:
Everyone has to take a greater responsibility in energy issues, both individuals and companies. There have been some major developments in the construction industry but there is still a lot to be done.   This diploma work thesis presents different methods of making existing construction shed establishments to use less energy. What is possible to do and how much energy is there to be saved? A shed establishment consisting of 8 shed units were studied in this project and the building simulation tool VIP-Energy was used to simulate different types of material in the sheds. An infrared camera was used to locate thermal bridges.   There are several factors that can be improved to make a construction shed establishment to use less energy. Replacement of old windows and doors, using more insulation, replacement of old less efficient insulation with new improved materials, using a different heating system, and more. One factor, which is more important than all the others, is to insulate between the 8 different shed units in the establishment. It is also important to make the construction wind proof.   The results show that if you replace the old wall and roof insulation with new better insulation, replace the old windows and doors and seal the gaps between the shed units the total energy reduction would be as high as 60%. All companies working with the handling or utilization of shed establishments at building sites should consider these improvements.

Abstract:
This thesis examines the importance of the musical score in creating a film's meaning, through an analysis of the musical scores to several recent Hollywood movies. To show how film music is ideologically loaded, this thesis takes the case of the new, ultra-violent film (anti)heroine and demonstrates how the musical score works to explain, excuse or undermine her violence so that she can remain situated within the standard gender codes of Hollywood film. To do this, the theoretical section first discusses issues about the representation of femininity and the pleasures of movie violence, and then continues with a discussion of theories of musical meaning, both within and without film scores. The analysis section examines the scores to several well-known mainstream Hollywood movies, dividing the heroines into either action heroines or fatal femmes, to show firstly how these women potentially transgress gender codes, and how the music is employed - along with other filmic elements - to lessen the threat of the female character's transgression. It examines the presentation of women as mothers, victims, lesbians and jezebels - all of which roles act to frame the active heroine within some acknowledged stereotype. Finally, the thesis addresses the question of how specific this phenomenon is to Hollywood, by examining the scores to a test-case French film and its Hollywood remake. Through the analyses contained within this thesis, I show how violent Hollywood women are still subject to ideologies which position women as passive and specifically as non-violent, and that the musical score is a particularly effective means of neutralising their potential transgression of gender norms.

Abstract:
Data structured in the Resource Description Framework (RDF) are increasingly available in large volumes. SPARQL is the widely used and well supported standard query language for RDF data. In parallel to query language evolutions, schema languages for expressing constraints on RDF datasets also evolve. Shape Expressions (ShEx) are increasingly used to validate RDF data, and to communicate expected graph patterns. Schemas in general are important for static analysis tasks such as query optimisation and containment. Our purpose is to investigate the means and methodologies for SPARQL query static analysis and optimisation in the presence of ShEx schema constraints. Our contribution is mainly divided into two parts. In the first part we consider the problem of SPARQL query containment in the presence of ShEx constraints. We propose a sound and complete procedure for the problem of containment with ShEx, considering several SPARQL fragments. Particularly our procedure considers OPTIONAL query patterns, that turns out to be an important feature to be studied with schemas. We provide complexity bounds for the containment problem with respect to the language fragments considered. We also propose alternative method for SPARQL query containment with ShEx by reduction into First Order Logic satisfiability, which allows for considering SPARQL fragment extension in comparison to the first method. This is the first work addressing SPARQL query containment in the presence of ShEx constraints. In the second part of our contribution we propose an analysis method to optimise the evaluation of conjunctive SPARQL queries, on RDF graphs, by taking advantage of ShEx constraints. The optimisation is based on computing and assigning ranks to query triple patterns, dictating their order of execution. The presence of intermediate joins between the query triple patterns is the reason why ordering is important in increasing efficiency. We define a set of well-formed ShEx schemas, that possess interesting characteristics for SPARQL query optimisation. We then develop our optimisation method by exploiting information extracted from a ShEx schema. We finally report on evaluation results performed showing the advantages of applying our optimisation on the top of an existing state-of-the-art query evaluation system

The recent legitimization of Landscape Urbanism as a theory for architectural design may signal a growing cultural shift toward environmental custodianship. Design strategies that blur buildings and landscape have become architecture's new orthodox response to this trend, promising continuity between architecture and landscape on the new "green" urban surface. However, the infrastructural and organizational demands of this blurring of city and country may actually require an architecture that is more flexible over its lifespan -more appropriately accommodating the on-going bureaucratic alterations required to manage this utopian complexity. This thesis offers a modest proposal for big-box architecture, one capable of delivering continuity and flexibility for the city, even for programs that require functional separation. An exploration of this super-sized typology, the Mega-Shed demonstrates the timeless desire to manage the environment, while resisting the urge to re-create picturesque landscape. Whereas modern cities banished their support systems to the periphery, the Mega-Shed is a passive machine for the current age, an organizational strategy capable of bringing these support systems back into view, producing a sublime utility in the heart of the city.

Abstract:
Eskom Holdings SOC Limited (referred to as Eskom), a public state owned company, is required to continually demonstrate environmental duty of care and report on its environmental performance. In furthering its commitment to sustainable development and in line with environmental compliance of its activities, Eskom is planning to have the Distribution Division certified against the International Organisation for Standardization (ISO) 14001:2004 environmental management system (EMS) standard by 2014. In general, management review may quickly turn into a dry review of the items on the agenda, done only to show an auditor the review was held. As such, the aim of this study was to assess the effectiveness of management reviews as one of the key continual improvement elements of the ISO 14001:2004 environmental management system within Eskom Distribution: Western Region. This research has been designed to take place in three stages. These stages are planning, execution and publication, and preparing the dissertation for marking. The research methodology included a literature study on various national and international articles and journals on EMSs that relate to environmental performance. The databases of Eskom Distribution: Western Region ‒ i.e. the management review presentation results for the 2008/09, 2009/10 and 2010/11 financial years, key significant environmental aspects for the Technical Service Centres (TSCs) and Minor/Major Engineering Works (MEW), key performance indicators (KPIs), Risk Audit System (RAS) results, and results of the external EMS audit conducted from 23–25 April 2007 were used for this research. Questionnaires were sent to the TSCs’ senior supervisors and Safety, Health, Environment and Quality (SHEQ) management. An analysis of the questionnaires completed by the TSC senior supervisors showed that all the participants have seen an improvement in environmental performance due to annual management reviews, except TSC11, who have not seen management review results. It can be concluded from the results of the SHEQ management questionnaires that an improvement (continual improvement as per the ISO 14001 requirement) was observed. The improvements were identified through improved processes, improved gap analysis, increased environmental awareness, and addressing risks. Most of the ISO 14001:2004 requirements for the management review for all financial years covered in this study were met. In terms of the objectives, targets and KPIs, Eskom Distribution: Western Region performed effectively and most of the outcomes were implemented according to the recommendations. Further, based on the external audit conducted from 23 to 25 April 2007, it is clear that Eskom Distribution: Western Region conformed to the minimum requirements of the ISO 14001:2004 EMS. Their management reviews also met all requirements of the ISO 14001:2004 relating to management review. This was proven by minutes of previous meetings that covered all the necessary issues that were audited. Thus, the existence of management reviews contributed towards ensuring that the ISO system was scrutinised and improved where it worked, and that aspects that did not work well were removed.

This paper is a case study based on Beng Shan Ba She in the middle- north in Taiwan from Qing Dynasty to Japanese colonial period. In addition to the study on the history of rental situations in Pingpu Tribe, this paper tries to integrate the cadastre files in the beginning of Japanese colonial period all together via GIS so as to present the whole picture about Beng Shan Ba She and the private files of the rental situations. Further, in order to present the development process of Beng Shan Ba She’s rental situations, this paper tries to check and compare the related files about the land contracts between the aboriginal and Han people. This paper tries to point out the exact locations of the various groups and also shows the distribution and the rental sum of money. Meanwhile, till the beginning of Japanese colonial period, there were up to sixty percent of cultivated lands under the aboriginal’s rights. More than seventy percent of rental were still owned by the aboriginals. This discovery is different from Chih-ming Ka’s observation on the aboriginal’s land rights in Hsinchu but similar to John R. Shepherdi’s studies on the ones distributed among Shulin and ShanXie in Taipei. Different from Chih-ming Ka’s and John R. Shepherdi’s studies which are based on case studies and develop the theories like rational nation and politics of tribal group respectively, this purpose of this paper is to figure out the meaning of Beng Shan Ba She’s existence and their social autonomy. In order to understand the development process of Beng Shan Ba She, this paper divide the land rights of Beng Shan Ba She into different parts, namely mixed cultivated area between the aboriginal and Han, Han cultivated land and Ai Tun Areas so as to understand the development process of these three types of land rights. Among them, the mixed cultivated lands were mostly passed down from the aboriginal’s ancestors, granted by Qing government and group purchasing. And they teamed up to reserve the lands which are the precious and rare lands in Beng Shen Ba She nowadays. In Ai Tun Areas , the land rights were mainly based on Ai rental and Tun rental formed in Qianlong 25th year and end of Qianlong years. Ai Tun Areas were mainly cultivated under the ways of join stock but the lands were disappeared gradually since the tenants declined to pay the original landlords after the policy of land measurement by Liu Mingchuan in Guang Xu 13th year. Last, the land rights of Han cultivated lands were mainly affected by the policy of fixed aboriginal rental in Qianlong years. Among Han people, Cai Zhenfeng is the most powerful land right holders. He accumulated his properties by buying Ri Nan’s and Ri Bei’s lands and further carried out the rental business. To sum up, these three rental areas in Beng Shan Ba She completely reflected a variety of land rights. Basically, rental was treated as assets under the aboriginal rental policy in Beng Shen Ba She in the middle years of Qianlong and for aboriginal, organization staff , government staff and the capitalists of Han people, it carried different meanings for different positions. In fact, rental business not only proved crop for the aboriginal but also was the tax source for the local government. As for the organization staff like aboriginal heads and interpreters, involved in rental business management usually used rental as guarantee to borrow money from Han people. That is to say rental was treated as a medium to accumulate their fortunes and also working in the rental organization was the goal for people in and out of the aboriginal tribes. For the capitalists of Han, they could borrow money from the tribes and further practice the society duty and than embezzled the rental at last in the society even though they are not allowed to work in the organization by law. Therefore, the government , capitalists of Han, the aboriginal and the tribe staff as well competed with one another and further tried to seize the rental business. On the other hand, seeing from the rental sum of money recorded from Japanese colonial period, we can find that the reform of aboriginal affairs carried out by Liu Ming Chuan was a protection for the rental business which helped the tribes to maintain their social operation and their income. Last, this paper finds that both the private land rights and crops were mostly under the ones who had served as staff in the rental organization. When judging form the situation that most of the public rental lands spreaded near to the aboriginal tribes, we can infer that the situation may be affected by Da Jia event and reflected the management strategies among the various tribes. Last, even though Beng Shan Be She could get the compensation from Japanese government under the policy of rental right., land rental business had provided the aboriginal income and maintained the whole society’s operation for the past hundreds of years. Once the rental business is eliminated by the government, the economy of the aboriginal may face a great pressure and the social operation and organizations among the aboriginal tribes may be destroyed As a result, the aboriginal tribes were gradually assimilated by Han people.

Abarinov, Elena. "Progressive restriction of CNS cell-fate potential by the transiently expressed transcription factor Nkx2.2." Thesis, 2019. https://doi.org/10.7916/d8-shpq-2d83.

Full text
Abstract:
The progressive loss of developmental potential is a hallmark of all differentiating cells in multicellular organisms. At the chromatin level, this restriction in cell-fate plasticity is established through the silencing of active and poised lineage-specific genes that are incompatible with the terminal fate of the maturing cell type. The effective and stable inhibition of gene expression relies on the coordinated action of transcriptional repressors. These repressors are often transiently expressed only at the time of cell-fate specification and direct lineage decisions by suppressing alternative developmental programs. However, compared to the numerous studies examining the mechanisms by which cell-type specific transcriptional activators program cellular identity, little is currently known regarding how transient repressors execute permanent silencing of gene regulatory networks. To address this question, I have examined the mechanisms through which the transiently expressed transcription factor (TF) Nkx2.2 represses the acquisition of motor neuron (MN) identity in V3 neuronal progenitors. While it is well-established that Nkx2.2 functions as a transcriptional repressor through its interactions with the Groucho (Grg) family of co-repressors, how these interactions manifest in gene silencing has remained unknown. Moreover, the effects of Nkx2.2 occupancy on chromatin modifications have not been determined. In this dissertation, I demonstrate that surprisingly, Nkx2.2 decommissions enhancers of the MN developmental program not through the recruitment of additional co-repressor proteins but rather through the eviction of co-activator complexes. While this displacement is dependent upon an intact Grg-interacting domain, Nkx2.2 binding does not increase Grg enrichment. In addition, extensive profiling of Nkx2.2 genome-wide binding events in neural precursors unexpectedly revealed that Nkx2.2 occupies not only enhancers of MN progenitor genes acutely repressed by Nkx2.2 but also enhancers of genes expressed exclusively in postmitotic MNs, long after Nkx2.2 expression has been down- regulated. In vivo lineage tracing experiments and in vitro genomic analyses demonstrated that Nkx2.2 also functions in a repressive capacity at these poised regulatory regions. Here, Nkx2.2 binding prevents the activation of postmitotic genetic networks through a preferential enlistment of histone deacetylase complex 2 (HDAC2) proteins. However, this binding is not accompanied by the deposition of repressive chromatin modifications, and removal of Nkx2.2 in differentiating V3 neurons leads to the ectopic expression of the postmitotic MN TFs Isl1 and Hb9. Collectively, these studies indicate that transiently expressed repressors may establish gene suppression by counteracting the activities of transcriptional activators, rather than by directly establishing repressive chromatin signatures. As transcriptional reprogramming of differentiated cell linages often fails to adequately silence the expression programs of the starting population, these results may help to inform new methodologies for instructing cell conversions.

Abstract:
This project attempts to translate my experience of the everyday street for the viewer; to reveal my experience of walking and looking through drawing. The everyday is an elusive and slippery subject that has been used for various and sometimes radical agendas by many writers and artists in the last two centuries. It is seen as having potential for finding the authenticity that is lacking in other spheres of human activity. However, there is an inherent contradiction for the artist engaged with the everyday in that, by definition, the mundane is barely noticed yet it can swiftly become extraordinary when examined closely. This project uses a toolbox of everyday tactics, processes and methods to explore and devise appropriate modes of representing the everyday street, while taking this contradiction into account.

Abstract:
This investigation used a one-group pretest-posttest design to examine the relationship between a tier three reading intervention program (i.e., a program designed for students that have failed to respond to regular (tier one) and resource (tier two) supports), and grade-level reading gains, as measured using the Fountas and Pinnell Benchmark Assessment System (Fountas & Pinnell, 2010), for 140 students between grades two and four. All students included in this study began well-below their expected grade-level in reading (i.e., two or more grade-levels below). In order to describe the gains made in grade-level reading ability, the Fountas and Pinnell Benchmark Assessment System (Fountas & Pinnell, 2010) was used pretest and posttest to track reading gains. Results from descriptive statistics, Kruskal-Wallis H tests, Mann-Whitney U tests, t-tests, and a multiple regression were indicative of positive reading gains. Approximately 74% of participants achieved grade-level reading gains that had the potential to close the reading gap (i.e., 0.75 grade-levels or more as measured using the Fountas and Pinnell Benchmark Assessment System (Fountas & Pinnell, 2010)). These results were similar for all participants regardless of gender, diagnosis status (i.e., garden-variety poor reader, physical disability, and learning/attentional disability) or first language status (i.e., English as an additional language or English). This study provides preliminary evidence that this intervention is improving reading outcomes for tier three students. Moving forward, stakeholders of this division would be encouraged to develop a well-designed, structured mixed-methods research study with standardized assessment measures and planned follow-up.

(6984413), Tiwaladeoluwa Adekunle. ""Shea Moisture is Cancelled": Racialized Identification in the 2017 Shea Moisture Crisis." Thesis, 2019.

Find full text
Abstract:

In 2017, Shea Moisture, a company that created natural hair products targeted primarily toward Black women, released an online video in which ostensibly white and multiracial women discussed the struggles they encountered in accepting their natural hair. This video led to a public relations crisis for the organization as a result of its perceived exclusion of the organization’s core public--Black women with 4C hair, who arguably experience the highest levels of discrimination on account of their natural hair. This study explored the role of identity and identification in this crisis by conducting a qualitative content analysis of identification types in Black men and women’s online responses to the video. Emotions present in the online posts were used as rhetorical indicators of deidentification, ambivalent identification, identification or disidentification. The findings of this study, contextualized within the socio-political context of the crisis, suggest that responses to Shea Moisture’s video were informed by : its public’s identification with one another, their construal and co-construction of the organization’s identity as a Black business, and their identification with the organization on the basis of this identity. This study reinforces the role played by publics in co-constructing an organization’s identity and reveals the importance of sociopolitical realities and uneven power relations to publics’ identification. This study also introduces the concept of “protected identification” to describe a mode of identification that informed by a socio-political context wherein marginalization exists, comes with a unique set of expectations for the actions of an organization.
