Abstract Background: Homologous recombination deficiency (HRD) affects cancer types including breast, ovarian, pancreas and prostate and patients with HR deficient tumors can respond to poly ADP-ribose polymerase (PARP) inhibitors and platinum-based therapies. HRD is associated to the loss of either germline or somatic, BRCA1 or BRCA2 and other associated variants. However, a large proportion of tumors do not have an identifiable variant in BRCA1 or BRCA2 and other related genes. Patterns of mutations—Mutational Signatures—of HRD can identify changes in DNA sequences due to defective DNA repai

Abstract BCL6 is a preclinically validated oncogenic driver of DLBCL historically considered to be undruggable. ARV-393, a PROteolysis TArgeting Chimera (PROTAC) degrader, directly binds an E3 ubiquitin ligase and BCL6 to induce ubiquitination of BCL6 and its subsequent proteasomal degradation. ARV-393’s iterative activity overcomes rapid resynthesis of BCL6 resulting in potent, single-agent activity and tumor regressions in many cell- and patient-derived xenograft (CDX and PDX) DLBCL models. ARV-393 monotherapy is being evaluated in a phase 1 trial (NCT06393738) in patients with non-Hodgkin l

Abstract Background Breast-conserving surgery is a critical step in treatment with the goal of removing all cancer cells while minimizing the removal of healthy tissue. 15% to 25% of lumpectomy patients have positive margins and require a second surgery to achieve negative margins and reduce the risk of local recurrences. These positive margins are poorly predictive (35% PPV) of cancer left in the cavity, so most second surgeries find no residual cancer. Better detection tools are needed to guide in real-time the removal of cancer missed during the initial lumpectomy to reduce the number of se
