Abstract:
In China, economic anthropology started relatively later compared with that in the West; however, under the guidance of Marxist theory and based on the needs of China's economic development, it gradually formed unique advantages in its development process. In China's economic and social development practice, Fei Xiaotong and Shi Zhengyi, among other pioneers, creatively applied the economic anthropological paradigm to solve the specific issues, starting from the local characteristics, and completed some excellent academic results. This paper probes and analyzes the scholarly contributions of the pioneers of Chinese economic anthropology; especially, it takes Marxist economics as the starting point to examine Professor Shi Zhengyi's theoretical contributions to the development of Chinese economic anthropology and its practical significance.

AbstractThe article presents the findings of a survey of the imprecation phrase ma yi fei shi 麻夷非是 and its variations, as written (using brush and ink) on several thousand excavated covenant texts (mengshu 盟書) from Houma 侯馬 and Wenxian 溫縣. I argue that the findings support Zhu Dexi 朱德熙 and Qiu Xigui 裘錫圭's analysis of the phrase as mi yi bi shi 靡夷彼氏 “Wipe out that shi” (shi, I suggest, referring to the covenantor and his direct male descendants). Through comparison of scribal hands, I demonstrate that those variations which do not fit this analysis were produced by a small number of scribes and, in almost all cases, can be shown to be errors. I conclude that such variations are generally unreliable and do not require us to reject Zhu and Qiu's analysis. These examples suggest that formulaic, possibly archaic, stock phrases, such as this imprecation, were liable to be misinterpreted, even during the period in which they were in use. Identification of scribal hands and scribal errors was essential to this analysis, demonstrating not only the importance of this methodology in such research, but also the potential value of these particular materials for furthering our understanding of scribal habits and text reproduction in early China.

Abstract:
Purpose: Prescriptions of Han-Shi-Yu-Fei (HSYF), Han-Shi-Zu-Fei (HSZF), and Yi-Du-Bi-Fei (YDBF) were effective in treating COVID-19. Based on network pharmacology and molecular docking, overlapping Traditional Chinese medicines (TCMs), their active components, and core targets were explored in this study. Methods: First, the overlapping TCMs and their active components were collected from the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP) by evaluating Oral Bioactivity (OB) and Drug Likeness (DL). The overlapping targets of potential components and COVID-19 were collected by SwissTargetPrediction, Gene Cards, and Venn 2.1.0 databases. Gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment were analyzed via DAVID6.8.1 database. Through comprehensive analysis of the “prescriptions-TCMs-components” (P-T-C), “components-targets-pathways” (C-T-P) and “protein–protein interaction” (PPI) networks constructed by Cytoscape 3.7.1 software, the active components and core targets were obtained. Finally, the binding energies of these components with ACE2 and SARS-CoV-2 3CL were analyzed by AutDockTools-1.5.6 and PyMOL software. Results: In all, five overlapping TCMs, 40 potential active components, and 47 candidate targets were obtained and analyzed in these prescriptions. There were 288 GO entries ( P < 0.05), including 211 biological process (BP), 40 cell composition (CC), and 37 molecular function (MF) entries. Most of the 105 KEGG pathways ( P < 0.05) were involved with viral infection and inflammation. Through “PPI” and “C-T-P” networks, the core targets (EGFR, PTGS2, CDK2, GSK3B, PIK3R1, and MAPK3) and active components (Q27134551, acanthoside B, neohesperidin, and irisolidone) with high degrees were obtained. Molecular docking results showed that the above-mentioned four components could inhibit the binding of ACE2 and SARS-COV-2 3CL to protect against COVID-19. Conclusion: In this study, the active components and core targets of three prescriptions in the treatment of COVID-19 were elaborated by network pharmacology and molecular docking, providing a reference for their applications.

By establishing a fuzzy mathematical evaluation model based on physical and chemical indicators, this paper investigates the genetic toxic pollution of Chaohu Lake in 2016 through the micronucleus technology of the Vicia faba root tip and evaluates the water quality of Chaohu Lake. The results show that, in recent years, the overall water quality of the lakes around Chaohu has improved slightly, but the pollution levels of some areas (such as Nan Fei He and Shi Wu Li He) are still serious. The results can also provide a more reliable theoretical basis for the planning and management of environment protection.

The Hainan Pygmy Toad Parapelophryne scalpta (Liu & Hu) is the single species of the genus Parapelophryne Fei, Ye & Jiang in the Family Bufonidae (Frost 2020) and is endemic to China’s Hainan Island, which lies within the tropics at 18°09′–20°10′N, 108°37′–111°03′E, with a land area of ca. 33,900 km2. The taxonomy status of this species has obtained much debates since its discovery: it was originally described by Liu et al. (1973) as a member of the genus Nectophryne Buchholz & Peters then allocated to Pelophryne Barbour by Ye & Fei (1978). On the basis of its unique morphological characters, Fei et al. (2003) erected a new genus Parapelophryne to discriminate P. scalpta from other bufonids. Some researchers, however, still listed the species under the genus Pelophryne (Stuart et al. 2008; Shi et al. 2011). Recently, the validity of the genus Parapelophryne is further confirmed by Matsui et al. (2015) on the basis of molecular analyses. Interestingly enough, Matsui et al. (2015)’s result showed that this extremely small-sized toad is a sister taxon of the large-sized toads of the genus Bufo Garsault, which is mainly distributed in temperate and subtropical regions of Eurasia (Frost 2020). Parapelophryne scalpta is a forest-dependent species and only inhabits in well-preserved evergreen broadleaf forests (Fei & Ye 2016). It is the smallest toad species in China with snout-vent length of 19–23 mm in adult males and 24–27 mm in females (Fei & Ye 2016). Due to their small size and elusive habits, little is known about the natural history of P. scalpta and its acoustic characteristics are still undocumented (Fei & Ye 2016). During a herpetological survey in Hainan in 2015, we luckily detected male calls of P. scalpta and obtained a short but clear record of the advertisement call of the species. Herein, we describe the acoustic characteristics and calling behavior of Parapelophryne scalpta.

Leptobrachella (Anura: Megophryidae) is currently composed of seventy-one species distributed in China and Southeast Asia (Frost 2018). Fourteen species of Leptobrachella are currently known from southern China: L. alpina (Fei, Ye & Li, 1990), L. laui (Sung, Yang & Wang, 2014), L. liui (Fei & Ye, 1990), L. mangshanensis (Hou, Zhang, Hu, Li, Shi, Chen, Mo & Wang, 2018), L. maoershanensis (Yuan, Sun, Chen, Rowley & Che, 2017), L. oshanensis (Liu, 1950), L. purpura (Yang, Zeng & Wang, 2018), L. tengchongensis (Yang, Wang, Chen & Rao, 2016), L. wuhuangmontis (Wang, Yang & Wang, 2018), L. yingjiangensis (Yang, Zeng & Wang, 2018), L. yunkaiensis (Wang, Li, Lyu & Wang, 2018), L. ventripunctata (Fei, Ye & Li, 1990), L. pelodytoides (Boulenger, 1893) and L. sungi (Lathrop, Murphy, Orlov & Ho, 1998) and the first eleven species are endemic of this region (AmphibiaChina 2018). Many new species in this genus were discovered based on molecular, morphometric and bioacoustic data in recent years (Frost 2018). In comparison to molecular and morphological data, bioacoustics is especially useful in species identification of Leptobrachella in field surveys (Rowley et al. 2016). Although the advertisement calls of many Leptobrachella species were described along with the species descriptions (e.g., Rowley et al. 2016), the advertisement calls of only three Chinese endemic species (L. alpinus; L. laui and L. oshanensis) are known (Jiang et al. 2002; Matsui 2006; Xu et al. 2005). Here, we describe the advertisement calls of L. liui, which inhabits rocky streams in southeastern China (Fujian, Jiangxi and Zhejinag) and is morphologically diagnosed by the combination of a set of external color traits (Fig. 1).

Abstract Objective To realize the characteristics of “zheng” differentiation-treatment for hand, foot and mouth disease (HFMD), a new methodology of syndrome differentiation for different stages of HFMD has been explored. Methods Total of 2 325 cases with HFMD were recorded by distributing them into exterior syndrome stage, interior syndrome stage, severe syndrome stage and recovered syndrome stage, respectively, and the main symptoms and subsidiary symptoms of different stages of HFMD have been observed. The major and minor pathogenesis of HFMD in different stages were obtained, and compared with the “2010 Guideline for the Diagnosis and Treatment of HFMD”. Results It was found that the major pathogenesis of exterior stage was defined as “the invation of the wenevil to the defender of the body with the collaterals got involved”, and the minor as “qi deficiency”; in interior stage, “the fury of Gan-Yang” was the main pathogenesis, and “qi in chaos and qi deficiency” was the minor; in severe syndrome stage, “the damage of heart, liver and lung” was the main pathogenesis, and “qi in chaos” was the minor; and the pathogenesis of recovered stage was “qi-yin deficiency”. Compared with the “2010 Guideline for the Diagnosis and Treatment of HFMD”, it showed that “the obstruction of the fei-pi qi by the mixture of shi-re evil” and “the mixture of shi-re” in vivo was quite difficult to be explained in completely different context in the general situation; in the severe stage, the TCM clinical characteristics of syndrome differentiation might lose; in the early acute severe cases, the phenomenon that xin-yang and fei-qi almost ran out was difficult to be observed, then, the line between the severe and the acute severe became vague. Conclusions The theory of syndrome differentiation by stages of HFMD was reasonable in the actual situation of clinical description on HFMD which was expected to be further tested and widely applied in the “zheng” differentiation-treatment of HFMD in the future.

Aristolochic acid nephropathy is caused by aristolochic acid (AA) and AA-containing herbs. In traditional Chinese medicine, a principle called “Jun-Chen-Zou-Shi” may be utilized to construct a remedial herbal formula that attempts to mitigate the toxicity of the main ingredient. This study usedBu-Fei-A-Jiao-Tang(BFAJT) to test if the compound remedy based on a principle of “Jun-Chen-Zou-Shi” can decrease the toxicity of AA-containing herbs. We compared the three toxicities of AA standard,Madouling(anAristolochiaherb), and a herbal formula BFAJT. AA standard was given for BALB/c mice at a dose of 5 mg/kg bw/day or 7.5 mg/kg bw/day for 10 days.Madoulingand BFAJT were given at an equivalence of AA 0.5 mg/kg bw/day for 21 days. Nephrotoxicity was evaluated by metabolomics and histopathology. The urinary metabolomics profiles were characterized by1H NMR spectroscopy. The spectral data was analyzed with partial least squares discriminant analysis, and the significant differential metabolites between groups were identified. The result showed different degrees of acute renal tubular injuries, and metabolomics analysis found that the kidney injuries were focused in proximal renal tubules. Both metabolomics and pathological studies revealed that AA standard,Madouling, and BFAJT were all nephrotoxicants. The compositions of the compound remedy did not diminish the nephrotoxicity caused by AA.

This study aimed to investigate the distribution and characteristics of traditional Chinese medicine (TCM) syndrome and its elements on respiratory diseases (RDs) based on real-world data (RWD). Methods. A real-world study was performed to explore the relationships among TCM syndrome and RDs based on electronic medical information. A total of 26,074 medical records with complete data were available for data analysis. Factor analyses were used to reduce dimensions of TCM syndrome elements and detect common factors. Additionally, cluster analyses were employed to assess combinations of TCM syndrome elements. Finally, association rule analyses were performed to investigate the structures of TCM syndrome elements to estimate the patterns of TCM syndrome. Results. A total of 27 TCM syndromes were extracted from RWD in this work. There were four TCM syndromes with >5.0% frequency based on the distribution frequency. The top five pathogenesis TCM syndrome elements were Tan, Huo, Feng, Qi_Xu, and Han. Factor analysis, cluster analysis, and association rule analysis demonstrated that Tan, Huo, Feng, Qi_Xu, Shen, and Fei were the core TCM syndrome elements. Conclusion. Four common Shi TCM syndromes on RDs were identified: Tan_Re_Yong_Fei, Tan_Zhuo_Zu_Fei, Feng_Re_Fan_Fei, and Feng_Han_Xi_Fei; two core common Xu TCM syndromes (Fei_Shen_Qi_Xu and Fei_Yin_Xu) and two core common Mix TCM syndromes (Fei_Pi_Qi_Xu-Tan_Shi_Yun_Fei and Fei_Shen_Qi_Xu-Tan_Yu_Zu_Fei) were also determined. The core TCM syndrome elements of Tan, Huo, Feng, Qi_Xu, Shen, and Fei were identified in this work.

Factor analyses, cluster analyses, and association rule analyses demonstrated that Tan, Huo, Feng, Han, Qi-Xu, Yin-Xu, Fei, and Shen were the core TCM syndrome elements. Conclusion: The four common Shi TCM syndromes of Tan_Zhuo_Zu_Fei, Tan_Re_Yong_Fei, Feng_Han_Xi_Fei, and Feng_Re_Fan_Fei for CB were detected in the real world study, and the two Xu TCM syndromes of Fei_Yin_Xu and Fei_Shen_Qi_Xu were identified. Abstract:
Abstract Background While whole breast radiotherapy has been standard of care for invasive breast cancer patients treated with breast conserving surgery, not all women may benefit from radiotherapy. Recently, we demonstrated that a 16-gene signature named Profile for the Omission of Local Adjuvant Radiotherapy (POLAR) could identify breast cancer patients with HR+, HER2- tumors treated with breast conserving surgery alone with a 10-year locoregional recurrence risk of less than 10%. Methods In this study, we apply the signature to patients enrolled in the Princess Margaret Trial, a randomized trial where patients age 50 years or older were randomized to radiotherapy and tamoxifen or tamoxifen alone after breast conserving surgery. Tissue from 132 patients with HR+, HER2- tumors were available for gene expression analysis. Results For women treated with tamoxifen alone after breast conserving surgery, POLAR identified low risk women with a 7% risk of locoregional recurrence at 10 years. Comparison to POLAR-low patients treated with adjuvant radiotherapy did not demonstrate a significant benefit from radiotherapy (HR=1.5[0.14-16], p=0.74). POLAR-high patients not treated with radiotherapy had a 22% risk of locoregional recurrence at 10 years. Comparison to POLAR-high patients treated with adjuvant radiotherapy demonstrated a significant benefit from standard radiotherapy (HR=0.25[0.07-0.92], p=0.038). Conclusions These data suggest that the POLAR genomic signature may be used to identify patients with a low risk of locoregional recurrence without significant benefit from adjuvant radiotherapy. Patients with low POLAR scores may potentially be candidates for radiation therapy omission when treated with breast conserving surgery and tamoxifen. Citation Format: Anthony Fyles, S. Abstract Tumor-infiltrating lymphocytes (TILs) has demonstrated significant clinical benefits for patients with solid tumor. A robust expansion process is needed in order to produce a large number of cells for clinical usage. We modulated the PI3K-AKT signaling pathway in TILs isolated from cervical and ovarian cancer patients and found the inhibition of either PI3K or AKT can promote TIL proliferation during the REP stage. Inhibiting PI3K-AKT led to an increase of the effector CD8+T cell population with higher CD25+ and CD28+ in the CD8+ population, more TCF-1+ and CD39-CD69- T cells in both CD4+ and CD8+ T cell population, and downregulated exhaustion markers CD39+ and TIM-3+ in CD8+ T cell population. This inhibition significantly increased cytotoxicity when cocultured with tumor cell lines and patient-derived tumor samples. Moreover, dual knockout of ATK1 and ATK2 promoted TIL proliferation, long-term survival and decreased apoptosis of TIL, which were further confirmed by the RNA-seq results. Thus, inhibition of PI3K-AKT signaling represents a promising strategy to generate large numbers of TIL with enhanced stemness, activation and cytotoxicity, and could be used to support adoptive cell therapy to treat solid tumors. Background: Dyslipidemia is an abnormal lipid metabolism characterized by an increase in total cholesterol, LDL (Low-Density Lipoprotein), triglycerides (TG), and a decrease in HDL (High-Density Lipoprotein). Increased levels of total cholesterol, TG, LDL, and decreased HDL levels can increase the risk of cardiovascular disease. According to TCM, dyslipidemia is categorized into “Tan Zheng” (phlegm syndrome), “Shi Zuo” (damp retention), “Xuan Yun” (vertigo), and “Fei Pang” (obesity). Purpose: The purpose of this review is to analyze the literature study on TCM within the treatment of dyslipidemia with different acupuncture methods and points. The primary data search was conducted using online e-resources (Google Scholar, Proquest, and Pubmed). Review: The results of the study should include at least one of the outcomes indicators for total cholesterol, TG, LDL, and HDL. The result there were 20 journals consisting of studies that used several acupuncture methods, manual acupuncture (AM), electroacupuncture (EA), moxa (MOK), auricular acupuncture (AA), Cupping (CUP), and found additional interventions such as nutritional interventions, and exercise; body points often used are Zusanli (ST 36), Fenglong (ST 40), Quchi (LI 11), Daheng (SP15), Zhongwan (CV 12), Qihai (CV 6), Guanyuan (CV 4), Sanyinjiao (SP 6), Shenque (CV 8), Tianshu (ST 25), and therefore for AA are Spleen (CO 13), Stomach (CO 4), Shenmen (TF 4), and hunger point. Twenty journals showed a decrease in total cholesterol, TG, LDL levels, and an increase in HDL levels. Conclusion: In conclusion, all types of acupuncture methods can reduce total cholesterol, TG and LDL levels, and increase HDL levels.

With the increase of portable power sources demand, new technologies, e.g. wearable and flexible electronics, are projected to generate $1.25 billion market by 2022.[1] New storage energy devices are more than ever in demand which requires new specifications in order to be used in those future applications. To achieve this development, we have to minimize the environmental impact in the whole battery life cycle, from conception to degradation of the system, and reduce production costs. Polymer hydrogel electrolyte are one of the promising alternative for processing new flexible batteries.[2] A great hydrogel electrolyte should promise excellent ionic transport pathways and sufficient mechanical strength, not to cause short-circuits. As a matter of fact, hydrogel electrolytes are well-known for their good ionic conductivity. Nevertheless, the original polymers used in these systems don’t take into account the cost of the environmental impact and safety due to the processing or biodegradability of those hydrogels.[3] In this study, we report a new hydrogel-based electrolyte material made by apple pectin. This presentation will mainly focus on the interactions between pectin functional groups, water and ions using solid NMR spectroscopy. Thermal properties will be discussed based on differential scanning calorimetry analysis. Electrical and electrochemical characterisctics obtained by electrochemical impedance spectroscopy, galvanostatic cycling and cyclic voltametry will demonstrate the applicability of such hydrogel electrolyte. This study could promote a great innovation in the energy storage field, by recycling one of apple peel’s component (which is the main waste in preserves manufacturing[4]) into a hydrogel electrolyte. Background:IgG4-related disease (IgG4-RD) is an autoimmune disorder and frequently involve multiple organs. The respiratory tract is one of the most frequently involved sites.Objectives:This study aimed to compare the demographic and clinical characteristics of IgG4-related respiratory disease (IgG4-RRD) and non-IgG4-RRD patients in a large cohort.Methods:We carried out a retrospective study of 452 cases of IgG4-RD (104 IgG4-RRD patients and 348 non-IgG4-RRD patients) diagnosed at Peking University People’s Hospital between 2003 and 2020.Results:IgG4-RRD patients had an elder age of disease onset and diagnosis. Multiorgan involvement and hypocomplementemia were more common in IgG4-RRD. Besides, the level of ESR, eosinophilia, IgG and IgG4 were higher in IgG4-RRD patients. In IgG4-RRD group, salivary gland, lacrimal gland, lymph nodes, biliary system and kidney were more commonly involved than those in the non-IgG4-RRD group. Abstract Background: Pancreatic adenocarcinoma (PAC) is a malignant tumor with less than 5% of 5-year overall survival. Although immunotherapy has shown excellent and lasting efficacy in many solid tumors, immunomonotherapy is ineffective in PAC. The combinations of immune checkpoint inhibitors (ICIs) with radiotherapy/chemotherapy has shown encouraging results. Identifying predictive biomarkers is of great sense for PAC patients to benefit from immunotherapy. Methods: Formalin-fixed, paraffin-embedded (FFPE) tumor tissues and matched blood samples of 83 Chinese PAC patients were collected from Shandong Provincial Hospital Affiliated to Shandong First Medical University. Next-generation sequencing (NGS) of targeted-450 cancer genes was performed in OrigiMed (Shanghai, China), a College of American Pathologists (CAP) accredited and Clinical Laboratory Improvement Amendments (CLIA) certified laboratory. Genomic alterations, tumor mutational burden (TMB), and microsatellite instability (MSI) were assayed. PD-L1 expression was determined using Combined Positive Score (CPS). Results: A total of 83 PAC patients were enrolled in this cohort, including 50 (60.2%) males and 33 (39.8%) females. Among them, 79.5% (66/83) and 20.5% (17/83) were primary and metastatic lesions, respectively. The most common mutant genes were KRAS (84.3%, 70/83), TP53 (75.9%, 63/83), CDKN2A (27.7%, 23/83), SMAD4 (26.5%, 22/83) and ARID1A (12.0%, 10/83). In addition, some gene alterations associated with immune hyperprogression (HP) were detected in 9.6% (8/83) patients, including 11q13 amp (2.4%, 2/83), and CDKN2A/B gene loss (7.2%, 6/83). There are 32 (38.6%) PD-L1 positive patients, including 25 patients with CPS 1-10 and 7 patients with CPS≥20. We found that SPTA1 mutations were specifically detected in PD-L1 positive group, while OBSCN and ATM mutations were specifically detected in PD-L1 negative group. The median TMB value was 3.3 muts/Mb and only 4 (4.8%) patients had TMB-values higher than 10 muts/Mb (TMB-H). There are 3 patients (3.6%) showed MSI-H. Interestingly, all 3 MSI-H patients were TMB-H, and 2 of them also showed PD-L1 positive without any HP related mutations. Both patients received immunotherapy and were benefited for more than 11 months, including one of whom had received immunomonotherapy. Conclusions: We described the characteristics of TMB, MSI and PD-L1 expression in Chinese patients with PAC. In the absence of HP related mutations, comprehensive consideration of TMB, MSI and PD-L1 status may guide PAC immunotherapy and benefit PAC patients. Abstract Background For a long time, the use of immune checkpoint inhibitors(ICIs)has been limited in sarcomas and bone tumors, and is only recommended as second or later line therapy for patients who meet specific requirements such as dMMR/MSI-H. DNA damage response (DDR) pathways aim to protect cells against some acquired genome changes and monitor exogenous or endogenous DNA damage. Studies have reported that genomic instability caused by DDR gene mutations effects on the immune system, and may benefit from ICI treatment. Although some studies have reported that DDR gene mutations in solid tumors such as non-small cell lung cancer and bladder cancer predicted response to ICI and correlated with patient prognosis, reports are lacking in sarcomas and bone tumors. Methods Formalin-fixed, paraffin-embedded tissues and matched blood samples were collected from patients with sarcoma or bone tumor for targeted next-generation sequencing assay at OrigiMed (Shanghai, China), a College of American Pathologists accredited and Clinical Laboratory Improvement Amendments certified laboratory. PD-L1 expression was detected by immunohistochemistry using 22C3 antibody. Results Fifteen patients including 12 male and 3 female with a median age of 22 years were recruited. Among them, 9 patients had bone tumors with a median age of 17 years. PD-L1 was tested in 13 patients and 10 patients were positive.Genetic testing showed that TP53, BRCA1, and CCND3 were the most frequently mutated genes. The analysis of pathways showed that DDR (11/15), cell cycle (7/15), NOTCH (4/15), RTK.RAS (4\15) were the most affected pathways. The median TMB was 1.6 muts/Mb. In DDR genes, TP53 mutations occurred in 8 patients including 2 soft tissue sarcomas and 6 bone tumors. BRCA1 gene fusions occurred in 2 osteosarcoma patients, one is 7-year-old boy who carried 3 BRCA1 fusions with ASIC2, LUZP6 and OXCT1 fusion partners. The boy also carried TP53 germline pathogenic mutations. Another 17-year-old boy carried a BRCA1-TMEM271 fusion, and PTEN and STK11 DDR gene mutations. Among the 11 sarcoma patients with DDR mutations, 8 were PD-L1 positive and 1 was PD-L1 negative. PD-L1 testing was not performed in other 2 cases. Conclusion Our results showed that a proportion of sarcoma and bone tumor patients carried DDR gene mutations, especially in pediatric and adolescent osteosarcoma patients.The majority of sarcoma and bone tumor patients with DDR gene mutations in this cohort were PD-L1 positive. Since DDR gene mutation was associated with favorable response rate to ICIs, patients with DDR gene mutated sarcoma and bone tumor may benefit from ICIs therapy. Abstract Background Osteosarcoma (OS) is a highly aggressive malignant tumor of mesenchymal origin and prone to early hematogenous metastases. The 5-year overall survival of metastatic OS is only approximately 20% to 30%. Therefore, it is still clinical dilemma in the treatment of OS. Thus,understanding the molecular features of metastatic osteosarcoma become increasingly important. Methods Formalin-fixed, paraffin-embedded or fresh tissues and matched blood samples were collected from OS patients for whole exome sequencing using next-generation sequencing at OrigiMed (Shanghai, China), a College of American Pathologists accredited and Clinical Laboratory Improvement Amendments certified laboratory. Genomic alterations including single nucleotide variations (SNVs), short and long insertions/deletions (INDELs), copy number variations (CNVs), and gene rearrangements were assessed. Tumor mutational burden (TMB) and the number of tumor neoantigens (NEO) were also measured. Results In total, 38 patients including 29 males and 9 females were recruited with a median age of 19.5 years. Among them, 12 patients had first metastases to bone (group B). The metastatic sites included femur, fibula, pelvis, ribs, sacrum, and spine. The median age of group B was 19.5 years. Twenty-six (26) patients had first metastases to lung (group L). The median age of group L was 14 years.The most frequently mutated genes in OS was TP53 (55.3%), followed by MYC (36.8%), MUC16 (26.3%), PTK2 (21.1%), RAD21 (21.1%), and CDK4 (18.4%). However, genetic features between Groups B and Group L patients were different. In group B patients, the median percentage of SNVs and short INDELs was 94.7% of the overall mutations, and the median percentage of CNVs was 4.3%. In contrast, the median percentage of SNVs and short INDELs in group L was 58.8%, and the median percentage of CNVs was 36.7%. The mutations patterns were clearly different between Group B and Group L with a predominant SNV & INDEL in Group B and a mixture feature of SNV & INDEL and CNV in Group L. Inaddition the median TMB in group B was significantly higher than that in group L (4.85 muts/Mb vs 2.4 muts/Mb, P&lt;0.05). Also, the median number of tumor NEO in group B was nearly 6 times higher than that in group L (743 vs 128.5, P=0.0016). Conclusion Our study identified different molecular features of patients with OS firstly metastasizing to lung and to bone. OS with first bone metastases had a predominant SNV and short INDEL, high TMB level, and high NEO counts, while OS with first lung metastasis had an increased CNV, low TMB, and low NEO counts. Our results suggest that metastatic OS that firstly spread to bone and to lung may be two distinct subgroups and may adopt different treatment strategies. Abstract Background: Morpho-physiological alternations of platelets provided a rationale to harness RNA sequencing of tumor-educated platelets (TEPs) for preoperative diagnosis of cancer. Timely, accurate, and non-invasive detection of ovarian cancer in women with adnexal masses presents a significant clinical challenge. Patients and Methods: This intercontinental, hospital-based, diagnostic study included 761 treatment-naïve inpatients with histologically confirmed adnexal masses and 167 healthy controls from nine medical centers (China, n=3; Netherlands, n=5; Poland, n=1) between September 2016 and May 2019. The main outcomes were the performance of TEPs and their combination with CA125 in two Chinese (VC1 and VC2) and the European (VC3) validation cohorts collectively and independently. Exploratory outcome was the value of TEPs in public pan-cancer platelet transcriptome datasets. Results: The AUCs for TEPs in the combined validation cohort, VC1, VC2, and VC3 were 0.918 (95% CI 0.889-0.948), 0.923 (0.855-0.990), 0.918 (0.872-0.963), and 0.887 (0.813-0.960), respectively. Combination of TEPs and CA125 demonstrated an AUC of 0.922 (0.889-0.955) in the combined validation cohort; 0.955 (0.912-0.997) in VC1; 0.939 (0.901-0.977) in VC2; 0.917 (0.824-1.000) in VC3. For subgroup analysis, TEPs exhibited an AUC of 0.858, 0.859, and 0.920 to detect early-stage, borderline, non-epithelial diseases and 0.899 to discriminate ovarian cancer from endometriosis. Analysis of public datasets suggested that TEPs had potential to detect multiple malignancies (Table 1). Conclusions: TEPs had robustness, compatibility, and universality for preoperative diagnosis of ovarian cancer since it withstood validations in populations of different ethnicities, heterogeneous histological subtypes, early-stage ovarian cancer as well as other malignancies. Lithium metal batteries (LMB) have risen back into focus of battery research in resent years, due to the high theoretical specific capacity of the lithium metal anodes (3860 mAh g-1).[1] However, the utilization of lithium metal as an anode material requires an in depth understanding of the solid electrolyte interphase (SEI) on lithium metal electrodes, since its formation, composition and electrochemical characteristics significantly impact important LMB lifetime parameters, e.g. lithium stripping/plating behaviour, dendrite growth and in operando electrolyte consumption.[2] Lithium metal electrode performance is influenced by a variety of different, however often overlooked factors, e.g. the native passivation layer, inevitably present on each lithium metal electrode surface, which has only recently gathered more attention as an influencing factor in respect to SEI characteristics.[3] The presented research utilizes the preformation of an uncontaminated SEI on the lithium metal electrode surface to circumvent the influence of native lithium metal passivation. A systematic study of different preformed SEIs resulted in a better understanding of the influence by different electrolyte components on relevant SEI characteristics, namely, selected conducting salts (LiFSI, LiPF6) and functional additives (VC, FEC). Analyses of the SEIs electrochemical properties, composition, morphology and dynamics were conducted via stripping/plating experiments as well as in operando EIS, SEM (incl. cryogenic cross section and post mortem) and XPS (post mortem) measurements. Finally, careful tailoring of the preformed SEI was shown to have an enhancing effect on NMC811||Li cells, increasing the LMB lifetime by up to 25%. Furthermore, the application of a tailored preformed SEI has also shown a positive impact on the performance of solid state LMBs. The presented results highlight the importance of an in depth understanding regarding the cause and effect of varying electrolyte components on the SEIs characteristics. Kết quả cho thấy, khi Fe thay thế cho Ti, cấu trúc của vật liệu chuyển từ tứ giác (P4mm) sang lục giác (P63/mmc). Sự xuất hiện của các mức tạp chất Fe kết hợp với các mức tạp do sự khuyết thiếu ôxy, sai hỏng mạng tạo nên sự chồng chập, mở rộng dải hấp thụ và làm dịch bờ hấp thụ về phía bước sóng dài. Chúng tôi cũng chỉ ra rằng, các ion Fe3+ và Fe4+ đã thay thế cho ion Ti4+ trong cấu trúc tứ giác và lục giác của vật liệu BaTiO3. Chúng tôi cho rằng, tính chất sắt từ của vật liệu BaTi1-xFexO3 có nguồn gốc từ những sai hỏng mạng và tương tác trao đổi giữa các ion Fe3+ và Fe4+.

Abstract Background: Metronomic chemotherapy, defined as frequent administration of. chemotherapeutic agents at a non-toxic dose without extended rest periods, can overcome drug resistance and achieve disease control with reduced toxicity compared to conventional chemotherapy in maximum tolerated dose by shifting the therapeutic target from tumor cells to tumor endothelial cells. Some of the previous studies of oral vinorelbine have shown good data in efficacy and safety in advanced breast cancer. Methods: The multicenter, open-label, non-inferiority, randomized phase 2 study (NCT03854617) aimed to evaluate the efficacy and safety of oral metronomic vinorelbine in 13 hospitals in China. Eligible HER2-negative breast cancer patients previously treated with anthracycline or taxane regimens were randomized (1:1) to receive metronomic dosage of oral vinorelbine (50mg/3 times a week) or conventional dosage of oral vinorelbine (60mg/m2 weekly for cycle 1 and 80mg/m2 weekly for subsequent cycles in the absence of grade 3 or 4 toxicity) for first-line/second-line chemotherapy. The primary end point was Disease Control Rate (DCR) and a non-inferiority margin of 6% was defined for DCR. Patient characteristics, progression-free survival (PFS), overall survival (OS) and safety/adverse events (AEs) were among the parameters assessed. The expression of 27 cytokines was profiled longitudinally in these patients at baseline and at regular intervals during therapy. Results: Between February 2019 and September 2020, a total of 171 patients were enrolled and randomized to metronomic dosage group (86 patients) and conventional dosage group (85 patients). 136 patients were hormone receptor(HR)positive and 117 patients (68.4%) had visceral metastases. The DCR was 59.3% (95% CI:48.17% to 69.78%) in the metronomic dosage group and 67.1% (95% CI:56.02% to 76.87%) in the conventional dosage group. Whereas, the 18-month survival rate was higher in the metronomic dosage group than that in the conventional dosage group (68.7% vs 43.0%).The median progression-free survival in the metronomic dosage group was 2.8 months (95% CI:1.40 to 3.50) compared with 4.1 months (95% CI:2.80 to 6.20) in the conventional dosage group. Grade 3 or higher adverse events were significantly less frequent in patients in the metronomic dosage group than patients in the conventional dosage group (19.8% vs 48.2%, P&lt;0.001). By comparing the variation of cytokine profiles at baseline and after 6-week treatment in 122 patients, multilevel partial least squares discriminant analysis (PLS-DA) suggested the variation of VEGF, MIP-1α, IL-1B, IL-17, MCP-1, IL-13, PDGF-BB, IL-4 and RANTES were significantly different between the metronomic dosage group and the conventional dosage group during the treatment (all VIP values &gt; 1.2). As for the patients in the metronomic dosage group, GM-CSF, MCP-1, TNF-α, IL-10, IL-13 and MIP-1α were potential biomarkers between the response patients and non-response patients (all VIP values &gt; 1.2). Conclusions: Oral metronomic vinorelbine decreased the risk of severe toxicity significantly and may be an option for older patients and for those intolerable to standard chemotherapy with proper predictive biomarkers, though this study couldn’t prove to show the non-inferiority of oral metronomic vinorelbine for first-line or second-line chemotherapy previously treated with anthracycline or taxane in HER2-negative metastatic breast cancer. Despite recent concerns about the increasing influence of outside investors on the European Union (EU) and Western Balkans, the developed European countries are still a dominant source of foreign direct investment (FDI) in the region, confirming the benefits of EU membership. At the same time, fast-growing connectivity and lower trade costs in accession and neighboring countries determine the FDI growth from China, particularly via the Belt and Road Initiative (BRI). By applying panel data over 2000-2019 for 34 countries, which form 89% of all European FDI, we first examine FDI patterns around Europe, compare the EU, NMS, and Western Balkans; verify the importance of EU membership for FDI, caused reducing trade costs and improving connectivity. Thus, the new EU member states (NMS) and Western Balkans appear both as a home country and as a pre- entry destination to the EU. Then, we calculate trade costs indices for each selected country and partners over time and find that Europe and China are closely interconnected through trade and FDI. It means that stronger ties with China can be realized for the sample countries at the cost of easing relations with the EU. Finally, incorporating trade costs indices into the FDI model; we evaluate the impact of connectivity on FDI and estimate how BRI affected FDI in Europe. Additionally, we validate that the old framework of horizontal and vertical FDI not representative well and even new complex vertical or export-oriented FDI strategies are shifting today.

A total of 96 28-d-old healthy weaned male pigs (Duroc×Landrace×Yorkshire) with similar weight were randomly assigned into two groups. Six units were included in each treatment, with eight pigs in each unit. The trial lasted 35 d. Four pigs were exchanged between every two pens in the same treatment on day 28. No difference was observed on average daily feed intake, average daily gain and feed-to-gain ratio between the pigs fed 30 mg kg−1 GABA diets and the pigs fed basal diet, respectively. Longer feed consumption time and lower biting incidences were observed in pigs fed GABA, compared with those fed basal diet, respectively. Plasma growth hormone concentration was significantly increased and the plasma concentrations of adrenocorticotrophic hormone and cortisol were lower in weaned pigs in the GABA supplementation group on day 35. Dietary GABA significantly increased the plasma neuropeptide Y concentration on day 14 and day 35. It is indicated that dietary GABA reduced aggressive behavior and regulated endocrine hormones in weaned pigs, ultimately reducing the negative influences of stress (weaning and mixing) on the pigs.

We set out to assess the diagnostic utility of the split hand index (SHI) for amyotrophic lateral sclerosis (ALS) and also to see if and how it can be applied to severely atrophied muscles, a frequent finding in this setting. We enrolled 38 patients from our clinic, 19 diagnosed with ALS and 19 controls, matched for age and sex. The SHI was calculated, on both sides, for all the patients. We calculated a SHI of 0 when the abductor pollicis brevis muscle (ABD) or first dorsal interosseous muscle (FDI) compound muscle action potentials (CMAPs) were unobtainable, and we allotted a value of 0.1 mV to abductor digiti minimi muscle (ADM) CMAP, for mathematical purposes, when the value would have been 0. The means differences were large between groups, with a significant variance heterogeneity. We performed a ROC analysis and obtained an accuracy of 0.83 for a SHI of 7.2, p-value < 0.0001. In conclusion, we reaffirm the utility of the SHI in the diagnosis of ALS, especially in limb onset cases, and we think that it can be safely extended to severely atrophied muscles with absent or very low CMAP values, without endangering the sensitivity or specificity.

The developing erythron requires tremendous amounts of iron (Fe) for the synthesis of heme for hemoproteins, such as hemoglobin, and iron-sulfur (Fe/S) clusters of proteins, which are required to catalyze redox reactions and regulate Fe uptake and storage. The uptake of Fe from transferrin (Tf) involves the binding of Tf to its cognate receptor (TfR), followed by the endocytosis of the Tf-TfR complex.1,2 In the late endosome, the release of Fe3+ from TfR is achieved by acidification of the vesicle by the v-ATPase H+-pump. Steap3 reduces the liberated Fe3+ prior to its transport out of the endosome by the DMT1 transporter. In contrast to previous conventional models for a cytosolic intermediate state, new data have emerged showing the direct interorganellar transfer of Fe from the endosome to the mitochondria.3 Although it is assumed that the exported Fe is targeted to the mitochondria for eventual incorporation into heme and Fe/S clusters, our understanding of the precise mechanism of how Fe traverses the outer and inner mitochondrial membranes remains poorly understood. Work in yeast mutants have implicated the role of solute carriers, Mrs3/4p, in mitochondrial iron homeostasis and revealed that it is the reduced form of iron, Fe2+, that is imported into the mitochondria. Subsequent studies of the zebrafish mutant, frascati, led to the discovery of mitoferrin 1 (Mfrn1, slc25a37), the vertebrate ortholog of Mrs3/4, as the major iron importer across the inner mitochondrial membrane in developing erythroblasts.4 A structurally related paralog, mitoferrin 2 (Mfrn2, slc25a28), plays the analogous role of Fe importer in non-erythroid cells. Loss-of-function studies of Mfrn1 in the mouse have confirmed its requirement in mammalian primitive and definitive erythropoiesis and its essential role in heme and Fe/S biosynthesis. Several questions remain unanswered in mitochondrial Fe metabolism: Do the two Mfrn importers account for all Fe imported into the mitochondria? How does Fe get across the outer mitochondrial membrane to reach the Mfrn importers? How does the translocated Fe in the matrix ultimately reach ferrochelatase to form heme? How is Fe translocated across the inner mitochondrial membrane?

Purpose The purpose of this paper is to define the “fast evolving industry” (FEI) and its supply chain management (SCM) challenges. The authors review and structure the literature regarding integration strategies and implementation methods to develop a strategic decision-making framework for SCM in the FEI. Design/methodology/approach The authors conduct a review of SCM literature, including supply chain strategy, supply chain integration (SCI), agile and responsive supply chain and SCM for innovative and fast-changing industries. The authors develop a conceptual model and a decision-making framework and use four mini cases to provide support for the model and framework. Findings The FEI, characterised by a high level of innovation and differentiation, short products/services lifecycle and high variety, is yet to be fully defined. Inherent uncertainty in FEI supply systems makes SCM in these industries a complex but strategic task for their managers. The framework and the model offered in this study, which employ a core competency concept and provide risk management strategies, offer a strategic tool for managers and scholars in the field to optimise their integration strategies and to operationalise integration decisions. Originality/value Little research has been published on transferable and cross-industrial SCM in FEIs. This paper defines the FEI and its resource-related concerns and then offers a conceptual model and a strategic decision-making framework for SCI in FEIs.

Seaweed is a valuable coastal resource for its use in food, cosmetics, and other items. This study proposed new remote sensing based seaweed enhancing index (SEI) using spectral bands of near-infrared (NIR) and shortwave-infrared (SWIR) of Landsat 8 satellite data. Nine Landsat 8 satellite images of years 2014, 2016, and 2018 for the January, February, and March months were utilized to test the performance of SEI. The seaweed patches in the coastal waters of Karachi, Pakistan were mapped using the SEI, normalized difference vegetation index (NDVI), and floating algae index (FAI). Seaweed locations recorded during a field survey on February 26, 2014, were used to determine threshold values for all three indices. The accuracy of SEI was compared with NDVI while placing FAI as the reference index. The accuracy of NDVI and SEI were assessed by matching their spatial extent of seaweed cover with FAI enhanced seaweed area. SEI images of January 2016, February 2018, and March 2018 enhanced less than 50 percent of the corresponding FAI total seaweed areas. However, on these dates the NDVI performed very well, matching more than 95 percent of FAI seaweed coverage. Except for these three times, the performance of SEI in the remaining six images was either similar to NDVI or even better than NDVI. SEI enhanced 99 percent of FAI seaweed cover on January 2018 image. Overall, seaweed area not covered by FAI was greater in SEI than NDVI in almost all images, which needs to be further explored in future studies by collecting extensive field information to validate SEI mapped additional area beyond the extent of FAI seaweed cover. Based on these results, in the majority of the satellite temporal images selected for this study, the performance of the newly proposed index—SEI, was found either better than or similar to NDVI.

Labour market dependency on Foreign Direct Investments (FDI) inflow is very high in transition economies. This paper estimates the effects of FDI inflow on the employment rate and average net wages in Western Balkan economies in the period 2003-2017. The sample of economies (Albania, Bosnia and Herzegovina, Croatia, North Macedonia, Montenegro, and Serbia) was selected mainly because of the high legacy with FDI and unsaturated labour market. Presumably, FDI inflow has a positive impact on the employment rate and on average net wages in the Western Balkan countries. Employing linear mixed-effects models (LMM), the results indicate that FDI inflow changes have very low but positive and significant effects on both the changes in employment rate and on average net wages.

Thirty-two (60±2 kg) crossed-bred barrows (Duroc×Landrace×Yorkshine) were randomly assigned to one of four diets: (1) the control diet containing no corn DDGS (control group); (2) the diet containing 30% corn DDGS (DDGS-fed group); (3) the diet containing 30% corn DDGS and 10 g kg−1 CLA (CLA-fed group); (4) the diet containing 30% corn DDGS and 1 g kg−1 BET (BET-fed group). Dietary DDGS decreased the proportion of saturated fatty acids (SFA) (P<0.05), and increased the proportion of unsaturated fatty acids (UFA) and polyunsaturated fatty acids (PUFA) (P<0.05) in pork with respect to the control group, whereas dietary CLA and BET decreased the proportion of PUFA and increased the proportion of SFA in pork compared with DDGS-fed group. Pork treated with DDGS alone had higher total volatile basic nitrogen (TVB-N) value (P<0.05) and thiobarbituric acid reactive substances (TBARS) value (P<0.05) with respect to the control diet during display, whereas CLA-fed group and BET-fed group had lower TVB-N concentration and TBARS value (P<0.05) in pork than DDGS-fed group. There was no significant difference of instrumental color (L*, a*, b*) between DDGS-fed group and the control group (P>0.05). However, CLA-fed and BET-fed groups maintained lower L* values and higher a* values (P<0.05), but did not affect b* values with respect to the DDGS-fed group and the control group. Results confirmed that diets containing 30% DDGS have some adverse effects on pork freshness and lipid peroxidation; However, supplementation with CLA or BET can partially reverse these effects and improve color image of pork during display.

Within these reform acts budgets for SHI-expenditures were introduced in 1997, and reimbursement schemes were changed from fee-for-service-payments to capitation and flat rates per case. This article discusses the possible effects of these institutional changes on the quality of health care. It then demonstrates the weaknesses of the regulatory strategy for quality assurance pursued by German health policy. Finally, it outlines a two-tier strategy based on regulatory and competitive elements to ensure a high quality of care.

AbstractChina has achieved nearly universal social health insurance (SHI) coverage by implementing three statutory schemes, but gaps and differences in benefit levels are apparent. There is wide agreement that China should merge the three schemes into a universal and uniform SHI. However, data on the medical expenses of all inpatients in 2014 at a public Tier-three hospital suggests that supply-induced demand (SID) is a serious concern and that, under the design of the current schemes, a higher benefit level has a greater impact on the total expenses of insured patients. Thus, if SID is not effectively controlled, a universal and uniform SHI may be more harmful than beneficial in China. Finally, we suggest that China should substitute the existing fee-for-service design with a suite of bundled provider payment methods; furthermore, China should replace its current system of pricing drugs that encourages hospitals and doctors to use costlier medications.

A feeding trial with broilers was conducted to evaluate the effects of amino acids (lysine and methionine) supplementation of diets low in protein content on the voluntary intake, feed conversion efficiency, broiler performance, and cost and return of broiler production. Results showed cumulative voluntary feed intake was not significantly affected by lowering the protein content of the diet. Cumulative weight gain of broilers was lower with diet when supplemented iwht lysine and methionine to meet requirements. Birds fed with diets low in protein has less efficient feed converstion, but became comparable with those receiveing diets high in protein when supplemented with amino acids. Feed cost per kilogram broiler produced was not significantly affected by diets used in the study, although the low-protien diet with amino acid supplement had the lowest values. In terms of return above feed and chick cost, broilers fed with high-protein diet had the greatest value, but not significantly different from birds fed with low-protien diet with amino acid supplementation which gave about P10 per bird higher returns than those fed low-protein diet without amino acid supplementation.

The solid electrolyte interphase (SEI) film plays a significant role in the capacity and storage performance of lithium primary batteries. The electrolyte additives are essential in controlling the morphology, composition and structure of the SEI film. Herein, fluoroethylene carbonate (FEC) is chosen as the additive, its effects on the lithium primary battery performance are investigated, and the relevant formation mechanism of SEI film is analyzed. By comparing the electrochemical performance of the Li/AlF3 primary batteries and the microstructure of the Li anode surface under different conditions, the evolution model of the SEI film is established. The FEC additive can decrease the electrolyte decomposition and protect the lithium metal anode effectively. When an optimal 5% FEC is added, the discharge specific capacity of the Li/AlF3 primary battery is 212.8 mAh g−1, and the discharge specific capacities are respectively 205.7 and 122.3 mAh g−1 after storage for 7 days at room temperature and 55 °C. Compared to primary electrolytes, the charge transfer resistance of the Li/AlF3 batteries with FEC additive decreases, indicating that FEC is a promising electrolyte additive to effectively improve the SEI film, increase discharge-specific capacities and promote charge transfer of the lithium primary batteries.

In this study, a polymer with characteristics analogous to that of the humic acid was obtained through oxidative polymerization in alkaline medium using para-benzoquinone as precursor, and compared to natural humic acid (HA) extracted from a soil. Complexes of as-prepared synthetic humic acid (SHA) with the Cu2+, Fe3+ and Mn2+ ions were also prepared. Elemental analysis and thermogravimetry were used for comparing the SHA characteristics with those of HA and for evaluating the effect of the metal-type for complexing with SH and SHA. Element analysis showed that SHA presented almost the same atomic constitution of HA, except for or sulfur and nitrogen contents, fact that is expected due to the precursor compounds. TGA curves of HA and SHA are quite similar in spite the thermogravimetic index (TGI) indicates higher aromatic portion in SHA. SHA-metallic ions (SHA-Cu2+, SHA-Fe3+ and SHA-Mn2+) complexes presented different thermal stability as compared among them and with the precursor SHA. The data indicate that the complexation is dependent of metallic ion-type and provokes different rearrangements in the SHA matrix.

Abstract Abstract SCI-23 As the human body cannot excrete excess iron, its absorption needs to be finely regulated at the intestinal level. Ferric iron (Fe3+) is reduced to ferrous iron (Fe2+) by brush border ferric reductases, including duodenal cytochrome b (DCYTB), before being transported across the apical membrane by divalent metal transporter 1 (DMT1), which is the principal iron importer. Depending on body iron requirements, iron can be either stored bound to ferritin or exported across the basolateral enterocyte membrane into the plasma by the sole iron exporter ferroportin (FPN). Iron absorption responds to systemic signals reflecting body iron requirements and local signals in the enterocyte. At the systemic level, hepcidin is the key circulating peptide hormone maintaining iron homeostasis. Hepcidin controls plasma iron concentration by inhibiting intestinal iron absorption and iron recycling by macrophages. Hepcidin acts by inhibiting cellular iron efflux through binding to and inducing the degradation of FPN. Hepcidin transcription is upregulated by iron repletion and downregulated by iron deficiency, ineffective erythropoiesis, and hypoxia. Hypoxia-inducible factors HIF-1 and HIF-2 are heterodimeric transcriptional factors and central mediators of cellular and systemic adaptation to hypoxia. In the presence of oxygen, the HIF-α subunit is targeted to the proteasome, while in hypoxia (or iron deficiency), HIF-α is stabilized and induces the transcription of target genes. We hypothesized that HIFs, stabilized in the hypoxic intestinal epithelium, may also play critical local roles in regulating intestinal iron absorption. We generated conditional knockout mice that lacked either Hif1a or Hif2a specifically in the intestinal epithelium and found that HIF-1α was not necessary for iron absorption, whereas HIF-2α played a crucial role in maintaining iron balance in the organism by directly regulating the transcription of the genes encoding DMT1 and DCYTB. Specific deletion of Hif2a led to a decrease in serum and liver iron levels. Alterations in HIF-2 at the intestinal level can override systemic regulation via hepcidin. Interestingly, we further demonstrated that HIF-2α contributes to iron hyperabsorption in a genetic mouse model of hereditary hemochromatosis (HH). HH is a genetic disorder characterized by abnormally low hepcidin expression and excessive iron accumulation in the liver and parenchyma. These findings suggest a prominent role of HIF-2 in the physiopathological regulation of intestinal iron absorption and may provide new therapeutic perspectives for the treatment of anemias and iron overload-associated disorders. This research focuses on the nexus between foreign direct investments (FDI) and external balance in selected Emerging European Economies (EEEs). Emerging countries in convergence process tend to have problem of structural external imbalance that is covered with foreign capital inflows. FDI are long-term and sustainable source of financing the current account deficit. The aim of this paper is to test if FDI together with chosen macroeconomic indicators are relevant factors of trading balance in EEEs in order to give useful implications towards economic policy creators in emerging economies. The research hypothesis is tested with robust micro panel models for total sample and two subperiods: before and after the structural break caused with Global financial crisis (GFC). The results indicate that substantial FDI inflows are significantly related with negative sum of trading balance on the total sample level i.e. average external position deficit is financed with the FDI inflows. Also, dummy variable for the Western Balkans indicates that FDI are significant variable that finances external imbalance in this subsample. The government policy recommendations are directed towards incentive measures for attracting greater FDI inflows, especially greenfield investments motivated with greater efficiency and export stimulation in order to stabilize trading balance and foster economic growth.

Src homology 3 (SH3) domains have been suggested to play an important role in the assembly of the superoxide-forming nicotinamide adenine dinucleotide phosphate (NADPH) oxidase upon activation of phagocytes, which involves the association of membrane-bound and cytosolic components. We studied the translocation of the cytosolic proteins to the plasma membrane in neutrophils of a patient with a point mutation in the gene encoding the light chain of cytochrome b558. This mutation leads to a substitution at residue 156 of a proline into a glutamine in a putative SH3 binding domain of p22-phox (Dinauer, M., E. A. Pierce, R. W. Erickson, T. Muhlebach, H. Messner, R. A. Seger, S. H. Orkin, and J. T. Curnutte. 1991. Proc. Natl. Acad. Sci. 88:11231). In PMA-stimulated neutrophils and in a cell-free translocation assay with neutrophil membranes and cytosol, association of the cytosolic proteins p47-phox and p67-phox with the membrane fraction of the patient's neutrophils was virtually absent. In contrast, when solubilized membranes of the patient's neutrophils were activated with phospholipids in the absence of cytosol (Koshkin, V., and E. Pick. 1993. FEBS [Fed. Eur. Biochem. Soc.] Lett. 327:57), the rate of NADPH-dependent oxygen uptake was observed at a rate similar to that of control membranes. We suggest that the binding of an SH3 domain of p47-phox to p22-phox, and thus activation of the oxidase, does not occur in the neutrophils of this patient, although under artificial conditions, electron flow from NADPH to oxygen in cytochrome b558 is possible.

Study aims to analyze the macroeconomic effect of the Dow Jones index on IDX Composite on the Indonesia Stock Exchange. The variables used in the macro economy are inflation, kurs, SBI rate, FED rate. Another variable is the Dow Jones index. This study uses quarterly secondary data from 2010 to 2020. The data analysis model uses the Autoregressive Distributed Lag (ARDL) approach. The results of the ARDL model analysis show that in the long term inflation, kurs, and FED rate has a negative and insignificant effect on the IDX Composite, SBI rate variable has a positive and not significant effect on the IDX Composite, the Dow Jones index variable has a positive and no significant effect on the IDX Composite significant to the IDX Composite. In the short term, inflation has a negative and insignificant effect on the IDX Composite, kurs has a negative and significant effect on the IDX Composite, SBI rate has a positive and insignificant effect on the IDX Composite, FED rate and the Dow Jones index have a positive and significant effect on the IDX Composite.
