Abstract:
Abstract Modern Chinese confirmative shi (as in mei cuo, wo shi yao dusi ni (沒錯,我是要毒死你) ‘that’s right. I really wanted to poison you to death’) is not an auxiliary but an adverb. It derives from the adjective shi ‘true, real’ in Old Chinese (Yan zhi yan shi ye (偃之言是也) ‘what Yan said was true’). The grammaticalization pathway of the Modern Chinese confirmative shi is different from that of the copula shi (Laozhang shi huoche siji (老張是貨車司機) ‘Laozhang is a truck driver’) or the auxiliary shi (Laozhang shi kai huoche, wo shi kai keche (老張是開貨車,我是開客車) ‘Laozhang drives a truck and I drive a coach car’). Modern Chinese confirmative shi, copula shi, and auxiliary shi have the same morphological form because they all appear to derive from the adjective shi or demonstrative ( shi ke ren, shu bu ke ren ( 是可忍,孰不可忍) ‘if this could be endured, is there anything else that could not be endured’) in Old Chinese. Such a pattern of morphological sameness seems to be cross-linguistically rare, if not unique.

The genus Mongolotettix Rehn, 1928 is a small genus of family Adrididae. It has nine species discovered from the Far Eastern Asian countries, including China, Japan, Korea, Mongolia, and Russia [Bolívar, 1898; Uvarov, 1914; Caudell, 1921; Chogsomzhav, 1974; Li et Lian, 1994; Wan, Ren et Zhang, 1998; Xie et Li, 2000, Kim et Kim 2005; Dong, et al, 2015; Shi et al, 2016; Storozhenko, 2016; Eades et al, 2017; Zhang et al, 2017]. In the present paper, we describe a new species of the genus from Hubei, China. Abstract:
AbstractA new genus and new species of Nymphidae (Neuroptera) is described from the Ypresian Okanagan Highlands locality of Falkland, British Columbia, Canada: Epinesydrion falklandensisnew genus, new species. This is only the fourth known Cenozoic adult specimen, and all others are less complete. It is the second specimen from the Okanagan Highlands. Currently Nymphidae has two recognised subfamilies. All Cenozoic fossils are confident members of the Nymphinae, but the subfamily assignments of almost all Mesozoic genera are problematic. The Late Cretaceous Dactylomyius is the only genus that might belong to Myiodactylinae. The rest may belong to the undefined stem groups of the family or to the Nymphinae, with varying levels of probability. Mesonymphes sibirica is transferred to Nymphites Haase: N. sibiricus (Ponomarenko), new combination; Sialium minor to Spilonymphes Shi, Winterton, and Ren: Spilonymphes minor (Shi, Winterton, and Ren), new combination; “Mesonymphes” apicalis does not belong to Mesonymphes Carpenter and may not even belong to the Nymphidae. The fossil record of the family occurs across much of the globe, but today they are restricted to Australia, New Guinea, and possibly the Philippines. Modern Nymphinae is only found in Australia. This may result from a requirement of frost-free climates, which were more widespread in the past.

Reviewing his long reign in 1792, the Qianlong Emperor (r. 1736–1795) hailed his military triumphs as one of its central accomplishments. To underscore the importance he ascribed to these successes, he began to style himself ‘Old Man of the Ten Complete Victories’ (Shi Quan Lao Ren), after an essay in which he boldly declared he had surpassed, in ‘Ten Complete Military Victories’ (Shi Quan Wu Gong), the far-reaching westward expansions of the great Han (206 BCE–220 CE) and Tang (618–907) empires. Such an assertion, together with the program of commemoration discussed below, served to justify the immense expense incurred by frequent long-distance campaigning; to elevate all these wars to an unimpeachable level of splendor even though some were distinctly less glorious than others; and to align the Manchu Qing dynasty (16–191 i) with two of the greatest native dynasties of Chinese history and the Qianlong Emperor personally with some of the great figures of the past.

The Bacillus halodurans voltage-gated sodium-selective channel (NaChBac) (Ren, D., B. Navarro, H. Xu, L. Yue, Q. Shi, and D.E. Clapham. 2001b. Science. 294:2372–2375), is an ideal candidate for high resolution structural studies because it can be expressed in mammalian cells and its functional properties studied in detail. It has the added advantage of being a single six transmembrane (6TM) orthologue of a single repeat of mammalian voltage-gated Ca2+ (CaV) and Na+ (NaV) channels. Here we report that six amino acids in the pore domain (LESWAS) participate in the selectivity filter. Replacing the amino acid residues adjacent to glutamatic acid (E) by a negatively charged aspartate (D; LEDWAS) converted the Na+-selective NaChBac to a Ca2+- and Na+-permeant channel. When additional aspartates were incorporated (LDDWAD), the mutant channel resulted in a highly expressing voltage-gated Ca2+-selective conductance.

The paper is the first of a series, which aims to present a consistent interpretation of the suprageneric taxa of fossil beetles in the current century and their generic and species composition. Abstract References to native culture are frequently in the foreground of works by Chinese women artists. When they make contact with different cultures, although not necessarily connected with leaving their place of birth thanks to the transfer of information and cultural heritage that has developed extremely efficiently in the era of globalization (Gordon Mathews), they see their own entanglement in the cultural tradition. In the process of constructing their identity they try to find answers to the following question: which part of the cultural tradition is mine? Which one do I identify with? In the case of Chinese women artists, is it the legacy of literati? Classic ink painting and calligraphy? Or perhaps women's crafts that bear no name? Or perhaps a mixture of inspirations? Such questions about material heritage might also be augmented by others that consider aspects of the immaterial heritage of China. This article explores how Chinese women artists such as Chen Qingqing, Qin Yufen, Shi Hui, Wang Xiaohui, Cheng Caroline, Lin Tianmiao, Zhang Yanzi, Man Fung-yi, Liu Liyun, Peng Wei, Chen Lingyang, Chen Qiulin, Zhang Ou and Liu Ren refer to their cultural tradition.

TheHuainanziis a Former Han 前漢 dynasty (202 B.C.E.–9 C.E.) compendium of knowledge written at the court of Huainan and presented to Emperor Wu of the Han 漢武帝 in 139 by Liu An 劉安 (?179–122), the king of Huainan. Liu An was the grandson of the Han “progenitor” Gaozu 高祖 (Liu Bang 劉邦 r. 202–195), and he was the uncle of the reigning emperor Wu (r. 140–87). According to the author(s) of the text’s postface, “Yao lue” 要略 or “A Summary of the Essentials,” the work seeks to provide a comprehensive account or chronicle of thedao道 (conventionally translated as the “Way”), understood broadly to encompass the cosmos (tiandi天地 or “Heaven and Earth”), human beings (ren人) and their affairs (shi事), and the relationship between them. The account of thedaopresented in its chapters is not, however, purely descriptive. TheHuainanziis foremost a political treatise containing instructions worthy of a sage-king (shengwang聖王) to be employed by the ruler as the proper model or standard by which to govern the empire.

Asthma is one of the most common allergic respiratory diseases around the world and places great burden on medical payment. Chinese herbal medicine (CHM) is commonly used for Taiwanese children to control diseases. The aim of this study is to analyze the CHM prescriptions for asthmatic children by using a nationwide clinical database. The National Health Insurance Research Database (NHIRD) was used to perform this study. Medical records from 1997 to 2009 with diagnosis with asthma made for children aged 6 to 18 were included into the analysis. Association rule mining and social network analysis were used to analyze the prevalence of single CHM and its combinations. Ma-Xing-Gan-Shi-Tang (MXGST) was the most commonly used herbal formula (HF) (20.2% of all prescriptions), followed by Xiao-Qing-Long-Tang (13.1%) and Xing-Su-San (12.8%). Zhe Bei Mu is the most frequently used single herb (SH) (14.6%), followed by Xing Ren (10.7%). MXGST was commonly used with Zhe Bei Mu (3.5%) and other single herbs capable of dispelling phlegm. Besides, MXGST was the core formula to relieve asthma. Further studies about efficacy and drug safety are needed for the CHM commonly used for asthma based on the result of this study.

The bacterial sodium channel, NaChBac, from Bacillus halodurans provides an excellent model to study structure–function relationships of voltage-gated ion channels. It can be expressed in mammalian cells for functional studies as well as in bacterial cultures as starting material for protein purification for fine biochemical and biophysical studies. Macroscopic functional properties of NaChBac have been described previously (Ren, D., B. Navarro, H. Xu, L. Yue, Q. Shi, and D.E. Clapham. 2001. Science. 294:2372–2375). In this study, we report gating current properties of NaChBac expressed in COS-1 cells. Upon depolarization of the membrane, gating currents appeared as upward inflections preceding the ionic currents. Gating currents were detectable at −90 mV while holding at −150 mV. Charge–voltage (Q–V) curves showed sigmoidal dependence on voltage with gating charge saturating at −10 mV. Charge movement was shifted by −22 mV relative to the conductance–voltage curve, indicating the presence of more than one closed state. Consistent with this was the Cole-Moore shift of 533 μs observed for a change in preconditioning voltage from −160 to −80 mV. The total gating charge was estimated to be 16 elementary charges per channel. Charge immobilization caused by prolonged depolarization was also observed; Q–V curves were shifted by approximately −60 mV to hyperpolarized potentials when cells were held at 0 mV. The kinetic properties of NaChBac were simulated by simultaneous fit of sodium currents at various voltages to a sequential kinetic model. Gating current kinetics predicted from ionic current experiments resembled the experimental data, indicating that gating currents are coupled to activation of NaChBac and confirming the assertion that this channel undergoes several transitions between closed states before channel opening. The results indicate that NaChBac has several closed states with voltage-dependent transitions between them realized by translocation of gating charge that causes activation of the channel.

Purpose – In the face of diverse national and international threats, the purpose of this paper is to explore the transformational leadership challenges in emergency services systems in Canada. These adaptive complex systems respond to critical mass emergencies, disasters and catastrophes. Design/methodology/approach – This qualitative research study uses grounded theory to examine the phenomenology of emergency services leadership from systems perspective. Using the theoretical Wu-Shi-Ren (WSR)-Li systems paradigm, this key informant study of 103 emergency leaders from 81 organizations focusses on the systemic challenges that transformational leaders face in emergency services systems. The response rate was 83.5 percent using a semi-structured and open-ended questionnaire. Findings – This key informant study underscores the competencies for transformational leadership and identifies 12 key leadership challenges in the context of the future evolution of emergency services systems. From the use of a grounded theory methodology, the new theory of transcendental transformational emergency leadership is posited. Research limitations/implications – Qualitative studies such as this key informant study underscores the relevance of the WSR-Li systems paradigm and grounded theory approach in discerning leadership challenges that are specific if not unique to emergency services systems. Practical implications – This study underscores the theoretical and pragmatic implications of the transformational leadership challenges for leadership paradigms, innovation and inter-sectorial collaborative networks and possible future emergency services research. Social implications – This study stresses the importance of addressing leadership challenges of emergency services systems as instrumental in ultimately saving lives, minimizing injuries and assuring complete health and social recovery from the scourges of emergency events nationally and internationally. Originality/value – Qualitative studies of the perspectives of strategic emergency leadership of systemic emergency services systems has not hitherto been done in Canada nor internationally. This key informant study underscores the relevance of the WSR-Li systems paradigm and grounded theory approach in discerning transformational leadership challenges that are specific if not unique to emergency services systems. Moreover, from the use of a grounded theory approach, a new theory of transcendental transformational emergency leadership emerged.

Abstract:
The Red river system is a typical example of Southeast Asian rivers that is strongly impacted by human and climatic conditions, especially in the recent period. In this paper, we aim to investigate the longitudinal variation of the water quality of the Red river, in the section from Hanoi city to the Ba Lat estuary. The sampling campaigns were conducted in the dry seasons in 2017 and 2018. The monitoring results showed that the average concentrations of nutrients (NO2-, NO3-, NH4+, PO43-) were still lower than the allowed values of the Vietnamese standard limits for surface water quality (QCVN 08:2015/BTNMT, column A1) whereas the average concentrations of Cl- and TSS exceeded the allowed values of the QCVN 08:2015/BTNMT, column A1 4.6 and 2.3 times, respectively. NO3- and dissolved silica (DSi) concentrations showed a significant variation from the Hanoi site to the Ba Lat site (6.62 mg/l to 1.19 mg/l for NO3- and 5.21 mg/l to 2.14 mg/l for DSi) whereas SO42-, NO2- and Cl- increased markedly in this longitudinal section, especially from the point SH6 where the salinity started to increase. Based on the three different methods for classification of trophic levels and on the different variables observed during the dry seasons in 2017–2018, the nutrient concentrations of the Red river water tended to slightly increase from the site Hanoi (SH1) to the site SH5 at Nam Dinh, indicating the increase of nutrient external input along the river whereas it tended to decrease from the site SH6 (at mesotrophic/eutrophic level) to the last observed site SH9 (at oligotrophic/mesotrophic level) at the sea due to the dilution of seawater. Seawater in dry season could affect directly the river downstream about 35 km far from the sea. The results may be a guide for planning of water use including agricultural irrigation in the Red river estuary.

Background:IgG4-related disease (IgG4-RD) is an autoimmune disorder and frequently involve multiple organs. The respiratory tract is one of the most frequently involved sites.Objectives:This study aimed to compare the demographic and clinical characteristics of IgG4-related respiratory disease (IgG4-RRD) and non-IgG4-RRD patients in a large cohort.Methods:We carried out a retrospective study of 452 cases of IgG4-RD (104 IgG4-RRD patients and 348 non-IgG4-RRD patients) diagnosed at Peking University People’s Hospital between 2003 and 2020.Results:IgG4-RRD patients had an elder age of disease onset and diagnosis. Multiorgan involvement and hypocomplementemia were more common in IgG4-RRD. Besides, the level of ESR, eosinophilia, IgG and IgG4 were higher in IgG4-RRD patients. In IgG4-RRD group, salivary gland, lacrimal gland, lymph nodes, biliary system and kidney were more commonly involved than those in the non-IgG4-RRD group. Abstract:
Recently, our group has reported a novel Li-ion battery cathode, ‘LT-LiMn0.5Ni0.5O2’, where LT refers to its relatively low synthesis temperature (400 °C). By collecting high energy X-ray diffraction and high-angle, annular-dark-field (HAADF) scanning-transmission electron microscopy (STEM) data, we confirm that the LT-LiMn0.5Ni0.5O2 features a unique partially disordered rock salt structure with predominant lithiated-spinel-like character. The electrochemical data indicates that the Li/LT-LiMn0.5Ni0.5O2 cells shows good cycle stability when operate between 2.5-5.0V. The 1st cycle specific discharge capacity was determined to be 225mAh/g. Two distinct voltage plateaus can be identified at approximately at ~4.6 V and ~3.5 V during discharge. The ~1 voltage separation of the plateaus is caused by the octahedral and tetrahedral site energy difference during lithium extraction and insertion.1 The discovery of LT-LiMn0.5Ni0.5O2 not only expand the compositional space for the known lithated-spinel family but also provide a practical way to increase the operational capacity of traditional spinels such like LiMn2O4 and LiMn1.5Ni0.5O2.2-5 It is well known that when synthesized at 900°C, LiMn0.5Ni0.5O2 features a layered structure.6 The existence of lithated-spinel like LiMn0.5Ni0.5O2 at low temperature (400°C) immediately raise another question: How does the synthesis temperature affect the structure and electrochemical performance of LiMn0.5Ni0.5O2? We synthesized a series of different temperature of LiMn0.5Ni0.5O2 powders. By collecting high energy X-ray diffraction and high-resolution transmission electron microscopy (HR-TEM) data, we confirm that, the LiMn0.5Ni0.5O2 exhibits a lithated spinel → disordered layered → ordered layered structure transformation as temperature increase from 400-900 °C. The Rietveld refinements results also reveals a gradual decrease of the Li/Ni exchange ratio accompanied by the synthesis temperature changes. The evolution of specific discharge capacity for different temperature LiMn0.5Ni0.5O2 shows a parabola-like curve behavior with the lowest discharge capacity at 700 °C. This unique behavior is likely to be related to the diffusion channels reconstruction along with the structure changes. 3D diffusion channels are incorporated with 2D diffusion channels at low temperature (400°C) for lithated-spinel LiMn0.5Ni0.5O2. Within the middle temperature range (500°C-700°C), the 3D channels destroy rapidly while 2D diffusion channels are not well established due to the highly disorder ratio resulting in a decrease of capacity. Abstract Chemotherapy has significantly improved the survival of cancer patients. However, the vast variation in treatment responses remains a major issue in the clinic. It is imperative to understand the molecular mechanisms underlying treatment failure in specific populations of patients. Genetic diversity has been recognized to mainly account for the differences in drug responses among patients. Yet, it is not feasible to map the responsible genetic variants within millions of single-nucleotide polymorphisms in the human genome in clinical settings with a limited number of subjects and insufficient genomic information. Genetically diverse mice are an ideal tool to overcome the limitations in clinical studies and model drug responses in hosts that require genetic susceptibility and resistance factors. Recent studies have also implicated a critical role for host cells (i.e., the tissue microenvironment) in building a protective “niche” for tumor cells enabling their escape from chemotherapeutic treatments. Notably, the host regenerative response upon chemotherapy “injury,” which is regarded as an intrinsic host mechanism to repair damaged tissues, may be exploited by tumor cells for their local recurrence or distant metastases. In this study, using a lung injury model, we determined how drug-induced lung stromal wound healing responses differed among eight inbred mouse strains (BALB/cJ, NSG, C57BL/6J, CH3/HeJ, CBA/J, SJL/J, A/J, and NOD/ShiLtJ). Upon either cisplatin or doxorubicin stimulation, the wound healing-associated genes Il-6, Spp1, Cxcl1, and Ccl2 were all found upregulated in lung stromal cells but showed a great variation across different strains. Furthermore, we performed single-cell RNA sequencing on the lung stromal cells and immune cells isolated from three selected mouse strains (BALB/cJ, C57BL/6J, and NOD/ShiLtJ) without and with doxorubicin treatment in vivo. The transcriptomic analyses revealed strain-specific and non-specific therapy-elicited gene signatures in different types of lung tissue cells. Collectively, our results signify an association between genetic backgrounds and drug-induced host responses in mice. The next steps of the study aim to understand how genetic diversity would functionally impact drug-induced lung tissue regeneration, lung inflammation, and post-therapy tumor relapse or metastasis in the lung. Recently, new types of cation disordered rocksalt (DRS) have been reported which show good reversibility. In our study we combined the strategy of using high-valent cations with partial substitution of fluorine for oxygen anions in disordered rocksalt-structure phase to achieve optimal Mn2+/Mn4+ double-redox reaction in the composition system Li2MnxTi1-xO2F (1/3 ≤ x ≤ 1). we synthesized 4 different compositions (Li2MnIIIO2F, Li2MnII 1/3MnIII 1/3TiIV 1/3O2F, Li2MnII 1/2TiIV 1/2O2F and Li2MnII 1/3TiIII 1/3TiIV 1/3O2F). Two of them were synthesized for the first time, Li2MnII 1/3MnIII 1/3TiIV 1/3O2F and Li2Mn II 1/3TiIII 1/3TiIV 1/3O2F. By studying the electrochemical properties of different compounds we found that Ti+4 in the structure keeps Mn at the second state of charge, thus enabling a double redox reaction of Mn2+/Mn4+. By investigating the electrochemical properties of all samples we found that the sample with the composition Li2Mn2/3Ti1/3O2F showed the best electrochemical properties with initial high discharge capacity of 227 mAh g-1 in the voltage window of 1.5-4.3 V and 82% of capacity retentionafter 100 cycles. However, fluorination might lead to several issues such as synthesis limitation, lithium diffusion issues due to preferable strong Li-F bonds, etc. thus, two more different samples based on the Li2Mn2/3Ti1/3O2F composition were synthesized and their properties were investigated (Li1.5MnII 1/3MnIII 1/3TiIV 1/3O2F0.5 and Li1.25MnII 1/3MnIII 1/3TiIV 1/3O2F0.25) in order to find the proper amount of fluorine in the structure which promises the electrochemical behavior. In the following the effect of fluorine on lithium diffusion was investigated by ex-situ Raman studies. These studies shed light on the diffusion pathways of lithium ions during charge and discharge process. The structural characteristics are examined using X-ray diffraction patterns, Rietveld refinement, energy-dispersive X-ray spectroscopy and scanning electron microscopy, transmission electron microscopy and Raman spectroscopy. The oxidation states and charge transfer mechanism are also studied further using extended X-ray absorption fine structure and X-ray photoelectron spectroscopy in which the results approve the double redox mechanism of Mn2+/Mn4+ in agreement with Mn-Ti structural charge compensation. The findings pave the way for designing high capacity electrode materials with multi-electron redox reactions. Tin (Sn) is a metal that is commonly used in our daily life. With the rapid development of lithium-ion battery in the past decades, Sn and its alloy, such as Sn-Cu[1], and Sn-Ni[2], Sn-Co[3] and Sn-Fe[4] have been used as anode for lithium-ion battery because they can undergo alloying/dealloying process with lithium ions and exhibit high capacity and suitable working voltage of about 0.4 V vs. Li/Li+. Sn can also undergo an oxidation reaction to Sn2+, with an electrode potential of about -0.14 V vs. SHE. It is therefore possible to also use Sn as a cathode material. Herein, we are first to demonstrate a metal-metal battery made up of Sn metal as the cathode and Li metal as the anode in organic electrolyte (see Fig. a). Sn foil and Li foil are simply assembled with 3M LiTFSI in dimethoxyethane/propylene carbonate (DME/PC) electrolyte in an Ar-filled glove box to form a pouch cell. During charging, Sn will give out two electrons and dissolves into the electrolyte as Sn2+, while during discharging, the metal ions will be re-deposited onto the cathode. Thus, the energy is stored in the form of Sn2+ in the electrolyte. The charge-discharge curves in Fig. b show that the operating voltage of the battery is about 2.8 V. Since Sn2+ that is dissolved into the electrolyte from the cathode has higher potential than the Li metal anode, any Sn2+ ions cross-over to the anode will be spontaneously reduced, decreasing the efficiency of the battery. To suppress such self-discharge process, an anion exchange membrane based on poly(ionic liquid) polymer coated on common polypropylene separator is adopted. The Sn-Li battery with the modified separator tested in a current rate of 0.2 mA cm-2 with a capacity limitation of 0.1 mAh cm-2 gives an average Coulombic efficiency about 99.5% and can be cycled for more than 1500 cycles(See Fig. c). We found that the stripping/deposition of Sn on the cathode, and its polarization depend strongly on the type of electrolyte used. With 3M LiTFSI in DME/PC electrolyte, the discharge voltage is lowered by about 0.05 V when the current density is increased from 0.2 mA cm-2 to 1 mA cm-2. Rare earth elements (REE) are important raw materials for electronic products and green-energy technology and deep-sea sediment as an important potential source of REE mineral. It is of great scientific significance to understand the influence of REE in deep-sea sediments. In this paper, thirty sediments samples and eleven sediments samples were collected at the site SH3 and SH1 of Shenhu area, respectively. The trace elements, major elements, and different Fe-Mn minerals were analyzed to investigate the role of anaerobic oxidation of methane (AOM) in the enrichment of rare earth elements in the marine sediments. The result showed that the content of ∑REE/Ti increased at the top of sulfate and methane transition zone (SMTZ) at site SH1 and SH3. Combined with the analysis of different forms of ferromanganese minerals and the REE patterns, it is inferred that there is little influence of terrigenous input to the REE content. In contrary, the REE-rich carbonate phase and REE-rich Fe-Mn oxides (hydroxides) phase have important contribution of REE enrichment at the top of SMTZ. Meanwhile, the calculations of the quantitative relationship between the REE-rich carbonate phase and REE-rich Fe-Mn oxides (hydroxides) phase also confirm this conclusion. And the AOM caused the decrease of the REE-rich Fe-Mn oxides (hydroxides) phase and increase of the REE-rich carbonate phase. This is the potential explanation of ∑REE/Ti enrichment at the top of SMTZ. This study is of great scientific significance for the study of REE enrichment mechanism in the methane seeps and provided a new sight of REE enrichment mechanism in deep-sea sediments.

Climate change has stimulated great interest in developing clean and renewable energy conversion and storage solutions. Emerging technologies for solutions, like unitized regenerative fuel cells, metal-air batteries, and water electrolyzers, depend on the reliable catalyst materials suited for long-term application in alkaline environments[1]. In particular, the demanding reaction in these systems is the oxygen evolution reaction involving complex multielectron/proton transfer processes and thus sluggish kinetic.[2] At present, the state-of-the-art OER catalysts are IrO2 and RuO2. However, the high cost and scarcity of these materials severely prevent the wide-scale application of these systems.[3] Perovskite oxide (e.g., single perovskite expressed as ABO3) electrocatalysts are particularly considered next-generation OER catalysts due to their adjustable physicochemical properties and, as a consequence, their catalytic properties by substitution of ions in the A and B sites.[4] Via doping both at A and B site, double perovskite oxides structure can form (AA′B2O5+δ), which were shown to have stable structure during the OER due to proper O p-band center position relative to the Fermi level. The previous research shows that hexagonal perovskites exhibit enhanced catalytic activity rather than cubic or tetragonal perovskites, which is associated with the face-sharing octahedral unit. [5,6] In this work, we systematically investigate doping of parent La2CoMnO6 with Ba to tune the crystal structure and electronic structure. A series of the double perovskites with the chemical formula of La2-xBaxCoMnO6 (x = 0, 0.5, 1.5, 2) were fabricated via the modified Sol-Gel Pechini method. According to Goldschmidt tolerance factor, it is expected that when Ba amount increases in the A site, the crystal structure changes from cubic to hexagonal, which can contribute to electrocatalytic OER activity based above discussions. Furthermore, it is anticipated that increasing Ba content at the A site should result in higher valence state of transition metals due to stabilizing charge balance of the structure. It is widely reported that presence of higher valence state of transition metals is beneficial for OER. The crystal structure was confirmed by combining powder X-ray diffraction (XRD) patterns and selected area electron diffraction (SAED) patterns. The two-layer hexagonal structure of Ba2CoMnO6 (BCM) was further characterized by high-angle annular dark-field scanning transmission electron microscopy (HAADF-STEM). The results show doping of Ba into parent La2CoMnO6 resulted in the hexagonal phase formation and BCM achieves a current density of 10 mA cm–2 at a low overpotential of 288 mV and has the highest intrinsic activity in the series of BaxLa2-xCoMnO6 (x = 0, 0.5, 1, 1.5, 2). Furthermore, BCM exhibits outstanding stability of 12 h in a 0.1 M KOH electrolyte. The electronic structure and surface work function values of the catalysts were examined with X-ray photoelectron spectroscopy (XPS). XPS analysis revealed that valence state of Co and Mn increases through Ba amount in the structure, indicating the double perovskite oxide charge balance is maintained. In addition to valence state analysis, we performed work function analysis of the catalysts tested in this work. The result shows work function of the series of BaxLa2-xCoMnO6 (x = 0, 0.5, 1, 1.5, 2) substantially decreases with Ba content. Here, we show that the high intrinsic activity of BCM is not only related with the hexagonal structure and also may be related with exhibiting lower work function value in the series. Abstract Approximately 15% of all patients with non-small cell lung cancer (NSCLC) and nearly 35% of Asian patients with NSCLC harbor activating mutations within the epidermal growth factor receptor (EGFR). Although these patients are initially highly sensitive to first or second generation EGFR tyrosine kinase inhibitors (TKIs) including erlotinib or third-generation inhibitors including osimertinib, EGFR TKI-refractory disease inevitably emerges. While therapeutic strategies to target resistant disease that emerges though secondary EGFR mutations or MET amplification have been developed, there remains a void of therapeutic options for patients where resistance occurs through EGFR-independent mechanism such as epithelial to mesenchymal transition (EMT) or transformation to small cell lung cancer (SCLC). To identify cell surface proteins that could be targeted by antibody-based or adoptive cell therapy approaches we interrogated RNAseq data from EGFR mutant NSCLC cell lines (HCC827 and HCC4006) and their associated EGFR TKI resistant variants previously shown to have developed resistance through EMT and filtered gene expression data to include only genes which transcribed proteins localized to the cell surface. We identified CD70 as a being highly upregulated in EGFR TKI resistant cells (p = 7.2e-42). Given that CD70 expression is highly restricted and only transiently expressed on immune cells, CD70 was selected as a top candidate cell surface protein for targeting studies. Western blotting and flow cytometry analysis confirmed CD70 protein levels to be highly upregulated in EGFR TKI resistant cells that had undergone EMT but not in cells harboring secondary EGFR mutations or MET amplifications. We also observed CD70 upregulation in osimertinib-treated drug tolerant persister cells, indicating that CD70 upregulation is an early event in the evolution of TKI resistance. Moreover, patient-derived models of acquired EGFR TKI resistance also exhibited CD70 positivity. Our data also indicated that in EGFR mutant NSCLC cells, CD70 could be upregulated through decreased CD70 promoter methylation as well as by the EMT regulators, transforming growth factor-β (TGF-β) and ZEB1, both of which were upregulated in TKI resistant cells. In EGFR TKI resistant cells, CD70 knockdown impaired cell viability and invasiveness, and stimulation of CD70 using the exogenous binding partner CD27 resulted in activation of AKT and MAPK, pathways known to be re-activated with acquired TKI resistance. CD70-targeting approaches including anti-CD70 antibody drug conjugates (ADCs) and CD70-targeting CAR T cell and CAR NK cells showed promising in vitro and in vivo activity against CD70 positive tumor cells and in osimertinib drug-tolerant persister cells. These results identify CD70 as a novel therapeutic target for EGFR mutant tumors with acquired EGFR TKI resistance that merits further investigation in the clinic. Citation Format: Monique B. Novel Coronavirus 2019 (covid-19) is a generation of the coronavirus that humans have never identified. This virus was first reported to cause an outbreak in Wuhan, China, in December 2019. Until now, covid-19 infection cases have been reported in many countries, such as Thailand, Japan, South Korea, Iran, Italy, Germany, and more than 90 countries worldwide. Until November 1, 2020, a total of 412.784 positive cases of covid-19 were recorded, with 341.942 recovered patients and 13.943 deaths in Indonesia. The purpose of this event is to anticipate the spread of Covid-19 by reminding the public to maintain health protocols and other alternatives in preserving health, also increasing body immunity through massage and traditional Indonesian herbal medicine. This event received a good response from the public. It was evidenced by the number of participants exceeding the predetermined quota and enthusiastic in following the speaker's material. The event was a success and was well received by the participants as well as the local government. Abstract:
The novelty of the study is that the ordering that occurs in a PET film under the action of SHI irradiation manifests itself as an increase in the integral intensity of intrinsic luminescence. The Urbach behaviour of the red shift of the absorption edge is used as a baseline for further analysis of experimental optical transmission spectra of PET films irradiated by swift heavy ions (SHI) previously published by the authors. Negative deviations of the experimental spectra from the Urbach baseline in the visible and UV parts of the spectrum are attributed to enhanced by SHI irradiation intrinsic luminescence. The observed dependence of the integral intensity of luminescence of irradiated PET films on the SHI fluence and ion charge provides further confirmation of the presence of SHI-induced ordering of the molecular structure in SHI latent tracks.

The work is devoted to development of a smart lighting system that is able to change correlated colour temperature and consists of tunable 4-components RGBW clusters controlled via three channels. It is shown that fixing a ratio between the intensities of white and red LEDs at a level of 100:9 enables obtaining high values of colour rendering in a wide range of correlated colour temperatures. SRC family kinases (SFKs) function in initiating Ca2+release at fertilization in several species in the vertebrate evolutionary line, but whether they play a similar role in mammalian fertilization has been uncertain. We investigated this question by first determining which SFK proteins are expressed in mouse eggs, and then measuring Ca2+release at fertilization in the presence of dominant negative inhibitors. FYN and YES proteins were found in mouse eggs, but other SFKs were not detected; based on this, we injected mouse eggs with a mixture of FYN and YES Src homology 2 (SH2) domains. These SH2 domains were effective inhibitors of Ca2+release at fertilization in starfish eggs, but did not inhibit Ca2+release at fertilization in mouse eggs. Thus the mechanism by which sperm initiate Ca2+release in mouse eggs does not depend on SH2 domain-mediated activation of an SFK. We also tested the small molecule SFK inhibitor SU6656, and found that it became compartmentalized in the egg cytoplasm, thus suggesting caution in the use of this inhibitor. Our findings indicate that although the initiation of Ca2+release at fertilization of mammalian eggs occurs by a pathway that has many similarities to that in evolutionarily earlier animal groups, the requirement for SH2 domain-mediated activation of an SFK is not conserved.

Fyn and other Src-family kinases play an essential role at several steps during egg activation following fertilization of externally fertilizing species, such as marine invertebrates, fish, and frogs. Recent studies demonstrate that the requirement for Src-family kinases in activation of the mammalian egg is different from lower species, and the objective of this study was to test the role of the Fyn kinase in the mouse egg activated by intracytoplasmic sperm injection (ICSI). An Src homology 2 (SH2) domain containing fusion protein was used to suppress Fyn function in the mouse zygote following ICSI. Eggs injected with the Fyn SH2 domain at an intracellular concentration of 4–8 μM exhibited reduced developmental potential with 100% of the zygotes being arrested following the first or the second cleavage. At higher concentrations, the protein blocked pronuclear congression and the zygotes remained at the pronuclear stage. The SH2 domain had no effect on sperm-induced calcium oscillations in distinct contrast to its effect on the eggs of lower species. The results indicate that the SH2 domain of Fyn kinase plays an important role in pronuclear congression as well as early cleavage events and that this effect appears not to involve disruption of calcium oscillations.

Abstract Abstract SCI-46 Blood transfusion, the earliest form of cell replacement therapy, has become indispensable for modern medicine making the safety and adequacy of the blood supply a national priority. The US blood supply is adequate overall because in 2006 the number of blood units collected exceed by 7.8% the number of those transfused. However, issues surrounding blood transfusion, such as sporadic shortages and potential adverse events to recipients (related to changes in red cell physiology during storage and alloimmunization in chronically transfused patients) prompted past and current efforts to develop alternative transfusion products. Recently, the culture conditions to generate erythroid cells have greatly improved making the production of a transfusion product ex-vivo a theoretically possible, although expensive, proposition. This recognition is inspiring several investigators to develop production processes for ex-vivo generation of red cell transfusion products. A proof-of-concept demonstrating that ex-vivo generated red cells protect mice from experimentally induced lethal anemia has been obtained. Alternative sources of stem cells which include human embryonic stem cells (hESC) and induced pluripotency stem cells (iPS), are being explored. Since red cells do not have a nucleus, safety considerations suggest that they may represent the first cell therapy product to be generated from hESC and iPS. In addition, discarded hematopoietic stem cells present in adult and cord blood donations may theoretically generate numbers of red cells ex-vivo sufficient for transfusion. Affordable clinical grade humanized culture media have also been developed. Possible differences in immunological and biological properties of erythroid cells from different sources are under investigation. These differences include size, levels of activity of glycolytic enzymes and carbonic anhydrase, expression of different isozymes, hemoglobin and antigenic profiles (HLA class II antigens). This last aspect is particularly important because ex-vivo expanded red cells pose the same risk for infection and incompatibility as any transfusion product but pose unique antigenic risks. Since expression of blood group antigens is susceptible to post-transcriptional modifications, the ex-vivo expansion process itself may induce antigenic variability. Therefore, even cells generated from completely matched stem cell sources may induce auto-immunity and/or appear incompatible. Regarding the identity of ex-vivo generated red cell transfusion products, a conservative approach would be to define them as “enucleated red cells”. In principle, however, ex-vivo generated erythroblasts may also serve as transfusion product. Since they undergo 4–64 further divisions and reduce iron overload, they may represent a more potent transfusion product for patients that require chronic transfusion. The clinical use of these cells, however, may involve development of specific procedures to facilitate their homing/maturation in the erythroid niches of the recipients. In summary, on the basis of these cost, logistic and safety considerations we hypothesize that the clinical application of ex-vivo expanded erythroblasts will involve in sequence, drug discovery for personalized therapy, systemic drug delivery, genotypically matched transfusion for alloimmunized patients and then transfusion in the general population. Abstract Abstract SCI-24 Erythroid precursors in the bone marrow require transferrin-bound iron for hemoglobin synthesis. Therefore, it is not surprising that the regulation of erythropoiesis and iron metabolism is interlinked. Iron demand for erythropoiesis is communicated to the iron-regulatory machinery through incompletely understood mechanisms. At the core of systemic iron homeostasis is the peptide hormone hepcidin, restricting cellular iron export to plasma by inducing the endocytosis and proteolysis of ferroportin. Hepcidin, a liver-synthesized peptide hormone, is released in response to increased iron load, and there is early evidence that circulating hepcidin concentrations affect the distribution of iron between the macrophage storage compartment (favored by higher hepcidin concentrations) and parenchymal cells, including cardiac myocytes and hepatocytes (favored by low hepcidin). Furthermore, ferroportin has recently been identified on erythroid precursors. Its purpose in this cell type and its function in the interface between erythropoiesis and iron metabolism are unclear. Additionally, in response to bleeding or the administration of erythropoietin, expansion of erythroid precursors suppresses hepcidin, most likely through one or more mediators released by the bone marrow and acting on hepatocytes. Iron-loading anemias with ineffective erythropoiesis, in particular β-thalassemia, demonstrate the effects of pathological “erythroid regulators” of hepcidin. Although erythrocyte transfusions are the main cause of iron loading in patients who receive them (β-thalassemia major), lethal iron overload is seen also in patients who are rarely or never transfused (β-thalassemia intermedia). Here, iron hyperabsorption is the cause of iron overload and, as in hereditary hemochromatosis, is caused by low hepcidin. Decreased hepcidin expression in β-thalassemia, with concurrent ineffective erythropoiesis and iron overload, indicates that the “erythroid regulator” may play an even more substantial role in iron metabolism than the “stores regulator.” Two members of the bone morphogenetic protein (BMP) family, growth differentiation factor (GDF) 15 and Twisted Gastrulation (TWSG1), have been implicated as candidate bone marrow-derived hepcidin suppressors in β-thalassemia. Neither factor is responsible for the physiologic hepcidin suppression in response to hemorrhage-induced stress erythropoiesis, and the physiologic suppressor is not known. We focus here on the current state of knowledge regarding the regulation of iron metabolism and attempt to elucidate the interface between iron regulation and erythropoiesis using evidence in part derived from animal models of β-thalassemia. A more complete understanding of the coregulation of erythropoiesis and iron metabolism may lay the foundation for improving diagnosis, increasing treatment options, and ultimately impacting the well-being of patients afflicted with different anemias and/or iron overload related-disorders. Until recently it has been assumed that red blood cells (RBCs) play mainly a passive role in hemostasis and thrombosis, but now some studies have uncovered several novel roles for them in both of these processes. The best-known effects of RBCs in clotting in vivo are rheological, involving laminar shearing plus aggregation and deformability of RBCs. In addition, RBCs have specific receptors for platelets and fibrin(ogen) that may be involved in thrombosis. Furthermore, in the contraction of blood clots, the trapping of RBCs in the platelet-fibrin network and subsequent physical changes may be important for hemostasis and thrombosis. Contracted blood clots develop a remarkable structure, with a meshwork of fibrin and platelet aggregates on the exterior of the clot and a close-packed, tessellated array of compressed polyhedral RBCs within. The structure and properties of contracted clots vary depending on the relative amounts of platelets, fibrinogen and RBCs and the conditions of clotting. Such close-packed arrays of polyhedral erythrocytes, or polyhedrocytes, have also been observed in human arterial and especially venous thrombi taken from patients. The mechanical nature of this RBC shape change was confirmed by polyhedrocyte formation from the forces of centrifugation of blood without clotting. Many experiments suggest that polyhedrocytes are more prevalent under conditions associated with thrombosis and could be a marker of prothrombotic conditions. Consistent with this hypothesis, RBC membranes have been found to be a major source of exposure of phosphatidylserine, which forms a procoagulant surface to generate thrombin. Platelets (with their cytoskeletal motility proteins) and fibrin(ogen) (as the substrate bridging platelets for contraction) are required to generate the forces necessary to segregate platelets/fibrin from RBCs and to compress these cells into a tightly packed array. Clots formed after puncture of the mouse saphenous vein are composed primarily of polyhedrocytes, suggesting that they may be important in venous hemostasis. Moreover, the permeability of contracted clots is nearly the same as that of the endothelial cell lining, consistent with their functional significance. These results demonstrate how contracted clots form an impermeable barrier important for hemostasis and wound healing and also to restore flow past obstructive thrombi. On the other hand, they also help explain how fibrinolysis is greatly retarded after clots contract, as a result of these striking structural changes. In summary, RBCs may perform a dual role, both helping to stem bleeding but at the same time contributing to thrombosis in a variety of ways, including when the tightly packed array of RBCs in a contracted clot blocks blood flow and resists dissolution.
