Abstract:
A malária é transmitida por mosquitos que, preferencialmente, picam no interior (endofágicos) ou no exterior (exofágicos) das habitações. Atualmente, os métodos mais sucedidos no controlo vectorial são a pulverização residual intra-domiciliar (PIDOM) e as redes mosquiteiras tratadas com insecticidas de longa duração (REMILD), que têm como alvo principal os vectores endofágicos. No entanto, as espécies que picam no exterior (exófagicas) têm sido as mais difíceis de controlar e, atualmente, não existem métodos satisfatórios para o controlo de populações destes vectores. A transmissão da malária feita por vectores exofágicos é comum fora de África; contudo, o fenómeno vem, progressivamente, ganhando importância também no continente Africano. Daí que, para que os vectores exofágicos sejam controlados de forma efetiva, é necessário entender melhor como ou porquê alguns vectores preferem procurar refeições sanguíneas em ambientes fechados, enquanto os outros evitam entrar em tais compartimentos? Ou seja, como é que um vector localiza e reconhece um ambiente fechado (ou intra-domiciliar) ou aberto (extra-domiciliar)? Com vista a obter respostas à tais questões, varias experiências de campo foram realizadas na aldeia de Massavasse, localizada na região leste do distrito de Chóckwè, província de Gaza, sul de Moçambique. As experiências assentaram-se em dois objetivos principais: 1. Projetar, construir e avaliar a performance de uma armadilha de grelha elétrica, apelidada de armadilha Shock-wè (SHK-wè), que poderá ser utilizada como uma alternativa fiável às colheitas com isca humana, na amostragem de vectores da malária que procuram um hospedeiro humano em ambientes interiores ou exteriores. 2. Determinar a contribuição dos elementos estruturais básicos que compõe uma habitação (ou seja, tecto, paredes, pilares, etc.) para a ocorrência e entrada de mosquitos que procuram uma refeição sanguínea em casas experimentais. Para cumprir o primeiro objectivo, foram feitas colheitas emparelhadas de mosquitos através de armadilhas SHK-wè e colheitas com isca humana (CIH) durante 35 noites consecutivas. Os resultados indicam que a SHK-wè é uma ferramenta segura e fiável para a amostragem e vigilância das populações de vetores importantes da malária em África. SHK-wè mostrou um bom desempenho tanto no interior como no exterior e, portanto, é potencialmente um substituto prático, robusto e seguro para as capturas CIH convencionais. Para cumprir o segundo objetivo, foram igualmente utilizadas armadilhas SHK-wè numa série de experiências de tratamentos randomizados para determinar a resposta das populações de mosquitos locais à presença de componentes estruturais de uma casa experimental construída para o efeito. A experiência foi realizada durante 50 noites consecutivas e constituiu em montagens e desmontagens aleatórias da casa experimental nos seus elementos básicos (isto é, o suporte de metal, paredes inferiores e superiores e o tecto). Os resultados desta experiência sugerem que, contrariamente ao que se conhecia, o tecto da casa é um componente chave para o reconhecimento e entrada dos vectores endofágicos do complexo A. gambiae s.l em ambientes intra-domiciliares. Os dados indicaram que a probabilidade de A. gambiae s.l. entrar numa casa experimental aumenta 4.5 vezes [IRR = 1.50 (0.63 – 2.37; p = 0.001) quando o tecto está presente em comparação com outros tipos de tratamentos
Malaria is, preferentially, transmitted by mosquitoes that bite indoors or outdoors. To date, the most successful vector control methods, namely: indoors residual spraying (IRS) and long lasting insecticide treated bed nets (LLINs) have targeted indoor species. Outdoor biters (exophagic) are difficult to target and no satisfactory control methods exist. Already common outside Africa, outdoor-transmitted malaria is growing in importance within Africa. To target exophagic mosquitoes we need to understand more how/why they choose to enter or to avoid houses, i.e. what do mosquitoes perceive as indoors or outdoors? Field experiments were undertaken in Massavasse, a village located in the eastern region of Chóckwè district, Gaza province, southern Mozambique, to investigate that question and addressing two objectives: 1. To design, build and evaluate an electric net trap (ENT), named the “Shock-wè (SHK-wè) trap” for reliable sampling of mosquitoes responding to human bait, indoors and outdoors; 2. To determine the contribution of simple elements of a house (i.e. roof, walls, partial walls) to occurrence and entry rates of host seeking mosquitoes. To accomplish the first objective, a series of paired field tests were performed to evaluate the performance of SHK-wè trap compared to Human landing catch (HLC) method. Results showed that SHK-wè trap is a safe and reliable sampling and surveillance tool for African malaria vector populations. The trap performed well indoors and outdoors and is potentially a robust and safe practical replacement for the conventional HLC method. To accomplish the second objective, SHK-wè trap was used in a series of randomized treatment-assigning experiments to determine the response of local mosquitoes populations to presence of structural components of a purpose-built experimental house. The experiment consisted on randomly dismantling or reassembling an experimental house into its basic elements (i.e. a frame on a base, lower walls, upper walls and roof). Results from this experiment suggest that, contrarily to current belief, the roof of the house was a key component that triggered endophagic malaria vectors, such as members of A. gambiae complex, to enter. Data indicated that the probability of A. gambiae s.l. entering an experimental house increased 4.5 [IRR = 1.50 (0.63 – 2.37); p = 0.001)] times when the roof was in place compared to other types of treatments.

Abstract:
Human - Technology - Organization (HTO) is a well-established, general unifying concept in the Swedish Accident Investigation Authority (SHK) that represents an approach, knowledge and use of various tools regarding interactions between people, technology and organizational factors. The HTO-perspective is well described in literature but there are few studies on how SHKs staff experiences working according to this method in their investigations. The aim of this study was therefore to describe their HTO-perspective, examine how it is used in the investigations at SHK and describe the investigators experience of working with the HTO-perspective as well as the method's usefulness compared to old methods in accident investigations. A literature study has been conducted in the areas of HTO, Theory of planned behavior (TPB) and safety culture. TPB and safety culture are described in this paper since they highlight the different aspects of a HTO-perspective. The hypothesis was answered by using semi-structured interviews. The interviews were analyzed by a content analysis and themes were identified. Furthermore the material from the interviews was subsequently structured through a Strength – Weakness – Opportunities – Threats analysis (SWOT), i.e. the informants' view on the HTO-perspective was structured based on the strengths, weaknesses, opportunities and threats SHKs staff experience that the method has in the investigative work. The informants consisted of two investigators at SHK who 3have worked with accident investigations for a long time in various roles. The results yielded that the investigators had a positive attitude towards conducting investigations according to the HTO-perspective because they feel that this perspective provides them with cross-competence. A flaw is though that the perspective lacks a structured approach. As a result of this, the investigators own experiences and expertise play a major role in the quality of the investigation. As the study's aim is met and the informants are considered to be experts the validity requirements are also fulfilled.

Abstract:
A. A study of Stichodactyla helianthus toxin’s potential to penetrate the buccal mucosa Introduction:  Buccal mucosa is an alternative route for drug administration and has advantages over other conventional routes by avoiding both enzymes in the gastro intestinal system and the hepatic first passage mechanism. Stichodactyla helianthus toxin (ShK) is a peptide toxin that blocks potassium channels in T lymphocytes and could be a future treatment for autoimmune diseases when finding a suitable way of administration. Aim:  The purpose of this part of the study was to develop a robust and reproducible assay for identification and quantification of ShK. The method was then employed for a proof of principle study; determining the concentration of ShK following an in vitro permeability experiment, to evaluate the potential of ShK penetrating the buccal mucosa in porcine tissue.  Materials and Methods:  An HPLC method was developed and validated. A piece of porcine buccal mucosa was used as a membrane because of its similarities with human buccal mucosa, and cinched in between a modified Ussing Chamber consisting of a donor and a receptor chamber. Samples were withdrawn from the receptor chamber to determine the amount of ShK that had penetrated the membrane. Results: The HPLC method developed for quantification of ShK demonstrated high accuracy and precision. No concentrations of ShK were able to be quantified from the receptor chambers. Conclusions:  A robust assay for quantification of ShK was developed but the results from the experiment indicated that ShK could not penetrate the buccal mucosa membrane. B. An investigation of the permeability alterations in the blood-brain barrier associated with Alzheimer’s disease   Introduction:  The blood brain barrier (BBB) protects the brain from potential dangerous substances by different barrier properties such as tight junctions and efflux transporters such as P-glycoprotein. Previous studies have showed that the barrier functions may be altered in Alzheimer’s disease and thereby increase the exposure to substances that are normally excluded from the brain parenchyma. This could be an issue regarding safety and toxicity of medications used among Alzheimer patients. Aim:  The aim of this part of the study was to investigate the difference in brain uptake of verapamil, digoxin, loperamide, propanolol, diazepam and sucrose between 3xTg-AD mice and wild type control mice. Materials and Methods: Female 3xTg-AD mice and control mice of the age 11.5-13.5 months were used. In Situ brain perfusion with radiolabeled substances (n=5-12) was performed and the brain uptake ratio of the substances was compared and statistically analyzed.  Results: No difference in the vascular volume was found when comparing 3xTg-AD with control mice. The ratio of diazepam was observed to be higher in the cortex and propranolol higher in the hippocampus, of 3xTg-AD mice. The uptake ratio of verapamil was higher in both the hippocampus and cortex of 3xTg-AD mice whereas digoxin appeared to be lower in the cortex of 3xTg-AD mice. There was no difference in uptake ratio of loperamide between 3xTg-AD and control mice. Conclusions:  This study in addition to previously executed studies in our laboratory, showed that the membrane thickness is age dependent in 3xTg-AD and that further studies needs to be conducted on the expression of P-glycoprotein in the BBB in 3xTg-AD and control mice.

In this experiment, fluorescent N- (1-pyrenyl) iodoacetamide modified the two reactive thiols, SH1 (Cys 707) and SH2 (Cys 697) on myosin to detect SH1-SH2 a -helix melting. The excimer forming property of pyrene is well suited to monitor the dynamics of the SH1 and SH2 helix melting, since the excimer should only form during the melted state. Decreased anisotropy of the excimer relative to the monomeric pyrene fluorescence is consistent with the disordering of the melted SH1-SH2 region in the atomic model. Furthermore, nucleotide analogs induced changes in the anisotropy of the excimer, suggesting that the nucleotide site modulates the flexibility of SH1-SH2 region.

L'interleukine-2 (IL-2) est produite par les lymphocytes T activés et joue un rôle crucial dans les processus de prolifération, de différentiation en cellules effectrices et d'apoptose secondaire à l'activation. Ses fonctions biologiques reposent sur son interaction avec un récepteur membranaire composé de trois chaînes α, β et γc, dont seules β et γ sont nécessaires à la signalisation intracellulaire. Nous nous sommes intéressés à la signalisation précoce en réponse à l'IL-2 dans des lymphocytes T humains, et en particulier au rôle de la protéine adaptatrice plate-forme Gab2. Nous avons montré que cette protéine est tyrosine phosphorylée en réponse à l'IL-2 et qu'elle s'associe à différents partenaires possédant des domaines SH2 comme la tyrosine phosphatase SHP-2, la lipide kinase PI3K et la protéine adaptatrice CrkL. Nous avons choisi d'étudier plus particulièrement l'interaction Gab2/SHP-2. Nous avons montré que le recrutement et l'activation de SHP-2 via son interaction avec Gab2 sont nécessaires en amont de MEK pour l'activation de la voie Ras/ERK MAPK et permettent à SHP-2 une action en parallèle de la voie RhoA/SRF dans la régulation de l'expression du gène c-fos impliqué dans la prolifération cellulaire. Nous avons également montré que ERK est capable de phosphoryler Gab2 sur sa sérine 623. Cette phosphorylation entraîne une diminution de l'interaction Gab2/SHP-2 qui se traduit par une participation à l'inhibition de la voie ERK et une levée de l'inhibition exercée par l'activité phosphatase de SHP-2 sur STAT5, ce qui module la prolifération des lymphocytes. Ces résultats suggèrent l'implication du complexe Gab2/SHP-2 dans la régulation fine du processus de prolifération cellulaire en réponse à l'IL-2. Ces résultats permettent de mieux comprendre les mécanismes de contrôle de la prolifération des cellules leucémiques humaines et identifient la protéine Gab2 comme un élément régulateur central des voies de signalisation activées par l'IL-2
Interleukin-2 (IL-2) is a major growth factor for T lymphocytes which plays a critical role in cellular proliferation, differentiation and activation-induced cell death. The effects of IL-2 on its target cells are mediated through specific cell-surface receptors, which comprise three subunits α, β and γc. Only β and γc are required to trigger downstream signalling events. We were interested in early signalling events in response to IL-2 in human T lymphocytes and have studied the role of scaffolding adaptor protein Gab2. We report here that Gab2 becomes tyrosine-phosphorylated in response to IL-2 and forms multimolecular complexes with SH2 domain-containing partners such as the tyrosine phosphatase SHP-2, the lipid kinase PI3K and the adaptor protein CrkL. 8ince the role of SHP-2 in IL-2 signalling has not been fully elucidated, we investigated in details the function of Gab2/SHP-2 interaction. We have shown here that IL-2 activates SHP-2 via Gab2. Furthermore, SHP-2 appears to be required upstream of MEK in activation of the Ras/ERK pathway and in parallel to the RhoA/SRF pathway for c-fos promoter regulation and therefore cellular proliferation. Moreover, we have shown that ERK phosphorylates Gab2 on a consensus phosphorylation site, Ser 623. This phosphorylation downregulates the interaction between Gab2 and SRP-2 and consequently the activity of SHP-2, which modulates ERK and STAT5 activation. So, at the initiation of the response, Gab2-activated SHP-2 is required for ERK activation. At later time points, secondary to ERK-mediated phosphorylation of Gab2 S-623, decreased SHP-2 activity helps down-regulating ERK, and allows for STAT5 activation to persist. These results demonstrate that ERK-mediated phosphorylation of Gab2 S623 is involved in fine tuning the proliferative response of T-lymphocytes to IL-2. These results provide new insight on the control of human leukemic cell proliferation and on the critical role of Gab2 in IL-2 signal transduction

Abstract:
The adaptor protein CT10-Regulator of Kinase (CRK) and the closely related CRK-Like (CRKL) are adaptor proteins that play important roles in many signaling pathways regulating cell proliferation and cell motility. A notable example is their required role in Reelin signaling during development of the laminated structures of the vertebrate central nervous system, including the cerebral cortex, cerebellum, hippocampus, and retina. As adaptors, CRK/CRKL are important in coupling phosphotyrosine signaling to G protein activity to regulate both cell proliferation and changes in the actin cytoskeleton, thereby exerting control over cell motility, and migration. While many proteins that interact with CRK/CRKL have been identified, the diverse roles of these molecules suggest that more remain to be found. Herein is described a novel CRK/CRKL interacting protein, Src Homology 2 domain-containing protein D (SHD), which demonstrates a phosphorylation-dependent interaction with the CRK/CRKL SH2 domain in HEK 293 cells stimulated with hydrogen peroxide, which globally boosts tyrosine phosphorylation by inhibiting tyrosine phosphatases. Treatment with an inhibitor for Src family kinases (SFKs), Src-1, or an inhibitor of Abl/Arg kinases, STI571, reduces peroxide-induced binding of the CRKL-SH2 domain to SHD. We show that overexpression of Abl kinase, but not the SFK Fyn is sufficient to induce binding of the CRKL-SH2 to SHD and that this interaction requires at least one of the five tyrosines in YxxP motifs found in SHD. Using mass spectrometry, we found that Abl phosphorylates SHD on Y144, which is located in a YxxP motif. Mutation of this site to phenylalanine reduces, but does not prevent, Abl-induced binding of SHD to the CRKL-SH2 domain, suggesting that other YxxP sites also facilitate the interaction. A discussion of the cellular consequences of the interaction between SHD and CRK/CRKL is presented. To explore the biological role of SHD, we used the zebrafish to study shdb, a putative ortholog of human SHD. The expression of shdb was unknown and so we performed in situ hybridization and determined that shdb was expressed in the developing nervous system. To study the function of this gene, we used a morpholino to knock down expression of shdb which resulted in significantly reduced eye size. Possible roles of Shdb in eye development are discussed as is future research aimed to elucidate the cellular and developmental mechanisms by which Shdb functions in the developing eye.

Abstract:
Le travail presente dans cette these a porte sur l'implication des proteines contenant des domaines sh2 et sh3, dans le mecanisme d'action de l'insuline et de l'igf-i. Nous avons montre que la phosphatidylinositol-3-kinase (pi-3-kinase) est stimulee par l'insuline dans des cellules insulino-sensibles, les adipocytes de rat. De plus, l'insuline induit une translocation de l'enzyme depuis le cytosol vers la membrane plasmique. Dans des fibroblastes, l'insuline et l'igf-i induisent la phosphorylation sur tyrosine de irs-1 (insulin receptor substrate-1), et son association avec la pi-3-kinase. Cette association est responsable de l'activation de l'enzyme. Dans des fibroblastes transfectes avec l'adnc de shc, les proteines shc sont phosphorylees sur tyrosine en reponse a l'insuline et a l'igf-i et interagissent avec irs-1. La surexpression des ces proteines ne modifie pas l'activation de la pi-3-kinase, mais induit une augmentation de l'activite basale de la mek. De plus, les cellules sont plus sensibles a l'action mitogenique de l'insuline

La tyrosine kinase de type non-recepteur c-abl est exprimee ubiquitairement chez tous les metazoaires. En plus de son domaine catalytique, c-abl possede des domaines d'interaction proteine-proteine qui sont essentiels pour sa fonction. Si la fonction precise de c-abl est inconnue, elle est impliquee dans une grande variete de processus. L'inactivation du gene a un phenotype pleiotropique conduisant a une mort embryonnaire precoce. L'activation constitutive de c-abl la transforme en un oncogene puissant implique dans des leucemies humaines. Il est donc vital pour la cellule de reguler de maniere extremement precise son activite catalytique. Au cours de ce travail de these, nous avons etudie les mecanismes moleculaires regulant c-abl. La tyr 412 est situee dans la boucle d'activation du domaine catalytique de c-abl. Ce residu est au cur du mecanisme de regulation de l'activite catalytique de c-abl. Son positionnement a l'interieur du domaine catalytique est essentiel au maintien de la forme inactive de c-abl. La phosphorylation de la tyr 412 est requise pour activer l'enzyme. La tyr 412 est un site de trans-autophosphorylation qui permet une boucle d'amplification du signal. Une particularite de la regulation de c-abl est le role d'un domaine de 80 acides amines situe en n-terminal de la molecule et contenant le site de myristoylation. Il interagit de maniere intramoleculaire avec le reste de la molecule. Cette interaction est necessaire pour la regulation de c-abl. Le point commun des formes oncogeniques de c-abl est l'absence de ce domaine. La deletion de ce domaine suffit a rendre c-abl oncogenique. C'est peut-etre la que reside la difference fondamentale entre la forme sauvage et les formes oncogeniques d'abl. L'activite catalytique de c-abl est donc sous le controle d'une combinaison de facteurs -interactions intramoleculaires, interactions proteine-proteine, evenement de phosphorylation- qui permettent d'assurer une reponse appropriee a un stimulus donne.

Establishment of precise neuronal connections requires the proper guidance and targeting of growing axons during embryogenesis. The growth cone, a sensori motor structure at the leading edge of the axon, expresses cell surface receptors for recognizing guidance and targeting signals. It also possesses the intracellular signal transduction machinery to convert the extracellular signals into the reorganization of cytoskeletal structures in inducing directed movement (guidance) or the cessation of movement (targeting). Accumulated evidence points to the importance of protein tyrosine phosphorylation in growth cone signaling. The signaling cascade linking tyrosine phosphorylation into the changes in growth cone motility, however, remains largely undefined. Previous studies suggest that the Dock/Nck adapter protein play a role in transducing tyrosine phosphorylation signals to the actin-based cytoskeleton to regulate growth cone motility. To gain insight into the Dock signaling pathway, we have sought to identify the downstream effector of Dock in photoreceptor (R-cell) growth cones in the Drosophila visual system. Our recent results indicate that Misshapen (Msn), a Ste20-like Ser/Thr kinase, functions downstream of Dock to terminate R-cell growth cones in the target region. First, like loss of dock, mutations in msn cause R-cell growth cone targeting defects. Second, Msn is expressed in R-cells and predominantly localized in R-cell growth cones. Third, overexpression of Msn induces the early growth cone termination, a phenotype that is opposite to the msn loss-of-function phenotype. Finally, Msn interacts with Dock physically and genetically. We propose that Dock-linked target-derived signals activate Msn, which in turn modulates cytoskeletal-related proteins within the growth cones in decelerating their motility when the axons reach their targets. Abstract:
The Sixteen Kingdoms was a period of political disintegration in medieval China. Foreign rulers of Wu Hu (“Five Barbarian Tribes”) captured the northern China during the 3rd – 5th centuries and established independent states through continuous military actions. This thesis aims at studying the relationship between the emperors and Buddhist monks in the Northwest empires in the Sixteen Kingdoms Period, including the Former Liang (320-376), the Western Qin (385-400; 409-431), the Later Liang (386-403), the Northern Liang (397-439) and the Xia (407-431), by developing a theoretical model called “Hourglass Model”. The model provides a holistic framework for investigating not only the emperors’ changing attitudes towards Buddhism but also the dissemination of the Dharma by monks. In addition, the concept of “data visualization” is implemented to re-interpret various historical sources. The complex interaction among imperial clans and Buddhist practitioners is further analyzed with computer-aided historical research methods. This thesis hopes to reveal the inspiring nature of the political religion in China’s early medieval age and expand the scope of Chinese Studies by rethinking about foreign rulers and Buddhist monks’ roles in reshaping and vitalizing the Chinese civilization. Moreover, this study develops alternative approaches to the research on the history of Chinese Buddhism, and attempts to shed new lights on theories and research methodology.

Abstract:
In tracing the experience of Chinese Canadians in the 20th Century, we need to look further back into its history. Most people believe that the increasing number of immigrants from Hong Kong, Taiwan and China in the 1980s and 1990s played the most important roles in the social and economic changes during the latest decades of the 20th Century. The contribution of the Chinese Canadians settlement throughout the 20th Century should also be considered as it marks the beginning of the rise of their social status and identity in Canada. Although the Chinese Canadians earned their fame and status since the 1980s, they had been racially discriminated for more than a century. To probe into the situation, the social and political situations in the Chinese Canadian community will be meticulously analyzed and their contribution in difference aspects examined. In addition, other minority ethnic groups, such as the Japanese, Jewish and Indian, will be used as a comparison to demonstrate the change of policies towards the Chinese in Canada. In doing so, both English and Canadian Chinese newspapers will be used to illustrate the cultural difference between the “whites” and “non-whites”. To illustrate the changes, the 20th Century will be break into three parts. In most of the pre-Second World War period, the Chinese community was isolated from the mainstream community with their activities largely confined to Chinatowns in cities, such as Vancouver, Victoria, Toronto and so on. For the Chinese living in small townships, such as Prince Rupert, Richmond and so on, their daily life will also be examined. When Canada declared war on Japan on 7th December 1941, Canada became an ally of China during the war. A sentiment of acceptance of the Chinese in the mainstream society began to take shape. Some of the Chinese chose to contribute their efforts to Canada by joining the Canadian Armed Forces and went into battle alongside the White Canadians. After the Second World War, Canada adopted a new policy towards the minority ethnic groups and Chinese Canadians started to enjoy political equality. In May 1947, the Canadian Government repealed the Chinese Immigration Act. In 1967, after the liberalization of the Canadian immigration policy, the Chinese, once again, were allowed to immigrate freely to Canada as an individual. With granted full citizenship, the Chinese social and political status began to change. In 1957, Douglas Jung, a Canadian born Chinese, was elected a Member of the Parliament, which can be seen as the beginning of the Chinese involvement in the political arena of the Canadian community. Since then, Chinese Canadians were able to achieve equality in the society. Based on documentary accounts and oral history research, this thesis re-constructed the history of Canadian Chinese involvement in the 20th Century and the change of their identity and social status thereafter.
