Abstract:
Relevance. The current paper has presented the study results of the effect of arid growing conditions on water deficit and foliage pigment (chlorophyll) of winter wheat varieties with various productivity.Methods. The study was conducted in 2017–2019. The objects of study were the winter durum wheat varieties ‘Eyrena’, ‘Yubilyarka’, ‘Yakhont’, ‘Yantarina’, ‘Oniks’, ‘Lazurit’ developed in the FSBSI “Agricultural Research Center “Donskoy”. There have been used such methods in the current study as the testing of varieties on drought tolerance in conditions of simulated drought (“zasushnik”) proposed by V.V. Maimistov (1988); the method of identification of chlorophyll content in winter wheat leaves proposed by I.G. Shmatko (1976); the method of identification of residual water deficit proposed by L.S. Litvinov (1988).Results. According to the study results there have been identified the most stable, productive winter wheat varieties ‘Yantarina’ and ‘Lazurit’, which possess mechanisms of adaptation to the water stress conditions. The varieties ‘Yantarina’ and ‘Lazurit’ also possess large productivity and chlorophyll content, as well as the smallest rise of residual water deficit in the increasing aridity with 169.7 and 159.5 g/m2, 1.4 and 1.9 mg/100 g of raw weight, 24.6 and 25.7%, respectively.

Abstract:
Abstract This article overviews community-based water monitoring (CBWM) in Aotearoa-New Zealand (NZ). CBWM was strongly boosted in NZ around 2000 by the development of the Stream Health Monitoring and Assessment Kit (SHMAK) and the Wai Care community initiative. Reform of freshwater management in NZ may be one driver for renewed interest recently in CBWM. Because professionals perceive volunteer-monitoring data as unreliable, they currently give little support to volunteer monitoring. To address their concern, we compared CBWM with measurements by regional authorities (RAs) – the main water management agencies in NZ. Agreement was encouragingly close for a comprehensive range of variables, including the important state-of-environment (SoE) indicators: visual clarity, Escherichia coli and macroinvertebrates. Community volunteers need and want ongoing professional support and encouragement, and, fortunately, there are important benefits for water management agencies, including engagement of citizens in water management and use of volunteer data. Professional support for CBWM in NZ could include: advice and encouragement, training, database development and quality assurance. Current research and development is focused on improving resources and systems for volunteer monitoring, notably with upgrading of the SHMAK. We are enlisting volunteers, equipped with improved tools and support systems, to assess the benefits of riparian rehabilitation and the suitability of water for swimming.

Abstract:
Binge drinking is a severe health problem faced by many US colleges and universities. College students often post drinking-related text and images on social media, portraying their alcohol use as socially desirable. In this project, we investigated the feasibility of mining the heterogeneous data (e.g. text, images, and videos) on Facebook to identify drinking-related contents. We manually annotated 4266 posts during 21 October 2011 and 3 November 2014 from “I’m Shmacked” group on Facebook, where 511 posts were drinking-related. Our machine learning models show that by combining heterogeneous data types, we were able to identify drinking-related posts with an F1-score of 0.81. Prediction models built on text data were more reliable compared to those built on image and video data for predicting drinking-related contents. As the first step of our efforts in this direction, this feasibility study showed promise toward unleashing the potential of mining social media to identify students who binge drink.

Abstract:
Gastric cancer is one of the most frequently diagnosed cancer and poses a serious threat to health system in the world. Upregulation of meningioma-associated protein (MAC30) has been found in many solid tumors and can regulate the proliferation, differentiation, and apoptosis of different tumor cells. Quantitative polymerase chain reaction (qPCR) was used to detect the expression of MAC30 in 68 patients with gastric cancer and their adjacent tissues. Lentiviral vector pGCSIL-shMAC30-GFP of the RNA interference (RNAi) of the MAC30 gene was transfected into gastric cancer BGC-823 cell line and the expression of lentivirus label protein GFP was observed via fluorescence microscope, while cell proliferation and apoptosis were determined with flow cytometry and MTT assay, respectively. Also, related protein expressions on Wnt/β-catenin signaling pathway were analyzed by Western blot method. The expression of MAC30 was abnormally elevated in gastric cancer tissues, while interfering of its expression could significantly inhibit the proliferation of gastric cancer BGC-823 cell line. However, the promotion of apoptosis by mitochondrial pathway was mediated by Bax/Bcl-2 upregulation. Present work showed the effect of downregulated MAC30 expression on proliferation and apoptosis of gastric cancer cell through Wnt/β-catenin signaling pathway. Thus, this investigation provides an experimental basis for future development of chemotherapeutic agent on gastric cancer.

Abstract:
Abstract Multiple myeloma (MM) remains incurable due to the development of a drug-resistant phenotype after prolonged therapy. Myristoylated alanine-rich C-kinase substrate (MARCKS) is a protein kinase C (PKC) substrate that plays an important role in cell adhesion, spreading and invasion. Our previous studies found that overexpression of phospho-MARCKS (pMARCKS) was detected in developed drug resistant MM cell lines (RPMI-8226 R5, MM.1R) relative to their parental drug sensitive cell lines (RPMI-8226S, MM.1S). We hypothesized that pMARCKS is involved in chemo- and novel drug resistance in MM. To further evaluate the drug resistance, we exposed both RPMI-8226 R5 and MM.1R cell lines to varying dosages of bortezomib, dexamethasone, doxorubicin, and lenalidomide. By MTT assay, both resistant cell lines were found to have significantly higher viability to all 4 drugs compared to their respective non-resistant lines. In addition, Western blot analysis showed increased pMARCKS expressions in all 3 bortezomib resistant cell lines 8226.BR, OPM2.BR, and ANBL-6.BR as compared to their respective bortezomib sensitive cell lines. We next acquired MM patient samples collected at diagnosis and at relapse after bortezomib treatment, and investigated their pMARCKS expression with immunoblotting analyses. The patient samples collected from relapse after bortezomib treatment had higher pMARCKS expression than those collected at diagnosis. Furthermore, we studied additional 3 primary MM patient samples with high pMARCKS expressions and 3 with low expressions for their vaibility after a 36 hour bortezomib treatment, and found that the samples with high pMARCKS expressions were more resistant to bortezomib than those with low pMARCKS expressions (mean IC50 of 7.1 nM and mean IC50 of 4.8 nM, respectively; p = 0.042). Importantly, combination of a low dosage of bortezomib (5.0 nM) with either 2.5 uM or 5.0 uM of enzastaurin (an inhibitor of phospho-PKC), displayed a synergistic cytotoxicity on myeloma cells with high pMARCKS expressions. To further elucidate the role of pMARCKS in drug resistance, we knocked down pMARCKS expression by transfecting siMARCKS into 8226 R5 and MM.1R cells. Following the knockdown, both cell lines had significantly lower viability after treatment with either bortezomib, dexamethasone, doxorubicin, or lenolidomide, in comparison to empty vector controls. FACS analysis and annexin V assay of the knockdown cells and the control cells from both cell lines showed significantly induction of G1/S cell cycle arrest and apoptosis in the knockdown cells. The immunoprecipitation (IP) and chromatin immunoprecipitation (ChIP) DNA-qPCR analysis further demonstrated that pMARCKS regulates SKP2 expression through binding with E2F1, mediating SKP2/p27Kip1 cell cycle pathway. Finally, we investigated the effect of inhibition of pMARCKS in a 8226 R5 xenograft model of SCID mice. Mice injected with shMARCKS-transfected 8226 R5 cells and received bortezomib showed significant retardation of tumor growth and prolonged survival compared to the control groups. Taken together, our data indicate that pMARCKS is constitutively activated in resistant and relapsed MM cells and contributes to drug resistance by regulating E2F1 mediated SKP2/p27Kip1 cell cycle pathway, thus providing a preclinical rationale for targeting pMARCKS as a promising approach in patients with refractory/relapsed MM. Abstract:
Problem setting. The modern world is characterized by rapid changes towards democracy, which have a positive impact on the humanitarian component of society. Education reforms are affected by many negative factors, including unstable political and economic situation in the country, and 2020 introduced the concept of “pandemic learning” into the educational process, which certainly did not improve the overall situation. However, despite these problems in the field of education, the issue of involvement of children with mental and physical disabilities into the educational environment of Ukraine remains extremely important. This trend of integration of children with mental and physical disabilities into the educational environment is relevant around the world, but in Ukraine attention has been paid and appropriate steps has been taken to improve the critical educational situation for this category of children only in the last few years. Also, the Association Agreement with the EU, which came into force in 2017, prompted the government to take certain steps for political, economic, social and cultural reforming of all spheres of life that affect the quality of provision and receipt of educational services. Recent research and publications analysis. Elaboration of theoretical and methodical basics of inclusive education can be found in the works by: N. Bastun, S. Bohdanova, L. Zdanevych, T. Zubarieva, E. Ketrysh, T. Kozhekina, K. Kolchenko, O. Kuzmina, T. Kuchai, N. Lalak, O. Lesher, O. Martynova, G. Onykovych, M. Semago, N. Semago, I. Smirnova, N. Sofiy, M. Tomchuk, N. Shmatko, O. Yarska-Smirnova, etc.Highlighting previously unsettled parts of the general problem. Analysis of current legislation and scientific literature allows us to conclude that inclusive education in Ukraine opens up great opportunities for children/people with special educational needs, but is not explored fully. These sources emphasize that creation of optimal conditions for qualitative education for children with mental and physical disabilities has its social, economic and psychological characteristics and problems, which are quite complex and require further research and development. That is why there is a need to work systematically and purposefully on introduction of inclusive process in educational institutions to implement the right to education of every citizen of Ukraine. Paper main body. The article considers specific features of introduction of inclusive education in Ukraine in the context of European integration. Dynamics of implementation of inclusive education in the context of education system reform is analyzed. Components of preparation to implementation of the inclusion process both for teachers and for children with special educational needs and main problems of the educational environment were considered. Conclusions of the research and prospects for further studies. Analysis of development of inclusive education in Ukraine indicates a positive trend in the number of children with special educational needs enrolled in comprehensive schools, as indicated by the dynamics from 2720 pupils in the 2015 – 2016 school year to 18643 pupils in the 2019 – 2020 school year. Opening of 638 inclusive resource centers is an important step towards introduction of inclusive education not only in big cities but also in the regions. However, existing economic (ensuring architectural accessibility of interiors, accessibility of appropriate transport and transport infrastructure for children/persons with mental and physical disabilities, campuses, sports and cultural facilities, dormitories, etc.) and social (complex process of socialization both for children with special educational needs (as well as their family members) and for relatively healthy children; change of psychological and value attitudes of teachers and increase of their professional competencies in an inclusive form of education, etc.) problems are complicated by partial or complete distance learning during a pandemic, and solving these problems will facilitate the reception of educational services by the specified category of citizens of Ukraine.

Abstract:
The article draws on a broad interpretation of the invective as a non‑standard (non-literary) vocabulary known in linguistics as jargonisms, expletives, vulgarisms; foul, pejorative, negatively coloured, disparaging, slang, obscene, coarse, abusivу, taboo words and other lexical units that contain the meaning of an insult in their semic structure; less often the invective is understood as a codified (literary) vocabulary which acquires the insulting meaning in a context as an expression of the speaker’s communicative intention and pragmatic tactics of consciously offering a public affront to a specific addressee of communication. The aim of the research is the lexico-semantic and communicative-pragmatic characteristics of the invective vocabulary in a modern media discourse and social networks as a specific verbal means of a psychological impact on the consciousness of the recipients. By resorting to the method of free word association test, the authors have studied a conscious and/or subconscious reaction of Ukrainian females and males to pejorative by-words that stir up a feeling of insult. 100 people have been selected as respondents (50 people of each gender). All of them were Ukrainian native speakers including female and male lecturers and students of Lesya Ukrainka Eastern European National University (Ukraine) and Pereiaslav-Khmelnytskyi Hryhorii Skovoroda State Pedagogical University (Ukraine); choosing the stimuli, the authors proceeded from the frequency of their use in the texts of modern mass media (out of 300 detected nominations the authors used 100 units). According to the extent of the insult caused by the given words they were rated on a scale of 1 to 4 which made it possible to combine the analyzed stimuli into four groups with the following scores: 1) 2.65–2.93; 2) 1.67–2.31; 3) 1.03–1.54; 4) 0 (zero). The experiment gave a clear structure of the invective – a psycholinguistic category including a communicative-pragmatic intention of the insult.
