AbstractProtein-focused research has been challenging in Drosophila melanogaster due to few specific antibodies for Western blotting and the lack of effective labeling methods for quantitative proteomics. Herein, we describe the preparation of a holidic medium that allows stable-isotope labeling of amino acids in fruit flies (SILAF). Furthermore, in this chapter, we provide a protocol for mitochondrial enrichments from Drosophila larvae and flies together with a procedure to generate high-quality peptides for further analysis by mass spectrometry. Samples obtained following this protocol can be used for various functional studies such as comprehensive proteome profiling or quantitative analysis of posttranslational modifications upon enrichment. SILAF is based on standard fly routines in a basic wet lab environment and provides a flexible and cost-effective tool for quantitative protein expression analysis.

AbstractThe frequent fluctuations in global climate variability (GCV), decreases in farmland and irrigation water, soil degradation and erosion, and increasing fertilizer costs are the significant factors in declining rice productivity, mainly in Asia and Africa. Under GCV scenarios, it is a challenging task to meet the rice food demand of the growing population. Identifying green traits (tolerance of biotic and abiotic stresses, nutrient-use efficiency, and nutritional grain quality) and stacking them in high-yielding elite genetic backgrounds is one promising approach to increase rice productivity. To this end, the Green Super Rice (GSR) breeding strategy helps to pool multi-stress-tolerance traits by stringent selection processes and to develop superior GSR cultivars within a short span of 4–5 years. In the crossing and selection process of GSR breeding, selective introgression lines (SILs) derived from sets of early backcross BC1F2 bulk populations through both target traits and non-target traits were selected. Genotyping of SILs with high-density SNP markers leads to the identification of a large number of SNP markers linked with the target green traits. The identified SILs with superior trait combinations were used for designed QTL pyramiding to combine different target green traits. The GSR breeding strategy also focused on nutrient- and water-use efficiency besides environment-friendly green features primarily to increase grain yield and income returns for resource-poor farmers. In this chapter, we have highlighted the GSR breeding strategy and QTL introgression of green traits in rice. This breeding strategy has successfully dissected many complex traits and also released several multi-stress-tolerant varieties with high grain yield and productivity in the target regions of Asia and Africa.

This issue is particularly significant in areas such as Remote Condition Monitoring, where the ability to analyse the network at a whole-systems level is being blocked by the business cases around the purchase of systems as silos. Blockchains are an emerging technology that have the potential to revolutionize the management of transactions in a number of industrial sectors. This chapter will address the outstanding issues around the fair attribution of costs and benefits of data sharing in the rail industry by proposing blockchains as a forth enabler of the rail data revolution, alongside ESB, ontology, and open data.

Two popular initiatives are supply chain management (SCM) and customer relationship management (CRM). SCM focuses on optimizing the materials, information, services, and financial flows through a supply network. CRM focuses on marketing, sales, and customer service, and aims to maximize the value of customer relationships. Furthermore, the real potential lies in the integration of SCM and CRM. Disconnected implementations can result in IT “silos” with redundancies in hardware, software and staff, breaks in the information chain, and disappointing performance. The forecasting function in customer service drives the downstream resource planning processes to provide the best customer experience at optimal costs. For a mature, global organization, its existing systems and processes have evolved over time and become complex. These complexities result in informational silos that result in sub-optimal use of data thereby creating inaccurate forecasts that adversely affect the planning process in supporting the customer service function. For addressing this problem, the authors argue for the use of frameworks that are best suited for a big data ecosystem. Drawing from existing literature, the concept of data lakes and data value chain have been used as theoretical approaches to devise a road map to implement a better data architecture to improve the forecasting capabilities in the given organizational scenario.

The emergence of e-services benefited the stakeholders due to ease of access to data, information and knowledge sources. Service-based applications have evolved into flexible and adaptable systems capable of coping with changes in user requirements and business processes. The shift from monolithic application silos towards service-oriented approaches is evident in the literature today. The benefits of service-oriented approaches include cost effectiveness, improved inter-operability, reusability, and flexibility. The benefits are not enjoyed without the threat of cognitively overloading managers in their decision making activities. Tools for effective management of information are necessary. Effective and efficient service-oriented applications need to operate within their situational boundaries. As such, decision support type tools require tight integration with the service-based approach. This study proposes an integrated Situated Service-Oriented Model and demonstrates its value via a case study of an e-learning service-based application used over a period of 15 months. Two designs were used; component-based and service-oriented. The significance of this study is in the tangible value of the model proposed and demonstrated by the e-learning case study.

Las celdas solares de CdTe/CdS en película delgada depositadas sobre substratos flexibles y en la configuración sustrato no han tenido buena respuesta fotovoltaica, dando menores eficiencias que sus homologas desarrolladas en configuración superestrato. Son dos los procesos relacionados con el deterioro de las propiedades de la película absorbente CdTe, estos son: 1) el tratamiento con cloruro de cadmio que se deposita sobre el CdTe antes del CdS no permite que se mejore la unión entre estos semiconductores; 2) el contenido de cobre que se deposita en el contacto trasero de la celda o durante el depósito de CdTe degradando las propiedades de la película. Sin embargo, en el año 2013 se logró desarrollar una celda solar CdTe/CdS sobre sustrato de molibdeno flexible, incorporando de manera controlada capas atómicas de cobre entre el CdTe y el CdS, alcanzando una eficiencia de 11.5 %. Estos resultados dejan claro que aún hay mucho por investigar en cuanto a los materiales disponibles para celdas solares y la importancia que tiene el conocer sus propiedades para entender y dirigir la investigación hacia desarrollo de dispositivos fotovoltaicos eficientes, de bajo costo y que brinden beneficio a largo plazo. En el presente trabajo se presentan los resultados de estudios de las películas delgadas de CdTe depositadas mediante la técnica de sublimación por transporte de vapor en espacio cercano (CSVT, por sus siglas en inglés), sobre sustratos de molibdeno flexible. En estos depósitos se variaron espesores para evaluar la interacción del oxígeno usado durante el depósito en las propiedades estructurales, ópticas, eléctricas, químicas y morfológicas del CdTe. Los resultados mostraron que el oxígeno forma óxidos de teluro y teluratos de cadmio en la superficie de la película de CdTe. Además, se encontró evidencia de defectos comunes del CdTe tales como maclas y fallas de apilamiento.
