Abstract:
Current evidence-based guidelines recommend that 2% (w/v) chlorhexidine digluconate (CHG), preferentially in 70% (v/v) isopropyl alcohol (IIPA), is used for skin antisepsis prior to incision of the skin. In this current study, the antimicrobial efficacy of CHG, six essential oils [tea tree oil (TTO), thymol, eucalyptus oil (EO), juniper oil, lavender oil and citronella] and novel benzylidenecarboxamidrazone and thiosemicarbazone compounds were determined against a panel of microorganisms commonly associated with skin infection (Staphylococcus epidermidis, S. aureus, meticillin-resistant S. aur

The aim of this thesis was to A) study drug delivery through the skin with a liquid formulated to promote absorption, B) develop and validate methods to analyze the drug in the samples obtained, C) assess appropriate methods to measure the transdermal delivery of drug, and D) apply to pharmacodynamics. The stability of a rectal formulation of diazepam, Diastat®, and a quality control of a topical form, TDS® diazepam, were studied using high performance liquid chromatography (HPL

Abstract:
In order to develop safe and effective topical and transdermal formulations to treat either local skin disorders or for systemic drug delivery, it is first imperative to assess skin permeation using a reliable and comprehensive analysis method. The assessment of drug permeation into/across the skin is traditionally accomplished using Franz diffusion cells with subsequent analysis by conventional chromatographic methods such as HPLC and more recently using advanced imaging techniques. In this context, time of flight-secondary ion mass spectrometry (ToF-SIMS) offers distinctive advantages in map

Abstract:
The sympathetic division of the autonomic nervous system is known to play a role in the genesis of neuropathic pain. In the skin of the rat lower lip (hairy skin), sympathetic and parasympathetic fibres normally innervate the same blood vessels in the lower dermis but do not occur in the upper dermis. However, we have shown that sympathetic fibre migration into the upper dermis occurs following mental nerve lesions (1. Compo Neurol. 422:287-296, 2000). As sensory denervation has a dramatic effect on sympathetic fibre innervation patterns in the rat lower lip skin, we decided to investigate the

Neuropathic pain presents itself with many faces. It can be devastatingly painful leading to a person's inability to lead a productive life or it can be mild. In this thesis I discuss one of the countless possible mechanisms which could underlie the development and maintenance of this disorder; namely the involvement of peripheral changes in innervation. The approach taken in this thesis was mainly based on protein analyses, based on the assumption that protein expression (or lack thereof) is indicative of changes in the physiological properties of structures that either express or secrete pr

In this thesis, the innervation patterns by substance P- (SP), dopamine-beta-hydroxylase- (DbetaH) and the vesicular acetylcholine transporter- (VAChT) immunoreactive (IR) fibres were investigated in the skin of the rat lower lip, in order to provide a morphological basis for the possible interactions between sensory and autonomic fibres in the periphery. Our immunocytochemical approach revealed that SP-IR sensory fibres were widely distributed throughout the epidermis and dermis. In the dermis, they were associated with blood vessels, hair follicles, sebaceous glands and mast cells. Electron

Epidermal Langerhans cells (LCs) and multiple subsets of dermal dendritic cells (dDCs) make skin a valuable route for vaccination, offering the potential for antigen-sparing immunisation. The interconnected immunological functions of dDC subsets and LCs are not fully understood however. This Thesis therefore aimed to explore the interactions of skin immune cells with viral pathogens and vaccines to inform the development of future therapeutics and intradermal vaccines. LCs and dDCs were isolated from ex vivo human skin tissue using a walkout protocol which allowed the enrichment of the migrato
