Full textThrough the desire of understanding the Internet better, we focused our attention on the modelling of the way data move in the Internet. Our main goal here was to give ideas on what does a route followed by packets look like.
In most of the simulations, researchers model routes by shortest paths, we try to provide more realistic models to improve simulations accuracy out of concern for keeping it relatively simple.
To achieve this study, we used the data from the Skitter network measurement platform that runs about thirty monitors which probe tens of thousands destinations per day around the world in order to collect routes packets have followed with the path forwarding method. It is the largest known effort of route collection and also a mean to get a realistic topology of the Internet.
With the help of the Skitter data we pointed out lots of global and local routes characteristics and we have designed some algorithms to generate fake routes that fit the properties observed.

Full textThis essay will focus on drug addicts and the world as they see it. The essay is based on intervjues with three men and one woman and participant observation of the three men´s social life in the surroundings of Stockholm. The informants’ opinions and experiences are central as well as the observations made of them during an intensive fieldwork. The study which was of an exploring character has revealed a complex relationship between the informants and society. The concepts of subculture and stigmatization seem equally central in the social life and perception of life of these drug addicts. The intricate relationship between the two needs further study before a model of the interrelationship can be presented. In a final discussion of the results of the study with the informants they confirm that they see themselves both as part of a subculture and stigmatized by society.
The study also includes some ethical reflections on the role of the investigator in relation to informants who are drug addicts.

Full textCell polarity is a fundamental process that controls cell’s functional and physiological specificities. This process relies on membranous compartments differently composed both on proteins and on phosphatidyl-inositols (PIs). Indeed, through their asymmetric localization, polarity proteins, such as the PAR proteins, are essentials to establish and maintain polarity of the cells. During my PhD, I studied the interplay between the polarity proteins and the PIs. Using the Drosophila egg chamber, as a model, I aimed to characterized the upstream events that regulate the PI(4,5)P2 producing kinase (PIP5K), Skittles (SKTL), activity and localization. I also studied the downstream molecular process that link the PI(4,5)P2, SKTL and the PAR proteins in cell polarity. I contributed to the characterization of the importance of PI(4,5)P2, mainly produced by SKTL in maintaining the apical-basal polarity and during the morphogenesis of the follicle cells. The PI(4,5)P2 is ensuring PAR3 and adherens junctions but not PAR1 proper localizations. Next, through a precise quantification method, I showed that SKTL and the PI(4,5)P2, probably via vesicular traffic, were also ensuring PAR3 proper localizations (anterior accumulation and stage 9B posterior exclusion) in the oocyte. PAR3 accumulation also relies on a Dynein mediated transport and the IKKε kinase while its posterior exclusion relies on PAR1 phosphorylation. Finally, I studied SKTL post translational modifications and their relevance on cell polarity. I identified palmitoylation and phosphorylations that are regulated by the kinase PAR1 and the phosphatase PP1. SKTL phosphorylations seem to be related to its role on the vesicular traffic. Altogether these results clarify some mechanisms involving both PIs and PAR proteins in cell polarity maintaining and establishment

Full textCell division must be accurately controlled in time and space to permit the formation of two daughter cells whose genetic content is identical to that of the mother cell. This process requires successive modifications of cell shape, induced by cortical remodelling. Molecular mechanisms controlling cortical reorganization during mitosis remain partially uncharacterized. Our work in Drosophila cells demonstrates that PI(4,5)P2, a phosophoinositide of the plasma membrane, is enriched at the equatorial region at the onset of anaphase. This PI(4,5)P2 is necessary for the cortical stability of mitotic cells, and requires dMoesin activation. The dMoesin, linking actin to the plasma membrane, plays a critical role in the cortical organization of mitotic cells and in the regulation of its mechanical properties. We show that the interaction PI(4,5)P2/dMoesin participates in cellular contraction at the beginning of mitosis, then in cell elongation characteristic of subsequent steps. At the end of mitosis, the Pp1-87B phosphatase inactivates the dMoesin. By a systematic functional screen, we characterize the key role of two pathways acting in synergy to locally produce PI(4,5)P2, Skittles- and Pten-dependent, and the role of a third pathway requiring dOcrl activity to control PI(4,5)P2 homeostasis. Altogether, these results allow us to better understand the mechanisms controlling cortical remodelling and modifications of cell shape that occur during mitosis.

Full textGuimarães, Marta Barahona Simões da Fonseca. "Communication plan for Skittles." Master's thesis, 2015. http://hdl.handle.net/10071/12074.
Full textThe purpose of this master thesis was the development of a communication plan for one product that belongs to MARS Inc., Skittles. Nowadays sugar consumption is part of the daily life of each teenager, and mostly is consumed in classes or specific leisure moments. One thing is certain, these products are consumed every day and people have pleasure in eat them. However, there wide range of possibilities in this market, they can choose from chocolates, gums or even other products such as cakes, for example. Therefore, this kind of product is easily replaceable and the competition is very “aggressive” and everywhere. Skittles, with almost null communication yet has a significant amount of sales, which mean that represents a great opportunity to invest in a communication plan and make Skittles the “chosen candy”. Given the situation, the communication plan will strive to fulfil two main objectives: increase brand awareness and raise purchase intention of the product from the target audience. To support the plan it was necessary to perform a deep analysis of the external and internal environment of the product Skittles, using primary and secondary data, which allowed reaching a set of insights and conclusions. Therefore, the Skittles communication plan was designed for one year, consisting in promotional activities and brand activations, as well as, the combination of different communication tools in order to create an integrated and effective plan while taking into consideration the proposed goals.
