Development of a three dimensional (3D) HYBROT model with targeted in vivo like intact cellular circuitry in thick brain slices for multi-site stimulation and recording will provide a useful in vitro model to study neuronal dynamics at network level. In order to make this in vitro model feasible, we need to develop several associated technologies. These technologies include development of a thick organotypic brain slice culturing method, a three dimensional (3D) micro-fluidic multielectrode Neural Interface system (µNIS) and the associated electronic interfaces for stimulation and recording of/from tissue, development of targeted stimulation patterns for closed-loop interaction with a robotic body, and a deep-tissue non-invasive imaging system. To make progress towards this goal, I undertook two projects: (i) to develop a method to culture thick organotypic brain slices, and (ii) construct a multiphoton imaging system that allows long-term and deep-tissue imaging of two dimensional and three dimensional cultures.
Organotypic brain slices preserve cytoarchitecture of the brain. Therefore, they make more a realistic reduced model for various network level investigations. However, current culturing methods are not successful for culturing thick brain slices due to limited supply of nutrients and oxygen to inner layers of the culture. We developed a forced-convection based perfusion method to culture viable 700µm thick brain slices.
Multiphoton microscopy is ideal for imaging living 2D or 3D cultures at submicron resolution. We successfully fabricated a custom-designed high efficiency multiphoton microscope that has the desired flexibility to perform experiments using multiple technologies simultaneously. This microscope was used successfully for 3D and time-lapse imaging.
Together these projects have contributed towards the progress of development of a 3D HYBROT.

Die Entwicklung effizienter Desktop Publishing Systeme wird behindert durch den Mangel an leistungsfähigen, automatischen Layoutalgorithmen. Aktuelle Algorithmen zum Layout ganzer Dokumente oder einzelner Seiten erfordern entweder die Formulierung des Layoutproblems in einer formalen Beschreibungssprache, oder sie benötigen fertige, detaillierte Layouttemplates. Layoutprobleme mit schwacher Semantik lassen sich schlecht in formale Sprachen umsetzen, Layout Templates verschieben den manuellen Aufwand nur vom Endnutzer zum Template Designer. Das erste Ergebnis dieser Dissertation ist ein Layoutalgorithmus, der ein allgemeines Layoutproblem löst, in dem er es als Ressourcenallokationsproblem interpretiert. Die Fläche einer einzelnen Seite ist eine Ressource, die zwischen den visuellen Elementen einer Seite verteilt wird. Das Layoutproblem wird in ein lexikographisches min-ordering Optimierungsproblem übersetzt, das durch lineare Optimierung in Echtzeit gelöst wird. Die Lösungen manuell erzeugter Layoutprobleme sind häufig über- oder unterbestimmt. Wenn das Problem überbestimmt ist, also keine gültige Lösung besitzt, muss der Algorithmus die Lösung finden, die am n\"achsten an der intendierten Lösung ist. Der Algorithmus erkennt nicht eindeutig definierte Probleme mit unbefriedigenden Lösungen und fügt die minimal notwendige Anzahl von Constraints hinzu um das vom Nutzer beabsichtigte Layout zu erzeugen. Das zweite Ergebnis ist die Entwicklung einer intuitiven Benutzerschnittstelle, die es erlaubt, die vorhergehend beschriebenen Layoutprobleme zu erzeugen. Sie verbirgt die Komplexität des Constraintsystems und vermeidet die Komplexität constraint-basierter Grafikanwendungen der Vergangenheit. Diese Benutzerschnittstelle macht formale Beschreibungssprachen und manuell erzeugte Layouttemplates überflüssig. Die Evaluation zeigt, dass die besten Tabellenlayoutalgorithmen keine signifikant besseren Ergebnisse produzieren als der allgemeinere ICBM Layout Algorithmus.The efficiency of desktop publishing is severely limited by the lack of sophisticated automatic document layout systems. State-of-the-art algorithms either require the input to be written in a description language such as HTML and LaTeX, or to be a manually designed layout template. However, description languages are ill-suited to express layout problems with weak semantics and layout templates shift the burden from the end user to the template designer. The first contribution of this thesis is an algorithm that solves a general class of layout problems by treating them as equitable resource allocation problems. The available document area is a resource that is distributed among inter-element gaps. The layout problem is transformed into a lexicographic min-ordering optimization problem that is solved using linear programming techniques in real-time. If the layout problem is over-constrained, the quality of the solution layout degrades gracefully. The layout algorithm finds the solution layout with the most equitable distribution of constraint errors among the soft layout constraints, i.e., the solution closest to the user''s original intent. Conversely, the layout algorithm detects the under-constrained subproblems that adversely affect the solution layout. It adds the minimal number of constraints required to achieve the fully specified layout problem that is closest to the user''s input. The second contribution is the creation of an intuitive direct manipulation user interface that lets users create the aforementioned class of general constrained layout problems. It hides the complexity of the constraint system and avoids the usability problems that have plagued constraint drawing applications. It eliminates the need of document description languages and manually-created layout templates. In the evaluation, we show that the best state-of-the-art specialized table layout algorithms do not outperform the general ICBM layout algorithm by any significant margin.

The main goal of this work is the analysis of manufacturing costs for the component of wheel´s blower. Followed by setting up the size of specific cutting force for milling operation of the titanium alloy Ti-Al6-Mo2-Cr2-Fe-Si, the used tool was a milling cutter which is made out of sintered carbide with conical and spherical face. The final values which are at intervals of 1500 to 1800 MPa were compared with the values of the Sandvik Coromant firm kc = 1690 MPa, for titanium alloy with the strenght in tension Rm = 1050 MPa.

In this thesis we describe an automatic human interest detector that uses speech, physiology, body movement, location and proximity information. The speech features, consisting of activity, stress, empathy and engagement measures are used in three large experimental evaluations; measuring interest in short conversations, attraction in speed dating, and understanding the interactions within a focus group, all within a few minutes. In the conversational interest experiment, the speech features predict about 45% of the variance in self-reported interest ratings for 20 male and female participants. Stress and activity measures play the most important role, and a simple activity-based classifier predicts low or high interest with 74% accuracy (for men). In the speed-dating study, we use the speech features measured from five minutes of conversation to predict attraction between people. The features predict 40% of the variance in outcomes for attraction, friendship and business relationships. Speech features are used in an SVM classifier that is 75%-80% accurate in predicting outcomes based on speaking style. In the context of measuring consumer interest in focus groups, the speech features help to identify a pattern of behavior where subjects changed their opinions after discussion. Finally, we propose a prototype wearable 'interest meter' and various application scenarios. We portray a world where cell phones can automatically measure interest and engagement, and share this information between families and workgroups.

This diploma thesis deals with cerium-based conversion coatings for aluminium alloys. At first the mechanism of protection of metals by inhibitory substances is described in the theoretical section. The next chapter is devoted to application of lanthanide salts as cathodic corrosion inhibitors. Then the characterization of alloy AA 2024 follows. The main part is dedicated to the growth mechanism of the cerium-based mixed conversion coating on the alloy AA 2024. Finally, the factors affecting the character and the quality of obtained coatings are commented. In the experimental part an influence of the thermal activation together with the activation by a hydrogen peroxide was observed. The created film was characterized using SEM and EDS. At the introduction of this work is briefly commented phase composition of AZ91 alloy, which is followed by detailed description of five very different theories of Ni-P coatings formation. Theoretical part is ended by review aimed to current states of development in areas of chemical nickel coating, influence of AZ91 alloy microstructure to current less deposition and possibility of surfactants influence to decrease of surface roughness. In experimental part of this thesis was the morphology and composition of Ni-P coatings including interlayers dependence on chemical composition of nickel-plating bath and magnesium alloy surface pre-treatment investigated using scanning electron microscopy and inverted optical microscopy. Average phosphorus content was obtained using energy dispersive analysis. Closing part of experimental part was dedicated for mechanical properties of coatings by micro-indentation analysis.

The thesis is focused on the surface treatment used for the preparation of aluminium alloys for the adhesive bonding. The theoretical part can be divided into three chapters. The first of them deals with aluminium alloys, the second one speaks about the adhesive bonding. The surface treatment itself, especially the conversion coating issues, is discussed in the last chapter. The experimental part is focused on the optimalization of a two-step titanium and zirconium based conversion coating technology. In the first part, the aluminium alloy is analysed by the glow discharge optical emission spectroscopy. The second part of the experiment deals with the first step of the technology, acidic deoxidation. The objective was to examine the influence of the exposure time on the morphology of the alloy surface and the shear strength of adhesive joints. In the last part of the experiment, the conversion coatings are prepared and characterised by the scanning electron microscopy and energy dispersive spectroscopy.

The theoretical part of the thesis contains a brief overview of some experimental methods that are used in machining technologies. The experimental part at first elaborates on the analysis of two titanium alloys. The actual experiment, which consists of milling of the two aforementioned materials with the use of several tools and holders, is then described. It was conducted in collaboration with the company Frentech Aerospace s.r.o. When milling, forces were measured with piezoelectric dynamometer. Further, the analysis of acquired data that is followed with discussion about its main findings, is provided. The conclusion of the thesis deals with the found facts corresponding with the information obtained by analysis of materials. The main results arise out of the comparison of used combinations of tools and holders for milling of material Ti-Al6-V4, observation of impact of particular factors on measured force, or out of different values of measured forces when milling two given materials.

Video streaming is becoming a major channel for distance learning (or e-learning). A tremendous number of videos for educational purpose are capturedand archived in various e-learning systems today throughout schools, corporations and over the Internet. However, making information searchable and browsable, and presenting results optimally for a wide range of users and systems, remains a challenge.In this work two core algorithms have been developedto support effective browsing and searching of educational videos. The first is a fully automatic approach that recognizes slides in the videowith high accuracy. Built upon SIFT (scale invariant feature transformation) keypoint matching using RANSAC (random sample consensus), the approach is independent of capture systems and can handle a variety of videos with different styles and plentiful ambiguities. In particular, we propose a multi-phase matching pipeline that incrementally identifies slides from the easy ones to the difficult ones. We achieve further robustness by using the matching confidence as part of a dynamic Hidden Markov model (HMM) that integrates temporal information, taking camera operations into account as well.The second algorithm locates slides in the video. We develop a non-linear optimization method (bundle adjustment) to accurately estimate the projective transformations (homographies) between slides and video frames. Different from estimating homography from a single image, our method solves a set of homographies jointly in a frame sequence that is related to a single slide.These two algorithms open up a series of possibilities for making the video content more searchable, browsable and understandable, thus greatly enriching the user's learning experience. Their usefulness has been demonstrated in the SLIC (Semantically Linking Instructional Content) system, which aims to turnsimple video content into fully interactive learning experience for students and scholars.

In particular we define the Dehn-Thurston coordinates for the set S = S(Σ) of free homotopy class of multicurves on the surface Σ. We also prove new results, like the precise relationship between Penner's and D. Thurston's definition of the twist coordinate and the formula for calculating the Thurston's symplectic form using Dehn-Thurston coordinates. For Chapter 2, let Σ be a surface of negative Euler characteristic together with a pants decomposition PC. Kra's plumbing construction endows Σ with a projective structure as follows. Replace each pair of pants by a triply punctured sphere and glue, or `plumb', adjacent pants by gluing punctured disk neighbourhoods of the punctures. The gluing across the ith pants curve is denied by a complex parameter μi ∈ C. He proposed this as a technique to aid in debugging because he conjectured that this is what programmers did naturally when debugging. Here program slicing is applied to an experimental concurrent language called LF. Existing techniques are adapted to accommodate the unique features of the language.

We studies two examples of polytope slices, hypersimplices as slices of hypercubes and edge polytopes. For hypersimplices, the main result is a proof of a conjecture by R. Stanley which gives an interpretation of the Ehrhart h*-vector in terms of descents and excedances. Our proof is geometric using a careful book-keeping of a shelling of a unimodular triangulation. We generalize this result to other closely related polytopes. We next study slices of edge polytopes. Let G be a finite connected simple graph with d vertices and let PG C Rd be the edge polytope of G. We call PG decomposable if PG decomposes into integral polytopes PG+ and PG- via a hyperplane, and we give an algorithm which determines the decomposability of an edge polytope. Based on a sequence of papers by Ohsugi and Hibi, we prove that when PG is decomposable, PG is normal if and only if both PG+ and PG- are normal. We also study toric ideals of PG, PG+ and PG-. This part is joint work with Hibi and Zhang.

Using presentation software such as PowerPoint or Keynote to sup- port lectures and presentations has become ubiquitous in both academia and industry. However, designing the visuals of presentation slides is a time-consuming and laborious task; repetitive steps are required for selecting templates, organizing objects, and optimizing layouts. To al- leviate these laborious works and to allow users to focus on preparing the contents of presentations, we present SmartPPT, a framework that supports the automatic generation of presentation slides from textual outline. We built a Recommender System model inside the framework that could pick up slide templates for input textual outline. To test its functionality and efficiency, two sets of user study procedures were conducted and shown that SmartPPT is time efficient in generating slides and it outperforms in user satisfaction when compared to user- handcrafted slides and the baseline condition, which was PowerPoint suggested templates.Att använda presentationsprogram som PowerPoint eller Keynote för att stödja föreläsningar och presentationer har blivit allestädes närvarande både i akademin och industrin. Att utforma visuella bilder av presentationsmaterial är emellertid en tidskrävande och mödosam uppgift; upprepade steg krävs för att välja mallar, organisera objekt och optimera layouter. För att underlätta dessa mödosamma arbeten och låta användarna fokusera på att förbereda innehållet i presentationer presenterar vi SmartPPT, ett ramverk som stöder den automatiska generationen av presentationsbilder från en textvy. Vi byggde en Recommender System-modell inuti ramen som kunde plocka upp bildmallar för inmatning av textrutor. För att testa dess funktionalitet och effektivitet genomfördes två uppsättningar av användarstudieprocedurer och visade att SmartPPT är tidseffektivt för att generera bilder och användarna blev nöjdare jämfört med när man gjort handgjorda bilder eller använt PowerPoint-föreslagna mallar.

Study includes review of machining strategies of titanium alloys. The materials for cutting tools and various types of tool wear are included. The experimental part of study consists of two partial experiments focused on the face milling by the ball – end milling cutter. First part is focused on facial milling of the inside part of spherical surface by the ball – end milling cutter and thus its surface roughness rating. The second part of experiment focuses on the milling of flat surface by the ball – end milling cutter and consequential analysis of cutting forces when the chip thickness is variable.

The Diploma thesis deals with the verification of Jominy end-quench test when analysis of hardenability of aluminium alloys is considered. The problem was solved experimentally, by developing end-quench curves of selected grades of aluminium alloys. Degree of hardenability of alloy is hardness after complete strengthening heat treatment. Samples of aluminium alloys commonly used for aircraft structures were prepared and then subjected to end-quench tests in various quenchants. On the basis of performed experiments, suitability of Jominy end-quench test for comparison of hardenability of alloys was found. Part of the thesis deals with the creation of equivalent cooling rate diagrams using the developed end-quench curves.

The diploma thesis is focused on the description and the verification of a relatively young creep age forming method. This method is based on a relaxation of stress and a creep of material, which occur by forming a part in the clamping tool during the artificial precipitation heat treatment at elevated temperature. After removing from the tool, the spring back and the residual stresses in the part are smaller compared to the conventional forming methods. This is mainly reflected in the increased fatigue resistance of the structure. The creep aging is only applicable to the precipitation heat treatable alloys. This thesis is focused on aluminium alloys, which are typical for the aerospace industry. The first part of the thesis describes principles of technology, its use in the aviation and material processes in the aluminium precipitation heat treatable alloys during the creep age forming. In the second part, there is a methodology of the verification of technology on samples made of the aluminium alloys ČSN EN AL-P 7075 and ČSN 424203 (approximate equivalent of ČSN EN AL-P 2024). Furthermore, a forming tool is designed, and experiments are performed according to the proposed methodology. At the end of the thesis, the experiments are evaluated and changes in the spring back of the samples are compared for three different methods of forming: the conventional cold clamping forming, the creep age forming and the creep age forming with an overaging.

This thesis describes magnesium alloys, their properties and methods of their production. However, experimental part is focused on AZ91 magnesium alloy. Conversion coatings were prepared on the AZ91 surface – chromate coating, phosphate permanganate coating and fluoride coating. Corrosion resistance of the prepared coatings were compared by immersion tests and by potentiodynamic methods. Evaluation of corrosion resistance by immersion tests was successful only for phosphate permanganate coating. Only this coating was removed during cleaning of the samples. For comparison of corrosion resistance was important to find the correct procedures and optimize the measurements methods. For these purposes have been chosen galvanic zinc coating on steel sheets. Thickness of the zinc coating were 8 and 30 microns. This coatings provide uniform zinc surface without heterogenities which was important for the potentiodynamic measurements. After optimization methods for potentiodynamic measurements by galvanic zinc coatings have been measured magnesium alloy AZ91 uncoated and coated by conversion coatings. For uncoated AZ91 magnesium alloy was determined suitable method so called virgin cathodic curve. On the other hand, for the coated AZ91 magnesium alloys was determined suitable method so called combined curve, within which we measured cyclic voltammetry curve (± 5 mV) and then virgin anodic curve.

In my project, I researched the boundary in the field of architecture. I suppose that setting the boundary is the most primitive architectural activity. It is often said that the beginning of architecture was constructing the roof to prevent from rain. When the roof is constructed the boundary between inside and outside is generated. Setting boundary in space is, so to say making order in disorder or giving concreteness to abstraction. A boundary divides the space into two. However, the divided two spaces share a boundary. Therefore I think when the idea of “dividing” comes up we can always find a sense of “sharing” as well. As a starting point I believed that when the notion of “connecting” is added to boundary there could be a new value in the space. I surveyed how the idea of setting boundary in architecture has been changed through history. In my research I reached the concept of “gradational boundary” which divides a space but also connects the space at the same time. Furthermore I designed a piece of furniture which functions both as a shelf and as a room divider based on this concept.

The purpose of this thesis is to study the problem of a low Reynolds number
swimmer that is in very close proximity to a wall or solid boundary in a non-
Newtonian fluid. We assume that it moves by propagating waves down its length
in one direction, creating a thrust and therefore propelling it in the opposite
direction. We model the swimmer as an infinite, inextensible waving sheet.
We consider two main cases of this swimming sheet problem. In the first
case, the type of wave being propagated down the length of the swimmer is
specified. We compare the swimming speeds of viscoelastic shear thinning,
shear thickening and Newtonian fluids for a fixed propagating wave speed. We
then compare the swimming speeds of these same fluids for a fixed rate of work
per wavelength. In the latter situation, we find that a shear thinning fluid
always yields the fastest swimming speed regardless of the amplitude of the
propagating waves. We conclude that a shear thinning fluid is optimal for the
swimmer. Analytical results are obtained for various limiting cases. Next, we
consider the problem with a Bingham fluid. Yield surfaces and flow profiles are
obtained.
In the second case, the forcing along the length of the swimmer is specified,
but the shape of the swimmer is unknown. First, we solve this problem for a
Newtonian fluid. Large amplitude forcing yields a swimmer shape that has a
plateau region following by a large spike region. It is found that there exists
an optimal forcing that will yield a maximum swimming speed. Next, we solve
the problem for moderate forcing amplitudes for viscoelastic shear thickening
and shear thinning fluids. For a given forcing, it is found that a shear thinning
fluid yields the fastest swimming speed when compared to a shear thickening
fluid and a Newtonian fluid. The difference in swimming speeds decreases as
the bending stiffness of the swimmer increases.

Slices are digraphs that can be composed together to form larger digraphs.In this thesis we introduce the foundations of a theory whose aim is to provide ways of defining and manipulating infinite families of combinatorial objects such as graphs, partial orders, logical equations etc. We give special attentionto objects that can be represented as sequences of slices. We have successfully applied our theory to obtain novel results in three fields: concurrency theory,combinatorics and logic. Some notable results are: Concurrency Theory: We prove that inclusion and emptiness of intersection of the causalbehavior of bounded Petri nets are decidable. These problems had been open for almost two decades. We introduce an algorithm to transitively reduce infinite familiesof DAGs. This algorithm allows us to operate with partial order languages defined via distinct formalisms, such as, Mazurkiewicztrace languages and message sequence chart languages. Combinatorics: For each constant z ∈ N, we define the notion of z-topological or-der for digraphs, and use it as a point of connection between the monadic second order logic of graphs and directed width measures, such as directed path-width and cycle-rank. Through this connection we establish the polynomial time solvability of a large numberof natural counting problems on digraphs admitting z-topological orderings. Logic: We introduce an ordered version of equational logic. We show thatthe validity problem for this logic is fixed parameter tractable withrespect to the depth of the proof DAG, and solvable in polynomial time with respect to several notions of width of the equations being proved. An attempt to renegotiate the typology of a functionally separate administrative building by integrating the stacked infrastructure of movement built into the site and introducing new programs that are naturally occurring in Tensta in spite of the inflexible urban context. The building signifies a point of convergence between the different types of traffic and bridges between them, revealing latent connectivity and opening up for a gradual take-over of traffic systems as general patterns of behaviour change. The will to make the building an urban move stems from thoughts about the welfare state architecture as a continuous megastructure, including the subway, Systembolag, Folktandvården, and much more. The municipality building generally separates itself from this continuity, physically and in turn in the public’s perception of it. The goal was to investigate the implications when it comes to how a member of the public perceives a more municipality building that is more integrated in the urban fabric.Mitt projekt utgår från tankar kring välfärdsstatens arkitektur och hur den generellt ser ut och upplevs. Det finns många delar av det som vi inte reflekterar över utan bara passerar genom, i tunnelbanan, systembolag etc. medans det i andra fall blir mer påtagligt. Att besöka en myndighetsbyggnad är ett sådant tillfälle. Oavsett retoriken som används är en myndighetsbyggnad inte en aktivt demokratisk plats, utan en plats dit medborgarna går för att vara passiva mottagare av politiska beslut. Vid inträdandet passeras en subtil gräns där individen går från sin vanliga autonomi till att sätta sig i händerna på ett kollektivt strukturerande system. Den här gränsen där individen gör ett utträde ur sin upplevda individualism och tar steget in det kollektiva är en viktig gestaltningsfråga. I mitt projekt ville jag ifrågasätta stadsdelshusets typologi som funktionsseparerad byggnad och snarare se det som infrastrukturobjekt där rörelsen genom byggnaden och inslag av vardag skulle kunna marmorera strukturen och vara dess huvudsakliga motiv. Min förhoppning var att det skulle kunna förmildra känslan av att träda in i en främmande sfär där man gör avkall på sin autonomi.

Tissue slices can be made from organs, such as liver, kidney, brain, and heart, and from various species including humans, dogs, non-human primates, rats and mice. It has been demonstrated that human and rat liver slices are viable for up to 2 days, and liver slices have been extensively used as an in vitro method to study hepatic drug metabolism and toxicity in humans. The objective of this study was to determine the utility of canine hepatic slices as an in vitro model for studying drug metabolism and hepatotoxicity in dogs. Canine hepatic slices were incubated in media containing various drugs to determine the hepatotoxicity of the agents and the ability of the slices to metabolize the drugs. The toxicity of phenobarbital, primidone, lidocaine and carprofen to canine hepatic slices was assessed by determining changes in supernatant concentrations of potassium ions and adenosine triphosphate (ATP); histologic lesions were determined as necrosis, extent of vacuolation and severity of vacuolation. Xenobiotic drug metabolizing enzymatic activity was investigated by determining the metabolism of lidocaine to monoethylglycinexylidide (MEGX), and administration of phenobarbital plus primidone was used as a positive control for hepatotoxicity in dogs. The function of drug-metabolizing enzymes was demonstrated by the successful metabolism of lidocaine to MEGX. Carprofen, a drug which causes idiosyncratic hepatic disease in dogs, did not show any hepatotoxicity at concentrations of 10, 50 and 100 µg/ml using potassium ion levels, ATP concentrations and histology as indicators of hepatotoxicity. Slices incubated in media without drug showed no toxicity over 24 hours based on potassium ion and ATP supernatant concentrations while significant increases in histologic lesions were noted at 8, 12 and 24 hours. Canine hepatic slices were a useful model for examining drug metabolism and toxicity for up to 24 hours.

Nespornou vyhodou počítačem podporované výroby (CAM) je výrazná časová úspora při přípravě obráběcího programu. Asociativita je jedním z řešení, její místo v rámci přípravy výrobku se nachází mezi hlavní konstrukcí a technologií podniku. Pro studium asociativity byl použity CAD/CAM modely kompresoru, který díky své komplexní geomtrii posloužil jaky dobrý příklad. Tato práce se podtrhuje omezené možnosti asociativity, především pokud jde o realizaci obráběcího programu pro součást tvořenou více prvky. Dále v této studii budou prezentovány jisté možnosti zlepšení procesu. Studie asociativity v této práci je zaměřena především na obrábění, tedy její aplikace jsou využívány zejména ve oborech jako je automobilový průmysl, letectví, kosmický průmysl či stavebnictví. Kompresor studovaný v této práci je vyroben ze slitiny titanu Ti-6Al-4V a jedná se o součást využívanou v kosmickém průmyslu. Jelikož se jsou jeho rozměry velmi malé, jsou pro jeho obrábění nezbytné velmi přesné nástroje a vysoké řezné rychlosti. Tato práce představuje odlišné strategie obrábění navrhnuté pro výrobu kompresoru, společně s analýzou výsledků. Po počáteční přípravě výroby následují dvě hlavní etpy : editace programu v CATII V5 a jeho ověření v NCSimul8.

This Master’s thesis is focused on theoretical study of the high entropy alloy CoCrNi using ab initio calculations. The focus was on the effect of short range order on the relative stability of FCC and HCP structures and the value of stacking fault energy.The results show increase of stability in both types of structures wtih decreasing number of Cr-Cr nearest neighbours. The effect of the number of Cr-Cr nearest neighbours on the stacking fault energy previously shown in literature was not observed. However the strong dependency was found on the change of short range order caused by the shift of (1 1 1) planes after the transformation from the FCC to HCP structure. The effect of interstitial atoms C a N was also studied. Both these interstitials stabilise FCC structure and thus cause the increase of stacking fault energy. Both interstitials prefer octahedral positions with higher amount of Cr in their nearest neighbour shell.

Optimalizace procesu vrtání kovových materiálů vyžaduje minimální tvorbu otřepů a jednotný vzhled vrtaných děr. Cílem práce bylo pochopit roli různých klíčových faktorů, jako řezných podmínek, způsobu upnutí obrobku a geometrie vrtáku, na tvorbu otřepů při vrtání 2 mm plechů tvářené hliníkové slitiny Al99.7Mg0.5Cu-H24, za použití vrtáků o průměru 1,6 a 2 mm, pro dosažení minimalizace tvorby otřepů a zajištění jednotnosti získaných povrchů pro velký počet vrtaných děr v obrobku. Tohoto cíle bylo dosaženo provedením 3 experimentálních testů. První experimentální test byl proveden za účelem sledování vlivu řezné rychlosti a posuvu za otáčku, jakožto rozpoznaných řezných parametrů nejvýznamněji ovlivňujících tvorbu otřepů, na výsledné velikosti vzniklých otřepů a jednotnosti vrtaných děr. V druhém experimentu byl zkonstruován vakuový upínací přípravek, za účelem upnutí vrtaného hliníkového plechu a omezení prostoru pro tvorbu otřepů na výstupní straně vrtání na obrobku, a jeho vlivu na tvorbu otřepů bylo vyhodnoceno. V třetím testu byl vyhodnocen efekt rozdílných geometrií vrtáků na tvorbu otřepů, spolu s dosaženou jednotností vrtaných děr. Výsledky z prvního experimentu prokázaly, že výška a tloušťka otřepů je redukována na obou stranách vrtaného plechu za použití vyšších řezných rychlostí. Při použití vyšších posuvů byla viděna zvětšující se výška a tloušťka otřepů na obou stranách vrtání, s výjimkou výšky otřepů na vstupní straně vrtáku do obrobku, která byla minimálně snížena. Druhý experiment prokázal, že vhodně konstruovaný vakuový upínací přípravek může významně omezit vznik otřepů na výstupní straně vrtáku z obrobku. V třetím experimentu, za použití 3 břitého vrtáku a vhodně konstruovaného vakuového upínací ho přípravku, bylo dosaženo eliminace tvorby otřepů z obou stran vrtaného plechu, společně s naplněním požadavků na jednotný vzhled vrtaných děr a vysoké výrobní produktivity. Takovýto optimalizovaný proces, bez nutnosti použití přídavné operace na odstranění vzniklých otřepů, poskytuje významnou redukci výrobních nákladů. Mimoto jsou poskytnuta doporučení pro další výzkum k dosažení zlepšení ve výrobě.

Master's thesis deals with consideration possibility of conventional and unconvetional welding methods of wrought magnesium alloys. Methods of Friction Stir Welding, Pulsed Nd:YAG laser and Continuous Fiber laser was used to join magnesium alloy sheets AZ31, AZ61, ZE10 and ZE41. For studying influence of processing parameters on microstructure and mechanical properties were used methods of light microscopy, scanning electron microscopy, tensile tests and microhardness measurements. The results of tensile tests show that from the point of view of keeping ultimate tensile strength is the best methods for welding magnesium alloys Friction Stir Welding. Compared with Nd:YAG laser, fiber laser give better properties of welds. From comparing microstructure and mechanical properties . It wasn't found publications about Friction Stir Welding of magnesium ZE alloys or about FSW sheets with thickness only 0,8 mm and 1,6 mm.

This diploma thesis is focusing on possibilities of influencing casting’s structure of nickel superalloy Inconel 713LC by force effects caused by vibration and rotation during crystallization. In this research were cast three test castings of turbine wheels with different solidification conditions that have affected the size and distribution of the grains. The aim was to compare the different microstructure and macrostructure of castings and then compare the dependence of grain refinement on mechanical properties. Changes in the structure were analyzed by means of light and electron microscopy methods after the fracture of test bars.
