Abstract:
The Covid-19 crisis lockdown caused rapid transformation to remote working/learning modes and the need for e-commerce-, web-education-related projects development, and maintenance. However, an increase in internet traffic has a direct impact on infrastructure and software performance. We study the problem of accurate and quick web-project infrastructure issues/bottleneck/overload identification. The research aims to achieve and ensure the reliability and availability of a commerce/educational web project by providing system observability and Site Reliability Engineering (SRE) methods. In this research, we propose methods for technical condition assessment by applying the correlation of user-engagement score and Service Level Indicators (SLIs)/Service Level Objectives (SLOs)/Service Level Agreements (SLAs) measurements to identify user satisfaction types along with the infrastructure state. Our solution helps to improve content quality and, mainly, detect abnormal system behavior and poor infrastructure conditions. A straightforward interpretation of potential performance bottlenecks and vulnerabilities is achieved with the developed contingency table and correlation matrix for that purpose. We identify big data and system logs and metrics as the central sources that have performance issues during web-project usage. Throughout the analysis of an educational platform dataset, we found the main features of web-project content that have high user-engagement and provide value to services’ customers. According to our study, the usage and correlation of SLOs/SLAs with other critical metrics, such as user satisfaction or engagement improves early indication of potential system issues and avoids having users face them. These findings correspond to the concepts of SRE that focus on maintaining high service availability.

Abstract:
ABSTRACT Streptococcus mutans belongs to the viridans group of oral streptococci, which is the leading cause of endocarditis in humans. The LraI family of lipoproteins in viridans group streptococci and other bacteria have been shown to function as virulence factors, adhesins, or ABC-type metal transporters. We previously reported the identification of the S. mutans LraI operon, sloABCR, which encodes components of a putative metal uptake system composed of SloA, an ATP-binding protein, SloB, an integral membrane protein, and SloC, a solute-binding lipoprotein, as well as a metal-dependent regulator, SloR. We report here the functional analysis of this operon. By Western blotting, addition of Mn to the growth medium repressed SloC expression in a wild-type strain but not in a sloR mutant. Other metals tested had little effect. Cells were also tested for aerobic growth in media stripped of metals then reconstituted with Mg and either Mn or Fe. Fe at 10 μM supported growth of the wild-type strain but not of a sloA or sloC mutant. Mn at 0.1 μM supported growth of the wild-type strain and sloR mutant but not of sloA or sloC mutants. The combined results suggest that the SloABC proteins transport both metals, although the SloR protein represses this system only in response to Mn. These conclusions are supported by 55Fe uptake studies with Mn as a competitor. Finally, a sloA mutant demonstrated loss of virulence in a rat model of endocarditis, suggesting that metal transport is required for endocarditis pathogenesis.

Abstract:
Purpose The purpose of this paper is to describe the student learning outcomes (SLOs) for a sustainability major, evaluate faculty incorporation of the SLOs into the courses in the sustainability major curriculum and measure student performance on the SLOs from entry into the major to the senior capstone course. Design/methodology/approach Through an iterative approach with a faculty advisory committee, SLOs were developed for the sustainability major. Curriculum mapping followed by evaluation of course syllabi were used to determine the extent to which each course addressed the SLOs. Student performance on most SLOs was measured through student assessment in an introductory and capstone course to evaluate the change in performance over time. Findings The core courses of the sustainability major were more likely to address the SLOs of the major than that of the elective courses. Where measured, student performance on the SLOs increased from the introductory course to the capstone course. Sustainability majors participated in an average of almost ten experiential learning opportunities focused on sustainability. Originality/value This research provides a longitudinal assessment of student learning in an undergraduate sustainability major. Because undergraduate sustainability degrees are generally new, this research can serve as a base upon which to continue to improve sustainability curriculum design.

Abstract:
Social licence to operate (SLO) is an informal agreement that infers ongoing acceptance of an industrial or energy project by a local community and the stakeholders affected by it. Negotiation of SLOs centrally implicates language and communication, including scientific language and concepts. We first review the literature about the definition and communicative features of SLOs, and their relation to scientific communication. We describe communication accommodation theory and the ways that it can help understand (un)successful SLO negotiation, and describe examples of texts that show accommodative or nonaccommodative language around SLOs. We summarize some results which help indicate different ways of accommodating communities in the negotiation of SLOs. Finally, we describe a research agenda on communication accommodation and SLOs, in the service of improving their impact on energy, the environment, and the transfer of science.

Tregs are essential for maintaining peripheral tolerance, and thus targeting these cells may aid in the treatment of autoimmunity and cancer by enhancing or reducing suppressive functions, respectively. Before these cells can be harnessed for therapeutic purposes, it is necessary to understand how they maintain tolerance under physiologically relevant conditions. We now report that transcription factor Kruppel-like factor 2 (KLF2) controls naive Treg migration patterns via regulation of homeostatic and inflammatory homing receptors, and that in its absence KLF2-deficient Tregs are unable to migrate efficiently to secondary lymphoid organs (SLOs). Diminished Treg trafficking to SLOs is sufficient to initiate autoimmunity, indicating that SLOs are a primary site for maintaining peripheral tolerance under homeostatic conditions. Disease severity correlates with impaired Treg recruitment to SLOs and, conversely, promotion of Tregs into these tissues can ameliorate autoimmunity. Moreover, stabilizing KLF2 expression within the Treg compartment enhances peripheral tolerance by diverting these suppressive cells from tertiary tissues into SLOs. Taken together, these results demonstrate that peripheral tolerance is enhanced or diminished through modulation of Treg trafficking to SLOs, a process that can be controlled by adjusting KLF2 protein levels.

Abstract:
Scanning laser ophthalmoscopes (SLOs) have been available since the early 1990s, but they were not commonly used because their advantages were not enough to replace conventional color fundus photography. In recent years, color SLOs have improved significantly, and the colored SLO images are obtained by combining multiple SLO images taken by lasers of different wavelengths. A combination of these images of different lasers can create an image that is close to that of the real ocular fundus. One advantage of the advanced SLOs is that they can obtain images with a wider view of the ocular fundus while maintaining a high resolution even through non-dilated eyes. The current SLOs are superior to the conventional fundus photography in their ability to image abnormal alterations of the retina and choroid. Thus, the purpose of this review was to present the characteristics of the current color SLOs and to show how that can help in the diagnosis and the following of changes after treatments. To accomplish these goals, we will present our findings in patients with different types of retinochoroidal disorders.

In acute graft-versus-host disease (aGVHD), donor T cells attack the recipient's gastrointestinal tract, liver, and skin. We hypothesized that blocking access to distinct lymphoid priming sites may alter the specific organ tropism and prevent aGVHD development. In support of this initial hypothesis, we found that different secondary lymphoid organs (SLOs) imprint distinct homing receptor phenotypes on evolving alloreactive effector T cells in vivo. Yet preventing T-cell entry to specific SLOs through blocking monoclonal antibodies, or SLO ablation, did not alter aGVHD pathophysiology. Moreover, transfer of alloreactive effector T cells into conditioned secondary recipients targeted the intestines and liver, irrespective of their initial priming site. Thus, we demonstrate redundancy of SLOs at different anatomical sites in aGVHD initiation. Only prevention of T-cell entry to all SLOs could completely abrogate the onset of aGVHD.

Abstract:
Smith–Lemli–Opitz Syndrome (SLOS) results from mutations in the gene encoding the enzyme DHCR7, which catalyzes conversion of 7-dehydrocholesterol (7DHC) to cholesterol (CHOL). Rats treated with a DHCR7 inhibitor serve as a SLOS animal model, and exhibit progressive photoreceptor-specific cell death, with accumulation of 7DHC and oxidized sterols. To understand the basis of this cell type specificity, we performed transcriptomic analyses on a photoreceptor-derived cell line (661W), treating cells with two 7DHC-derived oxysterols, which accumulate in tissues and bodily fluids of SLOS patients and in the rat SLOS model, as well as with CHOL (negative control), and evaluated differentially expressed genes (DEGs) for each treatment. Gene enrichment analysis and compilation of DEG sets indicated that endoplasmic reticulum stress, oxidative stress, DNA damage and repair, and autophagy were all highly up-regulated pathways in oxysterol-treated cells. Detailed analysis indicated that the two oxysterols exert their effects via different molecular mechanisms. Changes in expression of key genes in highlighted pathways (Hmox1, Ddit3, Trib3, and Herpud1) were validated by immunofluorescence confocal microscopy. The results extend our understanding of the pathobiology of retinal degeneration and SLOS, identifying potential new druggable targets for therapeutic intervention into these and other related orphan diseases.

Abstract:
Healthcare resource utilization peaks throughout the first year following acute myocardial infarction (AMI). Data linking the former and outcomes are sparse. We evaluated the associations between subsequent length of in-hospital stay (SLOS) and primary ambulatory visits (PAV) within the first year after AMI and long-term mortality. This retrospective analysis included patients who were discharged following an AMI. Study groups: low (0–1 days), intermediate (2–7) and high (≥8 days) SLOS; low (<10) and high (≥10 visits) PAV, throughout the first post-AMI year. All-cause mortality was set as the primary outcome. Overall, 8112 patients were included: 55.2%, 23.4% and 21.4% in low, intermediate and high SLOS groups respectively; 26.0% and 74.0% in low and high-PAV groups. Throughout the follow-up period (up to 18 years), 49.6% patients died. Multivariable analysis showed that an increased SLOS (Hazard ratio (HR) = 1.313 and HR = 1.714 for intermediate and high vs. low groups respectively) and a reduced number of PAV (HR = 1.24 for low vs. high groups) were independently associated with an increased risk for mortality (p < 0.001 for each). Long-term mortality following AMI is associated with high hospital and low primary ambulatory services utilization throughout the first-year post-discharge. Measures focusing on patients with increased SLOS and reduced PAV should be considered to improve patient outcomes.

Psychology seniors at a Midwest university reflected on the student learning outcomes (SLOs) for the major. This study examined the relations among students' perceived usefulness of the SLOs, measures of learning, ratings of self-confidence in their knowledge of psychology, and evaluations of the department's helpfulness in students' skill development. Correlations showed few significant relations between usefulness of the SLOs and measures of learning and no relations with ratings of self-confidence. Perceived usefulness of the outcomes was a significant predictor of the department's perceived helpfulness in students' skill development. However, students' perceptions of the department dropped significantly by the end of the capstone course. We discuss implications related to activities teachers might use to enhance learning.

Abstract:
Despite the prevalence of student learning objectives (SLOs) in teacher evaluation systems throughout the United States, research on the validity of student and teacher SLO scores used for high-stakes decisions is lacking. For this reason, this dissertation is comprised of two chapters that examine student and teacher-level SLO performance data from select districts in one Race to the Top state. In Chapter 1, I describe the quality of student assessment data and the comparability of student scores across alternative growth targets. I find that in the first year of implementation, assessments from half of the courses in the sample contained indicators of poor data quality, including anomalous score distributions and small to negative correlations between student prescores and postscores. However, in the second year of implementation, when student SLO performance is incorporated into final teacher evaluation scores, far fewer assessments contained anomalous score distributions, and there is no evidence to suggest manipulation of student scores. In addition to the assessments, the choice of student growth target does have an impact on the comparability of student and teacher scores across districts and years. Chapter 2 describes the validity and reliability of teacher SLO scores. I find that while teacher SLO scores are moderately stable across courses, they are not stable over time, likely due to changes made to the assessments and targets used to determine student SLO scores. Further, for teachers with both SLO scores and an alternative metric of performance based on student growth, the two metrics do not converge. Finally, teachers in courses with higher average student prescores and lower proportions of students with disabilities have slightly higher SLO scores. In general, results on teacher SLO scores were similar to those found with value-added based metrics of teacher performance. Findings from both chapters suggest that improvement in the quality of the assessments administered as well as greater consistency in the growth targets assigned to students, both within districts over time and across districts, will improve the validity of student and teacher SLO scores in this state.

Abstract:
Large-conductance-Ca2+-activated potassium Slo1 channels are well characterised members of the Slo channel family. The other members, Slo3 and Slo2.χ, are less characterised and their physiological roles are elusive. Little is known of the pH-sensitive Slo3 channel, aside from a role regulating Ca2+ influx through CatSper channels during capacitation in spermatozoa. This thesis aims to investigate mSlo3 pharmacology and determine if mSlo3- EST’s encode novel isoforms. Na+-activated Slo2.χ channels (Slo2.1/Slo2.2) are thought to mediate adaptation of neuronal action potential firing rate and slow after hyperpolarisations following repetitive firing. We aim to characterise hSlo2.2a currents and their bithionol sensitivity, an activator of a rat isoform. Mutation of the conserved phenylalanine in the S6-domain (mSlo3-F304/hSlo1-F380) to tyrosine has similar effects on Slo channels, namely a negative shift in the voltagedependence of activation and an increase in membrane conductance. mSlo3 pharmacology matches the IKSper profile and suggests roles for mSlo3 as a pharmacological target in infertility treatment. This work identified a novel splice-variant of mSlo3; CV562866 lacks transmembrane domains and is more widely expressed than mSlo3. CV562866 protein is expressed in vivo, and contains the nominal RCK2 domain. CV562866 affects functional expression of mSlo3 and mSlo1 channels in Xenopus oocytes, decreasing and increasing expression respectively. This study examined the hSlo2.2a channel characteristics, which unlike the previously reported rSlo2.2a channel, mediates a non-rectifying K+ current. Bithionol activates hSlo2.2a channels with an EC50 of 1.29±0.12μM and activation was Na+ co-operative. We found a candidate bithionol binding site in the C-terminal domain near the “Cl- bowl”. Due to toxicity of hSlo2.1 expression; determination of the bithionol selectivity of Slo2.χ channels was unattainable. In vivo, Slo2.χ channels act as a background K+ conductance, activated by a variety of conditions, stabilising the resting membrane potential of the cell. This study suggests Slo2.χ are viable pharmacological targets for treatment of inflammatory pain.

En effet, le contrôle de la répartition spatiale de la lumière grâce à l'interaction résonante entre nanostructures et ondes électromagnétiques a conduit aux développements de nombreuses applications dans des domaines variés tels que les télécommunications,la spectroscopie et la détection d'objets. Le ralentissement de la lumière, sujet de la thèse, obtenue grâces à l'interférence d'ondes contre-propageantes dans des milieux périodiques ou le confinement sub-longueur d'onde dans des guides d'ondes plasmoniques, est associé à une compression des pulses lumineux et une forte augmentation du champ électrique, deux phénomènes clés pour la miniaturisation de composées optiques et l'augmentation de l'interaction lumière matière
Enhancing light-matter interactions at micro and nanoscales is one of the spearheads of nanophotonics. Indeed, the control of the field distribution due to the resonant interaction of nanostructures with electromagnetic waves has prompted the development of numerous optical components for many applications in telecommunication, spectroscopy or sensing. A promising approach lies in the control of light speed in nanostructures. Light slowdown, obtained by wave interferences in periodic structures or subwavelength confinement in plasmonic waveguides, is associated to pulse compressions and large field enhancements,which are envisioned as key processes for the miniaturization of optical devices and the enhancement of light-matter interactions.The thesis studies both fundamental aspects and possible applications related to slow light in photonic and plasmonic nanostructures. In particular, we study the impact of periodic system sizes on the group velocity reduction and propose a novelfamily of resonators that implement slow light on very small spatial scales. We then investigate the role of fabrication disorder in slow periodic waveguides on light localization and demonstrate how modal properties influence the confinement of localized modes. Also we propose a new hollow-core photonic crystal waveguide that provides efficient and remote couplings between the waveguide and atoms thatare trapped away from it. Finally we demonstrate the important role played by slow plasmons on the emission of quantum emitters placed in nanogap plasmonic antennas and explain how large radiation efficiency can be achieved by overcoming quenching in the metal. Additionally, one part of the thesis is devoted to thederivation of a novel modal method to accurately describe the dynamics of plasmonic resonators under short pulse illumination

Esta dissertação objetiva a percepção dos enclaves do ecodesenvolvimento no movimento slow. Para este diagnóstico, contextualizam-se as teorias tradicionais e os modelos de desenvolvimento pela racionalidade instrumental, que pressupõem o mercado como única arena das necessidades humanas, sociais e ambientais. Contudo, criticam-se as conseqüências negativas do nivelamento mercantil e sua unidimensão. Vários autores respondem que o desenvolvimento deve fomentar a racionalidade substantiva para uma mudança de paradigma em oposição ao maldesenvolvimento. O ecodesenvolvimento, enquanto planejamento, volta-se ao território para alavancar o desenvolvimento com respeito à diversidade ecossistêmica e à cultura local, o suporte de tecnologias para o uso de parcimonioso de recursos naturais e de tecnologias limpas. Também, percebe as relações de demanda pelos modelos culturais da sociedade, seu estilo de vida e suas estruturas de consumo como indicadores da legitimação de outro modelo de desenvolvimento. Os movimentos slow apontam a necessidade de uma desaceleração (quebra da lógica produtiva) e o resgate de valores integrativos e colaborativos pela sociedade, considerando várias esferas de necessidades humanas. Seus projetos têm forte ligação com o desenvolvimento territorial sustentado e com a preservação ambiental e cultural das localidades. Considera-se, então, o rumo de uma globalização virtuosa, em que se respeitem as pequenas produções e o comércio local com intuito da não massificação cultural e utilização responsável dos recursos.
This thesis aims at understanding the enclaves of ecodevelopment in slow movement. For this diagnosis, contextualized the traditional theories and models of development by instrumental rationality, which require the market as the only arena of human needs, social and environmental. However, criticize it the negative consequences of race and one-dimension market. Several authors reply that the development should encourage the substantive rationality for a change of paradigm in opposition to bad-development. The ecodevelopment as planning, returns to the territory to leverage the development with respect to ecosystem diversity and local culture, support technologies for the thrifty use of natural resources and clean technologies. Also, understands that the relationship of demand for society's cultural models, its lifestyle and its structures of consumption as indicators of legitimacy of another model of development. The slow movements suggest the needs for a deceleration (decline of productive logic) and the redemption of integrative value and collaborative by the society considering various spheres of human needs. Its projects have strong links with territorial sustainable development with environmental and cultural preservation of locations. It considers that the direction of a virtuous globalization, where is respect for small local production and trade with intention of no mass culture and responsible use of resources.

Abstract:
Research into how one can create architecture that will last beyond the foreseeable future. Part ONE is research based, building up a manifesto based on; structural integrity, aesthetic longevity, programmatic adaptability and cultural relevance. The research is based on observing historical architecture from the past 2,000 years, both looking into how the buildings have stood the test of time physically, but also why they might remain attractive, analysing the comings and goings of beauty through time. The research is compiled and represented through suggested ideal ways of designing each of the architectural elements, such as the wall, window, door, threshold or stair. Part TWO applies the research to a case study; the extension and renovation of the Royal Palace in Stockholm, in order to turn the residence into a public building, with the extension functioning as a connection point between the city landscape and the palace beyond. The design has no specific programme other than allowing the palace complex to be used by the public, as they wish, for the future to come. A large part of the design is essentially made up of sculptural urban landscaping, colonnaded and vaulted walkways both interacting with the waterfront, and bringing the public up to the level of the palace which today stands behind tall walls. Whilst internally the new building functions as an entrance hall with necessary facilities, and creates a route into the existing palace.

Like the ever-circling lines in the skin of trees, sometimes the whole of a person is peeled back, layer by layer, until only seeds remain. Names, faces, stories, and relationships are unmade and molded into new shapes. Without warning, those left at the base, at the roots, can’t recognize this maple’s form. They hold a pile of leaves, a bottle of glue, and the hope that something familiar will take sprout again. The tree becomes new, and its tangled branches reach out. These poems explore the lives of those living with and caring for those with dementia and Alzheimer’s disease. The hope is that they will touch on all the layers that make up a person, and the connection that nature and materiality have with building memory.

Ett av skolans viktigaste uppdrag är att lära eleverna att läsa och skriva, vårt samhälles kommunikationssystem litar på att alla har dessa färdigheter. (Lundberg, 2003) På skolorna finns det en mängd olika kartläggningsscheman som ska vara en hjälp för läraren att dokumentera elevernas läsutveckling så att skolsystemet inte missar elever som fastnar i utvecklingen. Ett nytt sådant kartläggningsschema som utvecklas i Älmhults kommun är SLUS, språk- och läsutvecklingsschema. Studiens syfte är dels att belysa utvecklingen av SLUS samt införandet av materialet i en rad kommuner dels att synliggöra hur verksamma pedagoger upplever arbetet med materialet SLUS. Jag använder mig av semistrukturerade intervjuer för att genomföra studien. SLUS är enbart ett kartläggningsschema med det finns ett material, God läsutveckling i praktiken, där det finns uppgifter som väl kan användas tillsammans med schemat för att bedöma barnets kunskaper. Tillsammans är det ett material som används aktivt i både Alvesta och Älmhults kommun och många pedagoger är mycket positiva till materialet.

Abstract:
Cloud computing providers offer multiple service classes to deal with workload heterogeneity. Classes are distinguished by their expected Quality of Service (QoS), which is defined in terms of Service Level Objectives (SLO). A priority-based scheduling policy is commonly used to guarantee that requests submitted to the different service classes achieve the desired QoS. However, the QoS delivered during resource contention periods may be unfair to certain users. In this paper, we present a SLO-driven scheduling policy which takes the SLOs and actual QoS delivered for each request into account when making decisions. We used simulation experiments fed with traces from a production system to compare the SLO-driven policy with a priority-based one. In general, the SLO-driven policy delivered a better service than the priority-based one.
