The deuterium content of sugar in fruits is influenced mainly by two factors – the botanical and also the geographical origin of the fruit. The information of the fruit origin is kept even after fermentation of the sugar, in the molecules of ethanol. The deuterium content on the methyl group of ethanol then varies depending upon the botanical origin of the fruit. The aims of this study were to characterise the molecular function of LSH and attempt to relate this to its role in DNA methylation <i>in vivo</i>. To address these aims the work described in this thesis uses a variety of approaches to address two key questions. (1) Is LSH an active SNF2 ATPase? (2) How does LSH interaction with, and modulate, the DNA methylation machinery? To determine if LSH is an active SNF2 ATPase, recombinant LSH was purified from insect cells and biochemically characterised. Genes encoding subunits of the SWI/SNF chromatin remodeling complex are frequently mutated in a wide variety of cancers, but the mechanism of cancer formation following SWI/SNF subunit mutation is not known. As a result, improved understanding of how mutation of these subunits promotes oncogenesis could have broad relevance for human cancer therapy. In this dissertation, two distinct lines of investigation were pursued with the common goals of understanding the mechanisms by which SWI/SNF subunit mutation contributes to cancer and identifying vulnerabilities specific to SWI/SNF-mutant cancers.
