They are type 1 and type 2 phosphatases, which are further classified as phosphatase 2A, 2B, and 2C. In this study, endogenous brain phosphatases that dephosphorylate sodium channels were partially purified and characterized. The thesis investigates pipeline plugging mechanisms that have occurred during interim stabilization transfers at Hanford. A laboratory-scale saltwell pumping test loop was designed to evaluate a surrogate of Hanford Tank 241-SX-104 supernate. The effect of surrogate flow rate, cooling water flow rate and phosphate concentrations on plugging mechanisms was investigated. Critical parameters like particle and agglomerate size, velocity and bed growth rate were determined. Theoretical models were used to compare the experimental pressure rise and temperature drop of the surrogate in the channel.

Inorganic phosphate (Pi) homeostasis is mainly regulated by absorption of dietary Pi in the small intestine and reabsorption of filtered Pi in the kidney. I have characterized the 5 '-flanking region and overall gene structure of the murine type IIb sodium-phosphate cotransporter (Na/Pi-IIb), a major apical Pi transporter. The Na/Pi-IIb cotransporter gene spans more than 18 kb and consists of 12 introns and 13 exons. Three promoter/reporter gene constructs, -159/+73, -429/+73 and -954/+73, showed significant luciferase activity when transfected into in rat intestinal epithelial (RIE-1) cells.

Sodium phosphate (NaP) is a low-volume, hyperosmolar laxative that has been shown to be an effective bowel cleansing agent in people. The purposes of this study were to evaluate the safety and efficacy of oral NaP in dogs. Standard (NaP and enemas; NaP1) and control preparations (polyethylene glycol [PEG] and enemas) were compared in a crossover design to determine safety and efficacy of NaP. To determine its precise role in the maintenance of Pi homeostasis, we cloned and characterized the renal-specific Na$ sp+$-Pi cotransporter/Npt2 gene and generated a gene targeting vector for the creation of a knockout mouse. The gene was cloned by screening a genomic DNA library with a rat Npt2 cDNA probe. The Npt2 gene is approximately 17kb and contains 13 exons and 12 introns. Processed turkey rolls were prepared with 1 or 3% whey protein concentrates WPC-50 (pH=5. 8 0) , WPC-60 (pH=4. 53) and WPC-75 (pH=6.85) containing 50, 60 and 75% protein along with controls (phosphate and no phosphate) . Control rolls made with 0.5% phosphate had the highest bind strength, and sensory evaluation scores. Only WPC-75 (1%) was acceptable as a binding agent and flavor enhancer. WPC-60 reduced pink discoloration of rolls, but flavor, bind and cohesiveness scores were unacceptably low. WPC-50 was not an effective binding agent. T cell protein tyrosine phosphatase (TC-PTP / PTPN2) is an enzyme that is essential for the proper functioning of the immune system and that participates in the control of cell proliferation, and inflammation. We previously observed that TC-PTP-/- mice display various immunodeficiencies, hypersensitivity to lipopolysaccharide (LPS) and die within three weeks of birth due to anemia and widespread inflammation. A recent analysis of the Wellcome Trust Case Control Consortium (WTCC) genome wide scan data, reported in 2007, indicated a potential role for TC-PTP in inflammatory bowel disease (IBD).
