Partindo de narrativas de mulheres de camadas médias, com alta escolaridade, profissionalizadas, "solteiras", sem filhos e que moram sozinhas, analiso noções gerais relacionadas à sociabilidade em sentido amplo, com especial ênfase à sexualidade e à amizade, e aos significados atribuídos à solidão, problematizando o valor social concedido ao par conjugal como símbolo de intimidade. À luz do ideário feminista de liberdade, igualdade, independência e autonomia, que ganhou força e se expandiu a partir dos anos 1960 nas "sociedades ocidentais modernas", as noções analisadas remetem a mudanças e permanências, revelando um cenário, ainda pouco explorado, das grandes cidades brasileiras.

Resumo: O artigo apresenta um estudo antropológico sobre integrantes dos grupos de ajuda mútua "Mulheres que amam demais anônimas", sediados na cidade do Rio de Janeiro. As entrevistadas apresentam um perfil social heterogêneo; autodenominam-se heterossexuais; e se concentram na faixa etária entre 40 e 50 anos. As narrativas assinalaram: dedicação intensa ao relacionamento; necessidade de controle do parceiro; medo da solidão; e sentimento de "baixa autoestima". A interação conjugal é marcada por competição e conflitos acerca da reciprocidade de atenção e cuidados. Essas mulheres parecem reatualizar um modelo de comportamento feminino tradicional, apesar de sua familiaridade com a proposta igualitária de conjugalidade.

A emigração internacional em Governador Valadares e região, iniciada na década de 1960, cujo ápice se deu na de 1980, continua como importante processo de deslocamento populacional, apesar das políticas restritivas impostas pelos Estados Unidos. Até a década de 1990, a emigração masculina cresceu consideravelmente, induzindo a formação de novas configurações familiares e novas maneiras de organização das rotinas diárias pelas parceiras que permaneceram no Brasil. O presente estudo investiga o sentimento de solidão vivenciado por mulheres de emigrantes submetidas ao isolamento conjugal decorrente da ida do parceiro para o exterior como emigrante. Trata-se de um levantamento no qual foram realizadas entrevistas domiciliares guiadas por um Roteiro Semiestruturado de Entrevista junto a 247 mulheres residentes na cidade de Governador Valadares/MG. A amostra foi intencional, não probabilística, os dados de natureza quantitativa foram submetidos à análise descritiva, e os de natureza qualitativa (fragmentos dos discursos) à Análise de Conteúdo proposta por Bardin. Embora o projeto de emigrar seja construído pelo núcleo familiar e apoiado pela Cultura Migratória característica da região, há evidências de que as entrevistadas vivenciam considerável sofrimento psíquico, sugerindo que o processo emigratório traz implicações indesejáveis para todos os envolvidos.

O presente estudo teve como objetivo compreender o que foi denominado pelas mães, nas redes sociais, de "Lado B da maternidade". Foi realizado um estudo qualitativo envolvendo falas de mães, ao longo do primeiro ano de vida do bebê, extraídas de 101 blogs, que tinham como tema central o "Lado B da maternidade". As falas das mães nos blogs foram analisadas por meio de análise temática e expressaram sentimentos de desamparo, solidão e ambivalência, característicos do período de dependência. Há um confronto das expectativas sociais idealizadas em relação à maternidade com a experiência materna cotidiana com o bebê, que acaba gerando na mãe, também, sentimentos de culpa por não estar correspondendo ao ideal social. As mães relataram dificuldades em lidar com as exigências que perpassam sua vida pessoal, social e conjugal. Os resultados sugerem que nos espaços de blogs com referências explicitas ao "Lado B" da maternidade as mulheres falam sobre aquilo que, normalmente, permanece oculto e negado no discurso social, rompendo, assim, com a visão idealizada da maternidade. Poder falar livremente sobre aspectos da experiência da maternidade usualmente cindidos e negados – o lado B da maternidade –, oportuniza, para além do alívio, diminuição de angústia e de culpa.

Este estudo tem como objetivo realizar uma revisão integrativa de artigos nacionais e internacionais sobre o tema da transição para a paternidade publicados entre 2006 e 2014. Foram consultadas as bases de dados EBSCO, PsycINFO, SciELO Regional e PEPSIC, resultando em 25 artigos indexados, que foram analisados em relação ao ano de publicação, país, metodologia, temática e resultados. Os resultados foram agrupados em quatro grandes grupos, intitulados: “o pai consigo mesmo”, “o pai, a mãe e o bebê”, “o pai e a rede de apoio formal” e “o pai, o trabalho e a sociedade”. No grupo, “o pai consigo mesmo”, destaca-se a vivência da paternidade como uma revolução interna, acompanhada de intensa sobrecarga emocional, ambivalência, solidão e idealização da relação pai-bebê. No grupo, “o pai, a mãe e o bebê”, percebe-se uma intensa transformação na vida conjugal, no qual o pai tende a ser mais ativo na relação com o bebê, porém modulado pela presença materna. No grupo “o pai, o trabalho e a sociedade”, percebemos a falta de preparo dos profissionais para lidar com as demandas paternas, bem como a inexistência de políticas sociais e de saúde voltadas à relação pai-bebê. No grupo “o pai, o trabalho e a sociedade”, destacamos o uso de redes informais de apoio e o surgimento de modelos horizontais de identificação paterna. Foram discutidos tendências e hiatos nas pesquisas atuais no campo da transição para a paternidade, no sentido da constituição de um novo pai.

A partir do final da década de 1940 do século XX, a prosa moderna brasileira foi impulsionada por um grupo de escritores que começava a definir o núcleo da ficção curta urbana. Como fruto desse momento, o conto se firma e amplia seus universos conteudísticos, visitando desde temáticas urbanas até os temas regionalistas, do relato documental às investidas intimistas. Dentre as temáticas urbanas e coletivas destacamos a velhice. Essa temática está presente nas narrativas de vários representantes da linha média que caracterizou a ficção brasileira dos anos de 1950 até essas primeiras décadas do século XXI. Das trinta histórias que constituem A guerra conjugal (1969) do contista curitibano Dalton Trevisan, algumas trazem protagonistas na velhice, expondo-as em uma fase do casamento na qual a solidão se instala incisiva e seca. Se em outras narrativas o malogro do casamento é espicaçado pelas ironias constantes, nos enredos em que a velhice é temática central o silêncio predomina como significado maior do distanciamento dos casais. Em "Batalha de bilhetes", a temática da velhice é problematizada através dos personagens protagonistas João e Maria. O conto joga alternadamente com a voz do narrador, que introduz o cenário e descreve as ações das personagens; com o diálogo entre eles, curto e estéril, sugerindo a falta de comunicação; e com a linguagem dos bilhetes, elemento primordial para nossos comentários e, como o próprio título incita, "instrumento bélico" usado pelos personagens. Neste artigo procuramos analisar as marcas que diferenciam as perspectivas adotadas e como a distância do discurso constrói a imagem da velhice isolada do casal. Para a leitura utilizaremos Bosi (1988) sobre a velhice e na análise do plano do enunciado e do plano da enunciação.

Portugal é um dos países europeus mais envelhecidos. A solidão e o isolamento nos idosos podem desencadear enorme sofrimento, com grande impacto na sua vida e na sua saúde. Este trabalho, pretende descrever e analisar a solidão e o isolamento em idosos em Portugal, identificando os determinantes e as estratégias adotadas no combate à solidão. É uma revisão sistemática da literatura. Os termos de pesquisa utilizados foram “solidão”,“isolamento” e “idoso”, nas bases de dados científicos RCAAP e Scielo, com intervalo temporal 2010-2018. Após aplicação dos critérios de inclusão e exclusão e avaliação da qualidade dos artigos, resultaram 8 artigos. Maioritariamente utilizaram metodologias quantitativas, com amostragem não probabilística. Foram identificados em todos os estudos baixos níveis de solidão e isolamento. Os determinantes de solidão e isolamento identificados nesta revisão são a morte do conjugue (50%), seguido da perda ou enfraquecimento da relação com os filhos (37,5%), o ser mulher (37,5%), o viver só (37,5%), a incapacidade de desempenho das AVD (37,5%), o aumento da idade (25%) e a presença de doenças (25%) e as carências económicas (12,50%). Estão presentes a adoção de adoção de mecanismos ou estratégias de combate à solidão e isolamento, que evidenciam a procura de atividades de relação, de inclusão e por outro lado, uma certa acomodação ao viver só. Concluímos que os idosos portugueses estudados apresentam em geral níveis baixos de isolamento e solidão. Sendo os principais determinantes o sexo, a idade, a presença do conjugue, o viver só, as relações com os filhos, a autonomia no desempenho das AVD, as comorbilidades e as carências económicas. Como estratégias de combate ou controlo da solidão surge a procura de atividades de relação, de inclusão e a adaptação ou acomodação ao viver só. O conhecimento desta realidade é de crucial importância ao nível da tomada de decisão.

La motivación pastoral que siempre animó a la figura de K. Wojtyla antes de ser papa, y más tarde como papa Juan Pablo II, le llevó a profundizar en la ciencia ética para intentar elaborar un corpus ético cuyas raíces y principios fueran capaces de dar razón de la dignidad de integralidad de la persona. Para ello analizó con profundidad las aportaciones que Kant y Scheler habían hecho a la ciencia ética, mostrando sus logros y sus deficiencias, y planteó la conveniencia de otra propuesta ética basada en las relaciones de interpersonalidad en el amor, tomando la fidelidad como elemento de contraste para probar su solidez. Aparece así un contexto novedoso en el estudio de la ética que conjuga lo mejor de la tradición filosófica con elementos radicalmente nuevos, como las nociones wojtylianas de antropología adecuada, teología del cuerpo, norma personalista, o hermenéutica del don, por citar sólo algunas. Su visión integradora conjugó magníficamente la dimensión filosófica y teológica de la antropología, sobre la base de la analogía del amor entre un Amor originario —Misterio trinitario— y un amor participado en la imago Dei que es la comunión interpersonal. En esta analogía, el matrimonio y la familia constituyen un verdadero tropos del Amor intratrinitario.

O trabalho foi dividido em quatro partes. A fim de introduzir os temas e esclarecer algumas idéias sobre os objetos da pesquisa, a primeira parte trata teoricamente, sobre algumas perspectivas da solidão e depois sobre a conjugalidade, destaca-se também um recorte da história da família e do casamento com o objetivo de contextualizar a construção da experiência do casamento e da família. Na segunda parte do trabalho foi desenvolvido o método utilizado na pesquisa qualitativa de orientação fenomenológica, que visa desvelar os elementos de significados envolvidos na relação dos sujeitos com suas experiências. A pesquisa foi realizada com três casais seis cônjuges por meio de entrevistas semidirigidas. Descreveu-se o momento empírico, o procedimento de análise de dados utilizado com base no método de Gomes (1997). Na terceira parte foram apresentados os resultados, organizados em treze categorias, das quais sete delimitaram estruturas constitutivas e operantes da vida conjugal, e seis delimitaram estruturas desconstrutivas e degradantes da relação conjugal. Na quarta e última parte, foi feita uma reflexão sobre a pesquisa, com base em alguns questionamentos. A conjugalidade é vivida como uma síntese de aspectos negativos e positivos vividos a dois, quais sejam: tolerância, solidariedade, respeito, estima, amizade, respeito, amor conjugal e amor ágape e, ainda, também pela vivência da intolerância, da falta de solidariedade, falta de estima, proteção individualista e falta de amor conjugal. Já a solidão conjugal foi compreendida pela vivência de aspectos negativos e degradantes da vida conjugal como: a intolerância, a falta de solidariedade, a falta de estima, a falta de amor conjugal, atitudes de proteção individual e pela solidão individual vivida no casamento. Assim, a solidão conjugal é resultado de um apanhado de aspectos desconstitutivos que são vividos de forma exasperada na relação a dois e que podem se intensificar por questões individuais de cada um.

RESUMO - O sentimento de solidão tem vindo a tornar-se cada vez mais frequente entre os indivíduos, fruto do desenvolvimento da sociedade moderna. Especificamente, ocorrendo durante a gravidez, situação que produz na mulher importantes alterações não só físicas, como também psicológicas, familiares e sociais, deve passar a ser alvo de uma atenção especial. De um ponto de vista da Saúde Mental, o desenvolvimento saudável da gravidez exige uma rede de suporte social e uma relação conjugal satisfatórias. Tais elementos devem ser considerados como factores protectores do aparecimento de sentimentos de solidão. Pelo contrário, um baixo sentido de coerência e o aparecimento de depressão durante a gravidez são factores de risco que, inevitavelmente, irão interferir na qualidade da ligação materno-fetal, com repercussões no desenvolvimento psico-social do futuro ser humano. Este protocolo de projecto propõe um estudo transversal, exploratório e de natureza quantitativa. Engloba dois sub-estudos, focando os determinantes da solidão e o impacto desta na ligação materno-fetal, e pretende identificar associações pertinentes entre as várias dimensões envolvidas. A amostra em estudo será constituída por 202 grávidas que frequentam o Centro de Saúde de Torres Vedras. As variáveis correspondentes serão operacionalizadas através de questionários estandardizados e validados para a população portuguesa, sendo eles a Escala de Solidão da UCLA, a Escala de Satisfação com o Suporte Social, a Escala de Avaliação de Áreas da Vida Conjugal, a Escala de Ligação Materno-Fetal, a Escala de Depressão Pós-Parto de Edimburgo e o Questionário de Orientação para Viver. Espera-se identificar e caracterizar as possíveis associações entre a solidão e a satisfação com o suporte social, a satisfação conjugal, a depressão durante a gravidez e o sentido de coerência, que a explicarão, e a ligação materno-fetal, que será influenciada por ela. Os questionários serão respondidos pelas grávidas seleccionadas de acordo com critérios de inclusão e exclusão. Além das descrições estatísticas iniciais, as análises de associação serão realizadas em função das distribuições encontradas, e tendo em conta dimensões do contexto sociodemográfico. Os resultados da investigação serão divulgados num relatório final. ----------------------- ABSTRACT - The feeling of loneliness is increasing as a result of developments in modern society. Specifically occurring during pregnancy, when important changes - physical, psychological and related to the family structure and interaction with society - take place, special attention should be devoted. To maintain good mental health during pregnancy it is important to have good social support and harmonious conjugal relations, both considered as factors preventing the emergence of feelings of loneliness. By contrast low sense of coherence and depression during the pregnancy are risk factors which, inevitably, will affect the quality of the maternal-fetal attachment and have repercussions on the psycho-social development of the future individual. This protocol of draft proposes a transversal exploratory study of a quantitative nature on two sub-studies, exploring the determinants of loneliness and its impact on the maternal-fetal attachment, which intends to identify some correlations between parameters. The study sample is made up of 202 pregnant women who are patients at the Health Centre of the city of Torres Vedras. They will be selected according to criteria of inclusion and exclusion. All variables will be measured through standardized and validated surveys illustrating the Portuguese population, like the Scale of Loneliness of the UCLA, Scale of Satisfaction with the Social Support, Scale of Evaluation of Areas of the Conjugal Life, Maternal-Fetal Attachment Scale, Edinburgh Postnatal Depression Scale, and Orientation To Life Questionnaire. We expect to identify correlations between loneliness and satisfaction with social support and conjugal relations, depression during pregnancy and sense of coherence, which will explain it, and the maternal-fetal attachment, which will influence it. Beyond the initial statistical descriptions, the data analysis will be executed according to the distributions found and will be carried taking into account the socio-demographic context. The results of the survey will be published in a final report.

In condensed matter physics, atomistic first principle calculations are often necessary to achieve a microscopic understanding of the observed experimental phenomena and to make quantitative predictions of physical properties. In practice, atomic scale systems have irregularities (e.g. surface roughness) or defects (e.g. substitutional atoms or vacancies) that are too strong to be ignored or treated as small perturbations. In this thesis, we report the development of a real space DFT code for studying atomic scale systems from first principles. Our code, named MatRcal, which stands for "Matlab-based real space calculator", is developed in the technical computing language Matlab. The physics is described by density functional theory. The method itself is based on projecting the Kohn-Sham Hamiltonian on a uniform Cartesian grid. High-order finite-differencing is used to discretize the Laplacian operator. The potential due to the atomic nuclei is approximated with ab initio pseudopotentials. The pseudopotentials are generated following the procedure proposed by Troullier and Martins. We use the fully separable form introduced by Kleinman and Bylander. We argue that the method is simpler and yet has many advantages compared with conventional spectral methods. We provide relevant mathematical techniques and implementation details. In particular, we present and compare different eigensolvers used to diagonalize the Kohn-Sham Hamiltonian. We validate our software by comparing the HOMO-LUMO gaps of many organic and inorganic molecules obtained using our method with those obtained with the commercial code Gaussian. Our results are in excellent agreement. Our method gains in computational speed and algorithm parallelism, and its power in handling real space boundary conditions will be a major advantage for future applications in nanoelectronic device modelling.<br>En physique de la matière condensée, les calculs numériques sont souvent nécessaires pour parvenir à comprendre les phénomènes microscopiques observés lors d'expériences ou à prédire quantitativement des propriétés physiques. En pratique, les systèmes d'échelle atomique sont irréguliers (rugosité de surface) ou comportent des défauts (atomes de substitution ou lacunes), ce qui induit des effets trop sévères pour être ignorés ou traités comme des perturbations. Dans cette thèse, nous présentons une méthode qui permet d'étudier des systèmes d'échelle atomique à partir des lois fondamentales de la physique. Notre logiciel, nommé MatRcal, qui signifie "Matlab-based real space calculator", est développé dans le langage Matlab. La physique est décrite par la théorie de la fonctionnelle de la densité. La méthode projette l'Hamiltonien de Kohn-Sham sur un maillage Cartésien uniforme. Le calcul des différences finies est utilisé pour discrétiser l'opérateur Laplacien. Le potentiel dû aux noyaux atomiques est approximé par des pseudopotentiels non-empiriques. Les pseudopotentiels sont générés en suivant la procédure proposée par Troullier et Martins. Nous utilisons la forme séparable introduite par Kleinman et Bylander. Nous soutenons que la méthode est plus simple et pourtant présente de nombreux avantages par rapport aux conventionnelles méthodes spectrales. Nous introduisons plusieurs techniques mathématiques pertinentes à notre étude et certains détails d'implémentation. Entre autres, nous présentons et comparons plusieurs algorithmes de calcul de vecteurs propres utilisés pour diagonaliser l'Hamiltonien de Kohn-Sham. Nous validons notre méthode en comparant la largeur de bande interdite "HOMO-LUMO" de nombreuses molécules organiques et inorganiques prédites par notre méthode avec celles prédites par le logiciel commercial Gaussian. Notre méthode permet des gains en rapidité et en parallélisme, mais la possibilité de traiter des conditions limites non-périodiques sera le principal atout pour de futures simulations de dispositifs nanoélectroniques.

On s'intéresse aux méthodes de calcul par sous-domaines et aux problèmes de contact entre deux solides déformables. Après une revue détaillée des méthodes de calcul par sous-domaines, l'étude porte sur les méthodes relatives à la décomposition sans recouvrement. Les mises en oeuvre numérique de ces méthodes sont présentées. Dans le cas d'une décomposition en série, nous proposons (i) une méthode itérative de relaxation, (ii) le préconditionnement ssor relatif à la méthode du gradient conjugué pour la résolution du système condensé sur les interfaces. Des comparaisons numériques des différentes méthodes sont données. Nous montrons ensuite que les problèmes de contact entre deux solides déformables sont équivalents à la recherche d'un point selle. Grâce à cette dernière formulation, nous aboutissons à deux sous-problèmes couples pour lesquels la pression normale, sur la zone de contact, joue le rôle de paramètre de couplage. Nous proposons deux méthodes de résolution (i) une méthode de dualité dans le cas d'un contact sans frottement (ii) une méthode de penalité dualité, basée sur la méthode de pénalisation bornée et la méthode duale, dans le cas d'un contact avec frottement. Des tests de validation numérique prouvant la validité des approches retenues sont présentés

Nowadays, around a third of the agricultural production is lost due to the damage caused by pests, resulting in a significant economic impact. It is estimated that about a third of these losses are provoked by phytopathogens. Currently, the solution to these problems focuses on the use of pesticides, mainly copper derivatives, antibiotics or fungicides. These compounds are effective, but they are highly polluting. In addition, antibiotics are not allowed in many countries, because abusive use causes the rapid appearance of bacterial resistant strains. The lack of non-polluting and effective treatments for plant disease control has raised the search for new compounds. Antimicrobial peptides are one of the most promising alternatives to traditional pesticides. With this aim, in this doctoral thesis the design, synthesis and characterization of BP100 derivatives was considered<br>Actualment, es calcula que, aproximadament, un terç de la producció agrícola es perd a causa dels danys provocats per les plagues, resultant en un impacte econòmic molt important. Al voltant d’un terç d’aquestes pèrdues són provocades per fitopatògens. S'ha estudiat la introducció de modificacions com D-aminoàcids o un residu d'histidina, per tal de millorar el perfil biològic d'aquestes seqüències. Així mateix, s'han dissenyat i sintetitzat ciclolipopeptidotriazoles i pèptids conjugats derivats de BPC194. S’han identificat ciclolipopèptids i pèptids conjugats amb activitat antimicrobiana enfront patògens de plantes. Aquests resultats són la base per al desenvolupament de nous compostos antimicrobians per al tractament de malalties de plantes d’importància econòmica.<br>Plant diseases caused by bacteria and fungi are responsible for significant losses in agriculture, being one of the major factors limiting worldwide crop production. Natural and synthetic antimicrobial peptides are considered as promising candidates to be used as pesticides. In this thesis, a library of cyclolipopeptides derived from the antimicrobial peptide BPC194 incorporating a range of hydrophobic chains was synthesized. It was studied the introduction of different modifications, such as D-amino acids or a histidine residue, to improve the biological profile of these sequences. Moreover, cyclolipopeptides and peptide conjugates derived from BPC194 were designed and prepared. Cyclolipopeptides and peptide conjugates active against plant phatogenic bacteria and fungi were identified. These results are the basis for the development of new antimicrobial compounds to treat plant diseases of economic importance.

Abstract:
This thesis describes different aspects of protein interactions. Initially the function of peptides and their conjugates with small molecule inhibitors on the surface of Human Carbonic Anhydrase isoenzyme II (HCAII) is evaluated. The affinities for HCAII of the flexible, synthetic helix-loop-helix motif conjugated with a series of spacered inhibitors were measured by fluorescence spectroscopy and found in the best cases to be in the low nM range. Dissociation constants show considerable dependence on linker length and vary from 3000 nM for the shortest spacer to 40 nM for the longest with a minimum of 5 nM for a spacer with an intermediate length. A rationale for binding differences based on cooperativity is presented and supported by affinities as determined by fluorescence spectroscopy. Heteronuclear Single Quantum Correlation Nuclear Magnetic Resonance (HSQC) spectroscopic experiments with 15N-labeled HCAII were used for the determination of the site of interaction. The influence of peptide charge and hydrophobicity was evaluated by surface plasmon resonance experiments. Hydrophobic sidechain branching and, more pronounced, peptide charge was demonstrated to modulate peptide – HCAII binding interactions in a cooperative manner, with affinities spanning almost two orders of magnitude. Detailed synthesis of small molecule inhibitors in a general lead discovery library as well as a targeted library for inhibition of α-thrombin is described. For the lead discovery library 160 members emanate from two N4-aryl-piperazine-2-carboxylic acid scaffolds derivatized in two dimensions employing a combinatorial approach on solid support. The targeted library was based on peptidomimetics of the D-Phe-Pro-Arg showing the scaffolds cyclopropane-1R,2R-dicarboxylic acid and (4-amino-3-oxo-morpholin-2-yl)- acetic acid as proline isosters. Employing 4-aminomethyl-benzamidine as arginine mimic and different hydrophobic amines and electrophiles as D-phenylalanine mimics resulted in 34 compounds showing IC50 values for α-thrombin ranging more than three orders of magnitude with the best inhibitor showing an IC50 of 130 nM. Interestingly, the best inhibitors showed reversed stereochemistry in comparison with a previously reported series employing a 3-oxo-morpholin-2-yl-acetic acid scaffold.

This work offers new computational methods for the optimal control of the conjugate heat transfer (CHT) problem in thermal science. Conjugate heat transfer has many important industrial applications, such as heat exchange processes in power plants and cooling in electronic packaging industry, and has been a staple of computational methods in thermal science for many years. This work considers the Dirichlet boundary control of fluid-solid CHT problems. The CHT system falls into the category of multi-physics problems. Its domain typically consists of two parts, namely, a solid region subject to thermal heating or cooling and a conjugate fluid region responsible for thermal convection transport. These two different physical systems are strongly coupled through the thermal boundary condition at the fluid-solid interface. The objective in the CHT boundary control problem is to select optimally the fluid inflow profile that minimizes an objective function that involves the sum of the mismatch between the temperature distribution in the system and a prescribed temperature profile and the cost of the control. This objective is realized by minimizing a nonlinear objective function of the boundary control and the fluid temperature variables, subject to partial differential equations (PDE) constraints governed by the coupled heat diffusion equation in the solid region and mass, momentum and energy conservation equations in the fluid region. Although CHT has received extensive attention as a forward problem, the optimal Dirichlet velocity boundary control for the coupled CHT process to our knowledge is only very sparsely studied analytically or computationally in the literature [131]. Therefore, in Part I, we describe the formulation of the optimal control problem and introduce the building blocks for the finite element modeling of the CHT problem, namely, the diffusion equation for the solid temperature, the convection-diffusion equation for the fluid temperature, the incompressible viscous Navier-Stokes equations for the fluid velocity and pressure, and the model verification of CHT simulations. In Part II, we provide theoretical analysis to explain the nonsmoothness issue which has been observed in this study and in Dirichlet boundary control of Navier-Stokes flows by other scientists. Based on these findings, we use either explicit or implicit numerical smoothing to resolve the nonsmoothness issue. Moreover, we use the numerical continuation on regularization parameters to alleviate the difficulty of locating the global minimum in one shot for highly nonlinear optimization problems even when the initial guess is far from optimal. Two suites of numerical experiments have been provided to demonstrate the feasibility, effectiveness and robustness of the optimization scheme. In Part III, we demonstrate the strategy of achieving parallel scalable algorithms for CHT models in Simulations of Reactor Thermal Hydraulics. Our motivation originates from the observation that parallel processing is necessary for optimal control problems of very large scale, when the simulation of the underlying physics (or PDE constraints) involves millions or billions of degrees of freedom. To achieve the overall scalability of optimal control problems governed by PDE constraints, scalable components that resolve the PDE constraints and their adjoints are the key. In this Part, first we provide the strategy of designing parallel scalable solvers for each building blocks of the CHT modeling, namely, for the discrete diffusive operator, the discrete convection-diffusion operator, and the discrete Navier-Stokes operator. Second, we demonstrate a pair of effective, robust, parallel, and scalable solvers built with collaborators for simulations of reactor thermal hydraulics. Finally, in the the section of future work, we outline the roadmap of parallel and scalable solutions for Dirichlet boundary control of fluid-solid conjugate heat transfer processes.

No<br>Development of therapeutic strategies for tumor-selective delivery of therapeutics through exploitation of the proteolytic tumor phenotype has significant scope for improvement of cancer treatment. ICT2588 is a peptide-conjugated prodrug of the vascular disrupting agent (VDA) azademethylcolchicine developed to be selectively hydrolyzed by matrix metalloproteinase-14 (MMP-14) within the tumor. In this report, we extend our previous proof-of-concept studies and demonstrate the therapeutic potential of this agent against models of human colorectal, lung, breast, and prostate cancer. In all tumor types, ICT2588 was superior to azademethylcolchicine and was greater or comparable to standard clinically used agents for the respective tumor type. Prodrug activation in clinical human lung tumor homogenates relative to stability in human plasma and liver was observed, supporting clinical translation potential. A major limiting factor to the clinical value of VDAs is their inherent cardiovascular toxicity. No increase in plasma von Willebrand factor (vWF) levels, an indicator of systemic vascular dysfunction and acute cardiovascular toxicity, was detected with ICT2588, thereby supporting the tumor-selective activation and reduced potential of ICT2588 to cause cardiovascular toxicity. Our findings reinforce the improved therapeutic index and tumorselective approach offered by ICT2588 and this nanotherapeutic approach.

[...] O casamento é uma fórmula narrativa de que MCC se serve menos para lhe desmontar a ideologia do que para dispensar a própria necessidade de uma narrativa – e assim usar o tempo de outra maneira. Histoplasma was initially described from a lesion in a horse by Rivolta in 1873, who named the organism Cryptococcus farciminosum. In 1905, Samuel Darling noted the presence of intracellular organisms in many tissues, including the lungs, of a patient suspected of succumbing to miliary tuberculosis (Darling 1906). Darling named the organism Histoplasma capsulatum , because it appeared to be an encapsulated protozoan-like organism. In 1912, mycologist Henrique da Rocha-Lima reviewed Darling’s slides and noted the cytological similarities between Darling’s Histoplasma organism and Cryptococcus farciminosum. Cryptococcus farciminosum was reclassified as Histoplasma farciminosum in 1934, and in 1985 it was again reclassified as a variant of Histoplasma capsulatum (var. farciminosum ) (Weeks et al. 1985).William De Monbreun cultured the organism from the blood of a child suffering from an unexplained febrile disease in 1934, and demonstrated it to be a dimorphic fungus (De Monbreun 1934). De Monbreun and others reported naturally occurring histoplasmosis in a dog in 1939, and subsequently demonstrated experimentally that clinically inapparent histoplasmosis occurred in dogs (De Monbreun 1939). De Monbreun and others speculated that animals might serve as the source of histoplasmosis in human beings. However, C.W. Emmons demonstrated in 1949 that Histoplasma capsulatum is a soil saprophyte, and that inhalation of aerosolized microconidia and mycelial fragments served as the source of infection (Emmons 1949).The prevalence of histoplasmosis in endemic regions was estimated to be more than 50% based on positive skin tests for histoplasmin (Edwards et al. 1969). Active histoplasmosis has been identifi ed in up to 50% of dogs in endemic regions based on culture at necropsy of healthy animals (Turner et al. 1972a). The case prevalence of disseminated histoplasmosis at a veterinary teaching hospital in an endemic region of the mid-western USA of 43 cases in cats and 12 cases in dogs per 100,000 hospital records per year has been estimated (Clinkenbeard et al. 1988; Kaplan 1973). Dogs and cats with outdoor exposure are reportedly at greater risk for histoplasmosis than those with minimal time outdoors. However, some completely indoor cats become ill with histoplasmosis (Davies and Troy 1996; Johnson et al. 2004). Young to middle-aged dogs of hunting and sporting breeds have historically been reported at greatest risk for acquiring histoplasmosis (Selby et al. 1981). Risk factors for cats have not been systematically studied.Infection by Histoplasma capsulatum var. capsulatum is not contagious except in unusual situations. Rare cases of horizontal transmission have been reported. Horizontal transmission is associated with conjugal contraction of individuals with cutaneous lesions of the genitalia (Sills et al. 1973) and by solid organ transplantation of infected organs (Limaye et al. 2000). No documented cases of transmission from animals to human beings or vice versa have been reported. In contrast to Histoplasma capsulatum var. capsulatum, equine infection by Histoplasma capsulatum var. farciminosum is contagious and is transmitted by bites of contaminated flies or ticks as well as through skin traumatized with contaminated tack (Kohn 2006).

For many applications, peptides should be conjugated to carriers. An important example is the conjugation of peptides to proteins, carbohydrates, or lipids for immunological studies. Further examples are the preparation of peptide affinity media and the conjugation of peptides to suitable coating compounds on surfaces for enzyme-linked immunosorbent assay and plasma resonance. The most important consideration in designing conjugates is that the peptide part of the conjugate must retain its biological activity. This means that the site of activity of the peptide must not be involved in the conjugation reaction. However, the active part of the peptide is often not precisely known, which complicates the design of a proper conjugation strategy. In addition, if the peptide is a fragment of a larger biologically active protein, the small peptide will frequently not adopt the conformation (for instance, a loop structure) of the corresponding sequence in the native protein. This might also abolish desired biological properties, such as the possibility to induce functional antibodies recognizing the native protein. In that case, appropriate artificial conformational restrictions should be introduced into the peptide - if possible. From the numerous conjugation methods available, only a few are described in this chapter. One example concerns the application of the homobifunctional cross-linker glutaraldehyde. Heterobifunctional cross-linking is illustrated by coupling of thiol-containing peptides or carriers to sulphydryl-reactive carriers or peptides, respectively. A general and easy method of conjugating peptides to proteins is to make use of homobifunctional cross-linkers. An example of a homobifunctional cross-linker is glutaraldehyde. This bis-aldehyde reacts with amine groups at neutral or basic pH to yield enamines, which can be reduced optionally to amines with sodium cyanoborohydride. The peptide and the protein react selectively via their N-terminus and/or lysine side chains to give not only a very complex mixture of products—peptide-peptide, peptide-protein, and protein-protein conjugates—but also large constructs containing peptide and protein. Typically, several peptide molecules per protein molecule are coupled. In view of the complex reaction pathway, the batch-to-batch reproducibility of the conjugate is difficult to control. If the peptide contains a lysine in the active part, it is recommended to extend the peptide N- or C-terminally with some additional lysines during synthesis in order to provide more amine groups for conjugation.

The quantum Hamiltonian represents the total energy of the system, the sum of kinetic energy plus potential energy, written in canonical coordinates and conjugate momenta, and where these variables become time independent quantum operators. The nature of the potential energy for the nano-vibrator, following Hooke’s law, serves to localize the particle. The relevance of the nano-vibrator Hamiltonian—sometimes called the harmonic oscillator Hamiltonian—is perhaps one of the most important Hamiltonians in quantum systems. Not only can it be extended to cover things like phonons in solids, vibrations in molecules, and the behavior of bosons, but it is also the basis for leading to the concept of a photon, the quantum radiation field, and the quantum vacuum. This chapter provides the basic introduction for vibration of a particle or a nano-rod and looks at the wave-like behavior that emerges from the solution to the time independent Schrödinger equation. When we include the time evolution, we can observe dynamical behavior and begin to examine the meaning of quantum measurement.

The present chapter describes how these residues are incorporated into peptides in the context of an Fmoc/tBu solid-phase synthesis strategy, as well as further considerations once the synthetic peptide is released from the support. Of added interest, some manipulations of Cys are advantageously performed at the level of the assembled peptide-resin, prior to cleavage. Many of the aspects discussed here also carry over to the preparation of peptides using a Boc/Bzl strategy. The major problems associated with management of Met reflect the susceptibility of the thioether to alkylation and oxidation. One of the merits of the Fmoc/tBu strategy, in contrast to Boc/Bzl, is that in the former strategy Met is usually introduced without recourse to a protecting group for the thioether side-chain. As documented in this chapter, a proper understanding of acidolytic cleavage conditions and the availability of selective procedures to reverse any inadvertent oxidation are likely to lead to success in obtaining homogeneous peptides containing Met. Management of Cys provides additional significant challenges. For some targets, Cys is required with its side-chain in the free thiol form, whereas for other targets, an even number of Cys residues pair with each other via disulphide linkage(s) to provide cystine residue(s). Disulphide bridges play an important role in the folding and structural stabilization of many natural peptides and proteins, and their artificial introduction into natural or designed peptides is a useful approach to improve biological activities/specificities and stabilities. Furthermore, use of a disulphide bridge is a preferred method to conjugate peptides to protein carriers for increasing the response in immuno-logical studies, to link two separate chains for developing discontinuous epitopes, and to generate active site models. This chapter describes Cys protecting groups, how they are removed to provide either free thiols or disulphides directly, and various strategies and practical considerations to minimize side reactions and maximize formation of the desired products. The thioether side-chain of Met is subject to alkylation and oxidation side reactions, either during the synthetic process or during subsequent handling of the Met-containing peptide.

The crinipellins are the only tetraquinane natural products. The enone crinipellins, including crinipellin A 3, have anticancer activity. Hee-Yoon Lee of the Korea Advanced Institute of Science and Technology (KAIST) envisioned (J. Am. Chem. Soc. 2014, 136, 10274) the assembly of 2 and thus 3 by the intramolecular dipolar cycloaddition of the diazoalkane derived from the tosylhydrazone 1. The initial cyclopentene was prepared from commercial 4 following the Williams procedure. Conjugate addition of the Grignard reagent 5 in the presence of TMS-Cl led to the silyl enol ether 6. Regeneration of the enolate followed by allylation gave 7. The preparation of the racemic ketone was completed by ozonolysis followed by selec­tive reduction and protection. Addition of hydride in an absolute sense led to separa­ble 1:1 mixture of diastereomers. Reoxidation of one of the diastereomers delivered enantiomerically enriched 8. A few steps later, after coupling with 10, the sidechain stereocenter was set by Sharpless asymmetric epoxidation. Oxidation of 11 gave the aldehyde, that was converted to the alkyne 12 by the Ohira protocol. Addition of the Grignard reagent 13 gave the allene 14 as an inconse­quential 1:1 mixture of diastereomers. Deprotection then led to the tosylhydrazone 1. The transformation of 1 to 2 proceeded by initial formation of the diazo alkane 15. Intramolecular dipolar cycloaddition gave 16, that lost N2 to give the trimethylene–methane diradical 17. The insertion into the distal alkene proceeded with remarkable stereocontrol, to give 2 as a single diastereomer—in 87% yield from 1. Direct α-hydroxylation of the ketone derived from 2 gave the wrong diastereo­mer, and hydride addition to 18 reduced the wrong ketone. As an alternative, the enantiomerically-pure sulfoximine anion was added to the more reactive ketone, and the product was reduced and protected to give 19. Allylic oxidation converted the alkene to the enone, and heating to reflux in toluene reversed the sulfoximine addi­tion, leading to 20. Epoxidation of 20 followed by α-methylenation delivered the enone 21, that proved to be particularly sensitive. Eventually, success was found with TASF. With a similarly sensitive substrate, Douglass F. Taber of the University of Delaware observed (J. Am. Chem. Soc. 1998, 120, 13285) that TBAF in THF buffered with solid NH4Cl worked well.

The applications of the conjugate stress and strain measures are in the development of the basic relations in nonlinear analysis of solids. In this paper using eigenprojection method, unified explicit basis-free relation between two arbitrary stress tensors T(f) and T(g), respectively conjugate to two measures of Hill’s strains is determined. The result is valid for arbitrary dimension of the Euclidean inner product space and for all cases of distinct and repeated eigenvalues of the right stretch tensor U.

Thermal stresses developed in electronic systems mainly depend, not only on average temperature values, but rather on wall temperature profiles. These profiles are difficult to predict unless one uses detailed finite element or finite difference modeling and simulation techniques. This type of analysis is only suitable at final design phases were geometrical details are available or being finalized. It is not suitable at early design phases to get a rapid estimate of wall thermal gradients to orient design appropriately. Standard approaches involving correlations for the heat transfer coefficient fail to predict temperature profiles for many reasons. In fact, these correlations depend on temperature profile as an input. In most engineering applications, walls are neither infinitely conducting nor of negligible conductivity to justify the usage of either uniform temperature or uniform heat flux assumptions. Correlations addressing conjugate heat transfer would not be able to solve the problem, unless a large number of them were available covering all possible combinations of fluid and wall conditions. Besides, the case of multiple heat sources, quite common in modern systems, can never be correctly handled by such an approach. The flexible profile technology was proposed earlier to model heat transfer in either solids (conduction) or fluids (forced convection. The model depends on domain (fluid or solid) geometry and physical properties, regardless of the particular set of applied boundary conditions, including that of multiple heat sources. Combining a fluid flexible profile model with a solid one, will allow predicting wall temperature profiles, with an adjustable level of precision, depending on the number of degrees of freedom retained. It will be applied in this paper to predict thermally induced stresses in some simple test cases as a demonstrator of the potentials behind this approach.

The primary computational bottle-neck in finite element based solid mechanics is the solution of the underlying linear systems of equations. Among the various numerical methods for solving such linear systems, the deflated conjugate gradient (DCG) is well-known for its efficiency and simplicity. The first contribution of this paper is an extension of the “rigid-body agglomeration” concept used in DCG to a “curvature-sensitive agglomeration”. The latter exploits, classic Kirchoff-Love theory for plates and Euler-Bernoulli theory for beams. We demonstrate that curvature-sensitive agglomeration is much more efficient for highly ill-conditioned problems arising from thin structures. The second contribution is a strategy for a limited-memory assembly-free DCG where neither the stiffness matrix nor the deflation matrix is assembled. The resulting implementation is particularly well suited for large-scale problems, and can be easily ported to multi-core architectures. For example, we show that one can solve a 50 million degree of freedom system on a single GPU card, equipped with 3 GB of memory.

Abstract The desire for increased engine efficiencies is driving higher firing temperatures. But this increases the risk for hot spots in solid components which can lead to durability issues. These may not be discovered until late in the design process through expensive and time consuming thermal paint tests. Historically, conjugate heat transfer simulations to predict solid temperatures have been done with steady RANS. However, Large Eddy Simulation (LES) is now being used in the early design process for gas turbine engines to account for the multiphysics of reacting flows. Incorporating the solid into these simulations poses a new challenge: the physical response time of the solid components can be orders of magnitude larger than the reacting gas phase. Running a fully coupled unsteady conjugate heat transfer analysis is therefore not tractable, but the high fidelity of the LES reacting solution is still desired. The objective of this paper is to demonstrate a multi-timescale simulation approach for conjugate heat transfer (CHT) in Simcenter STAR-CCM+ 2019.3. The combustor is solved using LES, including all relevant physics, while steady state conduction is determined in the metal liner and thermal barrier coating. Time averaged boundary conditions are transferred from the combustor to the solids, and temperature is returned through multiple exchanges until the solid temperatures reach a stable solution. A simplified case is used to verify the approach, and then results from a test combustor are compared against data. The investigation compares results obtained with PISO and SIMPLE numerical schemes.

For high pressure turbine heat transfer designs, a primary requirement is to predict blade metal temperature. There has been considerable recent effort in developing coupled fluid convection and solid conduction (conjugate) heat transfer prediction methods. They are so far, however, confined to steady flows. In the present work, a new approach to conjugate analysis for periodic unsteady flows of practical interest is proposed and demonstrated. This paper starts with a simple model analysis to quantify the huge disparity in time scale between convection and conduction and the implications of this for steady and unsteady conjugate solutions. To realign the greatly mismatched time scales, a hybrid approach of coupling between time-domain fluid solution and frequency-domain solid conduction is adopted in conjunction with a continuously updated Fourier transform at the interface. A novel semi-analytical harmonic interface condition is introduced, initially for reducing the truncation error in Finite-difference discretization. More interestingly, the semianalytical interface condition enables the unsteady conjugate coupling to be achieved without simultaneously solving the unsteady temperature field. This unique feature leads to a very efficient and accurate unsteady conjugate solution approach. The fluid and solid solutions are validated against analytical solutions and experimental data. The implemented unsteady conjugate method has been demonstrated for a turbine cascade subject to inlet unsteady hot streaks.

In existing gas turbine heat transfer literature, there are several correlations developed for the spanwise-averaged film-cooling effectiveness and heat transfer augmentation for inline injection on flat plates. More accurate and detailed predictions of film-cooling performance, particularly 3-D solid temperatures are needed for design purposes. 2-D correlations where effectiveness and heat transfer augmentation are functions of streamwise and spanwise directions are necessary to satisfy this need. Previously developed 2-D correlations for single row of cylindrical holes with inline injection have been improved to include the effects of shaped holes such as hole breakthrough width (t/D) and area ratio (AR). The correlations are improved to better match spanwise effectiveness of a single row of shaped cooling holes using data and spanwise-averaged correlations. Modifications to the correlations to improve application to compound injection (β) have been implemented. The blowing ratio is modified to account for the compound angle effect. The spanwise location of maximum film-cooling effectiveness and heat transfer augmentation are obtained as functions of the streamwise coordinate. Iterative Conjugate Heat Transfer Reduced Order Film Model (ICHT-ROFM) was used to obtain 3-D conjugate temperature distribution in film cooled solids. The developed correlations predicted a relative cooling effect in the near hole region for shaped holes (24 K) and for compound angle injection (20K) compared to cylindrical holes. Spanwise variations in the solid temperature in the near hole region are between 40–50K for a temperature difference of 250K between the surface and the main stream and are quite significant, showing the need for 3-D simulations. Shaped and compound angle holes increase this temperature difference due to the increased cooling. The comparisons of solid temperatures for conjugate and non-conjugate heat transfer cases show about 13–18K or 8–10% of the local temperature difference of 180K. Therefore it can be concluded that the calculations of 3-D temperature distributions using conjugate heat transfer are very important for design purposes.

Abstract This paper presents a novel approach that couples system modelling of both the thermo-fluid system and the thermal solid system by modelling conjugate heat transfer within a single 1D system analysis solver and applies it to the Secondary Air System of a Gas Turbine. The Secondary Air System design has to balance minimizing engine bleed whilst ensuring sufficient cooling. To achieve this, designers model both the secondary air flow and the temperature distribution in solid components. System CFD tools like Simcenter Flomaster may be used to solve flow, pressure and temperature distributions and a 3D thermal solver used to perform the thermal analysis of the blade and disc solids. The thermal interaction between the secondary flow system and the solid components is a key part of the model and is known as conjugate heat transfer analysis (CHT). This approach is problematic early in the design cycle when detailed or stable geometry information may not be available for the 3D thermal tool. An approach that couples the modelling of both the thermo-fluid system and the thermal solid system within a single 1D/system analysis tool offers the advantage of faster modelling and consistent model accuracy of both fluid and solid components, especially in the early concept design stage. This 1D-CHT approach has been implemented within Simcenter Flomaster and validated using an idealized analytical solution. It is shown that the model can be applied to the analysis of gas turbine secondary air systems including cavity flows and thermal analysis of the rotor and stator discs that form the thermal boundary of these cavities using Simcenter Flomaster alone.
