Abstract:
Sortilin-related receptor with LDLR class A repeats (SORLA, SORL1, or LR11) is a genetic risk factor associated with Alzheimer’s disease (AD). Although SORLA is known to regulate trafficking of the amyloid β (Aβ) precursor protein to decrease levels of proteotoxic Aβ oligomers, whether SORLA can counteract synaptic dysfunction induced by Aβ oligomers remains unclear. Here, we show that SORLA interacts with the EphA4 receptor tyrosine kinase and attenuates ephrinA1 ligand–induced EphA4 clustering and activation to limit downstream effects of EphA4 signaling in neurons. Consistent with these findings, SORLA transgenic mice, compared with WT mice, exhibit decreased EphA4 activation and redistribution to postsynaptic densities, with milder deficits in long-term potentiation and memory induced by Aβ oligomers. Importantly, we detected elevated levels of active EphA4 in human AD brains, where EphA4 activation is inversely correlated with SORLA/EphA4 association. These results demonstrate a novel role for SORLA as a physiological and pathological EphA4 modulator, which attenuates synaptotoxic EphA4 activation and cognitive impairment associated with Aβ-induced neurodegeneration in AD.

AbstractCurrent evidence indicates that resistance to the tyrosine kinase-type cell surface receptor (HER2)-targeted therapies is frequently associated with HER3 and active signaling via HER2-HER3 dimers, particularly in the context of breast cancer. Thus, understanding the response to HER2-HER3 signaling and the regulation of the dimer is essential to decipher therapy relapse mechanisms. Here, we investigate a bidirectional relationship between HER2-HER3 signaling and a type-1 transmembrane sorting receptor, sortilin-related receptor (SorLA; SORL1). We demonstrate that heregulin-mediated signaling supports SorLA transcription downstream of the mitogen-activated protein kinase pathway. In addition, we demonstrate that SorLA interacts directly with HER3, forming a trimeric complex with HER2 and HER3 to attenuate lysosomal degradation of the dimer in a Ras-related protein Rab4-dependent manner. In line with a role for SorLA in supporting the stability of the HER2 and HER3 receptors, loss of SorLA compromised heregulin-induced cell proliferation and sensitized metastatic anti-HER2 therapy-resistant breast cancer cells to neratinib in cancer spheroids in vitro and in vivo in a zebrafish brain xenograft model.

ABSTRACT Proper control of NaCl excretion in the kidney is central to bodily functions, yet many mechanisms that regulate reabsorption of sodium and chloride in the kidney remain incompletely understood. Here, we identify an important role played by the intracellular sorting receptor SORLA (sorting protein-related receptor with A-type repeats) in functional activation of renal ion transporters. We demonstrate that SORLA is expressed in epithelial cells of the thick ascending limb (TAL) of Henle's loop and that lack of receptor expression in this cell type in SORLA-deficient mice results in an inability to properly reabsorb sodium and chloride during osmotic stress. The underlying cellular defect was correlated with an inability of the TAL to phosphorylate Na+-K+-Cl− cotransporter 2 (NKCC2), the major sodium transporter in the distal nephron. SORLA functionally interacts with Ste-20-related proline-alanine-rich kinase (SPAK), an activator of NKCC2, and receptor deficiency is associated with missorting of SPAK. Our data suggest a novel regulatory pathway whereby intracellular trafficking of SPAK by the sorting receptor SORLA is crucial for proper NKCC2 activation and for maintenance of renal ion balance.

Charles sorel est un polygraphe. Cette etude propose une analyse analytique de l'ensemble de ses oeuvres romanesques, en envisageant chaque roman, pour en degager ses differentes caracteristiques. Les romans sont etudies selon l'ordre chronologique de leur production, en trois parties: - la premiere partie qui represente la periode des essais romanesques de sorel, comprend: - l'histoire amoureuse de cleagenor et de doristee (1621). - le palais d'angelie (1622). - les nouvelles francoises (1623). Le romancier commence tot a faire paraitre ses facultes, en contribuant a fixer en particulier un genre encore balbutiant: la nouvelle francaise. La deuxieme partie est l'etape florissante de sa carriere romanesque avec: -l'histoire comique de francion (1623-1633). - l'orphize de chrysante (1626). -le berger extravagant (1627-1628). Dans cette periode l'auteur a donne son chefd'oeuvre l'histoire comique de francion qu'on peut considerer comme comique unique dans son temps. La troisieme partie est celle du declin, avec: -la vraye suite des advantures de la polyxene (1634). - la solitude et l'amour philosophique de cleomede (1640). -la maison des jeux (1642). -nouveau recueil des pieces les plus agreables de ce temps (1644). - polyandre, histoire comique (1648). - le parasite mormon, histoire comique (1650). Sorel donne a la litterature francaise un nouvel elan, il ouvre la porte du realisme en essayant de combattre les invraisemblances qui regnaient dans la production litteraire de l'epoque
Charles sorel is an polygraph. This work propose an analyse of all his novels, in considering every novel appearing by its different characteristics. The novels are studied by their chronologic order of their production in three parties: the first party wich shows the novels essays period of sorel, is composed: - l'histoire amoureuse de cleagenor et de doristee (1621). - le palais d'angelie (1622). - les nouvelles francoises (1623). The novelist starts to appear earlier by its characteristics in contributing particularly a fixed syllabus which is still an earlier trial. The second party is the succesful step of his novels carrier with: l'histoire comique de francion (1623-1633). - l'orphize de chrysante (1626). - le berger extravagant (1627-1628). In this period, the author had given his prosperious work : l'histoire comique de francion wich can be considered as unique of this period. The third party is one of declination with: -la vraye suite des advantures de bla polyxene (1634). - la solitude et l'amour philosophique de cleomede (1640). - la maison des jeux (1642). - nouveau recueil des pieces les plus agreables de ce temps (1644). -polyandre, histoire comique (1648). - le parasite mormon; histoire comique (1650). In the present work, we seek to elucidate the relations between the problem of moral luck and our assignments of responsibility. The problem of moral luck emerges from two dimensions of human life. On the one side, we are autonomous and rational beings, we have control over our actions and are moral agents. On the other side, we are vulnerable to every sort of external contingency that eliminates the complete control we have over our actions and their results. The contingency, also, has a significant weight on the formation of our character and personality. Therefore, the problem of moral luck takes a real importance: how can we assign responsibility to the agents, given that a lot of what configures a moral action are contingent elements? The research was elaborated based on Bernard Williams and Thomas Nagel s articles on Moral luck. Williams, in his article, seeks to show that morality, as we conceive it, is (in fact) distant from our moral evaluations. Williams introduces the role of regret and recognizes the need to understand moral justification as retrospective. Nagel, in turn, finds the center of the moral luck problem in the control principle. In trying to understand how we assign responsibility to an agent for things beyond his control, Nagel defines four methods in which luck influences our moral judgements, and lists the kinds of moral luck: resultant, circumstantial, constitutive and causal luck. Finally, we take a look at critiques pertinent to the moral luck and responsibility problem, both negating and accepting the influence of luck in moral responsibility. From the epistemic argument and Zimmerman s postulates to Walker s pure agency critique and Otsuka s strawsonian considerations about reactive attitudes.
No presente trabalho, buscamos elucidar as relações entre o problema da sorte moral e nossas atribuições de responsabilidade. O problema da sorte moral emerge a partir de duas dimensões da vida humana. Por um lado, somos seres autônomos e racionais, temos o controle sobre nossas ações e somos agentes morais. Por outro, somos vulneráveis a toda espécie de contingência externa que elimina o controle total que temos de nossas ações e de seus resultados. A contingência, também, tem um peso significativo na formação de nosso caráter e personalidade. Por isso, o problema da sorte moral assume importância real: como podemos atribuir responsabilidade aos agentes, visto que muito do que configura uma ação moral são elementos contingentes? Elaboramos esta pesquisa a partir dos artigos Moral luck de Bernard Williams e Thomas Nagel. Williams, em seu artigo, busca mostrar que a moralidade, como a concebemos, mostra-se distante de nossas avaliações morais. Williams introduz o papel do arrependimento e reconhece a necessidade de entendermos a justificação moral como retrospectiva. Nagel, por sua vez, encontra no princípio do controle o centro do problema da sorte moral. Na tentativa de entender como atribuímos responsabilidade a uma agente por coisas além de seu controle, Nagel define quatro meios pelos quais a sorte influencia nossos juízos morais e elenca os tipos de sorte moral: sorte resultante, circunstancial, constitutiva e causal. Por fim, procuramos as críticas pertinentes ao problema da sorte moral e responsabilidade, as quais neguem ou aceitem a influência da sorte na responsabilidade moral, indo do argumento epistêmico e os postulados de Zimmerman à crítica da agência pura de Walker e as considerações strawsonianas de Otsuka sobre as atitudes reativas.

Esta pesquisa tem como ponto focal a análise das relações de gênero na comunidade garimpeira de Datas, localizada no Estado de Minas Gerais. A análise da divisão sexual do trabalho, dos conflitos envolvendo honra e gênero e da cosmologia característica da atividade garimpeira em Datas aponta para uma configuração das relações de gênero na qual a assimetria de poder entre os sexos apresenta uma maior flexibilidade, devido ao fato da mulher também garimpar, participando assim das possibilidades da sorte e de valores e atributos hegemonicamente atribuídos à masculinidade.

Este trabalho busca testar a validade da hipótese de eficiência dos mercados no mercado futuro do índice lbovespa através do uso das chamadas estratégias de análise técnica. São utilizados testes de habilidade preditiva para verificar a hipótese de superioridade destas regras de decisão como forma de investimento. Estes testes possuem a vantagem de considerar a possibilidade de data-snooping na escolha da melhor estratégia, permitindo identificar se a aparente capacidade preditiva destes modelos é realmente significativa ou mero produto do acaso. Os resultados indicam que as estratégias de análise técnica não são capazes de gerar retornos estatisticamente significativos quando os efeitos de data-snooping são levados em conta. Estes resultados estão de acordo com o previsto pela hipótese fraca de eficiência de mercado.
