Full textThe lateral distribution of lipids and proteins in the plasma membrane is heterogeneous. Therefore specific lipids and proteins in membranes of mammalian spermatozoa are enriched in special domains of varying size and different time scales enabling the cell’s membrane functional variability. Being transcriptional inactive this is especially relevant for spermatozoa in responding to multiple challenges on their way to fertilization. Therefore so called detergent resistant membrane domains (DRMs) from boar spermatozoa of different developmental stages were investigated. Already in the sucrose density gradients differences were visible, so the opalescent bands of more maturated sperm had a higher density. An analysis of these bands by 31P-NMR showed an enrichment of glycerophosphatidylethanolamine and phosphatidylinositol during maturation and a decrease of sphingomyelin during maturation in the epididymis and even after ejaculation. This suggests destabilization of DRMs and hence of putative membrane domains. This could enable clustering to bigger membrane domain platforms in preparation for capacitation and acrosome reaction. First, however, seminal fluid proteins cover the spermatozoa protecting the membrane with the destabilized membrane domains. This could have led to the detected decrease of the lipid to protein ratio in DRMs of ejaculated sperm. Boar spermatozoa are sensitive to storage at cold temperatures and are therefore usually stored at 16°C, which is especially disadvantageous with regard to growing of bacteria. A screening of sperm from 64 boars showed a ratio of 10% individuals with cold resistant sperm which could be stored at 4°C without quality loss. The DRMs of cold resistant sperm had a higher proportion of longchained, polyunsaturated fatty acids, as shown by analysis with 31P-NMR und MALDI-TOF MS.

Full textCilia and eukaryotic flagella are long, thin extensions of cells that contain a structure known as axoneme. The key components of the axoneme are microtubule filaments and the motor proteins dynein. These dynein motors force the microtubules to slide in an oscillatory fashion leading to a wave pattern along the axoneme. How these motors are coordinated and how this phenomenon can be described quantitatively is not understood. I therefore studied the waveforms of sperm tails that contain such an axoneme. I observed these waveforms under different conditions with a high-speed camera and developed an automated image analysis tool allowing the extraction of long time series of this waveform. In a subsequent Fourier analysis I increased the precision by obtaining an averaged waveform. I then compared the data to the predictions of a theoretical framework (Camalet, Julicher et al. 1999) and found that they do not agree. I suggested extending this theoretical framework by considering a visco-elastic element at the base of the axoneme, which leads to a satisfactory agreement. This project leaves open questions hence further work is discussed. As a side finding, I discovered a new phenomenon on how spermatozoa form dynamic vortex arrays. I described this pattern in detail and introduced a novel order parameter to quantify the order among many particles. I showed that the array only forms above a critical sperm density. I suggested a model to explain the origin of the pattern and showed by simulation that the model can account for the main features of the pattern. Finally I estimated the typical interaction force between beating axonemes to be 0.1 pN and drew conclusions about their collective action in general that might be relevant for sperm cooperation or metachronal waves of cilia

Full textThe search for autonomously moving, highly functional and controllable microdevices is a purpose of current micro/nanobiotechnology research, especially in the area of biomedical applications, for which reason, biocompatible solutions are in demand. In this thesis, a novel type of hybrid microswimmer is fabricated by the combination of rolled up thin nanomembranes with bovine spermatozoa. The microbiorobot presented here uses the powerful motion of the sperm flagella as a propulsion source for the magnetic microtube. This work demonstrates how the microswimmer performs its motion and how several factors such as temperature, radius of the microtube, penetration of the cell inside the microtube and length of the tube have influence on its performance. Directional control mechanisms are offered by external magnetic fields and are presented to be useful for the on-chip separation of the microbiorobots from a mixture of cells and microtubes. Two surface modification methods are presented as means to improve the coupling efficiency between the microtubes and the sperm cells. By these surface functionalizations, the extracellular matrix protein fibronectin is attached on the inner microtube walls and serves as binding agent for the spermatozoa. Finally, a remote release mechanism for the sperm cells is demonstrated by the incorporation of thermoresponsive material into the microtubes, which makes them fold and unfold upon small temperature changes. This work discusses the potential of such microswimmers for the application in assisted reproduction techniques and gives an outlook on future perspectives

Full textThe pikeperch (Sander lucioperca) has great potential for European aquaculture diversification, but the propagation is impeded by a shortage of stocking material caused by variability in gamete quality. Here, maternal size rather than out-of-season spawning or spawning experience was associated with variability in egg quality with very large females showing decreasing reproductive performance. Maternal factors were linked to the egg composition mainly in regard to fatty acid (FA) profiles, which in turn were correlated with rates of fertilization and embryo survival. In addition, egg size, dry weight and cortisol content were associated with fecundity and embryo survival until hatching. Consequently, a substantial variability in egg developmental performance could be explained (47.1% fertilization; 58.2% 24 h, 47% 48 h, 43.9% 72 h embryo survival; 46.6% hatching; 88.9% hatched larvae) by a combination of maternal traits and egg quality parameters. Despite the overall high fertilization observed here (89.2% on average), there is still potential for fine tuning of protocols for in vitro fertilization in pikeperch with regard to the management of male gametes. Sperm of different males is often pooled prior to fertilization or stored for short periods (hours) until ovulated eggs become available. A loss of sperm quality (motility, velocity) could not be prevented during short-term storage using sperm extenders and enhancement supplements (melatonin, progesterone). Transfusion of seminal fluid between low and high quality sperm to assess pooling effects resulted in a significant decrease in sperm with high initial velocity, whereas the velocity of low quality sperm could not be improved. These findings deliver valuable management advice for hatcheries and indicate potential for optimization in regard to in vitro reproduction protocols in pikeperch.

Full textOne of the essential qualities of cell membranes in Eucaryotae is a stable transverse phospholipid asymmetry. It is regulated and maintained by ATP-dependent action of an aminophospholipid translocase and is a major prerequisite for cell homeostasis. The plasma membranes of several mammalian cells show moreover lateral lipid domains, which are imputed to play a significant role in signal transduction. The membranes of mammalian spermatozoa undergo significant changes during genesis. The localisation and dynamics of phosphatidylserine in the cell as well as acrosome membranes of boar sperm cells was studied during capacitation and acrosome reaction to assess the relevance of lipid asymmetry for sperm function. The localisation of endogenous phosphatidylserine in morphologically differentiated cells was followed using the selective calcium depending binding of annexinV. Information on the transverse dynamics of phospholipids in the plasma membrane was obtained by labelling the cells with a NBD-phospholipid analogues. The morphological status of the cells was assessed by flow cytometry, fluorescence and electron microscopy. The results of this study indicate both a transversal and lateral inhomogenous distribution of lipids in the cell membrane as well as in the outer acrosome membrane during sperm genesis. The plasma membrane of boar sperm shows a stable transversal lipid asymmetry characterised by an accumulation of phosphatidylserine in the cytoplasmic monolayer. Moreover a cytoplasmic localisation of phosphatidylserine on the outer acrosome membrane could be detected for the first time. Therefore the two facing cytoplasmic leaflets of the outer acrosome and cell membrane contain phosphatidylserine. Applying microscopy substantiated the hypothesis that there are close interactions between the cell membrane and the outer layer of the acrosome membrane because of capacitation. The cytoplasmic accumulation of phosphatidylserine in lateral lipid domains is probably essential for the strong association of plasma and outer acrosome membrane finally leading to local fusions of both membranes. An indication for the functional meaning of lateral membrane domains in sperm cells was futher deduced from the isolation of Triton-insoluble lipid domains from membranes of trout sperm cells.
