Full textIt is increasingly recognized that the oceans preserve a huge number of natural products and novel chemical entities, with biological activities that may be useful in the quest for finding drugs with greater efficacy and specificity for the treatment of many human diseases. In this light, the aim of my project was to isolate and characterize novel molecules from marine organisms with regard to the identification of new “lead compounds” for pharmaceutical applications. The organisms considered for this study were selected by using two different strategies. The first one was based on enhancement of the taxonomic diversity. In this process, an emphasis was placed on collecting specimens related to - but differing from - those known to contain bioactive natural products. The second approach was to evaluate ecological factors such as costumer pressure, growth form (e.g. thin encrusting), level of resource competition, presence or absence of biofouling, etc., and relate this to the expression of the secondary metabolism. Some invasive species have chemical defences, which may enhance their invasion success, so as many marine organisms are soft bodied and have a sedentary life style necessitating chemical means of defence. Therefore, they have evolved the ability to synthesize or to obtain from marine microorganisms bioactive compounds that help them in deterring predators, keep competitors at bay or paralyze their prey. Full textMy thesis work was based on the characterization of porous materials, paying particular attention to the research of dynamic elements within the structures and to the study of adsorbed gases. I was able to detect the presence of ultrafast paraphenylenic rotors in both porous molecular crystals and metal-organic frameworks (MOFs). A more detailed study has also revealed how these motions are influenced by the adsorbed gas. Specifically, the activation energy of the rotation increases as a function of the quantity of gas in the pores. To better understand this interaction, the knowledge of the behavior of gases in porous materials is fundamental. I turned my attention to the study of xenon and CO2 motion in different materials. The combined use of NMR and ab initio calculations proved to be fundamental for understanding these phenomena and it was possible to reveal particular characteristics both of the gases and of the materials. The complexity of the diffusion within the channels has also been presented in unusual ways as the helicoidal motion of carbon dioxide imposed by the electrostatic potential. To continue the study of pore gases, I characterized several porous aromatic frameworks (PAFs) with the hyperpolarized xenon technique. This not only allowed me to accurately measure the pore size but also to calculate the interaction energy between the xenon and the channel walls. To expand my knowledge on hyperpolarization as an NMR technique, I spent six months at the group of Prof. L. Emsley in Lausanne learning dynamic nuclear polarization (DNP) as well as its application to different materials.

Full textPlant extracts mainly consist in complex mixtures of chemically different bioactive compounds also known as secondary metabolites. In most cases, the bioactivity of natural products is not ascribable to a single compound, but rather to a multitude of them acting in a synergistic way. In this view, the complete definition of all the phytochemical constituents represents a key point to ensure the efficacy, reliability, and safety in the use of herbal medicines. Even today, the quality and authenticity assessment of herbal raw materials is frequently impaired by the lack of validated analytical methods and by the limited availability of certified references. Techniques for the analysis of herbal extracts and nutraceuticals need to be developed and upgraded continuously since different combinations of ingredients are employed in new formulations. In view of the above, the current PhD project was focused on the development and application of innovative analytical methods for the analysis of plant bioactive secondary metabolites. Particular attention was paid to plants of pharmaceutical (Cannabis sativa L., and Spathodea campanulata P. Beauv.) and nutraceutical interest (Morus alba L., Aloysia polystachya Griseb. et Moldenke). Different in-vitro assays for the bioactivity evaluation of these plants’ extracts were also the object of this PhD Thesis. In Chapter 1, the NMR technique was applied for the determination of the main non-psychoactive cannabinoids in fiber-type Cannabis female inflorescences. The analytical method was developed and fully validated to show compliance with international requirements. This technique finds application in the monitoring of the plant material, to guarantee a better reproducibility for biological assays, and to ensure the efficacy and safety of Cannabis-derived products. As a continuation of this project, the potentialities and drawbacks of separation and non-separation methods for the analysis of cannabinoids were considered and compared. Finally, C. sativa non-psychoactive varieties were selected for the preparation of cannabinoids rich extracts to evaluate their antiproliferative activity, by using cellular models. In Chapter 2 the chemical composition of Italian mulberry was investigated, as a source of phytochemicals in the context of new therapeutic approaches for diabetes mellitus. The first aim was to assess the quality of the plant material in comparison to Far Eastern Asia cultivations, where mulberry has been considered as a medicine for the treatment of diabetes for decades. The antiglycative capacity and hypoglycaemic effect of leaf extracts were evaluated on different in vitro models, to understand the potentialities of mulberry for the therapy of hyperglycaemia. Finally, the development of novel strategies for delaying the release of mulberry active constituents and enhancing therapeutic results will be discussed and are intended as the conclusion of this research project. In Chapter 3 the chemical composition of S. campanulata was investigated by HPLC-ESI-MS2, to clarify which are the most active components responsible for the inhibition of H. pylori growth. The aim was also to disclose the mechanisms underlying the biological activity. In Chapter 4 the phenolic composition of A. polystachya was investigated by means of three different MS techniques coupled to HPLC. The analytical method was developed and validated for the identification of metabolites and some phenylpropanoids were detected in this plant for the first time.

Full textNear-infrared spectroscopy is a mature technique continuing to demonstrate steady progress. This is thanks to cutting-edge developments of new handheld spectrometers. These spectrometers can quickly generate a high volume of spectral data, requiring advanced methodologies such as artificial intelligence, big data, and deep learning to decipher the hidden content of the spectral data. This experimental research focuses on a handheld spectrometer based on MEMS technology and its application in various fields. We show that developing NIR models can be challenging upon application. We show the use case of this technology in the healthcare area and the industry (food). Plants and Fruits have been subjected to our research, such as Pothos and orange fruit. Organic powders have also found an application in our research. Furthermore, we extend our experimental research into animal wellness, particularly in dairy cow farms. We used advanced methodologies in specific use cases. We applied 1D-CNN when predicting quantities of content in mixtures of organic powder where a high volume of data was collected or a VAE model to predict water conditions as a health anomaly detection in the Pothos plant. We show spectra analysis and preprocessing approaches to remove signal artifacts. We implement different effective preprocessing methods: scaling methods, spectral derivatives, and the Savitsky Golay filter. We present the results of different data scaling and transformation methods using a convolutional neural network.

Full textNMR spectroscopy in weakly and highly orienting media is used as a route for dealing with orientational, positional, structural and conformational problems of a variety of small rigid and flexible organic molecules in solution. First, the very weak orientational order of a quasi-spherical molecule dissolved in a nematic phase is exploited for exploring the role of the different contributions to the observed dipolar coupling. In such a limit condition, a predominant effect of the non-rigid reorientationvibration coupling term emerges. Then, NMR data obtained from small rigid probes dissolved in smectic solvents are combined with a statistical thermodynamic density functional theory, in order to measure the positional order parameters of both solutes and solvent. The methodology gives good results when applied to a conventional smectic A liquid crystal and to the more delicate case of an interdigitated smectic Ad phase. The strategy is subsequently extended to the investigation of structure, order and conformational equilibrium of flexible bioactive or biomimetic molecules dissolved in various partially ordered NMR solvents. A first experimental and theoretical study is presented on the symmetric single-rotor molecule of biphenyl dissolved in a thermotropic liquid crystal. This test-case indicates molecular dynamics simulations are a promising tool for estimating a set of dipolar couplings of a solute in a thermotropic solvent, to be used as starting set of parameters in a standard operator-mediated NMR spectral analysis. Then, we report the conformational study of some single- and two-rotor nonsteroidal anti-inflammatory drugs, belonging to the families of salicylates and profens, dissolved in weakly orienting chiral nematic PBLG phases. A new pulse sequence, the Gradient Encoded heTeronuclear 1H-19F SElective ReFocusing NMR experiment (GET-SERF), is proposed here for the trivial edition of all 1H-19F couplings in one single NMR experiment, for a given fluorine atom. Starting from homo- and heteronuclear dipolar couplings, difficult to extract in thermotropic solvents because of a too complex spectral analysis, the torsional distributions of such molecules can be satisfactory described by the Additive Potential model combined with the Direct Probability Description of the torsional distribution in terms of Gaussian functions (AP-DPD approach). Finally, the conformational and orientational study of two stilbenoids displaying cooperative torsions is discussed in both a highly and weakly ordering liquid crystal phase. This comparative study allows to draw some conclusions on reliability, accuracy and accessibility of desired data in the two phases. Overall, this work proves NMR in liquid crystals is a flexible and meaningful tool for studying order, structure and conformation and it can greatly benefit from the availability of several aligning media inducing a different degree of order.
