Abstract:
Spire is a maternal effect locus that affects both the dorsal-ventral and anterior-posterior axes of the Drosophila egg and embryo. It is required for localization of determinants within the developing oocyte to the posterior pole and to the dorsal anterior corner. During mid-oogenesis, spire mutants display premature microtubule-dependent cytoplasmic streaming, a phenotype that can be mimicked by pharmacological disruption of the actin cytoskeleton with cytochalasin D. Spire has been cloned by transposon tagging and is related to posterior end mark-5, a gene from sea squirts that encodes a posteriorly localized mRNA. Spire mRNA is not, however, localized to the posterior pole. SPIRE also contains two domains with similarity to the actin monomer-binding WH2 domain, and we demonstrate that SPIRE binds to actin in the interaction trap system and in vitro. In addition, SPIRE interacts with the rho family GTPases RHOA, RAC1 and CDC42 in the interaction trap system. Thus, our evidence supports the model that SPIRE links rho family signaling to the actin cytoskeleton.

Abstract:
P168 Recently, clinical trials have demonstrated the efficacy of spironolactone (SPIRO) in men. Nevertheless the molecular mechanism of action not completely understood. Locally generated angiotensin II (ANG II) stimulates aldosterone. Therefore, we have tested the hypothesis that SPIRO ameliorates ANG II-induced renal damage. Furthermore, we investigated the effect of SPIRO on the transcription factors AP-1 and NF-κB. We treated transgenic rats overexpressing the human renin and angiotensinogen genes (dTGR) from week 5 to 7 with SPIRO (20 mg/kg/d). Plasma aldosterone was significantly increased in dTGR vs. SPIRO treated and non-transgenic (SD) rats (p<0.05). Untreated dTGR showed high systolic blood pressure (182±8 mm Hg), severe renal damage with 150-fold increased albuminuria, vasculopathy and perivascular and interstitial fibrosis. Chronic SPIRO treatment reduced mortality and vasculopathy completely, despite blood pressure levels of 161±11. 24-hour albuminuria was reduced from 59±15 in dTGR to 3.5±2 mg/d; p<0.01). Electrophoretic mobility gel shift analysis demonstrated a reduction of renal AP-1 and NF-κB DNA binding activity after SPIRO treatment. Immunohistological analysis showed that SPIRO also prevented the expression of AP-1 and/or NF-kB regulated matrix molecules fibronectin and laminin. The reno-protective effect of SPIRO was accompanied with a reduction of monocyte/macrophage infiltration. These findings show that blockade of aldosterone signaling ameliorates ANG II-induced renal damage. SPIRO action was at least partially mediated via AP-1 and NF-κB.

The structures of two novel 3′-spiro nucleosides analogues of the potent human immunodeficiency virus type 1 (HIV-1) reverse transcriptase (RT) inhibitor TSAO-m3T, in solution, as derived from NMR spectroscopy are described. In these TSAO analogues the spiro amino oxathioledioxide moiety has been replaced by spiro amino oxazolone or spiro amino oxathiazoledioxide moieties. A comparative study based on theoretical calculations of the hydrophobicity, the solvation free energies and molecular electrostatic potentials (MEP) of the three compounds is also described. No significant conformational differences were detected in solution between TSAO-m3T and its analogues that might account for the differencesobserved in their inhibitory activity against HIV-1 RT. The calculated hydrophobicity (log P) values, dipole moments and the electrostatic contributions to the solvation free energies of the three spiro ring systems were also similar. However, the differences found in the calculated MEPs of the spiro systems between TSAO-m3T and its analogues suggest that the different electrostatic surroundings of the 4″-amino group of the spiro moiety in the analogues may be responsible for a detrimental electrostatic interaction of the spiro rings with the Glu-B138 of RT.

Spire and Cappuccino are actin nucleation factors that are required to establish the polarity of Drosophila melanogaster oocytes. Their mutant phenotypes are nearly identical, and the proteins interact biochemically. We find that the interaction between Spire and Cappuccino family proteins is conserved across metazoan phyla and is mediated by binding of the formin homology 2 (FH2) domain from Cappuccino (or its mammalian homologue formin-2) to the kinase noncatalytic C-lobe domain (KIND) from Spire. In vitro, the KIND domain is a monomeric folded domain. Two KIND monomers bind each FH2 dimer with nanomolar affinity and strongly inhibit actin nucleation by the FH2 domain. In contrast, formation of the Spire–Cappuccino complex enhances actin nucleation by Spire. In Drosophila oocytes, Spire localizes to the cortex early in oogenesis and disappears around stage 10b, coincident with the onset of cytoplasmic streaming.

Ce travail décrit une approche générale et efficace des spiro-β-méthylène-γ-thiobutyrolactones. La stratégie utilisée s'appuie sur une étude développée précédemment dans notre laboratoire pour la synthèse des bakkénolides fonctionnalisés sur le cycle A ou sur les cycles A et B à la fois. La formation de ces spiro-thiolactones en deux étapes, repose sur une réaction de transestérification en milieu neutre, conduisant à des β-céto-thiolesters allyliques qui conduit directement aux spiro-β méthylène-γ-thiobutyrolactones de façon radicalaire, en présence du mélange Mn(OAc)3/Cu(OAc)2. A partir des β-cétoesters insaturés, il est possible de réaliser des réactions en cascades pour obtenir des composés polycycliques comportant une lactone. Le greffage de ce type de lactone insturé spiranique, en seulement deux ou trois étapes, a permis d'obtenir 9 nouveaux stéroïdes modifiés. Finalement, comme application synthétique, un analogue soufré de bakkénolide a été obtenu, (7 épi-9-acétoxyfukinanthiolide), en seulemnt 6 étapes avec un rendement global de 17%<br>This work deals with general and efficient approch of spiro-β-methylene-γ-thiobutyrolactones. The used strategy despends on a previously developed study in our laboratory concerning the synthesis of bakkenolides fonctionnalized on the cycle A or cycle A and B. The two steps formation of these spirothiolactones leads on a transesterification reaction in neutral condition to get allylic β-keto-thiolesters which permit to access directly to the spiro-β-methylene-γ-thiobutyrolactones by free radical cyclization with a mixture of Mn(OAc)3 and CU(OAC)2. From more substitued and insaturated β-keto-esters, polycyclic compounds having a lactone are obtained by cascade reactions. Grafting of this insaturated spirolactone with only two or three steps has permited to get 9 new modified steroïds. Finaly, as synthetic application, a sulfur analogue of bakkenolide (7-epi-9-acetoxyfukinanthiolide) has been obtained, by only 6 steps with 17% overyield

Abstract:
<P ALIGN="JUSTIFY">A novel antenna made of a spiral wire wrapped into a larger helical shape is introduced. The geometry of this antenna, which is a doubly helical structure, is fully described by five independent parameters, including two radial dimensions, two pitch angles, and the number of turns. Radiation properties of this antenna are examined both theoretically and experimentally. The Numerical Electromagnetic Code (NEC-2) is used to obtain simulation results. A large number of cases with different radii, pitch angles, and number of turns are investigated. Results for far-field patterns, gain, axial ratio, and bandwidth are presented. The influence of parameters on radiation properties are examined. Several prototypes of the antenna were constructed and tested using an outdoor antenna range. Measured far-field patterns are presented over a wide range of frequencies. The measured and computed radiation patterns are in good agreement.</P> <P ALIGN="JUSTIFY">The results of this study indicate that the proposed antenna provides circular polarization and high gain over a wide frequency range. For example, when the number of turns is 10, a gain of 11-14 dB, a boresight axial ratio of less than 3 dB, and a half-power beamwidth of about 40 degrees are achieved over a 30% bandwidth. The side-lobe level for most cases examined is better than 10 dB below the main beam. A unique advantage of this antenna is its much smaller size compared with a conventional helical antenna made of straight wire shaped into a helix. Having about the same radiation characteristics, including gain, circular polarization, bandwidth, and side-lobe level, this new antenna occupies a volume more than 2.5 to 3 times smaller than the conventional helix. This reduction in size, which in turn may imply smaller weight and lower packaging and manufacturing costs, makes the proposed antenna very appealing to many communications and aerospace applications. </P><br>Master of Science

Abstract:
Naphthoxazine-spiro-indolines (NOSIs) can exist in a colourless and a coloured form. These compounds can interconvert between these two forms either photochemically or thermally and are resilient to degradation even after repeated cycles of colouration and bleaching. Absorption spectra of both the stable colourless form and the energetically less stable coloured form have been measured. Several NOSI compounds have been shown to photoconvert to the coloured form with an efficiency of between 0.06–0.74, depending upon the compound conditions under which the conversion is brought about. The factors which have been varied in this work are: the basicity of the 6'-substituent; the size of the N-alkyl group within these compounds; and the nature solvent or matrix in which the reaction occurs. Electron donation from the 6'-group has been shown to increase the quantum yield of the forward reaction whereas solvent polarity increases have been shown to reduce the quantum yield.

L’obiettivo di questo elaborato è quello ricercare una tecnica diagnostica non invasiva, attraverso la quale identificare la presenza del corto circuito spira-spira in una fase di statore di una macchina brushless esafase asimmetrica. L’elaborato risulta essere diviso in 4 capitoli. In particolare, nel Capitolo 1 si descrive il modello matematico che permette di studiare la macchina in presenza del corto circuito in una fase di statore. Nel corso del Capitolo 2, si descrive il modello di simulazione realizzato mediante l’utilizzo del software Simulink. Il modello è sviluppato seguendo le equazioni del modello matematico, per la determinazione delle grandezze di interesse. La sua realizzazione risulta essere fondamentale per collaudare il funzionamento delle tecniche diagnostiche in simulazione, prima di provarle sulla macchina reale. Nel capitolo si descrivono i vari sottosistemi che compongono il modello, soffermandosi in particolare sull’implementazione della tecnica diagnostica utilizzata. Nel Capitolo 3 si descrive il banco utilizzato per le prove sperimentali sulla macchina. In particolare, nella prima parte è descritta la scheda dSpace, attraverso la quale si è interfacciato il modello di simulazione con l’azionamento reale. Successivamente sono descritti i componenti dell’hardware che compongono l’azionamento, soffermandosi sulle caratteristiche della macchina brushless. Infine, nel Capitolo 4 si illustrano le prove sperimentali realizzate sulla macchina. Questo capitolo è diviso in 3 parti. Nella prima parte vengono descritte le misure realizzate per la determinazione dei parametri di interesse della macchina, sia nel caso di macchina sana che di macchina guasta, utilizzate come dati per il modello di simulazione. Successivamente, si illustrano i risultati delle prove a vuoto e della prova a carico, relativamente alla tecnica diagnostica che ha fornito i risultati migliori in simulazione.

Abstract:
Une étude bibliographique a montré qu'il existait peu de 2H-chromènes hétéroannellés. Deux stratégies de synthèse sélectionnées ont permis l'obtention d'une large série de photochromes de ce type. L'accès à ces chromènes nécessite l'élaboration préalable d'hétérocycles hydroxyles (benzimidazole, benzoxazole, benzothazole, benzosélénazole, benzofurane, indole, isoquinolène, quinoléine, quinazoline, quinoxaline). Une autre série originale de composés photochromiques, les spirofluorène-2H-chromènes a été préparée. La détermination par photolyse à éclairs des paramètres caractérisant le photochromisme a permis de quantifier les effets respectifs de la substitution du carbone SP#3, du type d'annellation et des hétérocycles. Les paramètres de l'équilibre photochromique varient dans un large intervalle. Les composés hétéroannellés par des cycles à cinq chaînons présentent deux bandes pour les spectres d'absorption de leurs formes colorées couvrant ainsi une large plage du domaine visible. Les chromènes hétéroannellés par des cycles azotés à six chaînons manifestent de bonnes propriétés d'ensemble et notamment de résistance à la fatigue. La participation accrue des électrons de chacun des noyaux phenyle provoque une amélioration sensible des propriétés photochromiques, observées pour les spirofluorène-2H-chromènes. Une étude radiocristallographique a mis en évidence la non-équivalence des deux groupements phényle pour les formes incolores. Un phénomène d'interconversion photochimique entre différents stéréosomères de la forme colorée, a été observé lors de l'étude à l'état photostationnaire. L'amélioration technique récente du matériel d'investigation spectrocinétique a montré l'existence d'un équilibre thermique, à température ambiante entre plusieurs stéréoisomères de la forme colorée, traduisant bien la complexité du phénomène photochromique intime mais ouvrant de nouvelles perspectives quant à sa meilleure compréhension. Les spiroindoline-oxazines sont des partenaires de choix des chromènes et dérivés, en vue d'applications dans les matériaux à transmission optique variable. Le comportement spectroscopique de photomérocyanines issues de spiroindolinéphénanthrolinoxazines a été étudié et a montré leur caractère de structure délocalisée. Une estimation des absorptivités molaires a été également réalisée

LundbergDesign har tagit fram ett armaturkoncept för ateljé Lyktan som de vill börja tillverka. Att utveckla denna armatur skulle vara studenternas huvuduppgift under examens-arbetet. Samtidigt skulle de också utvärdera ateljé Lyktans nya produktutvecklingsprocess, FPM, som de fått av systerföretaget Fagerhult. Utgångspunkten var att processen var för stor och komplex för Lyktan och att processen således behövde anpassas till deras verksamhet, som är mindre än Fagerhults. Huvuduppgiften för examensarbetet kom att ändras, då utvecklingsprocessen för armaturen försenades på grund av problem med att hitta en leverantör av en komponent. Istället fokuserade examensarbete nu på att utvärdera processen, och armaturprojektet användes istället som en fallstudie. Två utvärderingsmetoder för processen studerades och vägdes mot varandra, varav den ena, PAIN, valdes. Processens ritades upp i en processkarta och analyserades sedan aktivitet för aktivitet med bland annat intervjuer av medarbetare och SIPOC. Utfallet från analyser resulterade i en önskvärd processkarta och ett antal rekommendationer. Under analysens gång fångades åsikter från medarbetare upp och allmänna rekommendationer som inte direkt berör processen är också en del av resultatet. Eftersom alla analyser, utöver litteraturstudier, bygger på medarbetares erfarenheter av processen vilken inte hunnit genomföras så många gånger på företaget, påverkas resultatet av detta. Faserna är olika djupgående analyserade beroende på studenternas erfarenhet av produktutvecklingsprocessen, fallstudien, samt företagets egen erfarenhet av processen.<br>LundbergDesign has designed a concept of a luminaire for ateljé Lyktan which they intend to manufacture. To develop this luminaire, was supposed to be the main task for the bachelor thesis. A parallel task was to evaluate ateljé Lyktan’s recently implemented product development process, FPM, which they obtained from their sister company, Fagerhult. The main task for the thesis was changed due to delays in the luminaire project, which had problems finding a supplier for a part. Instead, the evaluation of the development process was now the main focus, and the luminaire project was used as a case study of the process. For evaluating the process two methods were considered of which one was chosen, PAIN. The process was first drawn as a process map and then analyzed activity by activity with, among other, interviews of staff and SIPOC. The outcome from the analyses resulted in a "should be"- process map and recommendations. During the analysis opinions from the staff was taken under consideration and general recommendations, that not directly concern the process, are also a part of the result. All analyses, apart from being based on literature studies, are also based on the staff’s experience of the process within the company. The process has not been performed more than a few times and the results are affected by this. The analyses of the phases are varying in depth depending on the students’ experience in the product development process, the case study, and the company’s own experience of the process.

The European Space Agency's Herschel Space Observatory is comprised of three cryogenically cooled instruments commissioned to explore the far infrared/submillimetre universe. The Spectral and Photometric Imaging REceiver (SPIRE) is one of Herschel's instruments and consists of a three band imaging photometer and a two band imaging spectrometer. Canada is involved in the SPIRE project through provision of instrument development hardware and software, mission flight software, and support personnel. This thesis discusses Fourier transform spectroscopy (FTS) and FTS data processing. A detailed discussion is included on FTS phase correction, with results presented from the optimization of an enhanced Forman phase correction routine developed for this work. This thesis discusses the design, verification, and use of the hardware and software provided by Dr. Naylor's group as it relates to SPIRE verification testing. Results of the photometer characterization are presented. The current status of SPIRE and its future schedule is also discussed.<br>xvii, 239 leaves : ill. (some col.) ; 28 cm.

Single Point Incremental Forming (SPIF) has received considerable attention recently due to advantages such as part-shape-independent tooling, higher formability and higher process flexibility as compared to conventional forming. While significant research has been performed on SPIF of metals, recent work has also shown the feasibility of using SPIF for cold-forming of thermoplastic polymer sheets. However, the effects of incremental depth and part shape on the modes of failure during polymer SPIF have rarely been investigated. This paper examines the effects of part shape and incremental depth on the formability and failure modes in polymer SPIF. It is shown that greater incremental depths result in greater formability in polymer SPIF. Furthermore, it is shown that increasing the rate of change of the wall angle with the Z depth of the part increases the maximum formability achievable using a given incremental depth. At the same time, it is observed that this dual advantage of greater formability and reduced forming time, possible with higher incremental depths, is limited by the occurrence of sheet wrinkling when the incremental depth becomes too high. Additionally, the dependence of sheet wrinkling on the overall shape of the part being formed is also shown.

Abstract:
O presente relatório contém resultados parciais do projeto “Definição de áreas prioritárias para recuperação florestal”, referentes a atividade “Uso e comparação da acurácia de diferentes modelos preditivos de desmatamento na Amazônia”. O objetivo deste estudo foi a implementação de modelos preditivos de desmatamento na Amazônia brasileira com base nas técnicas de Random Forest (RF), Spatial Random Forest (SpRF) e Integrated Nested Laplace Approximations (INLA) e comparação dos erros obtidos com cada modelo. Uma base de dados geográficos foi gerada por meio da integração de dados de diversas instituições brasileiras, como IBGE, MMA e INPE, utilizando células de 25 x 25 km e uma janela temporal de um ano. Os principais drivers de desmatamento identificados estão relacionados à fragmentação florestal e à expansão de áreas de pastagem na Amazônia, corroborando com outros trabalhos encontrados em literatura. A modelagem obteve melhores resultados com o uso dos modelos RF e SpRF em relação aos modelos do tipo INLA, com menores valores de erro médio quadrático obtido em conjuntos de dados de treinamento e validação dos algoritmos. A previsão de desmatamento para o ano de 2020 foi de 31 mil km2 , dados que apresentam uma superestimava devido ao método utilizado para o cálculo do desmatamento. Entre as ações identificadas que podem ser adotadas em trabalhos futuros para melhorar a previsão do desmatamento, cita-se o uso da abordagem CLUE e a melhoria de algumas bases de dados utilizada, a exemplo da malha viária.
