A promising approach to measuring the level of diagnostically significant serum proteins is the development of test systems based on aptamers. In this work, we generated a series of new DNA aptamers for the bone metabolism biomarker DKK-1. Several sandwich systems based on aptamers and DKK-1-specific antibodies for heterogeneous colorimetric detection of DKK-1 were developed and tested. The optimal aptamer/antibody system have shown its suitability for measuring DKK-1 levels in serum samples from patients with spondyloarthritis.

In an attempt to aid the subtyping of spondyloarthritis (SpA), this work assessed neural nets and magnetic resonance imaging (MRI) features to predict SpA. Patients underwent SPAIR and STIR MRI sequences. Radiologists manually segmented sacroiliac joints images for extracting MRI features. A neural net used these features to predict SpA. The STIR-based model yielded higher performance than SPAIR to diagnose SpA, although no statistical difference was found between them. The SPAIR model yielded an area under the curve of 0.83 to differentiate axial and peripheral subtypes, while STIR yielded 0.

Abstract:
Around one third of the UK population live with a rheumatic and musculoskeletal disorder (RMD)1. Inflammatory RMDs such as rheumatoid arthritis, psoriatic arthritis and axial spondyloarthritis, and rarer rheumatic conditions such as systemic vasculitis and lupus, are looked after by hospital-based specialists. Other common non-inflammatory conditions such as osteoarthritis and fibromyalgia are mainly looked after in primary care. Some people have more than one type of RMD, for example, rheumatoid arthritis and osteoarthritis.

Supporting people with health conditions to remain in work is a UK government priority. Rheumatic and musculoskeletal conditions greatly impact people’s working lives. This includes inflammatory arthritis (e.g., rheumatoid arthritis, psoriatic arthritis and axial spondyloarthritis), rare autoimmune conditions such as lupus and vasculitis, and other painful conditions such as osteoarthritis and fibromyalgia. They are one of the most common causes of days lost from work in the UK and people deciding to stop work earlier than intended.
