Abstract:
A case of acquired severe genu valgum and genu recurvatum in a patient with rheumatoid arthritis (RA) is here reported. The pain and genu valgum progressed because of poor RA control. The patient had no history of major trauma of the knee before or after the onset of RA. The most reasonable hypothesis to explain this patient‘s pathology is that occult genu recurvatum developed after a minor trauma and progressed; poor RA control then worsened the stability, the osteopenia and genu valgum, in the end the patient acquired a severe genu valgum with extreme values of the tibiofemoral axes (TFM), with multiplanar knee laxity and important bone destruction. Total knee arthroplasty (TKA) was successfully performed using a constrained hinged prosthesis. One year after the surgery, the patient exhibited improvement in her Knee Society Score from preoperative scores of 19 to postoperative scores of 91, no early or late signs of infection occurred, no early signs of loosening were noted either. Acquired global ligamentous deficiency with hyperextension instability (genu recurvatum) associated with severe genu valgum in patients with RA is rare. Excellent results were obtained with TKA, but the use of the most constrained prosthesis due to massive bone loss and ligamentous deficiency comes with the disadvantages of early aseptic loosening as a result of increased constrained compared to an unconstrained design.

Abstract:
La lésion du ligament croisé antérieur est une atteinte sévère et fortement prévalente chez les joueuses de sports pivot (handball, basketball). Le développement des sports féminins incite à en comprendre les causes et les mécanismes. Cette thèse a pour objectif d’étudier le risque de lésion du ligament croisé antérieur de la joueuse de sport pivot et son évolution lors de l’exposition à une charge de travail. La première partie est bibliographique et expose le caractère dynamique du risque de blessure en sport. La complexité des interactions entre facteurs de risque et mécanisme lésionnel dans la lésion du LCA est présentée selon une approche épidémiologique puis biomécanique. Ces éléments mettent en valeur l’importance des facteurs de risque neuromusculaires dans la transition vers l’évènement incitatif d’une lésion du LCA. L’effet d’une charge de travail aigue sur ces facteurs est peu décrit chez les sportives pivot ce qui soulève plusieurs questions de recherche. La deuxième partie est expérimentale, avec deux études prospectives et une revue systématique de la littérature utilisant des outils d’évaluation de la force musculaire, d’analyse systématique de la littérature et de mesures de biomécaniques (cinétique, cinématique et laximétrie). Nos résultats montrent la spécificité de réponse à la fatigue isocinétique de la joueuse de handball. La cinétique de la réception de saut est la plus impactée par un protocole de fatigue neuromusculaire, les changements de direction semblant les moins modifiés. Nous identifions le caractère précoce, entre 46 et 48 ms après contact initial, de la mise en contrainte des structures activo-passives du genou lors de 766 changements de direction. Nos travaux montrent également que les joueuses abordent le changement de direction avec des amplitudes de genou compatibles avec le mécanisme lésionnel, une posture du tronc plus érigée et une vitesse angulaire de flexion de genou que nous estimons plus élevée que les hommes. La troisième partie de ce travail propose une discussion et une mise en lien de ces résultats, ainsi que leurs perspectives pour la prévention des lésions du LCA chez les joueuses de handball et basketball
ACL injuries are severe and highly prevalent in female handball and basketball players. The increasing participation of women in multidirectional sports conduces to a better understand the mechanism and causes of the injury. The aim of this thesis is to investigate anterior cruciate ligament injury risk among female players in multidirectional sports and its workload-induced changes. The first part is a narrative review about the dynamic process of sports injury. Epidemiologic and biomechanic approaches were used to explain the complexity of risk factors and injury mechanism interaction. These data showed the key role of neuromuscular risk factors in building the incitative event of anterior cruciate ligament injuries. The lack of knowledge about the impact of an acute workload on those risk factors in female multidirectional sports needs to be addressed.The second part is an experimental report, including two prospective studies and a systematic review using muscular assessment, systematic tools for literature review and biomechanical analysis (kinetics, kinematics and laximetry). The results showed the specificity of female handball players when performing an isokinetic fatigue protocol. The jump landing kinetic was the most impacted by a fatigue protocol. Cutting techniques were the less impacted. The analysis of 766 side-step-cutting showed that knee joint loading is anticipated (46 to 48 ms after initial contact) in female handball and basketball players. It was also shown that women performed side-step-cutting with similar knee amplitude than the injury mechanism, a less flexed trunk position and a highest angle-velocity than men.The third part of this work proposes a discussion of those results in relation to their perspectives for the prevention of anterior cruciate ligament injury risk in female handball and basketball players

Abstract:

The engine of JAS 39 Gripen, RM12, has recently been equipped with a new flameholder and a new engine inlet. This change has caused an increased noise level from the engine when the afterburner is in use. The noise level reaches its peak when the afterburner is partially used. The problem is thought to be unstable combustion in the regions around the flameholder. A solution to the problem has been proposed by Volvo Aero. The idea is to modify the software in the engine control unit (FADEC) that changes the airflows in the engine. This master thesis implements the proposed changes and analyzes the effects of this implementation. The analysis is done by simulations of a model of the engine that Volvo Aero has created and implemented in MatrixX/SystemBuild. Simulations showed that it is possible to achieve the desired result by the software changes. Further the simulations showed some problems with stability for the engine when controlled by the modified control unit. This was solved by implementing a filter in the FADEC software.

Abstract:
Gotland har länge präglats av mycket speciella lösningar och legat i framkant vad gäller ny teknik. Under ca 20 år har Gotland haft en VSC-HVDC-station som har stabiliserat spänningen i det gotländska nätet. HVDC Light har löst en del av de tekniska begränsningarna som hindrat utvecklingen av vindkraftverk. Anläggningen börjar lida mot slutet av sin tekniska livslängd och examensarbetet är ett första steg i utredningen för en eventuell ersättning. Med hjälp av elkvalitetsmätare kartlagdes prestanda och eventuella förbättringsområden. Den svenska stamnätsoperatören Svenska kraftnät har sedan 2016 haft en VSC-HVDC-station som använts för att utväxla energi till asynkrona systemet i Östeuropa. Svenska kraftnät har på senare tid velat utnyttja spänningsregleringsmöjligheterna och utreda mättekniska metoder. Elkvalitetsmätare placerades ut på lämpliga mätpunkter för att utreda anläggningens beteenden. ELSPEC G4500 elkvalitetsmätare installerades på Nordbalt och Gotlands HVDC Light för att mäta under sensommar till hösten 2017. Skillnader mellan CVT och IVT samt Rogowskispole och CT mättes. Valet av ELSPEC lämpade sig bra eftersom att inga triggningsvillkor behövdes som tillåter att man kan upptäcka små men viktiga avvikelser. Gotlands HVDC Light stabiliserar nätet främst mot spänningsdippar efter kortslutningar i 10 kV-elnätet och flimmer ifrån vindkraftsparkerna i ände 2. Märkeffekten för en uppdaterad anläggning kommer baseras på den kortslutningseffekt i 10 kV-nätet som kan utvecklas under anläggningens livstid. Spänningsregleringen ska baseras på en PI-regulator och ska kunna reglera fullt på 40 ms. Komponentspänningar kan användas för att ge reaktiv effekt på de faser som behöver det. Behovet mot flimmer ska baseras på en mätning i närmaste konsumtionscentrum, två mil från ände 2. Om behovet finns, ska en separat loop för flimmerkontroll som motverkar 1,5 Hz-komponenter implementeras utifrån en punkt ca 1,5 mil ifrån ände 2. Teknikvalet står mellan two-level generation 3 eller MMC, beroende på uppgradering eller ersättning. Många olika framtida scenarion påverkar HVDC Lights roll och oavsett, kommer mycket resurser behövas för Gotlands och HVDC Lights framtid. Det har observerats två beteenden på Nordbalt varav ena är långsam och det andra beteendet är snabbt. Det snabba beteendet uppstår när lågohmiga fel sker som också synkronmaskinerna tar hand om. Nordbalt kan hjälpa till för mindre spänningsvariationer om den varit snabbare likt beteendet vid lågohmiga fel. En stabilitetsbedömning behövs dock eftersom att snabbare beteende ökar risken för instabilitet. Eftersom att data mellan CVT och IVT skiljde sig mycket, upplystes mättekniska problem. Några lösningar diskuteras varav RCVT och PQSensor gås igenom grundligt. Alternativa lösningar som MoW och mobila enheter presenteras också vagt. Samtliga lösningar visade sig ha praktiska hinder, vilket försvårar genomförandet. Det uppmanas därför att man bör testa teknikerna i laborationsmiljö eftersom begränsat med studier gjorts på dessa samt att konkurrerande tillverkare uppger olika uppgifter.
Gotland has long been known for various special solutions and been on the leading edge regarding new technology. For the past 20 years Gotland has had a VSC-HVDC station that has stabilized the voltage in the Gotlandic grid. HVDC Light has solved some of the technical limitations that has hindered the growth of wind turbines. The station has almost reached the end of its technical lifespan and the thesis is a first step in the investigation for an eventual replacement. With power quality analyzers performance and improvements were investigated. The Swedish transmission system operator Svenska kraftnät have since 2016 had a VSCHVDC station that have been used to exchange energy to the asynchronous system in Eastern Europe. Svenska kraftnät has of lately wanted to take advantage of the voltage control capabilities and investigate measurement technologies. Power quality analyzers were installed on suitable connection points to investigate the behavior of the station. ELSPEC G4500 power quality analyzers were installed on Nordbalt’s and Gotland’s HVDC Light to measure during the late summer to fall of 2017. The differences between CVT and IVT, and Rogowski coil and CT were captured. The choice of ELSPEC suited well since no trigger conditions are needed which allows small but important errors to be discovered. Gotland’s HVDC Light stabilizes the grid mostly against voltage sags for faults in the 10 kVgrid and flicker from the wind farms in node 2. The rated power for an upgrade should be based on the 10 kV-grid short-circuit power to be developed during the station’s life span. The voltage control should be based on a PI-controller and should be able to transmit full power in 40 ms. Component voltages can be used to produce reactive power on the phases in need. The need against flicker should be based on measurements in the closest center of consumption, 2 miles from node 2. If it’s needed, a separate loop for flicker-control that prevents 1.5 Hz-components based on a point 1.5 mile from node 2 should be implemented. Depending on an upgrade or replacement, the topology can either be “two-level generation 3” or MMC. Many different future scenarios affect HVDC Light’s role and more resources will be required for Gotland’s and HVDC Light’s future. Two behaviors have been observed on Nordbalt where one is slow and the other behavior is fast. The fast behavior was only observed when low impedance faults occurs, that the synchronous generators also handles. Nordbalt can contribute to small voltage variations if it was faster, like the behavior for low impedance faults. A consideration in stability is needed since a faster behavior can lead to stability problems. Since the data between CVT and IVT differed a lot, metrology problems were discovered. Some solutions were discussed, of which RCVT and PQSensor was thoroughly reviewed. Alternative solutions like MoW and mobile units were also presented vaguely. All solutions showed practical difficulties, which complicates the implementation. It is therefore encouraged to test the technologies in a laboratory environment since few studies have been made on these and competing manufactures state different information.

Inledning: Balans eller postural kontroll är en fundamental sensomotorisk funktion för att kunna hålla sig upprätt. Ett viktigt sensoriskt system för den posturala kontrollen är proprioceptionen, vilken bidrar till god precision, ledstabilitet, koordination och balans vid rörelse. Den mekanoreceptor som anses ha störst betydelse för proprioceptionen är muskelspolen, vilken har till uppgift att registrera och reglera längd och förändring av längd hos en muskel. Det går att träna upp sin balans genom träning på instabilt underlag men det är oklart huruvida ankelproprioception påverkas. Syfte: Studiens syfte var att genomföra en initial pilotstudie för att undersöka eventuella indikationer på effekter på dels postural kontroll generellt och dels mer specifikt ankelproprioception. Detta mättes i stillastående balanstest efter balansträning på instabilt jämfört med stabilt underlag med och utan lokal muskelvibration på en grupp unga friska män och kvinnor. Metod: Nio deltagare rekryterades varav fyra stycken slumpades till den grupp som tränade på stabilt underlag (SG) och fem stycken till den grupp som tränade på instabilt underlag (IG). Fyra balanstester utfördes innan och efter träningsperioden genom test av postural svaj mätt med Wii Balance Board (WBB). Resultat: För IG sågs en sänkning av Velocity under och efter vibration av vaderna. Inga tydliga skillnader framkom gällande Anterio-Posterior amplitud inom eller mellan grupperna.Konklusion:Resultatet indikerar att träning på instabilt underlag kan ha påverkan på proprioceptionen i anklarna vad gäller att minska svajets hastighet under och efter vadmuskelvibration. Dessa resultat motiverar större studie med fler deltagare för statistiska analyser av eventuella skillnader inom och mellan grupperna.

Abstract:
L’objectif de cette étude est de vérifier si un ciment ostéoconducteur résorbable utilisé comme technique de fixation de greffons ostéochondraux permet d'obtenir une stabilité initiale supérieure à celle obtenue avec la technique de mosaicplastie originalement décrite. Il s’agit d’une étude biomécanique effectuée sur des paires de fémurs cadavériques bovins. Pour chaque paire de fémurs, des greffons ostéochondraux autologues ont été insérés et stabilisés au moyen d’un ciment biorésorbable (Kryptonite, DRG inc.) sur un fémur alors qu’au fémur controlatéral, les greffons ont été implantés par impaction selon la technique usuelle de mosaicplastie. Des greffons uniques ainsi que des greffons en configuration groupée ont été implantés et soumis à une évaluation biomécanique. Les charges axiales nécessaires pour enfoncer les greffons de 1, 2 et 3 mm ont été comparées en fonction de la technique de stabilisation utilisée, ciment ou impaction, pour chaque configuration de greffons. Les résultats démontrent que les greffons ostéochondraux cimentés uniques et groupés ont une stabilité initiale supérieure à celle de greffons non cimentés sur des spécimens cadavériques bovins. L’obtention d’une plus grande stabilité initiale par cimentation des greffons ostéochondraux pourrait permettre une mise en charge précoce post-mosaicplastie et mener à une réhabilitation plus rapide.
The objective of this project is to compare the primary stability of osteochondral autografts stabilized with a resorbable osteoconductive bone cement to that of bottomed press fit grafts inserted according to the original mosaicplasty technique. Biomechanical testing was conducted on pairs of cadaveric bovine femurs. For each femoral pair, osteochondral grafts were inserted and stabilized with an osteoconductive bone cement (Kryptonite, DRG inc.) on one bone whereas on the controlateral femur, grafts were inserted in a press fit fashion. Grafts were inserted in 2 different configurations, single grafts as well as groups of 3 adjacent grafts, and submitted to biomechanical testing. Axial loads needed to sink the grafts to 1, 2 and 3 millimeters below cartilage level were recorded and compared according to the fixation technique, cement or press-fit impaction, for each graft configuration. According to those results, cemented osteochondral autografts appear more stable than press fit grafts for both single and 3-in-a-row configurations. Using such a cementation technique could potentially prevent the initial loss of stability that has been shown to occur with osteochondral grafts in the post-operative period, allowing patients to perform early weight bearing and rehabilitation.

Cells in our body are challenged every day by DNA damage arising as a result of both endogenous and exogenous insults. The ability of cells to repair DNA lesions is essential for correct propagation of genetic information. The most cytotoxic DNA lesion is DNA double-strand break (DSB) while oxidative DNA damage is one of the most frequent lesions. The aim of this thesis was to improve current the knowledge of the molecular mechanisms underlying the repair of DSBs and oxidative DNA damage. The major source of oxidative damage in cells are reactive oxygen species that are constantly generated as by-products of cell metabolism. One of the most frequent lesions is 7,8- dihydro-8-oxo-guanine (8-oxo-G) that gives rise to 8-oxo-G:A mispairs during DNA replication and if left unrepaired, results in accumulation of DNA mutations. We found that Werner helicase (WRN) physically interacts with DNA polymerase λ (Polλ) and stimulates DNA gap-filling by Polλ opposite to 8-oxo-G followed by strand displacement synthesis in MutY DNA glycosylase homolog (MUTYH) initiated base excision repair (BER) of 8-oxo-G:A mispairs. There are two major pathways involved in repair of DSBs: homologous recombination (HR) and non-homologous end joining (NHEJ). NHEJ is highly error-prone while HR is error-free. There are two...

Abstract:
DNA replication is the most vulnerable process of DNA metabolism in proliferating cells and therefore it is tightly controlled and coordinated with processes that maintain genomic stability. Human RecQ helicases are among the most important factors involved in the maintenance of replication fork integrity, especially under conditions of replication stress. Collisions between replication and transcription machineries represent a significant source of genomic instability. RECQ5 DNA helicase binds to RNA-polymerase (RNAP) II during transcription elongation and suppresses transcription-associated genomic instability. Here we show that RECQ5 also associates with RNAPI and enforces the stability of ribosomal DNA arrays in cells exposed to replication stress. We demonstrate that RECQ5 associates with transcription complexes in DNA replication foci and counteracts replication fork stalling in RNAPI- and RNAPII-transcribed genes, suggesting that RECQ5 exerts its genome stabilizing effect by acting at sites of concomitant replication and transcription. BACKGROUND: The inactivation of the tumor suppressor gene BRCA1 is a predisposing factor for a breast/ovarian cancer development. Formation of cancer-specific alternative splicing variants with aberrant biological properties can represent additional mechanism decreasing the overall BRCA1 activity in DNA double strand break (DDSB) repair. In this study, we analyzed BRCA1 alternative splicing variants BRCA114-15 and 17-19 ascertained previously during the screening of high-risk breast cancer individuals. METHODS: We established a stable MCF-7 cell line-based model system for an in vitro analysis of BRCA1 variants. Using this system, we analyzed the impact of BRCA114-15 and 17-19 variants on DNA repair kinetics using comet assay and confocal immunomicroscopy. The capacity of DNA repair was assessed directly by an in vitro NHEJ assay and indirectly by a mitomycin C sensitivity test. The proliferation activities were determined by a clonogenic assay and growth curves. RESULTS: Overexpression of BRCA114-15 and 17-19 increases the endogenous level of DNA damage, slows down the DDSB repair, and decelerates the initial phase of radiation-induced foci formation and prolongs their persistence. Abstract:
Ionisierende Strahlung (IR) ist in der medizinischen Diagnostik und in der Tumortherapie von zentraler Bedeutung, kann aber Genominstabilität und Krebs auslösen. Strahleninduzierte Genominstabilität (RIGI) ist in den klonalen Nachkommen bestrahlter Zellen zu beobachten, die zugrundeliegenden Mechanismen sind jedoch noch unverstanden. Zur Erforschung von verzögerten Strahleneffekten wurden primäre embryonale Fibroblastenkulturen mit 2 Gray bestrahlt und für 20 Populationsverdopplungen klonal expandiert. Zellen, die keiner Strahlung ausgesetzt waren, dienten als Kontrolle für normale Alterungsprozesse. Die Klone wurden durch klassische Chromosomenbänderungstechniken analysiert und in Abhängigkeit der Stabilität ihres Genoms in Gruppen eingeteilt. Ein Klon wurde als stabil gewertet, wenn die analysierten Metaphasen keinerlei Auffälligkeiten zeigten, während instabile Klone ein Mosaik aus normalen und abnormalen Metaphasen waren. Die Zellen von zwei Spendern wurden untersucht, um interindividuelle Strahleneffekte zu beurteilen. Nach Bestrahlung hatten mehr als die Hälfte der Klone Metaphasen mit strukturellen Aberrationen und wurden dementsprechend als instabil eingestuft. Drei Klone zeigten zudem numerische Aberrationen, die ausschließlich das Y Chromosom betrafen. Fluoreszenz in situ Hybridisierungen verifizierten diese Beobachtung in weiteren Klonen und deuteten an, dass der Verlust des Y Chromosoms mit RIGI assoziiert ist. Molekulare Karyotypisierungen mit SNP Arrays ergaben, dass IR in den Klonen Veränderungen der Kopienzahl auslöst. Ein Unterschied zwischen chromosomal stabilen und instabilen Klonen konnte jedoch nicht detektiert werden. Chromosomale Regionen, in denen sich bekanntermaßen fragile Stellen befinden, zeigten eine Anhäufung von CNVs. Ein RIGI Effekt konnte für die fragile Stelle 3B, in der sich das Gen FHIT befindet, identifiziert werden. Exom Sequenzierungen von Klonen und der entsprechenden Massenkultur zeigten eine alterungsassoziierte Entstehung von Varianten. Der Effekt wurde durch die Einwirkung von Strahlung erhöht. Auf Ebene von einzelnen Nukleotiden konnten ebenfalls Anhäufungen von Schäden in bestimmten genomischen Bereichen detektiert werden, dieser Effekt ging ohne die typischen RIGI Endpunkte einher. Die Ergebnisse der vorliegenden Arbeit zeigen, dass strahlenbedingte Veränderungen auf verschiedenen Ebenen (Chromosomen, Genkopienzahl und einzelnen Nukleotiden) beobachtet werden können, welche, unabhängig von RIGI, die Tumorentstehung begünstigen. Speziell Veränderungen im FRA3B Lokus und der Verlust des Y Chromosoms scheinen jedoch über die Destabilisierung des Genoms zur Krebsentstehung beizutragen
Ionizing radiation is important in medical diagnosis and cancer treatment but can lead to genomic instability and secondary malignancies as a late effect. Radiation induced genomic instability (RIGI) can be observed in the progeny of irradiated cells but the underlying cellular processes remain to be elucidated. To study delayed genetic radiation effects, single cell fibroblast clones from two different male donors were established after a single X ray dose of 2 Gray. Non irradiated cells were used as controls to account for aging related effects. Clones were characterized using chromosome banding analysis. Stable clones were endowed with no karyotype abnormalities in all analysed metaphases after 20 Population doublings, whereas unstable clones showed both normal and abnormal metaphases. Two fibroblast strains were used to detect differences in radiation sensitivity. More than half of the irradiated clones were chromosomally abnormal and thus classified as unstable. Both banding analysis and fluorescence in situ hybridization revealed an increased rate of Y chromosome loss in irradiated clones which might be associated with RIGI. Using SNP Arrays, an increased rate of de novo copy number variations (CNVs) was detected in the progeny of irradiated cells when compared to non irradiated controls. However, a significant difference between chromosomally stable and unstable clones was not found. CNVs clustered in chromosomal regions that are associated with known fragile sites. The fragile site 3B, which encompasses the gene FHIT, was found to be affected by CNVs in unstable clones suggesting a RIGI related effect. Exome sequencing of clones and the respective primary cultures revealed an increased rate of variants during in vitro aging. This effect was further increased by radiation exposure. Analysis of single nucleotides showed a RIGI independent accumulation of damage in specific regions. The results of the present study show that radiation induced changes can manifest as chromosomal abnormalities, copy number variations and DNA sequence mutations promoting tumorigenesis independent from RIGI. However, changes in FRA3B and loss of Y chromosome might trigger cancer through destabilizing the genome

Abstract:
This work presents the re-engineering of the TRANSAT 1.0 code which was developed to perform off-design and transient condition analysis of Saturators and Direct Contact Heat Exchangers. This model, now available in the 2.0 release, was originally implemented in FORTRAN language, has been updated to C language, fully coded into MATLAB/Simulink® environment and validated using the extensive set of data available from the MOSAT project, carried out by the Thermochemical Power Group of the University of Genoa. The rig consists of a fully instrumented modular vertical saturator, which is controlled and monitored with a LABVIEW® computer interface. The simulation software showed fair stability in computation and in response to step variation of the main parameters driving the thermodynamic evolution of the air and water flows. Considering the actual mass flow rates, a geometric similitude was used to avoid calculation instability due to flows under 100 g/s. Overall the model proved to be reliable and accurate enough for energy system simulations.

Abstract Buoyancy-driven flows are widespread in diverse engineering applications. Such flows have been studied in great detail theoretically, experimentally, and numerically. However, the fluid-dynamic instabilities and flow reversals of thermosiphon are still actively investigated. The presence of such instabilities limits the effectiveness of such devices for decay heat removal. Traditionally the stability analysis of natural convection loops has been confined to one-dimensional calculations, on the argument that the flow would be mono-dimensional when the ratio between the radius of the loop and the radius of the pipe is much larger than 1. Nevertheless, accurate velocity measurements of the flow in toroidal loops have shown that the flow presents three-dimensional effects. Previous works of the authors have shown that these structures can be seen in thermosiphons. In this paper, we aim to use modern CFD methods to investigate the three-dimensional flow in thermosiphons. This paper focuses on rectangular thermosiphons. In particular, we perform a series of high-fidelity simulations using the spectral element code Nek5000 to investigate the stability behavior of the flow in a rectangular thermosiphon. We compare the results with available existing experimental data from the L2 facility in Genoa. We examine in detail the flow structures generated. Moreover, in the past various authors have demonstrated that the overall behavior of the thermosiphon depends strongly on the boundary conditions (BCs). The simulation campaign was carried out with different BCs to investigate and confirm this effect. In particular, simulations with Dirichlet, Neumann and Robin BCs for heater and sink were performed.

Abstract:
In this paper two different advanced control approaches for a pressurized SOFC hybrid system are investigated and compared against traditional proportional–integral–derivative (PID). Both advanced control methods use model predictive control (MPC): in the first case, the MPC has direct access to the plant manipulated variables, in the second case the MPC operates on the setpoints of PIDs which control the plant. In the second approach the idea is to use MPC at the highest level of the plant control system to optimize the performance of bottoming PIDs, retaining system stability and operator confidence. Two MIMO (multi-input multi-output) controllers were obtained: fuel cell power and cathode inlet temperature are the controlled variables; fuel cell by-pass flow, current and fuel mass flow rate (the utilization factor kept constant) are the manipulated variables. The two advanced control methods were tested and compared against the conventional PID approach using a SOFC hybrid system model. Then, the MPC controller was implemented in the hybrid system emulator test rig developed by the Thermochemical Power Group (TPG) at the University of Genoa. Experimental tests were carried out to compare MPC against classic PID method: load following tests were carried out. Ramping the fuel cell load from 100% to 80% and back, keeping constant the target of the cathode inlet temperature, the MPC controller was able to reduce the mismatch between the actual and the target values of the cathode inlet temperature from 7 K maximum of the PID controller to 3 K maximum, showing more stable behavior in general.

In the recent years, as the technical developments in the field of GT related technology are more and more driven by regulations on environmental pollution control, a whole series of different industrial evolution and innovation lines are investigated so to make combustion processes ever “cleaner”. Among those, there is for sure the adoption of lean and ultra lean combustion processes to be pursued by means of air-fuel premixing combustion technologies. Within this scenario, at DIMSET/SCL (Savona Combustion Laboratory, Dept. of Thermal Machines, Energy Systems and Transportation, Univ. of Genoa) since several years research activities are carried out, mainly within the frame of EC-funded Research Programmes (ICLEAC, MUSCLES, TLC, H2-IGCC) and cooperation with industrial companies of the energy sector (Ansaldo Energia S.p.A.) and aero-propulsion (Avio Group) sectors. Research activities can take advantage of a close integration between experimental facilities, such as several reactive and non-reactive dedicated burner test-rigs, instrumented with LDV, PDA and PIV laser-based equipment, as well as of in-house continuously improved reactive Navier-Stokes solvers for combustor analysis (NastComb solver) and design (TPM method). The paper deals with the stability characterisation of the different combustion-processes taking place within several GT power plants, namely, the heavy duty AE64-3A heavy duty gas turbine (Ansaldo Energia), already present on the market, the so-called Liquid and Gas Rapid Pre-Mix burners, LRPM and GRPM, designed at DIMSET/SCL and still prototypical, and the Avio-designed LPP (Lean Premixed Prevaporised) burner, for aero-engine applications. The research has been addressed at in-depth characterising the stability behaviour of the burner’s operation. In particular, those aspects have been investigated deemed of greatest importance in affecting a stable performance profile, such as swirlers’ design, burner’s internal aerodynamics, premixing duct configuration, fuel typology and injection modalities, etc. The paper gives a synoptic view both of the research approaches (experimental, instrumental, numerical analysis and design) jointly pursued by DIMSET/SCL team in investigating the combustion instability, as well as of the obtained results, which help in pointing out those burner design and operational parameters which appear as most critical in affecting instability insurgence and self-sustainment.

Abstract:
Plasmin and leucocyte elastase are regarded as the two medically most important fibrin(ogen)-degrading proteolytic enzymes. There is, however, a considerable difference in information available about the cleavage specificities and fragmentation pathways of these two enzymes. Degradation by plasmin has been studied already for a long time in great detail so that now the time course of the degradation, the cleavage sites and the functional properties of many fragments are well known. In contrast, relatively little is known about the degradation by leucocyte elastase, except that the overall cleavage pattern resembles that obtained with plasminIn this investigation the leucocyte elastase-mediated degradation of fibrinogen has been examined by means of proteinchemi-cal methods. Human fibrinogen was incubated with human enzyme material for various periods of time and at some different enzyme concentrations. The split products formed at the various stages were isolated in pure form by gel filtration followed by reversed-phase high-performance liquid chromatography. The fragments were identified by N-terminal amino acid sequence and amino acid composition. The course of the degradation was also monitored by sodium dodecylsulfate-polyacrylamide gel electrophoresis. All cleavage patterns were compared with the corresponding patterns from plasmic degradation. It could be confirmed that X-, D- and E-like fragments are formed also with elastase. However, several early elastolytic Aα-chain fragments are characteristically different from plasmic fragments. The previously identified N-terminal cleavage site in the Aα-chain, i.e. after position 21, was found to be the most important site in this region of fibrinogen. The very early degradation of the Aα-chain N-terminus by elastase is in strong contrast to the stability against plasmin. Several cleavage sites in N-terminal region of the Bβ-chain were observed, though the low amino acid specificity of elastase partly hampered the identification. The γ-chain N-terminus was found to be as highly stable towards elastase as towards plasmin. The results are expected to contribute to the understanding of the role of leucocyte elastase in pathophysiologic fibrino(geno)lysis
