Abstract:
AbstractThe double-stranded multifunctional RNA-binding protein (dsRBP) Staufen was initially discovered in insects as a regulator of mRNA localization. Later, its mammalian orthologs have been described in different organisms, including humans. Two human orthologues of Staufen, named Staufen1 (STAU1) and Staufen2 (STAU2), share some structural and functional similarities. However, given their different spatio-temporal expression patterns, each of these orthologues plays distinct roles in cells. In the current review, we focus on the role of STAU1 in cell functions and cancer development. Sinc

Mammalian Staufen2 (Stau2) is involved in mRNA transport in neurons. Here,we report that Stau2 is a double-stranded RNA-binding protein that is mainly expressed in the brain. We show that Stau2 is found in the somatodendritic compartment of neurons. In dendrites, Stau2 is aligned on individual tracts and colocalizes with microtubules. Stau2 is expressed as at least three splice isoforms, which can be observed in several subcellular complexes. Although a 62 kDa isoform (Stau262) fractionates in ribosome-free fractions of light density, Stau259 and Stau252 are found in high-density complexes. Th

Abstract:
RNA-binding proteins (RBPs) act as posttranscriptional regulators controlling the fate of target mRNAs. Unraveling how RNAs are recognized by RBPs and in turn are assembled into neuronal RNA granules is therefore key to understanding the underlying mechanism. While RNA sequence elements have been extensively characterized, the functional impact of RNA secondary structures is only recently being explored. Here, we show that Staufen2 binds complex, long-ranged RNA hairpins in the 3′-untranslated region (UTR) of its targets. These structures are involved in the assembly of Staufen2 into RNA granu

In this study, I have shown that XStau 1 and 2 are expressed throughout oogenesis, embryogenesis and in the adult Xenopus. Immunostaining and confocal microscopy has demonstrated that both XStaul 1 and 2 localise to the vegetal cortex during oogenesis and colocalise with the ER but not the MC, at a time consistent with a role in the vegetal localisation of late pathway mRNAs. Biochemical analysis has shown that on meiotic maturation XStau 1 and 2 become phosphorylated and that this phosphorylation coincides with their rearrangement at the vegetal cortex in mature eggs, where they no lon

Staufen1 is a double-stranded RNA-binding protein known to be involved in the transport, localization, decay and increased translation of some mRNAs. The goal of the present study is to determine the role of Staufen1 during myogenic differentiation by characterizing the effects of Staufen1 over-expression in C2C12 cells. Immunofluorescence experiments revealed that Staufen1 over-expression causes a decrease in the fusion and differentiation indices and leads to the formation of myotubes with significantly fewer nuclei. Human Immunodeficiency Virus type 1 (HIV-1) is a member of the Retrovirus family and is the causative agent of Acquired Immunodeficiency Syndrome (AIDS). As an obligate intracellular parasite, HIV-1 uses cellular proteins and machinery to ensure transmission to uninfected cells. One such cellular protein involved in the late stages of viral replication is Staufen1. It is a main component of mRNA ribonucleoproteins (RNPs) and plays distinct roles in mRNA transport, translation and decay. In the context of HIV-1, Staufen1 was initially found to be selectively encapsidated into the virions. Subse
