Abstract:
The invisible past of St. Eustatius Saint Eustatius, a small Dutch Caribbean island, was one of the busiest transit harbors in the 18th-century Atlantic World. While its importance as an entrepot in colonial trade networks has been well researched, much less is known about the less visible past on the island, which includes its plantation landscape and the lives of people belonging to the lowest social classes. Recent archaeological and documentary research have produced valuable data that sheds more light on these topics. This article discusses the significance of various elements in the island’s rural landscape, how they affected the experiences of the people living in it, and how these have shaped the history of the island.

The ste mutants (ste2, ste4, ste5, ste7, ste11, and ste12) are insensitive to mating factors and are, therefore, sterile. Roles of the STE gene products in the GPA1-mediated mating factor signaling pathway were studied by using ste gpa1 double mutants. Mating efficiency of a ste2 mutant defective in the alpha-factor receptor increased 1,000-fold in a gpa1 background, while G1 arrest and aberrant morphology (shmoo) caused by gpa1 were not suppressed by ste2. Furthermore, the steady-state level of the FUS1 transcript, which normally increases in response to mating factors, was also elevated when the GPA1 function was impaired. These results suggest that the GPA1 protein functions downstream of the STE2 receptor. Conversely, the sterility of ste4, ste5, ste7, ste11, and ste12 mutants was not suppressed by gpa1, but the lethal phenotype of gpa1 was suppressed by these ste mutations. Northern (RNA) blotting analysis revealed that the ste7, ste11, and ste12 mutations caused reductions of 50 to 70% in the steady-state levels of the GPA1 transcript, while ste4 had a slight effect and ste5 had no effect. This implies that the suppression by ste7, ste11, and ste12 could be due to reduced syntheses of additional components, including an effector, and that suppression by ste4 and ste5 may result from direct effects on the signaling pathway. The STE4, STE5, STE7, STE11, and STE12 products, therefore, appear to specify components of the signal transduction machinery, directly or indirectly, which function together with or downstream of GPA1.

The STE5 gene encodes an essential element of the pheromone response pathway which is known to act either after the G subunit encoded by the STE4 gene or at the same step. Mutations in STE5, designated STE5Hyp, that partially activate the pathway in the absence of pheromone were isolated. One allele (STE5Hyp-2) was shown to cause a single amino acid substitution near the N terminus of the predicted STE5 protein. Immunoblotting with anti-Ste5 antibodies indicated that the phenotype was not due to an increased level of the mutant STE5 protein. A multicopy episomal plasmid containing a STE5Hyp allele partially suppressed both the block in pheromone-inducible transcription and the sterility phenotype caused by null alleles of the STE2, STE4, or STE18 gene, indicating that the STE5 product acts after the receptor (STE2 product) and after the G protein beta and gamma subunits (STE4 and STE18 products, respectively). However, the phenotypes of the STE5Hyp mutations were less pronounced in ste4 and ste18 mutants, suggesting that the STE5Hyp-generated signal partially depends on the proposed G beta gamma complex. The STE5Hyp alleles did not suppress ste7, ste11, ste12, or fus3 kss1 null mutants, consistent with previous findings that the STE5 product acts before the protein kinases encoded by STE7, STE11, FUS3, and KSS1 and the transcription factor encoded by STE12. The mating defects of the ste2 deletion mutant and the temperature-sensitive ste4-3 mutant were also suppressed by overexpression of wild-type STE5. The slow-growth phenotype manifested by cells carrying STE5Hyp alleles was enhanced by the sst2-1 mutation; this effect was eliminated in ste4 mutants. These results provide the first evidence that the STE5 gene product performs its function after the G protein subunits.

Die beginsel van partyoutonomie is ’n belangrike beginsel van die kontraktereg en word vandag ook beskou as die belangrikste verbindingsfaktor in die internasionale kontraktereg. In gevorderde ekonomieë word partye toegelaat om die reg te kies wat van toepassing moet wees op die regte en verpligtinge van hul internasionale kontrakte ter bevordering van grensoorskrydende handel en beleggings. Die outeur ondersoek in hierdie artikel die teoretiese basis vir die handhawing van die beginsel van partyoutonomie met betrekking tot regskeuse in internasionale kontrakte. Afgesien van die tradisionele regverdiging vir die bevordering van die beginsel, word in die artikel ook grondwetlike verankering oorweeg. In hierdie verband stel dit voor dat grondwetlike bepalings wat die vrye ontwikkeling van persoonlikheid of menswaardigheid wil beskerm, ook die reg van private partye omvat om die reg van toepassing op hulle internasionale kontrakte te kies. Dieselfde geld met betrekking tot die bepaling wat in sekere grondwette gevind word ter bevordering van aktiwiteite wat kan lei tot die vestiging van ’n gesonde ekonomie. Die artikel is daarop gemik om regsekerheid en voorspelbaarheid te bevorder deur middel van partyoutonomie in die regskeuse vir kontrakte. Sodanige erkenning sal na verwagting die nodige vertroue in private sakepersone en beleggers inboesem en gevolglik internasionale handel en beleggingsaktiwiteite binne ’n bepaalde jurisdiksie bevorder. In die lig van die bogenoemde, stel hierdie artikel voor dat, tensy daar dwingende redes bestaan om partye die reg te ontsê om die stel regsreëls te kies wat hulle verkies om hul internasionale kommersiële kontrakte te reguleer, die howe oor die algemeen hulle rol moet beperk tot die interpretasie en toepassing van kontraktuele bedinge.

Abstract:
Various types of hazardous substances are generated at fire scenes. Firefighters usually use the self-contained breathing apparatus (SCBA) during firefighting; however, SCBA is very inconvenient to use in other works (e.g., fire investigation and fire scene commands). Therefore, firefighters can be exposed to numerous chemicals. In this study, concentrations of hazardous gases were measured by utilizing gas analyzers with seven sensors during fire investigations. Six fire investigators measured the concentrations of hazardous gases directly as they worked. This included capturing the maximum concentrations of SO2 at seven places, HCHO at 29 places, NO2 at one place, HCN at 13 places, and CO at two places where the concentration exceeded the short-term exposure limit (STEL). When reconstruction experiments were performed, the maximum allowable concentrations for most hazardous chemicals fell below the STEL approximately 90 min after the fire occurrence. Therefore, we determined that fire investigators should wear proper respiratory protective equipment when working.

Die beslissing van die hoogste hof van appèl verdien instemming. Dit maak egter nie sin dat hierdie hof met sy beslissing voortgegaan het nadat bevind is dat nalatigheid afwesig was en dat deliktuele aanspreeklikheid dus ontbreek het nie.

Abstract:
Safely achieving the goals of the Paris Climate Agreement requires a worldwide transformation to carbon-neutral societies within the next 30 y. Accelerated technological progress and policy implementations are required to deliver emissions reductions at rates sufficiently fast to avoid crossing dangerous tipping points in the Earth’s climate system. Here, we discuss and evaluate the potential of social tipping interventions (STIs) that can activate contagious processes of rapidly spreading technologies, behaviors, social norms, and structural reorganization within their functional domains that we refer to as social tipping elements (STEs). STEs are subdomains of the planetary socioeconomic system where the required disruptive change may take place and lead to a sufficiently fast reduction in anthropogenic greenhouse gas emissions. The results are based on online expert elicitation, a subsequent expert workshop, and a literature review. The STIs that could trigger the tipping of STE subsystems include 1) removing fossil-fuel subsidies and incentivizing decentralized energy generation (STE1, energy production and storage systems), 2) building carbon-neutral cities (STE2, human settlements), 3) divesting from assets linked to fossil fuels (STE3, financial markets), 4) revealing the moral implications of fossil fuels (STE4, norms and value systems), 5) strengthening climate education and engagement (STE5, education system), and 6) disclosing information on greenhouse gas emissions (STE6, information feedbacks). Our research reveals important areas of focus for larger-scale empirical and modeling efforts to better understand the potentials of harnessing social tipping dynamics for climate change mitigation.

ABSTRACT We study the relationship between supermassive black hole (BH) feedback, BH luminosity, and the kinematics of stars and gas for galaxies in IllustrisTNG. We use galaxies with mock MaNGA observations to identify kinematic misalignment at z = 0 (difference in rotation of stars and gas), for which we follow the evolutionary history of BH activity and gas properties over the last 8 Gyr. Misaligned low-mass galaxies ($\mathrm{\mathit{ M}_{stel} \lt 10^{10.2}\, \mathrm{M}_{\odot }}$) typically have boosted BH luminosity and BH growth, and have had more energy injected by BHs into the gas over the last 8 Gyr in comparison to low-mass aligned galaxies. These properties likely lead to outflows and gas removal, in agreement with active low mass galaxies in observations. Splitting on BH luminosity at z = 0 produces consistent distributions of kinematic misalignment at z = 0; however, splitting on the maximum BH luminosity over the last 8 Gyr produces statistically significant different distributions. While instantaneous correlation at z = 0 is difficult due to misalignment persisting on longer time-scales, the relationship between BH activity and misalignment is clear. High-mass quenched galaxies ($\mathrm{\mathit{ M}_{stel} \gt 10^{10.2}\, \mathrm{M}_{\odot }}$) with misalignment typically have similar BH luminosities, show lower gas fractions, and have typically lower gas phase metallicity over the last 8 Gyr in comparison to the high mass aligned.

BACKGROUND: The safety and health of healthcare workers employed in pathology laboratories and exposed to formaldehyde (FA) is a matter of concern worldwide, as several health effects have been observed in workers resulting from exposure to FA, both short and long-term. OBJECTIVE: The study was aimed to describe the strategy implemented in a hospital pathology laboratory to minimize workers’ exposure to FA through interventions to working environment and workforce. METHODS: The NIOSH 2016 method for detecting gaseous FA was adopted to perform personal and area active sampling of FA. The samples were subsequently analyzed by High Performance Liquid Chromatography. The exposure to FA was measured before and after improvement interventions. RESULTS: The pre-intervention step showed FA levels exceeding the threshold limit values (TLV) established by ACGIH, both the time-weighted average (TLV-TWA) and short term exposure limit (TLV-STEL); after the improvement interventions, the median concentrations of personal and area FA sampling were respectively of 0.025 ppm (Range = 0.023–0.027) and 0.023 ppm (Range = 0.022–0.028) and significantly lower than pre-intervention step (p < 0.05) and below the TLV-TWA and TLV-STEL established by ACGIH. CONCLUSIONS: In our study the workers’ involvement in the risk management of FA exposure together with engineering improvements revealed a strategic way to minimize the FA pollution in the studied laboratory. Healthcare companies should consider the need to ensure the workers’ participation in the management of occupational hazards, including FA, to reach the goal of healthy workplaces.

Trimethylamine (TMA) is a gas at ambient temperature, which has a pungent, fishy odour. It is commercially available as a compressed gas, 40% aqueous solution or a 33 % solution in ethanol. It is used in organic synthesis, especially of choline salts, as a warning agent for natural gas and flotation agents, in the production of cationic starches, quaternary ammonium compounds, intense sweeteners and strongly basic anion exchange resins. Moreover, it is used in the production of disinfectants and insect attractants. TMA is irritating to the human respiratory tract, skin and eyes. The threshold of irritation was reported to be 1481 mg/m3 (median) after a single dose. No effects were observed in workers exposed to 0.24-19.5 mg/m³, most measurements being below 12.1 mg/m3. A LOAEC of 48 mg/m³ was established for human based on eyes, nose and throat irritation. Animal data with repeated inhalation exposure over 2 weeks revealed a LOAEC of 183.75 mg/m³ based on respiratory irritation in a rat study. Embryotoxic effects were observed in mice (NOAEL of 150 mg/kg bw/day). The Scientific Committee for Occupational Exposure Limits to Chemical Agents (SCOEL) has established OEL of 4.9 mg/m³ and STEL of 12.5 mg/m³. A MAC value has been derived using the RD50 value (147.62 mg/m3 for TMA) and multiplying it by a factor of 0.03. The Expert Group for Chemicals Agents has proposed to reduce the current MAC value from 12 mg/m³ to 4.9 mg/m³ and the current STEL value from 24 mg/m³ to 12.5 mg/m³, which is also in accordance with the values recommended by SCOEL. It has been proposed to remain the “I” (irritant) labelling of TMA. No bases for a BEI value have been found.

Mutations in STE4, 5, 7, and 11 partially reduced the number of binding sites, but this reduction was not sufficient to explain the loss of responsiveness; the products of these genes appear to affect postreceptor steps of the response pathway. As a second method of distinguishing the roles of the various STE genes, we examined the sterile mutants for suppression. Mating of the ste2-3 mutant was apparently limited by its sensitivity to alpha-factor, as its sterility was suppressed by mutation sst2-1, which leads to enhanced alpha-factor sensitivity. Sterility resulting from each of four ste4 mutations was suppressed partially by mutation sst2-1 or by mutation bar1-1 when one of three other mutations (ros1-1, ros2-1, or ros3-1) was also present. Abstract. Prescribed burning represents a serious threat to the personnel fighting fires because of smoke inhalation. This study aims to increase the knowledge about foresters exposure to the prescribed burning smoke by focusing on exposure to volatile organic compounds (VOCs). We initially assessed the methodology for smoke sampling. Then, we identified potentially dangerous molecules among the VOCs identified at 4 prescribed burning sites located around Corsica. The values measured were very high, exceeding the exposure limits, particularly for benzene, phenol, and furfural, whose concentrations were above short-term exposure limit (STEL) values. In conclusion, obvious but necessary recommendations were made for the protection of the personnel involved in fighting fires on a professional basis.

Abstract The mating process in yeast has two distinct aspects. One is the induction and activation of proteins required for cell fusion in response to a pheromone signal; the other is chemotropism, i.e., detection of a pheromone gradient and construction of a fusion site available to the signaling cell. To determine whether components of the signal transduction pathway necessary for transcriptional activation also play a role in chemotropism, we examined strains with null mutations in components of the signal transduction pathway for diploid formation, prezygote formation and the chemotropic process of mating partner discrimination when transcription was induced downstream of the mutation. Cells mutant for components of the mitogen-activated protein (MAP) kinase cascade (ste5, ste20, ste11, ste7 or fus3 kss1) formed diploids at a frequency 1% that of the wild-type control, but formed prezygotes as efficiently as the wild-type control and showed good mating partner discrimination, suggesting that the MAP kinase cascade is not essential for chemotropism. In contrast, cells mutant for the receptor (ste2) or the β or γ subunit (ste4 and stel8) of the G protein were extremely defective in both diploid and prezygote formation and discriminated poorly between signaling and nonsignaling mating partners, implying that these components are important for chemotropism.

Ste5 is essential for the yeast mating pheromone response pathway and is thought to function as a scaffold that organizes the components of the mitogen-activated protein kinase (MAPK) cascade. A new method was developed to isolate missense mutations in Ste5 that differentially affect the ability of Ste5 to interact with either of two MAPK cascade constituents, the MEKK (Ste11) and the MEK (Ste7). Mutations that affect association with Ste7 or with Ste11 delineate discrete regions of Ste5 that are critical for each interaction. Co-immunoprecipitation analysis, examining the binding in vitro of Ste5 to Ste11, Ste7, Ste4 (G protein, β subunit), and Fus3 (MAPK), confirmed that each mutation specifically affects the interaction of Ste5 with only one protein. When expressed in a ste5Δ cell, mutant Ste5 proteins that are defective in their ability to interact with either Ste11 or Ste7 result in a markedly reduced mating proficiency. One mutation that clearly weakened (but did not eliminate) interaction of Ste5 with Ste7 permitted mating at wild-type efficiency, indicating that an efficacious signal is generated even when Ste5 associates with only a small fraction of (or only transiently with) Ste7. Ste5 mutants defective in association with Ste11 or Ste7 showed strong interallelic complementation when co-expressed, suggesting that the functional form of Ste5 in vivo is an oligomer.
