Breast tumours have intrinsic heterogeneity. The cancer stem cell hypothesis is currently challenging the notion that all cancer cells are equally malignant, suggesting that it is important to evaluate the efficacy of potential anti-cancer drugs by their ability to target the stem-like population. TRAIL is a cytotoxic agent the efficacy of which has been limited by a lack of patient stratification in clinical trials (Lemke 2014). In pre-clinical studies TRAIL has shown specificity towards mesenchymal-like breast cancer cell lines (Rahman et al. 2009) We show here that TRAIL is able to target t

Introduction: Glioblastoma (GBM) is characterised by local recurrence following surgery, radiotherapy and chemotherapy. GBM has a poor prognosis and novel approaches are required. Recently, a hierarchical organisation of tumour cells in GBM has been proposed. This hypothesis suggests only a subset of cancer cells, termed ‘cancer stem-like cells’ (CSCs) drive tumour growth and possess properties of self renewal and unlimited proliferative capacity. CSCs have been described as radioresistant, implicating CSCs as a determinant of tumour recurrence following therapy. Therefore improved patient out

To gain an increased understanding of the mechanisms of GvHD I measured frequencies of Th1, Th17 and Treg subsets in the blood of 32 GvHD patients (during and outside of disease episodes) and in 21 patients who did not suffer GvHD. Introduction: Recent studies have revealed that cancer stem cells (CSCs) exist in malignant disease. Additionally, it is proposed that these cells may survive following chemotherapy, and hence contribute to tumour relapse. A significant mechanism of drug resistance in CSCs is believed to be the expression of ATP-binding cassette (ABC) transporters that efflux cytotoxic agents out of cells. The objective of this study was to study the existence of CSCs in a panel of primary paediatric brain tumours (PBTs) and to determine if these were drug resistant via functional ABC transporters. Colorectal cancer is the 2nd most common cause of death by cancer in the UK, but it is treatable if diagnosed early. In order to increase the likelihood of early diagnosis, more must be understood about the early stages of colorectal tumourigenesis. It is known that intestinal stem cells (ISCs) are the cells of origin of colorectal tumourigenesis, and that an expansion of undifferentiated cell types, akin to ISCs, is one of the earliest events in mouse models of tumourigenesis. This indicates the importance of the relationship between the ISC compartment and tumourigenesis. In order to underst

The induced pluripotent stem cells (iPSCs) technology has revolutionized disease modelling by enabling the generation of patient-specific pluripotent stem cells for the study of complex disorders such as cancer. Somatic cell reprogramming through iPSCs induces global epigenetic reconfiguration of the chromatin which converts cancer cells to an embryonic stem cell-like state with potential reversion of tumorigenicity. Therefore, reprogramming can be used to answer the question as to whether epigenetic alterations alone can be sufficient to induce carcinogenesis, independent of genetic defects.

Abstract:
The natural daily cycles of light and dark have played a fundamental role in shaping the development of an adaptive intrinsic clock mechanism which allows organisms to coordinate the function of multiple organs by setting the correct circadian timing of cellular processes ensuring proper homeostasis. In mammalian skin, homeostasis is maintained by epidermal stem cells (epSCs). EpSCs localize to specialized niches where they undergo cycles of quiescence and proliferation. Several pathways are known to play essential rol
