Abstract:
The Stiles-Crawford effect (SCE) is the differential effectivity of light entering the eye at different positions in the pupil. Its parameters are directionality (rate of change across the pupil) and peak location in the pupil. Using a spatial light modulation system, I investigated influences of field size, accommodation (focusing), and flicker frequency on the subjectively-measured SCE. SCE directionality increased with increase in field size, while peak location shifted temporally in the pupil with increase in focusing, but the SCE was not affected by a range of flicker frequencies. The field size results support a photoreceptor absorption model of the SCE.

Abstract:
The purpose of this study was to examine the reliability and validity of the Child Life Style Scale (CLSS) and clarify the underlying dimensions of the scale. Dr. Kathleen Stiles designed the 90 item CLSS to quantitatively identify life style typologies for children between eight and twelve. This questionnaire consisted of 6 scales based on Adlerian constructs of personality. They are pleasing, getting, controlling, rebelling, inadequacy, and socially useful. Ten items were deleted after an inter-judge reliability/validity study. The 80 item CLSS was administered to 314 third, fourth, and fifth graders in public and private schools in Dallas, Texas. Internal consistency coefficients for the six subscales ranged from .72 to .76 and test-retest reliability coefficients ranged from .70 to .80 (p < .001). Results from a factor analysis confirmed the original six scales but factors five and six were not strong. Exploratory factor analysis found four clear factors with internal consistency coefficients ranging from .76 to .84 and test-retest reliability coefficients ranging from .73 to .83 (p < .001). Underlying dimensions of the factors, which reflect Alfred Adler's four typologies exactly, were: Factor 1: Rebelliousness Factor 2: Social Usefulness Factor 3: Control Factor 4: Fear of Failure. Results of analysis of variance indicated that age and socioeconomics made significant differences while gender and place in the family were not as significant. This study showed the revised 64 item version of the CLSS reflects Adler's tenets exactly, and has a more concise format with better reliability and validity. The CLSS is a solid questionnaire worth being further investigated for use in schools and therapy.

Abstract:
Background: Mechanical forces either due to accommodation or myopia may stretch the retina and/or cause shear between the retina and choroid. This can be investigated by making use of the Stiles-Crawford effect (SCE), which is the phenomenon of light changing in apparent brightness as it enters through different positions in the pupil. The SCE can be measured by psychophysical and objective techniques, with the SCE parameters being directionality (rate of change across the pupil), and orientation (the location of peak sensitivity in the pupil). Aims: 1. To study the changes in foveal SCE with accommodation in emmetropes and myopes using a subjective (psychophysical) technique. 2. To develop and evaluate a quick objective technique of measuring the SCE using the multifocal electroretinogram. Methods: The SCE was measured in 6 young emmetropes and 6 young myopes for up to 8 D accommodation stimulus with a psychophysical technique and its variants. An objective technique using the multifocal electroretinogram was developed and evaluated with 5 emmetropes. Results: Using the psychophysical technique, the SCE directionality increased by similar amounts in both emmetropes and myopes as accommodation increased, with an increase of 15-20% with 6 D of accommodation. However, there were no significant orientation changes. Additional measurements showed that most of the change in the directionality was probably an artefact of optical factors such as higher-order aberrations and accommodative lag rather a true effect of accommodation. The multifocal technique demonstrated the presence of the SCE, but results were noisy and too variable to detect any changes in SCE directionality or orientation with accommodation. Conclusion: There is little true change in the SCE with accommodation responses up to 6 D in either emmetropes or myopes, although it is possible that substantial changes might occur at very high accommodation levels. The objective technique using the multifocal electroretinogram was quicker and less demanding for the subjects than the psychophysical technique, but as implemented in this thesis, it is not a reliable method of measuring the SCE.

Abstract:
Notre objectif a été d'analyser l'influence de la variabilité spatio-temporelle de la réflectance des cônes sur le comptage des cônes par imagerie à haute résolution par optique adaptative plein champ, et de proposer des méthodes pour pallier ces difficultés. La première partie des travaux porte sur l'étude de la variabilité temporelle des cônes. Nous avons confirmé la présence d'un phénomène de scintillement des cônes à une fréquence au moins égale à celle de notre système d'acquisition. Nous avons proposé une procédure d'analyse d'images incluant restauration par déconvolution myope, recalage rigide, fusion d'images pour la construction de cartographies de cônes. Nous avons montré que dans le cas des sujets sains, la procédure de fusion d'image permet de rapprocher la mesure de la densité des cônes des références histologiques. La directionalité de la réflectance des cônes est appelée effet Stiles-Crawford optique. Nous en avons identifié une nouvelle manifestation clinique, à travers une étude portant sur l'analyse d'images d'optique adaptative en périphérie de la rétine. Nous avons montré, à travers l'alternance des mosaïques positive/négative, que la réflectance des cônes peut devenir inférieure à celle des bâtonnets. La comparaison avec les données de la tomographie de cohérence optique a montré une forte corrélation de cette commutation avec la couche des segments externes des cônes. Nous avons proposé une procédure d'intégration de la variabilité spatiale pour la construction des cartographies. Cette procédure a été évaluée sur des sujets sains et pathologiques et a montré un gain significatif (en termes de dénombrement des cônes) chez les cas pathologiques
Our objective was to analyze the importance of spatial and temporal variability of the reflectance of cones in counting for adaptive optics high resolution imaging, and propose ways to overcome these difficulties.The first part of the work focuses on the study of the temporal variability of cones. We confirmed the presence of a scintillation phenomenon at a frequency at least equal to that of our acquisition system. We have proposed an image analysis process including restoration by deconvolution, rigid registration, and image fusion for cone mapping. To validate this approach, we conducted a validation test for two deconvolution methods. We have shown that in the case of healthy subjects, the image fusion procedure to bring the measurement of cone density and histological references. The angular variation of cone reflectance is called optical Stiles-Crawford effect. We have identified a new clinical manifestation of this phenomenon, through a study on the analysis of adaptive optics images periphery). We have shown, through the alternation of the positive / negative mosaics, that the reflectance of the cones may become lower than that of rods. Comparison with the data of the optical coherence tomography showed a strong correlation of the switching with the outer segment layer. We then proposed a procedure of integration of spatial variability for cone mapping. This procedure was evaluated in healthy and diseased subjects and showed a significant gain (in terms of cone enumeration) in pathological cases

Abstract:

The recent and rapid growth of horizontal drilling in the Anadarko basin necessitates newer studies to characterize reservoir and source rock quality in the region. Most oil production in the basin comes from the Granite Wash reservoirs, which are composed of stacked tight sandstones and conglomerates that range from Virgillian (305–299 Ma) to Atokan (311–309.4 Ma) in age. By utilizing geophysical well logging data available in raster format, the Granite Wash reservoirs and their respective marine flooding surfaces were stratigraphically mapped across the regional fault systems. Additionally, well log trends were calibrated with coincident core data to minimize uncertainty regarding facies variability and lateral continuity of these intervals. In this thesis, inferred lithofacies were grouped into medium submarine fan lobe, distal fan lobe, and offshore facies (the interpreted depositional environments). By creating isopach and net sand maps in Petra, faulting in the Missourian was determined to have occurred syndepositionally at the fifth order scale of stratigraphic hierarchy.

A estrongiloidíase é uma doença parasitária que tem distribuição mundial heterogênea, cuja prevalência é dividida em três categorias: esporádica (<1%), endêmica (1-5%) e hiperendêmica (>5%). Na região de Araraquara, estudos de frequência de enteroparasitoses vêm sendo conduzidos desde 1970 e mostram que a estrongiloidíase é endêmica na região. Com o intuito de realizar caracterização morfométrica de diferentes isolados humanos desse helminto, provenientes dos municípios de Araraquara, Américo Brasiliense, Gavião Peixoto, Motuca e Rincão foi feita a mensuração de larvas rabditóides e filarióides e sequenciamento da região ITS-1 do rDNA das larvas filarióides. Com o auxilio do programa de captura e mensuração de imagens Motic Images Advanced 3.2., os parâmetros das larvas rabditóides e filarióides propostos por TEIXEIRA, 1996, foram mensurados após fixação das mesmas em TAF. A extração do DNA genômico foi realizada segundo protocolo descrito por NILFOROUSHAN et al., 2007 e em seguida realizou-se a amplificação do ITS-1 por meio de NESTED-PCR. Os fragmentos de aproximadamente 680 pb obtidos foram purificados, sequenciados e a comparação de sequências foi realizada pelo programa ClustalW. Strongyloidiasis is a parasitic disease that occurs worldwide heterogeneous, whose prevalence is divided into three categories: sporadic (<1%), endemic (1-5%) and hyperendemic (> 5%). In the region of Araraquara, studies the frequency of intestinal parasites have been conducted since 1970 and show that strongyloidiasis is endemic in the region. In order to perform morphometric characterization of different isolates of human intestinal worms, from the municipalities of Araraquara, Américo Brasiliense, Gavião Peixoto, Motuca and Rincão the measurement was made of rhabditiforms and filariforms larvae and sequencing of the ITS-1 rDNA filariform larvae. With the help of the program capture and measurement of images Motic Images Advanced 3.2., the rhabditiforms and filariforms larvae parameters proposed by TEIXEIRA, 1997, were measured after fixed on the TAF. The extraction of genomic DNA was performed according to protocol described by NILFOROUSHAN et al., 2007 and then held the amplification of ITS-1 by nested-PCR. Fragments of about 680 bp obtained were purified, sequenced and the sequence comparison was performed by the program ClustalW. Only the results of the morphometric analysis was not possible to characterize different S. stercoralis strains, since the values of the parameters measured were quite homogeneous in larvae... Este trabalho foi desenvolvido com o objetivo de se avaliar a eficácia do ELISA ProSpecT Giardia (Alexon, Inc. BioBrás) anti-GSA65 (antígeno fecal específico de Giardia - peso molecular 65.000 Da) como instrumento de controle de cura da giardíase e o seu limiar de positividade. A contagem de cistos por grama de fezes foi feita utilizando a câmara de Neubauer em amostras positivas para G.lamblia de 100 pacientes. Os resultados mostraram eliminação de 3,6 x 106 cistos por grama de fezes em média. Em gráfico esses resultados desenharam uma curva assimétrica positiva, sugerindo a ocorrência de falso-negativos. Avaliou-se também a eficácia do ELISA anti-GSA65 e o método de Faust e Cols (MFc) em detectar mínimas concentrações de cistos de G. lamblia em amostras fecais. Fezes positivas foram diluídas em fezes negativas de modo a se obter amostras com 100, 1.000 e 10.000 cistos por grama de fezes. O ELISA anti-GSA65 mostrou uma positividade significativamente maior que o MFc. Amostras contendo 10.000 cistos por grama de fezes apresentaram 84,7% de positividade pelo ELISA anti-GSA65 e 34,7% pelo MFc. Due to the low sensitivity of the fecal examination (EPF) in detecting cysts of Giardia lamblia, the imunoenzimatic assay (ELISA enzyme linked immunosorbent assay) to detect G.lamblia coproantigens have been used and have showed high sensibility and specificity. With the ain of evaluating the efficacy of ELISA ProSpecT Giardia (Alexon, Inc. BioBrás) anti-GSA65 (Giardia stool specific antigens - molecular weigh 65.000 Da) as monitoring of the cure of giardiasis and its threshold of positivity it was developed this research. The cysts count per grama of faeces was calculated using Neubauer camera in positive fecal samples for Giardia in 100 patients. It was found a concentration of 3.6 X 106 cyst per grama of faeces as average. In a graph the data draw an assymmetrical positive curve, suggesting occurrence of false negative results. It was evaluated the efficacy of ELISA anti-GSA65, and fecal examination by Faust and colls. procedure (MFc), in detecting minimum concentrations of Giardia cysts in fecal samples. To obtain samples with 100, 1.000 and 10.000 cysts per grama, positive samples were diluted in negative samples. ELISA showed a positivity significative higher than MFc. Samples with 10.000 cystis per grama showed 84,7% of positivity by ELISA anti GSA65 and 34,7% by MFc. The efficacy of ELISA anti-GSA65 as cure monitoring after treatment with metronidazole in 91 patients with giardiasis, was evalued and showed that ELISA anti-GSA65 can be used to monitor the cure of patients, using only single sample of each patients, collected between the 4th and 7th days after treatment, whereas with MFc it would be necessary the exam of two or tree samples. It was also evaluated the efficacy of ELISA-anti-GSA65 and MFc in fecal samples of 96 patients. BioBrás) anti-GSA65 (Giardia stool specific antigens - molecular weigh 65.000 Da) as monitoring of the cure of giardiasis and its threshold of positivity it was developed this research. The cysts count per grama of faeces was calculated using Neubauer camera in positive fecal samples for Giardia in 100 patients. It was found a concentration of 3.6 X 106 cyst per grama of faeces as average. In a graph the data draw an assymmetrical positive curve, suggesting occurrence of false negative results. It was evaluated the efficacy of ELISA anti-GSA65, and fecal examination by Faust and colls. procedure (MFc), in detecting minimum concentrations of Giardia cysts in fecal samples. To obtain samples with 100, 1.000 and 10.000 cysts per grama, positive samples were diluted in negative samples. ELISA showed a positivity significative higher than MFc. Samples with 10.000 cystis per grama showed 84,7% of positivity by ELISA anti GSA65 and 34,7% by MFc. The efficacy of ELISA anti-GSA65 as cure monitoring after treatment with metronidazole in 91 patients with giardiasis, was evalued and showed that ELISA anti-GSA65 can be used to monitor the cure of patients, using only single sample of each patients, collected between the 4th and 7th days after treatment, whereas with MFc it would be necessary the exam of two or tree samples. It was also evaluated the efficacy of ELISA-anti-GSA65 and MFc in fecal samples of 96 patients. Stiles and Crawford (1933) showed sixty years ago that the sensitivity of the retina varies with the place of entry of the light in the pupil, and thus with the direction of incidence of light upon the retina. Sensitivity is generally greatest near the centre of the pupil and progressively decreases for more eccentric entry points. This phenomenon is known in the literature as the Stiles-Crawford effect of the first kind (SCE I). The hue shift associated with the SC-effect was discovered by both investigators (Stiles 1937) in a further study of the retinal direction effect. This effect is called the chromatic Stiles-Crawford effect, often also referred to as the Stiles-Crawford effect of the second kind (SCE II).

The Stiles-Crawford function (Stiles, 1937) is a psychophysical measure of photo receptor orientation and directional sensitivity. It is found that the Stiles-Crawford functions are affected in various pathologies. Birch and his group (Birch, Sandberg, and Berson, 1982) found in patients with different genetic types of retinitis pigmentosa (RP; visual acuity of 20/40 or better, no visible opacities in the media) that parafoveal Stiles-Crawford Effect of the first kind (SCE I) functions were either significantly flattened or have displaced peak positions. Morphological changes have been found in photoreceptors of RP patients (post-mortem). These morphological changes could lead to an increase in the numerical aperture of the cones (acceptance angle hypothesis) and result in reduced directional sensitivity. Alternatively, patients with RP may show increased variation in the alignment of photoreceptors (with normal acceptance angles) due to fractional forces on the retina, loss of supporting structures, and/or alterational forces maintaining alignment (splaying hypothesis). If the peak of the SCE I remains near center in the pupillary aperture, one assumes that photoreceptor alignment mechanism(s), perse, are still functional even if affected. Of course, both options, changes in optical properties, and splaying can occur concurrently.
