Abstract:
The aim of this thesis is to analyse a business model that is defined in two ways: company client and company company. The main point is ensuring of vehicle and emission testing of cars. In the first case, car owners are the clients, and in the second case, car repair shops are the clients. The theoretical part includes definitions of terms and descriptions of individual analyses and serves as a basis for the practical part where we can find internal and external analyses as well as descriptions of both forms of the business model. The outcomes of the analyses are then used to create a SWOT matrix. The conclusion of the thesis contains an evaluation of the data and results of all analyses, which can present a useful tool when deciding about any further development of the business idea.

Abstract:
The GPS data from Challenging Mini-satellite Payload (CHAMP) is used for its orbit determination for the epoch day of January 1st 2002.  The orbit of CHAMP is computed from the GPS data and ionospheric effects are removed by frequency combination. Further, the orbits of CHAMP for the same epoch day are computed using the satellite tool kit (STK) employing simplified general perturbations (SGP4) and a high precision orbit propagator (HPOP). Results from both techniques (GPS computed orbit and STK computed orbit) are compared. Furthermore, orbits computed using GPS data are also compared with jet propulsion laboratory’s published CHAMP spacecraft orbit and we have found that root mean square difference in ECEF position X component is below 0.01km other than some spikes at poles. The standard deviation of the difference in ECEF position X coordinate is 11.7m. The accuracy of our computed satellite positions (using GPS data) is about 12 metres for other than polar areas. However there are some occasional spikes, especially at poles, having maximum errors (about 0.055 km).

Abstract:
The StkF protein of Salmonella enterica serovar Paratyphi A, a putative adhesion-associated protein, was used to develop a candidate adhesin-based vaccine against Salmonella infections, particularly S. Paratyphi A-associated enteric fever. Subcutaneous immunisation of BALB/c mice with recombinant StkF (rStkF) showed that this protein was strongly immunogenic as revealed by the presence of high-titre (1:50,000) anti-StkF antibody in sera from immunized mice. Furthermore, as IgG1 was the major antibody isotype induced, it would appear that rStkF generates a strong Th2-type humoral immune response. The induction of a Th2-dominant immune response was further confirmed by splenocyte-associated cytokine profile analysis which demonstrated greater up-regulation of IL-4 than that of IFN-γ or IL-12-p70. In terms of the protective potential of this antigen, in vitro assays demonstrated that StkF-specific murine antiserum markedly enhanced opsonophagocytosis-mediated uptake of StkF-expressing Salmonella bacteria by human neutrophils. Augmented antibody-specific complement-mediated lysis targeting StkF-expressing Salmonella specifically was also shown. These data suggest a likely direct contribution of StkF-specific antibodies to in vivo killing and clearance of Salmonella and strongly support further investigation of StkF as a potential Salmonella vaccine candidate. BlastN-based interrogation of the NCBI bacterial genome database and PCR investigation of a larger set of strains has shown that the S. Paratyphi A stkF gene and/or the whole stk fimbrial gene cluster is carried by ~1/3 of examined Salmonella serovars. Additionally, bioinformatics and phenotypic characterisation has revealed that the stk fimbrial operon belongs to the chaperone/usher-γ4-fimbrial clade and that it encodes a mannose-sensitive haemagglutinating fimbrial structure. The latter trait is typical of type 1 fimbriae. ELISAs based on rStkF, rStaF and rSipA showed variable sensitivity and specificity for the serodiagnosis of invasive Salmonella infections depending on the particular UK or region-specific cut off applied for each antigen. Further refinement and/or merging of these assays may allow for development of simple and cost effective tests with higher sensitivity and/or specificity than the Widal test. Importantly, despite some minor reactivity with serum from patients with other Gram-negative bacterial infections, the rStkF-based ELISA exhibited no reactivity with serum from patients with dengue fever supporting its potential as a discriminatory diagnostic tool between fevers caused by S. Paratyphi A and dengue virus.

Thesis (MTech (Environmental Health))--Cape Peninsula University of Technology, 2016.<br>Environmental remediation of organic and inorganic contaminants such as hydrocarbons has been a research focus area of interest. Chemical surfactants have been extensively used for the remediation of contaminated sites for immobilisation of hydrocarbons from environmental matrices. The focus has been on the impact of chemical surfactants on the environment. These petroleum-based chemical surfactants have raised serious environmental concerns as: 1) they are toxic, 2) they deteriorate the environment owing to their non-biodegradability, 3) they are costly, and 4) most are not intended for environmental applications. As such, alternatives had to be found to mitigate concerns associated with the application of such synthetic surfactants in bioremediation. Biosurfactants produced by microorganisms are a potential alternative to these synthetic surfactants. They have minimal environmental impact, are biodegradable and can withstand extreme conditions. However, biosurfactants are associated with high production costs and low production yield. Currently, large-scale production of biosurfactants cannot be achieved. Most research focuses on improving production yield which will contribute to the reduction in production costs. A lichenysin lipopeptide biosurfactant producing Bacillus sp., which grew exclusively on Beta vulgaris agrowaste, was identified. The microorganism was found to be an effective emulsifier for high molecular weight hydrocarbons such as, lubricant oil and diesel. The aim of this study was to improve biosurfactant production yield from this Bacillus sp., including emulsification efficacy by optimising fermentation conditions by supplementing the broth with biocompatible nanoparticles synthesised using a green chemistry approach with B. vulgaris (B. vulgaris) extracts. This study also aimed at reducing production costs by using B. vulgaris agrowaste exclusively as the production medium, both for the biosurfactant and the nanoparticles.

Abstract:
Myxococcus xanthus, a Gram-negative bacterium with a developmental cycle, displays a type IV pili (TFP) mediated surface motility known as social (S) gliding. Beside the polarly localized TFP, the fibril or extracellular polysaccharide (EPS) is also required for S-motility to function. It is proposed that S-motility, along with the related bacterial twitching motility in other species, is powered by TFP retraction. EPS is proposed to anchor and trigger such retractions in M. xanthus. EPS production is known to be regulated by TFP and the Dif signal transduction pathway. Two genetic screens were performed previously to identify additional genes important for EPS production. The first was for the isolation of pilA suppressors, the second for the identification of mutants underproducing EPS in a difA suppressor background. Both screens identified transposon insertions at the stk locus. In particular, StkA, a DnaK homolog, was identified as a possible negative regulator of EPS production by a stkA transposon insertion that suppressed a pilA mutation. A stkB transposon insertion was found to have diminished EPS production in a difA suppressor background. In this study, in-frame deletion mutants of the five genes at the stk locus, stkY, stkZ, stkA, stkB and stkC, were constructed and examined. In addition, mutations of rbp and bskL, two genes downstream of the stk locus, were constructed. Like transposon insertions, the stkA in-frame deletion resulted in overproduction of EPS. The stkB and to a less extent the stkC mutants underproduced EPS. Mutations in the other genes had no obvious effects on EPS production. Genetic epistasis suggests that StkA functions downstream of TFP and upstream of the Dif sensory proteins in EPS regulation in M. xanthus. Epistasis analysis involving stkB was inconclusive. It is unresolved whether StkB plays a role in the biosynthesis or the regulation of EPS production in M. xanthus.<br>Master of Science

We are in an era of massive spending cuts in educational institutions, aerospace companies and governmental entities. Educational institutions are pursuing more training for less money, aerospace companies are reducing the cost of gaining ight heritage and the government is cutting budgets and their response times. Organizations are accomplishing this improved efficiency by moving away from large-scale satellite projects and developing pico and nanosatellites following the CubeSat specifications. One of the major challenges of developing satellites to the standard CubeSat mission requirements is meeting the exceedingly tight power, data and communication constraints. A MATLAB toolbox was created to assist the CubeSat community with understanding these restrictions, optimizing their systems, increasing mission success and decreasing the time building to these initial requirements. The Toolbox incorporated the lessons learned from the past nine years of CubeSats' successes and Analytical Graphics, Inc. (AGI)'s Satellite Tool Kit (STK). The CubeSat Mission Planning Toolbox (CMPT) provides graphical representations of the important requirements a systems engineer needs to plan their mission. This includes requirements for data storage, ground station facilities, orbital parameters, and power. CMPT also allows for a comparison of broadcast (BC) downlinking to Ground Station Initiated (GSI) downlinking for payload data using federated ground station networks. Ce travail porte sur l’étude des mécanismes de phosphorylation des protéines par les sérine/thréonine kinases chez Staphylococcus aureus. Nous avons, tout d’abord, mis en évidence et caractérisé une seconde Ser/Thr-kinase, nommée Stk2. Cette kinase présente peu d’homologies avec les autres Ser/Thr-kinases bactériennes décrites à ce jour, en particulier avec la première Ser/Thr-kinase mise en évidence précédemment chez S. aureus, Stk1. Nous avons ensuite caractérisé dix sites d’autophosphorylation de Stk2 et nous avons montré que trois sites sont nécessaires à son activité. Enfin, nous avons montré que le régulateur global de virulence, SarA, est phosphorylé à la fois par Stk1 et Stk2. La phosphorylation de SarA influence sa capacité de liaison à l’ADN. Cette étude contribue à mieux appréhender, au niveau moléculaire, le rôle des Ser/Thr-kinases dans le métabolisme des bactéries et, plus particulièrement, dans la régulation de leur virulence<br>We report that protein phosphorylation on serine and threonine is required for controlling staphylococcal virulence. We identified and characterized a second serine/threonine kinase, Stk2, in S. aureus. Biochemical analyses revealed that this enzyme displays autokinase activity on both threonine and serine residues. Stk2 is atypical in the sense that it exhibits a weak similarity with the first Ser/Thr-kinase previously detected, Stk1, and its undergoes a different mechanism of activation compared to the other bacterial Ser/Thr-kinases described so far. We also showed that SarA, a major transcription factor that regulates more than a hundred virulence genes, is phosphorylated by both Stk1 and Stk2. Phosphorylation of SarA leads to strong effects on its ability to bind DNA. The study of Stk1 and Stk2, at the molecular level, provides a better understanding of the role of these staphylococcal Ser/Thr-kinases in bacterial metabolism and, in particular, in the regulation of virulence

No atual estágio evolutivo, representado por um processo de aproximação dos sistemas e valoração dos direitos fundamentais; impôs-se uma adequação de institutos tradicionalmente formais, a partir de princípios voltados à justiça; não se admitindo mais interpretações restritivas de dispositivos jurídicos a limitar suas aplicações práticas. O desenvolvimento da internet propiciou a formação de redes de dados mais amplas e acessíveis. Nesse contexto, é comum a utilização de dados pertencentes ao passado do indivíduo, o qual muitas vezes já está, por ele, superado. O assunto do presente trabalho abarca instituto apto a proteger a pessoa humana e o seu livre desenvolvimento na sociedade atual, qual seja, o direito ao esquecimento. A questão cerne é se essa rede de dados pode armazenar informações em um espaço de tempo indefinido e, também, se esses dados podem ser republicados a qualquer tempo. O direito ao esquecimento visa coibir a veiculação de informações pretéritas, que digam respeito a questões privadas da pessoa, a partir de alguns critérios. Este trabalho investigou como o direito ao esquecimento vem sendo disciplinado no Brasil. Analisou-se o conceito em tela de acordo com os fundamentos jurídicos brasileiros contemporâneos, conceituando-o com base na doutrina e jurisprudência pátrias. Foram analisados os critérios normalmente utilizados para propiciar sua aplicação, bem como sugeridos outros que sejam aptos a justificar a sua aplicação nos casos concretos. O estabelecimento de critérios se mostra de extrema relevância para permitir a aplicação justa e equilibrada desse direito, sem prejudicar a liberdade de expressão ou a memória nacional.<br>Nowadays individuals are embedded in the communication society, which is characterized by the enormous flow of information made available and transmitted worldwide, without temporal or territorial barriers. The development of the Internet has fostered the formation of broader and more accessible data networks. In this context, the use of old data from an individual’s past is not unusual, a past which is often overcome. The aim of this paper encompasses an institute capable of protecting the human person and their free development in today's society, namely, the right to be forgotten. The question to be posed is whether this data network can store information over an indefinite period of time and whether that data can be republished at any time. This right is aimed at preventing the disclosure of past information that concerns a person's private affairs, in the presence of certain criteria. The purpose of this work is to investigate how the right to be forgotten has been regulated in Brazil. Therefore, it will be analyzed according to the present Brazilian legal grounds for its protection. Then it will be conceptualized based on the country’s doctrine and jurisprudence. The commonly used criteria to provide its enforcement will also be analyzed, as well as others that may be used to justify its enforcement in specific cases. Setting criteria is extremely relevant to allow a fair and balanced enforcement of this right, without harm to freedom of expression or national memory.

International Telemetering Conference Proceedings / October 27-30, 1997 / Riviera Hotel and Convention Center, Las Vegas, Nevada<br>This paper details the interaction of a small satellite with a space network and estimates the number of contacts and duration of contacts between the small, spin-stabilized satellite and a TDRS satellite. The simulations were performed using Satellite Tool Kit (STK) version 3.0, an orbital analysis software program. STK was configured for the four vehicles representing the spin-stabilized satellite and three TDRS satellites, TDRS East, West, and TDRS Zone of Exclusion. A set of simulations were run in which the spinstabilized satellite was given orbital elements corresponding to an orbital altitude between 600 km and 1200 km. The orbital inclination angle for the set of simulations was also varied from 20° through 100° along with the antenna cone angle of 10° through 40° to account for the effective beamwidths. In each of the simulations, the access to each TDRS satellite in the SN constellation was examined as a function of orbital altitude, orbital inclination angle, and antenna cone angle.

Abstract:
RESUMO - Objetivos: Estudos anteriores demonstraram a existência de uma relação inversa entre melhores outcomes e volume hospitalar. Tendo este fenómeno em consideração, o objetivo deste estudo foi analisar esta relação a um nível populacional em Portugal. Métodos: Análise da base de dados Portuguesa dos GDH relativos ao ano financeiro de 2009. Todas as STA e STJ foram contabilizadas e critérios de exclusão determinados, para selecionar duas amostras homogéneas, analisadas por volume hospitalar. O volume hospitalar foi definido em três grupos: HVB, HVM, HVE de acordo com o volume anual de procedimentos efetuado por cada hospital individual durante o período do estudo. Os outcomes avaliados incluíram a mortalidade, presença de pelo menos uma complicação cirúrgica, estadia prolongada assim como a média de dias de internamento. A idade e sexo foram controlados como possíveis fatores de confundimento. Resultados: A investigação principal consistiu na análise de 4615 STA e 5904 STJ. Os resultados indicam que os eventos adversos (mortalidade e complicações cirúrgicas) são pouco comuns nestes procedimentos, produzindo resultados sem significância estatística. A estadia prolongada estabeleceu uma clara e significativa relação inversa com o volume hospitalar. A estadia prolongada foi superior em hospitais de volume baixo (STA OR 2.71; STJ OR 2.17) e hospitais de volume médio (STA OR 1.72; STJ OR 1.73) quando comparados com hospitais de volume elevado. Os dias de internamento médios produziram uma associação semelhante, sendo possível estimar quase oito milhões e meio de euros de custos evitáveis no ano de 2009, em ambos os procedimentos. Conclusões: O volume hospitalar não apresentou relações significativas com a mortalidade e complicações cirúrgicas, devido ao baixo número de eventos adversos registados. Verificou-se, no entanto, uma associação significativa com os dias de internamento e estadia prolongada. As STA e STJ realizadas em hospitais de volume elevado poderão reduzir custos, ao diminuir os dias de internamento.<br>ABSTRACT - Objective: Prior work from other authors has shown a significant inverse relationship between improved outcomes and hospital volume. Bearing this in mind, the present study had the objective of examining this relation following THR and TKR at a population level in Portugal. Methods: Analysis of the Portuguese DRG database in the 2009 financial year. All THR and TKR were accounted for, and criteria used to select two homogeneous samples according to hospital volume. Hospital Volume was defined as three groups LVH, MVH and HVH according to the annual volume performed by each hospital during the study period. Assessed outcomes were mortality, presence of at least one complication, as well as average length of stay and prolonged length of stay. Age and sex were controlled for as potential confounders. Results: Main analyses consisted of data examined from 4615 THR and 5904 TKR. Results show that adverse events (mortality and complications) were uncommon, producing no significant association with hospital volume. Prolonged length of stay established a clear and significant association with hospital volume, greater in low volume hospitals (THR OR 2.71; TKR OR 2.17) and medium volume hospitals (THR OR 1.72; TKR OR 1.73) when compared to high volume hospitals. Average length of stay produced similar results and we were able to estimate almost eight and half million Euros in avoidable costs during 2009. Conclusions: Hospital volume showed no significant associations with mortality and complications, due to low number of adverse events, but significant relationship with prolonged and average length of stay. THR and TKR performed in high volume hospitals may reduce costs by shortening the length of stay.

Lo studio inizia con l\'analisi dello sviluppo dell\'istituto fin dalla sua origine, attraverso la sua costituzionalizzazione alla sua previsione nel progettodi nuovo Codice di Procedura Civile, che generalizza la sua idoneità come un mezzo per controllare l\'efficacia vincolante di quanto sopra esposto.Poi ci spostiamo l\'analisi del sistema brasiliano per quanto sopra, con un breve confronto con i paesi di common law, e ha concluso che, in Brasile, l\'effetto vincolante, che essendo una forza che impone il rispetto di standard di estrazione delle decisioni giudiziarie esiste solo se è previsto espressamente dalla Costituzione o dalla legge. O estudo se inicia com a análise do desenvolvimento do instituto desde sua origem correicional, passando por sua constitucionalização até sua previsão no novo Código de Processo Civil, que generaliza seu cabimento como meio de controle da eficácia vinculante dos precedentes. Em seguida, passamos à análise do sistema brasileiro de respeito aos precedentes, fazendo uma breve comparação com países do common law, e concluímos que, no Brasil, o efeito vinculante, assim entendida a força que torna obrigatória a observância da norma extraível das decisões judiciais, só existe se houver previsão expressa na Constituição ou na lei. Também constatamos que o sistema adotou a reclamação a ser ajuizada diretamente perante o STF e o STJ como instrumento processual de controle da observância dessa força vinculante. Verificamos que, além de valorizar a segurança jurídica, a isonomia e a justiça das decisões, a adoção de um sistema de respeito a precedentes no Brasil tem como confessado objetivo a otimização do serviço judiciário e a redução dos processos pendentes nos tribunais superiores. Todavia, a utilização da reclamação como meio de controle da eficácia vinculante dos precedentes vai de encontro àqueles objetivos, pois reatomiza os litígios sem que isso signifique maior respeito à obrigatoriedade dos precedentes.

Vývoj tohoto rozhraní je proveden pomocí GEMstudia, softwaru firmy Amulet Technologies a grafických programů. Dále tato studie pojednává o softwarové úpravě FTZ AS-i Slave Smilulátoru. Jedná se o úpravu řídícího FPGA v jazyce VHDL zajišťující komunikaci s dotykovým displejem. Poslední kapitola se týká problematiky spojené s návrhem uživatelsky přívětivé aplikace.

Com este intuito, primeiramente, no Capítulo II, serão expostas as vulnerabilidades socioeconômicas extramuros enfrentadas pelas mulheres (grávidas) em privação de liberdade no Brasil, as quais são agravadas pelas pressões emocionais, físicas e materiais decorrentes das dificuldades por elas vivenciadas no ambiente prisional. STF is a large ensemble work in three movements: "Fatwa," "Hitchens," and "Albino Sex." It is scored for piano, double bass, cello, viola, violin, French horn, clarinet in Bb, bass clarinet in Bb, flute, baritone, tenor, alto, soprano, narrator, and electronics. Its underlying concept is how religious worldviews can collide with freedom of speech, atheism, and fundamental human rights. In extreme cases, zealots will interpret disease as evidence of divine intervention or even demand the death of blasphemers. The text in the first movement, “Fatwa,” expresses freedom of speech and incorporates passages from Salman Rushdie’s “The Satanic Verses.” The second movement, “Hitchens,” centers on atheism and incorporates two audio excerpts from Christopher Hitchens’s lecture at the 2009 Festival of Dangerous Ideas. In the final movement, “Albino Sex,” I construct a narrative using media headlines, paraphrased biblical verses, and my own text to depict the human rights crisis during the 1980s A.I.D.S. epidemic. Various organizational approaches to text and pitch distinguish one movement from the other; the three movements cohere through unifying devices that I draw from set theory.

Esta pesquisa elege o materialismo histórico-dialético como método de análise de ques-tões contemporâneas da homossexualidade e sua relação com o Judiciário brasileiro. Articula paradigmas marxianos, marxistas e feministas, por meio da interdisciplinaridade do direito, da sociologia, da antropologia, da filosofia e das ciências da linguagem, para compreender o fenômeno LGBT (lésbicas, gays, bissexuais, transexuais e transgêneros) que mobiliza indivíduos na reivindicação por promoção, efetivação e proteção de direitos humanos. Recorre aos conceitos de fetichismo da mercadoria, fetichismo jurídi-co, forma jurídica, contrato e gênero, na perspectiva da História, para compreender e promover a desnaturalização de processos sociais e situá-los na base econômica da so-ciedade em face da luta de classes. Posiciona o direito na superestrutura, onde localiza também o direito civil, o direito de família e os direitos humanos, para desnudar seus processos discursivos ideológicos e, logo, práticos. Os sistemas analisados contemplam os up-grades das taxas de 2,48 Gbps para 10 Gbps e também da taxa de 10 Gbps para 40 Gbps. Para tal, foram introduzidos módulos de transmissão e recepção, capazes de utilizar arquiteturas quase totalmente ópticas. É avaliado então, através de simulações, o comportamento da arquitetura proposta em infra-estruturas de enlaces já instalados no Brasil, destacando os pontos mais críticos. No que se refere ao up-grade de 10 Gbps para 40 Gbps, foi dado enfoque especial para as penalidades relativas à PMD (Polarization Mode Dispersion).<br>This work aims at up grading the bit rate of optical links through the OTDM technology. The analyzed up-grades change the bit rate of 2,48 Gbps up to 10 Gbps and also from the bit rate of 10 Gbps up to 40 Gbps. To reach these objectives, transmission and reception modules were introduced, using all optical networks topologies. The performance of the proposed architecture was simulated using a infrastructure of links already installed in Brazil. The most critical issues were pointed out. Concerning the up-grade from 10 Gbps to 40 Gbps, a special focus was given to the penalties due to PMD (Polarization Mode Dispersion).

Abstract:
Salmonella enterica serovar Typhimurium genome encodes 13 fimbrial operons. Most of the fimbriae encoded by these operons are not produced under laboratory conditions but are likely to be synthesized in vivo. We used an in vivo expression technology (IVET) strategy to identify four fimbrial operons, agf, saf, sti, and stc that are expressed in the spleen. When any three of these operons were deleted, the strain retained wild-type virulence. However, when all four operons were deleted, the resulting strain was completely attenuated, indicating that these four fimbriae play functionally redundant roles critical for virulence. In mice, oral doses of as low as 1 x 10(5) CFU of the strain with four fimbrial operons deleted provided 100% protection against challenge with 1 x 10(9) CFU of wild-type S. Typhimurium. We also examined the possible effect of these fimbriae on the ability of a Salmonella vaccine strain to deliver a guest antigen. We modified one of our established attenuated vaccine strains, chi 9088, to delete three fimbrial operons while the fourth operon was constitutively expressed. Each derivative was modified to express the Streptococcus pneumoniae antigen PspA. Strains that constitutively expressed saf or stc elicited a strong Th1 response with significantly greater levels of anti-PspA serum IgG and greater protective efficacy than strains carrying saf or stc deletions. The isogenic strain in which all four operons were deleted generated the lowest anti-PspA levels and did not protect against challenge with virulent S. pneumoniae. Our results indicate that these fimbriae play important roles, as yet not understood, in Salmonella virulence and immunogenicity. IMPORTANCE Salmonella enterica is the leading cause of bacterial food-borne infection in the United States. S. Typhimurium is capable of producing up to 13 distinct surface structures called fimbriae that presumably mediate its adherence to surfaces. The roles of most of these fimbriae in disease are unknown. Identifying fimbriae produced during infection will provide important insights into how these bacterial structures contribute to disease and potentially induce protective immunity to Salmonella infection. We identified four fimbriae that are produced during infection. Deletion of all four of these fimbriae results in a significant reduction in virulence. We explored ways in which the expression of these fimbriae may be exploited for use in recombinant Salmonella vaccine strains and found that production of Saf and Stc fimbriae are important for generating a strong immune response against a vectored antigen. This work provides new insight into the role of fimbriae in disease and their potential for improving the efficacy of Salmonella-based vaccines.

Data insamlade med hjälp av högstrukturerade telefonintervjuer. En enkät med standardiserade frågor och ikryssbara svarsalternativ som kommer har använts vid telefonintervjuerna.Resultaten från de 52 intervjuerna visade att de flesta patienter inte överskred Numerical Rating Scale (NRS) 3 i vila eller rörelse dag ett. På dag tre hade de flesta mer smärta vid rörelse än dag ett. De flesta patienterna följde ordinationen de fick av läkaren avseende smärtstillande tabletter. Generellt sett visade studien på få postoperativa komplikationer och hög tillfredsställelse ur operationssynpunkt hos patienterna.<br>The purpose of this study was to evaluate the post-operative progress for one-day surgical inguinal hernia patients. Data was collected using structured telephone interviews three days after surgery. A questionnaire with standardized questions and multiple-choice response options was used.The results from the 52 interviews showed that most patients did not exceed Numerical Rating Scale (NRS) 3 day one when at rest or moving around. On day three most patients had more pain while moving than day 1. Most patients complied with the doctor’s prescription of painkillers. Generally the follow-up study showed the patients had few complications and felt high satisfaction post surgery.

<p>Under hösten 1997 och våren/hösten1998 har ett pilotprojekt bedrivits i Linköpings kommun. Det är drivet av Kungliga Vetenskapsakademien och Kungliga Ingenjörsakademien och handlar om ett amerikanskt undervisningsmaterial inom naturvetenskap och teknik. På engelska heter det"Science and Technology for Children"vilket givit namnet STC. Det är ett temaarbete med 24 olika teman att använda under de 6 första skolåren.</p><p>För att använda materialet i skolan har vissa utvalda delar översatts till svenska. Det är endast en direktöversättning och inget speciellt anpassat till den svenska skolan. Detta arbete har jag gjort för att undersöka hur det går att använda STC materialet Motion and Design enligt våra svenska styrdokument. Jag har även tittat på varför lärare fastnar för det.</p><p>Resultatet visar att det fungerar relativt bra utifrån de förutsättningar materialet hade när det provades. I detta projekt löses konflikten mellan program och plats meddels en långsmal byggnad som bevarar naturvärden och rörelsemönster.<br>Galarvarvet is in many ways an great site for a new and prominent cultural building in Stockholm. At the same time the complexity and size of the program of an opera poses a great challenge. In this project the conflict between program and site is solved with a stretched out building that preserves environmental values and movements.

From the idea of justice through the process, we seek to understand the basic conceptual resources, talks are on the requests for clarification about the features and extraordinary and special, having prequestionamento as a conducting wire that is exposed. After some brief remarks on the origin and role of each of the aforementioned features and the doctrinal and jurisprudential understanding about prequestionamento, seeks to analyze intelligence about the requests for clarification as elements able to substantiate this requirement, mainly the position espoused by the S T F and the S T J because of the lack of judgment a quo to examine the point raised by the objector. Seen these points, the next step was to verify the discipline of requests for clarification in the draft text of the new Code of Civil Procedure, approved by the Senate of the Republic and through the House of Representatives. At this point, what matters is to see what the new code brings to light, especially in regard to Article 979, which is called the positive findings prequestionamento ficto and the spirit that pervades the Explanatory Memorandum of the new CPC, taking into account its main objectives. Afterwards, we sought to investigate the relationship between Article 979 and the two most important methodological conceptions of civil procedure in contemporary Brazil: instrumentalism and formalism-evaluative. After that, the objective was to verify the ontological, deontological, axiological, principled and constitutional dimensions of prequestionamento ficto, taking into account their positivization on the new CPC, article 979, and the implications of this positivization. Finally, one comes to the conclusion that no further provenance 211 from the S T J 211 Precedent, praising the initiative to the legislature put an end to a disagreement between the STF and STJ which had lasted for years and seemingly endless, thus ensuring, knowledge of special and extraordinary appeal, therefore, justice, legal certainty and effectiveness of the process<br>Esta dissertação trata dos embargos de declaração e da sua utilização, no direito brasileiro, como forma de fazer aflorar o requisito de prequestionamento das matérias constitucional ou infraconstitucional inerente, segundo entendimento jurisprudencial (acórdãos e Súmulas 282 e 356 do STF e 211 do STJ) e doutrinário, à interposição de recurso extraordinário ao STF ou de recurso especial ao STJ. A partir da ideia de justiça por meio do processo, busca-se a compreensão conceitual básica dos recursos, discorre-se sobre os embargos de declaração e sobre os recursos extraordinário e especial, tendo o prequestionamento como fio condutor do que se expõe. A seguir, buscou-se verificar a relação entre o artigo 979 e as duas mais importantes concepções metodológicas de processo civil no Brasil contemporâneo: o instrumentalismo e o formalismo-valorativo. Depois disso, o objetivo foi verificar as dimensões ontológica, deontológica, axiológica, principiológica e constitucional do prequestionamento ficto, levandose em consideração sua positivação no novo CPC, no artigo 979, e as implicações dessa positivação. This thesis report presents a method to analyze the communication architecture for the Intelligent Transportation Systems (ITS). The report also includes a case study on ASN.1 notation and analysis of its encoding rules. Included in the report is also: (i) accompanying instruction on how to use ASN.1 compilers to produce the C/C++ message encoder/decoder, and (ii) analysis of Non-IP communications of Communication Access for Land Mobiles (CALM-FAST) protocol stack in ITS. The thesis is a part of the research project entitled “SCOOP”, a joint project between SCANIA CV AB and KTH. The purpose of this thesis is to contribute to the ultimate goal, which is to equip a vehicle with necessary hardware and software technology to provide a platooning behavior in the GCDC 2011 competition. This goal is achieved by the means of wireless communication system for both vehicle to vehicle and vehicle to road side units communications in the platoon. Overall, this thesis introduces the important usage of ASN.1 in implementation of cut-edge telecommunication systems especially in V2V and V2I communication; and clarifies the CALM-FAST protocol stack in mobile nodes.<br>Kartlägga CALM-FAST protokollet och hur det användes tillsammans med den i tävlingen GCDC 2011 fastslanga kommunikationsprotokollet. Background: Several studies have shown there has been significant lack of knowledge about women who have sex with women cross infect each other with STIs. It has been less likely that women who have sex with women visit health care than heterosexuals, they often felt badly treated, invisible and did not always know where to turn in case of an STI. Objective: The objective was to examine if STI information from Kalmar and Växjö county websites and GCK-summit, targeted women who have sex with women. Method: With a queer theoretical perspective a critical discourse analysis was made by STI information on the internet. Results: Women who have sex with women have been excluded in the STI information currently available through Kalmar and Växjö county websites. The public notices for screening tests have not been designed in such way it includes women who have sex with women. Women who have sex with women have been an obscure group. Conclusion: The conclusion was that women who have sex with women have not been included in the STI information available on the internet within the county councils of Kalmar and Kronoberg.

The inappropriate expression, localization, or activation of kidney protein kinases may precipitate renal dysplasia or cystic diseases. The expression and activation of the Ste20-like kinase, SLK, is increased during renal development and recovery from ischemic acute renal failure (ARF). Activation of SLK promotes apoptosis and, during development and healing, SLK may regulate cell growth. Alternatively, in glomerular diseases, the activation of SLK and apoptosis of glomerular epithelial cells (GEC) may be harmful and lead to sclerotic glomerular injury. The overall aim of the project is to elucidate the regulation of SLK activity in order to better understand the role of SLK in kidney development and injury. It is proposed that activation of SLK may be due to upregulation of expression, which may then favor homodimerization and autophosphorylation. Serine and threonine residues were mutated in the putative activation segment of the SLK catalytic domain. Wild type (WT) and mutant proteins were expressed in COS-1 cells or GEC. Compared with SLK WT, the S189A mutant and the T183A/S189A double mutant showed trivial in vitro kinase activity, while kinase activity was reduced significantly by the T183A mutation. Expression of SLK WT increased activation-specific phosphorylation of c-Jun N-terminal kinase (JNK) and p38 kinase, reflecting enhanced signaling via stress kinase pathways. In contrast, activation of JNK by SLK T183A, S189A, and T183A/S189A and p38 by T183A/S189A was impaired. Similarly, expression of SLK WT stimulated activator protein-1 (AP1) reporter activity, but activation of AP1 by the three SLK mutants was ineffective. To test if homodimerization of SLK affects phosphorylation, the cDNA encoding the SLK catalytic domain (amino acids 1-373) was fused with a cDNA for a modified FK506 binding protein, Fv (SLK-Fv). After transfection, addition of AP20187 (an FK506 analog) induced regulated dimerization of SLK-Fv. AP20187-stimulated dimerization enhanced the kinase activity of SLK-Fv WT. In contrast, kinase activities of SLK-Fv T183A/S189A and activation site mutants E79A, K63R, T193A were weak, and were not enhanced after dimerization. Surprisingly, phosphomimetic mutants of SLK-Fv, including T183D, T183E, S189D, and S189E showed weak kinase activity, which was unaffected by dimerization. Co-transfection of SLK and T183A/S189A failed to silence the kinase activity of the WT in an in vitro kinase assay. Finally, apoptosis and late apoptosis were increased after expression of SLK-Fv WT, but not T183A/S189A. Thus, phosphorylation of T183, S189 and T193 plays a key role in the activation and signaling of SLK and could represent a target for novel therapeutic approaches to renal injury.<br>Une expression, une localisation ou une activation inappropriée des protéines kinases rénales peut provoquer une dysplasie rénale ou une maladie cystique prématurée. L'expression et l'activation de la protéine Ste20-like kinase (SLK) sont plus élevées pendant le développement des reins et durant la guérison d'une insuffisance rénale ischémique aiguë. Une activation de la protéine SLK entraîne l'apoptose des cellules in vitro et in vivo. Au cours du développement et de la régénérescence cellulaire, SLK peut réguler la croissance cellulaire. Par contre, dans les glomérulonéphrites, l'activation de la SLK et une apoptose des cellules épithéliales glomérulaires (GEC) peuvent être dommageables et peuvent provoquer des lésions glomérulaires sclérotiques. L'objectif de ce projet est d'élucider la régulation de l'activité de la SLK dans le but de mieux comprendre son rôle dans le développement et les lésions ou maladies glomérulaires rénales. Ce projet postule que l'activation de la protéine SLK peut être due à une régulation à la hausse de l'expression, qui peut alors favoriser une homodimérisation et une autophosphorylation. Pour étudier le rôle de la phosphorylation dans la régulation de l'activation, les résidus sérine (S189) et thréonine (T183) ont été mutés dans le segment putatif du domaine catalytique de la SLK. Deux mutants simples (T183A et S189A) et un double mutant (T183A/S189A) ont été créés. La forme sauvage (WT) et les mutants de la SLK ont été exprimés dans les cellules cos-1 et GEC. SLK WT phosphorylais la protéine basique de myéline (MBP) contrairement aux mutants S189A et T183A/S189A qui montraient une activité kinase in vitro non significative. La mutation T183A diminuait la phosphorylation de MBP significativement. L'analyse des voies de signalisation de la kinase N-terminal c-jun (JNK) et de la p38 par la SLK a été étudiée sachant que l'expression de la SLK WT stimule la phosphorylation de la JNK et de la p38. Tant que T183A/S189A n'a pas pu activer la JNK et la p38, les mutants T183A et S189A ont activé la JNK à un degré moindre que la SLK WT. Ces résultats démontrent que la phosphorylation de T183 et de S189 est importante pour l'activation de la SLK. De façon similaire, l'expression de la SLK WT a stimulé l'activation de la protéine activatrice 1 (AP1), un facteur de transcription de la famille c-fos et c-Jun, tandis que les trois mutants ont été incapables de stimuler AP1. Pour déterminer si l'homodimérisation de la SLK affecte la phosphorylation, l'ADN complémentaire de la SLK codant le domaine catalytique (1-373) a été fusionné avec l'ADN complémentaire d'une protéine modifiée qui s'attache à des molécules FK506 (Fv), pour créer SLK-Fv. Après la transfection, l'addition du AP20187, un analogue de FK506, induit la dimérisation de la SLK-Fv. La dimérisation occasionnée par l'AP20187 a augmenté l'activité catalytique de la SLK-Fv. Par contre, tous les mutants Fv fusionnés T183A/S189A, E79A, K63R et T193A démontraient une activité catalytique faible, que la dimérisation n'a pas pu augmenter. Ce qui est particulièrement intéressant, les mutants phosphomimétiques T183D, T183E, S189D et S189E montraient aussi une activité catalytique faible qui n'a pas été affectée par la dimérisation. Finalement, l'apoptose et la nécrose sont accrues suite à une surexpression de la SLK-Fv WT, mais pas avec le mutant T183A/S189A-Fv. Ainsi, les acides aminés K63 et E79 et la phosphorylation de T183, S189 et T193 jouent un rôle important dans la régulation de la SLK et pourraient être ciblés par des approches thérapeutiques pour le traitement des néphropathies.

Questa tesi tratta il metodo di produzione del Latte Fieno STG, un prodotto alimentare di nicchia riconosciuto come specialità tradizionale garantita nel 2016 dalla UE; sono, inoltre, indicate le principali differenze col metodo di produzione del latte standard e in riferimento alle caratteristiche dell’alimento stesso. Viene, infine, trattato un esempio di trasformazione di questa materia prima reperibile sul mercato e delle sue caratteristiche, in riferimento anche esse con un prodotto analogo derivante da latte prodotto da stalle convenzionali. Il Latte Fieno STG è un prodotto alimentare originario dall’Austria che è stato riconosciuto, nel 2016, specialità tradizionale garantita (STG) dall’Unione Europea; deriva da un processo di produzione diverso da quello con cui si ottiene dal latte convenzionale, difatti le differenze principali si riscontrano nell’alimentazione: sono somministrati erba, legumi, cereali e fieno, senza l’utilizzo di alimenti fermentati e mangimi OGM secondo il regolamento di esecuzione UE 2016/304. La distribuzione degli alimenti segue l’andamento stagionale e molto spesso si associa anche l’utilizzo del pascolo in estate. Un’ulteriore differenza è relativa ai tipi genetici utilizzati, dato che nelle zone montane è possibile sfruttare, oltre alle classiche razze da latte, anche razze autoctone più idonee a tali ambienti. Ciò porterà a differenze dal punto di vista qualitativo e compositivo del latte: il contenuto di proteine e lipidi risulta maggiore nel Latte Fieno rispetto al latte tradizionale. Inoltre, si ritrovano caratteristiche organolettiche peculiari, derivanti principalmente dai vegetali ricavati dal pascolo, che apportano una colorazione gialla. Anche la composizione in acidi grassi è variabile. Il Latte Fieno STG rimane attualmente una piccola realtà, utile soprattutto per il turismo gastronomico delle zone montane, ma non ancora così remunerativo in un mercato più ampio.

Abstract:
We present the development of a novel, UHV-compatible device fabrication strategy for the realisation of nano- and atomic-scale devices in silicon by harnessing the atomic-resolution capability of a scanning tunnelling microscope (STM). We develop etched registration markers in the silicon substrate in combination with a custom-designed STM/ molecular beam epitaxy system (MBE) to solve one of the key problems in STM device fabrication ??? connecting devices, fabricated in UHV, to the outside world. Using hydrogen-based STM lithography in combination with phosphine, as a dopant source, and silicon MBE, we then go on to fabricate several planar Si:P devices on one chip, including control devices that demonstrate the efficiency of each stage of the fabrication process. We demonstrate that we can perform four terminal magnetoconductance measurements at cryogenic temperatures after ex-situ alignment of metal contacts to the buried device. Using this process, we demonstrate the lateral confinement of P dopants in a delta-doped plane to a line of width 90nm; and observe the cross-over from 2D to 1D magnetotransport. These measurements enable us to extract the wire width which is in excellent agreement with STM images of the patterned wire. We then create STM-patterned Si:P wires with widths from 90nm to 8nm that show ohmic conduction and low resistivities of 1 to 20 micro Ohm-cm respectively ??? some of the highest conductivity wires reported in silicon. We study the dominant scattering mechanisms in the wires and find that temperature-dependent magnetoconductance can be described by a combination of both 1D weak localisation and 1D electron-electron interaction theories with a potential crossover to strong localisation at lower temperatures. We present results from STM-patterned tunnel junctions with gap sizes of 50nm and 17nm exhibiting clean, non-linear characteristics. We also present preliminary conductance results from a 70nm long and 90nm wide dot between source-drain leads which show evidence of Coulomb blockade behaviour. The thesis demonstrates the viability of using STM lithography to make devices in silicon down to atomic-scale dimensions. In particular, we show the enormous potential of this technology to directly correlate images of the doped regions with ex-situ electrical device characteristics.

This thesis examines aspects of the prevention of sexually transmitted diseases (STDs) in the Australian community, with a particular emphasis on HIV/AIDS in the context of general practice (or primary care settings). The work has four broad aims: i) To describe the primary prevention of sexually transmitted diseases, following from the arrival of the HIV/AIDS pandemic in Australia ii) To describe HIV/STD risk behaviour iii) To summarise previously known evidence of interventions to reduce risk and to raise awareness of HIV and other sexually transmitted diseases iv) To contribute new evidence addressing the potential of the general practitioners' role in HIV/STD prevention The first chapter gives a brief review of the history of HIV/AIDS from its discovery in the United States of America to its appearance in Australia and New Zealand, and discusses the Australian response strategies, both initial and continuing, to confine the epidemic. Specifically, the arrival of HIV/AIDS gave rise to increasing awareness of sexually transmitted diseases, which hitherto, although sometimes chronic, were rarely fatal. The public health risk of HIV necessitated swift government action and led to wider acceptance of publicity about sexual behaviour. Although the thesis does not concentrate solely on HIV, this is still an emphasis. This chapter provides useful background to ensuing chapters. Chapter Two provides an overview of behavioural risk in sexually transmitted diseases. It gives a review of risk factor prevalence studies, and introduces risk behaviour and cognitive models of behaviour change, as applied to STD risk. Sexual behaviour is a complex social interaction, usually involving more than one person, and relying on the personality and behaviour patterns intrinsic to the individuals taking part. It is therefore perhaps more challenging to alter than behaviour which is undertaken alone, being dependent on the behaviour and intentions of both parties. Moreover, comprehensive assessment of sexual risk behaviour requires very detailed information about each incident. Its private nature makes accurate data difficult to obtain, and sexual risk behaviour is, correspondingly, difficult to measure. Chapter Three reviews the effectiveness of interventions tested in primary health care settings to reduce sexual risk behaviour. The candidate uses a replicable method to retrieve and critique studies, comparable with standards now required by the Cochrane Collaboration. From 22 studies discussed, nine health interventions were short, 'one-shot', efforts owing to limited time, resources and other practical constraints. This review demonstrates the scarcity of interventions with people who may be perceived as 'low-risk'. Only four interventions were carried out in community health centres and two in university health clinics. One of the university interventions showed no change in sexual behaviour in any of three arms of the intervention (Wenger, Greenberg et al 1992) while the other showed an increase in condom use in both groups, although the intervention group's self-efficacy and assertiveness also improved (Sikkema, Winett & Lombard 1995). The rationale for the intervention, where given, is described. Chapter Four analyses the content, format and quality of sexual health information brochures available in New South Wales at the time of the candidate's own planning for an interventional study. One of the most effective ways to disseminate information widely is by the use of educational literature, especially when the subject material is potentially sensitive or embarrassing to discuss in person. In this chapter, the candidate reviews the literature available at the time of designing the intervention used in Chapter Five. Readability, attractiveness, clarity and the accurate presentation of facts about sexually transmitted disease risk are examined for each pamphlet. Forty-seven pamphlets were scored according to the Flesch formula, and twenty-four of these scored in the 'fairly' to 'very difficult' range. There was, therefore, a paucity of easy-to-read material on these subjects. Chapter Five evaluates a general practitioner-based counselling intervention to raise awareness of sexually transmitted diseases and to modify HIV/STD risk behaviour. While adults aged 18-25 are less likely than older cohorts to have a regular general practitioner or to visit often, most people visit a general practitioner at least once a year. This could provide an opportunity for the general practitioner to raise preventive health issues, especially with infrequent attendees. As the effectiveness of an opportunistic intervention about sexual risk behaviour was yet to be tested, the candidate designed an innovative randomised controlled trial to raise awareness of risk and increase preventive behaviour. The participation rate was 90% and 76% consented to followup; however the attrition rate meant that overall only 52% of the original participants completed the follow-up questionnaire. The intervention proved easy and acceptable both to GPs and to patients, and risk perception had increased at three months' follow-up; however this occurred in both the control (odds ratio 2.6) and the intervention group, whose risk perception at baseline was higher (odds ratio 1.3). In order to establish some markers of risk in the general population, Chapter Six analyses data resulting from questions on sexual behaviour asked in the Central Sydney section of the NSW Health Survey. The candidate advocated for inclusion of relevant questions to determine some benchmarks of sexual risk behaviour and to provide an indication of condom use among heterosexuals. Although limited in scope as a result of competing priorities for questions in the survey, results demonstrate that, while a small percentage of people were at risk, those with higher levels of partner change or of alcohol use were the most likely to always use condoms. Specifically, 100% of those with more than four new partners in the last 12 months had used condoms with every new partner. In addition, 'heavy' alcohol users were more likely to report condom use every time with new partners (odds ratio 0.34). To furnish data to inform future planning of educational activities for general practitioners, Chapter Seven presents the results of a survey of Central Sydney general practitioners' opinions and current practices in HIV risk reduction with in the broader context of sexually transmitted disease prevention. The general practitioner is in an ideal position to provide information and advice, especially if future research affirms the impact of such advice on STD risk behaviour. General practitioners in this study said they would be slightly more likely to discuss sexual health matters with young patients than with older ones (p=0.091), but this was not significant. The most cited barrier to discussing sexual health was inadequate remuneration for taking time to do so (over 50% gave this reason). The next most cited obstacle was difficulty in raising the subject of STDs or HIV in routine consultations, but this reason was given by less than half the sample. Forty-six percent had participated in continuing medical education programs in STDs, HIV/AIDS, or hepatitis diagnosis or management; 32% of GPs had patients with HIV, and 55% of all GPs indicated they would like more training in management and continuity of care of HIV patients. Approximately half (51%) wanted more training in sexuality issues, including sexual dysfunction. Chapter Eight reviews the whole thesis and discusses future directions for the research agenda.

Abstract:
The Scanning Tunneling Microscope (STM) was invented by Gerd Binnig and Heinrich Rohrer and gave them the Nobel Prize in Physics 1986. STM can give us atomic resolution of a surface by applying a voltage between a very sharp tip (STM-tip) and the surface of a material that we want to examine. The STM-tip is moving over the surface and a computer is collecting the tunnel current in every single point to create a digital image. This diploma work is focused on the preparation of the STM-tip. The preparation method that is used is electrochemical etching of a tungsten wire. The sharper the STM-tip is the better resolution in the STM images we can get. With the purpose to get as sharp tip as possible and with a well-defined geometry, we prepared several tips by systematically varying the etching parameters such as voltage, current, concentration and wire length. A new method has been tested to minimize the oxidation on the surface and finally the tips were characterized with scanning electron microscope (SEM).

Abstract:
The fundamental atomic and electronic behaviour of clean silicon surfaces has been studied within a simple tight-binding picture of bonding in solids. Of the various contributions to the surface binding energy, the lowering in the promotion energy (i.e. rehybridization) which accompanies localized Jahn-Teller distortions has been identified as a major electronic driving force underlying the stability of silicon surfaces. The structure of Si(113) has been experimentally determined by the technique of scanning tunnelling microscopy (STM). Despite its high index, the Si(113) surface is found to be highly stable. STM images of both empty and filled states provide strong evidence for a particular structural model with a 3x2 unit cell. The STM results are explained in terms of a general rehybridization principle, suggested by the earlier theoretical study, which accounts for the low surface energy as well as the observed spatial distribution of empty and filled states. In addition, the STM images reveal a high density of domain boundaries which introduce energy states that pin the Fermi level and explain earlier reports of a 3x1 reconstruction for this surface. Voltage-dependent STM image simulations for the Si(113)3x2 surface have been carried out using a simple tight-binding description of surface electronic structure. Quantitative agreement with experiment is obtained confirming the qualitative rehybridization arguments used previously. The local barrier for tunnelling electrons is shown to have an important effect on the interpretation of STM images. The high stability of clean Si(l 13) is shown by STM to be disrupted by adsorption of submonolayer amounts of atomic hydrogen which saturates dangling bonds. Mass transport of silicon occurs and structural models are proposed for the resultant mixed 2x2 and 2x3 surface.

Abstract:
Many deductions made about STM images are based upon the model of Tersoff and Hamann, in which images are given in principal by a combination of surface atomic positions and local charge density. There is a now a need for a fuller understanding of this technique in order to explain experimental evidence which indicates that the tip and sample can interact strongly during normal imaging. In order to investigate the fundamental STM imaging process, a method for deducing the tunnel barrier height has been developed which is based on corrugation height measurements of constant current topographs. From experiments on clean Cu(100), values of the tunnel barrier height have been shown to be somewhat below the workfunction (~ 1-2.5eV) but are in good agreement with other reports of atomically resolved barrier height data. At large values of the tunnel conductance (~ 1μS), a fall-off (based upon extrapolation of large separation data) in the corrugation heights is observed with increasing conductance. This effect is quantitatively explained using a Molecular Dynamics simulation of the tip approaching the sample. The simulation gives a good estimate of both the absolute tip-sample separation and site-dependent tip-surface forces. Distributions of corrugation heights indicate that variations in both tip geometry and chemistry are likely to occur in practice and strongly influence the phenomena described above. Similarly, it is found that increased local tunnel barrier heights are measured when the Cu(100) surface is modified with small numbers of single halogen atoms. This data has been used to estimate the contributions to the increase in local barrier height of both adsorbate induced dipoles and geometric topography. Values for the charge transfer between the surface and adsorbate have been established. The process of tip-induced adsorbate manipulation has also been demonstrated at room temperature.

Moore’s Law appears to be drawing to an end. With it, the assumption that software written today will be more efficiently executed in the future simply due to processors’ evolution is being challenged. On the other hand, parallel applications can still be made more efficient by distributing work among different processors to be executed at the same time, thus reducing overall execution time. To enable parallelization, we must have multiple processor cores. This has led to the popularization of multicore architectures. However, writing parallel applications is not trivial. A program must be either written from the start to be executed in parallel, or later adapted for parallel execution. The programmer has the error-prone task of parallelizing the application through use of concurrency and parallelism constructs. Locking, the most common concurrency option, presents risks for inexperienced programmers, such as the famous Deadlock and Livelock problems. As we move from single core architectures to multicore, our programming languages need to make it easier for the programmers to use concurrency. Many researchers have pointed at Software Transactional Memory (STM) as an answer to that issue, as it is a lock-free, abstract way to guarantee isolated access to shared resources. But adapting for STM a program that uses lock is not simple. Besides being an error-prone task, technical details of the language might require special attention to preserve the program’s behavior. In this dissertation, we propose a set of program transformations for concurrency constructs in Haskell, a purely functional programming language. They may be used to refactor a program’s existing locks into transactional constructs from Haskell’s STM implementation. This allows a programmer to gain the benefits of working on STM even for programs which were already developed using locks. Each transformation is accompanied by execution examples and a discussion on its ability to preserve program behavior. We also present a supporting study, in which a controlled experiment was used to evaluate the benefits of locks or STM for the development of Haskell programs. Although subjects’ opinions tended to favor lock-based concurrency, those which used STM overall committed significantly fewer mistakes and required on average 12% less time to finish their assignments.<br>Recentemente, a queda na taxa de crescimento da quantidade de transístores integráveis em processadores tem desacelerado o crescimento de poder computacional. A lei de Moore parece aproximar-se de seu fim. Com isso, é desafiada a premissa de que software escrito hoje terá melhor desempenho no futuro simplesmente devido à evolução dos processadores. Ainda assim, aplicações paralelas ainda podem se tornar mais eficientes ao se distribuir trabalho entre diferentes processadores para execução simultânea. Para permitir a paralelização, são necessários múltiplos núcleos de processamento, o que tem levado à popularização de arquiteturas multinúcleo. Entretanto, a escrita de aplicações paralelas não é trivial. Deve-se escrever um programa para execução paralela desde sua concepção, ou adaptá-lo posteriormente para execução paralela. O programador tem a difícil tarefa de paralelização da aplicação através do uso de construções de concorrência e paralelismo. Travas, a mais comum opção para concorrência, apresentam riscos para programadores inexperientes, tais quais os famosos problemas de Deadlock e Livelock. Ao adaptarem-se de arquiteturas de um único núcleo para as de multinúcleo, as linguagens de programação precisam facilitar o uso de concorrência para os programadores. Muitos pesquisadores têm indicado Memória Transacional em Software (STM, do inglês Software Transactional Memory) como a resposta para esse problema, por ser uma forma abstrata e não bloqueante para garantia de acesso isolado a recursos compartilhados. Mas adaptar para STM programas que usam travas não é simples. Além de ser uma atividade propensa a erros, detalhes técnicos da linguagem podem requerer cuidados para se preservar o comportamento do programa. Nesta dissertação, é proposto um conjunto de transformações de programas para construções de concorrência em Haskell, uma linguagem de programação puramente funcional. Elas podem ser usadas para refatorar travas de um programa para uso de construções transacionais da implementação de STM em Haskell. Isso permite ao programador aproveitar os benefícios do trabalho com STM mesmo para programas já desenvolvidos com uso de travas. Cada transformação é acompanhada de exemplos de execução e uma discussão sobre sua capacidade de preservar o comportamento do programa. Também é apresentado um estudo de apoio, no qual um experimento controlado foi usado para avaliar os benefícios do uso de travas ou STM no desenvolvimento de programas em Haskell. Apesar das opiniões dos participantes terem favorecido o uso de travas, aqueles que usaram STM cometeram em geral menos erros e em média precisaram de 12% a menos de tempo para terminar suas tarefas.
