Abstract:

Gegenstand der Dissertation ist die Darstellung der Entwicklung der im Südharz und in Nordthüringen ansässigen Grafen zu Stolberg seit ihrer Ersterwähnung im Jahre 1210 bis zum Wiener Kongress 1815. Über einen Zeitraum von mehr als 600 Jahren wird das Beziehungsverhältnis zwischen diesem Harzgrafengeschlecht und den Landgrafen von Thüringen und den nachfolgenden Herzögen, Kurfürsten bzw. Königen von Sachsen näher untersucht. Breiten Raum nimmt dabei der handbuchartige Überblick über die Entwicklung der Lehnsverhältnisse in den einzelnen stolbergischen Besitzungen im thüringisch-sächsischen Raum ein. Kurze Vergleiche zu einigen anderen adeligen Herrschergeschlechtern und den Prozessen der modernen Staatsbildung in ausgewählten Kleinterritorien des Untersuchungsgebietes bilden den Abschluss der Arbeit.

Abstract:

Les phytoplasmes sont des bactéries phytopathogènes, sans paroi, qui appartenant à la classe des Mollicutes. Ils ne peuvent pas etre cultivés in vitro et sont limités à des tubes du phloème. Ils provoquent des centaines de maladies chez de nombreuses espèces végétales dans le monde entier, ce qui conduit à des pertes de récolte importantes. Les phytoplasmes sont transmis naturellement par des insectes suceurs de sève dans laquelle ils se multiplient. Ils induisent des symptômes graves, notamment le jaunissement, la croissance limitée, déclin, ainsi que des anomalies des fleurs et des fruits. L'infection par le phytoplasme du stolbur, en particulier, affect fortement la morphologie florale. Dans la tomate, deux isolats différents du phytoplasme du stolbur, nommé C et PO, induisent des symptômes différents. La tomate infectée par le phytoplasme du stolbur PO montrent des malformations florale telles que les sépales hypertrophiés, les pétales et les étamines avortées ce qui conduit à la stérilité. En revanche, la tomate infecté par le phytoplasme du stolbur C ont de petites feuilles de tomate en retrait, mais les fleurs presque normale, et produisent des fruits. Nous avons précédemment montré que SlDEF, un gène impliqué dans la formation des pétales est réprimé dans des plantes de tomate infectée par le stolbur phytoplasme PO. Toutefois, l'expression de son facteur de transcription, codée par le gène FA, est resté stable ou voir légèrement augmentée. Nous avons donc émis l'hypothèse que la répression de SlDEF pourrait être dû à une méthylation de l'ADN. Pour tester cette hypothèse, nous avons étudié l'expression des gènes de méthylases et de déméthylases. Ils étaient en général réprimés dans les tomates infectées par le phytoplasme du stolbur PO, ce qui était en accord avec l'hypothèse De plus, nous avons étudié les voies de défense activée chez les tomates infectées par le phytoplasme du stolbur. Pour se défendre, les plantes utilisées des molécules de signalisation comme l'acide salicylique (SA), l'acide jasmonique (JA) et d'éthylène (ET). Nous avons étudié l'expression de 21 gènes de défense dépendants SA / JA / ET, des gènes de biosynthèse et les facteurs de transcription chez les tomates infectées par les phytoplasmes du stolbur C et PO. Nous avons également étudié l'effet de la pré-activation des voies de SA et JA sur la production des symptômes. Nos résultats montrent clairement que les voies de défense ont été activées différemment dans les tomates infectés par le phytoplasme du stolbur C et PO. En effet, les voies de défense dépendantes de SA, ET et JA ont été activées chez les tomates infectées par le phytoplasme du stolbur C alors que seulement les voies dépendantes SA et ET ont été activés dans les tomates infectées par stolbur PO . En outre, la pré-activation de la voie de défense dépendante SA par l'application de BTH modifie légèrement l'évolution des symptômes de maladies causées par le phytoplasme du stolbur PO<br>Phytoplasma are cell wall-less, phytopathogenic bacteria belonging to the class Mollicutes. They have not been cultured in vitro and are restricted to the phloem sieve tubes. They cause hundreds of diseases in many plant species worldwide, resulting in important crop losses. Phytoplasmas are naturally transmitted by sap-sucking insects in which they multiply. They induce severe symptoms including yellowing, restricted growth, decline, as well as major flowers and fruits abnormalities.The stolbur phytoplasma infection, in particular, has been reported to strongly affect floral morphology. In tomato, two different isolates of stolbur phytoplasma, named C and PO, induce different symptoms. The stolbur PO phytoplasma-infected plants show abnormal flower development such as hypertrophied sepals, and aborted petals and stamens leading to sterility. In contrast, stolbur C phytoplasma-infected tomato have small indented leaves but nearly normal flowers, and produce fruits. We have previously shown that SlDEF, one gene involved in petal formation, was repressed in stolbur PO phytoplasma-infected tomato. However, the expression of its transcription factor, encoded by the gene FA, was unchanged or slightly up-regulated. So we hypothesized that SlDEF repression could be due to DNA methylation. To test this hypothesis, we studied the expression of DNA methylases and demethylases genes. They were in general down-regulated in stolbur PO infected tomato, which was in agreement with the hypothesis. However, the regulation of SlDEF expression could not be firmly correlated to the DNA methylation status of its promoter region. In addition, we studied the plant defense pathways activated in stolbur phytoplasma-infected tomato. To defend themselves, plants used signalling molecules like Salicylic acid (SA), Jasmonic acid (JA) and Ethylene (ET). We studied the expression of 21 SA/JA/ET regulated defense and biosynthesis genes including transcription factors in stolbur C and PO phytoplasma-infected tomato as compared to healthy ones. We also studied the effect of pre-activation of SA and JA mediated defense pathways on symptom production. Our results clearly showed that defense pathways were activated differently in stolbur C and PO phytoplasma-infected tomato. Indeed, SA ET and JA dependant pathways were activated in stolbur C-infected tomato while only SA and ET dependant pathways were activated in stolbur PO-infected plants. In addition, pre-activation of SA-dependent defense pathway by application of BTH slightly modify the evolution of disease symptoms caused by stolbur PO phytoplasma whereas no effect was observed after treatment with an analogue of JA

Abstract:

Les phytoplasmes, bactéries phytopathogènes sans paroi de la classe des mollicutes, sont localisés exclusivement dans le phloème et ont résisté à la mise en culture. Responsables de nombreuses maladies de plantes, aucun moyen de lutte curative n'existe à ce jour. Une strátégie de lutte par transgénèse a été entreprise au laboratoire. Elle consiste à exprimer une immunoglobuline Ig) recombinante de type scFv spécifique du phytoplasme du stolbur, dans le tabac, hôte du phytoplasme. Afin d'évaluer l'efficacité de cette stratégie, nous avons étudié l'action des Ig sur le mollicute phytopathogène cultivé Spiroplasma citri. Nous avons montré que, tant les Ig natives que les scFv, dirigés contre la protéine majeure de sa surface, inhibent la multiplication de ce microorganisme. Nous avons ensuite réalisé l'ingénierie de scFv anti-stolbur pour leur expression dans le tabac suivant les voies d'adressage apoplastique (tabacs 35-pel-scFv et RSS-scFv-KD), afin d'évaluer la production des anticorps dans la sève élaborée où sont localisés les phytoplasmes. Nous avons mis en évidence que les tabacs 35-pel-scFv produisent les scFv en quantité importante mais ne montrent qu'une résistance transitoire à la maladie, les anticorps n'étant probablement pas ou peu adressés vers la sève élaborée. En effet, il a été récemment montré que les protéines de la sève sont produites dans le cytoplasme des cellules compagnes pour transiter via les plasmodesmes vers le tube criblé. Les tabacs pour lesquels l'expression du scFv est ciblée vers le cytoplasme de façon constitutive (35-scFv) ou spécifiquement dans les tissus du phloème (RSS-scFv-KD), malgré un bon niveau de transcription des ARNm du scFv, ne produisent pas d'anticorps détectables et résistent peu à la maladie. Les anticorps sont donc efficaces in vitro pour inhiber la croissance des mollicutes phytopathogènes, mais leur ciblage vers la sève élaborée s'avère difficile. Différentes stratégies envisagées pour optimiser ce ciblage sont discutées<br>Phytoplasmas are phloem-restricted wall-less bacteria of the class mollicutes which have resisted in vitro cultivation so far. They are responsible for numerous plant diseases and there is no curative method for disease control. An antibody-based resistance method is evaluated in our laboratory for the stolbur phytoplasma. It consists in expressing scFv recombinant immunoglobulins (Ig) specific for this phytoplasma in tobacco plants, natural hosts for this pathogen. First we demonstrated that both native Ig and recombinant scFv, specific for the major surface protein of the culturable phytopathogenic mollicute Spiroplasma citri, inhibit its growth in vitro. Secondly, we engineered various anti-stolbur scFv constructions for their expression in tobacco plants via the apoplastic (35-pel-scFv plants) or symplasmic (35-scFv and RSS-scFv-KD) pathway, to evaluate their production into the phloem sap were phytoplasmas are located. Our results indicate that scFv, although produced in large amounts in 35-pel-scFv tobaccos, were probably not, or only slightly, present in the phloem sap. Only a transient disease resistance was observed in these plants. Indeed, it has been recently demonstrated that many phloem sap proteins are produced in the cytoplasm of companion cells and that they move towards sieve tubes via plasmodesmatas. Transgenic tobaccos plants in which scFv expression was targeted to the cytoplasm either constitutively (35-scFv) or specifically to phloem tissues (RSS-scFv-KD), in spite of a good transcription of scFv mRNAs, did not produce detectable amounts of scFv. These tobaccos, as previously, showed little resistance to the disease. As scFv inhibit in vitro growth of phytopathogenic mollicutes, the plantibody strategy appears to be well adapted to control phytoplasma diseases. However, targeting scFv in the phloem sap has to be optimized and various alternatives for sap targeting are discussed

Abstract:

Les phytoplasmes sont des bactéries phytopathogènes de la sève phloémienne non dsponibles en culture. Une technique dérivée de l'hybridation soustractive (SSH nous a permis de purifier et d'isoler l'ADN du phytoplasme du stolbur à partir de pervenche infectée. L'analyse comparée de 181 loci génétiques (113 kpb) indique que le génome du phytoplasme du stolbur contient environ 26 % G+C et que 83 % des 217 séquences codantes mises en évidence correspondent à une séquence homlologue chez 'Candidatus Phytoplasma asteris' (OY-M). Les gènes qui codent la riboflavine kinase et la protéine 1H10 homologue à une protéine de surface de mycoplasme sont absents chez 'Ca. P. Asteris'. La protéine de 59 kDa prédite in silico possède un peptide signal putatif et un domaine transmembranaire C-erminal qui permettraient sa sécrétion et son exposition à la surface du phytoplasme. Ces gènes pourraient jouer un rôle dans l'adaptation du phytoplasme à une famille spécifique d'insectes vecteurs<br>Phytoplasmas are phloem-restricted bacterial plant pathogens unvailable in culture. A modified Suppression Subtractive Hybridization (SSH) method allowed to isolate a nearly pure stolbur phytoplasma DNA from infected Catharanthus roseus periwinkle plants. Comparative analysis of 181 stolbur phytoplasma genetic loci (113 kbp) indicated that the G+C content of the stolbur phytoplasma genome should be about 26 %. Among 217 protein coding sequences, 83 % were homologous to 'Candidatus Phytoplasma asteris' (OY-M) genes. Two genes encoding proteins homologous to a mycoplasma surface protein and riboflavin kinase but absent in 'Ca. P. Asteris' were fully characterized. The stol-1H10 gene encodes a 59 kDa protein harbouring a putative signal peptide and a C-terminal transmembrane domain which should allow the matured 1H10 protein to be exposed at the phytoplasma surface. Such genes could play a role in the adaptation of stolbur phytoplasma to a specific family of insect vector

Abstract:

Gegenstand der Dissertation ist die Darstellung der Entwicklung der im Südharz und in Nordthüringen ansässigen Grafen zu Stolberg seit ihrer Ersterwähnung im Jahre 1210 bis zum Wiener Kongress 1815. Über einen Zeitraum von mehr als 600 Jahren wird das Beziehungsverhältnis zwischen diesem Harzgrafengeschlecht und den Landgrafen von Thüringen und den nachfolgenden Herzögen, Kurfürsten bzw. Königen von Sachsen näher untersucht. Breiten Raum nimmt dabei der handbuchartige Überblick über die Entwicklung der Lehnsverhältnisse in den einzelnen stolbergischen Besitzungen im thüringisch-sächsischen Raum ein. Kurze Vergleiche zu einigen anderen adeligen Herrschergeschlechtern und den Prozessen der modernen Staatsbildung in ausgewählten Kleinterritorien des Untersuchungsgebietes bilden den Abschluss der Arbeit.
