Full textThis work is focused on a biological process which controls bacterial protein synthesis, trans-translation. This all-in-one process allows the rescuing of ribosomes stalled on defective mRNA, the degradation of the problematic peptides and mRNA. It is driven by two principal actors that interact with the ribosome: transfer-messenger RNA (tmRNA) and Small protein B (SmpB). In a first chapter, by a cryo-electron microscopic (cryo-EM) study, two near-atomic resolution structures, involving the ribosome and various trans-translation actors, were obtained. The first one highlights the interactions between RNase R, an enzyme responsible for mRNA degradation during trans-translation, and the bacterial ribosome. The second one corresponds to the characterization of two early trans-translation states at a near-atomic resolution. New interactions have been observed between SmpB and tmRNA H5 helix. In a second chapter, trans-translation is used as a target for the development of new antibiotic molecules. Indeed, this pathway is often necessary for bacterial survival and pathogenicity. Towards this aim, we designed and set up a new in vitro assay for high-throughput screening assays. This efficient system is based on fluorescence measurements of a GFP reassembled through trans-translation by a mutated tmRNA. This system has been validated and will be used for the discovery of new anti-trans-translation compounds

Naturally occurring lignans of the 1-aryltetralin type or their synthetic derivatives present a wide range of biological activities. The 1-aryltetralin scaffold represents a privileged structure which remains to be exploited by combinatorial chemistry. Several small libraries of novel heterolignans have been synthesized from thuriferic methyl ester, a podophyllotoxine derivative. Parallel solution phase synthesis and new synthetic methodologies centered on indium chemistry have been developed. Full textThe undertaken studies relates to the determination of the positions relating of the vortical structures to the upper surface of simple or modified delta and ogival wings, cones, warheads of revolution and complex bodies (fuselage) with the application of the angular criteria of stability of the vortical systems. A wide documentation and visualizations in wind tunnel made it possible to observe the various angles which between them the vortices above these various configurations can form. For geometries such as the delta, simple, double or ogival wings, the phenomena going with the appearance and the development of the vortical structures begotten by carried surfaces of feeble aspect ratio come relatively elementary, whilst they can become very complex at the time of the study of the configurations gathering a fuselage (warhead-cylinder), wings, ducks plans. . . The intent of this study is the optimization of the marine, air and terrestrial forms such as the veils, the wings of planes, the cars, the trains, the solar collectors (the shape of the baffles); optimization related to the stability of the fluids flow around these forms

Full textOne of the challenges facing the pharmaceutical industry in the post genomic area is the rational identification and inhibition of protein-protein interactions. The approach is to incorpor characteristics of privileged structure in the development of inhibitors. This strategy is employed with MCH-R1 antagonists as novel antiobesity agents and with proteins inhibitors of Bcl-2 families as therapeutic agents in cancer. Synthesis of privileged structures: phenyl substituted monocycles (pyridines, piperidines and piperazines) and derivatives of piperazino-azaindolines are described. In vitro inhibition is reported for each compounds, and the structure-activity relationship for each series is discussed. Privileged structures have been successfully exploited throught two different target families and promises to be an effective approach to the discovery and optimization of novel bioactive molecules

Full textThe pyrrolodiazepinones have interesting biological activities on various biological receptors, which makes them a prime target for developing new biologically active small molecules. A methodology in solution had been developed for synthesizing pyrrolo[3,2-e][1,4]diazepin-2-ones, which utilized the Pictet-Spengler condensation as the key reaction to form the diazepinone ring. Pyrrolo[3,2-e][1,4]diazepin-2-ones were found to mimic an inverse γ-turn conformation by X-ray crystallographic analysis. The methodology was subsequently implemented on three types of support: Merrifield resin, Wang resin and the soluble TAP support. The urotensinergic system plays a role in certain diseases of the cardiovascular system, such as hypertension, heart failure and atherosclerosis. The urotensinergic system is expressed in the circulatory system, excretory and central nervous systems and includes the endogenous ligands urotensin II (UII) and urotensin II-related peptide (URP), and the urotensin receptor UT. The ligands UII and human URP are composed of the respective amino acid sequences: H-Glu-Thr-Pro-Asp-c[Cys-Phe-Trp-Lys-Tyr-Cys]-Val-OH and H-Ala-c[Cys-Phe-Lys-Tyr-Trp-Cys]-Val-OH. The peptide UII is the most potent vasoconstrictor known to date. The two peptides have different biological effects and may exhibit distinct roles in certain diseases. Their common Trp-Lys-Tyr sequence is believed to play an important role in the activity of UII and URP, and has been suggested to adopt an inverse γ-turn conformation. Notably, the laboratory of Professor David Chatenet developed the UT receptor antagonist peptide urocontrin by replacing the Trp residue by biphenylalanine (Bip) in URP. A library of pyrrolo[3,2-e][1,4]diazepin-2-one analogs was thus designed to mimic the inverse γ-turn sequence and targeted against UT. The pyrrolo[3,2-e][1,4]diazepin-2-one library was designed based on the Trp-Lys-Tyr sequence of UII and URP, and Trp-Lys-Bip sequence of urocontrin. The synthesis of the pyrrolo[3,2-e][1,4]diazepin-2-one library was achieved on Wang resin. The side chain of Tyr was mimicked using tyramine, Lys and Orn were used as the basic amino acid component, and the side chain of Trp was replicated using biphenyl (as in urocontrin) 1-naphthyl and 2-naphthyl groups that were introduced by employing their respective aldehydes in a Pictet-Spengler reaction, which furnished unsaturated and saturated S- and R-pyrrolo[3,2-e][1,4]diazepin-2-ones. Evaluation of the biological activity of the pyrrolo[3,2-e][1,4]diazepin-2-ones on the UT receptor was performed in vitro and ex vivo. Tests in vitro measured binding in CHO-cells which expressed UT by employing hUII-125I as radiolabeled control. In rat aorta, ex vivo tests measured capacity to induce contraction, or modulate the contractions induced by hUII and URP. Certain R-pyrrolo[3,2-e][1,4]diazepin-2-ones caused an up to 50% reduction of the radioactive signal of hUII-125I. Pyrrolo[3,2-e][1,4]diazepin-2-ones exhibited little activity ex vivo; however, they modulated contractions induced by hUII and URP. For example, the saturated R-analog possessing lysine and a biphenyl side chain inhibited completely hUII-induced contractions of the aorta at 14 µM with a pKb of 5.54 at 4 µM, without influencing URP-induced contractions. Pyrrolo[3,2-e][1,4]diazepin-2-ones were selective for the urotensinergic system and inactive on the related receptor endothelin-1. Pyrrolo[3,2-e][1,4]diazepin-2-ones represent the first small molecules that can differently modulate the biological activities of UII and URP, and offer interesting potential as tools for studying the urotensinergic system.

Full textThe heterocycle chemistry is one of the most important fields in organic chemistry. Indeed, 65% of the literature in organic chemistry is about heterocycles. They are widely present in nature, essential to life and their applications are endless. Among them, the so-called privileged structures, attract particular attention. Privileged structures, of which quinolines and indoles are a part, play a crucial role in medicinal chemistry, for the production of libraries of therapeutic targets. Therefore, the development of new synthetic methods, simple, efficient and sensitive to environmental impact remains a challenge for chemists. Many synthetic methods require the use of o-acylated anilines, but these substrates are not easily available. The copper catalyzed addition reaction of a Grignard reagent on methyl anthranilate allows to form an o-acylated aniline, never reported before, with a quantitative yield. The latter will then serve as starting material for the synthesis of several privileged heterocycles including quinolines and indoles. The Friedlander reaction has been used for several centuries but suffers from the use of extreme reaction conditions which are harmful to the environment. It has therefore been modernized to give access to a series of eight trisubstituted quinolines, without solvent, strong acid or high temperatures. Furthermore, Heumann's process consists of the synthesis of indoles substituted in position by o-acylated anilines alkylation, followed by saponification and intramolecular cyclization steps. Patented by B.A.S.F. in 1895, this process quickly became obsolete due to low yields and difficult reaction conditions. Continuous flow chemistry has resurrected this process through the development of a more efficient and safer protocol. All in all, the work presented in this thesis contributes to the development of simple, effective methodologies, sensitive to environmental issues and leading to increasingly complex structures.
