Full textThe human vocal fold owns exceptional vibratory properties. It is capable of withstanding large deformations, for different types of loading, in a repeated and reversible manner. These particular vibro-mechanical properties are closely linked to its microstructure: a multi-layer complex structure composed of highly heterogeneous protein fibre networks. However, it is still difficult today to describe precisely the implication of the microstructural specificities of the fold in its biomechanical behaviour.In order to clarify this link and to move towards a better understanding of the behaviour of the vocal tissue, this study proposes to approach the problem under three complementary approaches, combining microstructural characterization, mechanical characterization and numerical modelling. First, the microstructure of the fold was studied emph{ex vivo} using an original technique based on X-ray tomography. The use of synchrotron tomography in phase retrieval mode has revealed the structure of the tissue at different scales. In particular, high-resolution 3D images of the fibrous structure of the upper and muscular layers of the tissue were acquired. These images gave rise to a quantitative 3D analysis of the fibrous arrangement, allowing the determination of descriptors of orientation and 3D geometry of the fibers.In a second step, the mechanical behaviour of the fabric under different loading conditions was studied. A protocol has been proposed to characterize the same sample in tension, compression and shear. These tests have complemented existing knowledge on fold biomechanics, and constitute important reference data for the construction and validation of digital models.Finally, based on the data acquired experimentally, a micro-mechanical model was developed. This model has the specificity to take into account the 3D arrangement of the tissue through an idealized but relevant representation of its fibrous microstructure. The macroscopic responses predicted for different loading conditionds could be compared to the experiment for validation. At the microscopic scale, the kinematics of the fibres during the loading could be simulated. The micromechanisms that occur during the deformation of the fibrous network could thus be identified, opening new perspectives in the understanding of the multi-scale properties of the tissue

Full textWe have examined a Safaniya Vacuum Re sidue (VR) and its fraction s (asphaltenes, resins, aromatics and saturates). Ln order to describe the macrostructure of these cornplex colloidal systems, we used different characterisation techniques : small angle X-ray and neutron scattering (SAXS and SANS), rheology and electron microscopy. Scattering techniques allows us to precise the model for asphaltenes and resins in solution. These macromolecules are polydisperse disk-like particles with thickness and diameter which are respectively close to 1 and 10 nm. The average molecular weight is equal to 106 000 for asphaltenes and 3 300 for resins. We have shown that SAXS is more sensitive to the scattering of the aromatic part of the molecule. Adding n-heptane induces first a desolvation of the molecules and then an aggregation up to f flocculation. These solutions present large heterogeneities due to concentration fluctuations. A huge difference in the chemical composition of the different asphaltene molecules can explain these fluctuations. These heterogeneities are stable as a function of temperature; this means that exist strong molecular interactions. Ultracentrifugation gives two different fractions which contain different chemical structures. More or less aromatic. Solutions, in good solvent, of these two fractions are homogeneous but a mixture of these two fractions exhibits heterogeneities. SAXS gives information about the structure of VR. We have observed the presence of large density fluctuations up to 300°C. Rheological measurements confirm threedimensional organisation

Full textTo study the topology of the active site of human CYP 2C8 and 2C9 we used the first RX structure of a membrane cytochrome P450, the rabbit CYP 2C5, which presents a strong homology with the human CYP 2Cs. First, a 3D model of CYP 2C9 was constructed by virtual mutagenesis of CYP 2C5. This work, completed by a study of CYP 2C9 mutants, underlined some CYP 2C9 residues, important for the interaction with substrates and the catalytic efficiency of the enzyme. In a second step, a 3D model of CYP 2C8 was developed. Meanwhile a pharmacophore of CYP 2C8 substrates was constructed. By confronting the results of those two approaches we proposed a model of the interaction of these substrates with CYP 2C8. Moreover, CYP 2C8 seems to be implicated in the regulation of vasodilatation. By studying the effects on CYP 2C8 of derivatives from the other ways of control of vasodilatation (the way of NO biosynthesis and the arachidonic cascade), we identified a few products able to inhibit CYP 2C8. Finally, we found CYP 2C5 substrates and inhibitors common to human CYP 2Cs. Those molecules were crystallised with CYP 2C5 by the team of E. F. Johnson (La Jolla). The RX structures obtained showed that the enzyme adapts its structure to the substrate. By comparison, they also gave information on the human CYP 2Cs

Full textDuring the past decade, a world-wide industrial expansion of nanosystems has been taking place at the junction between fundamental and applied sciences. Among these nanosystems, clusters, defined as small particles whose constitution ranges from few atoms (or molecules) to several thousand units, constitute "the missing link" between the isolated atom and the bulk solid. Studying the interactions in the aggregates would allow to understand how the atom is influenced by its neighborhood when it is included in a dense system within other atoms. "Nanoparticle" (NP) is usually defined as a system extanding from several thousands to millions of atoms, and its size is expressed in nanometer (nm, 1 nm = 10^{-9} m).This interest for nanosystems mainly results from their particular physical properties at the nanometer scale, but also from their large "surface to bulk" ratio. Hence, more active sites are available at the surface, enhancing their reactivity. This characteristic is used in numerous fields of applications (heterogeneous catalysis, cleanup, photovoltaics, nanomedicine).% and is also relevant in natural processes (atmospheric sprays, volcanic eruptions).This expansion of the "nanotechnological" activities involves the development of new methods for the characterization of these new kinds of materials, which take a considerable part in our daily life. Among the methods of matter characterization, synchrotron radiation based soft X-ray spectroscopy has shown to be a powerful technique for the study of atoms and molecules as well as materials, and appears today as an efficient technique of characterization in surface science.The idea of this thesis is to combine the atomic selectivity of the spectroscopy applied to dilute matter, with the surface sensitivity of soft X-ray spectroscopy, to study isolated nanosystems (clusters and NPs) in the gas phase, by XPS in the energy range of soft X-ray (typically 100 eV to 1000 eV). We present in this thesis the first results obtained by this approach at the PLEIADES beamline of SOLEIL synchrotron radiation facility. PLEIADES is a ultra-high resolution beamline dedicated to the study of dilute matter. The experiments carried out in this work relate to materials often used in the industrial and biomedical fields: TiO2 NPs, Silicon and glycosylated polystyrene NPs. We show how XPS on isolated NPs allows us to answer concrete questions, as the surface reactivity of the NPs, processes and oxidation dynamics, the doping efficiency or the interface ligand/NP. The results presented here not only show the feasibility of the method, but much more than that, prove that it is possible to give insight about surface chemical processes occuring on isolated, randomly oriented NPs

Full textOnly a few studies of (eco)toxicology linked the physico-chemical properties of nanoparticles to the toxicity mechanisms or the stress they induce. Moreover, no clear conclusions can be drawn at present because of the variability of nanoparticles used in studies. The present study used the inorganic Ge-imogolite nanotubes as a model compound. The toxic effects of length, number of walls, structural defects, and chemical composition were assessed towards the soil bacteria Pseudomonas brassicacearum. Several mechanisms modulating the toxicity of Ge-imogolite were then identified. Indeed, reactive sites at the tube ends induce a slight toxicity via a strong cell interaction and the generation of reactive oxygen species. Creating vacant sites on the surface of Ge-imogolite (ant thus increasing the number of reactive sites), appears to cause a deficiency of nutrients in the culture media correlated with a higher degradation of the tubes, leading to a high bacterial growth decrease. Finally, structural iron incorporation into Ge-imogolite transforms them into an iron source, being degraded and becoming growth promoters. Full textGlycosidases inhibitors are potential antiviral, antimetastatic, antidiabetes, antihyperglycemic or antiimmunostimulatory agents, whose design is a task of major interest in therapeutical chemistry. A series of glyconoamidines, recently prepared in our laboratory and displaying inhibitory potency against mannosidases, have been co-crystallized within the active site of a Golgi α-mannosidase II from Drosophila melanogaster, allowing a fine understanding of the interactions between the inhibitors and the protein residues. This initial structural study allowed us to design and synthesise glyconoamidines derivatives, more moderate inhibitors (micromolar range) but having a really improved selectivity against alpha-mannosidases, compared to the parent active compounds. These prepared derivatives possess a delta-amidine ring, with D-manno, D-gluco, D-xylo or D-lyxo configuration, or a gamma-amidine ring with D-arabino configuration. Other polyhydroxylated structures, based on a pyrido-oxazine scaffold, NJ and MNJ analogues, were also prepared. A new C2 isomerisation of glyconolactones promoted by magnesium iodide has also been developed. Studies conducted in deuterated medium showing the incorporation of a deuterium atom only on C2 position of the isomerized product reveal an epimerization rather than a racemization reaction of this position. A mechanism involving a chelation with magnesium species is proposed to explain this C2 inversion reaction
