Abstract:

The presence of an o-type cytochrome oxidase in the membranes of the denitrifying soil bacterium <i>Pseudomonas stutzeri</i> 224 was suggested by difference spectroscopy and by carbon monoxide binding spectra. A small amount of a d-type cytochrome was also detected. The sensitivities of the oxidase activities of the membranes of <i>P.stutzeri</i> to cyanide leads to the conclusion that cytochrome d is associated with NADH oxidase activity and cytochrome o is associated with ascorbate-TMPD oxidase activity. Pseudomonas stutzeri is evaluated in this thesis for its ability to form a coral, corrugated biofilm. It begins with an overview of biofilm formation in other species, then offers a more concentrated view on the pseudomonad group, to give context for this topic. Pseudomonas stutzeri has been shown to produce extracellular amylases of different specificity, as well as a carbohydrase which hydrolyzed oligosaccharide but not starch. Pseudomonas stutzeri NCIB 11359 and NRRL B-3389 were adapted to, and then cultured in five different growth media. Both culture collections showed similar patterns in cell morphology and glucosidase secretion. Abstract:

Pseudomonas stutzeri OXI utilizza come unica fonte di carbonio e di energia l’oxilene, il toluene, i cresoli ed alcuni dimetilfenoli (DMP). Pertanto il microorganismo potrebbe essere efficacemente utilizzato per interventi di biorisanamento di aree inquinate da composti di natura aromatica. Lo stadio iniziale dell’ossidazione di molecole aromatiche è operato dalla Toluene/o-xilene Monoossigenasi (ToMO), e dalla Fenolo ossidrilasi (PH), che costituiscono l’upper pathway della via degradativa. Le molecole prodotte sono poi convertite dagli enzimi del lower pathway in intermedi del ciclo di Krebs
