Abstract:

Broadcast encryption is a fairly new area in cryptology. It was first addressed in 1992, and the research in this area has been large ever since. In short, broadcast encryption is used for efficient and secure broadcasting to an authorized group of users. This group can change dynamically, and in some cases only one-way communication between the sender and receivers is available. An example of this is digital TV transmissions via satellite, in which only the paying customers can decrypt and view the broadcast.

The purpose of this thesis is to develop a general Java framework for implementation and performance analysis of broadcast encryption algorithms. In addition to the actual framework a few of the most common broadcast encryption algorithms (Complete Subtree, Subset Difference, and the Logical Key Hierarchy scheme) have been implemented in the system.

This master’s thesis project was defined by and carried out at the Information Theory division at the Department of Electrical Engineering (ISY), Linköping Institute of Technology, during the first half of 2004.

Abstract:
This study argues that a theory of the distinctively human Individual lies latent within Deleuze's readings of Hume and Bergson and his two major metaphysical treatises. This evolving theory derives from efforts to re-think the concept of 'the subject' In terms of 'difference', 'becoming', 'repetition' and 'event'. Using critical exegesis, the study shows that Deleuze's model is precise and workable, capable of supplanting discredited accounts of the subject and nullifying charges that Deleuze is an 'anti-humanist'. Deleuze's subject is neither pre-existent nor stable, but always in the process of becoming-other, Individuated by Inherent differences. Chapter 1 argues that Deleuze's account (and several theoretical resources) can be traced to an early engagement with empiricism, where he uses Humean atomism to define a field of difference 'within which' associationist psychological tendencies define the subject as a 'fiction'. As Chapter 2 shows, weaknesses in this model lead Deleuze to Bergson. Having adopted Bergsonlan Intuition as his method, Deleuze seeks after the preconditions of the flow and temporality of consciousness. He determines that the subjects constitutive moment is the virtual point of intersection between the physicality of material objects and the 'inner life' of consciousness. Chapter 3 turns to questions of ontology and ethics, arguing that Deleuze's theory of internal difference accounts for the role of contingent circumstances In subject-formation whilst his theory of the event establishes each lived moment as unique. Deleuze Interprets Nietzsche's eternal return as an ontological device entailing the recurrence of difference in the lived time of the subject's 'becoming', and as the means for coherence between the moments of a life. This theory leads Deleuze to an 'ethics of the event' with the goal of transforming human thinking from a concentration on unity and identity towards a more creative and fulfilling life of becoming.

Abstract:
Introduction and Aims: The progressive nature of heart failure (HF) is reflected by its complex pathophysiology, featured by imbalance of damaging and reparative factors. The overall aim was to assess the implication of endothelial (dys)function, monocyte subsets, different types of endothelial progenitors and plasma microparticles in subjects with HF. A special focus was an investigation of possible ethnic differences in these parameters. Methods: Parameters of vascular function, monocyte subsets, endothelial progenitors, and cellular microparticles were compared between South Asian subjects with systolic HF, and those with heart disease without HF and healthy controls. Ethnic differences in HF were assessed in three ethnic groups: South Asians, Whites, and African-Caribbeans. Additionally, leukocyte counts were compared between subjects with HF with reduced or preserved ejection fraction, whose outcome (mortality) was recorded during follow-up. Results: South Asian subjects with HF had significantly impaired micro- and macrovascular endothelial function, reduced levels of endothelial progenitors, and monocytes with reparative potential, but increased levels of microparticles. In HF patients, a high count of monocyte microparticles was associated with low ejection fraction. There were significant ethnic differences in characteristics of microvascular endothelial function, counts of CD14++CD16+ and CD14+CD16++ monocytes and monocyte-derived endothelial progenitors. On multivariate analysis, a high monocyte count was a significant predictor of death in HF with preserved ejection fraction unlike in those with systolic HF. Conclusions: Significant impairment of microvascular endothelial function is present in South Asian subjects with HF. High monocyte count is an independent predictor of death in HF with preserved ejection fraction. The value of the tested biological markers as therapeutic targets should be explored in future studies.

Abstract:
This thesis examines the role of affect, subjectivity, and intertextuality in the major works of Djuna Barnes (1892-1982). I propose a theory of Barnes’s modernist textuality based on the traumatic structure of belated understanding; specifically, I consider how remembering trauma involves a re-enactment and witnessing of something that, paradoxically, did not exist prior to that re-enactment and witnessing. This structural logic provides a suggestive metaphor for Barnes’s performative re-iterations of literary history that have been diversely categorized as parody, pastiche, and satire. I suggest that Barnes is a modernist ‘witness’ whose performance of literary history enables us to see that history as if for the first time. This structure of witnessing, dependent on a notion of non-dichotomous difference, exceeds the affective dimensions of the traumatic scenario and can even, I argue, be used to describe Barnes’s understanding of happiness and the pleasure of the modernist reader. My thesis contributes to the critical understanding of how modernism may be conceived in terms other than a simple repudiation or continuation of literary history. Barnes’s witnessing includes literary history in its broadest sense, as she performs the histories often rejected by high modernists for their engagement with bodies and feelings and their association with a feminized mass culture, such as the sentimental novels of the nineteenth century. Chapter 1 examines traumatic testimony and witnessing in Barnes’s 1958 play, The Antiphon. I consider how Barnes engages ideas of theatricality, ritual, and ‘antiphony’ to capture the performative and affectively complex nature of traumatic memory. I consider how Barnes’s emphasis on the importance of the witness figure relates to her own ‘initiated’ readership of the literature of the past. In Chapter 2 I consider Barnes’s representation of the affective ambivalence of childhood trauma, and her equally ambivalent treatment of the figure of the abusive father, in her 1928 novel, Ryder. I also suggest that, through Ryder, Barnes ‘witnesses’ the sentimental tradition, allowing us to see its full complexity and richness. In Chapter 3 I discuss the ambivalent role of shame in Nightwood (1936), using Silvan Tomkins’s theory of the structural dependence of shame on initial positive affect. I observe the traces of pleasure within scenes of shame in the novel, consider the text’s embarrassing place in the modernist canon, and suggest that the novel might be regarded as a ‘guilty pleasure’ of modernism. In Chapter 4 I consider the radically ubiquitous happiness of Ladies Almanack (1928). I suggest that the Almanack challenges Roland Barthes’s theories in The Pleasure of the Text by suggesting that happiness does not depend on novelty but can be understood as a dynamic reiteration of the past in the present. I also argue that Barnes turns to the logic of commodity and fashion culture to find a language for the pleasures of modernist textuality.

Abstract:
The North Caspian Basin (NCB) contains a significant number of major oil fields, some of which are yet to be put into production. The reason why some of these fields are not yet put into production is the exploration challenge that the NCB poses. In particular, the complex geological structure of this region makes it quite difficult to image its oil fields with conventional seismic techniques. This thesis sheds more light on difficulties associated with acquiring and processing seismic data in the NCB. The two central tools for investigation of these imaging challenges were the construction of a geological model of the NCB and the use of an accurate elastic wave-propagation technique to analyze the capability of seismic to illuminate the geological structures of the NCB. Using all available regional and local studies and my knowledge gained with oil companies, where I worked on subsalt and suprasalt 2D and 3D seismic data from the North Caspian Basin, I constructed a 2D elastic isotropic 10-by-6 km geological model of a typical oil field located on the shelf of the Caspian Sea in the southeastern part of the North Caspian Basin, which has the largest oil fields. We have propagated seismic waves through this model. The technique we used to compute wave propagation is known as the Finite-Difference Modeling (FDM) technique. Generating 314 shot gathers with stationary multicomponent OBS receivers that were spread over 10 km took two weeks of CPU time using two parallel computers (8 CPU V880 Sun Microsystems and 24 CPU Sun Enterprise). We have made the data available to the public. The dataset can be uploaded at http://casp.tamu.edu in the SEGY format. The key conclusions of the analysis of these data are as follows: - Combined usage of P- and S-waves allows us to illuminate subsalt reef, clastics and complex salt structures despite the 4-km overburden. - Free-surface multiples and guided waves are one of the key processing challenges in NCB, despite relatively shallow (less than 15 m) shelf water.

Rheumatoid arthritis (RA) is an autoimmune disease characterized by chronic inflammatory polyarthritis as consequence, at least in part, of a T cell-driven inflammation in the synovial membrane, frequently associated with the production of pro-inflammatory cytokines, contributing for ongoing inflammation. CXCR3 have been implicated in RA, since it is known its ability to modulate migration and in addition an increased concentration of its ligands has been reported in RA. Hence in this study it was evaluated the frequency and absolute number of circulating CD4+, CD8+ and γδ T cells, as well as the distribution between naïve, effector, central memory and effector memory based on the CD27 and CD45RA expression. Furthermore minor T cells subsets were characterized according to the expression of CD62L and CD28 combined with CD27 and CD45RA; additionally the expression of CXCR3 was assessed in all the studied subsets. The frequency and absolute number of CD8+ and γδ T cells were significantly decreased in RA patients; moreover CD4+effector memory and CD8+ naïve T cells were also decreased in frequency and absolute number in RA patients compared to healthy controls. In addition through the new T cells analysis, combining the expression of the CD45RA, CD27, CD28 and CD62L, it was found an impairment of five subsets within CD4+ and CD8+ T cells and nine subsets within γδ T cells in RA patients. Finally, the analysis of CXCR3 overall seems to indicate an increased expression in CD4+ and CD8+ T cells, conversely γδ T cells from RA patients showed lower CXCR3 expression. In conclusion, this study highlighted the importance of a detailed analysis of the T cell subsets through the combination of different phenotypic parameters. Our findings suggest an abnormal distribution of specific T cells subsets together with altered frequencies of T cell subsets expressing CXCR3 might contribute to a better knowledge of the migration pattern of these cells and therefore for the inflammatory status verified on RA patients.
A artrite reumatóide (AR) é uma doença autoimune caracterizada por poliartrite e inflamação crónica, resultado, pelo menos em parte, de uma resposta excessiva das células T na membrana sinovial associada a uma excessiva produção de citocinas próinflamatórias, que contribuem para a perpetuação da inflamação. O CXCR3 parece também estar envolvido na AR, por um lado pela sua capacidade de promover a migração, mas também porque têm sido descritos elevados níveis dos seus ligandos na AR. Neste sentido, o objetivo do presente estudo foi avaliar a frequência e o valor absoluto das células T CD4+, CD8+ e γδ, bem como a distribuição entre os compartimentos de células naïve, efectoras, memória central e memória efectora tendo por base a expressão do CD27 e do CD45RA. Adicionalmente propôs-se uma análise que incluiu a expressão do CD28 e do CD62L em combinação com o CD27 e o CD45RA. Avaliou-se ainda a expressão do CXCR3 nas diferentes subpopulações identificadas. Observou-se uma diminuição na frequência e no valor absoluto das células T CD8+ e das células T γδ nos doentes com AR, para além disso verificou-se também uma diminuição da frequência e do valor absoluto das células T CD4+ com fenótipo de memória efectora e das células T CD8+ naïve. Através da análise conjunta da expressão dos marcadores CD45RA, CD27, CD28 e CD62L identificou-se cinco subpopulações dentro das células T CD4+ e CD8+ e nove subpopulações nas células T γδ diminuídas nos doentes com AR. Relativamente à expressão de CXCR3, os resultados parecem apontar na sua globalidade para um aumento da expressão nas diferentes subpopulações de células T CD4+ e CD8+ dos doentes com AR, no entanto nas células T γδ dos doentes com AR observou-se uma menor expressão de CXCR3. Em suma, o presente estudo evidencia a importância de uma análise detalhada das subpopulações de células T com recurso à combinação de diferentes parâmetros fenotípicos. Os resultados parecem sugerir que as células T dos doentes com AR se encontram distribuídas de forma diferente entre os diferentes subtipos. Estas evidências em conjunto com o aumento da frequência de alguns subtipos celulares a expressar CXCR3 podem ajudar a perceber a migração das diferentes subpopulações de células T e o seu contributo para a destruição dos tecidos verificada nos doentes com artrite reumatóide.

s thesis is to analyze the place assigned to human subjectivity by French philosophers Michel Foucault and Gilles Deleuze. In order to fulfil the requirements of this objective, what is focused on is their shared critique which is exercised against the traditional conceptions of humanity and subjectivity. Through the thesis, first Foucault&rsquo
s analyses which demonstrate that universal man as a construction emerges as an effect of discursive practices and power relations, and his archaeological method that illustrates knowledge process is not dependent on transcendental consciousness are explained and discussed. Then it is argued that Deleuzian philosophy of becoming which does not submit to any transcendent unity that governs experience is an actual alternative to subject-centered understandings of the world. Throughout the course of arguments it is emphasized that according to both Foucault and Deleuze the human subject is an effect of network type relations that occur in a non-subjective fashion.

The epistemological inheritance of economics is 'rooted' in the enlightenment tensions over knowledge, thus demonstrating how the endeavour of economics is not a universal timeless objective science but a 'routed' body of knowledge whose underlying foundations are structured by the contingent emergence of ideas in a geohistorical-temporal-ideological context in line with a wider discursive fixing of objectivity and representation in knowledge. This modernist rendition of knowledge relies upon - an elision of difference; a separated view of the domains of the economic, political, social; a particular version of subjectivity which is narrowly obtained but unjustifiably universalised. A postcolonial moment in epistemology is needed to place difference at the heart of self and identity in order to disrupt knowledge based upon manufacturing conceptual abstractions and universalising their essence. One such intervention is the juxtaposition of identity with the economic. The problematics of identity in economics are discussed and the wider ways of attempting a reconciliation of the diversity of subjects with the desire for systematic knowledge are evaluated. A detailed critical assessment of economists' rare discourse on identity is followed by a differentiation of the concentric and the translational views on identity. Addressing the separation of culture and economy involves attending to the slippage between economics, economy, economic; rethinking the link between the value and values; and considering identity as a translation. Finally, writing economic theory another way is presented as a rewriting the conditions of theory itself. Implications of economic theories as textual productions are analysed and the complexities of emancipation and epistemology are explored. The dominant methods of economics do not have a universal purchase on understanding the economic aspects of human life. Overcoming the economic logic that permeates all aspects of existence and yet remains unquestioned in the terrains of knowledge production is essential.

Subject headings for works of fiction are one way to increase access to these books. Increased access should result in greater use for these books. Using a multi-branch, metropolitan public library, fiction books with subject headings were compared against fiction books without subject headings for overall use and circulations (use adjusted for time available to circulate). These same two categories were then compared to determine the affect of subject headings with respect to: fiction collection size, genre, and publication date for overall use and circulations.

Abstract:
This thesis describes a body of work undertaken to improve the power of surface-based analysis for anatomical MRI of the human brain. Chapters one and two introduce the topic and present a thorough background to the research. Chapters three through six describe the four main contributions of the t hesis. The first contribution was to increase the accuracy of mesh-based representations of the cortical grey/white and pial surfaces. The second contribution was to enhance the quality of between-subject cortical alignment using iterative strategies to build an unbiased group alignment template. The third was to develop a new technique for whole brain analysis of cortical position and surface area. The final contribution was to use multivariate statistical techniques to identify and parcellate sources of artefactual and anatomical variance. Taken together, these contributions constitute a significant improvement to the power of surface-based analysis of inter-subject differences in anatomical MRI.
Cette thèse décrit un ensemble de travaux entrepris pour améliorer l'efficacité de l'analyse de surface du cerveau humain par IRM. Les chapitres un et deux introduisent le sujet et présente un contexte approfondi à la recherche. Les chapitres trois à six décrivent les quatre principales contributions de la thèse. La première contribution a été d'accroître la précision des représentations à base de mailles des surfaces grise/blanche et de la pie-mère. La deuxième contribution a été d'améliorer la qualité de l'alignement cortical entre-sujet en utilisant des stratégies itératives pour construire un modèle d'alignement de groupe impartial. La troisième a été de développer une nouvelle technique d'analyse de la position et de la surface corticale du cerveau complet. La dernière contribution a été d'utiliser des techniques statistiques multivariées pour identifier et parcelliser les sources de variance artéfactuelle et anatomique. Prises ensemble, ces contributions constituent une amélioration significative de l'efficacité de l'analyse de surface basée sur les différences inter-sujets par IRM anatomique.

Abstract:
Nesse trabalho foi realizada uma avaliação microestrutural da influência do ciclo térmico em três aços da classe API5LX80: Nb-Cr, Nb-Cr-Mo e Nb-Cr-Mo-V fabricados no Brasil. Estes aços foram fabricados através de processo termo-mecânico controlado sem passarem por uma etapa subseqüente de resfriamento acelerado. Foi realizada uma caracterização microestrutural em duas condições, pós- laminação e austenitização a 900oC seguida de três (3) ciclos térmicos: resfriamento ao ar (1,5oC/s), ao óleo (30oC/s) e a água (115oC/s). Na condição de pós-laminado, os aços Nb-Cr, Nb-Cr-Mo, e Nb-Cr-Mo-V não apresentaram diferenças significativas quanto ao TG, distribuição de fases (ferrita, bainita e AM), dureza e microdureza mas foram verificadas diferenças em suas subestruturas (subgrãos, células e precipitação fina) observada em MET. Concluise que a inclusão de Mo e V nos aços Nb-Cr-Mo e Nb- Cr-Mo-V não ocasionou influência a nível microestrutural, mas ocasionou mudanças a nível subestrutural verificadas em MET que talvez possam justificar a diferença entre propriedades mecânicas.Verificamos a diminuição do tamanho de grão com o aumento da taxa de resfriamento nos aços Nb- Cr, Nb-Cr-Mo, e Nb-Cr-Mo-V. Os valores de dureza e microdureza são os mesmos para os aços Nb-Cr e Nb-Cr-Mo na condição de após aplicação dos ciclos térmicos, e uma associação pode ser feita com a quantidade de fases de cada um, pois a distribuição quantitativa das fases para ambos os aços é muito similar. O aço Nb-Cr-Mo-V apresenta uma distribuição diferente, com a formação da fase martensítica já no resfriamento ao óleo (30oC/s) e o aumento da quantidade desta fase a medida que a taxa de resfriamento diminui.
The present work presents a microstructural evaluation of three different chemical compositions of API5LX80 classified as Nb-Cr, Nb-Cr-Mo e Nb-Cr- Mo-V steel in two conditions, as received and after three applied cooling rates: air-cooled (1,5oC/s), oil-cooled (30oC/s) e water- cooled (115oC/s). This material under study is a microalloyed HSLA steel used for pipeline purposes produced in Brazil. Usually this steel is processed by thermo- mechanical controlled hot rolling followed by accelerated cooling. However, the material under study was not accelerated cooled after thermo-mechanical controlled hot rolling. Instead, chemical composition was changed to obtain similar mechanical properties of this class of steel. The Nb-Cr, Nb-Cr-Mo, e Nb-Cr-Mo-V as-received steels studied in this work presented the same quantitative distribution of grain size and volumetric fraction of the phases (ferrite, bainite and MA), as well as, the same hardness Rockwell (HRB) and Vickers (HV100). However, the substructures (subgrains, cells and fine precipitation) of the three steels were different when examed in Transmission Electron Microscopy - TEM. The Nb-Cr, Nb-Cr- Mo, e Nb-Cr-Mo- V steels, after submitted to the three cooling rates, presented a decrease in grain size with the increasing of the cooling rate. The same hardness Rockwell (HRB) and Vickers (HV100) values were found for Nb-Cr e Nb- Cr-Mo steels and this fact can be associated to the volumetric fraction of both steels, because both have very similar phase distribution. In Year 9, when boys and girls are expected to make choices regarding what they want to become when they grow up, many take a crucial decision to drop or side-line IT as an academic subject, which in turn steers them away from a possible future IT career. This thesis examines the reasons why IT careers are not well-imagined or popular amongst teenagers at this critical time of their lives. Taking the widely acknowledged ‘women in IT' problem as a starting point, it focuses specifically on gender differences that exist in relation to how teenagers form their ideas about IT as an academic subject, as a possible career and in everyday life. 79 boys and 85 girls participated in this study from a mixture of 12 state-maintained and nine independent secondary schools (single-sex and co-educational) in Southeast London Borough. This research was exploratory and used an age-appropriate, participatory and mixed-methods framework incorporating: a questionnaire, a creativity map exercise, group and individual interviews, mini-focus groups, and observations. During the interviews, students were also provided with information and opportunities regarding IT careers. I argue this has been of benefit to the students as well as the research, as it has prompted them to think about a career they previously had not even considered. The findings of my study indicate boys were more likely than girls to say that they liked and enjoyed IT/ICT1 as a subject and would consider IT as a career choice for the future. Evidence throughout the study does not suggest girls lack confidence with regard to their general engagement with and use of technology, compared to the boys. Rather, the findings suggest more needs to be done in the area of role models, mentors and careers advice to inform more girls (and boys) about IT careers. The thesis concludes with recommendations for further research, especially in light of the new computing curriculum, which commenced in September 2014.

Group 1 major urinary protein (MUP) genes can be divided into subgroups named Mup-1.1A, Mup-1.1B, Mup-1.1C and Mup-1.1D. Allelic genes in Balb/c and C57BL/6 inbred strains of mice are referred to as Mup1.1A{2/} and Mup-1.1A{b/} respectively. Expression of MUP genes in the liver is sexually dimorphic, however, the relative expression in males and females varies between subgroups and between allelic genes within a subgroup. Genes from two subgroups, Mup-1.1A and Mup-1.1C code for proteins which are synthesized at higher levels in male than female liver. In contrast, genes from two other subgroups, Mup-1.1B and Mup-1.1D code for proteins synthesized in male liver but undetectable in female liver. The Mup-1.1C subgroup contains two single copy allelic genes, Mup-1.1C{a/} and mup-1.1C{b/}. In C57BL/6 expression of Mup-1.1C{b/} in the liver of males is twice as high as females, whereas in Balb/c, expression of Mup-1.1{Ca}/ in males is seven to twenty times as high as females. Several genes in the Mup-1-B subgroup differ between Balb/c and C57BL/6 mice, however, expression in the liver of these genes is the same in both strains and is approximately one hundred times higher in males than females. In the livers of growth hormone (GH) deficient lit/lit C57BL/6 mice MUP mRNA levels are approximately one hundred fold lower than in phenotypically normal lit/+ C57BL/6 males. The sexually dimorphic pattern of MUP expression is also absent. Administration of GH to male lit/lit mice by one regime caused a large increase in the levels of particular MUP mRNAs in the liver, whereas administration by another regime did not. Altering the GH regime affected some MUP mRNAs more than others.

This research explores sex and discipline-area differences in empathy across theInterpersonal Reactivity Index developed by Davis. A descriptive cross-sectionalstudy was performed with the participation of 680 university students, divided into351 women and 329 men belonging to Physics-Mathematics, Social-Humanisticand Biological-Chemistry areas. The results show sex differences in threedimensions of the inventory with higher scores for women. No differences inrelation to the subject area of the students were observed. DNA methylation is involved in various biological phenomena, and its dysregulation has been demonstrated as being correlated with a number of human disease processes, including cancers, autism, and autoimmune, mental health and neuro-degenerative ones. It has become important and useful in characterising and modelling these biological phenomena in or-der to understand the mechanism of such occurrences, in relation to both health and disease. An attempt has previously been made to map DNA methylation across human tissues, however, the means of distinguishing between methylated, unmethylated and differentially-methylated groups using DNA sequence features remains unclear. The aim of this study is therefore to: firstly, investigate DNA methylation classes and predict these based on DNA sequence features; secondly, to further identify methylation-associated DNA sequence features, and distinguish methylation differences between males and females in relation to both healthy and diseased, sta-tuses. This research is conducted in relation to three samples within nine biological feature sub-sets extracted from DNA sequence patterns (Human genome database). Two samples contain classes (methylated, unmethy-lated and differentially-methylated) within a total of 642 samples with 3,809 attributes driven from four human chromosomes, i.e. chromosomes 6, 20, 21 and 22, and the third sample contains all human chromosomes, which encompasses 1628 individuals, and then 1,505 CpG loci (features) were extracted by using Hierarchical clustering (a process Heatmap), along with pair correlation distance and then applied feature selection methods. From this analysis, author extract 47 features associated with gender and age, with 17 revealing significant methylation differences between males and females. Methylation classes prediction were applied a K-nearest Neighbour classifier, combined with a ten-fold cross- validation, since to some data were severely imbalanced (i.e., existed in sub-classes), and it has been established that direct analysis in machine-learning is biased towards the majority class. Hence, author propose a Modified- Leave-One-Out (MLOO) cross-validation and AdaBoost methods to tackle these issues, with the aim of compositing a balanced outcome and limiting the bias in-terference from inter-differences of the classes involved, which has provided potential predictive accuracies between 75% and 100%, based on the DNA sequence context.

To investigate the mechanism and functional significance of infection of CD8 lymphocytes by human immunodeficiency virus type 1 (HIV-1) in vivo, frequencies of infection and genetic relationships between HIV-1 variants infecting native (CD45RA+) and memory (CD45RO+) CD4 and CD8 lymphocytes was carried out. CD4 and CD8 lymphocytes were purified from peripheral blood mononuclear cells from 16 study subjects by negative selection, followed by positive selection of each of the two subsets with anit-CD45RA and -RO monoclonal antibody-coated beads. The contribution to total proviral load by the naive and memory subsets was estimated by quantitative PCR combined with measurement of their frequency in total T lymphocytes. In this analysis we found that HIV preferentially infects the naive subset of CD8+ lymphocytes along with the naive and memory populations of CD4+ cells. Sequence comparisons of the V1/V2 and V3 region of the envelope gene was also carried out on the naive and memory subsets of CD4 and CD8 cells. Variants infecting CD8 lymphocytes were partially or completely genetically distinct in the V3 region from those recovered from CD4 lymphocytes, and showed a greater degree of compartmentalisation than observed between naive and memory subsets of CD4 lymphocytes. A preferential distribution of syncytium-inducing, CXCR4-dependnet variants was also observed within CD4 lymphocytes. Even more marked sequence differences were observed upon sequence comparison of the V1/V2 hypervariable region, with some evidence for recombination between V1/V2 and V3 regions. Population differences may have originated through different times of infection rather than necessarily indicating a difference in their biological properties. In this study we provide evidence for the widespread infection of CD4 and CD8 naive and memory cell populations within the peripheral blood of HIV seropositive individuals, indicating that HIV has a broader tropism for cell types in vivo than described previously. The preferential distribution of HIV-1 in naive CD8 lymphocytes suggests that infection occurred early in T lymphocyte ontogeny, such as during maturation in the thymus. Destruction of cells destined to become CD8 lymphocytes may be a major factor in the decline of CD8 lymphocytes frequencies and function on disease progression, and contribute directly to the observed immunodeficiency in AIDS.

In this study, four calibration experiments were conducted for the DEM validation: filling of a model silo, confined compression in a cylinder, rod penetration into a granular bulk and silo discharging through an outlet. DEM simulations were conducted using two commercial software: PFC3D and EDEM, the latter being the DEM software that originated from Edinburgh University. A set of 8 benchmark tests were carried out to validate the codes and to evaluate the fundamental aspects of DEM. Following that, a large number of DEM simulations were conducted and comparisons between DEM simulations and experiments were made. For corn grains: the study shows that 4-sphere representation together with the measured corn properties produced satisfactory match with experiments for silo filling (normal wall pressure distribution), confined compression (normal wall pressure distribution, load transfer to boundary surfaces, and silo design parameter K and m), rod penetration (force-displacement response) and silo discharge (mass flow rate and angle of repose). For glass beads: the DEM simulations also gave good agreement with experiments for silo filling (normal wall pressure distribution), confined compression (normal wall pressure distribution, load transfer to boundary surfaces), rod penetration (force-displacement response) and silo discharge (mass flow rate). These findings provide sound verification that DEM is capable of producing quantitative predictions of the problems studied. They also suggest that very accurate representation of the non-spherical particle shape may not be necessary to produce satisfactory predictions and capturing the linear dimensions of a particle may be adequate. Two DEM results that produced larger discrepancies with experiments are filling density (~17% lower for corn grains and ~8% lower for glass beads) and loading stiffness (stiffer response). Plausible explanations for these are given in this thesis, which should be explored further.

It describes the extent of the debates and the role of British law that arose after conflict between Aboriginal people and settlers in the context of political and economic contests between settlers and government on land issues. While the British government continually maintained that the legal basis for annexation was settlement, by the mid 1830s Stirling regarded it as an ‘invasion,’ but was neither prepared to accept that Aboriginal people had to consent to the imposition of British law upon them, nor to formally recognise their rights as the original owners of the land. Instead, Stirling’s government applied an archaic form of outlawry to Aboriginal people who resisted the invasion. This was despite proposals for agreements in the 1830s. During the early 1840s there was a temporary legal pluralism in Western Australia where Indigenous laws were officially recognised. However, by the mid 1840s the administration of British law in Western Australia was increasingly dictated by settler interests and mounting settler-magistrate pressure to modify the legal position of Aboriginal people which resulted in the development of colonial law to construct a landless subject status with minimal rights based on their value as a useful labour force for the pastoral economy. This separate legal status deliberately departed from ‘equality’ principles and corresponded with the diminished status of Indigenous laws and the abandonment of legal pluralism in settled districts, during a period of rapid pastoral expansion in the 1850s. This entrenched discriminatory practice in colonial law would be the prelude to the ‘protectionist’ and discriminatory legislation of the early twentieth century which formalised inequality of legal status.

The development of insecticides resistance among many insect species and the ecological damage occasionally caused by the lack of specificity in the toxic effects of insecticides have provided the impetus to seek alternative methods of insect control. This observation led to the development of bioinsecticides based on the insecticidal action Bacillus sphaericus (Bs), Bacillus turingiensis (Bt). The discovery of biolarvicidal actions of Bacillus thuringiensis and Bacillus sphaericus opened a new perspective for insect control. In the first part of the study was initiated to determine a suitable fermentation medium formulation and optimal fermentation conditions for large scale, low cost production of Bs. Bs 2362 was tested in whey and soy flour based media. These media was reformulized form of NYSM (Nutrient Broth Yeast Extract Sporulation Medium). Soy flour based medium, SYSM, gave the promising results in terms of cell yield, sporulation frequency and toxin production. In the second part of the study, fermentation productivity anlaysis of a local isolate Bacillus thuringiensis subsp. kurstaki 81 was evaluated. In order to compare different C:N ratios (1:1, 2:1, 4:1, 8:1, 10:1 20:1 and 30:1) of YSM medium. Btk 81 were run for 72 h and cell growth, sporulation and toxin protein profile of Btk 81 were determined for each. When all the quantitative toxin data for both glucose and sucrose varying C:N ratios were compared, it was determined that the crystal protein concentrations had the highest value in sucrose based medium when C:N ratio was 10:1. Regulation by C:N ratio of crystal protein biosynthesis was investigated for improving the production of this protein by our third candidate strain Bacillus thuringiensis subsp. israelensis ONR60. The experiments were performed by using TBL medium, at three different C:N ratios, 2:1, 4:1 and 8:1 respectively. In view of the cell growth characteristics and bioassy results, TBL medium designed with 2:1 C:N ratio was chosen as the best for further steps. In addition, running time of the culture determined as 60 hours as was also determined in the previous experiment. As the last step of this study, the pre-determined optimal conditions were applied to a 30L batch type fermentor for toxin production by using Bacillus thuringiensis subsp. israelensis ONR60. Unfortunately, the toxicity was not satisfactory, being much below the level of that expected as based on the results of the laboratory scale studies.

Abstract:
Running performance has increased immensely during the last few years, coinciding with multiple shattered world records in relatively short amount of time. Improvements in footwear material and design are likely reasons for this increase in running performance. Previous studies on the effect of footwear on running economy (RE), a determinant of running performance, have not included participant-blinding. Furthermore, they have yet to compare multiple carbon-fiber plated running shoes available for purchase, what differences there are across price ranges and shoe categories, and if there is such a thing as a placebo-effect. Aim: (1) Descriptively compare a set of heterogeneous running shoes, with regards to running economy, Foot Strike Type (FST), vertical oscillation, ground contact time, stride length and cadence; including (2) a ‘sham’ and ‘normal condition’ of the same running shoe model; and (3) explore the participant’s perception of the study-specific blinding protocol. Method: A Single Subject Experimental Study (N=1), comparing nine different shoe conditions using a crossover design. The assessment of RE was conducted using indirect calorimetry with mixing-chamber in a climate-controlled facility. Spatiotemporal parameters were assessed using a Garmin HRM-Run™, and foot strike type was visually assessed using a frame-by-frame approach based on 2D-video at 240 fps. Results: The average running economy across all shoe tests varied between 16.02 to 17.02 W/kg, with the ‘worst’ shoe costing 6.24% W/kg more than the ‘best’ shoe. The descriptive difference between the ‘sham’ and ‘normal condition’ were negligible and within the range of measurement error. Spatiotemporal parameters were overall descriptively similar between the shoes, with a few outliers who differed with regards to measure of central tendency or dispersion. FST differed between the shoes including the ‘sham’ and ‘normal condition’, but were overall consistent with the participant’s habitual FST. The study-specific blinding procedure was perceived to work well, but may also be improved in some remarks. Conclusion: Descriptive difference in some, but not all, physiological and biomechanical parameters were observed between the shoe conditions in this study, including the ‘sham’ and ‘normal condition’. Blinding procedures in experimental footwear research may be feasible and adopted with future studies.

Abstract:
This essay investigates the feminist philosophy of Rosi Braidotti with particular focus on the alternative feminine feminist nomadic subject that she creates. I also introduce Braidotti’s theoretical inspiration from Gilles Deleuze and Luce Irigaray. I argue that Braidotti creates an alternative figuration for feminism through synthesizeing Deleuze’s concept of ”becoming” with Irigaray’s sexual difference-theory. Braidotti highlights the importance of understanding the concept of difference differently. She also argues for the difference between subjectivity and identity, for the materialistic foundation of the subject, for the fundamental asymmetry between the sexes and for the nomadic mode of thinking. Braidottis suggests that the way out of the phallogocentric system, which she means we are encapsulated in, consist in working through the images that patriarchy has produced of women, through mimetic repetitons, in order to create new representations of women. I argue that the feminist philosophy of Braidotti is both humble and subversive which makes it an interesting and useful alternative for everyone who is interested in alternative, complex and thrilling ways of theorizing female feminist subjectivity.

BACKGROUND:In addition to their well-documented ocular therapeutic effects, glucocorticoids (GCs) can cause sight-threatening side-effects including ocular hypertension presumably via morphological and biochemical changes in trabecular meshwork (TM) cells. In the present study, we directly compared the glucocorticoid receptor (GR) potency for dexamethasone (DEX), fluocinolone acetonide (FA) and triamcinolone acetonide (TA), examined the expression of known GRalpha and GRbeta isoforms, and used gene expression microarrays to compare the effects of DEX, FA, and TA on the complete transcriptome in two primary human TM cell lines.METHODS:GR binding affinity for DEX, FA, and TA was measured by a cell-free competitive radio-labeled GR binding assay. GR-mediated transcriptional activity was assessed using the GeneBLAzer beta-lactamase reporter gene assay. Levels of GRalpha and GRbeta isoforms were assessed by Western blot. Total RNA was extracted from TM 86 and TM 93 cells treated with 1 muM DEX, FA, or TA for 24 hr and used for microarray gene expression analysis. The microarray experiments were repeated three times. Differentially expressed genes were identified by Rosetta Resolver Gene Expression Analysis System.RESULTS:The GR binding affinity (IC50) for DEX, FA, and TA was 5.4, 2.0, and 1.5 nM, respectively. These values are similar to the GR transactivation EC50 of 3.0, 0.7, and 1.5 nM for DEX, FA, and TA, respectively. All four GRalpha translational isoforms (A-D) were expressed in TM 86 and TM 93 total cell lysates, however, the C and D isoforms were more highly expressed relative to A and B. All four GRbeta isoforms (A-D) were also detected in TM cells, although GRbeta-D isoform expression was lower compared to that of the A, B, or C isoforms. Microarray analysis revealed 1,968 and 1,150 genes commonly regulated by DEX, FA, and TA in TM 86 and TM 93, respectively. These genes included RGC32, OCA2, ANGPTL7, MYOC, FKBP5, SAA1 and ZBTB16. In addition, each GC specifically regulated a unique set of genes in both TM cell lines. Using Ingenuity Pathway Analysis (IPA) software, analysis of the data from TM 86 cells showed that DEX significantly regulated transcripts associated with RNA post-transcriptional modifications, whereas FA and TA modulated genes involved in lipid metabolism and cell morphology, respectively. In TM 93 cells, DEX significantly regulated genes implicated in histone methylation, whereas FA and TA altered genes associated with cell cycle and cell adhesion, respectively.CONCLUSION:Human trabecular meshwork cells in culture express all known GRalpha and GRbeta translational isoforms, and GCs with similar potency but subtly different chemical structure are capable of regulating common and unique gene subsets and presumably biologic responses in these cells. These GC structure-dependent effects appear to be TM cell-lineage dependent.

A composição química de cortiça, floema e xilema foi determinada em árvores jovens de Quercus suber L. de três proveniências (Alcácer do Sal, Azeitão e Santiago do Cacém) de uma importante região de produção suberícola em Portugal, tendo em vista a análise de eventuais diferenças entre proveniências. Foram estudadas três árvores por proveniência, determinando-se o conteúdo em cinzas, extractivos (solubilizados em diclorometano, etanol e água), suberina (no caso da cortiça), lenhina e polissacáridos. Os três tecidos mostraram grandes diferenças na sua composição química. A composição química média da cortiça foi a seguinte: 0,66 % em cinzas, 11,7 % extractivos, 42,3 % suberina, 24,1 % lenhina e 16,2 % polissacáridos; do floema 2,9 % cinzas, 4,5 % extractivos, 38,0% lenhina e 49,1 % polissacáridos; e do xilema 1,1 % cinzas, 5,6 % extractivos, 23,4 % lenhina e 64,6 % polissacáridos. A análise estatística mostrou que a proveniência apenas foi um factor de variação significativo para os extractivos em etanol no caso da cortiça e os polissacáridos no floema. A composição monomérica da lenhina de todas as amostras foi analisada por pirólise analítica a 650 ºC. A lenhina dos três tecidos difere substancialmente: o rácio S/G foi 0,12 na cortiça, 1,1 no floema e 2,3 no xilema. Os compostos obtidos por pirólise foram também identificados e, sempre que foi possível determinar a sua origem, agrupados em açúcares, lenhina e suberina (apenas nas amostras de cortiça). Os açúcares representaram 58,6 %, 63,1 % e 25,4 %, a lenhina 14,4 %, 10,4 % e 12,6 % respectivamente no floema, xilema e cortiça, e a suberina na cortiça representou 33,0 % do total dos picos dos pirogramas

Abstract:
This study explores the nature of writing and the sorts of presence that writing gives us access to. This understanding of writing includes not only all speaking and all writing in the narrow sense of marks on a page, but goes beyond this to include the sense in which Derrida uses the term ‘writing’ in Of Grammatology, to mean a broad and complex process of the construction of textual traces or presences necessarily brought about through the structural mechanism of difference inherent in the writing process (Derrida, 1997). This study argues that writing is a system that creates Subjects or selves as the writing happens. It suggests that writing is a remarkable site from which to explore the construction of selves, because it gives us access to (partially) identifiable presences, in the apparent absence of the writer. It goes on to demonstrate that this identity can be distinguished through written traces of difference left for the reader to decipher, by analysing different aspects of the plot and writing devices in Peter Greenaway’s film The Pillow Book and in Kafka’s short story In the Penal Colony. These two texts are considered particularly relevant to this study, in that they both explicitly deal with the contradictory nature of writing and how it relates to the Being (there or the contextualised Being of Dasein) and being (in general), the life and death, the empowerment and destruction of the Subjects that writing sets up. Both texts explore salient aspects of writing on the human body. The study uses these texts as a platform for speculation about the kind of presence that can be traced through writing, and proposes that the written Subject is multiple, contradictory and reflexive, connected and related, and that it is impermanent and has a deferred presence. Finally, this written Subject is also explored in the context of Foucault’s expositions of the self in texts such as Technologies of the self (Foucault, 1994) and ‘What is an Author?’ (Foucault, 1977) in answer to his question Who are we in the present, what is this fragile moment from which we can’t detach our identity and which will carry our identity away with itself? The concept of desire has been central to most recent philosophical debates, in various forms and styles. I have argued in the present study that, one of the main motivations for this apparent interest in the concept of desire is the result of the increasing awareness of the shortcomings of those presuppositions revolving around an &ldquo
autonomous subject&rdquo
, &ldquo
transcendence&rdquo
, &ldquo
representation&rdquo
, and &ldquo
moral subjectivity&rdquo
. Desire, in this vein, is conceived and put into practice by the traditional philosophy as one among the other attributes that cannot be considered without reference to man. Desire as such is conceived as something that is necessarily controlled and managed by reason. Ethics and politics, in terms of these ill-conceived presuppositions, are narratives erected upon this tension that necessarily refers to a self-conscious subject and her subversive desires. I argue, in this study, for the possibility of imagining other variants of desire, i.e., something other than traditionally established debates, where desire is no longer conceived in strict reference to human beings. These novel accounts, which I will attempt to uncover, hope, will help us see in what ways desire can be considered within the concept of pure immanence and the realm post-humanist ethico-politics. Spinoza, Nietzsche and certainly Deleuze and Guattari are on this side. Desire, according to this non-tradition, belongs to immanence. In arguing for the legitimacy of two affirmative notions of desire, namely, that of immanent desire and embodied desire, I tried to establish a continuity between immanence (totality of bodies and constant differing) and embodied desire (singular intensities), and by means of which I have drawn attention to the importance of a new vision of ethics and politics that might work, not through the already established form of subjectivities, but through new forms of individuation and flow-like encounters of bodies.
