Abstract:
This study aimed to develop a superovulation protocol for the spiny mouse (Acomys cahirinus). The spiny mouse is a desert-adapted rodent species, with a long oestrus cycle (11 days) compared with rat and mouse, and gives birth to few (mean litter size is 3) precocial offspring after a relatively long gestation (39 days). We successfully optimised a superovulation protocol that elicited a 5-fold increase in the normal ovulation rate of this species. To induce superovulation in the spiny mouse 2 injections of equine chorionic gonadotrophin (eCG, 10 IU each), 9 h apart, were required, followed by

To limit the number of times the bison were handled, we tested a two-injection superovulation scheme using sodium hyaluronate (MAP-5, Bioniche Animal Health Inc., Belleville, Ontario, Canada) in the FSH diluent, which serves to slow the absorption o

Abstract:
SUMMARY A superovulatory and surgical protocol was developed for recovery of bovine zygotes. Holstein cows and heifers were given follicle-stimulating hormone and cloprostenol to induce superovulation. Surgical cannulation and lavage of the uterine tube was performed 40 to 48 hours after the start of standing estrus. In general, cows had more corpora hemorrhagica than did heifers, but a higher percentage (P < 0.05) of ova recovered from cows were infertile. Several heifers were subjected to the procedure twice, and embryo recovery rates were equivalent both times.

The administration of sustained-release FSH-MAP5 in a 2-injection protocol has been shown to be as effective as a multiple-FSH injection protocol in inducing superovulation in cattle (Bo and Mapletoft 2014 Theriogenology 81, 38) and in sheep (Fry et al. 2016 Reprod. Fertil. Dev. 28, 250); however, the effect on embryo quality in the latter experiment was unclear. The following experiment further investigated the effect of FSH-MAP5 on ovulation rate and embryo quality in a sheep multiple-ovulation embryo transfer (MOET) program conducted in the breeding season. The appropriate dosage regimen for equine FSH (eFSH) (dose, dosing interval) administration in a superovulation treatment in pony mares was determined by a kinetic approach using production rates and kinetic parameters of elimination of the hormone. Two dosage regimens were then tested in superovulation protocols. The eFSH production rates were determined by sampling four ovariectomized pony mares every 10 min for 8 h during the breeding season. Kinetic parameters were determined by administering four dose levels of a preparation of eFSH (4.4, 8.8, 17.6 and 35.2 micro g/kg) by the i.v. route t

The feasibility of application of a standardized protocol for estrous synchronization, superovulation and embryo transfer in young pubertal and pre-pubertal gilts under farm conditions was investigated. Two groups of Large white X Landrace crossbreed gilts were used. Multiple ovulation (superovulation) and embryo transfer has been used extensively in cattle. In the past decade, superstimulatory treatment protocols that synchronise follicle growth and ovulation, allowing for improved donor management and fixed-time AI (FTAI), have been developed for zebu (Bos indicus) and European (Bos taurus) breeds of cattle. There is evidence that additional stimulus with LH (through the administration of exogenous LH or equine chorionic gonadotrophin (eCG)) on the last day of the superstimulatory treatment protocol, called the ‘P-36 protocol’ for FTAI, can increase embr

Following the performance of a superovulation protocol, multiple nodules were observed bilaterally in the uterine horns of 31 of 276 (11.2%) C57BL/6 J female mice aged 8.5 ± 0.6 (mean and standard error of mean) weeks. These lesions prevented embryo collection, and the uterine decidual reaction was suspected. Insemination of superovulated bovine donors in due time is of central importance for fertilization and embryo viability. A preliminary test focusing on LH surge detection during superovulation (unpublished datas) indicated that one quarter of the donors present LH surges 12 to 24 h before heat observation (which could correspond, in the case of AI after heat observation, to post ovulation AI). Therefore, it was hypothesised that the average number of embryos per flush could be improved by inseminating donors with early LH surge 12 h after the beginning of the LH peak whenever the heat occurs.I

Background: High dose gonadotropin stimulation protocols may increase costs, patient discomfort, likelihood of ovarian hyper stimulation syndrome (OHSS), ovarian torsion and increased bleeding after multiple ovarian punctures. In addition, it is possible that high dose stimulation may result in impaired oocyte “quality” with adverse effect of embryonic ploidy. This may negate any benefits seen from a higher oocyte yield after high dose stimulation. Aim: To determine the proportion of euploid blastocysts resulting from higher versus lower dose ovarian stimulation protocols. Method: Multicentre

The conventional superovulation treatment using follicle-stimulating hormone (FSH) comprises 2 intramuscular injections per day for 3 to 4 days. This conventional method requires frequent handling of donors—which may cause increasing stress in donors—and high labour costs. Therefore, to alleviate these issues, a treatment protocol using a single FSH injection under on-farm conditions is needed. The objective of this study was to evaluate the feasibility of the single subcutaneous FSH injection protocol (Sugawara et al. 2017 Reprod. Fertil.
