Abstract:
Many bio-pharmaceutical production processes are based upon the use of mammalian cell lines, such as Chinese hamster ovary (CHO) cells, capable of proliferation as single cells in suspension in a synthetic environment. Routine use of CHO cells as production vehicles requires a lengthy “adaptation” process from the wild-type adherent clone to clones capable of proliferation in a suspension environment depleted of exogenous growth factors and cell-matrix contacts. Endotransglycosylation of xyloglucan may cut and reform cross-links, enabling limited cell expansion through temporary loosening of the cell wall. Three distinct K$ sp+$ currents were identified in corn (Zea mays) protoplasts using the whole-cell patch-clamp technique. Inward-rectifying K$ sp+$ currents were evoked at membrane potentials more negative than $-$100 mV. The activation range was sensitive to external K$ sp+$ and shifted in the positive direction as the K$ sp+$ concentration was elevated. The second K$ sp+$ current was voltage-independent and contributed to the resting membrane conductance of the protoplast. Finally, a voltage- and Ca$ sp{2+}$-dependent K$ sp+$ current was observed at potentials positive to $-$60 mV. This cur

But in order to be competitive with fossil fuels, understanding the mass-transfer limitations in PEC systems is critical. Hemodialysis is a renal replacement therapy that removes waste solutes from the blood stream using concentration gradients across a membrane. In order to overcome several shortcomings and increase the waste removal rate, a new dialyzer (filter) design is proposed in this study. Because the blood stream directly contacts the sheath stream, it is important to prevent blood cell migration from the blood stream to the sheath stream while providing enough time for the waste solutes to diffuse

Poly(A) tail deadenylation is a rate-limiting step of mRNA degradation. CCR4-associated factor 1s (CAF1s) is one of major enzymes to catalyze mRNA deadenylation in eukaryotes. Four members of CAF1 family, designated OsCAF1A, B, G and H, were analyzed and characterized in rice (Oryza sativa). Northern experiments and RT-PCR revealed diverse transcription patterns in response to sugar and various a

After rice cells were cultured with or without sucrose for 1 to 5 days, the cells and media were collected, and total protein were isolated, respectively. The expression level of total protein in cells is higher when cells were cultured in +S than -S by SDS-PAGE analysis. About the total protein expression in medium, there were 2 groups of proteins, 30- and 37-kDa expressing in +S and -S media, but there were others prot

Up to date, no recombinant human transcription protein is produced in plant cells, which is known as safety hosts for recombinant protein production. In this study, an inducible expression of recombinant Oct4 by transformed rice suspension cell culture was established. The recombinant DNA insertion into rice chromosomes and

Recent clinical studies indicate that the reduction of TM in cancer is associated with poor prognosis and tumor metastasis. However, the functional consequence of decreased TM expression in tumor metastasis remains unclear. To better understand the mechanism underlying the role of TM in tumor metastasis, HeLa cellswhich express heterogeneous TM were used. Unlike the traditional method, cells cultured by this method will aggregate to a sphere and form a 3D structure, which compared to other systems like the hanging drop method and the hydrogel method, our system is simple, easy and good in long term culture. Our research used fibroblast and keratinocyte in co-culture, and we adopted Hs68 cell line from fibroblast and HaCaT cell line from keratinocyte. The alkaline invertase (ITa) could be released by 1.0 M NaCl and purified further by ammonium sulfate fractionation, Con A-Sepharose affinity chromatography (non-retained), gel-filtration (Sepharose CL-6B) and DEAE-Sepharose chromatography. Another bound-form invertase (ITb) can be released by 5% EDTA from cell fragments after treated 1.0 M NaCl and purified further through the same steps. The molecular masses of ITa and ITb estimated by Superose 6 (FPLC) were 250 kD, a
