Full textMy work of thesis mainly concerned the study of the impact of a recombinant glycosylated IL-7 simian (R- slL7gly) on lymphocytary homeostasis T, in the model rhesus monkey. We showed that this IL-7 was not immunogenic and had a half-life longer than the preceding proteinic preparation (not glycosylated). We noted a general increase in the number of circulating cells in all the T subpopulations T CD4 and CD8 (RTE, naive, memories and effector). We showed that the expansion observed were due to an increase in the potential of survival of the circulating lymphocytes like with a massive increase in the expression of Ki-67 in the majority of the lymphoid subpopulations T. The analysis of the population of the recent thymique emigrants (RTE), and the quantification of the thymique function, enabled us to show that the injection of R-slL-7gly has also an important effect on stimulation of the intrathymic proliferation of the precursors of the cells T, therefore on the capacity of the thymus to produce new cells T. The results of these studies in vivo, underline the diversity and the importance of the roles filled by the IL-7 in control of lymphocytary homeostasis T. They consolidate the idea to use the IL-7 in private clinic, as auxiliary immunomodulator within the framework of treatment against the lymphopenia

Full textMajor modifications of T-cell homoeostasis and type I interferon (IFN) response occurring during the acute phase of human immunodeficiency virus (HIV) infection are key elements of the evolution of physiopathology. Indeed, these phenomena are poorly understood due to the difficulty to study this phase in humans. This thesis work contributes to a better definition of these modifications in simian immunodeficiency virus (SIV) infected rhesus macaques, an animal model of infection presenting a similar physiopathology as HIV infection in humans. First, in Humans, I showed that recent thymic emigrants significantly participate to HIV reservoir. In our animal model, early infection of thymocytes is observed. This infection induces, by 3 days post-infection, an acceleration of thymopoiesis that leads to a massive cell release in blood. Furthermore, this phenomenon is dependent on diversification of local IFN-α secrétion. The study of antiviral and anti-proliferative effects of the different simian IFN-α subtypes demonstrates that these cytokines have 4 different profiles of antiviral activity against SIVmac₂₅₁. Within the 12 simian IFN-a subtypes, 3 strongly inhibit viral replication in the cultures, 7 allow late viral escape after an initial inhibition of viral replication and the last 2 show no activity. Finally, I demonstrated that the observed diversification of IFN-α subtypes secretion is also found in lymph nodes and in various parts of the intestine by day 3 post-infection. This IFN-α secretion coincides with chemokines production in these organs probably leading to a modification of cellular migration within these organs. Thus, naive and central memory T-cells are recruited into lymph nodes while effector memory and effector T-cells are preferentially recruited in the gut

Full textIn the macaque/ simian Immunodeficiency virus (SIV) animal model, cytotoxic T lymphocytes (CTL), present during primary infection, play a major role in controlling viral load and delaying disease onset. The aim of our work was to optimize our vaccination strategy. In order to incluse mufti-specific CTL responses in macaques using a Upopeptide-based SIV vaccine which ean be adaptecl for human use. Our results show that lipopeptide vaccination is highly immunogenic and is able to Induce multi- specific CTL responses. The presence of such responses should broaden the target spectrum of the host and may prevent viral escape. These results warrant more testing of the efficacy of based HIV vaccine candidates in humans; in vaccination strategies in which lipopeptides are alone or combined with other viral vectors

Full textThe principal impact of this ethnography is that it provides an in-depth examination of the institutional processes, categories and stages through which the United Nations have have defined and elaborated the notions of "civil society" and "good gouvernance" in their HIV prevention programs in Central Asia and Transcaucasia, and then analyses how these notions come to be translated and applied on the ground (between 1994 and 2001). As such, the etnography 1) decribes the waysin which these discourses and practices have, in recent years, become normalized and have penetrated people's daily lives, 2) the various modes of questionning, problematizing and destabilizing that have been formulated by local actors in opposition to these normalizing discourses; and 3) how these local resistances have, in part, been incorporated into changing ideological and practical fields at the United Nations

Full textInfection by HIV and SIV immunodeficiency virus induce multiple hematopoietic defects in Primates that reflect central hematopoietic disorders, which contribute to immunodeficiency in infected individuals. Here we show that CD34+ progenitor cells lost their clonogenic potential upon SIV infection, in parallel with down-regulation of STAT5A and STAT5B expression. This defect was entirely rescued by forced expression of STAT5B. We demonstrate that HIV/SIV Nef recapitulates SIV action on CD34+ progenitors ex vivo and is required for SIV action on hematopoietic progenitors in vivo. We further demonstrate that Nef activity strictly depends on thé presence of the peroxisome proliferator-activated receptor gamma (PPAR-y). Our findings extend the crucial role of Nef in HIV/SIV pathogenicity. They further reveal the pivotal role of PPAR-y/STAT5 pathway in regulating early hematopoiesis. They provide an avenue for exploration of the therapeutic benefits of PPAR-y antagonists in AIDS patients, and also of PPAR-y agonists in hematopoietic disorders. In this context, we particulary focused on therapeutic potential of PPAR-y/STAT5 pathway in Bcr/Abl leukemic stem cells
