Abstract:
La digestion intestinale des protéines alimentaires fait intervenir des protéases d’origine pancréatique et des peptidases intestinales. Les produits de la digestion sont constitués d’acides aminés libres et de peptides relativement abondants. Acides aminés et peptides sont transportés dans l’entérocyte où ces derniers subissent une hydrolyse. Les acides aminés libres présents dans la veine porte présentent un profil bien différent de celui des protéines alimentaires. En effet, le métabolisme intestinal des acides aminés est très actif. Afin d’assurer la synthèse des protéines constitutives et sécrétées, l’intestin prélève des acides aminés à la fois dans la lumière intestinale et dans le sang artériel. Cet organe renouvelle plus de 50 % de ses protéines par jour et la synthèse de protéines bien particulières comme les mucines engendre des besoins élevés en certains acides aminés comme la thréonine. L’intestin est le principal tissu utilisant la glutamine artérielle et le glutamate alimentaire. Le catabolisme intestinal de ces acides aminés produit de l’alanine, de l’acide aspartique, de la proline et, par l’intermédiaire des enzymes du cycle de l’urée, de l’ornithine, de la citrulline et de l’arginine. Les acides aminés indispensables n’échapperaient pas non plus au catabolisme intestinal. Le rôle de l’intestin ne se limite donc pas à la digestion des protéines et à l’absorption des acides aminés. Son métabolisme modifie profondément la disponibilité des acides aminés alimentaires pour le reste de l’organisme.

Abstract:
The present study reports the synthesis, crystal structure determinations, and the conformational analysis of N-tertiobutyloxycarbonyl N′-methylthioalanine (Boc-AlaS-NHCH3, C9H18N2O2S) and of N-tertiobutyloxycarbonyl N′-methylthioglycine (Boc-GlyS-NHCH3, C8H16N2O2S). The particular feature of these compounds is the replacement of the classical oxopeptide linkage by a thioamide bond. Crystals of Boc-AlaS-NHCH3 are tetragonal, space group P43212. Those of Boc-GlyS-NHCH3 are monoclinic, space group P21/c. Both structures were solved by direct methods and refined by full-matrix least-squares methods to Rw = 0.045 and 0.035 for 827 and 1335 reflections respectively, with intensities greater than 2.5σ(I). The conformations of both compounds correspond to conformational energy minima, calculated for classical amino acids. The C=S bond lengths of 1.665(9) and 1.650(3) Å constitute the major difference compared to oxopeptides; the crystal structures reveal that the presence of the sulfur atom does not change the electronic properties of the peptide bond. Using a classical method for the study of peptides (ECEPP/2), conformational energy maps were computed for derivatives of dithioalanine and dithioglycine and are compared to the oxo residues. We conclude that the synthesis and conformational analysis of thionated amino acids allow us to introduce the thioamide linkage into more complex peptide structures and to predict the conformational behaviour. In the following review article, we present arguments for and against the use of postoperative troponin surveillance and preoperative natriuretic peptide testing. This article covers the evidence that informed the CCS 2016 perioperative guidelines and research published since then. This review is based on the debate held at the Canadian Society of Internal Medicine/American College of Physicians Rocky Mountain Chapter Annual Meeting (CSIM/ACP RM) held in October 2018 in Banff, Alberta.

 Dans l'article de synthèse qui suit, nous présentons des arguments pour et contre l'utilisation de la surveillance postopératoire de la troponine et des tests préopératoires de peptides natriurétiques. Cet article porte sur les données probantes qui ont éclairé les lignes directrices périopératoires de la SCC 2016 et les recherches publiées depuis lors. Abstract:
La modification de la liaison amide dans les peptides a plusieurs conséquences potentielles: une biodégradabilité réduite, une possible modulation structurale due aux perturbations du réseau de liaisons hydrogène, et éventuellement, une bio-activité modulée pour l'analogue pseudopeptidique résultant, sans nécessiter de changer les chaines latérales. Nous avons étudie les perturbations structurales induites par la substitution d'une liaison amide par un groupe hydrazide dans diverses séquences mono-, di- et tripeptidiques protégées à leurs deux extrémités par une fonction amide, et contenant l'analogue hydrazine de la proline ou de l'alanine. L'analyse conformationnelle a été conduite à l'état solide par diffraction des rayons X, en solution par spectroscopie infrarouge et résonance magnétique nucléaire du proton, et par modélisation moléculaire. Après des considérations générales sur les structures des peptides et pseudopeptides, l'aspect chimique et technique de ce travail est présenté dans le second chapitre. Le troisième chapitre rassemble les données spectroscopiques et les conclusions sur les structures présentes qui sont ensuite discutées en référence aux séquences peptidiques originelles. La conclusion principale est que le groupe hydrazide induit localement un repliement du à la nucléophilie de l'oxygène du carbonyle et de l'azote alpha qui participent tous les deux a une liaison hydrogène bifide avec le site N-H amide immédiatement situe en aval. La conformation repliée qui en résulte est très stable, et capable de donner naissance à une forme globalement repliée de la molécule, de façon très semblable au repliement beta dans les peptides

Le tryptophane est un acide aminé essentiel qui, en plus de son rôle dans la biosynthèse des protéines, est le précurseur biochimique de nombreux composés. Beaucoup de peptides biologiquement actifs, qui possèdent dans leurs séquences cet acide aminé contenant un substituant indole, ont des propriétés physiologiques importantes. On peut citer par exemple des dérivés comme la sérotonine, le tryptamine, le sumatriptan qui ont des effets neurophysiologiques. Les analogues de tryptophane sont aussi des « building block » important pour la synthèse de peptidomimétiques, d'analogues de produits naturels et de composés biologiquement actifs. Une autre propriété importante du tryptophane et de ses analogues concerne la fluorescence du noyau d`indole qui peut être utilisée par exemple pour l`étude de changements conformationnels d'une protéine et aussi l'étude des interactions protéine-membrane.Dans ce contexte, cette thèse est consacrée à la mise au point d'une nouvelle stratégie de synthèse asymétrique permettant d'accéder efficacement à d`analogues énantiopurs du tryptophane : les beta 2-tryptophanes (2-indolyl-beta-alanines) ainsi qu'à la synthèse et l'étude de nouveaux analogues de peptides à activité opioïdes contenant ces analogues. Nous avons développé une nouvelle stratégie de synthèse asymétrique permettant d'accéder efficacement à des analogues de beta 2-tryptophane (2-indolyl-beta-alanines) énantiopurs. - Nous avons effectué la synthèse d`un auxiliaire chiral le (R)-4-(3-hydroxy-4,4-diméthyl-2-oxopyrrolidin-1-yl) benzoate de benzyle ainsi que celle du dérivé nitroacrylate correspondant : le (R)-4-(3-(3-nitroacryloyloxy)-4,4-diméthyl-2-oxo pyrrolidin-1-yl) benzoate de benzyle ((R)-10). Les composés racémiques ont également été synthétisés pour préparer les témoins racémique des molécules cibles.- Nous avons mis au point des conditions optimisées de la réaction d`alkylation de Friedel-Crafts entre ce nitroacrylate chiral et divers indoles.- Nous avons également étudié pour chaque produit ainsi obtenu les meilleures conditions de transformation conduisant à la synthèse des analogues des N-Fmoc-beta 2-tryptophanes racémiques et optiquement purs.- Nous avons contrôlé l'absence d'épimérisation pouvant se produire pendant les réactions de transformation des produits optiquement purs et déterminé sans ambiguïté la configuration absolue des 2-indolyl-beta-alanines isolées.- Nous avons synthétisé sur support solide par stratégie-(Fmoc) 4 nouveaux peptides potentiellement biologiquement actifs et 2 peptides de référence - analogues raccourcis de la nociceptine<br>Tryptophan, an essential amino acid, both functions as a building block in protein biosynthesis and as a biochemical precursor. It is abundantly found in most biologically active peptides that exhibit various physiological properties in particular hormonal and antimicrobial activities. Some of its natural derivatives like serotonin, tryptamine, and also unnatural derivatives such as sumatriptan, have neurophysiologic effects. Tryptophan analogues are also important building blocks for the synthesis of peptidomimetics, natural products and biologically active compounds. Another important property of tryptophan and tryptophan analogues is related to the fluorescence of the indole ring that can be used to study conformational changes in protein and in protein-membrane interactions. The asymmetric Friedel-Crafts alkylation of various indoles with a chiral nitroacrylate provides optically active beta-tryptophan analogues after reduction of the nitro group and removal of the chiral auxiliary. This reaction generally occurs in good yield and high diastereoselectivity (up to 90:10). We have established a new route to prepare enantiopure beta-tryptophan analogues ((S)-2-indolyl-beta-alanines). We showed that beta-nitroacrylate (R)-2 is a good chiral auxiliary for asymmetric Friedel-Crafts alkylation of indoles. (R)-2-indolyl--alanines were obtained by the same synthetic route by using the chiral compound (S)-2. beta-tryptophan analogues are delivered in their N-Fmoc-protected form, ready to use for instance in solid phase peptide synthesis, which is one of the most popular method for peptide synthesis. This study provides a new example of asymmetric beta-tryptophan analogues preparation and further studies concerning their applications in medicinal chemistry and in organic synthesis are now in progress

Une conception générale de synthèse de phosphonopeptides renfermant un motif phosphore du cote c-terminal ou du cote n-terminal est proposée à partir d'un même substrat acide 2-dialkylphosphonoalcanoique. L'objectif est de préparer des inhibiteurs de peptidases présentant une activité thérapeutique. L'approche la plus efficace pour introduire le motif phosphonate sous forme enantiomeriquement pure dans les phosphonopeptides n-terminaux consiste à utiliser la chaine peptidique chirale pour induire une asymétrie sur le carbone directement lié au phosphore. Le couplage peptidique a également été mis au point entre l'acide phosphonoacetique et l'acide 6-aminopenicillanique dans le but de préparer de nouveaux antibiotiques. L'étude du comportement des phosphonopeptides n-terminaux en tant que réactifs de Honer a été abordée pour préparer des peptides insaturés, substituts possibles de peptides à usage thérapeutique. Enfin, la réactivité de thiols sur les chlorures d'acides 2-dialkylphosphonoalcanoiques constitue une voie de synthèse de dialcoxyphosphorylalcane thioates de s-alkyle, qui conduisent eux-mêmes par réaction de Horner à des thioesters éthyléniques, cette réaction des généralisable aux dérivés chrysanthemiques

Face à l’explosion démographique mondiale actuelle, les rendements agricoles se doivent d’être les plus élevés possible. Afin de relever ce défi, l’utilisation de pesticides efficaces et éco-compatibles est devenue indispensable. Dans le cadre de la recherche de nouveaux composés répondants à ces objectifs, les produits naturels continuent de retenir l’attention des chercheurs. Parmi eux, nous nous sommes intéressés à la gougérotine, un peptidonucléoside découvert au sein d’une bactérie dans les années 60 et possédant un large spectre d’activités biologiques. Dans le but d’optimiser l’activité antifongique et de diminuer la toxicité de la molécule, plusieurs analogues ont été synthétisés en jouant notamment sur la nature de la base nucléique. Une des réactions clé au cœur de la synthèse de nucléoside est la N-glycosylation permettant la liaison glycosidique entre la base nucléique et un donneur de glycosyle. Au cours de la voie de synthèse des analogues peptidonucléosidiques, une étude méthodologique de la N-glycosylation a été réalisée mettant en jeu différents donneurs de glycosyle et bases nucléiques. Des tests préventifs ont ensuite été réalisés sur les composés obtenus afin d’évaluer leur potentiel antifongique. Grâce aux analogues de la gougérotine et aux intermédiaires synthétisés, une étude de relation structure-activité a été menée<br>In the current context of the world demographic explosion, agricultural yields need to be as high as possible. The use of efficient and environment friendly pesticides has therefore become essential. In the framework of the discovery of new compounds, natural products remain an important source of inspiration. Among them, we focused on gougerotin, a peptidylnucleoside isolated from a bacteria in the 60’s which possess a broad spectrum of biologic activities. In order to improve antifungal potential and decrease the phytotoxicity of gougerotin, several analogues have been synthesized replacing the natural nucleobase. N-glycosylation is one of the key-reaction in peptidylnucleoside synthesis. During the synthesis of the analogues, a study of the N-glycosylation was carried out with three different donors and several bases. Protectives tests were realized with many pathogens to evaluate antifungal activity of our compounds and a structure-activity relationship was established

Les bêta-peptides montrent une grande stabilité métabolique et protéolytique. Ce travail s'est principalement focalisé sur leur utilisation en tant que plateformes multivalentes pour la présentation de "sucres" et en particulier d'antigènes saccharidiques spécifiques des tumeurs, connus sous le nom de TACAs (Tumor-Associated Carbohydrate Antigen). Les TACAs, et parmi eux l'antigène Tn (GalNAc-Alpha-O-Ser/Thr), sont considérés comme des marqueurs des cellules tumorales qui peuvent être intégrés dans des structures synthétiques destinées à mimer la surface des cellules cancéreuses en vue d'induire une réponse immunitaire antitumorale. Dans la première partie de la thèse, nous avons étudié la synthèse de bêta-peptides glycoconjugués, obtenus par cycloaddition 1,3-dipolaire catalysée au cuivre entre un alcyne et un azoture (CuAAC - Copper-catalysed Alkyne-Azide Cycloaddition). Ces glycoconjugués, présentant jusqu'à six unités saccharidiques, ont été élaborés à partir de bêta-peptides fonctionnalisés par des groupes azoture ou alcyne, préparés au départ d'acide aspartique. Les trimères glycoconjugués ont également été couplés à de la biotine, par l'intermédiaire d'un espaceur (d'acide aminocaproïque) et l'antigénécité de ces édifices a été testée. Les bêta-peptides présentent également une tendance prononcée à adopter des structurations secondaires. C'est pourquoi les oligomères synthétisés ont été étudiés d'un point de vue conformationnel afin de déterminer la nature d'une éventuelle préférence conformationnelle

Les extraits naturels tels que les venins animaux constituent une source importante de peptides bioactifs à visée thérapeutique. Parmi les peptides dérivés de venins utilisés en médecine, citons l’Eptifibatide, un médicament antiplaquettaire développé à partir de l’echistatin provenant d’une vipère, le Ziconotide, un puissant analgésique identifié dans le venin d’un cône ou encore l’Exenatide, un agoniste du récepteur du glucagon-like peptide 1 isolé de la salive du monstre de Gila et utilisé pour le traitement du diabète de type 2. Ces peptides riches en ponts disulfures provenant des venins présentent une structure tridimensionnelle contrainte et une stabilité plasmatique accrue comparé aux peptides linéaires. La conservation et la ressemblance des récepteurs de la proie/du prédateur avec les récepteurs humains, fait des peptides de venins une source unique de composés tête de série pour le design d’outils pharmacologiques et de composés thérapeutiques. Il est estimé que moins de 1% des peptides de venins ont été caractérisé pharmacologiquement. Ce projet a pour objectif d’explorer la pharmacologie de nouveaux peptides isolés de venins en utilisant la synthèse peptidique en phase solide basée sur la chimie Fmoc (Fmoc-SPPS) ainsi que des stratégies de repliement oxydant et régiosélectif afin de produire le peptide correctement replié responsable de l’activité biologique pour sa caractérisation ultérieure. Alors que la première partie de ce projet est dédiée à la synthèse de peptides de venins linéaires et pauvre en pont disulfures, la seconde partie sera consacrée à la synthèse de peptides riches en pont disulfures via des stratégies de repliement oxydatif et régiosélectifs. Dans un dernier chapitre, nous allons explorer les approches de protéomique intégrées à la transcriptomique pour l’identification de nouvelles séquences à partir des venins. Dans l'ensemble, ce projet permettra de mieux comprendre la pharmacologie des peptides de venins et mènera à la conception de nouveaux outils pharmacologiques et de potentiels candidats médicaments<br>Natural extracts such as animal venoms are an important source of bioactive peptides for therapeutic purposes. Peptides derived from venoms currently used in medicine include Eptifibatide, an antiplatelet drug developed from echistatin, a toxin isolated from a viper, Ziconotide, a potent analgesic identified in the venom of a cone snail and Exenatide , a glucagon-like peptide 1 receptor agonist isolated from the saliva of the Gila monster and used for the treatment of type 2 diabetes. These disulfide-rich venom peptides exhibit a constrained three-dimensional structure and increased plasma stability compared to linear peptides. Conservation of prey / predator receptors with human receptors makes venom peptides a unique source of lead compounds for the design of pharmacological tools and therapeutic compounds. It is estimated that less than 1% of the venom peptides have been pharmacologically characterized. Thus, this project aims to explore the pharmacology of novel venom-isolated peptides using solid phase peptide synthesis based on Fmoc chemistry (Fmoc-SPPS) as well as oxidative and regioselective folding strategies to produce the correctly folded and biologically active peptide for subsequent characterization. While the first part of this project is dedicated to the synthesis of linear and disulfide-poor venom peptides, the second part will be dedicated to the synthesis of disulfide-rich peptides via oxidative and regioselective folding strategies. Finally, we will use proteomic approaches integrated with transcriptomic data for the identification of new sequences from venoms. Overall, this project provides a better understanding of the pharmacology of venom peptides and identifies leads for the development of new pharmacological tools and potential drug candidates

Récemment, beaucoup de recherches ont démontré l'importance des peptides dans le domaine de la chimie médicale. Le développement de médicaments basés sur la modification des peptides, mimétiques des protéines natives, est une cible très importante. La synthèse de peptides contraints est une stratégie très efficace pour des études structure-activité ayant la finalité de caractériser les sites actifs des protéines. Au cours de cette thèse, les peptides ont été modifiés par introduction de différentes contraintes. Dans une première partie de ces études, une série d'acides aminés non naturels contraintes ont été synthétisées. Les acides aminés ont été introduits dans la séquence peptidique de l'antagoniste puissant de la bradykinine HOE 140. Des analogues de ce peptide ont été synthétisés en phase solide et des tests biologiques ont été effectués. Au cours de la deuxième partie de la thèse une stratégie efficace de synthèse de cyclopeptides contraints en phase solide a été développée<br>Many researches have recently demonstrated the importance of peptides for medicinal chemistry. The development of new drugs based on peptide modification is an important goal, as peptides can mimic proteins. The synthesis of constrained peptides represents a very efficient strategy for structure-activity relationship studies, in order to characterize the protein active site. During this thesis, peptides were modified introducing different constraints. In the first part of these studies, a series of unnatural amino acids were synthesized. These amino acids were introduced in the peptide sequence of the potent BK antagonist HOE 140. These peptide analogues were synthesized by a solid-phase approach and were then tested for their biological activity. Abstract:
Control of agro-associated pathogens is becoming increasingly difficult due to increased resistance and mounting restrictions on chemical pesticides and antibiotics. Likewise, in veterinary and human environments, there is increasing resistance of pathogens to currently available antibiotics requiring discovery of novel antibiotic compounds. These drawbacks necessitate discovery and application of microorganisms that can be used as biocontrol agents (BCAs) and the isolation of novel biologically-active compounds. This highly-synergistic one year project implemented an innovative pipeline aimed at detecting BCAs and associated biologically-active compounds, which included: (A) isolation of multidrug-resistant desert soil bacteria and root-associated bacteria from medicinal plants; (B) invitro screening of bacterial isolates against known plant, animal and human pathogens; (C) nextgeneration sequencing of isolates that displayed antagonistic activity against at least one of the model pathogens and (D) in-planta screening of promising BCAs in a model bean-Sclerotiumrolfsii system. The BCA genome data were examined for presence of: i) secondary metabolite encoding genes potentially linked to the anti-pathogenic activity of the isolates; and ii) rhizosphere competence-associated genes, associated with the capacity of microorganisms to successfully inhabit plant roots, and a prerequisite for the success of a soil amended BCA. Altogether, 56 phylogenetically-diverse isolates with bioactivity against bacterial, oomycete and fungal plant pathogens were identified. These strains were sent to Auburn University where bioassays against a panel of animal and human pathogens (including multi-drug resistant pathogenic strains such as A. baumannii 3806) were conducted. Nineteen isolates that showed substantial antagonistic activity against at least one of the screened pathogens were sequenced, assembled and subjected to bioinformatics analyses aimed at identifying secondary metabolite-encoding and rhizosphere competence-associated genes. The genome size of the bacteria ranged from 3.77 to 9.85 Mbp. All of the genomes were characterized by a plethora of secondary metabolite encoding genes including non-ribosomal peptide synthase, polyketidesynthases, lantipeptides, bacteriocins, terpenes and siderophores. While some of these genes were highly similar to documented genes, many were unique and therefore may encode for novel antagonistic compounds. Comparative genomic analysis of root-associated isolates with similar strains not isolated from root environments revealed genes encoding for several rhizospherecompetence- associated traits including urea utilization, chitin degradation, plant cell polymerdegradation, biofilm formation, mechanisms for iron, phosphorus and sulfur acquisition and antibiotic resistance. Our labs are currently writing a continuation of this feasibility study that proposes a unique pipeline for the detection of BCAs and biopesticides that can be used against phytopathogens. It will combine i) metabolomic screening of strains from our collection that contain unique secondary metabolite-encoding genes, in order to isolate novel antimicrobial compounds; ii) model plant-based experiments to assess the antagonistic capacities of selected BCAs toward selected phytopathogens; and iii) an innovative next-generation-sequencing based method to monitor the relative abundance and distribution of selected BCAs in field experiments in order to assess their persistence in natural agro-environments. We believe that this integrated approach will enable development of novel strains and compounds that can be used in large-scale operations.
