Herein we describe efforts to reconstitute parts of the Escherichia coli cell division machinery inside of water-in-oil emulsion compartments and synthetic phospholipid vesicles. The system was built with DNA and purified transcription and translation machinery housed in a compartment. Theoretical models of mutation-selection balance predict that the maximum tolerable mutation rate is much lower for organisms growing in colonies than for those in well-mixed liquid media due to drift imposed by competition for position along the growing colony front. If a kinetochore does not correctly attach to a microtubule the spindle checkpoint stops cell cycle progression until all chromosomes are attached to microtubules or tension is experienced while pulling the chromosomes. In der Dissertationsschrift wird ein neuartiges Konzept der Realisierung der Direkten Digitalen Frequenzsynthese (DDS) vorgestellt. Ausgehend von der analysierten Literatur werden das Wirkprinzip eines Standard-DDS-Synthesizer analysiert und Möglichkeiten zur Aufwandsreduktion untersucht. Ein neuartiger Ansatz zur Realisierung einer vollständig digitalen DDS ergibt sich in der Anwendung der Pulse-Output-DDS. DNA synthesis, as monitored by ('3)H-thymidine incorporation, in the germinative stem cells of juvenile Hymenolepis diminuta, appears to be negatively modulated by increases in cyclic-3'5'-adenosine monophosphate (cAMP). Among the endogenous substances which elevate cAMP levels in these worms is serotonin, a.k.a. 5-hydroxytryptamine (5HT). High concentrations of exogenous 5HT, or treating worms with reserpine which presumably induces a secretion of endogenous 5HT, similarly depresses DNA synthesis.
