Abstract:

Abstract:
Jute is a bio-degradable, agro-renewable, and widely available lingo cellulosic fiber having high tensile strength and initial modulus, moisture regain, good sound, and heat insulation properties. For these unique properties and eco-friendly nature of jute fibers, jute-based products are now widely used in many sectors such as packaging, home textiles, agro textiles, build textiles, and so forth. The diversified applications of jute products create an excellent opportunity to mitigate the negative environmental effect of petroleum-based products. For producing the best quality jute products, the main prerequisite is to ensure the jute yarn quality that can be defined by the load at break (L.B), strain at break (S.B), tenacity at break (T.B), and tensile modulus (T.M). However, good quality yarn production by considering these parameters is quite difficult because these parameters follow a non-linear relationship. Therefore, it is essential to build up a model that can cover this entire inconsistent pattern and forecast the yarn quality accurately. That is why, in this study, a laboratory-based research work was performed to develop a fuzzy model to predict the quality of jute yarn considering L.B, S.B, T.B, and T.M as input parameters. For this purpose, 173 tex (5 lb/spindle) and 241 tex (7 lb/spindle) were produced, and then L.B, S.B, T.B and T.M values were measured. Using this measured value, a fuzzy model was developed to determine the optimum L.B, S.B, T.B, and T.M to produce the best quality jute yarn. In our proposed fuzzy model, for 173 tex and 241 tex yarn count, the mean relative error was found to be 1.46% (Triangular membership) and 1.48% (Gaussian membership), respectively, and the correlation coefficient was 0.93 for both triangular and gaussian membership function. This result validated the effectiveness of the proposed fuzzy model for an industrial application. The developed fuzzy model may help a spinner to produce the best quality jute yarn.

Abstract:
Helminth infections are among the neglected tropical diseases affecting billions of people globally, predominantly in developing countries. Helminths’ effects are augmented by coincident tuberculosis disease, which infects a third of the world’s population. The role of helminth infections on the pathogenesis and pathology of active tuberculosis (T.B.) remains controversial. Parasite-induced suppression of the efficacy of Bacille Calmette-Guerin (BCG) has been widely reported in helminth-endemic areas worldwide. T.B. immune response is predominantly proinflammatory T-helper type 1 (Th1)-dependent. On the other hand, helminth infections induce an opposing anti-inflammatory Th2 and Th3 immune-regulatory response. This review summarizes the literature focusing on host immune response profiles during single-helminth, T.B. and dual infections. It also aims to necessitate investigations into the complexity of immunity in helminth/T.B. coinfected patients since the research data are limited and contradictory. Helminths overlap geographically with T.B., particularly in Sub-Saharan Africa. Each disease elicits a response which may skew the immune responses. However, these effects are helminth species-dependent, where some parasites have no impact on the immune responses to concurrent T.B. The implications for the complex immunological interactions that occur during coinfection are highlighted to inform government treatment policies and encourage the development of high-efficacy T.B. vaccines in areas where helminths are prevalent.

Abstract:
Instant healing, performance of miracles, public exorcism, supply of blessed objects for protection, and prophetic abilities are major religious practices that account for the popularity of certain Pentecostal leaders in contemporary Africa. Prophet T.B. Joshua, a renowned Nigerian televangelist, is one of them. Through his Emmanueltvchannel, he informs ‘viewers all over the world’ that invisible but influential evil forces are responsible for their daily challenges and that people can enjoy prosperity when they are purged of malevolent spirits. This article describes the practices and narratives of liberation from unknown bondage by Cameroonians who draw inspiration from T.B. Joshua’s prophetic messages and displays. The aspects covered include the desire to embark on a pilgrimage to T.B. Joshua’s church in Nigeria and actors’ use of his blessed religious objects to neutralise, destroy, or fight off invisible evil forces suspected of halting their socioeconomic progress.

Abstract:
Abstract Procyclic culture form (PCF) trypanosomes were established from a bloodstream form population of cloned Trypanosoma brucei rhodesiense and were used to immunize mice for hybridoma production. Indirect immunofluorescence was used to select 10 hybridomas which secreted antibodies that bound to the surface of homologous living PCF. The antibodies reacted with PCF of several clones of T.b. brucei, T.b. gambiense, and T.b. rhodesiense, but not with PCF of T. congolense or T. vivax, or with promastigotes of several species of Leishmania parasites. The antigens were not detectable in ethanol/acetic acid-fixed bloodstream forms or in lysates of bloodstream forms of any of the T. brucei subspecies, and are thus species-specific and stage-specific markers. Selected monoclonal antibodies bound to procyclic trypanosomes taken directly from the midgut of infected tsetse flies, and to immature epimastigote forms in salivary probes, and may therefore be useful in epidemiologic investigations.

Abstract:
The primary aim of this work was to investigate phospholipid metabolism in trypanosomes, including a phosphoinositide signalling, and the metabolism of the VSG and its GPI anchor. The phosphoinositides of <I>T.b brucei</I> were labelled using 32P (from [32P)Pi or [γ32P]ATP) and [3H)inositol through a variety of methods. Most work was carried out with phospholipids labelled using [γ32P]ATP as the donor, with cells permeabilised by being placed in a buffer of low osmotic strength (swell dialysis). Species labelled with 32P were identified as PI 4-P and PI 4,5-P2 through HPLC analysis. There was no evidence of 3-phosphorylated phosphoinositides under the conditions used. Results showed that <I>T.b brucei</I> has GTP- and Ca2+-regulated phosphoinositide metabolism, similar to that seen in vertebrate cells. Additionally, an unidentified factor in rabbit serum caused a 2-fold increase in the phosphorylation of the polyphosphoinositides. Although Ca2+ is known to cause the release of VSG through the action of GPI-PLC, it is not clear whether this enzyme plays an important role in membrane-form (mf) VSG metabolism in intact cells. The relationship between VSG release and subsequent GPI anchor metabolism was examined using a GPI-PLC inhibitor (pCMPSA) and conditions known to cause the release of soluble form (s) VSG (Ca2+). Metabolism of the mfVSG GPI anchor was taken to correspond with [32P]PA generation in cells incubated in the presence of [γ32P)ATP. Anti-VSG antibodies were used to detect VSG in supernatant samples of <I>T.b. brucei</I>. A great deal of phospholipid metabolism sensitive to pCMPSA, so possibly related to GPI-PLC activity, was shown to occur in the absence of sVSG release. The primary role of GPI-PLC in viable trypanosomes may therefore be in the regulation of processes other than mfVSG metabolism. <I>T.b. brucei </I>was incubated with anti-VSG IgGs to observe whether interaction with anti-VSG antibodies caused the destruction of mfVSG and concomitant increase in PA labelling. Anti-VSG IgGs had no effect on PA metabolism. mfVSG endocytosed in bloodstream from <I>T. brucei</I> may therefore be returned intact to the cell surface.

Abstract:

Abstract:

Abstract:
Objectives: Trastuzumab and pertuzumab bind to HER2 synergistically at different subdomains, improving the clinical benefit of breast cancer treatment. Recently, trastuzumab biosimilars were approved after comparison with the originator, proving high similarity in structure and activity, and equivalent efficacy, safety, and immunogenicity. This study aimed to compare in vitro antibody-dependent cell-mediated cytotoxicity (ADCC) and HER2-signaling inhibition elicited by biosimilar trastuzumab (BS-TZB) or originator trastuzumab (TZB) in combination with pertuzumab (PTZ). Methodology: Human breast cancer cell lines BT474 (HER2+/estrogen [ER] and progesterone receptor [PR]-positive) and HCC1954 (HER2+/ER and PR-negative) were used. Anti-HER2 antibodies: TZB (Herceptin® 440 mg, Roche, batch: N7194B11-B3064); BS-TZB (Zedora® 440 mg, Libbs/Biocon [trastuzumab-dkst], batch: 18F0072); and PTZ (Perjeta® 420 mg, Roche, batch: H0260B01) were commercially acquired. ADCC assay: cells were incubated with TZB or BS-TZB, with/without PTZ. To determine the cytotoxicity percentage, the LDH activity using the CytoTox-ONETM was measured. Western blot: HCC1954 cells were incubated with TZB or BS-TZB, with/without PTZ to analyze the HER2 signaling cascade. We analyzed that Akt, ERK, STAT3, and MKK3/6 were the downstream molecules. Results: ADCC activity of BS-TZB was higher than that of TZB in both cell lines (EC50 [μg/mL]: 1.01 and 1.32, respectively, for BT474: 0.16 and 0.23, for HCC1954). Combined with PTZ, BS-TZB also showed slightly higher ADCC activity than TZB (EC50 [μg/mL]: 0.90 and 1.15, for BT474: 0.13 and 0.19, for HCC1954). BS-TZB and TZB inhibited the phosphorylation levels of Akt, ERK, STAT3, and MKK3/6 in a dose- dependent manner. PTZ enhanced inhibition effects of BS-TZB and TZB on p-AKT and p-ERK, while it had no effect on the p-STAT3 and p-MKK3/6. Conclusions: In association with pertuzumab, trastuzumab biosimilar showed non-inferior biological potency and equivalent signaling inhibition compared with originator trastuzumab. This study explored mechanisms elicited by dual HER2 blockade, showing the same pattern for BS-TZB as well as TZB, combined with PTZ.

Abstract:
To decrease the weight of auto panels and reinforce the safety of vehicles, the tailor-welded blank (TWB) has been used widely in the auto panels manufacturing. Basic principles and advantages of TWB are described at first. Then the technologies of numerical simulation modeling for TWB are researched. By means of numerical simulation, the weld-bead movement and formability of TWB with the same mechanical property and multi-gauges are investigated. After analyzing the weld-bead movement in three different drawing depth combinations with same total deformed depth, some conclusions of weld-bead movement are drawn finally. It reveals the relationships between the weld-bead movement and formability of multi-gauge TWB.

Abstract:
Japan Atomic Energy Agency (JAEA) established the thermodynamic database (JAEA-TDB) for performance assessment of geological disposal of high-level radioactive waste (HLW) and TRU waste. Twenty-five elements which were important for the performance assessment of geological disposal were selected for the database. JAEA-TDB enhanced reliability of evaluation and estimation of their solubility through selecting the latest and the most reliable thermodynamic data at present. We evaluated and estimated solubility of the 25 elements in the simulated porewaters established in the “Second Progress Report for Safety Assessment of Geological Disposal of HLW in Japan” using the JAEA-TDB and compared with those using the previous thermodynamic database (JNC-TDB). It was found that most of the evaluated and estimated solubility values were not changed drastically, but the solubility and speciation of dominant aqueous species for some elements using the JAEA-TDB were different from those using the JNC-TDB. We discussed about how to provide reliable solubility values for the performance assessment.

Abstract:
The main contributor to the high-cycle fatigue of compressor blades is the response to aerodynamic forcing functions generated by an upstream row of stators or inlet guide vanes. Resonant response to engine order excitation at certain rotor speeds is especially damaging. Studies have shown that flow control by trailing edge blowing (TEB) can reduce stator wake strength and the amplitude of the downstream rotor blade vibrations generated by the unsteady stator-rotor interaction. In the present study, the effectiveness of TEB to reduce forced blade vibrations was evaluated in a modern transonic compressor rig. A row of wake generator (WG) vanes with TEB capability was installed upstream of the rotor, which was instrumented with strain gages. Data was collected with and without TEB at various rotor speeds involving resonance crossings. Using 0.8% of the compressor core flow for TEB along the full WG-span, rotor blade strain was reduced by 66% at the first torsional resonance crossing. Substantial reductions were also achieved with only partial span TEB. The results demonstrate the effectiveness of the TEB technique for reducing rotor vibrations in the complex flow environment of a closely-spaced transonic stage row. Moderate increases in stage performance were also measured.
