Abstract:
The primary aim of this work was to investigate phospholipid metabolism in trypanosomes, including a phosphoinositide signalling, and the metabolism of the VSG and its GPI anchor. The phosphoinositides of <I>T.b brucei</I> were labelled using 32P (from [32P)Pi or [γ32P]ATP) and [3H)inositol through a variety of methods. Most work was carried out with phospholipids labelled using [γ32P]ATP as the donor, with cells permeabilised by being placed in a buffer of low osmotic strength (swell dialysis). Species labelled with 32P were identified as PI 4-P and PI 4,5-P2 through HPLC analysis. There was no evidence of 3-phosphorylated phosphoinositides under the conditions used. Results showed that <I>T.b brucei</I> has GTP- and Ca2+-regulated phosphoinositide metabolism, similar to that seen in vertebrate cells. Additionally, an unidentified factor in rabbit serum caused a 2-fold increase in the phosphorylation of the polyphosphoinositides. Although Ca2+ is known to cause the release of VSG through the action of GPI-PLC, it is not clear whether this enzyme plays an important role in membrane-form (mf) VSG metabolism in intact cells. The relationship between VSG release and subsequent GPI anchor metabolism was examined using a GPI-PLC inhibitor (pCMPSA) and conditions known to cause the release of soluble form (s) VSG (Ca2+). Metabolism of the mfVSG GPI anchor was taken to correspond with [32P]PA generation in cells incubated in the presence of [γ32P)ATP. Anti-VSG antibodies were used to detect VSG in supernatant samples of <I>T.b. brucei</I>. A great deal of phospholipid metabolism sensitive to pCMPSA, so possibly related to GPI-PLC activity, was shown to occur in the absence of sVSG release. The primary role of GPI-PLC in viable trypanosomes may therefore be in the regulation of processes other than mfVSG metabolism. <I>T.b. brucei </I>was incubated with anti-VSG IgGs to observe whether interaction with anti-VSG antibodies caused the destruction of mfVSG and concomitant increase in PA labelling. Anti-VSG IgGs had no effect on PA metabolism. mfVSG endocytosed in bloodstream from <I>T. brucei</I> may therefore be returned intact to the cell surface.

Abstract:

Full text
Abstract:
The fast development of the tourism industry has created both positive and negative impacts on host communities. Reacting to these impacts, host residents hold various attitudes towards tourism and tourists. It has come to common agreement that a supportive host community plays a vital role in the successful and long-term development of community tourism. In order to explain the antecedents of community attitudes towards tourism, researchers have investigated the topic from both the extrinsic perspective (such as stage-based models) and the intrinsic perspective (such as the Social Exchange Theory). Nevertheless, there are still several important research gaps and deficiencies within the existing literature. First, the influence of psychological factors (personality) on attitudes towards tourism is somewhat neglected. Second, when examining the relationship between community attitudes towards tourism and the influential factors, the majority of previous studies do not distinguish the orthogonal dimensions of attitudes (such as positive dimension and negative dimension). Third, the existing literature has not addressed the issue of whether specific attitudes towards tourism will lead to a corresponding behaviour when interacting with tourists, and what other factors are determinant in host-guest interactive behaviour. Aiming to address such research gaps and deficiencies, this study has a major objective of developing a theoretical model (encompassing attitude element) to understand the antecedents of host-guest interaction. A quantitative approach was employed for the entire project. A self-administrated questionnaire survey was used to collect primary data. A total of 878 useful questionnaires were returned for analyses. Stratification sampling methods were utilized in communities where population database was accessible, while random sampling methods were used in other communities. The findings from this study confirmed the two major hypotheses in terms of community attitudes towards tourism. First, there was a significant relationship between an individual's personality and his/her attitudes towards tourism. Residents being high on Openness, Conscientiousness, Extraversion and Agreeableness traits and low on the Neuroticism trait tended to be more positive towards tourism than their counterparts. Second, while some factors were found to be influential on both positive and negative dimensions of attitudes towards tourism, others only demonstrated influences on one dimension. The Resident-Tourist Interaction Model developed in this study provided a comprehensive theoretical framework in modelling and predicting host residents' interactive behaviour towards tourists. Drawing on the evaluation results of three leading behavioural theories belonging to the school of cognition, the Model identified attitudinal, volitional, social, motivating and habitual factors for the prediction of resident-tourist interactive behaviour. Motivating factor (intention) was the critical and immediate element for action, which, in turn, was best predicted by subjective norms. External factors (gender, age and personality traits) only moderated the predictive power of the Model by less than two percent. The Model was valid and reliable for the current data. The present study advanced resident-tourist interactive behavioural study by establishing a sound theoretical framework. It also consolidated the body of knowledge in understanding community attitudes towards tourism. Findings from this study have significant practical implications for community tourism planning and management.

Gray, Will. "T.S. Eliot among the Metaphysicals." Thesis, University of St Andrews, 2011. http://hdl.handle.net/10023/3123.

Full text
Abstract:
Eliot's admiration for the poetry of the seventeenth century is well known. However, the several documents that explore the subject (thinly scattered across decades) fail to constitute a full account. Drawing on manuscript and print sources, and tracing particularly Eliot's prose poetics, this thesis redresses the scholarly need for a nuanced account of Eliot's role among the Metaphysical poets. The relationship ran in both directions, most famously in Eliot's championing of the poets and his urging that they find a new readership. His part in the revival of Metaphysical poetry, though, has been greatly exaggerated and the record is here faithfully adjusted. He was not in any way responsible for that revival, though he is its most important product, as is shown by a careful reconstruction of turn-of-the-century transcontinental publishing and reception. Eliot's criticism tells its own, largely unexplored story about the Metaphysicals and their influence on his critical and poetic sensibility. Most scholars, for instance, know that Eliot loved Donne, but few know the origin of that interest, let alone its brief nature or the personal reasons that drove him to appreciate the poet's audacity. Most also know the Modernist dicta of Tradition, objective correlative and the dissociation of sensibility, but not the fact that each owes something to Eliot's thinking about Donne. Engaging with Harvard class notes, under-consulted textbooks and a close study of Eliot's articles from the 1910s, two separate chapters investigate his education and early prose, along with their delicate dance between impersonality and confessional criticism. 1921-1926 marks a crucial stage in Eliot's writing, both for his poetry and his criticism. The Metaphysicals provide the clearest barometer of that change as well as the space where he approached conversion. This thesis is the first to trace the poets throughout Eliot's criticism, one of the first to engage with his Metaphysical-themed Clark Lectures, and the first to move far past Eliot's conversion, interpreting George Herbert as typical of his late mindset. In 1961 Eliot claimed no one had been as influenced by the Metaphysical poets as he had been. What this thesis offers is not only a more nuanced portrait of that influence but also a glimpse into the educational, critical and reading cultures of the early 1900s.

Bahbahani, A. "The invisible wagnerite : T.S. Eliot." Thesis, University of Sussex, 1999. http://ethos.bl.uk/OrderDetails.do?uin=uk.bl.ethos.264554.

Full text
Abstract:
The present thesis is about aspects of Wagnerism in the works of T.S. Eliot, in terms of both influence and affinities. The opening chapter offers a brief historical background to Eliot's familiarity with and relation to Wagner, as well as an account of the principal issues to be discussed in the following chapters. The two artists' volumes of criticism mostly show in theory how they work in practice. One key theme there is the maintenance of the idea of tradition and at the same time that of revolutionizing the arts (poetry, opera and drama). Eliot's interest in music, notably in the use of the Wagnerian leitmotif, is one of the highlights of this study. Then, Eliot's concern for myths: his 'mythical method' is discussed in a separate chapter for comparison with Wagner's way of handling myths in his operas. Other important topics feature drama, the Greeks and Shakespeare, and poetry, especially Dante's, the Romantics' and that of the French Symbolists. Certain themes common in both (like salvation and love) are tackled in more than one chapter because of their relevance throughout, but a selection of motifs is singled out in a separate chapter. The study also investigates Eliot's and Wagner's relation to art and life from religious (Buddhism and Christianity) and philosophical (Schopenhauer and Nietzsche) paints of view. Where relevant, some biographical data shedding light on their arts are touched upon--e.g. their personal (including marital) experiences and their anti-Semitism. The concluding chapter rounds off the subject by mainly offering some possible reasons for Eliot's obscure and neglected Wagnerism.

Toda, K. "T.S. Eliot and Renaissance drama." Thesis, University College London (University of London), 2015. http://discovery.ucl.ac.uk/1463322/.

Full text
Abstract:
The crucial importance of non-Shakespearian Renaissance dramatists to T.S. Eliot is evident in both his poetry and his prose. Eliot himself drew attention to this: he credited his own ‘poetic formation’ to the ‘minor Elizabethan dramatists’, and when reviewing his ‘critical output for the last thirty-odd years’ in 1951, he confessed himself ‘surprised to find how constantly I have returned to the drama, whether by examining the work of the contemporaries of Shakespeare, or by reflecting on the possibilities of the future.’ As C.S. Lewis disapprovingly wrote, Eliot’s ‘sympathy with depraved poets (Marlowe, Jonson, Webster) is apparent’. This thesis will trace Eliot’s engagement with these dramatists; it is a topic that has been comparatively neglected despite the central role it played in the evolution of his poetic and critical sensibility. The first section, which is largely biographical, explores Eliot’s background and education for clues to the development of his great interest in Renaissance drama, as well as detailing the ways in which he pursued this interest when he moved to England. The second comprises a detailed study of Eliot’s many essays on Renaissance dramatists. The last section examines his poetry, from the juvenilia to The Waste Land of 1922, when the outward signs of his ‘saturation’ were particularly prominent. In my conclusion I discuss how his engagement with Renaissance drama evolved in his post-1922 poetry and culminated in the composition of his own verse plays. The aim of this thesis is to explore the nature of the ‘profound kinship’ Eliot shared with Renaissance dramatists; their work appealed to him because it combines erudition with emotion, refinement with savagery, levity with the macabre and squalid. This appeal was so strong that it powerfully shaped both his poetic ideals and his vision of modernity.

Abstract:
The present PhD. thesis deals with the systems of building services (namely heating, ventilating and air conditioning, shading) which provide required indoor microclimate inside the immovable heritage. The work thus combines the exclusively technical field of building services (BS) and the general principles of heritage conservation. The aim of my PhD. thesis is the analysis of immovable heritage conservation processes, focusing on the current state of research and BS systems documentation. Furthermore the work concerns the possibilities of temporary measurements of indoor climate parameters inside the immovable heritage, and aims to develop a computer model (in BSim software) for the simulation of various working conditions in the selected buildings. For the stated aims were selected three representative historic houses: historical assembly hall at the Faculty of Civil Engineering, Brno University of Technology; the Palm Greenhouse at Lednice Chateau in Moravia; and Villa Tugendhat in Brno. These buildings had been surveyed for several years, and in this PhD. thesis I present their analysis, evaluation, and conclusions. The aims of this PhD. thesis broadly correspond to the transnational objectives. The present research is, for example, in accordance with the international document ICOMOS Charter (Zimbabwe, 2013) which is concerned with the analysis, conservation, and restoration of architectural heritage. The research of immovable heritage is also supported by European Union, e.g. by the Seventh Framework Programme of EU.

Abstract:

Full text
Abstract:
We propose dynamic aggregation of virtual tags in TLB to increase its coverage and improve the overall miss ratio during address translation. Dynamic aggregation exploits both the spatial and temporal locality inherent in most application programs. To support dynamic aggregation, we introduce the use of ternary-CAM (TCAM) cells at the second-level TLB. The modified TLB architecture results in an increase of TLB reach without additional CAM entries. We also adopt bulk prefetching concurrently with aggregation technique to enhance the benefits due to spatial locality. The performance of the proposed TLB architecture is evaluated using SPEC2000 benchmarks concentrating on those that show high data TLB miss ratios. Simulation results indicate a reduction in miss ratios between 59% and 99.99% for all the considered bench-marks except for one benchmark, which has a reduction of 10%. We show that the L2 TLB when enhanced using TCAM cells is an attractive solution to high miss ratios exhibited by applications.

Abstract:
The OS (operating system) is the primary target of todays attacks. A single exploitable defect can be sufficient to break the security of the system and give fully control over all the software on the machine. Because current operating systems are too large to be defect free, the best approach to improve the system security is to reduce their code to more manageable levels. This work shows how the security-critical part of the OS, the so called TCB (Trusted Computing Base), can be reduced from millions to less than hundred thousand lines of code to achieve these security goals. Shrinking the software stack by more than an order of magnitude is an open challenge since no single technique can currently achieve this. We therefore followed a holistic approach and improved the design as well as implementation of several system layers starting with a new OS called NOVA. NOVA provides a small TCB for both newly written applications but also for legacy code running inside virtual machines. Virtualization is thereby the key technique to ensure that compatibility requirements will not increase the minimal TCB of our system. The main contribution of this work is to show how the virtual machine monitor for NOVA was implemented with significantly less lines of code without affecting the performance of its guest OS. To reduce the overall TCB of our system, other parts had to be improved as well. Additional contributions are the simplification of the OS debugging interface, the reduction of the boot stack and a new programming language called B1 that can be more easily compiled.

Although in recent years Adorno's work has been the object of increased scrutiny within Anglo-American philosophical circles, as yet little sustained attention has been devoted to the concept of memory within Adorno's oeuvre. However, in Dialectic of Enlightenment Adorno and Horkheimer proclaimed that it is "by virtue of this memory of nature in the subject" that "enlightenment is universally opposed to domination. "Given that all of Adorno's work is concerned to redeem enlightenment from domination, the importance of a philosophical interpretation of the concept of memory is pivotal for an engagement with the legacy of Adorno's thought today. It will be argued that, for Adorno, memory always operates in relation to reification. The construal of this relation enjoins the consideration of a number of significant categories within Adorno's work: notably tradition, experience, mimesis and utopia; and further, it serves to situate and distance Adorno from those thinkers - Kant, Hegel, Heidegger and Benjamin - with whom he incessantly engages. Finally, by focusing on the relation between memory and reification, one can gauge the stakes of the Habermasian critique of Adorno, for it is Adorno's understanding of reconciliation (utopia) as the "remembrance (Eingedenken) of nature in the subject" that is the crux of the agon between Habermas and Adorno. I will argue that it is Habermas's failure to fully engage with the ramifications of Adorno's concept of memory that vitiates his critique, and indeed, that this failure provides the means for an Adornian critique of Haberman. It will be argued that memory is not an object of Adornian thought, but rather, that it provides the utopian texture of that thought.

Wyatt, Simon. "The drums of the southern TRB." Thesis, University of Edinburgh, 2007. http://hdl.handle.net/1842/29429.

Full text
Abstract:
This dissertation examines the clay drums found in the Neolithic contexts of the southern Trichterrandbecher culture (TRB). Chapter 1 introduces the subject and provides basic definitions Chapter 2 reviews the archaeological literature of the southern TRB, including typological, stratigraphic and dating concerns, followed by a survey of the settlement, economic and burial evidence. Chapter 3 reviews the classifications of the drums and proposes a revised version, based on the vessel form, the decoration and the archaeological context. In Chapter 4 we review the comparative analysis of the decoration and vessel form with a view to understanding their origins. Chapter 5 summarises the anthropological and archaeological literature on the subject of shamanism, and shamanistic practices, reviewing the concept of altered states of consciousness and providing definitions of the terms "trance", "ecstasy", "shamanism" and "possession". Then we discuss the neurological evidence of shamanism and brain structure, and explain the technical terminology. The chapter then proceeds to examine the methods of inducing changes in consciousness, specifically the use of auditory driving and imagery cultivation, finishing with a look at the evidence of European shamanisms. Chapter 6 examines the validity of applying a shamanistic approach using the comparison with entoptic imagery. Chapter 7 summarises the investigation, and after sifting the evidence draws conclusions on the interpretation of the Neolithic drums, the plausibility of shamanistic approaches and the nature of music in human culture.

Asciuto, Nicoletta. "T.S. Eliot : turning darkness into light." Thesis, Durham University, 2015. http://etheses.dur.ac.uk/11007/.

Full text
Abstract:
This thesis examines the use of light and dark imagery throughout T. S. Eliot’s literary career; in his poetry (Inventions of the March Hare, Prufrock and Other Observations, The Waste Land, Ash-Wednesday, Four Quartets) and in his dramatic works (The Rock, Murder in the Cathedral, The Family Reunion). The aim of this thesis is twofold. Firstly, it aims to show how, by an attentive analysis of light and dark imagery in Eliot’s works, it is possible to discern a continuous pattern of light and dark correlation and opposition throughout his oeuvre, which can be interpreted in historical, biographical, and existential terms. Secondly, this thesis seeks to explain poetry qua poetry, with the light and dark imagery proving a remarkable path of investigation, in which to understand Eliot’s poetry and its important relationship with his own literary “debts”. If, on the one hand, many critics have considered the importance of light and dark imagery in Eliot’s most significant poetical work after his religious conversion, Four Quartets, as an important turn to mysticism and religion, this has too often been perceived as a sudden change in his beliefs and in his poetics, rather than as a gradual development. The presence of light and dark images in Eliot’s early poetry, as well as in his earlier masterpiece The Waste Land, shows how Eliot had consciously imagined a path of light throughout his oeuvre, demonstrating his own preoccupations with regard to soul, rationality, and religion from his very early years as a poet. This thesis thus wants to fill a gap in the field of Eliot studies, where the importance of light and dark imagery in Eliot’s early poetry, as well as in his drama, and the connection of light and dark in Four Quartets with his other literary works, has been underestimated by previous scholars.

Micaković, Elizabeth Joan. "T.S. Eliot's voice : a cultural history." Thesis, University of Exeter, 2015. http://hdl.handle.net/10871/18902.

Full text
Abstract:
This thesis is a diachronic account of T. S. Eliot’s speaking voice, which, over fifty years, developed into the meticulously crafted tool of the twentieth-century author and critic and the politically and socially powerful instrument of the public intellectual. Eliot’s voice, although certainly the offspring of the nineteenth-century marriage of authorship as a bona fide profession and oral performance, was, however, unique in its responsiveness to twentieth-century legal and political debates on national identity and stability, copyright, and the powerful potential of recording technologies to both disseminate an author’s words almost exponentially whilst simultaneously encroaching on the traditional material of authorship: print. Indeed, what underpins this thesis is the argument that he was both fascinated by and actively involved in shaping those very discourses on the authority of the spoken voice in the belief that the power of the spoken word, and ultimately of his own voice, held an unrivalled ability to impact on social behaviour and national stability.

Abstract:
Twenty weeks ago I got the honor to pursue a project for ABB Crane Systems. After success with an innovative direction on their Remote Control Station they wanted a continuation of industrial design in another new product development. The aim was to specify the customer needs and create a concept to a Terminal Test Box (TTB), with hopes to gain momentum and funds for continuation of the project.   The terminals earn their money based on productivity of the quay, meaning that the number of ship-to-shore lifts per minute becomes very important. Clients have requested a solution where they can simulate their cranes to ensure function. At the moment approximately five prototypes have been delivered, for example to Peel Ports in Liverpool and Lazaro Cardenas in Mexico. The current Test box lack a proper casing and user adapted features. Based on interviews with personnel at Crane Systems representing linked/adjacent work with TTB the customer needs was determined to: a solution adapted to commissioning phase,a process panel easy to reach, access to components without exposing them, oversight of components and ability to move TTB. The test box is used either standing on the floor or placed on a table. Placed on the floor the operator must twist and bend to access the box, leading to an unpleasant working posture. Set on the table the box is bulky.   The initial idea development resulted in three potential positions and seven initial concepts. Of these seven three ideas was conceptualized and graded in Pughs matrix. The concept Stand high offered a convenient and ergonomic solution independent of the available interior in commissioning phase. However Stand high had weaknesses where the other two concepts Cross and Transparent showed strengths and therefore the three concepts were combined into one. Knowledge about the included components such as an industrial computer and power supply and their requirements has driven the process.   TTB will be produced in approximately ten units per year. The slim edition made it alluring to look into state of the art manufacturing such as Additive Manufacturing compared to the more traditional use of shielded metal. Possibilities regarding the small series can be used by ABB for continuously trying the product towards the customer and offer a flexible product that can be individually adapted to some extent.   The final concept of TTB is improved compared to the Test box looking at horizontal and vertical spacing between the components, which also enables cable channels. The process panel included has an inclination of 15 degrees to avoid reflections as much as possible and to create an ergonomic working position. It has a see through front door giving access and viewing of the components. A combined CNC and 3D-printing prototype/product from the company HLH in aluminum and Plexiglas would cost approximately 24 750 SEK. Manufacturing in shaped sheet metal at Sweco (without a see through door) would cost approximately 10 000 SEK. This is creating an exciting situation, where the recommendation is to create a prototype to further evaluate the concept into completion.

Frendo, Maria. "T.S. Eliot and the music of poetry." Thesis, Durham University, 1999. http://etheses.dur.ac.uk/4565/.

Full text
Abstract:
This thesis is a study of T.S. Eliot's poetry in the light of the different ways in which it can be considered 'musical'. Two concerns central to the thesis are: (1) Eliot's enduring interest in the musical quality of poetry; (2) the critical usefulness and viability of drawing analogies between his poetry and music. The thesis considers three important related topics: (1) Eliot's preoccupation with language, its inevitability and its inadequacy; (2) the figure of the seeker in his poetry; (3) his interest in mysticism. The thesis begins by exploring affinities between music and literature in the context of Wagner’s ideal of the 'Gesamtkunstwerk' and its influence on French Symbolist writers. It goes on to trace the development of T.S. Eliot's poetic style as influenced by the French Symbolist poets, by Dante and the mediaeval mystics, and by the music of Wagner, Stravinsky and other composers. Throughout, Eliot's poetry presents variations on the theme of detachment and involvement in relation to the figure of the seeker: consciousness is most engaged and challenged when it journeys. In the early poetry, music serves to emphasize failed relationships: the closer the physical proximity between protagonists, the greater the psychological distance. From The Waste Land on, Eliot makes use of myth and leitmotif to portray consciousness in the role of seeker urged on by the need for meaning. After his conversion to Anglo-Catholicism in 1927, Eliot's characters embark on a journey inward, where music, now "unheard", no longer signifies neurosis and despair, but becomes the only language for the ineffable.

The thesis is focused on the determination of the thermal conductivity coefficient on the group of thermoplastics used for 3D printing (PLA, ABS, PETG). The thesis describes the materials used for 3D printing and the design of air conditioning for the grocery store. A stationary method was used for the solution. The heat flux passing through the sample was measured and the thermal conductivity coefficient was calculated. By the chosen method, we determined the coefficient is = 0,11-0,13 W/(m·K). Comparison with the available results of other methods and authors has shown that the resulting coefficient is lower. Based on the data, it is possible to compare the thermal properties of 3D printing materials.
