Cytotoxic T lymphocytes (CTL) are major effector cells in the adaptive immune response against intracellular pathogens and cancers, killing targets with high precision. Precision is achieved through the specificity of the clonally expressed T cell receptor (TCR). TCRs recognise a specific peptide chain loaded into a major-histocompatability complex, triggering signalling, inducing the CTL to attach and kill target cells. TCR signal strength is critical for CD8+ T cell clonal expansion after antigen stimulation. Levels of the transcription factor IRF4 control the magnitude of this process through induction of genes involved in proliferation and glycolytic metabolism. The signaling mechanism connecting graded TCR signaling to the generation of varying amounts of IRF4 is not well understood. This thesis describes the evolution of Trellis Coded Modulation (TCM) technique and then develops the Analytically Described Trellis Codes with Partially Overlapped Signal Constellation for AWGN channels. This development is shown to obtain a performance gain over the TCM with conventional signal constellation. The new signal constellation is first introduced. A few examples (for one-, two-, and high-dimensional signal sets) are then given to show the performance gain due to the overlap of signal constellation. Bit error probability analysis is carried out for these examples.

Tsai(1996) reported that Ha-ras oncogene overexpression and Insulin growth factor-1(IGF-1) stimulation were both required for 2-12 cells to form colonies in soft agar containing 10%AC-2TM(collistrum, a substitute of serum). Using this model we analyzed colony formation related signal transduction of 2-12 cells in 10% AC-2TM containing medium.
