Abstract:
In this thesis, a survey design was used following closely the objective of descriptive research. The idea was to take a sample of the population of teachers in the area, gather demographic data, and find out if the reasons they fail to fully integrate technology into their classrooms can be attributed to skill deficiency, motivation, lack of environmental support or a combination thereof.

Abstract:
Modern manufacturing processes require better quality control of the manufactured products at a faster rate, for achieving good throughput. This is increasing the need for process-oriented precision metrology capable of providing faster inspection and yielding valuable feedback to the manufacturing system for quality control of the manufactured products. Over the past twenty years optical sensors have emerged as a preferred method for the measurement applications in manufacturing automation, owing to some inherent advantages, such as high speed, high resolution, non-contact operation, and low cost. Improved online optical sensors for surface measurement would enable incorporating measuring systems into production processes and machines, improving the production performance and the quality of products, which is highly desired in many high/ultraprecision manufacturing applications. This thesis presents a novel spatially dispersed short coherence interferometry (SDSCI) sensor system for online surface measurement applications on the nanoscale. The SDSCI sensor system uses a low-cost broadband super-luminescent diode (SLD) with an emission bandwidth of 25 nm. Two measurement methods, phase shifting interferometry and Fourier transform for surface profile measurements, have been investigated in this study. The metrology sensor system incorporated the Michelson interferometer configuration with an optical probe in the measurement arm, while the reference arm had a mirror with a piezo-electric transducer. The technique involves surface scanning by spatially dispersing the broadband light using a reflective grating and a scan lens, and recording the resulting interferogram by using a high-speed spectrometer. The first measurement method involved investigations of implementation of phase shifting interferometry and the Carré algorithm for phase retrieval from the measured phase-shifted interferograms for profile measurements. Standard diamond turned multi-stepped and NPL artefact samples with 550-nm and 100-nm-high steps, respectively, were measured and confirmed the capability of the measurement sensor. The measurement speed of this technique was limited by the spectrometer speed and by the piezo-electric transducer movement. The optimised system has a measurement time of 1s. The second method was then investigated based on the Fourier transform profilometry technique for further increasing the measurement speed of the sensor device, as it required a single-shot interferogram, alleviating the need of any phase shifting. With increased measurement speed, this technique further reduced the problem of environmental noise inherent to all interferometer-based systems. Similar artefacts were measured by using this technique for evaluating its applicability for surface profile measurements. Once the sensor system was optimised and calibrated, the resulting open-space system could be further miniaturised into a compact sensor system by using optical fibres with a remote probe connected via a fibre link for use in embedded metrology applications. This method will be very beneficial in online inspection of samples in rollto-roll manufacturing processes, where the measurand is constantly moving. An example of such a measurement challenge is detection of defects on vapour barrier films formed by depositing an aluminium oxide layer several tens of nanometres thick on a flexible polymer substrate. Effective detection and characterisation of defects in this layer requires a single-shot approach with nanometre-scale vertical resolution.

Abstract:
This thesis addresses a problem related to usage analysis in information retrieval systems. Indeed, we exploit the history of search queries as support of analysis to extract a profile model. The objective is to characterize the user and the data source that interact in a system to allow different types of comparison (user-to-user, source-to-source, user-to-source). According to the study we conducted on the work done on profile model, we concluded that the large majority of the contributions are strongly related to the applications within they are proposed. As a result, the proposed profile models are not reusable and suffer from several weaknesses. For instance, these models do not consider the data source, they lack of semantic mechanisms and they do not deal with scalability (in terms of complexity). Therefore, we propose a generic model of user and data source profiles. The characteristics of this model are the following. First, it is generic, being able to represent both the user and the data source. Second, it enables to construct the profiles in an implicit way based on histories of search queries. Third, it defines the profile as a set of topics of interest, each topic corresponding to a semantic cluster of keywords extracted by a specific clustering algorithm. Finally, the profile is represented according to the vector space model. The model is composed of several components organized in the form of a framework, in which we assessed the complexity of each component. The main components of the framework are: - a method for keyword queries disambiguation; - a method for semantically representing search query logs in the form of a taxonomy; - a clustering algorithm that allows fast and efficient identification of topics of interest as semantic clusters of keywords; - a method to identify user and data source profiles according to the generic model. This framework enables in particular to perform various tasks related to usage-based structuration of a distributed environment. As an example of application, the framework is used to the discovery of user communities, and the categorization of data sources. To validate the proposed framework, we conduct a series of experiments on real logs from the search engine AOL search, which demonstrate the efficiency of the disambiguation method in short queries, and show the relation between the quality based clustering and the structure based clustering

Abstract:
The pulp- and paper industry is a key industry globally and stands for a production of 600 million tonnes pulp and paper worldwide with a total revenue of 400 billion dollars. Due to high quality requirements on paper, it is important to use instruments that control that the produced paper fulfill the promised requirements. To control the quality, a sample strip needs to be taken along the full length of each paper reel. The purpose of this master’s thesis project was consequently to develop a unit that will be used when cutting out these samples. The project started with a pre-study on existing sample cutters to define all integrated subsystems and their functionalities, advantages and drawbacks. The essential subsystems were found to be: (1) paper reel cutting, (2) path movement, (3) motion generation and (4) mechanical transmission. The advantages and drawbacks were found by interviewing people who have used the cutters or in any way have encountered them for opinions, experience and knowledge. Sample winding is an additional subsystem which was found in a few of the existing cutters but was not further studied. After defining were the problems and development potentials lay, a range of concepts were generated for the subsystems. These concepts were presented to a defined target group to ensure a unit which will create costumer value. By taking their views and ideas into account, further concept development was made. After a few iterations, one concept was chosen for each subsystem in an evaluation with domain experts. A detail study and design were then made which incorporated all subsystems into one unit. The solution: (1) has two rotating circular blade pushed against sharp guide rails, (2) is hand-held with two pair of wheels on the cutter-head and a digital inclinometer, (3) has a manually generated motion and (4) uses a synchronous belt drive which transfer the manually generated motion to the rotating blades. In addition to this, complete 2D and 3D drawings, along with a bill of materials, were delivered for the future manufacturing of the unit. Since this thesis presents no solution for the sample winding, the next step is to develop a collector which is easily handled and should be able to co-use the manually generated power with the rotating blades. Further, a design which allows modularization of the unit should be developed.

Parton, Becky Sue. "Technology adoption and integration levels: A comparison study between technology-minded general educators and technology-minded deaf educators." Thesis, University of North Texas, 2006. https://digital.library.unt.edu/ark:/67531/metadc5352/.

Full text
Abstract:
The purpose of this study was to determine whether working in the field of deaf education, as opposed to general education, results in a higher level of technology integration. A secondary goal was to determine if deaf educators who are deaf integrate technology at a higher level than their hearing counterparts. The instrument chosen for this study was the LoTi Technology Use Profile, a tool used to explore the role of technology in the classroom. A total of 92 participates were included in the study of which 48 were regular educators and 44 were deaf educators. The participants were selected from a population pool whereby teachers were presumably pre-disposed to using technology based upon their attendance at a technology training session in the form of a conference or a class. Deaf educators as a whole did not perform as well as general educators on the LoTi scales. Given the fact that the technology-minded general educators who comprised the sample population of this study scored exceptionally high on the LoTi scales, further research is needed to ensure comparability between the two groups. The findings of the current study do suggest, though, that deaf educators who are deaf have the potential to integrate technology to a greater degree than deaf educators who are hearing. Thus, a primary recommendation is to conduct a national LoTi survey of typical, rather than technology-minded, deaf educators as a comparison to the 2004 national survey of typical general educators.

Al-Hinai, Sulaiyam. "Non-invasive velocity and volume fraction profile measurement in multiphase flows." Thesis, University of Huddersfield, 2010. http://eprints.hud.ac.uk/id/eprint/9094/.

Full text
Abstract:
Multiphase flow is the simultaneous flow of two or more phases, in direct contact, and is important in the oil industry, e.g. in production wells, in sub-sea pipelines and during the drilling of wells. The behaviour of the flow will depend on the properties of the constituent phases, the flow velocities and volume fractions of the phases and the geometry of the system. In solids-in-liquid flows, measurement of the local solids volume fraction distribution and the local axial solids velocity distribution in the flow cross section is important for many reasons including health and safety and economic reasons, particularly in oil well drilling operations. However upward inclined solidsliquid flows which are frequently encountered during oil well drilling operations are not well understood. Inclined solids-liquid flows result in non- uniform profiles of the solids volume fraction and axial solids velocity in the flow cross- section. In order to measure the solids volumetric flow rate in these situations it is necessary to measure the distributions of the local solids volume fraction and the local axial solids velocity and then to integrate the product of these local properties in the flow cross section. This thesis describes the development of a non-intrusive Impedance Cross-Correlation (ICC) device to measure the local solids volume fraction distribution and the local solids axial solids velocity distribution in upward inclined solids-water flows in which these distributions are highly non-uniform. The ICC device comprises a non-conductive pipe section of 80mm internal diameter fitted with two arrays of electrodes, denoted „array A‟ and „array B‟, separated by an axial distance of 50mm. At each array, eight electrodes are equispaced over the internal circumference of the pipe. A control system consisting of a microcontroller and analogue switches is used such that, for arrays A and B, any of the eight electrodes can be configured as an "excitation electrode" (V+), a "virtual earth measurement electrode" (Ve) or an "earth electrode" (E) thus enabling the local mixture conductance in different regions of the flow cross-section to be measured and thereby allowing the local solids volume fraction in each region to be deduced. The conductance signals from arrays A and B are also cross-correlated to yield the local solids axial velocity in the regions of flow under interrogation. A number of experiments were carried out in solids-in-water flows in a flow loop with an 80 mm inner diameter, 1.68m long Perspex test section which was inclined at three different inclination angle to the vertical ( o 0 , o 15 and o 30 ). The obtained results show good quantitative agreement with previous work carried out using intrusive local probes. Integration of the flow profiles in the cross section also yielded excellent quantitative agreement with reference measurements of the mean solids volume fraction, the mean solids velocity and the solids volumetric flow rate. Furthermore, this study also showed good qualitative agreement with high speed film of the flow. It is believed that the method of velocity and volume fraction profile measurement described in this thesis is much simpler to implement, more accurate and less expensive than the currently very popular technique of dual-plane Electrical Resistance Tomography (ERT). Finally, the thesis describes a mathematical model for predicting the axial velocity distribution of inclined solids-water flows using the solids volume fraction profiles measured by the ICC device. Good agreement was obtained between the predicted velocity profiles and the velocity profiles measured using the ICC device.

Möller, Niklas. "Development of an automatic autonomous sensor carrier for sound profile measurement in deep sea." Thesis, KTH, Marina system, 2016. http://urn.kb.se/resolve?urn=urn:nbn:se:kth:diva-203810.

Full text
Abstract:
This Master Thesis main purpose was to answer the question "Can you measure or calculate the velocity sound profile while performing a bathrymetric survey in an offshore environment like the North Sea, without any interaction nor modification of the existing equipment?".This, since today underwater surveys are a complex and expensive operation to perform where you either are mapping the sea floor or on a searching mission for a sunken wrecks. To achieve this successfully, one has to ensure that the accuracy of the position for every discovery or created map over the sea floor is entirely correct. This is an identified problem since the bathrymetric device sends its position by sonar, which relies in the sound propagation velocity, which in return varies with the water density. In order to increase the accuracy one need to determine the water density for all depths, i.e measure the salinity and temperature between the towing ship and the device that travels close to the sea bed. This because of layers consisting of fresh water and saltwater that never entirely mixes with each other in the ocean. The outcome of this project is a manufactured conceptual design of an autonomous sensor carrier that has the ability to measure temperature to a theoretical maximum depth down to 150m. It ascends and descends autonomously with an propagation speed of 0.54m/s in a static condition along an existing tether line, connected to a bathrymetric device that follows the sea floor. The sensor carrier ascend and descends its motion with help of two connected drive wheels powered by an electric motor, combined with two hall sensors to to reverse its movement when reaching desired depth. It has the ability to store sampled data onto a removable SD card, with a theoretical maximum endurance of 6,2km and it can be handled by one single person.Unfortunately, the concept as a whole is not entirely successful, and must therefore be supplemented within some areas. The major occurrence is that the drive mechanism tender to slip along the tether when climbing in vertical direction with a risk of damaging the tether coating. Furthermore one needs to increase the operational depth rating. This to be able to utilize the sensor carrier at all depths in the North Sea and also the Baltic Sea. However, the project as whole has achieved a solid framework and platform ready to be developed further in a future second version.

Abstract:
FundaÃÃo Cearense de Apoio ao Desenvolvimento Cientifico e TecnolÃgico
It aims to analyze the socioeconomic profile of producers of irrigated fruit in the Cariri and measure the technological level, identifying the factors that influence each level. The measurement of technological index of fruit growers allows you to identify the level of technology used by producers. Have the technical efficiency scores of caririenses fruit growers and the identification of the effects of socioeconomic variables on efficiency levels allowed capture the impact of these determinants along different points of the conditional distribution of technical efficiency. In preparing the technological index was used factor analysis, and to measure the index of efficiency scores were applied in determining the analysis of data envelopment analysis (DEA) models and quantile regression. Came indicators of primary sources, collected in 2014, directly, with 86 growers located in six municipalities of the Cariri. The results show that in most cases, the growers (53.49%) have only to primary education, and only 16.28% can only sign his name. You can still see that 31.40% of producers have secondary education and that is down the number of illiterate farmers (3.49%) and they have 30-50 years of age (66.67%) and also that 75 , 68% of growers have over five years experience in the activity. The results also show that 42.66% of the fruit growers of the region not participating unions have no access to any type of credit; 72.73% of these already, participating cooperatives or associations, hold some kind of credit. For the technological indexes classified as very low, there were two growers, 30 of them were considered low-tech, 53 fruit growers have considered average index and fruit grower demonstrated considered high index, 0.62 to 0.78, with relative frequency of 1, 16%. There were no fruit growers with very high rates for technology adoption. The variables that contributed most to indicate adoption of technological improvements in that index were Total Area, Gross Income and Total Production, result justified by the increased variance indicated by the data, a total of 24.058%, represented by the factor 1 (intensive Productivity) technology. The quantile regression was estimated, considering the technical efficiency scores obtained in the constant returns to scale model, the results show that the explanatory factors of technical efficiency for the group of more efficient producers are important in explaining the efficiency of this group. For the less efficient growers group, it is observed that among the six explanatory variables, only management index, credit and education played irrelevant role in explaining variations in technical efficiency level, since the coefficients of these variables were not statistically significant, in this case, there is no difference between fruit growers with access to credit and those without and between grade levels, ie, if the producer has secondary or higher education or do not have that level of education.
Abstract:
The competence approach is basic for modern technologies of personnel selection and assessment. In the current conditions of Ukrainian business this approach is actively applied in various areas of professional activity. Given the competition on the market of professions, we can see that people do not stay in one position for long and the search for specialists is constantly on. However, this is not the fault of employees, because a large percentage of employers do not use the services of, or do not have in their company, a specialist in the HR department. Thus, competence profiles, on the basis of which potential employees could understand their own professional competence and employers could register and evaluate the competences required by their organisation, are not always applied.

Abstract:
The methodology to quantify vegetation from airborne laser scanning (or LiDAR - Light Detection And Ranging) is somehow consolidated, but some concerns are still in the checklist of the scientific community. This thesis aims to bring some of those concerns and try to contribute with some results and insights. Four aspects were studied along this thesis. In the first study, the effect of threshold heights (minimum height and height break) in the quality of the set of metrics was investigated aiming the volume estimation of a eucalyptus plantation. The results indicate that higher threshold height may return a better set of metrics. The impact of threshold height was more evident in young stands and for canopy density metrics. In the second study, the stability of the LiDAR metrics between different LiDAR surveys over the same area was analyzed. This study demonstrated how the selection of stable metrics contributed to generate reliable models between different data sets. According to our results, the height metrics provided the greatest stability when used in the models, specifically the higher percentiles (>50%) and the mode. The third study was designed to evaluate the use of machine learning tools to estimate wood volume of eucalyptus plantations from LiDAR metrics. Rather than being limited to a subset of LiDAR metrics in attempting explain as much variability in a dependent variable as possible, artificial intelligence tools explored the complete metrics set when looking for patterns between LiDAR metrics and stand volume. The fourth and last study has focused upon several highly important forest typologies, and shown that it is possible to differentiate the typologies through their vertical profiles as derived from airborne laser surveys. The size of the sampling cell does have an influence on the behavior observed in analyses of spatial dependence. Each typology has its own specific characteristics, which will need to be taken into consideration in projects targeting monitoring, inventory construction, and mapping based upon airborne laser surveys. The determination of a converged vertical profile could be achieved with data representing 10 % of the area for all typologies, while for some typologies 2 % coverage was sufficient.

Abstract:
Thesis (MTech (Business Administration))--Cape Peninsula University of Technology, 2019
Background: Retail in South Africa is a growing industry, but new demands and the influx of foreign competition require retail business managers to be more creative in solving modern-day complex retail problems. An embedded outcome of the registered National Diploma: Retail Business Management qualification offered by the Cape Peninsula University of Technology is that students should be able to identify business problems and creatively make sound business decisions to solve these problems. However, because creativity is such a diverse topic, it seems that educational institutions and educators do not commit themselves to and evade the development of creativity. Consequently, this leads to graduates being ill-prepared in creatively solving the complex and often unique business problems they encounter in the Wholesale and Retail sector. Objectives: The objective of this study was to determine the creativity profile of Retail Business Management students and evaluate the progress of their creativity levels from their first- to third year. This approach evaluated if the embedded outcome of creativity was met for the National Diploma: Retail Business Management qualification. Methodology: Empirical research was conducted by collecting a combination of quantitative and qualitative data to determine the creativity levels of Retail Business Management students. A questionnaire consisting of both qualitative- and quantitative questions was used to evaluate the creativity levels of 159 full-time students and 16 students from the retail industry studying through the university’s Retail Academy. Quantitative data were analysed using both descriptive- and inferential statistics. Findings: Based on the analysed data, it was found that creativity is one of the key attributes, if not the most important, to ensure continued business success in the South African retail industry. The results indicate that participants were creative, but not at the required levels. It was further established that there was no growth in the creativity levels of the participants from their first- to third year of studies, thus indicating that the required outcome of creativity, as stipulated in the qualification criteria, was not achieved. Recommendations: It is recommended that further research should be conducted in an attempt to propose a suitable creativity instrument for developing the creativity of Retail Business Management students as this would ensure that graduates are confident and able to use their creativity to address the unique challenges that the South African retail industry face.

Abstract:
I dagens medievardag blir rörlig bild allt mer vanligt.
The use of moving pictures has become far more common. Since more people use moving pictures it has become more important for film producers to distinguish themselves from their competitors with a visual identity. This study has been conducted in collaboration with Rockmotion Film HB, a company specialized in making videos for music and entertainment businesses. Since the beginning Rockmotion Film have been missing a complete visual identity which for a company is necessary to be seen as serious. The purpose with this study was to create a new visual identity and redesign the existing logotype based on Rockmotion Film’s core values and vision. The study began with a semi-structured interview with the company to gather necessary information about the company. In addition, a visual content analysis was made to gain perception of the competitors' visual identities. Through surveys and a focus group, the produced material has been tested to obtain a final suggestion. The result of this thesis is a simple and catching visual identity that complies with the core values, personal, youthful and professional as well as the client's vision of the visual identity.

Abstract:

Numerical procedures for reliable wheel and rail wearprediction are rare. Recent development of simulationtechniques and computer power together with tribologicalknowledge do however suggest computer aided wear prediction. The objective of the related research field at the RoyalInstitute of Technology (KTH) is to arrive at a numericalprocedure able to simulate profile evolution due to uniformwear to a degree of accuracy sufficient for application tovehicle dynamics simulation. Such a tool would be useful formaintenance planning as well as optimisation of the transportsystem and its components.

The research contribution accounted for in this thesisincludes, in addition to a literature review, refinement ofmethods applied to uniform wheel wear simulation by inclusionof braking and improvement of the contact model. Further atentative application to uniform rail wheel simulation has beenproposed and tested.

The first part addresses issues related to braking andwheel-rail contact conditions in the context of wheel wearsimulation. The KTH approach includes Archard’s wear modelwith associated wear maps, vehicle dynamics simulation andrailway network definition. In previous work at KTH certainvariations in operating conditions have been accounted forthrough empirically estimated average scaling factors. Theobjective of the current research is to be able to include suchvariations in the set of simulations. In particular theinfluence of disc braking and varying friction and lubricationconditions are investigated. Both environmental factors likemoist and contamination and deliberate lubrication need to beconsidered. As part of the associated contact analysis theinfluence of tangential elastic deformation of the contactingsurfaces on the sliding velocity has been separatelyinvestigated and found to be essential in case of partial slipcontact conditions.

In the second part validation of the improvements related towheel wear simulation is addressed. Disc braking has beenincluded in the simulation set and a wear map for moist contactconditions based on recent tribometer tests has been draftedand tested. It has been shown that the previously used brakingfactor accounts for the combination of the contributions fromsurface elasticity and braking. Good agreement withmeasurements from the Stockholm commuter service is achieved. It is concluded that the model improvements accounted for aresufficient for adequate simulation of tread wear but thatfurther development of the flange / gauge corner contactmodelling may be needed.

In the final part a procedure for simulation of rail wearand corresponding profile evolution has been formulated. Asimulation set is selected defining the vehicles running on thetrack to be investigated, their operating conditions, andcontact parameters. Several variations of input data may beincluded together with the corresponding occurrenceprobability. Trial calculations of four non-lubricated curveswith radii from 303 m to 802 m show qualitatively reasonableresults in terms of profile shape development and difference inwear mechanisms between gauge corner and rail head. The wearrates related to traffic tonnage are however overestimated. Itis believed that model refinements in terms of environmentalinfluence and contact stress calculation are useful to improvethe quantitative results.

QC 20100531

Gilliam, David R. "A study of contact between a profile meter stylus and polymer films on roughened substrates." Thesis, Virginia Tech, 1985. http://hdl.handle.net/10919/45727.

Full text
Abstract:
When a polymer film is deposited onto a roughened substrate, the film masks over the underlying substrate roughness. Then when attempting to measure the roughness of the polymer film surface using a stylus-type profile meter, the pressure between the tip of the stylus and the film is sufficient to plastically deform the polymer surface. The result is a possible erroneous measurement of the surface roughness of the polymer film. This thesis reports on attempts to quantify the actual roughness of the polymer film from the measurement obtained by the profile meter. Extensive surface profile data were collected and analyzed to determine the characteristics of the surface of the substrate and of the coating on the substrate. The tracks made by the stylus in the film were then observed in a scanning electron microscope, from which the depth of the stylus tracks were measured. The effects of the film thickness, stylus traversing speed, and substrate roughness on the stylus penetration depth, the variation in the depth, and the measured film roughness are assessed and discussed. It was found that using the fastest stylus traversing speed minimizes the variation of the stylus penetration depth and thus results in the most accurate measurement of the film surface. To predict this stylus indentation depth, a plane strain plastic deformation model is developed using slip-line field theory. It is found that the slip-line model gives good estimates of the stylus indentation depth when the film thickness is large.
Master of Science

Venter, Tania. "Modelling the catabolite and microbiological profile of cheddar cheese manufactured from ayrshire milk." Thesis, Bloemfontein : Central University of Technology, Free State, 2010. http://hdl.handle.net/11462/127.

Full text
Abstract:
Thesis (D. Tech.) -- Central University of Technology, Free State, 2010
Branded dairy products have lately become a global trend. As a result of this, the origin of the milk used in the manufacturing of branded cheeses must be declared by the producer, since it is known that these products are highly adulterated with foreign milk. In South Africa, branded Ayrshire Cheddar cheese has become highly popular due to its unique organoleptic properties and in light of claims that it ripens much faster than cheese made from other milk (not including Ayrshire). This study was therefore directed to investigate the unique properties of branded Ayrshire Cheddar cheese versus Cheddar cheese manufactured from a mixture of other breeds’ milk (not including Ayrshire milk) and to establish a catabolite profile for each cheese type. The outlay of the thesis was constructed into six chapters each with its own outcomes. The first chapter focused on the variations between the two Cheddar cheese batches (produced from Ayrshire and other breeds’ milk) with regards to organic acid, selected chemical parameters and starter microbiotia. In the following three chapters mathematical models were developed that would predict organic-; fatty and amino acid fluxtuations respectively in the cheese made from Ayrshire and other milk. In the last chapter two artificial neural networks were designed with the two starter organisms, Lactococcus lactis and Streptococcus thermophilus as variable indicator respectively. Thirty-two cheese samples of each batch (pure Ayrshire (4) / mix breed with no Ayrshire (4)) were ripened and samples were analysed under the same conditions on the following days after production: 2, 10, 22, 36, 50, 64, 78, and 92. In the subsequent chapters, the following analysis were done on each day of analysis: organic acid by means of high performance liquid chromatography (HPLC); fatty acids by means of Gas Chromatography Mass Spectometry (GCMS); amino acids by means of GC-MS; microbial analysis by means of traditional methods, total DNA extraction and polymerase chain reaction (PCR); and standard chemical analysis for moisture, NaCl and pH. In the first research chapter, the minimum and maximum (min/max) values, standard deviations and proposed rel X values of organic acids were evaluated in Ayrshire and the mixed-breed Cheddar cheese, and showed that isovaleric acid is the organic acid with the least variation relative to concentration in both cheeses and it was assumed that this organic acid is the most effective indicator of cheese uniformity. Clear differences in organic acids, chemical variables and starter micro-organisms were also evident in the two cheese batches. Results obtained from the regression models which was defined for each organic -; amino - and fatty acid by means of mathematical equations can be used by the manufacturer to achieve i.e. the selection of cheese for specialist lines, the early exclusion of defective cheeses, and the establishment of brand origin (Ayrshire vs. mixed-breed Cheddar cheeses). The regression graphs also illustrate unique flux patterns in Ayrshire and the mixed-breed in terms of organic -, fatty -, and amino acid content. In the last chapter, the discrimination between the two batches was respectively done via artificial neural network (ANN) modelling of Lactococcus lactis and Streptococcus thermophilus as indicator organisms. The ANN consisted of a multilayered network with supervised training arranged into an ordered hierarchy of layers, in which connections were allowed only between nodes in immediately adjacent layers. The construction thereof allowed for two output nodes, connected to an input layer consisting of two nodes to which the inputs were connected. In both cheeses the results from the ANN showed acceptable classification of the cheeses based on the counts of L. lactis and S. thermophilus.

Full text
Abstract:
Asymmetric cell division is an important mechanism for the differentiation of cells during embryogenesis and cancer development. Saccharomyces cerevisiae divides asymmetrically and is therefore used as a model system for understanding the mechanisms behind asymmetric cell division. Ace2p is a transcriptional factor in yeast that localizes primarily to the daughter nucleus during cell division. The distribution of Ace2p is visualized using a fusion protein with yellow fluorescent protein (YFP) and confocal microscopy. Systems biology provides a new approach to investigating biological systems through the use of quantitative models. The localization of the transcriptional factor Ace2p in yeast during cell division has been modelled using ordinary differential equations. Herein such modelling has been evaluated. A 2-compartment model for the localization of Ace2p in yeast post-cytokinesis proposed in earlier work was found to be insufficient when new data was included in the model evaluation. Ace2p localization in the dividing yeast cell pair before cytokinesis has been investigated using a similar approach and was found to not explain the data to a significant degree. A 3-compartment model is proposed. The improvement in comparison to the 2-compartment model was statistically significant. Simulations of the 3-compartment model predicts a fast decrease in the amount of Ace2p in the cytosol close to the nucleus during the first seconds after each bleaching of the fluorescence. Experimental investigation of the cytosol close to the nucleus could test if the fast dynamics are present after each bleaching of the fluorescence. The parameters in the model have been estimated using the profile likelihood approach in combination with global optimization with simulated annealing. Confidence intervals for parameters have been found for the 3-compartment model of Ace2p localization post-cytokinesis. In conclusion, the profile likelihood approach has proven a good method of estimating parameters, and the new 3-compartment model allows for reliable parameter estimates in the post-cytokinesis situation. A new Matlab-implementation of the profile likelihood method is appended.

Abstract:

From an environmental- and health perspective, biogas and other biomass-based fuels have several advantages; nevertheless the majority of motorists fill their cars with petroleum-based fuels. This thesis is designed to explore the profile of biogas in relation to its perceptions. It is a study concerning the communication between the biogas producing company Svensk Biogas and their biogas users and non biogas users. To obtain a thorough understanding of the profile and perceptions of biogas a qualitative approach was considered appropriate. Biogas users and non-users were interviewed at gasoline stations, while Svensk Biogas was interviewed as a group.

The three interview segments were analyzed and compared in order to identify patterns, similarities and differences. Based on research data the thesis concludes that the profiling arguments of biogas correlates to that biogas is the most environmentally friendly fuel, the least expensive fuel, and locally produced. Furthermore, the company profile of Svensk Biogas is equal to sustainable alternative, locally produced, trustworthy, environmentally friendly and climate smart [klimatsmart]. Given the arguments of the company profile, environmental values seem to be the core communicating value. Profiling Svensk Biogas happens through events and by using communication material such as company logotype.

Motorists have an overall positive perception of biogas. Biogas users states environmental benefits as the key argument behind their commitment. Non-users are positive toward biogas although expressing a lack of knowledge confusing biogas with ethanol and bio-fuels in general. According to motorists the negative perceptions, in addition to the prerequisites of biogas, are connected to insufficient infrastructure of biogas filling stations, a short range of the biogas tank, a high investment cost of a biogas car, a biogas price increase, scarcity of cars, and information (lack of information and misleading information).

The overall perception of Svensk Biogas among biogas users is positive. Biogas users express a negative perception concerning the Svensk Biogas filling stations and also wish for a lower biogas price. Non-users express modest perceptions of the company. This research also concludes that perceptions of the biogas producer are correlated to the perceptions of biogas. Furthermore, biogas producer, users and non-users wish to be directed by political decisions, guiding them toward environmentally friendly fuel alternatives.

Abstract:
Mestrado em Biologia Molecular e Celular
The Attention Deficit Hyperactivity Disorder and Dyslexia are neurodevelopmental diseases with a very high prevalence in children which often persist into adolescence and adulthood. With a difficult diagnosis, based on scales quantifying the signs and symptoms, a method of reliable, noninvasive diagnosis, suitable for children is urgent. A diagnosis using saliva samples quantifying salivary biomarkers is ideal for this study population. The establishment of the standard operating procedure with procedures for collecting, processing and storage of samples is essential to use saliva. Furthermore, since there appears to be a relationship between cognitive diseases and inflammation, the evaluation of the protein profile and quantification of cytokines related to inflammation are a starting point to understand the pathology. The evaluation of the chemokine (c-c motif) ligand 13 (CCL13) and of the chemokine (c-c motif) ligand 3 (CCL3), present in inflammation, was done using multiplex technology. The levels of these two chemokines are increased in multicompromised individuals which leads us to infer that neuroinflammation is present in these pathologies. Future studies should be done to validate inflammatory biomarkers in these diseases.

Abstract:
With Northvolt’s aggressive plan to establishEurope’s largest battery factory in northern Sweden, the needto build a pilot plant to demonstrate the possibilities of thecompany was needed. Quality, performance and cycle-life arekey factors customers value particularly high when purchasingbatteries, thus, a need for validating these parameters emerges.Performance and life-cycle tests are performed in high powerconsuming cycling units divided into the major components -chamber and cycler. The cycler is programmed according tospecific charge and discharge patterns (flows).A validation facility has been established close to NorthvoltLabs in V¨aster°as, housing cycling units for different cell formfactors. The major scope of this thesis was to find an optimalway to effectively and energy conscientious conduct operationsin the facility. One part included testing the possibility tore-utilize energy from discharging cells in simultaneouslycharging cells as well as testing if the equipment can provideenough energy to sell back to the electricity distributor.Additionally, an analysis to see if advanced planning can beused to balance the load profile of the validation facility andreduce energy costs was researched. A second part was todetermine if the equipment imposed harmonics on theconnected grid, and to decide if a filter is needed forPQ-compensation/harmonic filtering. Estimating the powerdemand of validation facility was needed to provide the gridowner an energy forecast. Firstly, to be able to analyze thesequestions, the power profile of the cycling units had to beestablished through measurements performed by installingpower quality devices. It was determined that the chambersconsumption was considerably higher than previously assumedand was in fact higher than the average charge-, anddischarge-flow performed by the cyclers. A power profile wasestablished for each type of cycling unit which was utilized in aMonte Carlo model to predict the power need of the facilitybased on new equipment arriving over the next couple of years.A basic energy cost return-of-investment-calculation concludedthat installing an high energy battery energy system was noteconomically feasible due to prevalent circumstances.

Abstract:
The goal of this project was to investigate how elution buffer properties affectthe protein elution in the polishing purification step with a cation exchange chromatographyconsisting of POROS XS resin. Hence be able to better predict thebehavior of the resin, two model proteins were used for this investigation (K100and Y102). This project included lab experiments (small scale), and design ofexperiments using MODDE software to evaluate the optimal conditions. Thefactors pH, conductivity, sample load and buffer concentration were investigatedon antibody Y102 while only pH and conductivity were investigated as factorsfor antibody K100. The evaluated responses were the elution volume, aggregateamount and the amount of monomeric antibody (target protein). The results indicatedthat the elution behaviour for POROS XS resin was antibody-dependentand not general for the resin. The responses obtained different models for thetwo antibodies investigated. However, since the aggregate and monomeric proteinfor K100 had curvature indications more experiments should be performedto establish the different behaviour of aggregate and monomer for the respectiveproteins.

Abstract:
The main aim of this thesis was to understand the mechanism behind the classical transition scenario inside the boundary layer over an airfoil and eventually attempting to control this transition utilizing passive devices for transition delay. The initial objective of analyzing the transition phenomenon based on TS wave disturbance growth was conducted at 90 Hz using LDV and CTA measurement techniques at two different angles of attack. This was combined with the studies performed on two other frequencies of 100 and 110 Hz, in order to witness its impact on the neutral stability curve behavior. The challenges faced in the next phase of the thesis while trying to control the transition location, was to understand and encompass the effect of adverse pressure gradient before setting up the passive control devices, which in this case was miniature vortex generators. Consequently, several attempts were made to optimize the parameters of the miniature vortex generators depending upon the streak strength and stability. Finally, for 90 Hz a configuration of miniature vortex generators have been found to successfully stabilize the TS wave disturbances below a certain forcing amplitude, which also led to transition delay.

Fyra huvudteman kunde identifieras: 1. Målgruppsanpassning 2. Brist på information 3. Struktur och informationssökning 4. Tydlighet i regler.

Abstract:
The expression of visual design have sins long been important in graphic design because it conveys so much more than the eye see. A brand does not only represent the name of the company but also its purpose and idea. Therefore is it so important that the visual profile radiates the organization's vision in a clear way and gives the viewer a pleasant overall experience. Many times can it help to freshen up a brand's visual identity in order to successfully launch the company again and to some extent also increase its consumption. This essay presents the production and the designprocess of Sveriges Kommunikationsbyråers ( Komm! ) Titan award. Komm! thought the Titan award needed to have its own identity and gave me the task to create this. The current logo was not harmonious or particularly well executed and therefore did the company demand a re-production of the Titan award. The criteria for the reproduction of the Titan award was that they wanted a clean, clear and creative new design solution. An interview and content analysis have been performed to get the results presented at the end of the studie. These methods have been used to gain a deeper understanding of what constitutes visual profiles and logos. Also research of previous work and scientific papers have been read to acquire more knowledge about the topic.

Abstract:
This study investigated the problem of how to assess the visiting public's perceptions of the outcomes of their visit to an Interactive Science and Technology Centre. The study was carried out in several phases. First the researcher immersed himself as a participant observer over 15 months in the day-to-day activities of a science centre. Second, data to describe perceptions of visit outcomes were collected from one-to-one interviews with randomly selected visitors and professional staff, and four focus groups of explainers and visitors. Third, these data were used to structure the collection of further descriptions, in visitors' own words, about the visit experience. In the fourth and fifth stages, data from the second and third stages were pooled to develop two new instruments, the Perceptions of Visit Impact Instrument, a semantic differential survey designed to be administered at the point of exit from a science centre, and the Profile of Visit Outcomes Instrument, a nine part survey designed to be administered several weeks after the visit. The two instruments were refined through local review and field testing, and replication using data from large samples of randomly selected visitors at five and three centres, respectively, in Australia and New Zealand. Staff at these centres found the instruments easy to use and considered that they provided valuable data. Implications are drawn for future use of the instruments for evaluation and research.

Abstract:
Over the last few decades, the progressive reduction in the cost and size of computing resources has contributed to the emergence of different spatiotemporal configurations, making digital information and communication technologies increasingly present in educational scenarios. From the Laboratory of Informatics, based on the desktop model and managed by the Logic of Scarcity, to the mobile technology (notebooks, tablets and smartphones), devices that promoted the exercise of the connectivity, ubiquity and mobility concepts, this displacement allowed that conditions of possibility were conquered, forging the construction of differentiated pedagogical methodologies, but also, in parallel, conditioning a set of competences in the technological scope in the composition of the teacher profile. The conditions of possibilities established by technological mutability, however, they have not presented significant results in terms of qualification of the teaching and learning processes, which has often been evidenced in the underutilization of the potential of computational resources. From the expectation that technology itself will be able to transform this scenario and the absence of approaches that integrate it in a pedagogical perspective of the content, the expectation of the educational innovation mediated by the ICT rests on teachers. In this context, this study analyzes the influence of teacher and school identity elements in the composition of the profile of the Technological Pedagogical Content Knowledge (TPACK) of teachers and in the uses of the educational technology in a private network of teaching of national scope The research, with explanatory objective and qualitative and quantitative approach, was carried out with 606 subjects and 17 schools and is based on two theoretical frameworks, TPACK and Substitution Augmentation Modification Redefinition (SAMR). For the analysis and discussion of the data collected by an online questionnaire, the Discursive Textual Analysis technique and the application of statistical resources were used. The results indicate the existence of a relationship between the TPACK Profile and the SAMR Performance (p < 0.001); The TPACK Profile and the age group (p = 0.001); (p= 0.048), with the teaching qualification in the area of educational technology (p = 0.002) and the teaching area (p < 0.001); as well as a high index of pedagogical practices centered on the teacher and that mobilize the technology in the form of improvement of actions that could be carried out without technological support. The study provides to the scientific community extensions of the TPACK / SAMR models, new instruments with high internal consistency for the analysis of the Teaching Profile, the school methodological and technological context, as well as a mathematical model for the evaluation of the Technological Infrastructure of educational institutions.

This report describes the creative process of the website aiducate.se, which purpose is to support, motivate and guide high school students in their education. The project was conducted independently without any employers and lasted for a period of ten weeks. The basis of the project was a prior study that showed a lack of motivation among high school students. A supplementary survey showed that the students wanted to modernize the education with contemporary teaching methods. As a complement to high school education, the innovation Aiducate was made which in the future will include various tools for the studies. Everything from how to write a report to how to get inspiration. Within the framework of this thesis a simpler beta version of the website was presented. This included a complete graphic profile, responsive design and some features such as a forum and user account. The report also follows the development of the website in the popular CMS tool WordPress. Theoretical background has anchored all final decisions for the website’s design and function, which also reconnects through user testing.

Abstract:
With privacy settings on social networking sites (SNS) perceived as complex and difficult to use and maintain, young adults can be left vulnerable to others accessing and using their personal information. Consequences of not regulating the boundaries their information on SNS include the ability for current and future employers to make career-impacting decisions based upon their online reputation that may include disqualifying them as job candidates. On SNS, such as Facebook, LinkedIn, and Twitter, young adults must decide on how to manage their online reputation by regulating boundaries to their own personal and professional information and identities. One known practice for the regulation of boundaries is the use of multiple profile management (MPM), where users of SNS create and use multiple accounts on a SNS and separate the social and professional identities that they disclose publicly and privately. The purpose of the study was to understand the lived experiences of young adults in how they regulate boundaries on SNS, through the use of MPM, as they manage their online reputation to different audiences. The practice was studied by applying interpretative phenomenological analysis (IPA) through interviewing young adults of 18-23 years of age, who use MPM on a SNS. Semi-structured interviews permitted participants to provide in-depth descriptions of their lived experiences. Eight themes were identified and described based on the analysis of the interviews that include: SNS use with online audiences, motivations for using MPM, the processes for the presentation of self, online search results, privacy settings, untagging SNS posts, self-editing and censorship, and new features. The themes describe the complexity and challenges that young adults face with regulating boundaries with their professional and social identities online through the use of MPM. Findings from this study have implications for a variety of audiences. Through the findings of this study, SNS developers can introduce new features, improve usability related to privacy management, and further encourage use of their networks. Users of SNS can use this study to understand risks of using SNS and for learning of practices for how to manage their online reputation on SNS.

Abstract:
It becomes increasingly more difficult to stand out among the personal sites on the internet today and to become the one who actually gets remembered in the crowd. To be remembered, you need to have a useful website but also a creative appearance added to it. In this practical thesis I have focused on how a website should look like according to users and then added a creative look to the user-friendly website. The purpose of the study is to find out how to connect a useful website with a creative appearance. In order to do that, I first had to find out what users actually want. I used a survey in which I have asked questions about how a website should look like. Using the results of the survey I created prototypes and appearance sketches that I used in user tests. I used the results from the user test and created the final website that represents what users in this study believe is a useful website. To connect the useful website with a creative appearance, I created a short video clip of six seconds to loop on the website's front page. The welcome note describing the site's purpose is reached by means of an arrow, and therefore not directly visible when you enter the site. I have with this created a depth and a creative design solution to the website and thanks to the movement it got more interesting.
